The fine structure of the esophagus, including procorpus, metacorpus, isthmus, gland lobe, and esophago-intestinal junction, is examined in males of Sarisodera hydrophila. A cuticle-lined lumen extends most of the length of the esophagus, broadens to form a pump chamber in the metacorpus, and posteriorly is continuous with junctional complexes among four esophago-intestinal cells. These four cells are partially enveloped by the gland lobe which basically consists of three gland cells, one dorsal and two subventral. Each gland cell has an anterior process which opens into the lumen of the esophagus through a cuticle-lined duct. The dorsal gland joins the lumen in the anterior portion of the procorpus, whereas ducts of the subventral glands terminate at the base of the metacorpus pump chamber. The subventral glands are predominant in the posterior portion of the gland lobe and are partially ensheathed by a narrow portion of the dorsal gland which extends to within 5 μm of the posterior terminus of the gland lobe. Contents of the dorsal gland include primarily electron dense granules, although rough endoplasmic reticulum (RER) is predominant posteriorly. Secretory granules within the subventral glands vary in morphology and are evenly distributed throughout the two ceils among other organelles, including RER and a large Golgi apparatus. Innervation of the esophagus includes nerve processes which originate from several perikaryons (cell bodies) located in the anterior portion of the gland lobe. The esophagus of males of S. hydrophila is compared with that of other Heteroderoidea, Heterodera glycines and Meloidogyne incognita.  相似文献

Summary The dorsal and subventral esophageal glands and their secretory granules in the root-knot nematodeMeloidogyne incognita changed during parasitism of plants. The subventral esophageal glands shrank and the dorsal gland enlarged with the onset of parasitism. While secretory granules formed by both types of glands were spherical, membrane-bound, and Golgi derived, the granules differed in morphology and size between the two types of glands. Subventral gland extensions in preparasitic second-stage juveniles were packed with secretory granules which varied in diameter from 700–1,100 nm and had a finely granular matrix. Within the matrix of each subventral gland granule was an electron-transparent core that contained minute spherical vesicles. The size and position of the core varied within different granules. Few granules were present in the dorsal gland extension in preparasitic juveniles. The matrix of dorsal gland secretory granules formed during parasitism was homogeneous and more electron-dense than the matrix of subventral gland granules. Subventral gland secretory granules of parasitic juveniles and adult females appeared degenerate.  相似文献

The stomatostylet of Criconemoides curvature consists of three parts: tooth cone, shaft, and knobs. The tooth cone constitutes the outer conical covering and inner lining of the anterior half of the stylet lumen. The tooth cone is easily separated from the shaft by treating an isolated styler with 0.5% sodium hypochlorite. The posterior half of the shaft is cylindrical, tapering anteriorly to form the shaft extension, wedged between the inner and outer tooth cone. The shaft extension extends to the stylet lumen orifice, which is subterminal and ventral. Six ducts enter the shaft through the junction between the shaft and knobs. They extend anteriad toward the tip of the shaft extension. Cytoplasmic connections between the ducts and the cells surrounding the stylet occur near tile junction between the shaft and the basal knobs. Ribosome and membranous structures are observed in these ducts. The esophagus of the adult female consists of a fused procorpus and metacorpus with a large valve possessing thickened cuticular walls at the anterior and posterior ends. The dorsal esophageal gland reservoir is composed of many honeycomb-like compartments made up of two types of differing electron density. The subventral esophageal glands, however, consist of only one type of granules. Both dorsal and subventral esophageal glands open into the esophageal lumen through trachea-like branched multiple canals.  相似文献

Monoclonal antibodies to secretory granules in the dorsal or subventral esophageal glands were generated by injecting BALB/c mice with immunogens from preparasitic second-stage juveniles (J2) of Meloidogyne incognita. Antibodies specific for secretory granules in the J2 subventral esophageal glands or the dorsal gland were identified by indirect immunofluorescence microscopy. Only antibodies that reacted with granules in the J2 dorsal gland reacted with the esophageal gland lobe ofM. incognita adult females. The antibodies also reacted with secretory granules in both types of esophageal glands in M. javanica and M. arenaria J2 but not with granules in esophageal glands of Heterodera glycines J2.  相似文献

Ultrastructural cytochemical tests for several enzymes, proteins, carbohydrates, and nucleic acids were conducted on secretory granules o£ dorsal and subventral esophageal glands of preparasitic second-stage juveniles and the dorsal gland of adult females of Meloidogyne incognita. Secretory granules in the subventral glands of juveniles stained positive for acid phosphatase. Peroxidase, DNase, RNase, cellulase, and nucleic acids were not detected in these granules. Secretory granules in the dorsal gland of adult females stained positive for peroxidase (pH 7.6) in < 50% of the tests, Acid phosphatase, β-glucuronidase, DNase, RNase, polyphenoloxidase, cellulase, and carbohydrates were not detected in dorsal gland granules in adult females. Positive staining with cobalt thiocyanate, a stain for amino groups of basic proteins, occurred in secretory granules in the dorsal gland, ribosomes, and chromatin in adult females. Ribosomes, nuclei, and secretory granules of the dorsal gland of adult females intensely stained when incubated in three reagents specific for nucleic acid.  相似文献

Fine structure of the stoma, including the cheilostom, gymnostom, and stegostom of Bunonema sp. and Teratorhabditis palmarum was compared with Caenorhabditis elegans to consider fine structural characters that may be phylogenetically informative. The stegostom, enclosed by the anterior end of the pharynx, includes a triradiate lumen surrounded by radial cells (interradial or pairs of adradial cells) repeated in the dorsal and subventral sectors; in Rhabditina, typically the stegostom includes anteriorly two sets of epithelial and posteriorly two sets of muscular radial cells. These muscle cells are anteriorly m1 and posteriorly m2. In Bunonema sp., unlike T. palmarum and C. elegans, the stegostom has a third set of interradial epithelial cells. In Bunonema sp., m1 is expressed by three interradial cells, whereas in T. palmarum and C. elegans m1 is three pairs of adradial muscle cells (i.e., six cells). In all three taxa m2 is expressed as three pairs of adradial muscle cells. Posterior processes of adjacent adradial cells fuse, and closely apposed nuclei may present a figure-eight shape. However, in Bunonema the three interradial m1 cells each have a long posterior process enclosing two separate round nuclei. In combination with additional characters, these diverse stoma features may prove phylogenetically informative. Specifically, the radial epithelial cells of the stegostom appear to be a synapomorphy consistent with a bunonemid-diplogastrid-rhabditid clade, whereas a thickening in the dorsal sector of the stoma cuticle lining is interpreted as a synapomorphy supporting a bunonemid-diplogastrid clade.  相似文献

The ultrastructure of the dorsal oesophageal gland ampulla and its relationship with the oesophagus, oesophageal ultrastructure, and control mechanisms in oesophageal activity were studied. Terminal ducts of the sub-ventral glands open through the oesophageal crown at the base of the buccal cavity. The terminal duct of the dorsal oesophageal gland running through the dorsal gutter opens to the exterior at the rim 'groove' of the buccal capsule. The posterior oesophageal region is clavate and the cuticle of the lumen folds to form outlet valves, 'valvulae'. An inconspicuous oesophago-intestinal valve (three lobes) connects oesophagus and intestine and is visualized in the open and shut position. In the female reproductive tract, with the exception of the uterus, the cells lie on a thick, irregular (convoluted) basal lamina. The apical plasma membrane of the uterus, and seminal receptacle, extend into the lumen by microvilli-like projections with which spermatozoa make intimate contact. The lumen of the uterus is filled with oocytes, fertilized and unfertilized. Testicular cells have two parts linked by a rachis. Spermatocytes are elongated with a large nucleus, distinct nuclear membrane, and many granules. The apical membrane of the rachis forms long microvilli-like projections with balloon-like tips. The amoeboid spermatozoa contain membrane specializations, a nucleus devoid of a membrane, and are enclosed by a pseudopodial-like extension.  相似文献

Three monodonal antibodies (MAbs) that bound to secretory granules within the subventral esophageal glands of second-stage juveniles (J2) of the soybean cyst nematode (SCN), Heterodera glycines, were developed from intrasplenic immunizations of a mouse with homogenates of SCN J2. Two MAbs to the secretory granules within subventral glands and one MAb to granules within the dorsal esophageal gland of SCN J2 were developed by intrasplenic immunizations with J2 stylet secretions. Stylet secretions, produced in vitro by incubating SCN J2 in 5-methoxy DMT oxalate, were solubilized with a high pH buffer and concentrated for use as antigen. Three of the five MAbs specific to the subventral esophageal glands bound to stylet secretions from SCN J2 in immunofluorescence and ELISA assays. Two of these three MAbs also bound to secretory granules within both the dorsal and subventral esophageal glands of young SCN females. All five of the subventral gland MAbs bound to the subventral glands of Heterodera schachtii and one bound to the subventral glands of Globodera tabacum, but none bound to any structures in Meloidogyne incognita or Caenorhabditis elegans.  相似文献

The feeding apparatus of the marine nematode Rhabdodemania minima Chitwood, 1936 has been examined with light and transmission electron microscopy. The buccal capsule consists of a posterior region with smooth walls bearing three sets of three minute denticles at its posterior end and three large onchia in its mid region; a middle region with grooved walls; and an anterior region with costae and six odontia. The anterior and middle portions of the buccal capsule are enveloped by the cephalic cuticle, whereas the posterior region, which is set off from the middle region by a buccal seam, is partially enveloped by the anterior end of the esophagus. Two subventral esophageal glands open into the lumen of the esophagus. Secretions of each of three paraesophageal glands are conveyed through a duct in each of the three corresponding corners of the buccal wall to an opening between labia. A pair of wing-like thickenings, termed pterons, embraces the duct of each paraesophageal gland in the posterior and middle regions of the buccal capsule. A model of how the buccal capsule operates is proposed and tested. Morphological and functional aspects of the buccal apparatus and cephalic cuticle are compared with those of other taxa of the Enoplida, and their phylogenetic implications are discussed.  相似文献

Summary The structure of the male reproductive systems of two species ofHaplognathia cf.lyra andH. cf.rosacea was described. The structure of the testes and the anterior portions of the sperm ducts in both species was found to be similar. However, considerable species differences were found between the structures of the glands and muscles associated with the reproductive systems. These were more elaborate inH. cf.lyra than inH. cf.rosacea. The former species possessed an H-shaped sperm duct gland, three distinct groups of penis muscles and a penis with two cell types and with a lumen. The latter species had paired sperm duct glands, no specialized penis muscles and a penis with only one cell type and without a detectable lumen. No open gonopore was observed in either species. The sperm presumably exit through a ventral tissue connection observed connecting the penis and the ventral epidermis. Timema is the most basal genus of Phasmatodea and the sister group to the remaining stick and leaf-insects (Euphasmatodea). An autapomorphy of all phasmids is the paired prothoracic exocrine defence glands. In this study, the anatomy and innervation of the defence glands in Timema petita and Timema chumash are described and compared with the data on Euphasmatodea. In all phasmids, the glands consist of a cuticular epithelium, a secretory epithelium and muscular fibres that compress the lumen. In Timematodea, the muscular part of the gland is less developed than in Euphasmatodea and the ejection of the defence secretion depends on the dorsal longitudinal neck muscles. On the neuroanatomical level, Timema petita and Timema chumash lack neurons that are involved in the independent contraction of the gland in euphasmids. In both studied species of Timema, neck muscles play an active role in the gland function which is not observed in any other phasmid. Considering the basal position of this genus, this supports the hypothesis that in euphasmids, the muscular part of the gland evolved from the dorsal longitudinal neck muscles. Additionally, the same nerves that innervate the dorsal longitudinal neck muscles in all Polyneoptera also innervate the defence glands in phasmids.  相似文献

The ultrastructure of the anterior body region of the free-living marine nematode Deontostoma calilornicum was studied by electron microscopy. The body wall consists of a nine-layered cuticle, a cellular hypodermis containing eight nerve bundles, and a well-developed coelomyarian somatic musculature. Nerves in the dorsal, lateral, ventral, and submedian hypodermal chords anterior to the nerve ring were observed with regularity. Structure of subventral somatic setae suggests a mechanoreceptive function. The esophagus is cellular and consists of three marginal cells alternating with an equal number of radial muscle cells, three esophageal glands, and three enteric nerves. The membranes of adjacent esophageal cells are sinuous. Apices of the triradiate lumen are connected with the outer wall of each marginal cell by bands of electron-dense nonmyofibrils, whereas two types of myofilaments run radially between the apophyses of the lumen and the outer walls of radial cells. Each myofibril, which forms hemidesmosomes at both ends, is interpreted to be the morphological equivalent of one sarcomere. Synaptic junctions between the processes of muscles, gland cells, and axons of the enteric nerves are described in detail.  相似文献

Parodontophora limnophila sp. nov. is described from Poyang Lake, the largest freshwater lake of China. It is characterized by having an amphid with its posterior end close to the base of the stoma, relatively short cephalic setae, opisthocephalic setae arranged as two subdorsal groups of three longitudinally arranged setae and two single subventral setae, excretory pore at the level of the anterior part of the stoma and renette gland 34–47% of the oesophageal length. To date, the new species is the only Parodontophora species found in freshwater habitats.  相似文献

Differential interference contrast, transmission electron and epifluorescence microscopy techniques were employed to examine the ultrastructure of the rectal glands in Heterorhabditis bacteriophora hermaphrodites, with special attention to the location of Photorhabdus bacteria symbionts within these structures. Three rectal glands were clearly visualized in all examined specimens, with two glands positioned sub-ventrally and another gland located dorsally. The dorsal rectal gland in all examined specimens is larger than the subventral ones. Our observations indicate that Photorhabdus bacteria do not colonize the rectal glands of H. bacteriophora hermaphrodites, but rather are present in the most posterior-intestinal cells.  相似文献

The organization of the salivary glands in ad libitum-fed adult females of the microtrombidiid mite Platytrombidium fasciatum (C. L. Koch, 1836) was observed using transmission electron microscopy. In all, four pairs of large simple alveolar salivary glands were determined, which have been named due to their position as posterior, ventral, medial and dorsal. These glands occupy a body cavity behind, around the base and partly inside the gnathosoma. The posterior glands are largest and possess large nuclei with greatly folded nuclear envelope. Secretory granules are electron-light, containing fine granular material and are partly provided with various lamellar inclusions inside the granules. The latter tend to be placed predominantly in the middle parts of the gland around the central (intra-alveolar) cavity. The remaining glands, conversely, are typically filled with tightly packed electron-dense secretory granules, except for the ventral glands, the granules of which may show a compound organization. The nuclei of all these glands occupy a peripheral position and are mostly pressed between the granules. No prominent endoplasmic reticulum or conspicuous Golgi bodies were observed within the salivary glands. The salivary glands are provided with a complex apparatus of the intra-alveolar cavity (acinar lumen) with the excretory duct base provided by a system of branched special cells producing the duct walls. The ventral glands open by separate ducts into the most posterior part of the subcheliceral space. Ducts of the posterior glands immediately fuse with the ducts of the tubular (coxal) glands. The common duct of each side of the body joins with the ducts of the medial and dorsal glands respectively, and opens into the subcheliceral space far anterior to that of the ventral glands.  相似文献

Histolysis of pupal salivary glands and their further development during metamorphosis of Wilhelmia lineata were investigated by light microscopy. After pupal-adult apolysis the distal parts of the glands degenerate. The adult gland develops from a ring of tightly packed very small cells with small nuclei at the anterior end of the degenerating pupal gland. The gland of the female pharate adult consists of a spherical accessory gland with small cells and oval nuclei and a little U-shaped main gland with larger cells and round nuclei. The two U-shaped lobes are orientated horizontally. The gland of the male pharate adult has the same shape and cellular structure as the accessory gland of the female. During further development the main gland becomes greatly elongated as the cells increase in size, the cells become flattened, and the secretion of saliva into the gland lumen can be seen. The glands are fully differentiated at the time of pupal-adult ecdysis. Soluble proteins from the one-day-old female pharate adult show a very similar pattern to those of pupal and male glands. With growth of the female main gland and secretion of saliva into the gland lumen increasing amounts of proteins, which have been characterized earlier (Poehling et al., 1976) occur in the gland. This could suggest a specific rôle of these proteins during bloodsucking in the adult females.  相似文献

The structure of the pharynx of the adult female nematode Anguillicoloides crassus (Spirurina) has been studied for the first time using light and transmission electron microscopy. The cylindrical pharynx consists of a short anterior muscular corpus and an enlarged posterior glandular and muscular postcorpus. The main cellular components of the pharynx of A. crassus include the muscle cells, the marginal cells, the nerve cells, and 1 dorsal and 2 subventral glands. New observations for nematodes include: (1) the non-contractile regions of pharyngeal musculature in the corpus have specific appearance; (2) the ventrosublateral longitudinal nerve in the pharynx has an enlarged, enucleated anterior part, with a pronounced palmate projections; and (3) abundant lysosomelike membranous bodies consisting of myelinlike figures of varied size present in marginal cells and pharyngointestinal valve. The 2 subventral glands and, apparently, the single dorsal gland, have their openings at the same level, i.e., at the border between the corpus and postcorpus. The pharyngeal-intestinal valve joins the pharynx to the intestine. Knowledge of the ultrastructure of these complex characters may be useful in understanding of functional features, and for comparative morphology as well as evolutionary considerations within the Chromadorea.  相似文献

The unusual idiosomal glands of a water mite Teutonia cometes (Koch 1837) were examined by means of transmission and scanning electron microscopy as well as on semi-thin sections. One pair of these glands is situated ventrally in the body cavity of the idiosoma. They run posteriorly from the terminal opening (distal end) on epimeres IV and gradually dilate to their proximal blind end. The terminal opening of each gland is armed with the two fine hair-like mechanoreceptive sensilla (‘pre-anal external’ setae). The proximal part of the glands is formed of columnar secretory epithelium with a voluminous central lumen containing a large single ‘globule’ of electron-dense secretory material. The secretory gland cells contain large nuclei and intensively developed rough endoplasmic reticulum. Secretory granules of Golgi origin are scattered throughout the cell volume in small groups and are discharged from the cells into the lumen between the scarce apical microvilli. The distal part of the glands is formed of another cell type that is not secretory. These cells are composed of narrow strips of the cytoplasm leaving the large intracellular vacuoles. A short excretory cuticular duct formed by special excretory duct cells connects the glands with the external medium. At the base of the terminal opening a cuticular funnel strengthens the gland termination. At the apex of this funnel a valve prevents back-flow of the extruded secretion. These glands, as other dermal glands of water mites, are thought to play a protective role and react to external stimuli with the help of the hair-like sensilla.  相似文献
