As part of a program to synthesize the ceramide trisaccharide (1) related to Fabry's disease, methyl 4-O-(4-O-α-d-galactopyranosyl-β-d-galactopyranosyl)-β-d-glucopyranoside (12) was prepared. Methyl β-lactoside (2) was converted into methyl 4-O-(4,6-O-benzylidene-β-d-galactopyranosyl)-β-d-glucopyranoside (4). Methyl 2,3,6-tri-O-benzoyl-4-O-(2,3,6-tri-O-benzoyl-β-d-galactopyranosyl)-β-d-glucopyranoside (7) was synthesized from 4 through the intermediates methyl 2,3,6-tri-O-benzoyl-4-O-(4,6-O-benzylidene-2,3-di-O-benzoyl-β-d-galactopyranosyl)-β-d-glucopyranoside (5) and methyl 2,3,6-tri-O-benzoyl-4-O-(2,3-di-O-benzoyl-β-d-galactopyranosyl)-β-d-glucopyranoside (6). The halide-catalyzed condensation of 7 with 2,3,4,6-tetra-O-benzyl-d-galactopyranosyl bromide (8) gave methyl 2,3,6-tri-O-benzoyl-4-O-[2,3,6-tri-O-benzoyl-4-O-(2,3,4,6-tetra-O-benzyl-α-d-galactopyranosyl)- β-d-galactopyranosyl]-β-d-glucopyranoside (10). Stepwise deprotection of 10 led to 12, the methyl β-glycoside of the trisaccharide related to Fabry's disease.  相似文献

The possible modes of binding for methyl-α-d-mannopyranoside, methyl-β-d-mannopyranoside, 2-O-methyl-α-d-mannopyranoside, methyl-2-O-methyl-α-d-mannopyranoside and methyl-α-d-N-acetylmannosamine to concanavalin A have been investigated using theoretical methods. All these sugars, except methyl-α-d-N-acetylmannosamine, reach the active site of concanavalin A with a highly restricted number of binding orientations. Present investigations suggest that the failure of methyl-α-d-N-acetylmannosamine to bind to concanavalin A is not so much due to steric factors as to repulsive electrostatic interactions. Methyl-2-O-methyl-α-d-mannopyranoside can bind to concanavalin A in one mode whereas the other sugars can bind in more than one mode. The high potency of methyl-α-d-mannopyranoside over methyl-β-d-mannopyranoside is mainly due to the possibility of hydrophobic interactions of the α-methoxy group with Leu(99) or Tyr(100) and also due to the possibility of formation of better and more hydrogen bonds with the protein. A comparison of these data with those for the d-glucopyranosides suggests that the change of the hydroxyl at the C-2 atom from equatorial to axial orientation increases the stereochemically allowed region as well as the possible binding modes. From these studies it is also suggested that the overall shape of the oligosaccharides rather than the terminal or internal mannose alone affects the binding potency of saccharides to concanavalin A.  相似文献

(1) 4,6-O-Ethylidene-d-glucose is a good inhibitor of adipocyte sugar transport from the external surface. Using radioactively labelled 4,6-O-methylidene-d-glucose we have shown that this compound is not taken up into cells by the hexose transporter but through a route which is insulin insensitive, d-glucose insensitive, temperature sensitive and which is slightly inhibited by phloretin. When efflux of 3-O-methyl-d-glucose is studied with 4,6-O-methylidene-d-glucose only present inside the cells then no detectable inhibition is observed indicating that this compound is a good side-specific analogue with a high affinity for only the external site of the hexose transporter. (2) Radioactively labelled alkyl-β-d-glucosides have been prepared. These also penetrate the adipocyte membrane by an insulin and d-glucose insensitive route. The half-times for equilibration with methyl-, n-propyl-, and n-butyl-β-d-glucosides are 255, 9.45 and 3.3 min, respectively, indicating that the uptake rates are dependent upon the size of the alkyl group. (3) The glucosides show poor inhibition of 3-O-methyl-d-glucose transport when added to the external solution only. When cells are preincubated with n-propyl-β-d-glucoside and n-butyl-β-d-glucoside an increase in the amount of inhibition of 3-O-methyl-d-glucosez uptake is observed. This increase in inhibition correlates well with the glucoside uptake rates and indicates that availability of the glucosides at the internal surface of the transporter is required for inhibition. This has been confirmed by measuring 3-O-methyl-d-glucose exit in the presence of n-propyl-β-d-glucoside at the internal surface only. Thus, n-propyl-β-d-glucoside is a good side-specific analogue with high affinity only for the internal site of the hexose transporter. (4) n-Propyl-β-d-glucoside inhibition of d-allose transport is fully reversible. If cells are washed after a preincubation with the analogue then the inhibition is lost. The n-propyl-β-d-glucoside inhibition of d-allose transport is reduced competitively by 3-O-methyl-d-glucose. (5) 6-O-Propyl-d-galactose has low affinity for both internal and external sites.  相似文献

Condensation of 3,5-di-O-benzoyl-β-d-ribofuranosyl chloride severally with 3-acetyl-5-alkylpyridines, 5-alkyl-3-methoxycarbonylpyridines (alkyl = Me, Et, Pr, and iPr), 5-isopropylnicotinamide, and 3,5-diacetylpyridine bis(ethylene acetal) in acetonitrile at ?5° gave the corresponding 1-(3,5-di-O-benzoyl-β-d-ribofuranosyl))-3,5-disubstituted pyridinium chlorides in excellent yield (90%). From the reaction of a series of 2,3-O-isopropylidene-β-d-ribofuranosyl halides with 3-acetyl-5-methyl-pyridine at room temperature, the α-nucleosides were obtained.  相似文献

The syntheses of (±) 2α,6β-diethyl-7α-ethynyl-3α-(p-hydroxyphenyl)-trans-bicyclo[4.3.0]nonan-7β-ol (8), (±)2β,6β-diethyl-7α-ethynyl-3β-(p-methoxyphenyl)-trans-bicyclo[4.3.0]nonan-7β-ol (12) and (±) 2α,6β-diethyl-7α-ethynyl-3β-(p-hydroxyphenyl)-trans-bicyclo[4.3.0]nonan-7β-ol (18) and their derivatives, which are essentially B-seco-steroids having cis-anti-trans, cis-syn-trans and trans-anti-trans geometries have been carried out. A study of their antiimplantation activities (AI) and receptor binding affinities (RBA) show that trans-anti-trans compounds are biologically most potent, followed by the corresponding cis-anti-trans and cis-syn-trans compounds. The most potent compound 18 is active at 1 mg/kg in rats. Introduction of 7α-ethynyl group increases their AI activity; however, no significant effect on their RBA is observed.  相似文献

Both pairs of dl-ll-desoxy- and dl-13-cis-erythro-15, 16-dihydroxyprostaglandins have been synthesized via 1,4-conjugate additions of an appropriately functionalized cis-vinyl cuprate to the requisite cyclopentenone. A procedure for production of N-acetyl-l-methionine (N-Ac-l-Met) from its racemic mixture by enzymatic continuous hydrolysis of the l-enantiomer of N-acetyl-dl-methioninamide (N-Ac-dl-Met · NH2) is described. For this hydrolysis, whole cells of Erwinia carotovora immobilized by κ-carrageenan gel were employed. The complete conversion of N-Ac-dl-Met · NH2 to N-Ac-l-Met was achieved by using a column packed with the immobilized cells. The half-life of the enzyme activity of the column was calculated to be ~100 days. Pure crystalline N-Ac-l-Met was readily isolated from the effluent with >87.5% yield. N-Ac-d-Met · NH2 was easily racemized by incubating in ethanol containing 0.1 m NaOH for 30 min at 80°C, and used again for substrate.  相似文献

A series of 12α-hydroxy steroids with varying side chains was prepared, and their 24-hour acetylation yields were compared, l2α-Hydroxy-5β-pregnan-20-one (lb) was prepared from 3α, 12α-diacetoxy-5β~pregnan-20-one (2) and also by side chain degradation of 12α-acetoxy-5β-cholanoic acid (5d). 21-Benzyl-5β-pregnan-12α-ol (1g) was synthesized by hydrogenation of the 21-benzylidine derivative of ketone 1b. 23-Pheny1-5β-norcholan-12α-ol (1k) was obtained by the Grignard reaction of 2-phenyl-ethylmagnesium bromide and ketone 1b, dehydration, hydrogenation and hydride reduction; a similar sequence produced 20-methyl-5β-pregnan-12α-ol (lm). The acetylation results (Table 11) imply that branching at C-20 may be more significant for 12α-hydroxyl reactivity than side chain length or type. An additional compound with an unbranched side chain, 21-nor-5β-cholan-12α-ol (14), was synthesized by a Grignard reaction on the 21-bromo intermediate 11b. Acetylation rates determined by glc indicate (Table 111) That compounds with unbranched side chains have 12α-hydroxyl groups about ten times as reactive as their analogs with 20-methyl groups.  相似文献

In normal human subjects under basal conditions, we have reported that molar concentrations of immunoreactive β-lipotropin (IR-β-LPH) are approximately threefold greater than those of IR-β-endorphin (β-Ep). Following acute stimulation, there is a further two- to threefold disproportionate rise in plasma concentrations of IR-β-LPH as compared to those of IR-β-Ep. To begin to assess the possible factors involved in such altered IR-β-LPH/IR-β-Ep ratios in plasma, the metabolic clearance rate (MCR), volume of distribution (Vd), fractional rate of disappearance (Kd), and half-life (t12) of these peptides were determined by means of bolus injection of highly purified human β-LPH and synthetic human β-Ep in normal human subjects. β-Ep was found to have an MCR and a Kd greater than that of β-LPH, and a shorter t12. These differences, however, although they may in part be contributory, cannot solely account for the greater ratio of IR-β-LPH to IR-β-Ep in plasma, or for the disproportionate rise in plasma concentrations of these peptides after acute stimulation.  相似文献

Treatment of Sendai virus with p-(sec-butyl)-phenyl-6-chloro-6-deoxy-β-d-glucopyranoside, followed by freezing and thawing resulted in a loss of hemolytic and cell fusion activities as well as infectivity without affecting hemagglutinating and neuraminidase activities. The anti-hemolytic activity of this compound was reversed by the addition of phosphatidyl choline to the virus samples. p-Azidophenyl-6-chloro-6-deoxy-β-d-[3H]glucopyranoside was successfully used for photoaffinity labeling of a specific virion site, and we confirmed the affected site of the glucoside to be the lipid components in the viral envelopes.  相似文献

  • 1.1. In human plasma, an enzyme is present which hydrolyzes 4-methylumbelliferyl-tetra-N-acetylchitotetraoside. The function of this enzyme is unknown.
  • 2.2. We have examined whether hyaluronidase, neutral endoglucosaminidase, N-acetyl-β-d-hexosaminidase, aspartylglucosaminidase, β-d-glucosidase, and chitobiase could hydrolyze MU-TACT. The results obtained are detailed below.
  • 3.3. A purified commercial preparation of hyaluronidase does not hydrolyze MU-TACT.
  • 4.4. Substrate specificity requirements, pH optimum and subcellular localization indicate that neutral endoglucosaminidase is distinguishable from MU-TACT hydrolase. Also commercial neutral endoglucosaminidase D and H have no affinity towards MU-TACT.
  • 5.5. N-Acetyl-β-d-hexosaminidase is different from MU-TACT hydrolase for the following reasons: (a) a purified enzyme preparation does not hydrolyze MU-TACT; (b) there is no correlation in the activity of the enzymes; (c) MU-TACT hydrolase is not deficient in cells of a patient with a deficiency of total N-acetyl-β-d-glucosaminidase; and (d) the 2 enzymes have very different Chromatographic characteristics and Con A binding properties.
  • 6.6. Enzyme characteristics, substrate structural requirements and a lack of correlation with MU-TACT hydrolase activity suggest that aspartylglucosaminidase, β-d-glucosidase, and chitobiase are not involved in the hydrolysis of MU-TACT.
  • 7.7. None of the enzymes which we have considered corresponds to MU-TACT hydrolase. The exact nature and the function of the enzyme remains an enigma.

The capsular polysaccharide from Klebsiella K44 has been investigated by the techniques of methylation, base-catalyzed elimination, Smith degradation, and partial hydrolysis. The last-named yielded an oligosaccharide corresponding to one repeating unit. The anomeric configutations of the sugar residueswere determined by 1H- and 13C-n.m.r. spectroscopy. The products of nitrous acid deamination of per-O-methylated 2-amino-2-deoxy-d-glucitol and 2-amino-2-deoxy-3-O-β-d-galactopyranosyl-d-glucitol and its per-O-methylated derivative have been characterized by g.l.c.—mass spectrometry after treatment with sodium borodeuteride and further substitution by acetylation, methylation, or (trideuteriomethyl)ation. The results confirm that the most important reaction pathway (1) involves a 1 → 2-hydride shift to give 2-deoxy-d-arabino-hexoses, but that significant side-reactions include (2) solvolytic displacement at C-2, (3) a 3 → 2-hydride shift, to give 2-deoxy-d-erythro-3-hexuloses, and (4) a C-4→C-2 migration to give 2-deoxy-2-C-(hydroxymethyl)-d-ribose and -d-arabinose. Reactions (3) and (4) result in elimination of the original 3-O-substituents, with the exposure of new reducing groups, from oligosaccharides terminated by 3-O-substituted 2-amino-2-deoxyhexitols.  相似文献

Cellobiose hydrolysis by β-d-glucosidase (β-d-glucoside glucohydrolase EC 3.2.1.21) can become the rate-limiting step in the hydrolysis of cellulosic wastes because of inhibition phenomena involving other enzymes of the cellulase complex. Enhancement of the overall rate can therefore be obtained by increasing the amount of β-d-glucosidase present in the reactor. Unfortunately, the thermal stability of β-d-glucosidase is rather poor compared to endo-(1 → 4)-β-d-glucanase and cellobiohydrolase. A novel stabilization method is proposed that exploits the polarization phenomena that take place in an unstirred ultrafiltration membrane enzymatic reactor. As much as a 20-fold increase in half-life compared to the native enzyme is obtained by injecting small amounts of hydroxyethyl cellulose into the system. No reduction in enzyme activity levels is observed.  相似文献

The effect of P[N,N-bis(2-chloroethyl)amino]phenylacetate esters of 3β-hydroxy-methyl-17α-aza-d-homo-5α-androstan-17-one (compound 3) and 3β-hydroxy-17α-aza-d-homo-5α-androstane (compound 2) on sister-chromatid exchange (SCE) frequencies and on human lymphocytes proliferation kinetics was studied. The results are compared with those of the P[N,N-bis(2-chloroethyl)phenylacetate esters of 3β-hydroxy-17α-aza-d-homo-5α-androstan-17-one (compound 1). All compounds were found to be active in inducing markedly increased SCE rates and cell division delays. A correlation between potency for SCE induction, effectiveness in cell division delay and previously established antitumour activity of these compounds was observed.  相似文献

A new naturally occurring chalcone glycoside, 2′,-dihydroxy-4-methoxy-4′-O-β-d-glucosyl chalcone, and other chalcones, aurones, flavonols, and anthocyanins are reported from Megalodonta (Bidens) beckii. The plan is an amphibious aquatic with heterophyllous leaves, but the presence of certain flavonoids is correlated with vegetative versus floral tissues rather than vegetative leaf form. The implications of flavonoid chemistry with respect to the relationship of M. beckii to several sections of Bidens are discussed.  相似文献

The tetrasaccharide fraction obtained by gel chromatography after treatment of commercially available heparin with nitrous acid was reduced with NaB3H4 and then hydrolysed with 2m trifluoracetic acid at 70° for 3 days. By gel chromatography and electrophoresis, the 3H-labelled trisaccharide 1 bearing an unsubstituted 2-amino-2-deoxy-d-glucosyl group in the non-reducing position was obtained (18% from the 3H-labelled tetrasaccharide). By sequential, enzymic degradation, the structure α-d-GlcN-(1→4)-β-d-GlcA-(1→4)-[1-3H]aManol was obtained for 1, which is a substrate for acetyl-CoA: 2-amino-2-deoxy-α-d-glucoside N-acetyltransferase, an enzyme that is deficient in the Sanfilippo C syndrome. In human-skin fibroblasts, the pH optimum of acetyl transfer onto 1 was between pH 5.5 and 7.0, and dependent on the buffer. An apparent Km for 1 of 0.14mM was found.  相似文献

A number of lectins has been purified by affinity chromatography on O-glycosyl polyacrylamide gels. The lectins isolated (and the particular sugar ligands used in the affinity carriers) are as follows: Anguilla anguilla, serum (α-L-fucosyl-), Vicia cracca, seeds; Phaseolus lunatus, seeds; Glycine soja, seeds; Dolichos biflorus, seeds; Maclura pomifera, seeds; Sarothamnus scoparius, seeds; Helix pomatia, ablumin glands; Clitocybe nebularis, fruiting bodies (all N-acetyl-α-d-galactosaminyl-); Ricinus communis, seeds (β-lactosyl-); Onomis spinosa, root; Fomes fomentarius, fruiting bodies; Marasmius oreades, fruiting bodies (all α-d-galactosyl-), Canavalia ensiformis, seeds, (i.e., concanavalin A) (α-d-glucosyl-).Physicochemical properties of Glycine soja, Dolichos bifloras, Phaseolus lunatus, Helix pomatia and Ricinus communis lectins correponded well to properties of the preparations studied earlier by other workers. For the other purified lectins the essential physicochemical data (sedimentation coefficient, molecular weight, subunit composition, electrophoretic patterns, amino acid composition, carbohydrate content, isoelectric point) were established and their precipitating, hemagglutinating and mitogenic activities determined.  相似文献

A general method has been developed for determining the rate of entry of lactose into cells of Escherichia coli that contain β-galactosidase. Lactose entry is measured by either the glucose or galactose released after lactose hydrolysis. Since lactose is hydrolyzed by β-galactosidase as soon as it enters the cell, this assay measures the activity of the lactose transport system with respect to the translocation step. Using assays of glucose release, lactose entry was studied in strain GN2, which does not phosphorylate glucose. Lactose entry was stimulated 3-fold when cells were also presented with readily metabolizable substrates. Entry of o-nitrophenyl-β-d-galactopyranoside (ONPG) was only slightly elevated (1.5-fold) under the same conditions. The effects of arsenate treatment and anaerobiosis suggest that lactose entry may be limited by the need for reextrusion of protons which enter during H+/sugar cotransport. Entry of o-nitrophenyl-β-d-galactopyranoside is less dependent on the need for proton reextrusion, probably because the stoichiometry of H+/substrate cotransport is greater for lactose than for ONPG.  相似文献
