Efforts were made to eliminate the influence of other factors as far as possible in order to obtain reliable results on the effects of oxygen on the growth of baker's yeast. A cultivation method is presented which permits the study of the effects of aeration intensity under conditions where the influence of catabolite repression is eliminated. A completely synthetic medium with glucose as the only carbon and energy source is also described. The capacity of yeast to perform aerobic metabolism varies when cultivated under different intensities of aeration. A clear maximum is observed for growth with 10% oxygen in the aerating gas mixture. Under conditions where catabolite repression does not function yeast has the potential for oxidative metabolism even under oxygen-limited growth. The main agent controlling the ability of yeast to support growth using only the oxidative metabolism is the available oxygen. At high oxygen tensions the metabolism is disturbed.  相似文献

A series of baker's yeast continuous cultivations were made using different intensities of aeration. The experimental conditions were such as to eliminate the effects caused by high glucose concentrations in the medium on the formation of enzymes. The variation in activity of several enzymes was investigated and distinct changes were noted. The activities of hexokinase and alcohol dehydrogenase characterize the actual rate of glycolysis in yeast, the same being true, in part, for pyruvate decarboxylase. The activity of phosphofructokinase is nearly insensitive to the oxygen level at normal tensions. The activity of the cell to the phosphofructokinase can be limited in anaerobic conditions by its scarcity. The insensitivity of glucose-6-phosphate dehydrogenase to the oxygen tension together with its low activity suggests that this enzyme plays primarily a biosynthetic role and that the function of the pentose phosphate pathway as an energy-producing route is negligible.  相似文献

The activities of several oxidoreductases were measured in three fermentative and two nonfermentative Mycoplasma species that were grown under aerobic or anaerobic conditions. Acholeplasma laidlawii MG, Mycoplasma hyorhinis GDL, and Mycoplasma pneumoniae FH had very high apparent activities of pyruvate dehydrogenase and pyruvate dehydrogenase complex compared with the activities of mammalian fibroblasts or human platelet-enriched preparations, while Mycoplasma salivarium VV and Mycoplasma arthritidis 07 had very low apparent activities of these two enzymes. Strictly anaerobic growth diminished both enzymatic activities. The activity of alpha-ketoglutarate dehydrogenase complex was minimal in all five mycoplasmas that were grown under aerobic conditions, anaerobic conditions, or both. All the mycoplasmas that were examined exhibited lactate dehydrogenase and NADH-dichlorophenol indophenol oxidoreductase activities. The properties of mycoplasmal pyruvate dehydrogenase complex suggest that it differs from the mammalian enzyme.  相似文献

The subcellular location of hexose diphosphatase, phosphoenolpyruvate carboxykinase and pyruvate carboxylase in baker's yeast (Saccharomyces cerevisiae) was investigated by density gradient centrifugation of spheroplast lysates obtained by osmotic shock treatment of spheroplasts and centrifugation for 10000 g x min. In order to examine acid phosphatase (EC 3.1.3.2) and ATPase (EC 3.6.1.3) activities of baker's yeast (pH optimum 3.5) a protoplast-secreted enzyme preparation was purified and some physical and chemical properties were studied. Three protein fractions containing ATPase and acid phosphatase activities, in the same ratio as the initial preparation, were separated by ion-exchange chromatography. The first fraction which had the highest protein content yielded a homogeneous preparation after Sepharose 4B chromatography and was used in further studies. An attempt to estimate molecular weight of this protein was made. Attempts to separate acid phosphatase and ATPase activities by ion-exchange chromatography, gel filtration, isoelectric focusing and sucrose density gradient centrifugation have been unsuccessful. Both activities behaved the same way to heat and urea denaturation. These results suggest that the two activities are associated with the same protein molecule.  相似文献

The activity of some intracellular oxidative enzymes was studied histochemically in the cells of the thyroid follicles of teleost fishes of the genus Xiphophorus. The experimental material consisted of animals of the red swordtail and Mexican swordtail breeds of Xiphophorus helleri and of melanotic Xiphophorus maculatus fishes. Observations were carried out on adult specimens of both sexes, including pregnant femals of Mexican swordtail. Moreover, immature Mexican swordtails of both sexes were examined. Thyroid follicles were found to be present in the subpharyngeal region of all fishes studied. The distribution of these follicles as well as their number and form depended on sex, age and on the analysed stage of prenancy. A smaller number and size of thyroid follicles were characteristic of immature specimens, whereas they were most numerous in the thyroids of pregnant fishes. The follicles were arranged in characteristic dense aggregations, especially in the melanotic platyfish. The follicular eipthelium in the fishes under study was usually cubical, but pregnant and non-pregnant adult females also contained a considerable number of larger follicles with flattened epithelium. Besides, thyroid follicles of multilayer epithelium were rather frequently encountered, especially in male fishes, irrespective of their age. The thyroid follicle cells of these fishes demonstrated invariably high activities of reduced NAD and NADP dehydrogenases and of beta-hydroxybutyrate dehydrogenase, and a low activity of succinat dehydrogenase. The intensities of alpha-glycerophosphate and lactate dehydrogenases and of cytochrome oxidase varied with sex, age and breed of the studied fishes. The immature and pregnant fishes showed the most clearly pronounced differences in the intensity of enzymic activity, the thyroid follicles of immature specimens revealing a high activity of lactate dehydrogenase and low activity of cytochrome oxidase, an inverse picture being seen in pregnant fishes. The adult forms of both sexes exhibited an enhanced activity of cytochrome oxidase and a decline in that of lactate dehydrogenase. The observed differences in the intensities of enzymic acitivities in the thyroids of the studied fishes are related with functions of this gland which in the period of growth are different from those in the period of sexual maturity, and certainly also with individual metabolic characteristics of the studied fishes.  相似文献

Transketolase from baker's yeast was covalently bound to Sepharose via one subunit. Storage in glycine buffer pH 11 entailled loss of half of the protein and a 50% decrease in the activity of the immobilized enzyme. Addition to the system of free subunits of transketolase doubled the amount of the protein attached to the matrix and restored the catalytic activity to the initial level. Thermoinactivation of the initial immobilized dimer of transketolase and the renatured dimer formed on reassociation of the immobilized subunits with the free ones was the same and considerably differed from the thermoinactivation of the immobilized subunits. The conclusion is made that the individual subunits of transketolase are catalytically active.  相似文献

Accumulating evidence indicates that the enzymes involved in mitochondrial oxidative phosphorylation (OXPHOS) are co-assembled into higher-ordered supercomplexes within the mitochondrial inner membrane. This review will focus largely on the OXPHOS supercomplexes of the yeast Saccharomyces cerevisiae. The recent evidence to indicate that diversity in the populations of the cytochrome bc (1)-COX supercomplexes exist shall be outlined. In addition, the existence of dimeric/oligomeric F(1)F(o)-ATP synthase complexes and their proposed role in establishment of the cristae architecture of the inner mitochondrial membrane shall also be discussed.  相似文献

A reactor configuration consisting of two reactors with an exchange flow was used for the experimental simulation of large-scale conditions. The influence of fluctuations in oxygen concentration on the growth and metabolite production of baker's yeast was investigated by sparging one fermentor with air and one with nitrogen gas. It was found that the biomass yield decreased and the metabolite formation increased with rising circulation time (longer oxygen-unlimited and oxygen-limited periods). Not only was the performance of the oxygen-limited fermentor characterised by (partly) reductive metabolism, but that of the oxygen-unlimited fermentor as well. The results of the experiments in this reactor system were compared with those from the experiments carried out in a one-fermentor system with periodically changing oxygen concentrations. The formation of acetic acid, which is characteristic for transient states, showed a distinct difference between the two reactor systems.  相似文献

Malate synthase was investigated in solution by the small-angle X-ray scattering technique. The substrate-free enzyme was shown to have a molecular weight of 186000, a radius of gyration of 3.96 nm, a maximum particle diameter of 11.2 nm, a volume of 343 nm3, a radius of gyration of the thickness of 1.04 nm, and an axial ratio of 1:0.33. The enzyme molecule undergoes small changes in overall structure upon binding substrates. Investigation of the enzyme under prolonged exposure to X-rays led to an aggregation of the enzyme and allowed statements concerning the way of aggregation and factors influencing aggregation.  相似文献

Effects of digitonin, dimethylsulfoxide and protamine sulfate on yeast Pichia guilliermondii were studied in order to produce cells with increased permeability and possessing the GTP-cyclohydrolase, riboflavinsynthetase and riboflavinkinase activities. The digitonin-treated cells exhibited a higher cyclohydrolase activity than the cell-free extracts; the activities of riboflavinsynthetase and riboflavinkinase in the cells and cell-free extracts were found to be similar. Treatment of cells with dimethylsulfoxide proved to be most effective to determine the activity of GTP-cyclohydrolase and also helpful to determine that of riboflavinsynthetase. Protamine sulfate had no effect on the cells of P. guilliermondii. The methods developed were used to determine the activities of GTP-cyclohydrolase, riboflavinsynthetase and riboflavinkinase in the cells of flavinogenic (P. guiller-mondii, Torulopsis candida) and non-flavinogenic (Candida utilis, Candida pulcherrima) yeasts grown in iron-rich and iron-deficient media. Derepression of riboflavinsynthetase and GTP-cyclohydrolase syntheses under conditions of Fe deficiency in the flavinogenic yeast cells confirmed previously made assumptions.  相似文献
