The Ca++ and Mg++ contents of embryonic chick heart were studied by atomic absorption spectrophotometry during a period from 48 h of foetal development until 2-3 days post-hatching. The hearts were isolated and incubated for 40 min at 22°C in three different media aerated with 95% 02-5% C02. The media included: normal Ringer's; Ca+-free Ringer's with 3 mM EGTA; and Ca++-free Ringer's with 3 mM EDTA. At 48 h, the tubular myocardium contained 7-3 mM Ca++ per wet weight which decreased rapidly to 1-2 mM by 10 days of development and remained between 0-9 and 1-1 mM until hatching. The Ca++ content paralleled the changes in Na+ content reported earlier. Treatment with excess chelators, EGTA or EDTA, resulted in removal of 65-75% of the Ca++ content throughout development until the time of hatching, when 50% of the Ca++ became firmly bound. In contrast to the results with Ca++, myocardial Mg++ content rose rapidly from an initial value of 3.2 mM at 48 h to 6.7 mM by the 5th day of development, and then gradually declined throughout the remaining foetal development to 4.8 mM 2-3 days post-hatching. The Mg++ contents closely paralleled changes in K+ content during development, which were reported earlier. Treatment with EGTA and EDTA removed 13-22% and 19-28% of the myocardial Mg++, respectively, during development until just prior to hatching, when only 10-12% could be removed by chelation.  相似文献

Salts at high concentrations may cause oxidative damage to plant cells since many studies indicated the involvement of reactive oxygen species in salt-stress response. Recently, we have demonstrated that treatment of tobacco ( Nicotiana tabacum ) cell suspension culture with various salts result in an immediate burst of superoxide production via activation of NADPH oxidase by ions of alkali metals (Li+, Na+, K+), alkali earth metals (Mg2+, Ca2+) or lanthanides (La3+, Gd3+). In this study, we tested the effect of extracellular supplementation of Zn2+ and Mn2+ on the cation-induced oxidative burst in tobacco cell suspension culture, measured with a superoxide-specific Cypridina luciferin-derived chemiluminescent reagent. Extracellular supplementation of Zn2+ and Mn2+ inhibited the generation of superoxide in response to addition of salts. Although both Zn2+ and Mn2+ inhibited the salt-induced generation of superoxide, the modes of inhibition by those ions seemed to be different since Mn2+ simply inhibited total production of superoxide while Zn2+ inhibited the early phase of superoxide production and induced the slow release of superoxide. Roles of Mn2+ and Zn2+ in protection of plant cells from salt stress, as an effective superoxide scavenger and an effective inhibitor of plasma membrane-bound NADPH oxidase, respectively, are discussed.  相似文献

In Escherichia coli , lacZ operon fusions were isolated that were derepressed under iron repletion and repressed under iron depletion. Two fusions were localized in genes that formed an operon whose gene products had characteristics of a binding protein-dependent transport system. The growth defect of these mutants on TY medium containing 5 mM EGTA was compensated for by the addition of Zn2+. In the presence of 0.5 mM EGTA, only the parental strain was able to take up 65Zn2+. This high-affinity transport was energized by ATP. The genes were named znuACB (for zinc uptake; former name yebLMI ) and localized at 42 min on the genetic map of E. coli . At high Zn2+ concentrations, the znu mutants took up more 65Zn2+ than the parental strain. The high-affinity 65Zn2+ uptake was repressed by growth in the presence of 10 μM Zn2+. A znuA–lacZ operon fusion was repressed by 5 μM Zn2+ and showed a more than 20-fold increase in β-galactosidase activity when Zn2+ was bound to 1.5 μM TPEN [tetrakis-(2-pyridylmethyl) ethylenediamine]. To identify the Zn2+-dependent regulator, constitutive mutants were isolated and tested for complementation by a gene bank of E. coli . A complementing gene, yjbK of the E. coli genome, was identified and named zur (for zinc uptake regulation). The Zur protein showed 27% sequence identity with the iron regulator Fur. High-affinity 65Zn2+ transport of the constitutive zur mutant was 10-fold higher than that of the uninduced parental strain. An in vivo titration assay suggested that Zur binds to the bidirectional promoter region of znuA and znuCB .  相似文献

The Mg2+ requirement in fertilization was investigated in sea urchins. It was found that when sea urchin eggs were inseminated in sea water free of Mg2+, little fertilization took place. Even when spermatozoa pre-treated with dissolved egg-jelly to induce the acrosome reaction, which needs Ca2+, were used, the fertilization rate remained quite low in the absence of Mg2+. In Strongylo-centrotus intermedius , the lowest concentration of Mg2+ required for 50% fertilization was 0.05 mM in the presence of 10 mM Ca2+, whereas that of calcium was 3 mM in the presence of 49 mM Mg2+. These critical concentrations increased when the concentration of the other ion decreased. Removal of Mg2+ or Ca2+ or both from the suspending medium had little adverse effect on sperm motility. The elevation of the fertilization membrane was also induced by butyric acid independent of the presence or absence of Mg2+ and/or Ca2+. These results indicate that Mg2+ are required at least in some process(es) between acrosome reaction and fertilization membrane elevation, such as sperm penetration or membrane fusion.  相似文献

Indispensability of Iron for the Growth of Cultured Chick Cells   总被引:1,自引:0,他引:1  
In order to clarify the role of iron in the growth promoting effect of transferrin (Tf), the effects of the following substances were examined in cultured chick skeletal myogenic cells: transition metal ions (Fe2+, Fe3+, Cr3+, Cu2+, Mn2+, Co2+, Cd2+, Zn2+ and Ni2+), Tf complexes with these metals and metal-free apoTf.
The cells did not grow well when incubated in a culture medium composed of Eagle's minimum essential medium and horse serum. But they grew well in the presence of Fe2+ or Fe3+ (10–100 μM) or iron-bound Tf (10–500 nM) in the medium. None of the transition metal ions other than iron was effective. Neither apoTf nor Tf complexes with these metals showed the growth promoting effect. The generality of the requirement of iron for cell growth was ascertained in the primary culture of other types of chick embryonic cells: fibroblasts, cardiac myocytes, retinal pigment cells and spinal nerve cells.
The results show that iron is one of the indispensable substances for cell growth and suggest that Tf protein plays a role in facilitating the transport of iron into the cells.  相似文献

Abstract— The hypothesis that the ATPase and phosphatidyhnositol (PI) kinase activities of chromaffin vesicle membranes are catalysed by same enzyme was investigated. The two activities exhibited entirely different responses to variations in Mg2+ or Mn2+ concentrations. In the presence of 1 mM ATP, maximal ATPase activity occurred with 1 mM Mg2+ while maximal PI kinase activity required 100 mM Mg2+ Similar differences were observed with Mn2+ with the exception that maximal ATPase activity occurred with 0.5 mM Mn2+ and maximal PI kinase activity occurred with 5 mM Mn2+ Mn2+ was more effective than Mg2+ in stimulating PI kinase activity at low concentrations, but at optimal concentrations of each, the maximal activity obtained with Mg2+ was 5-fold greater than the maximal activity obtained with Mn2+ The heat stabilities of the two enzymes are vastly different. At 50°C the ATPase activity of the intact membranes was stable for up to 20 min while the t l/2 of PI kinase was less than 2 min. After solubilization in Lubrol PX or at higher temperatures both enzymes were less heat stable, but PI kinase was still inactivated at a much greater rate than the ATPase. The evidence suggests that the ATPase and the PI kinase are different proteins.
The major phosphorylated product was diphosphatidylinositol and once formed, it was stable. Phosphorylation of membrane protein accounted for less than 10% of the total 32P-incorporated into chromaffin vesicles. SDS gel electrophoresis of the solubilized membranes showed the presence of at least 2 major phosphorylated high molecular weight components.  相似文献

Pyoverdine (PvdI) is the major siderophore secreted by Pseudomonas aeruginosa PAOI in order to get access to iron. After being loaded with iron in the extracellular medium, PvdI is transported across the bacterial outer membrane by the transporter, FpvAI. We used the spectral properties of PvdI to show that in addition to Fe3+, this siderophore also chelates, but with lower efficiencies, all the 16 metals used in our screening. Afterwards, FpvAI at the cell surface binds Ag+, Al3+, Cd2+, Co2+, Cu2+, Fe3+, Ga3+, Hg2+, Mn2+, Ni2+ or Zn2+ in complex with PvdI. We used Inductively Coupled Plasma-Atomic Emission Spectrometry to monitor metal uptake in P. aeruginosa : TonB-dependent uptake, in the presence of PvdI, was only efficient for Fe3+. Cu2+, Ga3+, Mn2+ and Ni2+ were also transported into the cell but with lower uptake rates. The presence of Al3+, Cu2+, Ga3+, Mn2+, Ni2+ and Zn2+ in the extracellular medium induced PvdI production in P. aeruginosa . All these data allow a better understanding of the behaviour of the PvdI uptake pathway in the presence of metals other than iron: FpvAI at the cell surface has broad metal specificity at the binding stage and it is highly selective for Fe3+ only during the uptake process.  相似文献

Abstract Release of lipopolysaccharide from E. coli was studied in the presence of sub-minimal inhibitory concentrations of ethylenediaminetetraacetic acid (EDTA). In untreated cells no release was detected with 50 mM Mg2+ in the medium, but a steady release of over 50% of the synthesized lipopolysaccharide was observed with 0.1 mM Mg2+. EDTA at MIC/8 led to a 2- to 3-fold higher release, presumably by an adjustment of the concentration of unchelated Mg2+ to a value still sustaining normal growth but giving rise to a highly unstable outer membrane. No structural difference was observed between cell-bound and released lipopolysaccharide.  相似文献

Abstract: Following incubation with [14C]y-aminobutyric acid (GABA) or [3H]dopamine, slices of rat striatum were superfused with media containing 36 mM K+ or ethylenediamine (EDA), 1 or 5 mM. Both K+ and EDA induced a release of [14C]GABA, the K+-induced release being largely Ca2+-dependent, while the EDA-induced release was not. Whereas K+ also evoked a Ca2+-dependent release of [3H]dopamine, EDA evoked no release of dopamine. EDA may therefore have potential as a specific GABA releasing agent.  相似文献

Abstract: The present study reports the ion dependency of 2β-carbomethoxy-3β-(4-fluorophenyl)[3H]tropane ([3H]- CFT) binding to the dopamine transporter in the rat striaturn. The results indicate that [3H]CFT binding to synaptosomal P2 membranes requires low concentrations of Na+ (peak binding between 20 and 50 m M Na+), is stimulated by phosphate anion or l-, but is unaffected or only slightly affected by F-, Cl-, Br-, NO3-, or SO42-, Concentrations of Na+ of >50 m M become inhibitory except in the presence of l-, which shifts peak binding levels toward higher Na+ concentrations and also elevates the peak binding level. K+ strongly decreased [3H]CFT binding with a shallow inhibition curve, and Na+ could not overcome this effect. Saturation analysis of [3H]CFT binding revealed a single binding site changing its affinity for CFT depending on the concentration of sodium phosphate buffer (6, 10, 30, 50, 130, or 200 m M ; 1 mM plus 49 mM NaCIversus 10 m M plus 40 m M NaCI; or 1 mM plus 129 m M Nal versus 10 m M plus 120 m M Nal). No differences were observed in the density of CFT binding sites between any of the conditions examined.  相似文献

Pb and Cd are heavy metal pollutants that inhibit plant growth. Using a cultivated rice variety (Dongjin, Oryza sativa L.), we studied how the transport and toxicity of Pb2+ and Cd2+ are affected by the presence of K+, Ca2+ or Mg2+. K+ had a little effect on uptake or toxicity of Pb2+ and Cd2+. Ca2+ or Mg2+ blocked both Cd2+ transport into rice roots and Cd2+ toxicity on root growth, which suggested that their detoxification effect is directly related to their blocking of entry of the heavy metals. Similarly, Ca2+ blocked both Pb2+ transport into the root and Pb2+ toxicity on root growth. The protective effect of Ca2+ on Pb2+ toxicity may be related to its inhibition of the heavy metal accumulation in the root tip, a potential target site of Pb2+ toxicity. Mg2+ did not ameliorate the Pb2+ toxicity on root growth as much as Ca2+ did, although it decreased Pb2+ uptake into roots similarly as Ca2+ did. These results suggest that the protective effect of Ca2+ on Pb2+ toxicity may involve multiple mechanisms including competition at the entry level, and that Pb2+ and Cd2+ may compete with divalent cations for transport into roots of rice plants.  相似文献

In strictly anaerobic conditions in a culture medium adjusted to pH 5·2 with HCl and incubated at 30°C, inocula containing < 10 vegetative bacteria of Clostridium botulinum ZK3 (type A) multiplied to give > 108 bacteria per ml in 3 d. Growth from an inoculum of between 10 and 100 spores occurred after a delay of 10–20 weeks. Citric acid concentrations of 10–50 mmol/l at pH 5·2 inhibited growth from both vegetative bacteria and spore inocula, a concentration of 50 mmol/l increasing the number of vegetative bacteria or of spores required to produce growth by a factor of approximately 106. The citric acid also reduced the concentration of free Ca2+ in the medium. The inhibitory effect of citric acid on vegetative bacteria at pH 5·2 could be prevented by the addition of Ca2+ or Mg2+ and greatly reduced by Fe2+ and Mn2+. The addition of Ca2+, but not of the remaining divalent metal ions, restored the concentration of free Ca2+ in the medium to that in the citrate-free medium. The inhibitory effect of citric acid on growth from a spore inoculum was only partially prevented by Ca2+. Citric acid (50 mmol/l) did not inhibit growth of strain ZK3 at pH 6 despite the greater chelating activity of citrate at pH 6 than at pH 5·2. The effect of citric acid and Ca2+ at pH 5·2 on vegetative bacteria of strains VL1 (type A) and 2346 and B6 (proteolytic type B) was similar to that on strain ZK3.  相似文献

The hydraulic conductance ( L 0) of detached, exuding root systems from melon ( Cucumis melo cv. Amarillo oro) was measured. All plants received a half-strength Hoagland nutrient solution, and plants stressed either solely with NaCl (50 mM) or with NaCl (50 mM) following treatment (2 d) with CaCl2 (10 mM) were compared with controls and CaCl2-treated (10 mM) plants. The L 0 of NaCl-treated plants was markedly decreased when compared to control and CaCl2-treated plants, but the decrease was smaller when NaCl was added to plants previously treated with CaCl2. A similar effect was observed when the flux of Ca2+ into the xylem and the Ca2+ concentration in the plasma membrane of the root cells were determined. In control, CaCl2- and NaCl + CaCl2-treated plants, HgCl2 treatment (50 μM) caused a sharp decline in L 0 to values similar to those of NaCl-stressed roots, but L 0 was restored by treatment with 5 mM DTT. However, in NaCl roots only a slight effect of Hg2+ and DTT were observed. The effect of all treatments on L 0 was similar to that on osmotic water permeability ( P f) of individual protoplasts isolated from roots. The results suggest that NaCl decreased the passage of water through the membrane and roots by reducing the activity of Hg-sensitive water channels. The ameliorative effect of Ca2+ on NaCl stress could be related to water-channel function.  相似文献

After stimulation in a hypotonic solution (9.4 mOsm kg−1), inseminated eggs of the chum salmon Oncorhynchus keta initiate cleavages in isotonic salmon Ringer's solution (267.3 mOsm kg−1) containing 3.2 mM Ca2+ ions. Blastomeres of these eggs, however, separate from each other and the enveloping layer is not observed at the blastula stage. An increase in external divalent cations rescues the separation; the concentration of CaCl2 in the external medium should be 25 mM or more to induce close contact of blastomeres and the formation of an enveloping layer in isotonic salt solutions. The effectiveness of Ca2+ ions can be substituted by Mg2+, Sr2+ and Zn2+ ions; the same results are obtained in isotonic MgCl2 and SrCl2 solutions (100 mM) or in isotonic salmon Ringer's solution containing Zn ions (6.2 mM). The close contact of blastomeres and the formation of an enveloping layer are also observed in a low Ca2+ concentration (< 0.1 mM) in a hypotonic salt solution (9.4 mOsm kg−1). The Ca2+ level in the external medium to induce the enveloping layer formation seems to be correlated with the salinity of the incubation medium. It is suggested that adhesion molecules on the surface of blastomeres in the chum salmon eggs are different in properties from those found in sea urchin and other fish species.  相似文献

Strain Escherichia coli V38 resistant to 4 mM NiCl2 was isolated from the city sewage sludge. It showed low nickel accumulation by cells and nickel ion efflux. Cells were pregrown (induced) overnight in the presence of Ni2+, then the culture was kept on ice for 20–30 min and transferred to 37°C for further incubation. When the Ni2+ concentration during growth was the same as during incubation, there was no noticeable accumulation of Ni2+. When the Ni2+ concentration during incubation was higher than that used for induction, uptake of 63Ni2+ and delayed efflux were seen. The uptake and delay of both efflux and growth were directly proportional to the difference between the concentrations used for induction and incubation. Active nickel ion uptake was seen in cells taken from cultures in the delayed efflux period.  相似文献

Reed plants ( Phragmites communis Trinius) were grown at NaCl concentrations up to 500 m M and their growth, mineral contents and leaf blade osmotic potential were determined. Addition of NaCl up to 300 m M did not affect growth significantly. Sucrose, Cl-and Na+ concentrations in the shoots increased with the salinity of the medium and the shoot water content decreased. K+ always contributed most to the leaf osmotic potential. Even in the presence of 250 m M NaCl in the rooting medium, the leaf blade contained only 50 mM Na+, suggesting that the plants have an efficient mechanism for Na+ exclusion. 22Na+ uptake experiments suggested that the retranslo-cation of absorbed Na+ from shoots to the rooting medium lowered the uptake of Na+.  相似文献

Abstract Attempts to study bacterial resistance to metal ions in broth media can be complicated by precipitation reactions. Precipitation from Luria-Bertani (LB) broth occurred at Cd2+ concentrations higher than 1.6 mM and was complete within about 10 min. The precipitate obtained after addition of 3 mM Cd2+ was analysed by elemental analysis. 1H and 31P nuclear magnetic resonance (NMR) spectroscopy. It consisted of cadmium phosphates together with organic material containing valine- and glutamate-rich polypeptides. Precipitation significantly reduced the levels of the essential trace elements Fe and Zn in the growth medium.  相似文献

Abstract Accumulation of Li+ in Saccharomyces cerevisiae X2180-1B occured via an apparent stoichiometric relationship of 1: 1 (K+/Li+) when S. cerevisiae was incubated in the presence of 5 and 10 mM LiCl for 3 h. Other cellular cations (Mg2+, Ca2+ and Na+) did not vary on Li+ accumulation, although lithium chemistry dictates a degree of similarity to Group I and II metal cations. Compartmentation of Li+ was mainly in the vacuole which accounted for 85% of the Li+ accumulated after a 6-h incubation period. The remainder was located in the cytosol with negligible amounts being bound to cell fragments including the cell wall. Transmission electron microscopy of Li+-loaded cells revealed enlarged vacuoles compared with control cells. This asymmetric cellular distribution may therefore enhance tolerance of S. cerevisiae to Li+ and ensure that essential metabolic processes in the cytosol are not disrupted.  相似文献

Abstract: The metabotropic glutamate receptor mGluR5, but not the closely related mGluR1, is expressed in cultured astrocytes, and this expression is up-regulated by specific growth factors. We investigated the capability and underlying mechanisms of mGluR5 to induce oscillatory responses of intracellular calcium concentration ([Ca2+]i) in cultured rat astrocytes. Single-cell [Ca2+]i recordings indicated that an mGluR-selective agonist, (1 S ,3 R )-1-aminocyclopentane-1,3-dicarboxylate (1 S ,3 R -ACPD), elicits [Ca2+]i oscillations in good agreement with the growth factor-induced up-regulation of mGluR5 in cultured astrocytes. A protein kinase C (PKC) inhibitor, bisindolylmaleimide I, converted a 1 S ,3 R -ACPD-mediated oscillatory response into a nonoscillatory response. In addition, the PKC activator phorbol 12-myristate 13-acetate completely abolished the [Ca2+]i increase. These and other pharmacological properties of 1 S ,3 R -ACPD-induced [Ca2+]i oscillations correlate well with those of the cloned mGluR5 characterized in heterologous expression systems. Furthermore, the potential involvement of protein phosphatases in [Ca2+]i oscillations is suggested. The present study demonstrates that mGluR5 is capable of inducing [Ca2+]i oscillations in cultured astrocytes and that phosphorylation/dephosphorylation of mGluR5 is critical in [Ca2+]i oscillations, analogous to the cloned mGluR5 expressed in heterologous cell lines.  相似文献

Effects of mixtures of chloride salts of cadmium, copper and zinc on survival, whole body residues, and histopathology of mummichog, Fundulus heteroclitus (L.), were investigated in synthetic sea water at 20‰ salinity and 20°C. Mixtures of Cu2+ and Zn2+ as indicated by 96 h bioassay studies produced more deaths than expected on the basis of toxicities of individual components. Concentrations of Cd2+ not ordinarily lethal exerted a negative effect on survival of fish intoxicated by salts of copper, zinc, or both.
Atomic absorption determinations of Cd, Cu, and Zn residues in mummichog which survived 96 h exposures to each of these toxicants provided useful indices of total body burdens for these metals. Residues from survivors held in mixtures, especially Cd2+ and Zn2+ mixtures, did not conform to patterns observed for single elements. Whole body aggregates of Cd, Cu, and Zn from dead mummichogs were of limited worth owing to possible accumulation of these metals from the medium after death.
Renal and lateral line canal lesions were noted in all fish subjected to copper concentrations of 1 mg/1 and higher. Renal lesions observed in fish immersed in mixtures of Cu2+ and Cd2+ assumed a damage pattern characteristic of Cd2+; with mixtures of Cu2+ and Zn2+, lesion were typical of Cu2+-induced damage. Lesions induced in lateral line epithelium by Cu2+ were not affected by either Cd2+ or Zn2+. Epithelia lining the oral cavity were necrotized by the caustic action of high levels of Zn2+ (60 mg/1) and of Cu2+ (8 mg/1).  相似文献
