Summary The water permeability of astomatous cuticular membranes isolated from Citrus aurantium L. leaves, pear (Pyrus communis L.) leaves and onion (Allium cepa L.) bulb scales was determined before and after extraction of cuticular waxes with lipid solvents. In pear, the permeability coefficients for diffusion of tritiated water across cuticular membranes (CM) prior to extraction [P d(CM)] decreased by a factor of four during leaf expansion. In all three species investigated P d(CM) values of cuticular membranes from fully expanded leaves varied between 1 to 2×10-7 cm-3 s-1·P d(CM) values were not affected by pH. Extraction of cuticular waxes from the membranes increased their water permeability by a factor of 300 to 500. Permeability coefficients for diffusion of THO across the cutin matrix (MX) after extraction [P d(MX)] increased with increasing pH. P dvalues were not inversely proportional to the thickness of cuticular membranes. By treating the cutin matrix and cuticular waxes as two resistances acting in series it was shown that the water permeability of cuticles is completely determined by the waxes. The lack of the P d(CM) values to respond to pH appeared to be due to structural effects of waxes in the cutin matrix. Cuticular membranes from the submerse leaves of the aquatic plant Potamogeton lucens L. were three orders of magnitude more permeable to water than the cuticular membranes of the terrestrial species investigated.Abbreviations CM cuticular membrane - MX cutin matrix - WAX waxes This study was supported by a grant from the Deutsche Forschungsgemeinschaft.  相似文献

Thirty‐five codling moth (CM, Cydia pomonella L., Lepidoptera, Tortricidae) populations collected in different commercial orchards in six European countries were tested for their susceptibility to Cydia pomonella granulovirus (CpGV‐M). Including previously published data on CpGV‐M resistance, a total of 38 CM colonies showed considerably elevated LC50 values, independent of the country origin. When only few test individuals are available, determination of mortality of neonate larvae at a discriminating concentration range of 104 to 106 OB/ml (>log4) as a direct measure of percentage susceptible individuals in a CM population is more advisable than calculation of LC50 values. The >log4 mortality alone or in combination with the LC50 value can be used for identification of resistance in a population. Results indicated a locally separated but widely spread occurrence of CM populations with low susceptibility to CpGV‐M. The most plausible hypothesis for the emergence of CpGV‐M resistance is its selection by repeated use of CpGV products.  相似文献

Several indoleacetic acids, substituted in the benzene ring, were compared in the Avena straight growth bioassay. 4-Chloroindoleacetic acid, a naturally occurring plant hormone, is one of the strongest hormones in this bioassay. With an optimum at 10-6 mol l-1, it is more active than indoleacetic acid, 2,4-dichlorphenoxyacetic acid and naphthaleneacetic acid. 5-Chloro- and 6-chloroindoleacetic acids are very strong auxins as well. Other derivatives tested have a lower activity. 5,7-Dichloro- and 5-hydroxyindoleacetic acids have very low auxin activity at 10-4 mol l-1 and may be anti-auxins. Some of the derivatives were compared for their effect on pH decline in stem protoplast suspensions of Helianthus annuus L. and Pisum sativum L. The change of pH occurs without a lag period or with only a very short one. Investigations on the absorption of shortchain fatty acids across ruminal epithelium of sheep were performed both in vitro (Ussing chamber technique, using propionic acid representatively for shortchain fatty acids) and in vivo (washed, isolated reticulorumen). A pH-induced, nearly tenfold increase in the concentration of undissociated propionate led to an only twofold increase in mucosal-to-serosal flux of propionate (in vitro). Neither amiloride (1 mmol·l-1, in vitro) nor theophylline (10 mmol·l-1, in vivo), inhibitors of the ruminal Na+/H+ exchanger, exerted any significant influence on propionate fluxes or short-chain fatty acids absorption, respectively. Total replacement of luminal Na+ (by choline) did not alter short-chain fatty acids absorption (in vivo). Mucosal 4,4-diisothiocyanatostilbene-2,2-disulfonic acid (0.1 mmol·l-1) or mucosal nitrate (40 mmol·l-1) markedly reduced propionate net flux (in vitro). Increasing mucosal Cl- concentration brought about a significant drop in mucosal-to-serosal flux of propionate (in vitro) and in short-chain fatty acids net absorption (in vivo), respectively. The results obtained suggest that short-chain fatty acids are absorbed both as anions and as undissociated acids across ruminal epithelium of sheep. Effects of the gibberellins A4+7(GA4+7) and A3(GA3), benzyladenine (BA) and forchlorfenuron (CPPU) on deposition of the cuticular membrane (CM) in developing tomato (Lycopersicon esculentum L.) fruit were investigated. Growth regulators were applied when fruit development within trusses ranged from the flower to the mature stage. Developmental stage of fruit at the time of application was indexed by fruit diameter. Fruit were harvested at maturity, the CM isolated enzymatically on an individual fruit basis and mass of CM per unit fruit surface area calculated. In mature fruit, mass of CM per fruit increased with fruit size, but mass of CM per unit surface area was independent of fruit size, position within a truss and position of the truss on the plant. GA4+7 and GA3 increased CM mass per unit fruit surface area at concentrations up to 300 mg l−1. Young fruit (5–10 mm diam. at time of application) was most responsive. Responsiveness decreased as fruit development at application progressed towards maturity. There was no consistent effect of GA4+7 or GA3 on fruit mass. BA (up to 100 mg l−1) or CPPU (up to 3 mg l−1) had no significant effect on CM mass per unit surface area regardless of developmental stage. Higher concentrations of BA or CPPU decreased CM mass per unit surface area. There was no effect of BA or CPPU on fruit mass. Potential mechanisms and benefits of a gibberellin induced increase in CM deposition are discussed.  相似文献

Summary Carnitine was found to stimulate fatty acid oxidation by pea (Pisum sativum L.) cotyledon mitochondria. The stimulation was at a maximum for long chain (C16:0) and short chain (C4:0 and C6:0) fatty acids. Evidence was also provided which indicated that mid-chain (C10:0 and C12:0) fatty acid oxidation by mitochondria was stimulated by carnitine. The cyanobacteriumAnabaena siamensis Antarikanonda, isolated from rice paddies of Bangkok, Thailand, liberates substantial quantities of free amino acids into the external medium irrespective of whether it is growing on N2, NH4 +, NO3 or under nitrogen-starved conditions. Addition of such combined nitrogen causes changes in both intracellular and extracellular free amino acid pool patterns. No overall relationship exists between the amino acid efflux and the intracellular pools. The most abundant free amino acids found in the external media of N2, NO3 , NH4 +-grown and N-starved cultures were phenylalanine, threonine, glutamate, and glycine, respectively. These investigations suggest that amino acid liberation by the cyanobacterium is a selective diffusional process that is sensitive to environmental changes.  相似文献

The abilities of various sorbents to adsorb catalase (CAT; EC 1.11.1.6) from filtered culture liquid (FCL) of the fungus Penicillium piceum F-648 were compared. Potassium phosphate, hydroxyapatite (HAP), and coprecipitated sorbents containing calcium phosphate and magnesium hydroxide adsorbed extracellular CAT more efficiently than aluminum oxide, aluminum phosphate, or quartz sand. The enzyme was isolated from FCL of Penicillium piceum with the use of HAP and a binary coprecipitated sorbent, Ca3(PO4)2 + Mg(OH)2, 1 : 1 (CM). The CAT(CM) sample contained the least amount of protein admixture. Its spectra had absorption maximums at 279.6, 406.8 (Soret band), 540, 585, 636, and 703 nm and negative molar ellipticity minimums at 207 and 210–214 nm. The kinetic indices of the samples (K M, V max : K M, and specific activity) were intricately dependent on the protein concentration in the reaction mixture. In dilute solutions, the K M and specific activities of CAT(CM) and CAT(HAP) equaled 667 and 137 mM; 300.9 × 104 and 30.0 × 104 U/mg protein, respectively. The effective velocity constants of inactivation of CAT(HAP), CAT(CM), and FCL in the reaction of H2O2 decomposition increased dramatically after the dilution of samples. In the infinitely dilute solution, they were 4.30 × 10–2, 6.46 × 10–2, and 1.12 × 10–2 s–1, respectively.  相似文献

Ethylene treatment (approx. 20 l ·1-1 in air for 2 d) of tobacco (Nicotiana tabacum L. cv. Havana 425) plants markedly increases the endo--1,3-glucanase (EC 3.2.1.39) content of leaves. The antigenic form of the enzyme induced is the same one whose production is blocked by treating cultured cells with combinations of auxin (1.1 · 10-5 M -naphthaleneacetic acid) and cytokinin (1.4 · 10-6 M kinetin). Evidence is presented that cultured tobacco cells require ethylene for -1,3-glucanase accumulation: i) ethylene treatment increased the accumulation of \-1,3-glucanase in callus tissues >10 d after subculturing and in cell-suspension cultures; ii) callus tissues can produce ethylene; iii) conditions known to inhibit ethylene production (1 mM CoCl2; 33° C treatment) or ethylene action (approx. 1.6 mmol · 1-1 norbornadiene in air) inhibited -1,3-glucanase accumulation by callus tissues treated for 4 d following subculturing; and, these inhibitory effects were prevented by exogenous ethylene. Combinations of auxin and cytokinin blocked ethylene-induced accumulation of -1,3-glucanase by cell-suspension cultures. The potential of alginate-immobilized Anabaena doliolum and Chlorella vulgaris was assessed for removal of nutrients (NO inf3 sup- and NH inf4 sup+ ) and metals (Cr2O inf7 sup2- and Ni2+) at different biomass concentrations (0.05, 0.1, 0.25, 0.49 and 1.22 g dry wt l-1) and pH values (4 to 10). Though uptake of all these substances was higher in concentrated algal beads (0.25, 0.49 and 1.22 g dry wt l-1), their rate of uptake was significantly (P<0.001) lower than that of low (0.05 g dry wt l-1) cell density beads. For A. doliolum, there was no significant difference in uptake rates for beads having densities of 0.05 and 0.1 g dry wt l-1. Chlorella vulgaris, however, showed maximum efficiency at 0.1 g dry wt l-1. Uptake of both the nutrients and the metals was maximal at pH 7 followed by pH 8, 6, 9, 10, 5 and 4. Of the different substances (organic acids and divalent cations) used, humic acid was most efficient in decreasing metal uptake. Mg2+ was, however, more efficient than Ca2+ in decreasing Ni2+ uptake. Immobilized algae with a cell density of 0.1 g dry wt l-1 were the most efficient for nutrient and metal removal at pH 6 to 8.  相似文献

Using the system vapor/membrane/liquid, permeability coefficients of cuticular transpiration (P ct) were determined as functions of water activity in the vapor (a wv). Enzymatically isolated cuticular membranes (CM) of Citrus aurantium L. and nonisolated CM of onion bulb scales and eggplant fruits were investigated. P ct of Citrus and eggplant CM decreased with decreasing a wv, while permeability coefficients of CM of onion were independent of a wv. Extraction of soluble cuticular lipids (SCL) from the CM of Citrus increased permeability coefficients by a factor of approximately 500. This extraction had no effect on the dependence of P ct on a wv.Treating cuticular membranes as a resistance network consisting of SCL and the polymer matrix, it is shown that the permeability of onion CM is determined by the resistance of the SCL arranged in series with the polymer matrix. In this type of CM liquid and vapor are separated by a continuous, nonporous layer of SCL, and the driving force of transpiration is the gradient of partial pressure of water vapor across the SCL layer. In the CM of Citrus and eggplant, the SCL layer is traversed by polar pores that swell or shrink depending on a wv. However, liquid continuity is maintained across these membranes down to a wv=0.22, the lowest value used. Mobilities of lipophilic organic solutes in cuticular membranes (CM) isolated from mature leaves of Citrus aurantium L., Citrus grandis L., Hedera helix L., IIex aquifolium L., Ilex paraguariensis St.-Hil., Mains domestica Borkh., Prunus armeniaca L., Primus laurocerasus L., Pyrus communis L., Pyrus pyrifolia (Burm. f.) Nakai, Stephanotis florihunda Brongn. and Strophantus gratus Baill. were measured over a temperature range of 15–78°C. In this range, solute mobilities increased up to 1000-fold, which corresponds to temperature coefficients Q10 of 3 (IAA in P. armeniaca) to 14 (cholesterol in H. helix). For most species, Arrhenius graphs showed good linearity up to 40°C, and up to 78°C for some species, while for others activation energies declined with increasing temperature. However, no distinct phase transitions caused by sudden structural changes in the CM were observed. In three species we examined whether heating to 70°C changed solute mobility irreversibly by comparing Arrhenius graphs for two successive experiments with the same CM. The two graphs were very similar for P. laurocerasus, while mobilities in the second graph were somewhat reduced for C. aurantium and greatly increased (at 25 and 35°C) for H. helix. This indicates rearrangements of at least some wax constituents when heated to high temperatures. The activation energies of diffusion (ED) ranged from 75 to 189 KJ mol?11 depending on species and solute size. Size selectivity and variability between cuticles decreased with increasing temperature, and this is caused by differences in (ED). An excellent correlation between the pre-exponential factor of the Arrhenius equation and ED was observed, which is evidence that organic solutes differing greatly in molecular size (130–349 cm3 mol?1) and cuticle/water partition coefficient (25–108) use similar diffusion paths in the CM of all 12 plant species tested. Diffusion occurs in regions with identical physicochemical properties and differs only in magnitude.  相似文献

Non-mycorrhizal plants of the alpine sedge, Kobresia myosuroides, take up the amino acid glycine from nutrient solutions at greater rates than NO inf3 sup- or NH inf4 sup+ . The amino acids glutamate and proline were also taken up at high rates. Total plant biomass was twice as high after 4 months of growth on glycine, compared to NH4NO3, with significant increases in both root and leaf biomass. By taking advantage of differences in the 13C signature of air in the growth chamber and the glycine used for growth, a two-member mixing model was used to estimate that a significant amount of the glycine was taken up as intact molecules, enough to contribute 16% of the total carbon assimilation over a 4-month growing period. Glycine uptake was inhibited when roots were exposed to N2 in place of air, and when the protonophore carbonyl cyanide m-chlorophenylhydrazone (CCCP) was added to the root solution. From these results it is concluded that glycine uptake occurs through active transport. Glycine uptake exhibited a Q10 of 2.0 over the temperature range 5–15° C, with relatively high rates maintained at the lowest temperature measured (5° C). Roots of Kobreasia were not capable of taking up NH inf4 sup+ at measureable rates. To our knowledge, this is the first report of a plant whose non-mycorrhizal roots cannot take up NH inf4 sup+ . Measurements of three N fractions (NO inf3 sup- , NH inf4 sup+ , and total amino acids) in the soil pore water were made over two growing seasons in two Kobresia dry meadows using microlysimeters. At the West Knoll site, which is characterized by soils with average amounts of organic matter, the dominant forms of N in the soil pore water were NO inf3 sup- and NH inf4 sup+ (0–450 mol L-1). Amino acid concentrations were generally less than 20 mol L-1 at this site. At the East Knoll site, which is characterized by soils with higher than average amounts of organic matter, amino acids were generally present at higher concentrations (17–100 mol L-1), compared to NO inf3 sup- and NH inf4 sup+ . The most abundant amino acids were glycine (10–100 mol L-1), glutamate (5–70 mol L-1), and late in the season cystein (5–15 mol L-1). The results demonstrate that this sedge, which dominates dry meadow communities in many alpine ecosystems, is capable of taking up intact amino acids as a principal N source, and has access to high amino acid concentrations in certain alpine soils. Such uptake of organic N may accommodate plant N demands in the face of slow alpine N mineralization rates due to cold soil temperatures.  相似文献

Summary Mechanisms of proton conductance (G H) were investigated in phospholipid bilayer membranes containing long-chain fatty acids (lauric, myristic, palmitic, oleic or phytanic). Membranes were formed from diphytanoyl phosphatidylcholine in decane plus chlorodecane (usually 30% vol/vol). Fatty acids were added either to the aqueous phase or to the membrane-forming solution. Proton conductance was calculated from the steadystate total conductance and the H+ diffusion potential produced by a transmembrane pH gradient. Fatty acids causedG H to increase in proportion to the first power of the fatty acid concentration. TheG H induced by fatty acids was inhibited by phloretin, low pH and serum albumin.G H was increased by chlorodecane, and the voltage dependence ofG H was superlinear. The results suggest that fatty acids act as simple (A type) proton carriers. The membrane: water partition coefficient (K p ) and adsorption coefficient () were estimated by finding the membrane and aqueous fatty acid concentrations which gave identical values ofG H. For palmitic and oleic acidsK p was about 105 and was about 10–2 cm. The A translocation or flip-flop rate (k a ) was estimated from the value ofG H and the fatty acid concentration in the membrane, assuming that A translocation was the rate limiting step in H+ transport. Thek A 's were about 10–4 sec–1, slower than classical weak-acid uncouplers by a factor of 105. Although long-chain fatty acids are relatively inefficient H+ carriers, they may cause significant biological H+ conductance when present in the membrane at high concentrations, e.g., in ischemia, hypoxia, hormonally induced lipolysis, or certain hereditary disorders, e.g., Refsum's (phytanic acid storage) disease.  相似文献

Comparative 14CO2 pulse-12CO2 chase studies performed at CO2 compensation ()-versus air-concentrations of CO2 demonstrated a four-to eightfold increase in assimilation of 14CO2 into the C4 acids malate and aspartate by leaves of the C3-C4 intermediate species Panicum milioides Nees ex Trin., P. decipiens Nees ex Trin., Moricandia arvensis (L.) DC., and M. spinosa Pomel at . Specifically, the distribution of 14C in malate and aspartate following a 10-s pulse with 14CO2 increases from 2% to 17% (P. milioides) and 4% to 16% (M. arvensis) when leaves are illuminated at the CO2 compensation concentration (20 l CO2/l, 21% O2) versus air (340 l CO2/l, 21% O2). Chasing recently incorporated 14C for up to 5 min with 12CO2 failed to show any substantial turnover of label in the C4 acids or in carbon-4 of malate. The C4-acid labeling patterns of leaves of the closely related C3 species, P. laxum Sw. and M. moricandioides (Boiss.) Heywood, were found to be relatively unresponsive to changes in pCO2 from air to . These data demonstrate that the C3-C4 intermediate species of Panicum and Moricandia possess an inherently greater capacity for CO2 assimilation via phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) at the CO2 compensation concentration than closely related C3 species. The antibacterial peptide CM4 (ABP-CM4), isolated from Chinese Bombys mori, is a 35-residue cationic, amphipathic α-helical peptide that exhibits a broad range of antimicrobial activity. To explore a new approach for the expression of ABP-CM4 in E. coli, the gene ABP-CM4, obtained by recursive PCR (rPCR), was cloned into the vector pET32a to construct a fusion expression plasmid. The fusion protein Trx-CM4 was expressed in soluble form, purified by Ni2+-chelating chromatography, and cleaved by formic acid to release recombinant CM4. Purification of rCM4 was achieved by affinity chromatography and reverse-phase HPLC. The purified of recombinant peptide showed antimicrobial activities against E. coli K12D31, Penicillium chrysogenum, Aspergillus niger and Gibberella saubinetii. According to the antimicrobial peptide database (http://aps.unmc.edu/AP/main.html), 116 peptides contain a Met residue, but only 5 peptides contain the AspPro site, indicating a broader application of formic acid than CNBr in cleaving fusion protein. The successful application to the expression of the ABP-CM4 indicates that the system is a low-cost, efficient way of producting milligram quantities of ABP-CM4 that is biologically active.  相似文献

Monosodium urate (MSU) crystals stimulate the production of arachidonic acid metabolites by human neutrophils and platelets. Neutrophils exposed to MSU generated leukotriene B4(LTB4). 6- -LTB4, 12- -6- -LTB4, and 5S, 12S DHETE from endogenous sources of arachidonate. In addition to these metabolites both monohyroxyeicosatetraenoic acids (i.e., 5-HETE) and w-oxidation products (i.e., 20-COOH LTB4) were formed by neutrophils exposed to MSU. Addition of exogenous arachidonic acid led to increased formation of each of these metabolites. When neutrophils were treated with colchicine (10 uM), LTB4 but 5-HETE formation was impaired. (1-14C) Arachidonate-labeled platelets exposed to MSU released (1-14C)-arachidonate. (14C)-12 HETE, (14C)-HHT and (14C)-thromboxane B2. Results indicate that MSU stimulates arachidonic acid metabolism in both human neutrophils and platelets. Moreover, they suggest not only that metabolites of arachidonate may be considered as possible candidates for mediators of inflammation in crystal-associated diseases, but that colchicine blocks the formation of LTB4.  相似文献

Addition to rat liver mitochondria of 2 mM inorganic phosphate or 0.15 mM diamide, a thiol-oxidizing agent, induced an efflux of endogenous Mg2+ linear with time and dependent on coupled respiration. No net Ca2+ release occurred under these conditions, while a concomitant release of K+ was observed. Mg2+ efflux mediated either by Pi or low concentrations of diamide was completely prevented by EGTA, Ruthenium red, and NEM. These reagents also inhibited the increased rate of state 4 respiration induced both by Pi and diamide. At higher concentrations (0.4 mM), diamide induced an efflux of Mg2+ which was associated also with a release of endogenous Ca2+. Under these conditions EGTA completely prevented Mg2+ and K+ effluxes, while they were only partially inhibited by Ruthenium red and NEM. It is assumed that Mg2+ efflux, occurring at low diamide concentrations or in the presence of phosphate, is dependent on a cyclic in-and-out movement of Ca2+ across the inner mitochondrial membrane, in which the passive efflux is compensated by a continuous energy linked reuptake. This explains the dependence of Mg2+ efflux on coupled respiration, as well as the increased rate of state 4 respiration. This report presents evidence demonstrating that chick embryo chondrocyte cultures release into the culture medium a factor(s) which itself can act on chondrocytes to promote their own differentiation. Conditioned medium (CM) stimulates the synthesis of both sulfated mucopolysaccharides, as shown by increased incorporation of 35SO4 or glucose-14C into hyaluronidase-sensitive material, and collagen. However, protein synthesis, DNA synthesis, and cell number are not affected. While the identity of the factor is not yet known, it is nondialyzable, trypsin-and heat-sensitive. The factor is evidently a specialized product of chondrocytes, because it is not made by unexpressed chondrocytes or differentiated pigmented retina cultures. CM works rapidly on test cultures and has a significant effect on 35SO4 incorporation after 2 hr of treatment. In addition, the effect is relatively stable and is not reversed when CM is replaced with fresh medium. The results are significant in that they demonstrate that chondrocytes produce a factor that promotes their own differentiation, as defined in terms of the synthesis of two distinct specialized products.  相似文献
