Summary Detached pistils of the clonal variety, Lilium longiflorum Aral No. 5, were submerged before pollination in 50°C water for 0, 1, 2, 3, 4, 5, 6 or 7 min and then immediately compatibly and incompatibly pollinated. Incompatibility, as indicated by pollen tube length after 48 h at 23.5°C, was eliminated by a 1–2 min submersion while compatibility was removed by a 4–5 min one. The window of incubation temperatures at which incompatible and compatible pollen tubes are clearly differentiated occurred between 15 and 30°C.  相似文献

Temperature dependencies were established for the egg-to-mummy and mummy-to-adult phases, for mummy mortality, and for parasitism of Aphidius ervi Haliday, Aphidius rhopalosiphi De Stefani-Perez, and Praon volucre (Haliday) (Hymenoptera, Aphidiidae), three parasitoids of Sitobion avenae (Fabricius) (Homoptera, Aphididae), at 8°C, 12°C, 16°C, 20°C, and 25°C on winter wheat (cv. Haven). A physiological model described temperature-dependent development over the full temperature range, whereas a linear model was fitted for data above 8°C and used to estimate the lower temperature thresholds and day-degrees (° D) required for development. The thresholds for A. ervi were 2.2°C for egg-mummy development and 6.6°C for mummy-adult development, those for A. rhopalosiphi were 4.5°C and 7.2°C, and those for P. volucre were 3.8°C and 5.5°C. The time to develop into mummies and adults differed significantly between the three species: A. ervi development into mummies required an average of 159 ° D, while development into adults took an average of 73 ° D. The corresponding average times required for A. rhopalosiphi and P. volucre to develop mummies were 124° D and 126° D, while their development into adults required an average of 70° D and 150° D, respectively. Mummy mortality was 25–35% at 8°C and less at the higher temperatures tested, but began to increase again at 25°C, showing a quadratic relationship between mortality and temperature. Parasitization was very low or, in the case of P. volucre, absent up to 12°C and thereafter increased with increasing temperature. The relationship between parasitization, recorded as percent aphids mummified, and temperature was linear at the temperatures tested and depended on species. A. ervisuperparasitized 11.1% aphids at 20°C and 16.6% aphids at 25°C, whereas superparasitism was low in A. rhopalosiphi and absent in P. volucre. From 16°C to 25°C the P. volucre sex ratio increased. For A. ervi and A. rhopalosiphi there was no trend with temperature, but at 20°C and 25°C it was close to even. Field data for 1996 and 1997 allowed for a comparison of actual and expected emergence of overwintering mummies. In both years, parasitoids were predicted to have emerged from overwintering mummies well in advance of the onset of aphid infestation, and more than a month earlier than the first parasitized aphids were found in winter wheat. Observations from trap plants in other crops supported the predictions of the models. Other factors that can affect biological control by cereal aphid parasitoids are discussed.  相似文献

Laboratory studies were performed to assess the importance of temperature on sporulation and infection by the aphid-pathogenic fungus Pandora neoaphidis (Remaudière and Hennebert) Humber. Numbers of primary conidia discharged from mycelium formulated as alginate granules and unformulated mycelial mats were assessed, as well as infection of the potato aphid, Macrosiphum euphorbiae (Thomas) (Homoptera, Hemiptera, Aphididae), using culture plugs as inoculum sources. Sporulation from experiments at constant temperatures indicated the optimum temperature range was 10–20°C for both mycelial preparations and there was no or very little sporulation at 30°C. Infection of aphids kept at 15°C was 34–50%, while infection at 25°C was 11–44%. At 20°C, 77–79% of aphids were infected. Under fluctuating temperature cycles, conidia numbers did not differ when mycelial preparations were maintained at 18–25°C compared with 18–20°C, but fewer conidia were recorded when preparations were exposed continuously to 18–30°C. Infections of inoculated aphids kept for varying numbers of days at 18–25°C varied between 24–47%, but only 3–32% of aphids were infected when exposed to a cycle of 18–30°C for various times. Unformulated mycelial mats of P. neoaphidis appear to be superior to forumlated alginate granules for use in experimental greenhouse and field trials, since temperature stability is similar for both materials but mycelial mats are much easier to produce.  相似文献

A hetero-oligomeric glucose dehydrogenase (GDH) from a moderate thermophilic bacterium, SM4 was cross-linked with glutaraldehyde (GA) and it now showed only one optimum temperature for reaction at around 65°C, which approximately follows the Arrhenius equation. The native enzyme had shown optima at both 45°C and 75°C. In addition to the alteration of the optimum temperature for reaction, GA cross-linked GDH retained more than 90% of its initial activity even after 30 min of incubation at 65°C.  相似文献

I examined the effect of temperature and light on ecosystem composition was examined in a two factorial design using microcosms set up from natural rockpool communities. Furthermore I tested if the effect of temperature on different ecosystem components was dependent on the initial community composition by using communities from seven different rockpools that differed considerably in standing stocks of phytoplankton, zooplankton, zooplankton species composition, sediment mass and nutrient concentrations. Increased light caused phytoplankton biomass to decrease while zooplankton biomass and sediment dry weight was positively correlated to increased light levels. The effect of temperature on phytoplankon was largely determined by community type. Zooplankton biomass decreased with increasing temperature between 10°C and 25°C and this trend was not significantly different between different community types. A negative effect on zooplankton biomass was found at 7°C in one community. I propose, that the stronger temperature sensitivity of metabolical cost for herbivorous organisms compared to algae productivity might explain the decrease in zooplankton biomass at high temperatures. I discuss how edibility of algae and grazer characteristics may influence the response of ecosystem composition to temperature.  相似文献

Summary Hordeum spontaneum C. Koch emend. Bacht. varieties have been both intercrossed and crossed with two cultivated barley varieties ofH. vulgare (L.) emend Vav. et Bacht. with a view of eliciting the nature of inheriting the spikelet-pedicel of the lateral spikelets and the shape of their apex in the said wildgrowing barley. The investigations of F1 and F2 showed the inheritance of the spikelet-pedicel to have a dominating nature and to segregate in F2 in conformity with the Mendelian monohybrid type. In the second case the forms with shorter awn-like formations, or their rudiments, were dominating.As a result ofH. spontaneum x H. vulgare hybridization along with already known forms, new formations were received, they have been conditionally named by the author:sessiliproskowetzii, proskowfertillum, ischnofertillum, and pallipodum.
Zusammenfassung Im Rahmen größerer Untersuchungen über die Abstammung und Phylogenie der Gerste wurden mehrere Varietäten vonHordeum spontaneum C. Koch emend. Bacht. sowohl untereinander als auch mit zwei Varietäten der Kulturgerste,H. vulgare (L.) emend. Vav. et Bacht., gekreuzt. Es sollte geklärt werden, wie bei den genannten Wildgersten das Stielchen (pedicel) der Seitenährchen sowie die Ausbildung des Apex der Seitenährchen (d. h. ihre Begrannung) vererbt werden. Die Untersuchung der F1 und F2 zeigte, daß das Stielchen (gegenüber ungestielten Seitenährchen) dominant und gemäß einer monohybriden Mendelspaltung vererbt wird. Bezüglich der Ausbildung des Apex der Seitenährchen ergab sich im allgemeinen Dominanz der kürzeren oder rudimentären Grannen gegenüber längeren Grannen.Im Ergebnis der Hybridisation zwischenH. spontaneum undH. vulgare wurden, neben bereits bekannten, verschiedene neue Formen gefunden, die vom Autor vorläufig wie folgt benannt werden:sessiliproskowetzii, proskowfertillum, ischnofertillum, pallipodum.Die Ergebnisse werden im Zusammenhang mit Fragen der Abstammung der Kulturgerste diskutiert.

32 West African dwarf goats were exposed in respiration chambers to temperature treatments of 20, 25, 30, 35, 35, 35, 30, 25, 20°C. Each treatment lasted three days. 16 goats were kept in individual pens (I); the others in two group pens of eight animals each (G). During each treatment, heat production and activity were recorded continuously over 48 hours. In addition, feed and water intake, rectal temperature, skin temperature and respiratory rate were measured during each treatment.Compared to 20°C, at 35°C rectal temperature increased from 39.0°C to 39.9°C, respiratory rate from 30 to 260 times. min–1 and skin temperature from 37.1°C to 39.5°C. Hay intake decreased by 40%; concentrates (30 g. kg–0.75. d–1) were always completely consumed. Heat production was higher for the G animals at 20°C and higher for the I animals at 35°C. These differences in heat production between the two groups were reflected in differences in rectal and skin temperature and in respiratory rate but only very slightly in differences in hay intake.Tissue insulation was 0.014 K. m2. W–1 at 30°C and 35°C and 0.022 K. m2. W–1 at 20°C.It is concluded that the reactions of these dwarf goats to high ambient temperatures are not different in principle from those of other domestic ruminants and that they do not exhibit a specific suitability or unsuitability for ambient temperatures as prevailing in West Africa.  相似文献

Thirty-two West African dwarf goats were exposed to temperature treatments: 20, 25, 30, 35, 35, 35, 30, 25, 20°C each lasting three days.Sixteen goats were kept in individual pens (I), the others in two group pens (G). Heat production (HP) and activity were recorded during 48 hours in each temperature treatment.Mean HP and ME intake were similar for G and I animals, but I animals had lower values than G animals at low chamber temperatures and higher values than G animals at high temperatures. Upper critical temperature was between 25°C and 30°C under an increasing T and between 30 and 35°C under a decreasing T. Adaptation of heat production and ME intake to a change in temperature of 5°C required at least six and possibly more than nine days.Diurnal variation in HP was large, up to 44% between extremes. Summary The temperature dependence of the oxygen isotope fractionation factor during respiration has been examined for two different microorganisms, namelyTorulopsis utilis andEscherichia coli K12 representing a yeast and a bacterium, respectively. The investigation covered a temperature range of 18° C, that is from 16° C to 34° C forT. utilis and from 19° C to 37° C forE. coli K12. Development, survival and reproduction of Euseius finlandicus Oudemans were studied at seven constant temperatures (15, 20, 25, 27, 30, 32 and 34°C) in the laboratory. Within the temperature range tested, developmental period from egg to adult varied from 148 to 360.5h and 133.7 to 336.5h for females and males, respectively. The lower thermal threshold for immature development for females and males was 8.9 and 6.4°C, respectively. Survival during immature development exceeded 90% at all the temperatures from 15 to 32°C, but at 34°C an abrupt decline was recorded. Female longevity decreased gradually from 82.7d at 15°C to 12.2 d at 34°C. The mean generation time ranged from 44.3d at 15°C to 15.9d at 32°C. The highest r m value (0.2817) was obtained at 30°C and the lowest at 15°C (0.0976). Temperatures above 30°C had an adverse effect on population increase.  相似文献

Summary 1. Rates of growth (length increase of stolons) and of asexual reproduction (increase in number of polyps) were determined in secondaryClava multicornis colonies of a clone exposed to 12 different combinations of water temperature and salinity (12°, 17°, 22° C; 16 , 24 , 32 , 40 S). Sexual reproduction (via gonophores) has been observed only at 12° and 17° C; temperature and salinity ranges are narrower for sexual than for asexual reproduction.2. The data obtained are insufficient for a detailed analysis; they provide, however, interesting insights into the variability of growth and reproduction ofC. multicornis caused by different intensities of temperature and salinity.3. It appears that temperature requirements for maximum colony increase are reduced as the colony grows older.4. One feeding period per 24 hours seems insufficient for maximum growth and reproduction at the higher temperature levels, especially at 22° C.5. The different degrees of environmental stress endured during the initial period of transfer into the test combinations of temperature and salinity have affected the resulting colony size at least up to an age of 39 days. More appropriate criteria for assessment of rates of growth and reproduction are therefore the doubling times (number of days within which stolon length and polyp numbers taken 20 days after initiation of experiments have doubled).6. On the basis of doubling time values, increase in stolon length is progressively reduced with increasing water temperature (12°, 17°, 22° C). At 12° and 17° C stolons grow fastest in 32 , followed by 24 , 16 and 40 S; at 22° C stolon growth rates are identical in 32 and 24 S.7. Doubling times of polyp numbers per colony show a less obvious trend. In 56-day-old colonies, however, stolon length and polyp number are modified to similar degrees by the various temperatures and salinities offered. The sequence of temperatures causing fastest increase in polyp number is 12°>17°>22° C; the respective sequence of salinities reads: 24 , 32 , 16 , 40 S.8. Stolon length and polyp number per colony increase exponentially; most curves obtained exhibit undulations indicating endogenous growth rhythms.9. During the initial period of transfer into the final test media, asexual reproduction via budding seems to have been stimulated by a reduction in salinity.10. The doubling times obtained forC. multicornis are considerably longer than those found forCordylophora caspia and indicate that our culture conditions may have been suboptimal.

Kurzfassung Einzelpolypen eines Klons vonC. multicornis Forskål wurden schrittweise in 12 verschiedene Temperatur-Salzgehalts-Kombinationen überführt und — während sie zu neuen Kolonien heranwuchsen — das Längenwachstum ihrer Stolonen, die Geschwindigkeit ihrer asexuellen Vermehrung durch Knospung neuer Hydranthen sowie die Gonophorenausbildung (sexuelle Fortpflanzung) registriert. Die erhaltenen Daten sind unzureichend für eine detaillierte Analyse, gewähren jedoch interessante Einblicke in die Bedeutung der verschiedenen Temperatur- und Salzgehaltsbedingungen für Wachstum und Vermehrung. Die anfängliche, schrittweise Überführung in die Testmedien verursacht per se Leistungsunterschiede, deren Auswirkungen sich mindestens bis zu einem Alter von 39 Tagen verfolgen lassen. Doubling times stellen daher objektivere Kriterien dar als absolute Zuwachswerte. Die doubling times von Kolonien, welche länger als 20 Tage in den Testmedien gewachsen waren, zeigen eine Verringerung der Stolonenzuwachsrate mit steigender Temperatur (12°, 17°, 22° C). Die Reihenfolge der fördernden Wirkung der einzelnen Salzgehaltsstufen ergibt sich zu 32 , 24 , 16 , 40 S. Im Prinzip ähnliche Verhältnisse liegen hinsichtlich der asexuellen Vermehrungsrate vor. Bemessen an den getesteten Kriterien scheinen die Temperaturansprüche mit zunehmendem Koloniealter abzunehmen. Die errechneten doubling times sind wesentlich länger als beiCordylophora; möglicherweise deutet dieser Unterschied auf inadäquate Kulturbedingungen (Fütterung, Wasserbewegung) hin.

Activity and thermostability of total, tartrate-resistant (TR), and tartrate-sensitive (TS) acid phosphatases (APh) was determined by spectrophotometric and electrophoretic methods, using 1-naphtyl phosphate (1-NPh) as substrate in supernatant of homogenates of amoebae Amoeba proteus acclimated to 10 and 25°C. A decrease of the APh activity in the pH range from 3.5 to 5.8 is revealed in the amoebae cultured at 10°C (cold) in comparison with the cells cultured at 25°C (warm). The activity and thermostability both of total APh detected at pH 4.0 and of TS-APh detected at pH 5.5 are lower in cold amoebae, than in warm cells, while the activity and the heat resistance of TR-APh (pH 4.0) are identical at both temperatures of cultivation of the amoebae. Thus, acclimation of amoebae to cold is accompanied by a decrease of the activity and of the heat resistance of TS-APh. A decrease of the activity and the thermostability of this APh in cold amoebae correlate with a decrease of their primary cellular thermostability and phagocytic activity.  相似文献

A commercial (Cartazyme) and non-commercial (Asperzyme) xylanases were studied. Cartazyme stability in a 0–70% (v/v) methanol at 50°C and 65°C was carried out. No deactivation was found for Cartazyme in the presence of 15% methanol at 50°C. Half-life activity decay (t1/2) of Cartazyme at 50°C in 30%, 50% and 70% methanol solutions were 4.0 h, 2.3 h and 1.2 h, respectively. At 65°C, which is the ozone-alkali-peroxide (ZEP) bleaching temperature, only significant results on Kappa number reduction and selectivity were only observed in 15% methanol (t1/2 30 min) at the Z stage. For the Asperzyme, a t1/2 of 36.5 min at 50°C was found. In the Z stage with Asperzyme in the presence of 25% of methanol, a 20% Kappa number reduction and an improvement of the ZEP sequence of the brightness of 3.1 points were obtained. Life table parameters of Aphis gossypii Glover (Homoptera: Aphididae) on Gossypium hirsutum L. were determined at six temperatures (10, 15, 20, 25, 30, and 35 ± 0.5°C) in the laboratory. Relationships of life table parameters with temperature were described with mathematical equations. Development was fastest at 30°C, with a pre-larviposition period of 4.6 d. Survival to adult was greatest at 25°C (81%). Fecundity was highest at 25°C, with a total fecundity of 28.3 nymphs per female and a mean reproductive rate of 3.1 nymphs per female per day. Threshold temperatures for development in the first through fourth instar and the adult were 8.2, 8.0, 7.2, 6.2 and 7.9°C, respectively. The durations of these stages, expressed as temperature sums above these thresholds, were 24.2, 23.7, 23.0, 25.5 and 168.8 degree-days (D°), respectively. A. gossypii achieved its maximum net reproductive number (24.4 nymphs per female) and greatest intrinsic rate of increase (0.386 d–1) at 25°C. The high relative rate of population increase at 25°C results in a daily population increase of 47% and a doubling time of only 1.8 d, illustrating the tremendous growth capacity of A. gossypii populations under favourable conditions. Compared to literature sources, our source of A. gossypii, fed on cotton, showed a comparatively great heat tolerance.  相似文献

Summary Monhystera denticulata Timm, a free-living nematode present in the aufwuchs assemblages of several marine macrophytes located in North Sea Harbor, Southampton, New York, was isolated from Zostera marina and established in laboratory culture in order to study the influences of temperature and salinity on its life history. Under experimental conditions, M. denticulata has a generation time (Measured as the time elapsing between the first egg depositions of consecutive generations) of 10–12 days at 25° C and 26 S, which represent optimal growth conditions in the laboratory. The organism has a generation time of 20 days at 25° C and 13, 17 days at 25° C and 39, 18 days at 15° C and 26, 36 days at 15° C and 13 and 34 days at 15° C and 39. As conditions vary from the optimum of 25° C and 26 S, a decrease in temperature of 10° C and an increase or decrease in salinity of 13 results in a doubling of the generation time. At 5° C the generation time is about 180–197 days.Assuming optimum conditions and average generation time, about 15 generations of M. denticulata could occur in North Sea Harbor during the year. In the present study we investigated the response to iron (Fe) deficiency in two cultivars of Festuca rubra L. (Rubina and Barnica) used in correction of chlorosis of fruit trees cultivated on calcareous soils. We found that a Fe-chelating compound, identified as 2-deoxymugineic acid (DMA), was secreted from the roots in response to Fe-deficiency in both cultivars. The amount of DMA secreted into solution increased with the development of Fe-deficiency. The secretion showed a distinct diurnal rhythm characterized by a secretion peak at between 2 and 5 hours after sunrise at 20°C. However, this secretion peak was delayed by 3 hour at low temperature (<10°C) and occurred 3 h earlier at high temperature (30°C). When water used for the collection of root exudates was pre-warmed (25°C) or pre-cooled (10°C), this led to an earlier or a delayed secretion compared to control (15°C) under the same air temperature, respectively. Short-term shading treatment did not affect the secretion pattern of DMA. These results demonstrate that the secretion time of DMA from the roots is, at least partly controlled by the temperature in the root environment. Overall, these findings suggest that the ability of Festuca rubra to prevent Fe chlorosis symptoms (`re-greening effect') of associated fruit trees is partially related to the secretion of DMA which increase Fe availability in calcareous soils.  相似文献

Summary An alkalophilic bacterium producing high amounts of the cell-associated -mannosidase and extracellular -mannanase was isolated from soil. The isolate (AM-001) that grew well in alkaline pH media was identified as a strain of Bacillus sp. The optimal cultivation temperature for enzyme production was 31° C for -mannosidase and 37° C for -mannanase with the optimum production medium composed of 1% konjac powder, 0.2% yeast extract, 2% Polypepton, 0.1% K2HPO4, 0.02% MgSO4 · 7H2O and 0.5% Na2CO3. Optimum pH and temperature for -mannosidase were 7.0 and 55° C, and for -mannanase were 9.0 and 65° C.  相似文献

Morphometric variation in three clones of Brachionus plicatilis cultured at 20°C, 25°C and 30°C, and 9, 12 and 24 salinity was analysed. Size appeared to be largely under genetic control and this defined the narrow limits within which variation due to abiotic factors could occur. Temperature had a significant effect on size, but affected the three clones differently. The most general effect of temperature was a reduction in size which levelled off as the temperature rose. Of the measurements taken, only distance between the median spines was affected by salinity. An important inter-relationship between the effects of temperature and salinity was also detected.  相似文献

Four of the tetraspans family antigens expressed in B cells, CD37, CD53, TAPA-1, and R2/C33, as well as at least two other molecules, CD19 and CD21, coprecipitate with DR antigens from mild detergent lysates of human B-cell lines and tonsillar B cells. Coprecipitation and preclearing experiments indicate the existence of large multicomponent complexes containing jointly the seven components, although some incomplete complexes lacking some of the components may also exist. The complexes contain only a relatively small fraction of the total cellular pool of relevant molecules. The existence of these tetraspans-DR complexes may be related to the previously reported antiproliferative and signaling effects of anti-bodies against most of their components.  相似文献

The thermal denaturation of bovine serum albumin (BSA) was studied at pH 2.8 and 7.0 in the range of 2–65°C. The relative proportions of -helix, -structure, and disordered structure in the protein conformation were determined as a function of temperature, by the curve-fitting method of circular dichroism spectra. With the rise of temperature at pH 7.0, the proportion of -helix decreased above 30°C and those of -structure and disordered structure increased in the same temperature range. The structural change was reversible in the temperature range below 45°C. However, the structural change was partially reversible upon cooling to room temperature subsequent to heating at 65°C. On the other hand, the structural change of BSA at pH 2.3 was completely reversible in the temperature range of 2–65°C, probably because the interactions between domains and between subdomains might disappear due to the acid expansion. The secondary structure of disulfide bridges-cleaved BSA remained unchanged during the heat treatment up to 65°C at pH 2.8 and 7.0.  相似文献
