Although generation of ultrasound during courtship has been reported for an increasing number of moth species, the effect of the ultrasound on mating remains uncertain in many cases because of a lack of proper verification. Here we report that males of the yellow peach moth Conogethes punctiferalis (Crambidae) sexually communicate with females by emitting loud ultrasound (103 dB peak equivalent sound pressure level at 1 cm; dominant frequency 82 kHz) before attempting copulation. The male ultrasound consists of consecutive clicks (pulses) in the early phase of the sound train and consecutive pulses (burst) in the late phase. When females were deafened by puncturing the abdominal tympanic membranes, copulation never occurred. We found that deafened females did not assume the wing-raising posture, which, for normal pairs, always precedes successful copulation. Our findings indicate that male courtship ultrasound evokes wing-raising as an acceptance behavior from females, which in turn evokes a copulation attempt by a male.  相似文献

Conogethes punctiferalis (Guénee) is a critical pest that commonly infests castor (Ricinus communis Linnaeus) and cardamom (Elettaria cardamomum Maton) in India. The moths of both castor and cardamom appear to be similar in wing pattern and color. However, the results of behavioral studies elicited a doubt that there may be differences in terms of host specialization. In the present study, we conducted morphological studies and DNA barcode analyses using cytochrome oxidase I gene, which unraveled the mystery of C. punctiferalis breeding on castor and cardamom. The differences in male aedeagus and female bursae were prominent, yet, not sufficient enough to say that they are different species. The results showed high haplotype diversity (0.817 ± 0.073) and nucleotide diversity (0.0285 ± 0.002) in C. punctiferalis. In addition, topologies of neighbor-joining trees indicate that Conogethes sp. breeding on castor belongs to C. punctiferalis while those on cardamom are of a separate clade. Further genetic analysis revealed significant genetic differentiations among the two sampled populations, reflecting limited gene flow. Neutrality tests and mismatch distributions showed population expansion in C. punctiferalis, while the results of an analysis of molecular variance (AMOVA) indicated the existence of significant genetic variation among the examined host races. Conclusively, analysis using mitochondrial DNA showed an amount of genetic divergence between the two host-associated populations compatible with cryptic species rather than host races.  相似文献

Chestnuts and walnuts are ones of most important forest products in Korea. However, Conogethes punctiferalis has been increasingly causing economic damages to chestnuts and walnuts. Currently, its management is mainly relayed on aerial application of pesticides. To select pesticides of C. punctiferalis for aerial application, damage rate of chestnut and walnut fruits and death rate of C. punctiferalis were analyzed according to selected pesticides. Experiments were conducted during two years by randomly applying chemical pesticide (i.e., Chlorfluazuron 5 % in 2018 and Thiacloprid 10 % in 2019), organic pesticide (i.e., paraffin oil 8 % + plant extracts (90 %) from Styphnolobium japonicum, Melia azedarach, and Dysphania ambrosioides), and no treatment (i.e., control) into an area which were divided into three from a whole commercial field. All pesticides used in this study could suppress damages of walnuts and chestnuts by C. punctiferalis. However, these pesticides could not kill their larvae inside of fruits. Thus, aerial spraying these pesticides for C. punctiferalis should be applied at optimal timing or slightly earlier than optimal timing. By considering honeybee toxicity, cost, and management efficiency, Chlorfluazuron 5 % was the best as an aerial application pesticide among these three pesticide in this study.  相似文献

Mating disruption using synthetic sex pheromones is an effective pest-control method to prevent the evolution of insecticide resistance. Although the sex pheromone of the yellow peach moth Conogethes punctiferalis (Guenée) (Lepidoptera, Crambidae) has already been identified and artificially synthesized, mating disruption in the field has been less successful than expected. Such ineffectiveness may be caused by genetic variation in the attractiveness of the sex pheromone across moth populations. To evaluate this possibility, we evaluated the genetic population structure of this species across 10 populations in Japan. We found that two clades were present at the mtDNA COI region, each clade was sympatrically distributed, and no genetic structure was detected among the populations. In addition, the frequencies of the two clades did not differ between the samples in pheromone traps using two types of blend and those hand collected from infested fruit. Our mating experiment revealed random mating between the two clades. Our results strongly suggest that the genetic variation in the attractiveness of the sex pheromone is minimal and symmetrical gene flow likely occurs between the clades in the field.

Grapholita molesta (Busck) (the oriental fruit moth; OFM) and Conogethes punctiferalis (Guenée) are both fruit and stem borers with overlapping habitats, occurrences, and outbreak cycles. These two species are in different families and they have completely different sex pheromone components. Here, the effects of the sex pheromone components of C. punctiferalis, (E)-10-hexadecenal (E10-16:Ald) and (Z)-10-hexadecenal (Z10-16:Ald) and their mixture, were evaluated on the sexual communication of OFM by examining electrophysiological (EAG) and behavioral responses. We found that a considerably large amount of E10-16:Ald or Z10-16:Ald and a tiny amount of OFM pheromone elicited comparable EAG responses in OFM males, indicating the low antennal olfactory sensitivity of OFM males to the heterospecific pheromone compounds. In two different peach orchard parcels, captures of OFM by their pheromone lures baited in delta traps were increased by at least 1.5 times when OFM pheromone lures were combined with lures that contained 1000 μg of either E10-16:Ald, Z10-16:Ald or their mixture. In two other pear orchard parcels, both E10-16:Ald and Z10-16:Ald increased captures of OFM in Unitraps in a dose-dependent manner with more than a four-fold increase. Therefore, the enhanced attractiveness of OFM lures by the two interspecific pheromones suggests their potential application to improve mass trapping, population monitoring and mating disruption of OFM.  相似文献

The carrion beetle, Necrodes nigricornis Harold, 1875 (Coleoptera: Silphidae) is one of the most frequently encountered insect species among Silphidae in animal cadavers in Korea. Consequently, the postmortem colonization of the species can provide useful information regarding the time-since-death. In this study, we report the mitochondrial genome (mitogenome) characteristics of N. nigricornis with the aim of increasing the genomic data pools of the family Silphidae, which is of considerable forensic importance. The full-length circular genome is 18,503 bp, with 75.60 % A/T content. It contains a typical set of 37 metazoan genes, but the non-coding A + T-rich region is unusually long at 3,818 bp in length, containing two similar-sized repeats (539 and 542 bp) with a slight length and nucleotide variation. The gene arrangement of the species is identical to that of the ancestral arrangement found in the majority of insects. The biased A/T content in the genome is also reflected in the form of biased codon usage; six codons among 62 comprised solely of A/T nucleotides (TTA, ATT, TTT, ATA, TTA, and TAT) were highly used, accounting for 41.28 % of the total. The maximum-likelihood tree built using 12 PCG (excluding ND2) and lrRNA revealed monophyletic Silphidae, Silphinae, and Nicrophorinae, with relatively higher nodal supports (bootstrap support ≥ 80 %). N. nigricornis, belonging to Silphinae, was placed as the sister to congeneric N. littoralis with the highest nodal support. We believe this mitogenome sequences of the carrion beetle can become an important baseline information for future studies on phylogeny at various levels of taxonomic diversity, intra-specific variation, as well as species identification.  相似文献

Insect sound-producing apparatuses are mostly classified into two types: file–scraper and tymbal. Structures and locations of these organs are conserved in some phylogenetic groups, e.g., crickets, grasshoppers, and cicadas. However, moths have evolved diversified sound-producing organs, such as wing castanets and proboscis, in addition to the file–scraper and tymbal, in each species. Here we demonstrate that the yellow peach moth Conogethes punctiferalis (Guenée) (Lepidoptera: Crambidae) has developed mesothoracic tymbal organs never reported so far in insects. Tymbals are male specific and used for generating ultrasonic clicks in mating. We found eight to nine striae on the smooth surface of the tymbal membrane, suggesting the production of several clicks by a single buckle of the membrane in association with contraction/relaxation of the mesothoracic muscles. Acoustic data from click sequences support the idea that the series is generated by side-to-side asynchrony with an active/passive half cycle by an inward/outward buckle, and thus in click group (pulse) production, males emit 28 clicks with the right and left tymbals. The click-producing mechanism is similar, but not homologous, to those of other clicking species in five moth families. Thus, moths have acquired tymbal organs through independent and convergent evolution.  相似文献

We monitored hitchhiker insect pests on vessels arriving to Korea from foreign countries. A survey of 416 foreign vessels was conducted from June 1, 2018 to October 29, 2019. A total of 1,141 hitchhiker insects were collected by interception or by hand. Individuals were identified to the species level whenever possible using DNA barcoding and morphological re-examination. Of the 1,141 hitchhikers, a total of 439 species from 81 families and 12 orders, including 54 species (93 individuals) not-distributed in Korea, were identified. Among the 93 individuals from the 54 species not distributed in Korea, 78 individuals were collected alive. In particular, three regulated species in Korea, Erthesina fullo (Pentatomidae, Hemiptera) (hitchhiking route: China > Yeosu, Korea), Tessaratoma papillosa (Tessaratomidae, Hemiptera) (China > Yeosu, Korea), Noctua pronuba (Noctuidae, Lepidoptera) (Singapore > Pohang, Korea) were detected, nine species, Dicranocephalus wallichii (Scarabaeidae, Coleoptera), Palpita quadristigmalis (Crambidae, Lepidoptera), Lymantria xylina (Erebidae, Lepidoptera), Odontopera aurata, Semiothisa cinerearia (Geometridae, Lepidoptera), Mythimna pallidicosta (Noctuidae, Lepidoptera), Euhampsonia serratifera (Notodontidae, Lepidoptera), Arippara disticha (Pyralidae, Lepidoptera), and Psilogramma lukhtanovi (Sphingidae, Lepidoptera) were discovered as multiple detection over years or months, and three species, Lemyra rhodophilodes (Erebidae, Lepidoptera), Malacosoma dentata (Lasiocampidae, Lepidoptera), and Chondracris rosea (Acrididae, Orthoptera) were multiple detected from single vessel. Therefore, we suggest that hitchhiker monitoring on vessels travelling along navigated routes should be conducted in addition to performing a risk assessment of hitchhiker insect pest species not distributed in Korea that includes regulated species.  相似文献

本文应用ISSR分子标记技术对中国桃蛀螟Conogethes punctiferalis 11个地理种群的基因组DNA进行了遗传多样性和遗传分化分析。从34条引物中筛选出6条用于ISSR多态性分析, 共扩增出211条带, 其中209条具多态性, 总的多态性条带百分率为99.05%。11个种群的遗传距离在0.0059~0.0237之间, Nei氏基因多样性指数为0.1750, Shannon信息指数为0.2966, 遗传分化系数为0.053, 基因流高达8.8724。结果提示中国桃蛀螟地理种群因基因流水平较高而种群遗传分化水平保持较低。  相似文献

The mitochondrial genetic markers are considered useful tools for discrimination between more closely related lepidopteran taxa. Therefore, the present study aimed to investigate the role of mitochondrial (mt) 16 s rRNA gene in the determination of the taxonomic position for two moth species within Ditrysia clade. Maximum likelihood analysis has indicated a well-supported dendrogram based on the Tamura-Nei model for the recovered lepidopterans. The mt 16 s rRNA query sequences from 24 species within seven families were analyzed. This analysis and bootstrap confidence revealed two major clades representing Glossata suborder within Lepidoptera, with a close relationship of Noctuoidea + (Pyraloidea (Hesperioidea + Papilionoidea)). The subfamily Heliothinae forming a sister group with Risobinae (Noctinae + Hadeninae). In addition, there is a clear observation about the close relation between Phycitinae + Galleriinae within Pyraloidea and Cyrestinae + Limenitidinae within Papilionoidea. The present study supported that the Helicoverpa and Meroptera species are the first accounts of these genera inhabiting Saudi Arabia.  相似文献

The developmental biology of Trichogrammatoidea lutea Girault (Hymenoptera: Trichogrammatidae) was studied at six constant temperatures (18, 21, 24, 27, 30 and 35 °C) on eggs of three lepidopteran host species: Helicoverpa armigera (Hübner) (Noctuidae), Chilo partellus (Swinhoe) (Crambidae) and Cadra cautella (Walker) (Pyralidae). T. lutea did not complete development at 35 °C on any of the three host species. Parasitism levels were highest on H. armigera at 27 °C (58%), C. cautella at 27 and 30 °C (31% and 28%) and C. partellus between 24 and 30 °C (13–17%). Realized progeny of T. lutea per parasitized host egg was influenced by host size. The number of progeny of T. lutea per parasitized host egg was highest on H. armigera, followed by C. partellus and lowest on C. cautella. The sex ratio was female biased on C. partellus, female biased on C. cautella with the exception of 21 °C and close to 1:1 on H. armigera. The rate of development from egg to pupa and egg to adult was fastest on H. armigera and slowest on C. partellus. Lower thresholds for development and degree days (DD) of T. lutea from egg to adult were 12.8 °C and 105.4 DD on H. armigera, 11.3 °C and 141.6 DD on C. partellus and 12.9 °C and 118.2 DD on C. cautella, respectively. Based on these results, H. armigera is the most suitable host for mass rearing of T. lutea for biological control of Lepidoptera pests because of the relatively high parasitism levels, short development time, greater clutch size and balanced sex ratio. C. cautella may also be used although longer exposure times might be required due to lower parasitism levels.  相似文献

The perilla seed bugs, Nysius plebeius Distant and Nysius hidakai Nakatani, are emerging pests of several field crop species including perilla crop in Korea. Here, we investigated the oviposition, nymphal mortality, development, adult emergence, adult longevity, adult weight and female ratio of N. plebeius and N. hidakai on multiple (MC) and double (DC) combinations of five crop seed species: perilla [Perilla frutescens (L.) Britton.], sesame (Sesamum indicum L.), sorghum [Sorghum bicolor (L.)], proso millet (Panicum miliaceum L.), and foxtail millet (Setaria italic L.) in no-choice test. The life-history variables of N. plebeius and N. hidakai were significantly influenced by the crop seed combinations. The most preferred crop seed combinations for oviposition were MC (perilla + sorghum + sesame + proso millet + foxtail millet), followed by DC (perilla + foxtail millet and perilla + sorghum), whereas the least preferred seed combination was DC (proso millet + foxtail millet). Both Nysius sp. were developed faster in MC (perilla + sorghum + sesame + proso millet + foxtail millet), with minimal nymphal mortality, higher adult emergence, longer adult longevity and heavier adult weight. The positive adult bug’s performance on the MC (perilla + sorghum + sesame + proso millet + foxtail millet) may be due to the nutrient contents and physicochemical components associated with the crop seeds. Further, the findings of this study are discussed in terms of bug's behavioral response to crop seed species combinations and management strategies of Nysius sp. as an alternative to chemical management.  相似文献

Even though the spermatozoa of several strepsipteran species were described earlier, no data were available for the basal family Mengenillidae. Well-fixed material of the recently described Tunisian species Mengenilla moldrzyki was used for a detailed examination of the sperm ultrastructure. The total length is c. 30 μm. The head region contains a conical acrosome vesicle (0.3-0.35 μm) and an elongated nucleus (7.3 μm) with dense chromatin. Some granular material along with a uniformely dense centriole adjunct and two mitochondrial derivatives are visible at the posterior end of the nucleus. The material of the centriole adjunct does not extend along the flagellum and accessory bodies are absent. The mitochondrial derivatives are elongated structures crossed by a longitudinal crista but lacking parallel transverse cristae and paracrystalline material in the dense matrix. The mitochondrial derivatives gradually reduce their size and end at the most posterior tail region. The flagellar axoneme has a 9 + 9 + 2 pattern and originates beneath the nucleus. In the terminal tail region the axoneme gradually disintegrates. Despite the extreme specialization of the endoparasitc group, strepsipteran spermatozoa are mostly characterized by plesiomorphies. The pattern within the order is largely uniform, but Mengenilla displays several apomorphic features compared to the presumptive strepsipteran groundplan (e.g., absence of crystallizations and cristae in the mitochondrial derivatives). The subdivision of the intertubular material into two compartments with a dense beak-like structure adhering to the tubular wall supports a clade Coleopterida (=Strepsiptera + Coleoptera) + Neuropterida.  相似文献

The Dipsadidae contains more than 700 extant species belonging to 92 genera and is the largest family of American snakes. In this work, we built a data set including two mitochondrial genes (12S and 16S rRNA) for 125 dipsadid taxa belonging to 59 genera, in order to gain further insights on the phylogenetic relationships of this large group at the subfamilial and generic levels. Among dipsadines, the monotypic genus Nothopsis is the sister-group to Leptodeira. Among xenodontines, the monophyly of seven previously recognized tribes (Alsophiini, Elapomorphini, Hydropsini, Philodryadini, Pseudoboini, Tachymenini and Xenodontini) is confirmed. Among Xenodontini, the genus Liophis is paraphyletic with respect to Erythrolamprus and Umbrivaga and workers should be aware of the inadequacy of the current taxonomy. Finally, the following genera could not confidently be allocated to the above tribes: Caaeteboia, Echinantera and Taeniophallus, Tropidodryas, Manolepis and Pseudalsophis, Xenopholis, Psomophis, Hydrodynastes, Conophis and Crisantophis.  相似文献

Loxostege turbidalis, Loxostege aeruginalis, Pyrausta despicata, and Crambus perlellus belong to Crambidae, Pyraloidea. Their mitochondrial genomes (mitogenomes) were successfully sequenced. The mitogenomes of L. turbidalis, L. aeruginalis, P. despicata, and C. perlellus are 15 240 bp, 15 339 bp, 15 389 bp, and 15 440 bp. The four mitogenomes all have a typical insect mitochondrial gene order, including 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and one A + T rich region (control region). The PCGs are initiated by the typical ATN codons, except CGA for the cox1 gene. Most PCGs terminate with common codon TAA or TAG, the incomplete codon T is found as the stop codon for cox2, nad4, and nad5. Most tRNA genes exhibit typical cloverleaf structure, except trnS1 (AGN) lacking the dihydrouridine (DHU) arm. The secondary structure of rRNA of four mitogenomes were predicted. Poly-T structure and micro-satellite regions are conserved in control regions. The phylogenetic analyses based on 13 PCGs showed the relationships of subfamilies in Pyraloidea. Pyralidae, and Crambidae are monophyletic, respectively. Pyralidae comprises four subfamilies, which form the following topology with high support values: (Galleriinae + ((Pyralinae + Epipaschiinae)+ Phycitinae)). Crambidae includes seven subfamilies and is divided into two lineages. Pyraustinae and Spilomelinae are sister groups of each other, and form the “PS clade.” Other five subfamilies (Crambinae, Acentropinae, Scopariinae, Schoenobiinae, and Glaphyriinae) form the “non-PS clade” in the Bayesian inference tree. However, Schoenobiinae is not grouped with the other four subfamilies and located at the base of Crambidae in two maximum likelihood trees.  相似文献

Two new mammalian carnivoraform species, Uintacyon hookeri sp. nov. and Quercygale smithi sp. nov., are described from the early Eocene of Europe. U. hookeri sp. nov. is recorded in Mutigny (MP8 + 9, PE IV), Avenay (MP8 + 9, PE V), Brasles, Condé‐en‐Brie (MP8 + 9) and Cuis (MP 10), while Q. smithi sp. nov. comes from Mutigny and Mancy (MP10). Because the two species are not recorded in earliest Eocene localities such as Dormaal and Le Quesnoy (MP7, PE I), it is proposed that they dispersed after the main phase of the Mammal Dispersal Event. U. hookeri sp. nov. supports the existence of terrestrial connections with North America, while Q. smithi sp. nov. implies possible faunal exchange with Asia. This evidence for the evolution of the Carnivoraformes supports: (1) a rapid decrease in their diversity after the Mammal Dispersal Event; and (2) the existence of a mammal turnover event in Europe during the early Eocene. The discovery of a new species of Quercygale, which is generally considered as the closest carnivoraform to the crown‐group Carnivora, shows that the genus had already lost the M3 by the early Eocene and supports an important, but very poorly known, radiation of the carnivoraforms at least during the earliest early Eocene.  相似文献

The surface and structure of the chorion of eggs of Diatraea saccharalis (F.) (Lepidoptera: Crambidae), Anticarsia gemmatalis (Huebner), Heliothis virescens F., Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae), Sitotroga cerealella (Oliver) (Lepidoptera: Gelechiidae), Ephestia kuehniella Zeller and Corcyra cephalonica Stainton (Lepidoptera: Pyralidae), that are hosts of Trichogramma galloi Zucchi and Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) were studied on SEM and TEM. Other characteristics of these eggs, such as changes in their color during embryonic development, size and volume were also recorded. Sculpturing and texture of the surface of the chorion greatly varied among the species studied, as well as the number of layers of the chorion and their thickness. Eggs of the factitious hosts were among the smallest and their volume was very close to each other. All these characteristics would provide basic information for a better understanding of the host selection behavior and are useful for the development of a suitable artificial host egg for the in vitro rearing of these egg parasitoids.  相似文献

Lepidopterous stem borers are the main field insect pests that attack maize, Zea mays L. in tropical Africa. A survey was carried during the long and short rain cropping seasons of 2002 / 2003 across six main agro-climatic zones (ACZs) to determine the spatial distribution of important stem borer species in Kenya. A total of 474 visits were made in the seventy-eight localities conveniently chosen to represent each of the six ACZs. 189,600 stems were checked for infestation, of which 27,799 infested stems were destructively cut and dissected for stem borer larvae identification. An average of 1.4 stem borer larvae were recovered per infested plant. 54.5% of the recovered larvae were identified as Chilo partellus (Swinhoe) (Crambidae), 39.7% as Busseola fusca (Fuller) (Noctuidae), 4.5% as Sesamia calamistis Hampson (Noctuidae) and 0.8% as Chilo orichalcociliellus (Strand) (Crambidae). Minor species present included Eldana saccharina Walker (Pyralidae), Sesamia nonagrioides (Lefebvre) (Noctuidae), Sesamia cretica Lederer (Noctuidae), Sesamia sp. (Noctuidae), Sciomesa piscator Fletcher (Noctuidae), Busseola sp near phaia (Noctuidae), Chilo sp, Ematheudes sp 1 (Pyralidae) and Ematheudes sp 2 (Pyralidae). Farms were grouped into respective ACZs for statistical analysis and subsequent comparison of dominant species. Results indicated that B. fusca was the dominant stem borer species in high potential zones (highland tropics, moist transitional zone and moist midaltitude) while the exotic C. partellus dominated smallholder farms in low potential zones (dry midaltitude, dry transional and lowland tropical zone). Within each ACZs, there was evidence of variation in species proportions between seasons. These spatio-temporal differences in community structure are discussed in terms of agro-climatic biological adaptations.  相似文献

为明确寄主植物对桃蛀螟生长发育及产卵选择行为的影响,利用实验种群生命表和二项产卵选择试验,研究了玉米、大豆、棉花和桃等4种寄主植物对桃蛀螟种群生长发育及产卵选择性的影响。结果表明:取食棉花的桃蛀螟幼虫存活率最低、幼虫历期最长,取食玉米的幼虫存活率最高、幼虫历期最短,取食桃和大豆的幼虫存活率和历期居于棉花处理组和玉米处理组之间;玉米处理组的桃蛀螟化蛹率、蛹重和蛹历期均为最高,棉花处理组为最低,大豆和桃处理组的这些参数均显著小于玉米处理组而大于棉花处理组;发育至成虫后,取食玉米的桃蛀螟羽化率显著高于其他3个处理组;取食桃的桃蛀螟成虫寿命(雌虫和雄虫)及个体发育历期均显著高于其他3种处理组;同时取食桃的桃蛀螟单雌产卵量最高,其次是玉米处理组,两者均显著高于大豆和棉花处理组。二项产卵选择试验结果显示,桃蛀螟雌蛾在棉花和玉米处理组、玉米和大豆处理组或棉花和大豆处理组间的落卵量差异不显著;但在包含桃的处理组中,桃蛀螟在棉花、玉米或大豆处理区的落卵量均显著高于桃处理区。上述结果表明,供试4种寄主植物中,桃蛀螟偏好在棉花、玉米和大豆上产卵,其中玉米对桃蛀螟的适合度相对较高,棉花对桃蛀螟的适合度相对较低。  相似文献

Asymptomatic malaria parasite carriers do not seek anti-malarial treatment and may constitute a silent infectious reservoir. In order to assess the level of asymptomatic and symptomatic carriage amongst adolescents in a highly endemic area, and to identify the risk factors associated with such carriage, we conducted a cross-sectional survey of 1032 adolescents (ages 10–19 years) from eight schools located in Ibadan, southwestern Nigeria in 2016. Blood films and blood spot filter paper samples were prepared for microscopy and DNA analysis. The prevalence of asymptomatic malaria was determined using microscopy, rapid diagnostic tests and PCR for 658 randomly selected samples. Of these, we found that 80% of asymptomatic schoolchildren were positive for malaria parasites by PCR, compared with 47% and 9%, determined by rapid diagnostic tests and microscopy, respectively. Malaria parasite species typing was performed using PCR targeting the mitochondrial CoxIII gene, and revealed high rates of carriage of Plasmodium malariae (53%) and Plasmodium ovale (24%). Most asymptomatic infections were co-infections of two or more species (62%), with Plasmodium falciparum + P. malariae the most common (35%), followed by P. falciparum + P. malariae + P. ovale (21%) and P. falciparum + P. ovale (6%). Single infections of P. falciparum, P. malariae and P. ovale accounted for 24%, 10% and 4% of all asymptomatic infections, respectively. To compare the species composition of asymptomatic and symptomatic infections, further sample collection was carried out in 2017 at one of the previously sampled schools, and at a nearby hospital. Whilst the species composition of the asymptomatic infections was similar to that observed in 2016, the symptomatic infections were markedly different, with single infections of P. falciparum observed in 91% of patients, P. falciparum + P. malariae in 5% and P. falciparum + P. ovale in 4%.  相似文献
