Despite the use of Terminalia catappa (TC) leaf by traditional fish farmers around the world to improve the health status of cultured fish, there is a paucity of information on comprehensive metabolite profile and the maximum safe dose of the plant. This study aims at profiling the methanol leaf extract of T. catappa, quantifying total phenolic content (TPC) as well as the total flavonoid content (TFC) and evaluating its acute toxicity on blood, plasma biochemical parameters and histopathology of some vital organs in red hybrid tilapia (Oreochromis sp.). The experimental fish were acclimatised for 2 weeks and divided into six groups. Group (1) served as a control group and was administered 0.2 ml,g−1 of phosphate buffer saline (PBS). Groups 2–6 were orally administered T. catappa leaf extracts (0.2 ml.50 g−1) in the following sequence; 31.25, 62.5, 125, 250 and 500 mg.kg−1 body weight. The metabolites identified in T. catappa using liquid chromatography-tandem mass electrospray ionisation spectrometry (LC-ESI-MS/MS) revealed the presence of organic acids, hydrolysable tannins, phenolic acids and flavonoids. Phenolic quantification revealed reasonable quantity of phenolic compounds (217.48 μg GAEmg−1 for TPC and 91.90 μg. QCEmg−1 for TFC). Furthermore, there was no significant difference in all the tested doses in terms of blood parameters and plasma biochemical analysis except for the packed cell volume (PCV) at 500 mg.kg−1 when compared to the control. Significant histopathological changes were observed in groups administered with the extract at 125, 250 and 500 mg.kg−1 doses. To a very large extent it is therefore safe to administer the extract at 31.25 and 62.5 mg.kg−1 in tilapia.  相似文献

Soil salinization is a global issue impeding horticulture production and is approaching an alarming status due to climate change and urbanization. Breeding salt-tolerant rootstock varieties is an ideal strategy to mitigate stress due to salinity in mango and other perennial fruit species. Stress combating strategies employed by seedlings of 7 mango were studied under saline conditions (200 mM NaCl, EC: > 4.0 dSm−1, pH 8.5) in pot experiments. Significantly high accumulation of proline (19.07 µg g−1 FW in Bappakai), glycine betaine (55.11 µg g−1 FW in 13–1), and total sugars (17.33 mg g−1 FW in Kurukkan) were found to be the common mechanism employed by the tolerant cultivars to counter the osmotic stress, under suboptimal conditions. Non-enzymatic antioxidants viz., tannins (17.18 mg g−1), phenols (18.68 mg g−1), and anthocyanins (1.59 mg g−1) were increased in seedling of "13–1", the salt-resistant cultivar from Israel. Reactive oxygen species (ROS) regulation by increased activity of superoxide dismutase and catalase in the two polyembryonic cultivars of Indian origin (Kurukkan and Nekkare) suggests their potential use as rootstocks to combat oxidative stress. The tolerance index of various cultivars was calculated by averaging the scores of morphological stress indicators, and its correlation with studied parameters suggests that salinity resilience is more tightly linked to enhanced catalase accumulation (r2 = 0.8361) that is reduced ionic stress. This evidence assign the role of osmotic stress alleviation and redox regulation in salt tolerance mechanism operational in native Indian cultivars, Nekkare and Kurukkan at par with known salt tolerant rootstocks.

Plants have always been the prime focus in medicine industries due to their enormous ethnobotanical uses and multitude of biological and therapeutic properties. In the current study, preliminary phytochemical composition, Total phenolic content (TPC), and total flavonoid content (TFC) with the antioxidant and antibacterial activity of hydroalcoholic extract and n-hexane, chloroform and n-butanol fractions of five selected medicinal plants [Tephrosia purpurea (L.) Pers., Lavandula stoechas L., Aesculus indica (Wall. ex Cambess.) Hook, Iris ensata Thunb., and Kalanchoe pinnata (Lam.) Pers.] from Pakistan, have been evaluated. TPC and TFC were determined by Folin-Ciocalteu’s and AlCl3 methods respectively. The antioxidant activity was performed by DPPH, ABTS, FRAP, and CUPRAC while the antibacterial potential of these plants was determined by agar well diffusion assay. K. pinnata (Lam.) Pers. exhibited the highest TPC (695 ± 13.2 mg.GA.Eq.g-1DE ± SD) in n-butanol fraction and the highest TFC in its chloroform faction (615 ± 6.31 mg Q.Eq.g−1 DE ± SD). The n-butanol fraction and hydroalcoholic extract of I. ensata Thunb. exhibited strong antioxidant potential by DPPH and CUPRAC assays respectively, whereas K. pinnata (Lam.) Pers. n-butanol fraction exhibited the strongest reducing potential. The hydroalcoholic extract of all tested plants exhibited significant antibacterial activity against tested bacterial strains with ZI (12–18 mm). Conclusively, K. pinnata (Lam.) Pers. (Family: Crassulaceae) and I. ensataThunb. (Family: Iridaceae) exhibited the highest antioxidant and antibacterial potential. They can be explored for the isolation of phytoconstituents responsible for this potential and serve as a lead for the production of new natural antioxidants and antibacterial agents that can be used to cure various diseases.  相似文献

Little is currently known about the potential impact of silver nanoparticles (AgNPs) on estuarine microbial communities. The Colne estuary, UK, is susceptible to oil pollution through boat traffic, and there is the potential for AgNP exposure via effluent discharged from a sewage treatment works located in close proximity. This study examined the effects of uncapped AgNPs (uAgNPs), capped AgNPs (cAgNPs) and dissolved Ag2SO4, on hydrocarbon-degrading microbial communities in estuarine sediments. The uAgNPs, cAgNPs and Ag2SO4 (up to 50 mg L−1) had no significant impact on hydrocarbon biodegradation (80–92% hydrocarbons were biodegraded by day 7 in all samples). Although total and active cell counts in oil-amended sediments were unaffected by silver exposure; total cell counts in non-oiled sediments decreased from 1.66 to 0.84 × 107 g−1 dry weight sediment (dws) with 50 mg L−1 cAgNPs and from 1.66 to 0.66 × 107 g−1 dws with 0.5 mg L−1 Ag2SO4 by day 14. All silver-exposed sediments also underwent significant shifts in bacterial community structure, and one DGGE band corresponding to a member of Bacteroidetes was more prominent in non-oiled microcosms exposed to 50 mg L−1 Ag2SO4 compared to non-silver controls. In conclusion, AgNPs do not appear to affect microbial hydrocarbon-degradation but do impact on bacterial community diversity, which may have potential implications for other important microbial-mediated processes in estuaries.  相似文献

Aside from a companion investigation to this study, there are currently no peer-reviewed phosphorus (P) release rate data for northern North American (i.e., Canadian) reservoirs. Using Lake Diefenbaker, Saskatchewan, Canada as a case study, we tested the effect of variation in overlying water DO conditions on the P release rates from sediment cores. Sediment cores from four down-reservoir locations in Lake Diefenbaker were incubated under high (>8 mg l−1), low (2–3 mg l−1), or anoxic (<1 mg l−1) DO concentrations. Sediment cores were then analyzed for total P (TP) and three geochemical P fractions to assess how the DO regime influenced sediment P inventory. Maximum P release rates were highest under anoxic conditions and similar among sites (15.0–20.3 mg m−2 day−1), with the low-DO rates intermediate to the high-DO and anoxic P fluxes. Predictive internal P loading models considering only hypolimnetic anoxia may therefore oversimplify and thus underestimate P mobilization in situ. Non-apatite inorganic P (54 ± 10% across sites) from the top 1 cm of the sediment profile was the main source of P released during incubations, indicating that sampling on a coarser scale of resolution could obscure the relationship between sediment geochemistry and short-term P flux.

Endophytic fungi are the endogenous micro-organisms to interacting with the plant cells, which do not exhibit any symptoms on the host plant and may produce some of the main secondary metabolites of the host plant cells. Ferula assa-foetida is a perennial and endemic medicinal plant of Iran, which is a rich source of sesquiterpene, coumarins, polysulfides and phenolic acids. In this study, 28 endophytic fungi isolates including Fusarium (60·7%), Aspergillus (7·1%), Alternaria (17·9%) and Plectosphaerella (7·1%) were isolated from F. assa-foetida root (57·1%), stem (32·1%) and leaf (10·8%) collected from Parvand protected area. Subsequently, their ability to produce phenolic acids was evaluated. The high amounts of total phenol (326·09 mg g−1 of dry weight, DW), total flavonoid (901·11 mg g−1 DW) and antioxidant activity (247·96 mg l−1) were found in the supernatant fluid of SBUp1 isolate. The high-performance liquid chromatography analysis of 14 phenolic acids showed that rosmarinic acid (RA) is the main phenolic acid in the supernatant fluid of SBUp1 by 64·11 mg g−1 DW confirmed by the liquid chromatography coupled with mass spectrometric analysis. According to morphological identification followed by phylogenetic study based on internal transcribed spacer (ITS) sequencing (ITS1-5.8S-ITS2) analysis, the SBUp1 isolate was identified as Alternaria tenuissima. Eventually, to our knowledge, it is the first document confirming A. tenuissima as an endophytic fungus of F. assa-foetida, which is a rich source of RA.  相似文献

This study evaluated the production of cellulolytic enzymes from different agricultural residues. The crude enzyme extract produced was characterized and applied for saccharification of some agricultural residues. Maximum cellulolytic activities were obtained using soybean hulls. All enzymatic activities were highly stable at 40 °C at a pH range of 4.5–5.5. For stability at low temperatures, the enzyme extract was stored at freezing temperature and cooling for about 290 days without major loss of activity. The Km values found for total cellulase (FPase), endoglucanase (CMCase), and xylanase were 19.73 mg ml−1, 0.65 mg ml−1, and 22.64 mg ml−1, respectively, and Vmax values were 0.82 mol min−1 mg−1, 0.62 mol min−1 mg−1, and 104.17 mol min−1 mg−1 to cellulose, carboxymethyl cellulose, and xylan, respectively. In the saccharification tests, the total amount of total reducing sugars (TRS) released from 1 g of soybean hulls catalyzed by the enzymes present in the crude enzyme extract was 0.16 g g−1 dry substrate.

The effect of light on BPA degradation by an adapted bacterial consortium was investigated. BPA was completely degraded up to 50 mg l−1, and the degradation followed first-order reaction kinetics both in the light and in the dark. The degradation half-life of BPA when the consortium was grown in presence of light was 21.9, 17.2, and 12.6 h for concentrations of 10, 20, and 50 mg l−1, respectively; the degradation half-life of BPA in the dark was 13.1, 10.8, and 10.2 h for concentrations of 10, 20, and 50 mg l−1, respectively. Therefore, light inhibited BPA biodegradation. However, under both conditions, BPA was completely depleted. The bacterial consortium effectively utilised BPA as a growth substrate to sustain a cell yield of 0.95 g g−1 and 0.97 g g−1 in the light and dark, respectively. A total of ten and nine biodegradation intermediates were detected in the light and dark, respectively. Three bacterial metabolic pathways and one photodegradation pathway were proposed to explain their occurrence. This study demonstrated that bacterial consortia may assemble a wide range of catabolic pathways to allow for efficient degradation of BPA, converting BPA to principally bacterial biomass and metabolites exhibiting low or no oestrogenic activity.  相似文献

Warburgia ugandensis Sprague is a woody species in the family Canellaceae and an important source of medicines in Africa. Natural propagation of W. ugandensis is problematic due to its recalcitrant seeds and lack of an efficient in vitro regeneration system for this species. This study describes an efficient regeneration protocol. Petiole bases and shoot tips were used as explants. Callus tissue developed when the explants were cultured on Murashige and Skoog medium containing 30 g L−1 sucrose and 7 g L−1 agar (MS30 medium), supplemented with 1.0 mg L−1 indole-3-butyric acid (IBA), 1.6 mg L−1 6-benzylaminopurine (BA), and 0.1 mg L−1 thidiazuron (TDZ). Adventitious buds were efficiently induced from the callus when the MS30 medium was supplemented with 0.8 mg L−1 BA and 0.2 mg L−1 IBA. Root induction occurred within 7–10 d on half-strength MS30 medium supplemented with 0.8–1.0 mg L−1 1-napthalene acetic acid (NAA), 0.2 mg L−1 IBA, and 0.03% (w/v) activated charcoal (AC). Roots were followed by root elongation on the same medium but lacking NAA and IBA. Approximately 50% of the plantlets cultured produced roots, while more than 80% of the plantlets survived and successfully grew to maturity.

The development of a simple method to synthesize highly efficient and stable magnetic microsphere beads for sulfathiazole (STZ) removal from contaminated aqueous media was demonstrated in this study. Conocarpus (Conocarpus erectus L.) tree waste (CW) derived biochar (BC) was modified to fabricate chitosan-BC (CBC) and magnetic CBC (CBC-Fe) microsphere beads. Proximate, chemical, and structural properties of the produced adsorbents were investigated. Kinetics, equilibrium, and pH adsorption batch trials were conducted to evaluate the effectiveness of the synthesized adsorbents for STZ removal. All adsorbents exhibited the highest STZ adsorption at pH 5.0. STZ adsorption kinetics data was best emulated using pseudo-second order and Elovich models. The equilibrium adsorption data was best emulated using Langmuir, Freundlich, Redlich–Peterson, and Temkin models. CBC-Fe demonstrated the highest Elovich, pseudo-second order, and power function rate constants, as well as the highest apparent diffusion rate constant. Additionally, Langmuir isotherm predicted maximum adsorption capacity was the highest for CBC-Fe (98.67 mg g−1), followed by CBC (56.54 mg g−1) and BC (48.63 mg g−1). CBC-Fe and CBC removed 74.5%–108.8% and 16.2%–25.6% more STZ, respectively, than that of pristine BC. π-π electron-donor–acceptor interactions and Lewis acid-base reactions were the main mechanisms for STZ removal; however, intraparticle diffusion and H-bonding further contributed in the adsorption process. The higher efficiency of CBC-Fe for STZ adsorption could be due to its magnetic properties as well as stronger and conducting microsphere beads, which degraded the STZ molecules through generation of HO radicals.  相似文献

Bioactive compounds entrapped in plant materials can be effectively recovered using fungal enzymes. Cinnamomum zeylanicum Sri Wijaya (SW) and Sri Gemunu (SG) accessions and commercially available C. zeylanicum (CC) were subjected to fungal pretreatment and extracted with pressured water (PWE, 0·098 MPa). Thirteen fungal species were isolated and the substrate utilization ability of the species was tested using cellulose, pectin and lignin (indirectly). Total phenolic content (TPC, Folin–Ciocalteu method), proanthocyanidin content (PC, vanillin method) and α-amylase and α-glucosidase inhibitory potential of the extracts were evaluated. The anti-diabetic drug, Acarbose was used as the positive control. Trichoderma harzianum (MH298760) showed the highest cell lysis ability and hence was used for the microbial pretreatment process. Extracts of SW treated with T. harzianum species (Pre-SW) gave the highest percentage yield (4·08% ± 0·15%), significantly potent inhibition (P < 0·05) of α-amylase and α-glucosidase activities (IC50 57 ± 8 and 36 ± 8 μg ml−1 respectively), TPC (2·24 ± 0·02 mg gallic acid equivalent g−1), and PC (48·2 ± 0·4 mg of catechin equivalent g−1) compared to Pre-SG, Pre-CC and nontreated samples. Trichoderma harzianum treatment can enhance the hypoglycaemic properties, PC and TPC of Cinnamon extracts and provide new insights into the recovery of phytochemicals.  相似文献

The use of plants in treatments has been as old as humanity and it has preserved its popularity for centuries til now because of their availability, affordability and safeness. However, despite their widespread use, safety and quality issues have been major concerns in the world due to industrial- and anthropogenic-based heavy metal contamination risks. Thus, this study was attempted to analyze the heavy metal levels and mineral nutrient status of widely used medicinal plants in Turkey to have insights about their health implications on humans. The plant concentrations of B, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, Pb and Zn were analyzed by ICP-OES in the leaves of 44 medical plants purchased from herbal markets of three different districts of Istanbul/Turkey. The measured lowest to highest concentrations were 0.065–79.749 mg kg−1 B, 921.802–12,854.410 mg kg−1 Ca, 0.020–0.558 mg kg−1 Cd, 0.015–4.978 mg kg−1 Cr, 0.042–8.489 mg kg−1 Cu, 34.356–858.446 mg kg−1 Fe, 791.323–15,569.349 mg kg−1 K, 102.236–2837.836 mg kg−1 Mg, 4.915–91.519 mg kg−1 Mn, 10.224–3213.703 mg kg−1 Na, 0.001–5.589 mg kg−1 Ni, 0.003–3.636 mg kg−1 Pb and 2.601–36.102 mg kg−1 Zn. Those levels in plants were in acceptable limits though some elements in some plants have high limits which were not harmful. Variations (above acceptable limits) in element concentrations also indicated that these plants could be contaminated with other metals and that genetic variations may influence accumulation of these elements at different contents. Overall, analyzed medicinal plants are expected not to pose any serious threat to human health.

In this environmental-sample based study, rapid microbial-mediated degradation of 2,4,6-trinitrotoluene (TNT) contaminated soils is demonstrated by a novel strain, Achromobacter spanius STE 11. Complete removal of 100 mg L−1 TNT is achieved within only 20 h under aerobic conditions by the isolate. In this bio-conversion process, TNT is transformed to 2,4-dinitrotoluene (7 mg L−1), 2,6-dinitrotoluene (3 mg L−1), 4-aminodinitrotoluene (49 mg L−1) and 2-aminodinitrotoluene (16 mg L−1) as the key metabolites. A. spanius STE 11 has the ability to denitrate TNT in aerobic conditions as suggested by the dinitrotoluene and NO3 productions during the growth period. Elemental analysis results indicate that 24.77 mg L−1 nitrogen from TNT was accumulated in the cell biomass, showing that STE 11 can use TNT as its sole nitrogen source. TNT degradation was observed between pH 4.0–8.0 and 4–43 °C; however, the most efficient degradation was at pH 6.0–7.0 and 30 °C.  相似文献

Abstract Climatic conditions should not hinder nutrient release from decomposing leaf‐litter (mineralization) in the humid tropics, even though many tropical forests experience drought lasting from several weeks to months. We used a dry‐season irrigation experiment to examine the effect of seasonal drought on nutrient concentrations in leaf‐fall and in decomposing leaf‐litter. In the experiment, soil in two 2.25‐ha plots of old‐growth lowland moist forest on Barro Colorado Island, Republic of Panama, was watered to maintain soil water potential at or above field capacity throughout the 4‐month dry season. Wet‐season leaf‐fall had greater concentrations of nitrogen (N, 13.5 mg g?1) and calcium (Ca, 15.6 mg g?1) and lower concentrations of sulfur (S, 2.51 mg g?1) and potassium (K, 3.03 mg g?1) than dry‐season leaf‐fall (N = 11.6 mg g?1, Ca = 13.6 mg g?1, S = 2.98 mg g?1, K = 5.70 mg g?1). Irrigation did not affect nutrient concentrations or nutrient return from forest trees to the forest floor annually (N = 18 g m?2, phosphorus (P) = 1.06 g m?2, S = 3.5 g m?2, Ca = 18.9 g m?2, magnesium = 6.5 g m?2, K = 5.7 g m?2). Nutrient mineralization rates were much greater during the wet season than the dry season, except for K, which did not vary seasonally. Nutrient residence times in forest‐floor material were longer in control plots than in irrigated plots, with values approximately equal to that for organic matter (210 in control plots vs 160 in irrigated plots). Calcium had the longest residence time. Forest‐floor material collected at the transition between seasons and incubated with or without leaching in the laboratory did not display large pulses in nutrient availability. Rather, microorganisms immobilized nutrients primarily during the wet season, unlike observations in tropical forests with longer dry seasons. Large amounts of P moved among different pools in forest‐floor material, apparently mediated by microorganisms. Arylsulfatase and phosphatase enzymes, which mineralize organically bound nutrients, had high activity throughout the dry season. Low soil moisture levels do not hinder nutrient cycling in this moist lowland forest.  相似文献

Effects of heavy metals on aerobic denitrification have been poorly understood compared with their impacts on anaerobic denitrification. This paper presented effects of four heavy metals (Cd(II), Cu(II), Ni(II), and Zn(II)) on aerobic denitrification by a novel aerobic denitrifying strain Pseudomonas stutzeri PCN-1. Results indicated that aerobic denitrifying activity decreased with increasing heavy metal concentrations due to their corresponding inhibition on the denitrifying gene expression characterized by a time lapse between the expression of the nosZ gene and that of the cnorB gene by PCN-1, which led to lower nitrate removal rate (1.67∼6.67 mg L−1 h−1), higher nitrite accumulation (47.3∼99.8 mg L−1), and higher N2O emission ratios (5∼283 mg L−1/mg L−1). Specially, promotion of the nosZ gene expression by increasing Cu(II) concentrations (0∼0.05 mg L−1) was found, and the absence of Cu resulted in massive N2O emission due to poor synthesis of N2O reductase. The inhibition effect for both aerobic denitrifying activity and denitrifying gene expression was as follows from strongest to least: Cd(II) (0.5∼2.5 mg L−1) > Cu(II) (0.5∼5 mg L−1) > Ni(II) (2∼10 mg L−1) > Zn(II) (25∼50 mg L−1). Furthermore, sensitivity of denitrifying gene to heavy metals was similar in order of nosZ > nirS ≈ cnorB > napA. This study is of significance in understanding the potential application of aerobic denitrifying bacteria in practical wastewater treatment.

Phellodendron chinense Schneid is an important Chinese herb with berberine and phellodendrine in stems and leaves, but with little information available on in vitro culture of this species. Disinfection of explants in 75% alcohol for 45 s, sterilization in 0.1% HgCl2 for 20 min, and submersion in 1.0 mol L−1 gibberellin3 (GA3) solution for 24 h was the optimal condition for seed germination. Murashige and Skoog’s (MS) medium supplemented with 2.0 mg L−1 6-benzylaminopurine (6-BA) in combination with 1.5 mg L−1 1-naphthylacetic acid (NAA) was optimal for callus induction. MS medium supplemented with 2.0 mg L−1 6-BA was the appropriate medium for induction of adventitious shoots, and 1/2MS medium supplemented with 2.0 mg L−1 indole-3-butytric acid (IBA) and 0.5% active carbon was the optimal medium for root induction. The 15-d survival rate of regenerated plantlets after transplanting to basins containing perlite and peat moss (1:4) was greater than 80%, and the berberine and phellodendrine accumulation was lower in callus compared with regenerated plantlets. The establishment of highly efficient regeneration system provides technical support for genetic breeding of Phellodendron chinense Schneid.

New biosourced chiral cross‐linkers were reported for the first time in the synthesis of methyltestosterone (MT) chiral molecularly imprinted polymers (cMIPs). Isosorbide and isomannide, known as 1,4:3,6‐dianhydrohexitols, were selected as starting diols. The cMIPs were synthesized following a noncovalent approach via thermal radical polymerization and monitored by Raman spectroscopy. These cross‐linkers were fully characterized by 1H and 13C nuclear magnetic resonance (NMR) spectroscopy and high‐resolution mass spectrometry. The cross‐polarization magic angle spinning 13C NMR, Fourier transform infrared spectroscopy, scanning electron microscopy, and specific surface areas following the Brunauer‐Emmett‐Teller (BET) method were used to characterize the cMIPs. The effect of stereochemistry of cross‐linkers on the reactivity of polymerization, morphology, and adsorption‐recognition properties of the MIP was evaluated. The results showed that the cMIP exhibited an obvious improvement in terms of rebinding capacity for MT as compared with the nonimprinted polymer (NIP). The highest binding capacity was observed for cMIP‐Is (27.298 mg g−1) for high concentrations (500 mg L−1). However, the isomannide homologue cMIP‐Im showed higher recovery—up to 65% and capacity for low concentrations (15 mg L−1). The experimental data were properly fitted by the Freundlich adsorption isothermal model.  相似文献

The activities of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), as well as photosynthetic pigment contents and free malondialdehyde (MDA), were determined in senescent batch cultures of Tetraselmis gracilis (Kylin) Butcher, under a cyclic light regime. A 2.6-fold increase in SOD activity (from 53 to 137 U mg−1 protein) was observed in the light phase, contrasting with a 9-fold increase in CAT (from 1 to 9 μmol H2O2 min−1 mg−1 protein) and a 1.7-fold increase in APX (from 3 to 5 μmol ascorbate min−1 mg−1 protein) activities, both enzymes peaking in the dark phase. The β-carotene and lycopene content did not vary significantly with the light–dark cycle. The Chl a, Chl b, lutein, zeaxanthin, violaxanthin and neoxanthin pigments exhibited the highest values in the first half (3–6 h) of the light phase, followed by a declining trend and a plateau or a slight increase 3 h from the beginning of the dark phase onwards. The highest values for prasinoxanthin were observed in the second half of the dark phase and the first half of the light phase. None of the pigments displayed any discernible cyclic trend. The possibility of the xanthophyll cycle occurring during senescence is discussed in light of the high value (∼0.9) obtained for the zeaxanthin/(zeaxanthin + violaxanthin) ratio. The free MDA content was enhanced during the experimental period, which may be an indicator of oxidative stress in aging cell cultures. Our results indicated the occurrence of an imbalance between the production of reactive oxygen species and the antioxidant defense in stationary T. gracilis cells.  相似文献

This study investigated the potential functions of Pleurotus florida (an edible mushroom) in the biodegradation of gas oil at concentrations of 0 (control), 2.5, 5, and 10% (V: V) for 30 days. The gas oil increased dry weight and protein concentration in all treatments (by an average of 19.5 and 108%, respectively). Moreover, the pH, surface tension (ST), and interfacial tension (IFT) were reduced by the mushroom supplementation. The lowest surface tension (31.9 mN m−1) and the highest biosurfactant production belonged to the 10% gas oil treatment (0.845 ± 0.03 mg mL−1). The results demonstrated that the adsorption isotherm agreed well with the Langmuir isotherm. The maximum Langmuir adsorption capacity was calculated at 0.743 mg g−1 wet biomass of P. florida. The fungal supplementation efficiently remedied the total petroleum hydrocarbons (TPHs) by an average of 55% after 30 days. Gas chromatography (GC) analysis revealed that P. florida effectively detoxified C13–C28 hydrocarbons, Pristane, and Phytane, implying its high mycoremediation function. The toxicity test showed that mycoremediation increased the germination by an average of 35.82% ± 8.89 after 30 days. Laccase activity increased significantly with increasing gas oil concentration in the treatments. The maximum laccase activity was obtained in the 10% gas oil treatment (142.25 ± 0.72 U L−1). The presence of pollutants was also associated with induction in the tyrosinase activity when compared to the control. These results underline the high mycoremediation capacity of P. florida through the involvement of biosurfactants, laccase, and tyrosinase.  相似文献
