Thionins are antimicrobial peptides that are involved in plant defence. Here, we present an in‐depth analysis of the role of rice thionin genes in defence responses against two root pathogens: the root‐knot nematode Meloidogyne graminicola and the oomycete Pythium graminicola. The expression of rice thionin genes was observed to be differentially regulated by defence‐related hormones, whereas all analysed genes were consistently down‐regulated in M. graminicola‐induced galls, at least until 7 days post‐inoculation (dpi). Transgenic lines of Oryza sativa cv. Nipponbare overproducing OsTHI7 revealed decreased susceptibility to M. graminicola infection and P. graminicola colonization. Taken together, these results demonstrate the role of rice thionin genes in defence against two of the most damaging root pathogens attacking rice.  相似文献

The host response of 25 rice genotypes belonging to Oryza glaberrima and Oryza sativa to Meloidogyne graminicola infection was examined in a hydroponic system. The M. graminicola can build up high population densities in a hydroponic system. Resistance to this nematode species was found in O. glaberrima genotypes which supported significantly lower nematode numbers per plant and per unit root than O. sativa genotypes. The M. graminicola-infected O. sativa genotypes showed a higher root galling index than the O. glaberrima genotypes. The hydroponic system is efficient and reliable method to examine the host response of rice genotypes to M. graminicola infection, and can be useful for the fast screening of high numbers of rice genotypes for the selection of M. graminicola-resistant rice germplasm for breeding purposes.  相似文献

Salicylic acid (SA)‐mediated induction of systemic resistance by Pseudomonas aeruginosa strain 7NSK2 and P. fluorescens strain CHA0 against soil‐borne fungi and viruses have been reported. The role of SA biosynthesis in the enhancement of defence mechanism against plant‐parasitic nematodes by these bacterial strains in tomato is not known. To better understand the importance of SA in rhizobacteria‐mediated suppression of root‐knot nematodes, biocontrol potential of SA‐negative or SA‐overproducing mutants against Meloidogyne javanica was evaluated with their respective wild type counter parts. Culture supernatant of 7NSK2, CHA0 and their respective mutants caused significant mortality of M. javanica juveniles in vitro. SA deletion in 7NSK2 and SA overproduction in CHA0 did not influence bacterial efficacy to cause nematode deaths. Similarly, culture supernatants resulting from King's B liquid medium amended with FeCl3 did not influence nematicidal activity of the bacterial strains. Strain CHA0 induced juvenile deaths more than 7NSK2 did. In pot experiments, the bacterial strains applied in unsterilized sandy loam soil markedly reduced final nematode population densities in roots and subsequent root‐knot infection in tomato seedlings. SA‐negative or overproducing derivatives prevented tomato roots in kinetics similar to those with their respective wild types. When soil iron concentration was lowered by the addition of ethylenediamine di(o‐hydroxyphenylacetic acid), nematode biocontrol by the bacterial strains (both wild type and mutants) remained unaltered. To understand the mechanism involved in rhizobacteria‐mediated suppression of root‐knot nematode in tomato, bacterial performance was assessed in a split root trial in which one‐half of the root system was treated with bacterium while the other inoculated with nematode. Compared with the controls, application of the bacterial cell suspension to one‐half of the root system lowered the populations of root‐knot nematode in non‐bacterized nematode‐treated sections indicating enhanced defence in the non‐bacterized half. With respect to nematode infection, mutants induced systemic resistance to a similar extent as that caused by the wild types in both wild type tomato and NahG tomato plants. It is concluded that fluorescent pseudomonads induce systemic resistance against root‐knot nematode via a signal transduction pathway, which is independent of SA accumulation in roots.  相似文献

Rice (Oryza sativa) is an important staple food crop for majority of human population in the world in general and in Asia in particular. However, among various pests and diseases which constitute important constraints in the successful crop production, plant parasitic nematodes play an important role and account for yield losses to the extent of 90%. The major nematode pests associated with rice are Ditylenchus angustus, Aphelenchoides besseyi, Hirschmanniella spp., Heterodera oryzicola and Meloidogyne graminicola. However, rice root-knot nematode (M. graminicola) happens to be the most important pest and is prevalent in major rice producing countries of the world. In India, the distribution of M. graminicola in rice growing areas of different states has been documented in nematode distribution atlas prepared by All India Coordinated Research Project (Nematodes) and published by Directorate of Information and Publications of Agriculture, Indian Council of Agricultural Research, New Delhi, India during 2010. M. graminicola affected rice plants show stunting and chlorosis due to the characteristic terminal swellings/galls on the roots which ultimately result in severe reduction in growth and yield. Number of eco-friendly management technologies against M. graminicola have been developed and demonstrated, including the use of bioagents for minimising the losses due to rice root-knot nematode. This review is focused on collating information to understand the current scenario of rice root-knot nematodes with greater emphasis on its ecological requirements, damage symptoms, biology, morphology, host range and management strategies.  相似文献

[目的] 福建省福清市江镜镇与福安市潭溪镇水稻区直播稻苗期分别发生严重的根结线虫病,本研究对其病原物进行形态和分子鉴定,明确病原物种类,以期为福建省直播稻根结线虫病害防控提供理论依据。[方法] 通过根结线虫各虫态形态学特征进行观测;同时对其rDNA-ITS区进行测序,通过贝叶斯法与最大似然法构建了系统发育树来确定种类;利用拟禾本科根结线虫特异性引物Mg-F/Mg-R检测种群。[结果] 根结线虫的雌虫、雄虫和2龄幼虫的形态学特征与拟禾本科根结线虫原始描述种一致;rDNA-ITS区序列长度为576 bp,与GenBank拟禾本科根结线虫种群相似度均达99%以上;系统发育树明确了该根结线虫与拟禾本科根结线虫处于同一分支;特异性引物Mg-F/Mg-R检测进一步明确病原为拟禾本科根结线虫单一种群。[结论] 本研究通过形态与分子特征,明确了在福建直播稻上发现的根结线虫为拟禾本科根结线虫。拟禾本科根结线虫在福建省最早于2011年在政和县小范围水稻田发现,此后未在其他水稻种植区发现。本次在福建直播稻上首次发现大面积的根结线虫为害。随着直播稻的种植面积扩大,拟禾本根结线虫引起的水稻病害可能会成为制约其发展的新问题,应引起足够重视。  相似文献

Bacterial wilt, caused by Ralstonia solanacearum, is one of the most serious diseases of tomato (Solanum lycopersicum). Concomitant infection of R. solanacearum and root‐knot nematode Meloidogyne incognita increases the severity of bacterial wilt in tomato, but the role of this nematode in disease complexes involving bacterial pathogens is not completely elucidated. Although root wounding by root‐knot nematode infection seems to play an important role, it might not entirely explain the increased susceptibility of plants to R. solanacearum. In the present study, green fluorescent protein (GFP)‐labelled R. solanacearum distribution was observed in the root systems of the tomato cultivar Momotaro preinoculated with root‐knot nematode or mock‐inoculated with tap water. Fluorescence microscopy revealed that GFP‐labelled R. solanacearum mainly colonized root‐knot nematode galls, and little or no green fluorescence was observed in nematode‐uninfected roots. These results suggest that the gall induced by the nematode is a suitable location for the growth of R. solanacearum. Thus, it is crucial to control both R. solanacearum and root‐knot nematode in tomato production fields to reduce bacterial wilt disease incidence and effects.  相似文献

以根结线虫侵染的文冠果一年生苗木为材料,观察分析苗木根系的形态和显微结构,植株生长发育以及主要矿质养分在苗木中的分布特性,并分析不同矿质元素与线虫侵染的关联关系,以揭示根结线虫对文冠果苗木生长发育的影响机制。结果表明:(1)受根结线虫侵染的文冠果苗木根系形成根结,依据根结发生程度分为0(对照,正常植株)、2、3、4级;与对照相比,具有根结的文冠果苗木根系解剖结构特征主要表现为皮层相对较厚,木质部排列扭曲,导管较少,射线数目较少且分布不均匀;韧皮细胞内含物明显较多;多个巨细胞及细胞空腔主要存在于韧皮部。(2)具根结的苗木株高和地径较对照均增加,且株高增幅达显著水平(P<0.05)。(3)具根结苗木根、茎、叶部位的N、P含量均较对照下降,根部降幅达显著水平(P<0.05),其中具2~4级根结的根部N含量分别较对照显著下降7.8%、16.0%和29.5%,P含量分别显著下降15.6%、7.1%和43.3%;根部Fe含量、Zn含量显著上升,2~4级根结根系中Fe含量较对照分别增加1.56倍、0.81倍和3倍,Zn含量分别增加1.11倍、1.56倍和1.78倍;具3和4级根结的苗木根系中K含量较对照显著增加(P<0.05),具2和4级根结的苗木叶片中K含量较对照分别显著下降61.5%和47.0%。(4)苗木根部、茎部N元素含量对于根结线虫侵染的响应最明显,且其含量随侵染程度的增大而降低,叶部Mg元素含量与根结线虫侵染率、K元素含量与整株苗木生物量均呈显著负相关关系(P<0.05)。该研究为根结线虫对文冠果苗木养分吸收利用及运转的影响提供了一定的理论依据。  相似文献

The burrowing nematode (Radopholus similis (Cobb) Thorne) and the banana weevil (Cosmopolites sordidus Germar, Coleoptera: Curculionidae) are major pests of banana (Musa spp.) in the Lake Victoria basin region of Uganda. Among biological options to control the two pests is the use of non-pathogenic Fusarium oxysporum Schltdl.: Fries endophytes of banana. We investigated the ability of endophytic F. oxysporum isolates Emb2.4o and V5w2 to control the banana weevil and the burrowing nematode, alone and in combination. Plant colonization by the endophytes was determined by inoculating their chemical-resistant mutants separately and in combination, onto banana roots. Plant growth promotion was determined by measuring plant height, girth, number of live roots and fresh root weight at harvest, and control of the nematode and weevil was determined by challenging endophyte-inoculated plants with the pests 8 weeks after endophyte inoculation. Endophytic root colonization was highest in plants inoculated with both endophytes, compared with those inoculated with only one of the endophytes. Root colonization was better for isolate V5w2 than Emb2.4o. Dually inoculated plants showed a significant increase in height, girth, fresh root weight and number of functional roots following nematode challenge. Nematode numbers in roots were reduced 12 weeks after challenge of 8-week-old endophyte-inoculated plants. Significant reductions in weevil damage were observed in the rhizome periphery, inner and outer rhizomes, compared with endophyte non-inoculated controls. We conclude that dual inoculation of bananas with endophytic isolates Emb2.4o and V5w2 increases root colonization by the endophytes, reduces nematode numbers and weevil damage, and enhances plant growth in the presence of nematode infestation.  相似文献

The nematode trapping and mycoparasitic potential of Arthrobotrys oligospora was tested in vitro against Meloidogyne graminicola and Rhizoctonia solani, respectively. Five isolates of A. oligospora were isolated from different locations of India. Diversity of the trapping structures is large and highly dependent on the environmental condition and nature of the fungus. In A. oligospora, a three-dimensional adhesive net (in response to nematode) and hyphal coils developed around the hyphae of R. solani. In vitro trap formation and predacity were tested against second-stage juveniles of M. graminicola (J2) and the interactions between A. oligospora and R. solani were recorded. Under field conditions, we demonstrated the biocontrol potential of A. oligospora against R. solani causing sheath blight of rice (Oryza sativa) for the first time. All the isolates of A. oligospora parasitized and killed M. graminicola and R. solani. Application of A. oligospora, isolate VNS-1, in soil infested with M. graminicola and R. solani reduced the number of root knot by 57.58–62.02%, sheath blight incidence by 55.68–59.32% and lesion length by 54.91–66.66% under green house and miniplot (field) conditions. Applications of A. oligospora to the soil increased plant growth: shoot length by 56.4–68.8%, root length by 44.0–54.55%, fresh weight of shoot and root by 62.91–65.4% and 38.9–44.19%, respectively, as compared to the plants grown in nematode infested soil.  相似文献

Root galls of rice caused by Meloidogyne graminicola were examined for natural colonization by nematophagous fungi from four fields with different nematode infestations. Old galls from severely infested fields had a higher frequency of Monacrosporium eudermatum and Stylopaga hadra than young galls. The frequency of Arthrobotrys oligospora, Arthrobotrys dactyloides, Dactylaria brochopaga and Monacrosporium gephyropagum was lower. A greater proportion (%) of root galls were colonized by nematophagous fungi in those fields in which rice roots had a greater root gall index. This indicated that disease severity supported the colonization of galls by nematophagous fungi. In vitro predacity tests of four fungi showed that A. dactyloides was most effective in capturing and killing J2 of Mel. graminicola followed by D. brochopaga and Mon. eudermatum. Application of inocula of A. dactyloides and D. brochopaga in soil infested with Mel. graminicola, respectively, reduced the number of root galls by 86% and of females by 94%, and eggs and juveniles by 94%. The application of these fungi to soil increased plant growth: shoot length by 42.7% and 39.8%, root length by 45.5% and 48.9%, fresh weight of shoot by 59.9% and 56.7% and fresh weight of root by 20.3% and 25.1%, respectively, compared to these parameters for plants grown in nematode‐infested soil.  相似文献

In order to replace particularly biohazardous nematocides, there is a strong drive to finding natural product‐based alternatives with the aim of containing nematode pests in agriculture. The metabolites produced by the fungal endophyte Fusarium oxysporum 162 when cultivated on rice media were isolated and their structures elucidated. Eleven compounds were obtained, of which six were isolated from a Fusarium spp. for the first time. The three most potent nematode‐antagonistic compounds, 4‐hydroxybenzoic acid, indole‐3‐acetic acid (IAA) and gibepyrone D had LC50 values of 104, 117 and 134 μg ml?1, respectively, after 72 h. IAA is a well‐known phytohormone that plays a role in triggering plant resistance, thus suggesting a dual activity, either directly, by killing or compromising nematodes, or indirectly, by inducing defence mechanisms against pathogens (nematodes) in plants. Such compounds may serve as important leads in the development of novel, environmental friendly, nematocides.  相似文献

This study explores the genetic diversity and structure of Meloidogyne enterolobii in mulberry in China. The COI mitochondrial gene (mtCOI) in M.enterolobii populations in Guangdong, Guangxi, and Hunan Provinces was PCR‐amplified, sequenced, and analyzed for genetic diversity. The total number of variations, haplotypes (Hap), the average number of nucleotide differences (k), haplotype diversity (H), and nucleotide diversity (π) of mtCOI were 25, 11, 4.248, 0.900, and 0.00596, respectively. Insignificant differences in Fst value (0.0169) and a high level of gene flow (7.02) were detected among the 19‐mulberry root‐knot nematode populations, and high genetic variation within each population and a small genetic distance among populations were observed. Both phylogenetic analyses and network mapping of the 11 haplotypes revealed a dispersed distribution pattern of 19 mulberry root‐knot nematode populations and an absence of branches strictly corresponding to the 19 range sampling sites. The neutrality test and mismatch analysis indicated that mulberry root‐knot nematode populations experienced a population expansion in the past. The analysis of molecular variance (AMOVA) revealed that the genetic differentiation of M. enterolobii was mainly contributed by the variation within each group. No significant correlation was found between the genetic distance and geographical distance of M. enterolobii populations. The findings of this study provide a profound understanding of the M. enterolobii population and will inform the development of strategies to combat and manage root‐knot nematodes in mulberry.  相似文献

The effects of different water regimes on the pathogenicity of Meloidogyne graminicola on six rice cultivars were determined in two soil types in three greenhouse experiments. Two water regimes, simulating continuous flooding and intermittent flooding, were used with five of the cultivars. All cultivars were susceptible to the nematode, but IR72 and IR74 were more tolerant than IR20 and IR29 under intermittent flooding. All were tolerant under continuous flooding. UPLRi-5 was grown under multiple water regimes: no flooding; continuous flooding; flooding starting at maximum tillering, panicle initiation, or booting stage; and flooding from sowing until maximum tillering or booting. In sandy loam soil, M. graminicola reduced stem and leaf dry weight, root dry weight, and grain weight under all water regimes. In clay loam soil, the nematode reduced root weight when the soil was not flooded or flooded only for a short time, from panicle initiation, or booting to maturity, and from sowing to maximum tillering. In clay loam soil, stem and leaf dry weight, as well as grain weight, were reduced by the nematode under all water regimes except continuous flooding or when the soil was flooded from sowing to booting stage. These results indicate that rice cultivar tolerance of M. graminicola varies with water regime and that yield losses due to M. graminicola may be prevented or minimized when the rice crop is flooded early and kept flooded until a late stage of development.  相似文献

Aerobic rice is a production system where adapted rice varieties are established via direct seeding in non-puddled, non-flooded, non-saturated fields and grown under conditions similar to upland conditions. On land cultivated continuously with aerobic rice, a yield reduction has been observed. The rice root-knot nematode Meloidogyne graminicola is considered one of the possible causes of these yield reductions. Resistance to and tolerance for M. graminicola are essential traits for aerobic rice cultivars in alleviating this problem. In our study, the host response of nineteen aerobic, seven upland and four lowland rice genotypes which are either being used in the International Rice Research Institute’s aerobic rice breeding programme or already cultivated by farmers in Asia was evaluated under aerobic soil conditions in an outdoor raised-bed experiment. Our study showed a large variation in susceptibility and sensitivity to M. graminicola infection among the rice genotypes examined. Resistance comparable to the resistant reference genotypes included in the experiment (CG14, TOG5674, TOG7235) was not found but in terms of susceptibility, the upland genotype Morobereken may be an interesting less-susceptible genotype. Tolerance was found and in terms of sensitivity, the high yielding aerobic genotype IR78877-208-B-1-2 may be an interesting tolerant genotype. Our study also allowed the identification of rice genotypes that are either highly susceptible or sensitive to M. graminicola infection and of which the cultivation should be discouraged. On average, M. graminicola caused an almost 30% reduction in yield. Excluding the two susceptible and three resistant reference genotypes included in the experiment, most affected was dry-shoot biomass (23.6% reduction) followed by root length, which was more affected than fresh-root weight (19.8 vs. 8%) and grain filling (17.3%), while plant height and the number of spikelets/panicle were less affected (10.2 and 8.1%, respectively). Plant‐parasitic nematodes can secrete effector proteins into the host tissue to facilitate their parasitism. In this study, we report a novel effector protein, MgMO237, from Meloidogyne graminicola, which is exclusively expressed within the dorsal oesophageal gland cell and markedly up‐regulated in parasitic third‐/fourth‐stage juveniles of M. graminicola. Transient expression of MgMO237 in protoplasts from rice roots showed that MgMO237 was localized in the cytoplasm and nucleus of the host cells. Rice plants overexpressing MgMO237 showed an increased susceptibility to M. graminicola. In contrast, rice plants expressing RNA interference vectors targeting MgMO237 showed an increased resistance to M. graminicola. In addition, yeast two‐hybrid and co‐immunoprecipitation assays showed that MgMO237 interacted specifically with three rice endogenous proteins, i.e. 1,3‐β‐glucan synthase component (OsGSC), cysteine‐rich repeat secretory protein 55 (OsCRRSP55) and pathogenesis‐related BetvI family protein (OsBetvI), which are all related to host defences. Moreover, MgMO237 can suppress host defence responses, including the expression of host defence‐related genes, cell wall callose deposition and the burst of reactive oxygen species. These results demonstrate that the effector MgMO237 probably promotes the parasitism of M. graminicola by interacting with multiple host defence‐related proteins and suppressing plant basal immunity in the later parasitic stages of nematodes.  相似文献

Summary Catenaria anguillulae parasitized and killed the eggs and second stage juveniles (J2) of Meloidogyne graminicola under natural conditions. The percentage of infection in eggs was higher than J2 of M.␣graminicola, which ranged between 0–50.3% and 0–18.9% in 2004 and 0–46.6% and 0–21.7% in 2005, respectively. The higher parasitism of eggs and J2 was recorded from those fields in which plants were severely infected with M. graminicola. The degree of parasitism of eggs and J2 by C. anguillulae varied with severity of root knot disease. The fields with a higher root gall index recorded a higher percentage of infection in eggs and J2 of M. graminicola. In general, old galls when teased and incubated, recorded higher parasitism of eggs and juveniles than young galls.  相似文献
