根据人白细胞介素-2(IL-2)a螺旋B中氨基酸残基的空间分布选择性地突变了一些氨基酸残基,结果发现.57Gln→Gln,62Gln→Leu,62Gln→Arg和65Pro→Arg这些替换均使IL-2活性显著降低或丧失,而63Leu→Ser或64Lys→Ala对IL-2活性影响不大。从受体竞争抑制结合实验结果可知,上述不表现活性的突变体也同时丧失了与高亲和力受体的结合能力,这说明α螺旋B中这些位点对IL-2与受体结合有贡献,事实上,那些直接与受体β、γ亚基结合的残基所在螺旋为A、D螺旋而非α螺旋B,由此我们认为α螺旋B虽不直接参与与受体β、γ亚基结合,但它在空间结构上对IL-2与受体β、γ亚基的结合产生了有利的影响,而57Gln、62Gln、65Pro等残基则在此过程中发挥重要作用。  相似文献

猪圆环病毒2型(porcine circovirus type 2,PCV2)可读框2(open reading frame 2,ORF2)基因编码具有多个抗原表位的病毒衣壳(Cap)蛋白,能在机体内有效引发免疫反应并产生中和抗体,是预防和治疗猪圆环病毒病(porcine circovirus-associated disease,PCVAD)的主要抗原。但Cap蛋白高昂的生产成本限制了它的广泛应用。为了探索Cap蛋白新的生产途径,本研究选择烟草叶绿体为表达平台,构建了ORF2抗原基因烟草叶绿体表达载体pNK-a03,并通过基因枪法(gene biolistics)将表达载体导入烟草叶绿体中。转化植株经PCR和RT-PCR检测,证实ORF2抗原基因已整合进烟草叶绿体基因组中且正常转录。蛋白质印迹检测和ELISA分析表明,Cap蛋白在烟草叶绿体内获得有效表达,具有免疫活性。此外,种子萌发研究结果显示,转基因烟草植株F1代具有aadA抗性,目的基因可正常进行遗传。这些研究结果为探索Cap蛋白的新型表达途径提供了理论基础。  相似文献

Abstract: Interleukin (IL)-2, initially discovered for its mitogenic activity on T cells, also acts on monocytes, resulting in the activation of cytokine production, superoxide production, and tumoricidal activity. Because severe brain damage was observed in IL-2-transgenic mice, this cytokine may have some influence(s) on the cells of the CNS. We investigated IL-2 receptor-bearing cells in the CNS and found that activated microglia expressed α-chain mRNA and immunoreactive IL-2 receptor β-chain protein in culture. Although microglia did not express IL-2 receptors under normal culture conditions, they were induced to express these receptors by lipopolysaccharide (LPS) in a time-dependent manner. The IL-2 receptors were found to be functional because the viability and growth activity of LPS-treated microglia, but not untreated controls, increased in response to recombinant mouse IL-2 as determined by a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide colorimetric assay and bromodeoxyuridine uptake experiment, respectively. These effects of recombinant IL-2 were blocked by pretreatment with anti-mouse IL-2 receptor β-chain antibody. Our findings suggest that activated microglia in the CNS can respond to this T cell-derived factor regulating their growth, which may be an important mechanism of communication between nervous and immune systems in physiological and pathological conditions.  相似文献

通过定点诱变技术得到6个生物活性剧烈下降的人白细胞介素-2(IL-2)突变体,其中两个突变体即15Val-IL-2和126Asp-IL-2可以在一定浓度范围内使IL-2的生物效应降低.在对高亲和力IL-2受体(IL-2R)的竞争抑制实验中,15Val-IL-2和126Asp-IL-2又表现了一定的竞争能力.这些结果表明15Val-IL-2和126Asp-IL-2的部分拮抗天然IL-2的作用.结合IL-2二级结构分析及对IL-2与IL-2R相互作用的已有认识,可认为15Val-IL-2和126Asp-IL-2的部分拮抗作用产生的原因在于替换残基在空间上对IL-2与IL-2R βγ亚基结合微环境的轻微扰动,干扰了IL-2有关残基与IL-2R βγ亚基的结合,但尚不能完全阻止其与IL-2R βγ亚基的结合.  相似文献

Adenosine triphosphatase activity of tobacco leaf chloroplasts in the dark was measured, using leaves of different age as determined by the position of the leaves along the stem. The activity of the chloroplast preparations strongly decreased with age, regardless of the addition of Mg2+ or Ca2+. Opposite effects of Mg2+ and Ca2+ on the activity of the chloroplasts were noted in experiments where different ratios of Mg2+/Ca2+ were applied. They were related to the age of the leaves, Ca2+ strongly stimulated the activity of the preparations from old leaves but was practically without effect in young, just expanded leaves. Mg2+ slightly stimulated the activity from old leaves while it invariably inhibited the hydrolytic activity of preparations from young leaves.  相似文献

Human papillomavirus (HPV) is the causal agent of cervical cancer, one of the most common causes of death in women worldwide, and its E7 antigen is the major candidate for a therapeutic vaccine. The large scale production of E7 by molecular farming that would lead to the development of a safe and inexpensive vaccine is impaired by its low accumulation level in the plant cell. To enhance antigen production in the plastids, two alternative strategies were carried out: the expression of E7 as a translational fusion to β-glucuronidase enzyme and redirection of E7 into the thylakoid lumen. The use of the β-glucuronidase as a partner protein turned out to be a successful strategy, antigen expression levels were enhanced between 30 and 40 times relative to unfused E7. Moreover, best accumulation, albeit at a high metabolic cost that compromised biomass production, was obtained redirecting E7 into the thylakoid lumen by the incorporation of the N-terminal transit peptide, Str. Following this approach lumenal E7 production exceeded the stromal by two orders of magnitude. Our results highlight the relevance of exploring different strategies to improve recombinant protein stability for certain transgenes in order to exploit potential advantages of recombinant protein accumulation in chloroplasts.  相似文献

The expression of the green fluorescent protein reporter gene (gfp) from the bacterial trc and plastid rrn and psbA promoters has been compared in transplastomic tobacco plants produced by microprojectile bombardment. The homoplasmic nature of the regenerated plants was confirmed by Southern blot analysis. Northern blot analysis indicated that plants expressing gfp from the rrn promoter contained 3-fold more gfp RNA than plants containing the psbA promoter and 12-fold more than plants with the trc promoter. Immunoblot analysis and fluorescence spectroscopy indicated that plants expressing gfp from the rrn promoter contained approximately 90-fold more green fluorescent protein (GFP) than plants containing the psbA or trc promoters. This study demonstrates that the bacterial trc promoter is significantly weaker than the plastid rrn promoter for expression of gfp in tobacco chloroplasts.  相似文献

Abstract: Previous studies have shown that PC12 cells depend on growth factors for their survival. When deprived of growth factors, the cells undergo a dying process termed "apoptosis" (programed cell death). We show here that muscarinic agonists inhibited the apoptotic death of growth factor-deprived PC12M1 cells (PC12 cells stably expressing cloned m1 muscarinic acetylcholine receptors). This protective effect of the muscarinic agonists was observed in both proliferating and neuronal PC12M1 cells, was blocked by the muscarinic antagonist atropine, and was not observed in PC12 cells lacking m1 receptors. Muscarinic receptors therefore mediate inhibition of apoptosis in these cells. In addition to its effect on survival, the muscarinic agonist oxotremorine induced inhibition of DNA synthesis as well as growth arrest of exponentially growing PC12M1 cells at the S and G2/M phases of the cell cycle. Muscarinic receptors in these cells may therefore mediate inhibition of cell cycle progression.  相似文献

Background

A natural bispecific antibody, which can be produced by exchanging Fab arms of two IgG4 molecules, was first described in allergic patients receiving therapeutic injections with two distinct allergens. However, no information has been published on the production of natural bispecific antibody in animals. Even more important, establishment of an animal model is a useful approach to investigate and characterize the naturally occurring antibody.

Methodology/Principal Findings

We demonstrated that a natural bispecific antibody can also be generated in New Zealand white rabbits by immunization with synthesized conjugates. These antibodies showed bispecificity to the components that were simultaneously used to immunize the animals. We observed a trend in our test animals that female rabbits exhibited stronger bispecific antibody responses than males. The bispecific antibody was monomeric and primarily belonged to immunoglobulin (Ig) G. Moreover, bispecific antibodies were demonstrated by mixing 2 purified monospecific antibodies in vivo and in vitro.

Conclusions/Significance

Our results extend the context of natural bispecific antibodies on the basis of bispecific IgG4, and may provide insights into the exploration of native bispecific antibodies in immunological diseases.  相似文献

为了提高人重组白细胞介素 2的稳定性和活性以及减少毒副作用 ,有必要定向改造rhIL 2的分子结构 .用PCR法从白细胞介素 2 (IL 2 )cDNA全序列中扩增成熟的肽基因片段 ,并利用定点突变技术将人重组白细胞介素 2第 12 5位游离的半胱氨酸编码序列突变为丙氨酸序列 .编码 18位亮氨酸的序列突变为蛋氨酸序列 ;编码 19位亮氨酸的序列突变为丝氨酸序列 .突变型人白细胞介素 2 (MvIL 2 )基因与表达载体pPIC9K重组 ,酶切线性化后用Invitrogen转化毕赤酵母试剂盒导入酵母细胞进行整合 ,经筛选得到一高表达白介素 2的克隆 .SDS PAGE显示 ,表达量约占总量的4 5 7% .经Western印迹验证 ,重组人白介素 2有免疫活性 ;与野生型IL 2相比 ,所获得的突变型IL 2纯品的比活性为 4 0× 10 7IU mg蛋白 ,比天然型IL 2高 4~ 5倍  相似文献

质体作为植物细胞中一类重要的细胞器,控制其分裂的分子机制一直都不清楚。最近的研究表明,植物细胞中与原核细胞分裂基因fisZ类似的同源基因控制着质体的分裂过程。通过正反义转化分析了两个烟草的ftsZ基因(NtFtsZ1和NtFtsZ2)在转基因烟草中的功能。二的反义表达并未对转化烟草细胞中叶绿体的分裂和形态产生明显影响,但二过表达转化植株中叶绿体的数目和形态都发生了明显的变化,在某些转化植株的叶肉细胞中甚至只有1-2个巨大的叶绿体存在。对不同转化植株的电镜观察和叶绿素含量分析认为,NtFtsZs基因可能对叶绿体的正常发育和功能没有影响,叶绿体形态的变化是对其数目减少的一种补偿。正反义转化植株中叶绿体的不同表型暗示高等植物中同一家族的ftsZ基因可能在控制质体分裂方面具有相同的功能。同时,过表达植株中叶绿体形态的变化被认为是高等植物的FtsZ质体骨架功能的体现。  相似文献

按DNA-磷酸钙共沉淀法将人白细胞介素2受体(IL-2R)cDNA转染小鼠成纤维细胞L929,经RNA点渍杂交分析、荧光标记IL-2染色和抗Tac(人IL-2受体α链)特异性玫瑰花环试验,均证明转入的IL-2R cDNA在L929细胞中表达,其产物具有结合IL-2和抗Tac抗体的能力。本文还报道了T细胞白血病Jukat细胞和Molt-4等细胞系异常表达IL-2R的结果,并对此作了分析和讨论。  相似文献
