Methicillin-resistant Staphylococcus spp. (MRSS) are causing numerous forms of illness in humans ranging from mild to fatal infections. We need to investigate the resistant pattern for different clinical isolates to control the resistance phenomena. This study was designed to provide the resistance pattern of isolated Staphylococcus spp. from various clinical samples in Khartoum State and to elucidate the frequencies of Multidrug-resistant (MDR), Extensively drug-resistant (XDR) and pan-drug resistant (PDR). Two hundred and ten bacterial isolates were from different sources (catheter tip, sputum, vaginal swab, urine, tracheal aspirate, blood, pus, nasal swab, stool, throat swab, pleural fluid, and ear swab). Isolates were identified based on their morphological characters and biochemical reaction. Antibiotics susceptibility screening was performed using twenty-three antibiotics from eighteen classes against all isolated Staphylococcus spp. following the Clinical and Laboratory Standards Institute (CLSI) guideline. The result revealed that out of 63 Gram-positive isolated bacteria, 52 (82.5%) were Staphylococcus spp. with a high incidence of S. aureus 37(71.2%). Out of all Staphylococcus spp., 38 (73.1%) were Methicillin-resistant (MR). The prevalence of MDR was higher in S. aureus (89.2%) than in S. epidermidis (75%). All Staphylococcus spp. displayed resistance to ampicillin and penicillin, while all S. aureus were sensitive to daptomycin and fosfomycin. One isolate was XDR possible PDR, while no PDR was reported in all isolated bacteria. This study provided evidence for the antimicrobial-resistant (AMR) burden in Sudan and highlighted the need for a practical and functional stewardship program to reduce the unreasonable costs of antibiotics.  相似文献

植食性昆虫对寄主植物的选择适应性是研究昆虫和植物协同进化关系的核心内容之一。评估寄主植物对植食性昆虫种群的适合度,需要综合分析昆虫对寄主的产卵选择性和寄主对昆虫的取食适合性。以桔小实蝇和番石榴实蝇为研究对象,分别测定了这两种实蝇对6种寄主果实:番石榴、香蕉、杨桃、木瓜、甜橙、番茄的产卵选择性以及幼虫取食后对其生长发育的影响。寄主产卵选择性实验分别采用完整寄主果实直接供试产卵和块状寄主果实气味引诱产卵两种处理方式;在生长发育适应性实验中,以幼虫和蛹的存活和生长发育等相关参数作为评价指标。实验结果表明,寄主的供试方式不同,两种实蝇的选择性均有明显差异;对寄主气味选择性强的寄主更适合于两种实蝇后代的生长发育。两种实蝇对6种寄主果实的产卵选择性和后代发育适合性两者相关性不显著,与许多文献报道单一地采用发育适合性(如发育历期、存活率或蛹重等)作为评价寄主选择性的结果不一致。两种实蝇之间对6种寄主果实的产卵选择和幼虫取食适合性既具相似性也具差异性,表明这两种实蝇在寄主生态位上既有重叠性又有分化性。  相似文献

Co-60 radiation has been broadly used for pest management because it results in male sterility and a lack of emergence of unparasitized hosts due to oxidative damage. Insect life is significantly affected by abiotic stress factors, including Co-60 radiation. In the present study, we determined the influence of Co-60 radiation on an important pest in southern China, the oriental fruit fly Bactrocera dorsalis (Diptera: Tephritidae). We exposed 30-h-old B. dorsalis eggs to Co-60 radiation at doses of (0, 5, 10, 15, 20, 25, 30, 35, and 40?Gy) and determined the activity of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), and peroxidase (POX). CAT and POX activity significantly increased in B. dorsalis in the 1st larvae instar after high-intensity radiation doses (25–40?Gy) and played an important role in the antioxidant response to intensive radiation. The activity of POX in B. dorsalis at the 2nd larval instar sharply decreased after different doses of radiation, except for 10 and 40?Gy. Our results demonstrated that Co-60 radiation affected the activity of antioxidant enzymes and disturbed the physiology of B. dorsalis, especially at early stages (the 1st, 2nd larvae instar). These three antioxidant defense enzymes cooperatively play an important role in protecting B. dorsalis from oxidative damage.  相似文献

The aim of this study was to assess pit latrine samples from a peri-urban community in KwaZulu-Natal (South Africa) for the presence of multidrug-resistant (MDR) Staphylococcus spp. Standard procedures were used to isolate Staphylococcus spp. from pit latrine fecal sludge samples, with confirmation at genus level by polymerase chain reaction (PCR). Sixty-eight randomly selected pit latrine Staphylococcus spp. isolates were further characterized by using established disk diffusion procedures. An average Staphylococcus spp. count of 2.1?×?105 CFU per g fecal material was established using two randomly selected pit latrine samples. Of the 68-selected Staphylococcus spp. pit latrine isolates, 49% were identified as coagulase positive, 51% as coagulase negative and 65% (12 coagulase positive, 32 coagulase negative isolates) were categorized as MDR. The majority (66/68) of Staphylococcus spp. isolates displayed resistance to fusidic acid while only 5/68 isolates displayed resistance to chloramphenicol. The pit latrine samples analyzed in this study are a source of MDR Staphylococcus spp., highlighting the need for proper hygiene and sanitation regimes in rural communities using these facilities.  相似文献

Bactrocera dorsalis and B. correcta are serious pests of agricultural fruit crops. Both species are widely distributed in tropical and subtropical countries, especially in Asia. For integrated management of both Bactrocera species, understanding their olfactory behavior is vital for designing reliable control strategies. In this study, the response of female B. dorsalis and B. correcta adults to three main volatile components of fruits was evaluated using the Y-tube olfactometer. Our results showed that at higher concentrations all volatile components, except α-humulene and its combination with β-caryophyllene, attracted significantly more B. dorsalis than the untreated arm. For B. correcta, all the volatile components attracted significantly more females in the treated arm than the untreated arm. However, at 5% and 1% concentration, all the volatile components had similar attraction for B. dorsalis females. In the case of female B. correcta, the percentage of attraction was similar for all the individual volatile chemicals and their respective mixtures at 10% concentration. While, at 5% and 1% concentrations, the percentage of attraction was significantly higher for a mixture of β-caryophyllene and α-humulene than that of individual volatile components and all possible mixture of two and three volatile components. Based on the olfactometer results, this study concluded that 3-carene and the mixture of β-caryophyllene and α-humulene are strong attractants for female flies of B. dorsalis and B. correcta, respectively. This study might be helpful for the bait application against the female adults of B. dorsalis and B. correcta in farms and orchards.  相似文献

植物源引诱物质可显著提高橘小实蝇Bactrocera dorsalis(Hendel)蛋白饵剂的应用效果,本文测试了柑橘、番木瓜、芒果、番石榴、杨桃的叶片浸提物、果实以及已知植物次生物质对橘小实蝇的引诱效果。植物浸提物的生物测定结果表明,杨桃的二氯甲烷浸提物的引诱效果最佳,平均引诱率为20.83%,对于不同的萃取剂而言,二氯甲烷、乙醇浸提物效果明显优于石油醚,三者浸提的平均引诱率分别为15.67%、15.17%和10.50%;对于不同寄主植物而言,柑橘浸提物的效果最佳,3种有机溶剂浸提物的平均引诱率为18.33%,其中石油醚、二氯甲烷和乙醇浸提物的引诱率分别为20.00%、15.83%和19.17%,其他植物浸提物的引诱效果为杨桃>番木瓜>芒果>番石榴,3种有机溶剂浸提物的平均引诱率分别为17.22%、11.67%、11.11%和10.56%。寄主果实的生物测定结果表明,柑橘的平均引诱率为65.83%,其中雌虫和雄虫的引诱率分别为61.67%和70.00%,明显高于杨桃、番石榴和番木瓜,三者的引诱率分别为31.67%、31.67%和21.67%,与柑橘引诱率差异显著(P<0.05)。已知植物次生物质的生物测定结果表明,甲基丁香酚的引诱效果最佳,平均引诱率为45.00%,其中雄虫和雌虫的引诱率分别为86.67%和3.33%;乙酸乙酯和番石榴香精的引诱效果次之,平均引诱率分别为32.50%和28.33%,其中雌虫的引诱率均为36.67%;杨桃香精和柠檬酸的引诱效果较差,平均引诱率分别为25.00%和26.67%。  相似文献

Males of Bactrocera dorsalis (Hendel) are strongly attracted to methyl eugenol (ME), which occurs in over 450 plant species. Given this powerful attraction, ME is commonly used in surveillance and eradication programs against this invasive agricultural pest. Preliminary observations revealed that B. dorsalis males visited ME-bearing flowers of the fruit fly orchid (FFO) Bulbophyllum cheiri subsp. cheiri even when these occurred near traps baited with far greater quantities of ME. Based on this evidence, we undertook field experiments to assess the attraction of feral B. dorsalis males to FFO flowers relative to commercial sources of ME. At the edge of a secondary forest, an FFO flower was placed midway between two ME sources located 20 m apart, and attracted flies were collected over an entire day. When the ME sources were unenclosed (not in traps), the relative attractiveness of FFO flowers to B. dorsalis males varied with the amount of ME placed nearby. The FFO flower (i) attracted a similar proportion of males when 1 g ME was placed at the flanking sites but (ii) captured significantly smaller proportions when the nearby sites had 6 or 10 g commercial ME sources. Similar tests with the commercial sources enclosed in traps showed that (i) 6 g ME sources in Steiner traps attracted significantly more B. dorsalis males than FFO flowers but (ii) 10 g sources in Clear traps (1 L buckets with 4 entrance holes) did not outperform the FFO flowers, presumably owing to limited dispersion of volatiles from the trap. Implications of these results for surveillance programs are discussed.  相似文献

Traditional culturing methods take a long time for identification of pathogenic isolates. A protocol has been developed for the detection of Fusarium from soil samples in the early stage of infection. Seventeen soil samples from different locations were collected before the onset of rains to find out the presence of Fusarium spp. population present in the soil of guava orchards and to correlate its presence with incidence of wilt. A PCR based method was developed for the molecular characterization of Fusarium using Fusarium spp. specific primer. DNA extracted by this method was free from protein and other contaminations and the yield was sufficient for PCR amplification. The primer developed in this study was amplifying ∼230 bp in all infected samples while not in healthy soil. The specificity and sensitivity of primer were tested on several Fusarium spp. and found that this primer was amplifying 10−6 dilution of the fungal DNA. The present study facilitates the rapid detection of Fusarium spp. from infected soil samples of guava collected from different agroclimatic regions in India. A rapid detection method for pathogens and a diagnostic assay for disease would facilitate an early detection of pathogen and lead to more effective control strategies.  相似文献

Oriental fruit fly, Bactrocera dorsalis (Hendel), invaded French Polynesia in 1996. In 2002 a natural enemy, Fopius arisanus (Sonan), was released and established. By 2009 mean (±SD) F. arisanus parasitism for fruit flies infesting Psidium guajava (common guava), Inocarpus fagifer (Polynesian chestnut) and Terminalia catappa (tropical almond) fruits on Tahiti Island was 64.8 ± 2.0%. A second parasitoid, Diachasmimorpha longicaudata (Ashmead), was released and established in 2008. Although widespread, parasitism rates have not been higher than 10%. From 2003 (parasitoid establishment) to 2009 (present survey) numbers of B. dorsalis, Bactrocera tryoni (Froggatt), Queensland fruit fly, and Bactrocera kirki (Froggatt) emerging (per kg of fruit) declined. For example, for P. guajava there was a decline of 92.3%, 96.8%, and 99.6%, respectively. Analysis of co-infestation patterns (1998–2009) of B. dorsalis, B. tryoni, and B. kirki, suggest B. dorsalis is now the most abundant species in many common host fruits. Establishment of F. arisanus is the most successful example of classical biological control of fruit flies in the Pacific outside of Hawaii and can be introduced if B. dorsalis spreads to other French Polynesian islands, as was the recent case when B. dorsalis spread to the Marquesas Islands. These studies support F. arisanus as a prime biological control candidate for introduction into South America and Africa where Bactrocera carambolae Drew and Hancock and Bactrocera invadens Drew, Tsuruta, and White, respectively, have become established.  相似文献

Human α2-macroglobulin (A2M) is an abundant protease inhibitor in plasma, which regulates many proteolytic processes and is involved in innate immunity. A2M’s unique protease-trapping mechanism of inhibition is initiated when a protease cleaves within the exposed and highly susceptible “bait region.” As the wild-type bait region is permissive to cleavage by most human proteases, A2M is accordingly a broad-spectrum protease inhibitor. In this study, we extensively modified the bait region in order to identify any potential functionally important elements in the bait region sequence and to engineer A2M proteins with restrictive bait regions, which more selectively inhibit a target protease. A2M in which the bait region was entirely replaced by glycine-serine repeats remained fully functional and was not cleaved by any tested protease. Therefore, this bait region was designated as the “tabula rasa” bait region and used as the starting point for further bait region engineering. Cleavage of the tabula rasa bait region by specific proteases was conveyed by the insertion of appropriate substrate sequences, e.g., basic residues for trypsin. Screening and optimization of tabula rasa bait regions incorporating matrix metalloprotease 2 (MMP2) substrate sequences produced an A2M that was specifically cleaved by MMPs and inhibited MMP2 cleavage activity as efficiently as wild-type A2M. We propose that this approach can be used to develop A2M-based protease inhibitors, which selectively inhibit target proteases, which might be applied toward the clinical inhibition of dysregulated proteolysis as occurs in arthritis and many types of cancer.  相似文献

Severe oral mucositis occurs frequently in patients receiving hematopoietic stem cell transplantation (HCT). Oral mucosal bacteria can be associated with progression of oral mucositis, and systemic infection may occur via ulcerative oral mucositis. However, little information is available regarding the oral microbiota after HCT. Here, PCR-denaturing gradient gel electrophoresis (DGGE) was performed to characterize the oral mucosal microbiota, which can be affected by antibiotics, before and after HCT. Sixty reduced-intensity HCT patients were enrolled. Three patients with the least antibiotic use (quinolone prophylaxis and/or β-lactam monotherapy group) and three patients with the most antibiotic use (β-lactam-glycopeptide combination therapy group) were selected. Bacterial DNA samples obtained from the oral mucosa before and after HCT were subjected to PCR-DGGE. The trajectory of oral mucositis was evaluated. The oral mucosal microbiota in the β-lactam-glycopeptide combination therapy group was different from that in the quinolone prophylaxis and/or β-lactam monotherapy group, and Staphylococcus spp. and Enterococcus spp. were identified. Lautropia mirabilis was dominant in one patient. Ulcerative oral mucositis was observed only in the β-lactam-glycopeptide combination therapy group. In conclusion, especially with the use of strong antibiotics, such as glycopeptides, the oral mucosal microbiota differed completely from that under normal conditions and consisted of Staphylococcus spp., Enterococcus spp., and unexpectedly L. mirabilis. The normal oral microbiota consists not only of bacteria, but these unexpected bacteria could be involved in the pathophysiology as well as systemic infection via oral mucositis. Our results can be used as the basis for future studies in larger patient populations.  相似文献

The aim of this study was to identify the types and abundance of airborne bacteria of two health institutions (1 and 2) and to determine the genetic association between environmental and clinical isolates of Staphylococcus spp. Environmental sampling in institutions 1 and 2 was conducted for 1 year (dry and rainy seasons) using M Air T sampler. The bacteria and their susceptibility to antibiotics were identified. The colony-forming units per cubic meter (CFU/m3) of air were quantified for all the isolates, and the diversity and abundance of species were calculated. The genetic relationship between the clinical and environmental isolates of S. aureus obtained from institution 1 was established by the UPGMA based on RAPD markers. At both of the institutions, the genera most frequently isolated were Staphylococcus and Bacillus, and the greater concentration of airborne bacteria was detected during the dry season than the rainy season. The lowest diversity and highest dominance was found in the institution 2. On the other hand, at institution 1, the genus that was resistant to antibiotics was Staphylococcus, whereas at institution 2, no isolate was found to be resistant to antibiotics. Furthermore, no association between the clinical and environmental isolates of S. aureus was found. However, one clone was found in different areas of the institution 1. The presence of airborne pathogenic bacteria in institutions 1 and 2 is important to establish the measures for the prevention and control of nosocomial outbreaks.  相似文献

Systemic inflammation contributes to both the development of cancer and of cachexia. The microenvironment of bacterial habitats might be changed during the progression of cancer cachexia. The aim of this study was to quantitatively and qualitatively compare the composition of the skin microbiota between cancer cachexia patients and healthy volunteers. Cutaneous bacteria were swabbed at the axillary fossa of 70 cancer cachexia patients and 34 healthy individuals from China. Nested-PCR-denaturing gradient gel electrophoresis (PCR-DGGE) with primers specifically targeting V3 region and quantitative PCR (qPCR) for total bacteria, Corynebacterium spp., Staphylococcus spp., and Staphylococcus epidermidis were performed on all samples. Barcoded 454 pyrosequencing of the V3–V4 regions was performed on 30 randomly selected samples. By comparing diversity and richness indices, we found that the skin microbiome of cachectic cancer patients is less diverse than that of healthy participants, though these differences were not significant. The main microbes that reside on human skin were divided into four phyla: Firmicutes, Actinobacteria, Proteobacteria, and Bacteroidetes. Staphylococcus spp. and Corynebacterium spp. were the dominant bacteria at the genus level. Significantly fewer Corynebacterium spp. had been observed in cachexia patients compared to healthy subjects. These results suggest that the presence of cancer and cachexia alters human skin bacterial communities. Understanding the changes in microbiota during cancer cachexia may lead to new insights into the syndrome.  相似文献

Background

Observations that the airway microbiome is disturbed in asthma may be confounded by the widespread use of antibiotics and inhaled steroids. We have therefore examined the oropharyngeal microbiome in early onset wheezing infants from a rural area of tropical Ecuador where antibiotic usage is minimal and glucocorticoid usage is absent.

Materials and Methods

We performed pyrosequencing of amplicons of the polymorphic bacterial 16S rRNA gene from oropharyngeal samples from 24 infants with non-infectious early onset wheezing and 24 healthy controls (average age 10.2 months). We analyzed microbial community structure and differences between cases and controls by QIIME software.

Results

We obtained 76,627 high quality sequences classified into 182 operational taxonomic units (OTUs). Firmicutes was the most common and diverse phylum (71.22% of sequences) with Streptococcus being the most common genus (49.72%). Known pathogens were found significantly more often in cases of infantile wheeze compared to controls, exemplified by Haemophilus spp. (OR = 2.12, 95% Confidence Interval (CI) 1.82–2.47; P = 5.46×10−23) and Staphylococcus spp. (OR = 124.1, 95%CI 59.0–261.2; P = 1.87×10−241). Other OTUs were less common in cases than controls, notably Veillonella spp. (OR = 0.59, 95%CI = 0.56–0.62; P = 8.06×10−86).

Discussion

The airway microbiota appeared to contain many more Streptococci than found in Western Europe and the USA. Comparisons between healthy and wheezing infants revealed a significant difference in several bacterial phylotypes that were not confounded by antibiotics or use of inhaled steroids. The increased prevalence of pathogens such as Haemophilus and Staphylococcus spp. in cases may contribute to wheezing illnesses in this age group.  相似文献

Males of the oriental fruit fly, Bactrocera dorsalis (Hendel) and some of its sibling species have strong affinity for methyl eugenol (ME). Methyl eugenol ingested by male flies is biotransformed in the crop to two ME metabolites that eventually accumulate in the rectal gland, which is known to serve as a reservoir for B. dorsalis sex pheromones. When fed with ME, males of laboratory and wild B. philippinensis Drew and Hancock selectively accumulated two metabolites, 2-allyl-4,5-dimethoxyphenol and (E)-coniferyl alcohol, in the rectal gland, as was seen for B. dorsalis sensu stricto, B. invadens Drew, Tsuruta and White, and B. papayae Drew and Hancock. Phylogenetic analysis of COI and rDNA sequence data of these four taxa also revealed a close relationship among B. philippinensis, B. dorsalis s.s., B. invadens, and B. papayae (all four are members of the dorsalis species complex). This result corroborates pheromone analysis. The usefulness of pheromonal analysis as a chemotaxonomy tool to complement molecular and other analysis in differentiation of closely related sibling species within the Bactrocera dorsalis complex, for which use of morphological characters had been inadequate, is highlighted.  相似文献

Adult body size is one of the most ecologically relevant quantitative traits that underlies many other life-history traits of particular organism. In insects, there is positive intraspecific relationship between body size and female fecundity. In this study small scale temporal and spatial and space variability in structural body size of Poecilus cupreus and Anchomenus dorsalis was investigated. The beetles were collected in four fields near Prague-Suchdol in autumn 2009 and 2010, and in spring 2010, 2011 and 2012. In both species structural body size was significantly affected by sex (females were the larger sex). In A. dorsalis structural body size was also significantly affected by arable field identity, overwintering (post-overwintering individuals collected in spring were larger in comparison to pre-overwintering individuals collected in autumn), sampling year, overwintering by year and arable field by year interactions. Our results suggest that spatiotemporal variation in environmental conditions experienced by A. dorsalis during larval growth resulted in differences in adult structural body size among particular fields and particular sampling years. In addition, mean structural body size in A. dorsalis was affected by overwintering, which was probably caused by size-specific winter mortality. Moreover, effect of overwintering varied among years, probably according to the specific weather conditions during a particular winter.  相似文献

Skin microbiome main cultivable aerobes in human are coagulase-negative staphylococci and lipophilic corynebacteria. Staphylococcus strains (155) belonging to 10 species and 105 strains of Corynebacterium belonging to nine species from the skin swabs of healthy male volunteers were investigated to determine their enzymatic activity to main metabolic substrates: carbohydrates, proteins, lipids, and response to factors present on the skin such as osmotic pressure, pH, and organic acids. The results showed that lipophilic corynebacteria have different capacity for adaptation on the skin than staphylococci. Most of Corynebacterium spp. expressed lack of proteinase, phospholipase, and saccharolytic enzymes activity. Corynebacteria were also more sensitive than Staphylococcus spp. to antimicrobial agents existing on human skin, especially to low pH. These characters can explain domination of Staphylococcus genera on healthy human skin. It can be suggested that within these two bacterial genus, there exists conceivable cooperation and reciprocal protection which results in their quantitative ratio. Such behavior must be considered as crucial for the stability of the population on healthy skin.  相似文献
