The objective of this study was to determine the effect of a calcium-energy supplement at calving on the incidence of calving-related disorders (CRD), fertility, BCS and milk yield in cows fed anionic diets and to establish any associations among outcome variables. In Florida, from October to December 1997, 479 cows were assigned to three groups and treated at calving as follows: Group 1: 160 nontreated cows; Group 2: 158 cows, treated orally with 60g Ca as CaCl2; Group 3: 161 cows, treated orally with 110g Ca as calcium propionate (510g) plus propylene glycol (400g). No treatment effect was detected for any of the outcome variables. An association was found between dystocia and age and retained fetal membranes (RFM). Age and RFM were associated with metritis. RFM and displacement of the abomasum were associated with ketosis. Ketosis and age were related to displacement of the abomasum. Parity, BCS, ovarian cysts, RFM and metritis were associated with fertility.  相似文献

Nutrient balance affects the resumption of ovarian cyclic activity following calving in dairy cattle. However, few data are available about the relationships between nutrient balance and expression of oestrus or conception. It was hypothesised that previously anoestrous cows that conceived to first insemination and cows that expressed oestrus at the subsequent expected return to oestrus would be less likely to be in negative energy balance (i.e. would have higher body condition score, higher glucose, insulin-like growth factor (IGF), leptin or insulin concentrations, and lower non-esterified fatty acids (NEFA), blood urea nitrogen (BUN), beta hydroxy butyrate (BOH) concentrations 12-15 days after insemination) than herd mates not conceiving or expressing oestrus. Anoestrous cows were treated with progesterone and oestradiol benzoate (Day 0 = end of treatment period) and retrospectively categorised as conceiving (n = 108) or not conceiving (n = 108) to insemination. A subset of cows not conceiving to insemination were categorised as expressing (n = 44) or not expressing (n = 44) oestrus between Days 14 and 28 after initial insemination. Cows conceiving had a lower NEFA concentration (P = 0.014) than non-conceiving cows. Cows subsequently detected in oestrus had higher body condition scores (P = 0.016), IGF concentrations (P = 0.008) and milk protein percentages (P = 0.038), and lower BOH concentrations (P = 0.018) than cows not expressing oestrus. No difference in concentrations of leptin, insulin, glucose, blood urea nitrogen or milk yield were found between cows conceiving or not conceiving and those detected in oestrus or not detected in oestrus (P > 0.1). It is concluded that some measures of metabolic status at the time of pregnancy recognition affects the probability of pregnancy and of subsequent expression of oestrus in those treated, anoestrous cows not conceiving.  相似文献

The objective of this study was to investigate the effect of different body condition score (BCS) at 30 days before calving (-30 days) induced by a differential nutritional management from -100 days until -30 days on productive parameters, the interval to first ovulation and blood parameters in primiparous and multiparous Holstein cows under grazing conditions until 60 days post partum. The experimental arrangement was a randomized complete block design, where cows were blocked according to BW and expected calving date and then randomly assigned to different nutritional treatments from -100 to -30 days relative to calving to induce different BCS. As the assignment of cows to treatments was random, cows had to lose, maintain or gain BCS; thus, different planes of nutrition were offered with approximately 7, 14 or 20 kg dry matter per day. The BCS score was assessed every 15 days and animals were reassigned in order to achieve the desired BCS at -30 days. Only animals that responded to nutritional treatment were considered and this was defined as follows: primiparous and multiparous high cows (PH and MH) had to gain 0.5 points of BCS, primiparous low (PL) had to lose 0.5 points of BCS and multiparous low (ML) had to maintain BCS at least in two subsequent observations from -100 to -30 days. From -30 days to calving, primiparous and multiparous cows (P and M cows) were managed separately and cows were offered a diet once a day. From calving to 60 days post partum, cows of different groups grazed in separate plots a second year pasture. Cows were also supplemented individually with whole-plant maize silage and commercial concentrate. Cows had similar BCS at -100 days and differed after the nutritional treatment; however, all groups presented similar BCS at 21 days post partum. The daily milk production and milk yield at 60 days post partum was higher in M than P cows. The percentage of milk fat was higher in PH cows compared with PL cows. Concentrations of non-esterified fatty acids (NEFA) were affected by the BCS at -30 days within parity, and in PH cows the concentration of NEFA was higher than in PL cows. The concentrations of total protein were higher in M cows. A lower probability of cycling was found in PL than in PH cows (P < 0.05) and in ML than in MH cows (P < 0.05). Treatment affected various endocrine/metabolic profiles according to parity, suggesting that the metabolic reserves signal the productive/reproductive axis so as to induce a differential nutrient partitioning in adult v. first-calving cows.  相似文献

The present study examined ovarian changes preceding the resumption of the ovarian cycle in postpartum dairy cows with different parities under similar body nutritional conditions. In postpartum primi- (n=6), bi- (n=4), and multiparous (n=6) Holstein dairy cows, ovarian ultrasonographic observations starting at 7 days after calving were performed every other day and then daily after the confirmation of clinical signs of oestrus for the detection of postpartum first ovulation. Blood samples were collected at the same time as ultrasonography and analyzed for oestradiol and progesterone to monitor ovarian activity. To evaluate the nutritional condition of the cows, body weight and body condition score (BCS, 1=emaciated to 5=obese) were measured weekly and blood samples for the analysis of glucose, insulin, and non-esterified fatty acid (NEFA) were collected at the same time until postpartum second ovulation. Dominant follicles (>8mm in diameter) of the first follicular wave were detected at 7 days after calving in all cows. The first wave follicle ovulated in five of six multiparous cows, whereas no first wave follicle ovulated in any of the primiparous cows. The days to first ovulation after calving in primiparous cows (31.8+/-8.3 days) were significantly greater (p<0.05) than those in multiparous cows (17.3+/-6.3 days), but were not significantly different from biparous cows (28.8+/-8.6 days). There was a significant relationship between parity and days to first ovulation after calving (p<0.05). BCS was maintained at a level of more than 2.5 during the postpartum period in all cows and there was no influence of parity on postpartum changes in BCS, glucose, insulin, or NEFA throughout the experiment. The present study demonstrated a negative relationship between parity and number of days from calving to first ovulation in dairy cows under similar body nutritional conditions. It is possible that the influence of parity on the resumption of ovarian cycle is modulated by the factors different from the nutrition-related changes during the postpartum period in dairy cows.  相似文献

The objectives of this study were to describe the incidence of postpartum disease and to evaluate the association with serum cholesterol concentrations during the first 3 weeks after calving in grazing dairy cows. The association between non-esterified fatty acids (NEFA), β-hydroxybutyrate (BHBA), calcium and postpartum diseases was also evaluated. A total of 307 Holstein dairy cows from 6 commercial grazing herds in Osorno, Chile, were monitored from calving until 21 days in milk. Cases of retained placenta, clinical hypocalcemia and clinical mastitis were recorded by the farmer using established definitions. Twice weekly, cows were evaluated for metritis by the same veterinarian based on vaginal discharge and body temperature. Postpartum blood samples were collected weekly and analyzed for serum concentrations of cholesterol, NEFA, BHBA and calcium. Cows were considered as having subclinical ketosis if BHBA >1.2 mmol/L, and subclinical hypocalcemia if calcium <2.0 mmol/L in any of the 3 weekly samples. Overall, 56% of the cows studied developed at least one clinical or subclinical disease after calving. Incidence of individual diseases was 8.8% for retained placenta, 4.2% for clinical hypocalcemia, 11.7% for clinical mastitis, 41.1% for metritis, 19.9% for subclinical hypocalcemia and 16.6% for subclinical ketosis. Lower postpartum cholesterol in cows was associated with developing severe metritis or having more than one clinical disease after calving. For every 0.4 mmol/L decrease in serum cholesterol cows were nearly twice as likely to be diagnosed with multiple clinical diseases after calving. Higher BHBA concentrations and lower calcium concentrations during week 1 were associated with severe cases of metritis. Low serum calcium concentration during week 1 was also associated with developing more than one clinical disorder after calving. In conclusion, the incidence of postpartum diseases can be high even in grazing herds and lower serum cholesterol concentrations were associated with occurrence of clinical postpatum disorders.  相似文献

Cows in severe negative energy balance after calving have reduced fertility, mediated by metabolic signals influencing the reproductive system. We hypothesised that transition diet could alter metabolic status after calving, and thus influence fertility. Multiparous dairy cows were assigned to four transition groups 6 weeks pre-calving and fed: (a) basal control diet (n = 10); (b) basal diet plus barley (STARCH, n = 10); (c) basal diet plus Soypass (high protein, HiPROT, n = 11); or (d) no transition management (NoTRANS, n = 9). All cows received the same lactational diet. Blood samples, body weights and condition scores (BCS) were collected weekly. Fertility parameters were monitored using milk progesterone profiles and were not affected by transition diet. Data from all cows were then combined and analysed according to the pattern of post-partum ovarian activity. Cows with low progesterone profiles had significantly lower insulin-like growth factor-I (IGF-I) and insulin concentrations accompanied by reduced dry matter intakes (DMIs), BCS and body weight. Cows with prolonged luteal activity (PLA) were older and tended to have lower IGF-I. Analysis based on the calving to conception interval revealed that cows which failed to conceive (9/40) also had reduced IGF-I, BCS and body weight. Fertility was, therefore, decreased in cows which were in poor metabolic status following calving. This was reflected in reduced circulating IGF-I concentrations and compromised both ovarian activity and conception. There was little effect of the transition diets on these parameters.  相似文献

The objective of this field study with an automatic milking system was to evaluate the effects of omitting the dry period on health and productivity during the subsequent lactation in dairy cows. A total of 98 German Simmental cows of six Southern German farms were assigned randomly to two experimental groups: The first group was dried-off 56 days before calving (D for dried-off, n=49), and the second group was milked continuously during this period until calving (CM for continuous milking, n=49). From the latter a third group emerged, including cows that dried-off themselves spontaneously (DS for dried-off spontaneously, n=14). Blood serum values of glucose, β-hydroxybutyrate (BHBA), non-esterified fatty acids (NEFA) and IGF-1 showed most pronounced fluctuations in D cows. Over the entire study period, the concentrations of BHBA and NEFA were markedly lower in the CM and DS groups. Furthermore, IGF-1 concentration was lowest for D cows and also decrease in back fat thickness was more pronounced. Mean concentration of milk protein was markedly higher in CM and DS cows (3.70% and 3.71%) compared with D cows (3.38%). Owing to the lower 305-day milk yield (−15.6%) and the lower total milk yield (−3.1%), the total amount of produced protein in the subsequent lactation was 2.5% (6.8 kg) lower, although the additional protein amount in CM cows from week −8 to calving was 35.7 kg. The greatest benefit resulted from positive effects on fertility and the lower incidence of diseases: CM cows had their first oestrus 1 week earlier compared with D cows, they also conceived earlier and showed a significantly lower risk of developing hypocalcaemia, ketosis and puerperal disorders. The present study showed that the costs of medical treatment and milk losses were twice as high in D cows, compared with CM and DS cows, and thus the reduced costs because of the more stable health outweighed the financial losses of milk yield by +18.49 € per cow and lactation.  相似文献

The objective of this study was to investigate the relationships among body condition score (BCS), body weight and plasma metabolite concentrations, and resumption of ovarian cycles postpartum in high-producing dairy cows in a subtropical region in Japan. The study was conducted between May 2001 and January 2003 in a commercial dairy farm in Hiroshima. Cows were kept in free-stall barns with sawdust and wood shavings for bedding, and fed a total mixed ration consisting of alfalfa, timothy and oat hay, corn, tofu ground-wet, beet pulp, cottonseed and soybean. Milk samples were collected twice weekly from 2 to 11 weeks postpartum, and progesterone concentrations in skim milk were determined by ELISA. At 8 and 2 weeks before calving and 3, 5, 7, 9, and 11 weeks after calving, both BCS and body weights were determined and blood samples were collected to determine plasma concentrations of total cholesterol (T-cholesterol), non-esterified fatty acids (NEFA), total protein, and urea nitrogen. The average 305 days milk yield was 10,422 kg. Out of 110 cows, 44.5% had normal resumption (ovulation occurred < or =45 days after calving, followed by regular ovarian cycles), 33.6% had a prolonged luteal phase (luteal activity >20 days), 12.7% had delayed first ovulation (no ovulation until < or =45 days after calving), 4.6% had a short luteal phase (luteal activity <10 days, except in the first cycle), and the remaining 4.6% had cessation of cyclicity (no luteal activity > or =14 days between cycles). The BCS of cows with delayed first ovulation were lower than the BCS of cows with normal resumption and prolonged luteal phase at 5 weeks (2.9+/-0.1 versus 3.1+/-0.1 and 3.2+/-0.1; P<0.05), 7 weeks (2.8+/-0.1 versus 3.1+/-0.1 and 3.2+/-0.1; P<0.01), 9 weeks (2.7+/-0.1 versus 3.0+/-0.1 and 3.2+/-0.1; P<0.01) and 11 weeks (2.6+/-0.1 versus 3.0+/-0.1 and 3.2+/-0.1; P<0.01) after calving. None of the plasma parameters differed (P>0.05) among different groups of cows. Cows loosing > or =1 unit BCS after calving had a prolonged interval to commencement of luteal activity, and were at greater risk of having delayed first ovulation. Poor postpartum nutritional status was associated with delayed first ovulation postpartum. Regular monitoring of BCS before and after calving would be useful in nutritional management, and help in preventing delayed first ovulation in high-producing dairy cows.  相似文献

The study was aimed at assessing whether the peri-parturient period is associated with changes of intracellular and plasma inducible heat shock proteins (Hsp) 72 kDa molecular weight in dairy cows, and to establish possible relationships between Hsp72, metabolic, and immunological parameters subjected to changes around calving. The study was carried out on 35 healthy peri-parturient Holstein cows. Three, two, and one week before the expected calving, and 1, 2, 3, 4, and 5 weeks after calving, body conditions score (BCS) was measured and blood samples were collected to separate plasma and peripheral blood mononuclear cells (PBMC). Concentrations of Hsp72 in PBMC and plasma increased sharply after calving. In the post-calving period, BCS and plasma glucose declined, whereas plasma nonesterified fatty acids (NEFA) and tumor necrosis factor-alpha increased. The proliferative responses of PBMC to lipopolysaccharide (LPS) declined progressively after calving. The percentage of PBMC expressing CD14 receptors and Toll-like receptors (TLR)-4 increased and decreased in the early postpartum period, respectively. Correlation analysis revealed significant positive relationships between Hsp72 and NEFA, and between PBMC proliferation in response to LPS and the percentage of PBMC expressing TLR-4. Conversely, significant negative relationships were found between LPS-triggered proliferation of PBMC and both intracellular and plasma Hsp72. Literature data and changes of metabolic and immunological parameters reported herein authorize a few interpretative hypotheses and encourage further studies aimed at assessing possible cause and effect relationships between changes of PBMC and circulating Hsp72, metabolic, and immune parameters in dairy cows.  相似文献

Hypocalcaemia is a common metabolic disorder of post-parturient dairy cows; enhancement of our knowledge regarding Ca dynamics would improve cow health and welfare. This study investigated the presence of time- and severity-related subclinical hypocalcaemia (SCHCa) patterns in Holstein cows after calving and their association with diseases during the first week of lactation. A total of 938 cows from nine herds were blood sampled on days 1, 2, 4 and 8 post-partum, and serum Ca concentration was measured with atomic absorption. Subclinical ketosis (serum β-hydroxybutyrate≥1.2 mmol/L) and cases of retained foetal membranes, metritis (MET), mastitis, ketosis and displaced abomasum (DA) were recorded. Using receiver operating characteristic (ROC) analysis, the SCHCa cut-off was identified at serum Ca ≤ 2.09 mmol/L. Hierarchical and two-step cluster analysis classified cows to seven clusters based on test day cow Ca records and overall SCHCa status. Two clusters (NORM and HIGH) included 318 normocalcaemic cows and five clusters (SCH-1 to −5) those that were at least once subclinically hypocalcaemic (n = 620). A second ROC analysis was performed in order to distinguish mild from severe cases of SCHCa in these 620 cows; this cut-off was identified at 1.93 mmol/L. The associated risk of disease with Ca clustership was assessed with generalized linear mixed models. Overall incidence of SCHCa was 66.1%. Clusters SCH-1 and SCH-2 included cows with short-term SCHCa of day 1 and day 2, respectively, while SCH-3 included cows with severe and prolonged SCHCa extending to day 4 and beyond. Clusters SCH-4 and SCH-5 included cows with delayed SCHCa, which culminated on days 4 and 8, respectively. Compared to NORM cows in HIGH had lower risk of MET and no cases of DA. Cows in SCH-3 had higher risk of being diagnosed with retained foetal membranes, DA or any disease during the study period. Cows in SCH-5 had higher risk of being diagnosed with ketosis, subclinical ketosis or any disease. In conclusion, there are multiple normocalcaemic and hypocalcaemic patterns that are differently associated with disease risk.  相似文献

The objective of the present study was to quantify the relationships among body condition score (BCS; scale 1 to 5), live weight (WT) and milk production in Irish Holstein-Friesian spring calving dairy cows. Data were from 66 commercial dairy herds during the years 1999 and 2000. The data consisted of up to 9886 lactations with records for BCS or WT at least once pre-calving, or at calving, nadir or 60 days post-calving. Change in BCS and WT was also calculated between time periods. Mixed models with cow included as a random effect were used to quantify the effect of BCS and WT, as well as change in each trait, on milk yield, milk fat concentration and milk protein concentration. Significant and sometimes curvilinear associations were observed among BCS at calving or nadir and milk production. Total 305-day milk yield was greatest in cows calving at a BCS of 4.25 units. However, cows calving at a BCS of 3.50 units produced only 68 kg less milk than cows calving at a BCS of 4.25 units while cows calving at 3.25 or 3.00 BCS units produced a further 50 and 114 kg less, respectively. Cows that lost more condition in early lactation produced more milk of greater fat and protein concentration, although the trend reversed in cows that lost large amounts of condition post-calving. Milk yield increased with WT although the marginal effect decreased as cows got heavier. Milk fat and protein concentration in early lactation also increased with WT pre-calving, calving and nadir, although WT did not significantly affect average lactation milk fat concentration.  相似文献

The purpose of this survey was to evaluate the reproductive performance of dairy cows on problem farms in southern Iceland. In all 229 cows on 6 farms were studied. The animals were examined clinically by rectal palpation, once a month. Blood samples were taken 2–5 and 7–10 weeks after calving. The blood samples were analysed for the contents of glucose, urea, inorganic phosphate, calcium and magnesium. Milk samples for progesterone profiles were taken, by the farmer, every 4th day from day 10 postpartum until first oestrus. Because of this sampling method, 128 cows had no rise in progesterone levels, when milk sampling was stopped. These 128 animals were excluded from the study. The results are based on 101 animals. There was a large variation between cows in postpartum reproductive performance. In the total material 1st ovulation occurred later than reported in many other countries. Fifty percent of the cows had ovulated 35 days after calving and 90 percent 70 days after calving. The first luteal phase was short in about 60 % of the cows. The progesterone values assayed from those short cycles were lower than the values assayed from the following cycle. First artificial insemination (ai) was on the average 77 days postpartum (pp). The conception rate to first service was 49 %. Of 100 milk samples taken at the time of ai, 20 had high progesterone value. This indicates a high frequency of luteal phase inseminations. Clinical ketosis was diagnosed in 35 cows. Of these, 31 had a low glucose value. Cows with clinical ketosis ovulated, on the average, later than other animals and 24 ovulated later than 40 days pp. The results indicated that the fertility problems of these cows studied were late ovulations, low conception rate, probably in part, due to luteal phase inseminations, and a high frequency of ketosis which could be caused by low quality feedstuff.  相似文献

The endometrium regulates the inflammatory response after infection by production and release of cytokines and chemokines. The objective was to compare gene expression of important pro-inflammatory (TNFα, IL-1β, IL-6) and anti-inflammatory (IL-10) cytokines, and the main neutrophil chemokine (IL-8), from calving to Week 7 after calving, in cows that developed endometritis and healthy control cows. Uterine biopsies were obtained at calving and at Weeks 1, 3, 5 and 7. Endometritis was evaluated at Week 5 by uterine lavage and cytology; cows with ≥ 10% neutrophils were considered to have endometritis. Real-time RT-PCR threshold values (Ct) were used to calculate the fold difference in gene expression, using the 2−ddCt method, normalized to GAPDH and calibrated to the average dCt for all cows at calving. Serum IL-8 concentrations were measured with ELISA. The analysis included 28 cows (11 had endometritis) for the PCR data and 44 cows (20 had endometritis) for ELISA. Expression of the TNFα gene in uterine tissue was decreased in cows with endometritis compared to control cows at calving (P = 0.09) and at Week 1 (P = 0.05). Iterleukin-1β gene expression tended to be decreased (P = 0.08) in cows with endometritis compared to control cows at Week 1, but tended to be increased (P ≤ 0.10) at Weeks 5 and 7. Cows with endometritis had increased (P < 0.05) IL-6 gene expression at calving and at Week 7 compared to control cows. Interleukin-8 gene expression was increased (P = 0.03) in endometritic cows compared to control cows at Week 7. Uterine disease was not significantly associated with IL-10 gene expression. A lower local level of expression of pro-inflammatory cytokines in the endometrium soon after calving might impair activation of inflammation and clearance of bacteria, and lead to development of endometritis.  相似文献

The objective was to determine the effect of oxytocin treatments after calving on the incidence of RFM and reproductive performance in dual purpose cows under tropical conditions. Five hundred thirty six pluriparous, crossbred Zebu cows were randomly assigned to two groups: Oxy (n = 280): cows were given 30 IU of oxytocin im immediately after normal unassisted calving, and again 6 h later; C (n = 256): control. Expulsion of fetal membranes was evaluated 24 h after delivery. After a 30-d voluntary waiting period, AI was done 12 h after cows were detected in estrus. Oxytocin had no effect on the incidence of RFM (4.6 vs. 3.1% for Oxy and C, respectively, P > 0.05). Cows in Oxy and C had similar first service and overall pregnancy rates (54.0 vs. 47.8% and 75.4 vs. 73.4%; respectively, P > 0.05). There were no differences between Oxy and C for calving to first estrus (83.6 ± 3.7 vs. 77.2 ± 3.8 d) and calving to conception intervals (113.6 ± 5.0 vs. 110.5 ± 5.2 d), as well as rates of anestrus (13.6 vs. 13.7%), repeat breeding (21.8 vs. 20.7%), and culling (15.7 vs. 16.4%). In conclusion, oxytocin treatment after normal unassisted calving did not significantly reduce the incidence of RFM or improve reproductive performance in crossbred Zebu cows under tropical conditions.  相似文献

The overall reproductive performance has decreased over the last decades, involving changes in cyclicity, oestrous behaviour and fertility. High milk yield (MY), low body condition score (BCS) and large body condition (BC) loss have been identified as risk factors. However, these effects are often confounded, as high MY and body lipid reserve mobilization are correlated. The aim of this study was to evaluate the respective effects of MY and BC on post-partum ovarian cyclicity, oestrus and fertility of Holstein cows. This study provides novel information, as MY and BC change were uncoupled in the overall dataset that included 98 lactations and milk progesterone profiles. Cows were assigned to two feeding-level groups: high feed, which achieved high MY and moderate BC loss throughout lactation (8410 kg, -1.17 unit from calving to nadir BCS), and low feed, which limited MY and triggered a large BC loss (5719 kg, -1.54 unit). MY and BC had different effects at different stages of the reproductive process. Cyclicity as well as non-fertilization and early embryo mortality were mainly driven by body lipid reserves, whereas oestrous behaviour and late embryo mortality were related to MY. The results point to possible uncoupling between cyclicity, oestrus and early and late embryo survival allowing compensation along the reproductive process and leading to similar final reproductive performance. In compact calving systems, which require high pregnancy rates within a short period, higher MY strategies appear unsuitable even where BCS is maintained, owing to depressed oestrous behaviour and probably increased late embryo mortality, which delays rebreeding. Similarly, strategies that compromise cyclicity and fertility by excessively low BCS are unsuitable.  相似文献

The hypothesis was that supplementation during the late prepartum period will differentially affect reproductive and productive variables according to parity. Primiparous (n=22) and multiparous (n=22) pregnant autumn calving Holstein cows were stratified in two groups according to parity (primiparous or multiparous) and within each group were randomly assigned to two treatments: (a) low supplemented (LS) or (b) high supplemented (HS) prepartum diet. The LS group was offered 5.2 kg/cow/day (DM basis) of wheat silage, and the HS group 4.7 kg cow/day (DM basis)/of corn silage and 3.6 kg (DM basis) of wheat bran+12 g of urea. Both groups grazed on natural pastures. After calving, all cows received the same diet. The experimental period was from 3 weeks before calving to 7 weeks postpartum (PP); body condition score (BCS) and blood samples for hormonal analyses were obtained weekly and ovarian ultrasonography was conducted three times per week. The loss in BCS around calving was less pronounced in HS cows, but only multiparous supplemented cows maintained BCS throughout the study. Non-esterified fatty acids (NEFA) increased during the prepartum period in the LS but not in the HS cows, with peak values occurring on day 14 PP in all groups. During the remainder of the experiment NEFA was greater in LS than in HS cows. Prepartum treatment did not affect the proportion of cows that had ovulations from the first dominant follicle postpartum, but decreased the interval to first ovulation in multiparous cows (22.9 compared with 38.2 days; P<0.05). This was associated with greater plasma IGF-I concentrations at the time the dominant follicle of the first follicular wave reached its maximum diameter (8.0 compared with 3.6 nmol/L; P<0.05). However, prepartum treatment had no effect on onset of ovarian activity in primiparous cows. Supplementation had no effect on milk production or milk protein percentage but increased milk fat percentage. We conclude that feeding a high-supplemented prepartum diet to multiparous cows allowed them to maintain BCS around calving, and this was associated with greater concentrations of IGF-I and an earlier onset of estrous cycles after calving.  相似文献

The transition from gestation to lactation is marked by significant physiological changes for the individual cow such that disease incidence is highest in early lactation. Around the time of calving, cows rely on mobilisation of body energy reserves to fill the energy deficit created by an increase in nutrient demands at a time of restricted feed intake. It is well established that monitoring of body energy reserves in lactation is an important component of herd health management. However, despite their influence on future health and productivity, monitoring of body energy reserves in the dry period is often sparse. Further, there is increasing concern that current dry off management is inappropriate for modern cattle and may influence future disease risk. This study aimed to identify candidate indicators of early lactation production disease from body energy data collected in the dry period and production data recorded at the time of dry off. Retrospective analysis was performed on 482 cow-lactations collected from a long-term Holstein-Friesian genetic and management systems project, the Langhill herd in Scotland. Cow-lactations were assigned to one of four health groups based on health status in the first 30 days of lactation. These four groups were as follows: healthy, reproductive tract disorders (retained placenta and metritis), subclinical mastitis and metabolic disorders (ketosis, hypocalcaemia, hypomagnesaemia and left displaced abomasum). ANOVA, employing a GLM was used to determine effects for the candidate indicator traits. Cows which were diagnosed with a reproductive tract disorder in the first 30 days of lactation experienced a significantly greater loss in body energy content, body condition score and weight in the preceding dry period than healthy cows. The rate of change in body energy content during the first 15 days of the dry period was −18.26 MJ/day for cows which developed reproductive tract disorder compared with +0.63 MJ/day for healthy cows. Cows diagnosed with subclinical mastitis in the first 30 days of lactation had significantly greater milk yield at dry off in the previous lactation than cows that developed a reproductive tract disorder or metabolic disease in addition to a significantly higher yield to body energy content ratio at dry off than healthy cows. Physiological and production traits recorded in the lactation and dry period preceding a disease event differed between cows which developed different diseases post-calving. Differences in these traits allow the development of new disease indicators for use in models for the prediction of disease risk in the transition period.  相似文献

Relationships among GH genotype (AluI polymorphism), parity, metritis and interval from calving to first ovulation, milk production and body condition score (BCS) loss were determined in dairy cows (n = 307) on four large-scale farms in Hungary. Cows with systemic signs of puerperal metritis or mastitis were excluded. Time of the first postpartum (PP) ovulation was obtained from milk progesterone profiles. Based on GH genotype determination, groups of leucine homozygous cows (n = 246) and valine allele carriers (n = 61) were formed. All animals became cyclic during the study period. The average interval to first ovulation was 27.6 ± 0.69-d PP (mean ± S.D.). Genotype had no effect on the commencement of ovarian cyclicity. First ovulation occurred sooner after calving in pluriparous than in primiparous cows. The greater BCS loss cows had during the first 30-d PP, the longer they took to resume cyclic ovarian function. The interval from calving to first ovulation was substantially affected by farm, but not by mild cases of puerperal metritis. Genotype was not related to cumulative 30-d milk yield or BCS loss after calving. Primiparous cows had lower milk yield than pluriparous ones. Cows with metritis lost more body condition than healthy individuals in the first month postpartum. We concluded that, under field conditions, AluI polymorphism of the bovine GH gene had no effect on the interval from calving to first ovulation and could not be directly related to differences in milk yield and to the extent of BCS loss during the first month after calving in Holstein-Friesian cows.  相似文献

Dairy cows are highly susceptible after parturition to developing liver lipidosis and ketosis, which are costly diseases to farmers. A bovine microarray platform consisting of 13,257-annotated oligonucleotides was used to study hepatic gene networks underlying nutrition-induced ketosis. On day 5 postpartum, 14 Holstein cows were randomly assigned to ketosis-induction (n = 7) or control (n = 7) groups. Cows in the ketosis-induction group were fed at 50% of day 4 intake until they developed signs of clinical ketosis, and cows in the control group were fed ad libitum throughout the treatment period. Liver was biopsied at 10-14 (ketosis) or 14 days postpartum (controls). Feed restriction increased blood concentrations of nonesterified fatty acids and beta-hydroxybutyrate, but decreased glucose. Liver triacylglycerol concentration also increased. A total of 2,415 genes were altered by ketosis (false discovery rate = 0.05). Ingenuity Pathway Analysis revealed downregulation of genes associated with oxidative phosphorylation, protein ubiquitination, and ubiquinone biosynthesis with ketosis. Other molecular adaptations included upregulation of genes and nuclear receptors associated with cytokine signaling, fatty acid uptake/transport, and fatty acid oxidation. Genes downregulated during ketosis included several associated with cholesterol metabolism, growth hormone signaling, proton transport, and fatty acid desaturation. Feed restriction and ketosis resulted in previously unrecognized alterations in gene network expression underlying key cellular functions and discrete metabolic events. These responses might help explain well-documented physiological adaptations to reduced feed intake in early postpartum cows and, thus, provide molecular targets that might be useful in prevention and treatment of liver lipidosis and ketosis.  相似文献
