Procedures were developed for the in vitro elimination of Sugarcane mosaic virus (SCMV), Sorghum mosaic virus (SrMV), Sugarcane streak mosaic virus (SCSMV), Sugarcane yellow leaf virus (SCYLV) and Fiji disease virus (FDV) from infected sugarcane. In vitro shoot regeneration, elongation and virus elimination through meristem tissue culture originating from both apical and axillary shoots were compared. The average rates of regeneration and elongation from apical meristem tissues were 91 and 66%, respectively, with the virus-free rate among elongated shoots ranging from 61–92%. Mature axillary buds were cultivated in vitro to produce axillary shoots, from which meristem tissues were excised and cultured. These meristem tissues regenerated (77–100%) and elongated (55–88%) in culture medium at approximately the same rate as the apical meristems. The average virus elimination rate was 90% among elongated shoots derived from mature axillary buds. All five viruses can be eliminated by meristem tissue culture from both apical and axillary shoots using a standardized procedure. The overall average efficiency of virus-free plant production was 45 and 58% from apical and axillary shoots, respectively. There were no significant differences for shoot induction or virus elimination when the meristems were harvested from either the apical or the axillary shoots. This is the first report of SrMV or SCSMV elimination from sugarcane, as well as elimination of any mixed virus infections. This new method of harvesting meristems from axillary buds greatly expands the amount of material available for therapeutic treatments and thereby increases the probability of eliminating viruses from infected sugarcane.  相似文献

Degenerate Potyviridae primers were used to amplify and sequence the 3′‐terminal regions of viruses from traditional and modern cultivars of sugarcane with mosaic disease growing in different areas of Yunnan province, China. Seven samples contained Sugarcane mosaic virus (SCMV), 11 contained Sorghum mosaic virus (SrMV) and two contained both viruses. SCMV was only isolated from traditional cultivars. In a phylogenetic analysis of the partial NIb and complete coat protein coding regions, most SCMV isolates formed a distinctive phylogenetic cluster (named SO) that otherwise contained only three Vietnamese isolates. SCMV variation seems mostly related to host genotype. In the same analysis, the SrMV isolates formed three major groups, one of which is reported for the first time, but the significance of the grouping is unclear.  相似文献

Maize dwarf mosaic disease is one of the most important viral diseases of maize (Zea mays L.) throughout the world. It is caused by several virus species in the family Potyviridae, genus Potyvirus, including Maize dwarf mosaic virus (MDMV), Sugarcane mosaic virus (SCMV), Johnsongrass mosaic virus (JGMV) and Sorghum mosaic virus (SrMV). Resistance to another member of the family Potyviridae, Wheat streak mosaic virus (WSMV), is conferred by three alleles (Wsm1, Wsm2, Wsm3) in the maize inbred line Pa405, and these or closely linked genes were previously shown to confer resistance to the potyviruses MDMV and SCMV. In this study, we assessed whether Wsm alleles are linked to resistance to JGMV and SrMV. Near isogenic lines (NILs) carrying one or two of the Wsm alleles introgressed into the susceptible line Oh28 and F1 progeny from NIL × Oh28 were tested for their response to JGMV and SrMV. Our results indicate that Wsm1 provides resistance to both JGMV and SrMV in a dose-dependent manner. Wsm2 and Wsm3 each provide limited resistance, and combining Wsm alleles enhances that resistance.  相似文献

A sensitive duplex-immunocapture-RT-PCR (D-IC-RT-PCR) technique was developed for detection and discrimination of taxonomically distinct Sugarcane streak mosaic virus (SCSMV) and Sugarcane mosaic virus (SCMV) that naturally infect sugarcane. D-IC-RT-PCR was performed using polyclonal antisera for capture of virions. Oligo 5'-d(T)18(AGC)-3' as a common reverse primer for both viruses and virus specific forward primers, 5'-AAGTGGTTAAACGCCTGTGG-3' and 5'-ATGTC(GA)AAGAA(GA)ATGCGCTTGC-3' were used for amplifying approximately 1400 and approximately 900 bp fragments of SCSMV and SCMV genomes, respectively from their 3' termini. To assess the applicability of the developed technique, 67 mosaic affected sugarcane samples were initially screened by direct antigen coating-enzyme-linked immunosorbent assay (DAC-ELISA) followed by D-IC-RT-PCR. In DAC-ELISA, approximately 69% of tested samples were shown to be positive for presence of SCSMV, approximately 28% for SCMV and approximately 10% for both viruses. In D-IC-RT-PCR both viruses were detected up to the dilution of 10(-4). In D-IC-RT-PCR, approximately 76% of tested samples were found to be positive for SCSMV, approximately 37% for SCMV and approximately 16% for both viruses. The sequence analyses of D-IC-RT-PCR amplicons of 3 isolates of each virus revealed that the designed primers were virus-specific. The developed technique had potential application for sensitive parallel detection of two viruses in sugarcane.  相似文献

【目的】利用NIa基因,阐明甘蔗线条花叶病毒(Sugarcane streak mosaic virus,SCSMV)的种系发生关系,为预测SCSMV流行变异趋势及科学防控提供理论依据。【方法】从云南蔗区和国家甘蔗种质资源圃采集感病样品,RT-PCR扩增获得SCSMV NIa基因序列后,使用Splits Tree、RDP、Phy ML、Dna SP等软件分析SCSMV中国分离物的系统发生、选择压力及基因流动等特征。【结果】共获得23条SCSMV NIa基因序列。这些序列间未发生重组,云南蔗区的部分序列形成1个新簇,且云南蔗区与国家甘蔗种质资源圃之间的基因交流不显著。此外,选择压力分析表明,NIa基因受很强的负选择压力作用。【结论】与P1、HC-Pro和CP等基因类似,SCSMV在NIa基因上也包含5个簇;SCSMV云南分离物具有较高的遗传多样性和清晰的地理相关性。  相似文献

该研究以甘蔗条纹花叶病毒(Sugarcane streak mosaic virus,SCSMV)的P3蛋白(SCSMV-P3)为诱饵,采用酵母双杂交技术(yeast two-hybrid system,Y2HS)从甘蔗酵母文库中筛选到1个编码核酮糖-1,5-二磷酸羧化酶/加氧酶(ribulose-1,5-bisphosphate carboxylase/oxygenase,Rubisco)大亚基的基因,命名为ScRbcL。序列分析表明,该基因开放读码框(open reading frame,ORF)长度为1 337bp,编码478个氨基酸残基。Y2HS和双分子荧光互补(bimolecular flourescence complementation,BiFC)实验表明,ScRbcL与SCSMV-P3存在互作关系。研究认为SCSMV-P3与ScRbcL的互作可能是SCSMV侵染甘蔗导致花叶症状发生的分子基础。  相似文献

Field surveys were conducted during the 2012 wet season in five Local Government Areas (LGAs) each of Kaduna and Kano States, Nigeria, to determine the incidence of potyviruses infecting sorghum. Sorghum leaves (n = 450) showing mosaic, streak, dwarfism and asymptomatic ones were collected from 30 fields. Based on the fields, the virus disease incidence in Kaduna State was 19.3, 17.1, 53.7, 35.4 and 32.7% for Sabon Gari, Giwa, Lere, Kubau and Makarfi LGAs, respectively. In Kano State, the incidence was 13.3, 32.9, 28.9, 34.3 and 50.9% for Tsanyawa, Minjibir, Wudil, Sumaila and Garun Malam LGAs, respectively. Enzyme-linked immunosorbent assay (ELISA) was employed in the detection of the viruses. There was overall incidence of Sorghum mosaic virus 2.8%, Maize dwarf mosaic virus 2.0%, Sugarcane mosaic virus 3.3% and unidentified potyvirus 4.2% from the two states.  相似文献

Sugarcane streak mosaic virus (SCSMV), a member of the genus Poacevirus is an important viral pathogen affecting sugarcane production in India. The P1 gene of ten Indian isolates was sequenced and compared with previously reported SCSMV isolates. Comparative sequence analysis revealed a high level of diversity in the P1 gene (83–98% nucleotide sequence identity; 87–100% amino acid sequence identity), and the Indian SCSMV isolates were found to be the most variable (up to 9% diversity at the amino acid level). Phylogenetic tree analysis showed clustering of 17 SCSMV isolates into two groups: group I included isolates from India (except SCSMV-TPT) and Pakistan, and group II consisted of isolates from Japan, Indonesia, Thailand and SCSMV-TPT. The results obtained from phylogenetic study were further supported by the different in silico analysis viz. SNPs (single nucleotide polymorphism), INDELs (insertion and deletion) and evolutionary distance analysis. A significant proportion of recombination sites were observed at the N terminal region of P1 gene. Analysis of selection pressure indicated that the P1 gene of the Indian SCSMV isolates is under strong negative or purifying selection. It is likely that recombination identified in Indian SCSMV isolates, along with strong purifying selection, enhances the speed of elimination of deleterious mutations in the P1 gene. The evolutionary processes (recombination and selection pressure) together contributed to the observed genetic diversity and population structure of Indian SCSMV isolates.  相似文献

滇蔗茅杂交F_1双抗SCSMV和SrMV鉴定与评价   总被引:1,自引:0,他引:1  
以我国蔗区甘蔗花叶病的2种主要病原甘蔗条纹花叶病毒分离物(SCSMV-JP1,Gen Bank登录号JF488064)和高粱花叶病毒分离物(Sr MV-HH,Gen Bank登录号DQ530434)为接种毒源,采用人工切茎接种和RT-PCR检测相结合的方法,于2015-2016年2次对由热带种路打士与滇蔗茅云滇95-19杂交获得的41份滇蔗茅杂交F_1及亲本进行了双抗SCSMV和Sr MV鉴定与评价。结果表明,41份滇蔗茅杂交F_1及亲本中,对SCSMV表现1级高抗到3级中抗的有23份,占53.49%,4级感病到5级高感的有20份,占46.51%;对Sr MV表现1级高抗到3级中抗的有31份,占72.09%,4级感病到5级高感的有12份,占27.91%。综合分析结果显示,10份滇蔗茅杂交F_1对SCSMV和Sr MV均表现1~2级抗病,占23.26%,其中云09-604、云09-607、云09-619、云09-633、云09-656、云滇95-19等6份滇蔗茅杂交F_1对2种病毒均表现为1级高抗,占13.95%。研究结果明确了41份滇蔗茅杂交F_1及亲本对甘蔗花叶病2种主要致病病原的抗性,筛选出10份双抗SCSMV和Sr MV的滇蔗茅杂交F_1,为深入开展抗甘蔗花叶病育种提供了优良抗源种质和参考依据。  相似文献

Genes on chromosomes six (Wsm1), three (Wsm2) and ten (Wsm3) in the maize (Zea mays L.) inbred line Pa405 control resistance to Wheat streak mosaic virus (WSMV), and the same or closely linked genes control resistance to Maize dwarf mosaic virus (MDMV) and Sugarcane mosaic virus (SCMV). Near isogenic lines (NIL) carrying one or two of the genes were developed by introgressing regions of the respective chromosomes into the susceptible line Oh28 and tested for their responses to WSMV, MDMV, and SCMV in the field and greenhouse. F1 progeny from NIL × Oh28 were also tested. Wsm1, or closely linked genes, provided resistance to all three viruses, as determined by symptom incidence and severity. Wsm2 and Wsm3 provided resistance to WSMV. Wsm2 and/or Wsm3 provided no resistance to MDMV, but significantly increased resistance in plants with one Wsm1 allele. NIL carrying Wsm1, Wsm2, or Wsm3 had similar SCMV resistance in the field, but NIL with Wsm2 and Wsm3 were not resistant in the greenhouse. Addition of Wsm2 to Wsm1 increased SCMV resistance in the field. For all viruses, symptom incidence was higher in the greenhouse than in the field, and relative disease severity was higher in the greenhouse for WSMV and MDMV. An Italian MDMV isolate and the Ohio SCMV infected the Wsm1 NIL, while the Ohio MDMV and Seehausen SCMV isolates did not. Our results indicate that the three genes, or closely linked loci, provide virus resistance. Resistance conferred by the three genes is influenced by interactions among the genes, the virus species, the virus isolate, and the environment.  相似文献

Background

Sugarcane mosaic virus (SCMV) is responsible for large-scale economic losses in the global production of sugarcane, maize, sorghum, and some other graminaceous species. To understand the evolutionary mechanism of SCMV populations, this virus was studied in Shanxi, China. A total of 86 maize leaf samples (41 samples in 2012 and 45 samples in 2013) were collected from 4 regions of Shanxi.

Results

Double-antibody sandwich (DAS)-ELISA and RT-PCR showed 59 samples (30 samples in 2012 and 29 samples in 2013) to be positive for SCMV, from which 10 new isolates of SCMV were isolated and sequenced. The complete genomes of these isolates are 9610 nt long, including the 5′ and 3′ non-coding regions, and encode a 3063-amino acid polyprotein. Phylogenetic analyses revealed that 24 SCMV isolates could be divided on the basis of the whole genome into 2 divergent evolutionary groups, which were associated with the host species. Among the populations, 15 potential recombination events were identified. The selection pressure on the genes of these SCMV isolates was also calculated. The results confirmed that all the genes were under negative selection.

Conclusions

Negative selection and recombination appear to be important evolutionary factors shaping the genetic structure of these SCMV isolates. SCMV is distributed widely in China and exists as numerous strains with distinct genetic diversity. Our findings will provide a foundation for evaluating the epidemiological characteristics of SCMV in China and will be useful in designing long-term, sustainable management strategies for SCMV.  相似文献

Sorghum mosaic virus (SrMV), a causal agent of the destructive sugarcane mosaic disease, has a global presence. An isolate of SrMV infecting a commercially-grown sugarcane plant was recovered from the Hainan province of China. The virions were visualized by an electron microscope, and the coat proteins (CPs) were sequenced by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and tandem mass spectrometry. Discrepancies between the CP predicted and actual amino acid sequences were noted, which confounded the phylogenetic assignment of the isolate. The apparent variations may have physiological effects on the pathogenicity and virulence of SrMV.  相似文献

In line with recent reports of extended-spectrum beta-lactamases (ESBLs) in Escherichia coli isolates of highly virulent serotypes, such as O104:H4, we investigated the distribution of phylogroups (A, B1, B2, D) and virulence factor (VF)-encoding genes in 204 ESBL-producing E. coli isolates from diarrheic cattle. ESBL genes, VFs, and phylogroups were identified by PCR and a commercial DNA array (Alere, France). ESBL genes belonged mostly to the CTX-M-1 (65.7%) and CTX-M-9 (27.0%) groups, whereas those of the CTX-M-2 and TEM groups were much less represented (3.9% and 3.4%, respectively). One ESBL isolate was stx(1) and eae positive and belonged to a major enterohemorrhagic E. coli (EHEC) serotype (O111:H8). Two other isolates were eae positive but stx negative; one of these had serotype O26:H11. ESBL isolates belonged mainly to phylogroup A (55.4%) and, to lesser extents, to phylogroups D (25.5%) and B1 (15.6%), whereas B2 strains were quasi-absent (1/204). The number of VFs was significantly higher in phylogroup B1 than in phylogroups A (P = 0.04) and D (P = 0.02). Almost all of the VFs detected were found in CTX-M-1 isolates, whereas only 64.3% and 33.3% of them were found in CTX-M-9 and CTX-M-2 isolates, respectively. These results indicated that the widespread dissemination of the bla(CTX-M) genes within the E. coli population from cattle still spared the subpopulation of EHEC/Shiga-toxigenic E. coli (STEC) isolates. In contrast to other reports on non-ESBL-producing isolates from domestic animals, B1 was not the main phylogroup identified. However, B1 was found to be the most virulent phylogroup, suggesting host-specific distribution of virulence determinants among phylogenetic groups.  相似文献

Summary Soilborne wheat mosaic virus (SBWMV) is a member of the genusFurovirus of plant viruses. SBWMV is transmitted to wheat roots by the plasmodiophorid vectorPolymyxa graminis. Experiments were conducted to determine the path for SBWMV transport from roots to leaves. The results of immunogold labeling suggest that SBWMV enters and moves long distance through the xylem. SBWMV may enter primary xylem elements before cell death occurs and then move upward in the plant after the xylem has matured into hollow vessels. Electron microscopy of leaf samples displaying streak symptoms from enset (Ensete ventricosum), a banana‐like plant widely cultivated in Ethiopia, showed the presence of bacilliform shaped virions as known for badnaviruses. DNA extracts subjected to rolling circle amplification (RCA), polymerase chain reaction (PCR) and cloning and sequence analysis revealed that the virus has a circular double‐stranded DNA genome of 7,163 nucleotides encoding predicted proteins of 21.5 kDa, 14.5 kDa and 202.5 kDa, a genome organization known for badnaviruses. The virus is phylogenetically most closely related to Sugarcane bacilliform Guadeloupe D virus with a nucleotide sequence identity of 77.2% at the conserved RT/RNase‐H region and 73.6% for the whole genome. Following the current species demarcation criteria, the virus should be considered as an isolate of a new species in the genus Badnavirus for which the name Enset leaf streak virus (ELSV) is suggested. Leaf samples from enset and banana were screened using virus‐specific primers, and ELSV was detected in six of 40 enset but not found in any of 61 banana samples. On the other hand, Banana streak OL virus (BSOLV) was detected from the majority (60%) of symptomatic banana samples but not from enset samples. This paper reports the first full‐genome sequence of a putative new badnavirus species infecting plants in the genus Ensete. In addition, this is the first report of the occurrence of BSOLV in Ethiopia.  相似文献
