P64k is a Neisseria meningitidis high molecular weight protein present in meningococcal vaccine preparations. The lpdA gene, codifying for this protein, was cloned in Escherichia coli and the P64k protein was expressed in Escherichia coli K12 W3110 under the control of the tryptophan promoter. The recombinant bacteria were grown in batch or fed-batch cultures. P64k was expressed as an intracellular soluble form at about 40% of the total cellular protein. A final productivity of 215 mg l–1 h–1 and 11 g cell dry wt l–1 were obtained when the fed-batch culture conditions were optimised, compared to 30% of total protein, and a productivity of 76 mg l–1 h–1 and 5.1 g cell dry wt l–1 in batch cultivation.  相似文献

A study was made on the mass cultivation of Nannochloropsis sp. in newly designed annular reactors operated under natural, artificial or combined illumination. The annular reactor consists of two 2-m-high Plexiglas cylinders of different diameter placed vertically one inside the other so as to form an annular culture chamber. Artificial illumination is supplied by lamps placed inside the inner cylinder. Two annular reactors of different diameter (50 and 91 cm), light path (4.5 and 3.0 cm) and illuminated surface area (5.3 and 9.3 m2) were experimented with. The effect of two different artificial light sources (fluorescent tubes and metal halide lamps) on culture productivity was investigated in both systems. The highest productivity on a per reactor basis (about 34 g (d. wt) reactor–1 24 h–1) was achieved with the larger reactor illuminated by a 400-W metal halide lamp. From February to May a 91-cm reactor illuminated only with natural light was operated in parallel with a 91-cm reactor subjected to combined illumination. Under natural illumination productivity increased from 16.6 g (d. wt) reactor–1 d–1 in February to 34.1 g (d. wt) reactor–1 d–1 in May. Under combined illumination productivity was 41.3 g (d. wt) reactor–1 d–1 in February and increased up to 48.3 g (d. wt) reactor–1 d–1 in May. Although the culture exposed to combined illumination always attained higher yields, the productivity gap between the two cultures decreased gradually along the season as solar radiation and minimum night temperatures increased. A 1200-L plant made of ten 50-cm annular reactors was set up and operated for two years with combined illumination yielding an average of 270 g of dry Nannochloropsis sp. biomass per day. More than 2000 L of concentrate suspension (50 g (d. wt) L–1) of Nannochloropsis sp. were produced and successfully used by fish hatcheries as live feed for rotifers and for rearing seabream larvae with the green-water technique. This study indicates that the annular reactor can be profitably used for long-term cultivation of Nannochloropsis in temperate climates. Besides reliability and ease of operation, the main advantage of the system is that it can be used under natural illumination, yet artificial light can be also supplied to maintain high productivity levels in winter or on cloudy days.  相似文献

The transformed root culture of Polygonum tinctorium Lour. was established by infecting leaf explants with Agrobacterium rhizogenes A4. These cultures were examined for their growth and indigo content under various culture conditions. Among the four different culture media tested, SH medium showed the highest yield for root growth (28 mg dry wt/30 ml) and indigo production (152 g/dry wt). In SH medium, 30 g sucrose l–1, 2500 mg KNO3 l–1, 300 mg NH4H2PO4 l–1 were the best conditions for indigo production at pH 5.7. The production of indigo in hairy roots slightly increased with the addition of 200 mg chitosan l–1 (186 g/dry wt) and 20 U pectinase l–1 (181 g/dry wt).  相似文献

Saffron callus was grown in a two-stage culture on B5 medium supplemented with casein hydrolysate (300 mg l–1) at 22 °C in dark with naphthalene acetic acid (2 mg l–1) and 6-benzyladenine (1 mg l–1) to give maximum biomass (16 g dry wt l–1), and with indole 3-acetic acid (2 mg l–1) and 6-benzyladenine (0.5 mg l–1) for crocin formation. The maximum crocin production (0.43 g l–1) was achieved by this two-stage culture method, which was three times that by a one-stage method.  相似文献

Excised root cultures of Gloriosa superba reached 7.5 g dry wt l–1 and accumulated 240±40 g colchicine g–1 cell dry wt after 4 weeks growth. While all precursors (except trans-cinnamic acid) enhanced colchicine content of root cultures without adversely affecting root growth, treatment with p-coumaric acid + tyramine (each at 20 mg l–1) increased colchicine content to 1.9 mg g–1 cell dry wt.  相似文献

Hairy roots of Rauvolfia micrantha were induced from hypocotyl explants of 2–3 weeks old aseptic seedlings using Agrobacterium rhizogenes ATCC 15834. Hairy roots grown in half-strength Murashige & Skoog (MS) medium with 0.2 mg indole 3-butyric acid l–1 and 0.1 mg -naphthaleneacetic acid l–1 produced more ajmaline (0.01 mg g–1 dry wt) and ajmalicine (0.006 mg g–1 dry wt) than roots grown in auxin-free medium. Ajmaline (0.003 mg g–1 dry wt) and ajmalicine (0.0007 mg g–1 dry wt) were also produced in normal root cultures. This is the first report of production of ajmaline and ajmalicine in hairy root cultures of Rauvolfia micrantha.  相似文献

When, on the 15th day of growth, an elicitor from Fusarium solani was added at 40 mg l–1 to Cistanche deserticola cell suspension cultures, the contents of echinacoside, acteoside and total phenylethanoid glycosides (PeGs) in cultured cells all increased over the next 27 d by over 100% to 15 mg g–1 dry wt, 9 mg g–1 dry wt and 57 mg g–1 dry wt, respectively. Brevibacterium linens forms hydrolytic enzymes which can be used to accelerate the ripening of cheese without causing bitterness. B. linens ATCC 9172 was grown to a high cell density (50 g dry wt l–1 after 60 h) in a mineral medium containing lactic acid, soy-peptone and ammonium sulphate by applying a continuous feed of nutrients. The maximal activities of l-leucine aminopeptidase and cell-associated proteinase were 286 U l–1 and 202 U l–1, respectively. The cell-associated lipolytic activity exhibited a strong and sudden increase at 46 h, resulting in a maximum of 9.5 U g–1 dry wt; thus the volumetric productivity of proteolytic and lipolytic activity was 4220 U l–1 h–1 and 7.3 U l–1 h–1, respectively.  相似文献

Trichoderma reesei Rut-C30 is a highly derepressed mutant which synthesised active cellulases in culture media containing glucose and lactose as the only carbon sources. The maximum biomass, filter paper and specific filter paper activities for cell growth on 20 g glucose l–1 were 20 g dry cell wt l–1, 1.9 FPU ml–1 and 4.8 FPU mg–1 protein respectively, while on 40 g glucose l–1 were 25 g dry cell wt l–1, 4.5 FPU ml–1 and 6.2 FPU mg–1 protein, respectively. This strain had a higher specific filter paper activity (6.2 FPU mg–1 protein) than was produced by other T. reesei mutants (3.6 FPU mg–1 protein).  相似文献

Casein hydrolysate at 2.0 g l–1 significantly enhanced forskolin content (2.3 mg g–1 cell dry wt) in a rhizogenic tumourous line, GCO-RCH-2 of Coleus forskohlii. In rooty teratoma line, RC-ST-2/4, forskolin content enhanced to 1.7 mg g–1 cell dry wt in presence of 2.5 g l–1 casein hydrolysate. Unlike untransformed calli and rhizogenic/root cultures, all the forskolin yielding transformed cultures of C. forskohlii have been maintained for over 5 years.  相似文献

A unicellular marine picoplankton, Nannochloropsis sp., was grown under CO2-enriched photoautotrophic or/and acetate-added mixotrophic conditions. Photoautotrophic conditions with enriched CO2 of 2800 l CO2 l–1 and aeration gave the highest biomass yield (634 mg dry wt l–1), the highest total lipid content (9% of dry wt), total fatty acids (64 mg g–1 dry wt), polyunsaturated fatty acids (35% total fatty acids) and eicosapentaenoic acid (EPA, 20:53) (16 mg g–1 dry wt or 25% of total fatty acids). Mixotrophic cultures gave a greater protein content but less carbohydrates. Adding sodium acetate (2 mM) decreased the amounts of the total fatty acids and EPA. Elevation of CO2 in photoautotrophic culture thus enhances growth and raises the production of EPA in Nannochloropsis sp.  相似文献

In Taxus cuspidata callus, vanadyl sulfate (10 mg l–1) induced a high (146 g g–1 dry wt) production of 10-deacetylbaccatin III in comparison to 7 g g–1 dry wt of the control. The content of paclitaxel in this species increased from 16 g g–1 to 74 g g–1 dry wt when 20 mg phenylalanine l–1 was used. In T. media, p-aminobenzoic acid induced the highest content of 10-deacetylbaccatin III (481 g g–1 dry wt) versus 181 g g–1 in the control. Paclitaxel increased from 89 to 139 g g–1 dry wt after adding chitosan (20 mg l–1) to the cultures.  相似文献

To investigate the production potential of eicosapentaenoic acid (EPA) by the diatom Nitzschia laevis, the growth characteristics and fatty acid composition of the cells were studied under photoautotrophic, mixotrophic and heterotrophic conditions of growth. The specific growth rate and maximum biomass concentration were respectively 0.466 d–1 and 2.27 g l–1 for mixotrophic culture, 0.344 d–1 and 2.04 g l–1 for heterotrophic culture, and 0.167 d–1 and 0.5 g l–1 for photoautotrophic culture, respectively. As for EPA production, the yield and productivity were respectively 52.32 mg l–1 and 10.46 mg l–1 d for mixotrophic culture, 35.08 mg l–1 and 6.37 mg l–1 d for heterotrophic culture, and 6.78 mg l–1 and 3.39 mg l–1 d for photoautotrophic culture, respectively. Results suggest that mixotrophic culture is the most suitable growth mode for the production of EPA by the diatom Nitzschia laevis. The results are useful for the development of a cost-effective fermentation process for EPA production by Nitzschia laevis.  相似文献

The optimal growth of Cistanche deserticola callus and formation of phenylethanoid glycosides (PeG) was at 25°C with light irradiation intensity of 24 mol m–2 s–1 on solidified B5 media supplemented with 0.5 mg 6-benzylaminopurine l–1, 10 mg gibberellin l–1, 800 mg casein hydrolysate l–1 and 20 g sucrose l–1. After 30 d culture, the biomass reached 15.5 g dry wt callus l–1 medium and its PEG content was 10.7% (w/w). The PeG content was 42%–127% higher than those in explants.  相似文献

The sedative triterpene, galphimine B (1), was detected in cell suspension-batch cultures of Galphimia glauca. The effect of inoculum size, growth regulators and different concentrations of sucrose, nitrates and phosphates was evaluated. A two-stage batch process for biomass production and accumulation of compound 1 was established. Major cellular growth (15 g l–1 dry wt) was obtained in the first stage with naphthaleneacetic acid (2 mg l–1) + kinetin (2 mg l–1). Adding 4 mg 2,4-dichlorophenoxyacetic l–1 acid in the second stage resulted in the highest accumulation of 1 (0.21 mg g–1 dry wt) which was 36% higher with respect to calluses and comparable to that obtained from wild plants.  相似文献

Aspergillus niger was explored, for the first time, for the production of 2-phenylethanol (a rose-like aroma) using L-phenylalanine as precursor. Among the strains screened, A. niger CMICC 298302 was shown to produce, in a culture medium containing 6 g L-phenylalanine l–1 and 60 g glucose l–1, 1375 mg 2-phenylethanol l–1 with a productivity of 153 mg l–1 day–1 and a molar yield of 74%. 2-Phenylethanol concentrations of 1 to 2 g l–1 led to a two-fold and ten-fold decrease, respectively, in the mycelial radial growth rate. However, 2-phenylethanol was synthesized as the sole aromatic product and accumulated in the culture broth.  相似文献

A fed-batch fermentation of Lactobacillus lactis to produce l-lactic acid was developed in which the residual glucose concentration in the culture was used to control a continuous feeding strategy. Pseudomonas oleovorans was cultivated to produce medium chain length polyhydroxyalkanoates (MCL-PHAs) from octanoic acid and ammonium nitrate as carbon and nitrogen source, respectively, by a pH-stat fed-batch culture technique. The octanoate in the culture broth was maintained below 4 g l–1 by feeding the mixture of octanoic acid and ammonium nitrate when the culture pH rose above 7.1. The final cell concentrations of 63, 55 and 9.5 g l–1, PHA contents of 62, 75 and 67% of dry cell wt, and productivities of 1, 0.63 and 0.16 g l–1 h–1 were obtained when the C/N ratios in the feed were 10, 20 and 100 g octanoic acid g–1 ammonium nitrate, respectively.  相似文献

A high cell density cultivation method was developed to produce recombinant PvRII, a malaria vaccine candidate, in E. coli for use in vaccine studies. Cells were grown in completely defined media and glucose was fed to achieve a specific growth rate of 0.12 h–1 until cells reached 55 g dry wt l–1. Culture was then induced with 1 mm IPTG and cells were further grown for 4 h to reach 85 g dry wt l–1 at 0.1 h–1. Recombinant PvRII was purified from inclusion bodies under denaturing conditions using metal affinity chromatography which yielded 10 mg PvRII g–1 dry wt. Hairy roots of Astragalus membranaceus were grown in bioreactors up to 30 l for 20 d. Cultures from a 30 l airlift bioreactor gave 11.5 g l dry wt with 1.4 mg g–1 astragaloside IV, similar to cultures from 250 ml and 1 l flasks, but greater than yields from a 10 l bioreactor (dry wt 9.4 g l–1, astragaloside IV 0.9 mg g–1). Polysaccharide yields were similar amongst the different bioreactors (range 25–32 mg g–1). The active constituent content of the cells approached that of plant extracts, indicating that large scale hairy root cultures of A. membranaceus has the potential to provide an alternative to plant crops without compromising yield or pharmacological potential.  相似文献
