Schisandra chinensis (Turcz.) Baill. and Schisandra sphenanthera Rehd. et Wils. are well-known Chinese medicinal plants. The population genetic variation of the two species was studied using inter simple sequence repeat (ISSR) molecular markers. High levels of genetic diversity are revealed in both S. chinensis (P = 88.36%, h = 0.2894, I = 0.4396) and S. sphenanthera (P = 84.09%, H = 0.2782, I = 0.4280). However, the population genetic differentiation is significantly different between the two species. The S. sphenanthera harbors as high as 27% of the genetic variation among populations but 73% within populations, whereas in S. chinensis 17% of the genetic variation occurs among populations and 83% within populations. Both significant (P < 0.05) heterozygosity excess and shifted mode of allele frequency distribution are detected in four out of six populations of S. chinensis and one out of five populations of S. sphenanthera, suggesting the occurrence of recent population bottlenecks in the two species. The different patterns of genetic variation in S. chinensis and S. sphenanthera are discussed in relation to their differences in pollination mechanism, geographic distribution and historical events, and the level of gene flow and genetic drift.  相似文献

In this study, the herbal extracts of Schisandra chinensis were demonstrated to inhibit the contractions induced by acetylcholine (ACh) and serotonin (5-HT) in guinea pig ileum, and the 95% ethanol extract was more effective than the aqueous extract. Analysis with High Performance Liquid Chromatography (HPLC) indicated that schisandrin, schisandrol B, schisandrin A and schisandrin B were the major lignans of Schisandra chinensis, and the ethanol extract contained higher amount of these lignans than the aqueous extract. All four lignans inhibited the contractile responses to ACh, with EC20 values ranging from 2.2 ± 0.4 μM (schisandrin A) to 13.2 ± 4.7 μM (schisandrin). The effectiveness of these compounds in relaxing the 5-HT-induced contraction was observed with a similar magnitude. Receptor binding assay indicated that Schisandra lignans did not show significant antagonistic effect on muscarinic M3 receptor. In Ca2+-free preparations primed with ACh or KCl, schisandrin A (50 μM) attenuated the contractile responses to cumulative addition of CaCl2 by 37%. In addition, schisandrin A also concentration-dependently inhibited ACh-induced contractions in Ca2+-free buffer. This study demonstrates that Schisandra chinensis exhibited relaxant effects on agonist-induced contraction in guinea pig ileum, with schisandrin, schisandrol B, schisandrin A and schisandrin B being the major active ingredients. The antispasmodic action of schisandrin A involved inhibitions on both Ca2+ influx through L-type Ca2+ channels and intracellular Ca2+ mobilization, rather than specific antagonism of cholinergic muscarinic receptors.  相似文献

Twenty Schisandra samples were collected from different locations. Contents of 7 lignans in the samples were determined and analyzed by HPLC method coupled with hierarchical clustering analysis (HCA) and principal component analysis (PCA), and the antioxidant capacity of Schisandra from the different locations was evaluated by reducing power, ferric thiocyanate (FTC) and 2,2′‐diphenyl‐1‐picrylhydrazyl (DPPH) assay. The results showed that there was a significant difference in the content of lignans between Schisandra chinensis and Schisandra sphenanthera. The Schisandra sphenanthera samples in the southwest of China were significantly different from those from the other locations. The antioxidant capacity of Schisandra chinensis was significantly superior to that of Schisandra sphenanthera, and the main antioxidant components were schisandrol A, schisandrol B and schisandrin B based on the result of discrimination analyses. The differences in the chemical composition and antioxidant activity of lignans in Schisandra chinensis and Schisandra sphenanthera from the different locations were investigated in this study, which may provide an experimental basis for the quality control of Schisandra.  相似文献

Seven polymorphic and transferable nuclear microsatellites were used to investigate the population structure of genetic diversity of Schisandra chinensis and Schisandra sphenanthera for facilitating their conservation and sustainable utilization. High levels of gene diversity were revealed in these two medicinal species, the majority of genetic diversity was harbored within populations, and population structure was might due to restricted gene flow among populations. Isolation by distance was close to significance in S. chinensis but not in S. sphenanthera. In S. chinensis, null alleles were identified as a cause for excess of homozygotes at loci G24 and WGA60, but inbreeding might also be partly responsible for the positive F IS values in this species. In contrast, null allele frequencies were high at all the seven loci in S. sphenanthera and resulted in overestimation of fixation index. The strategy for ex situ conservation of these two medicinal species is discussed based on the genetic results.  相似文献

Schisandra chinensis has been regarded as a useful material in the preventive or treatment of several human diseases. The study of the Schisandra chinensis’ molecular identity has been prioritized because it has been found to regulate cellular responses. Here, we examined the effects from various extracts of Schisandra chinensis to monitor the relative intracellular calcium increase in human neutrophils. We identified an active molecule and structural configuration of a new active compound (α-iso-cubebene), based on the discovery of a cubebene backbone using NMR, MS, and CD spectral data. In terms of its functional aspect, we observed that α-iso-cubebene strongly stimulated CXCL8 production in human neutrophils. Also, α-iso-cubebene-induced CXCL8 production was almost completely inhibited by the calcium chelator, EGTA, thus highlighting the role of calcium signaling in the process. Taken together, our results demonstrate that α-iso-cubebene is a novel natural compound which stimulates intracellular calcium signaling and CXCL8 production. As a result, α-iso-cubebene should be useful for the development of an immune-modulating agent.  相似文献

The squids Uroteuthis (Photololigo) edulis and Uroteuthis (Photololigo) chinensis (family Loliginidae) are commercially important fishery species in many coastal regions of Asia. The morphologies of these two squids are very similar, and identification based on morphology has been inadequate. The occurrence of cryptic species in the family Loliginidae has been reported. The widely distributed U. (P.) chinensis and U. (P.) edulis are believed to comprise several cryptic species. In this study, the taxonomic status of the two species in East Asia was elucidated by morphological and genetic analyses. Analysis of U. (P.) chinensis from Hong Kong and Xiamen (China) and U. (P.) edulis from Yamaguchi (Japan) and Shanghai (China) was performed in order to determine the effectiveness of different morphometric variables in discriminating between the two species. Multivariate analysis of 27 morphometric indices revealed no new morphological characters for the taxonomic identification of the two taxa, which can be distinguished by the teeth shape and number on arm sucker rings, and the percentage of hectocotylized part of left arm IV of males. The morphometric differences between U. (P.) edulis individuals from the two localities is most probably due to differences in the maturity stages of the sampled individuals between the two localities. Genetic analysis based on the mitochondrial COI and 16S rRNA genes revealed a high divergence of 15.5% and 7.5% respectively, indicating that U. (P.) edulis and U. (P.) chinensis are distinct species.  相似文献

Schisandra chinensis (Turcz.) Baill. is a climbing woody vine found over a wide geographical range in northeast Asia. Its red five-flavored fruits have been used as drugs and food. In China and Korea, the dried fruit of S. chinensis has been used for a long time as a traditional medicine. Three species of the family Schisandraceae, S. chinensis, S. repanda, and Kadsura japonica, are distributed mainly in southern Korea. While S. chinensis is broadly distributed, S. repanda and K. japonica are restricted to the southern islands of Korea, especially Jeju. The amplification refractory mutation system (ARMS), one of molecular genotyping technique, is a simple and rapid method for detecting point mutations, restriction fragment length polymorphisms, and small deletions or insertions in a DNA sequence. This PCR-based technique utilizes a primer designed to react only when it anneals to specific target sequences in a DNA sample. In the present study, we analyzed rbcL of the chloroplast gene and ITS from S. chinensis and two related taxa, S. repanda and K. japonica, and developed a specific marker for distinguishing S. chinensis from the related taxa based on SNPs using multiplex PCR. Result showed that two species-specific fragments with 230 bp of rbcL and 278 bp of ITS region were additionally amplified only in all of individuals of S. chinenesis using multiplex PCR method. Here, we describe the details of the protocol for the molecular identification of S. chinensis and other related species. Our results cover five populations of S. chinensis in Korea.  相似文献

A novel α-iso-cubebenol, which has anti-inflammatory effects in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages, was isolated from the fruits of Schisandra chinensis. α-iso-cubebenol inhibited LPS-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production. Consistent with these findings, α-iso-cubebenol also reduced the LPS-induced expression of inducible nitric oxide synthase and cyclooxygenase-2 at the protein and mRNA levels in a concentration-dependent manner. α-iso-cubebenol also inhibited LPS-induced nuclear translocation of the NF-κB p65 subunit. Furthermore, α-iso-cubebenol suppressed the phosphorylation of ERK, JNK, and p38 kinase induced by LPS. Since the novel α-iso-cubebenol blocked the production of several pro-inflammatory mediators induced by LPS in macrophages, the molecule can be useful material for the development of anti-inflammatory agents against bacterial infections or endotoxin.  相似文献

Schisandra sphenanthera (Schisandraceae) is a traditional Chinese medicinal plant. In this study, nine polymorphic microsatellite loci were developed and characterized for the species. A genomic DNA enrichment protocol was used to isolate microsatellite loci and polymorphism was explored using 36 individuals sampled from a natural population. The observed number of alleles ranged from 2 to 10 with an average of 5.2. Expected and observed heterozygosity ranged from 0.30 to 0.82 and 0.33 to 0.97, respectively. These microsatellites have been directly applied to both population and conservation genetic studies of S. sphenanthera.  相似文献

Schisandra chinensis (Turz Baill) (S. chinensis) (SC) fruit is a hepatoprotective herb containing many lignans and a large amount of polysaccharides. A novel polysaccharide (called SC-2) was isolated from SC of MW 841 kDa, which exhibited a protein-to-polysaccharide ratio of 0.4089, and showed a characteristic FTIR spectrum of a peptidoglycan. Powder X-ray diffraction revealed microcrystalline structures within SC-2. SC-2 contained 10 monosaccharides and 15 amino acids (essential amino acids of 78.12%w/w). In a HepG2 cell model, SC-2 was shown by MTT and TUNEL assay to be completely non-cytotoxic. A kinetic analysis and fluorescence-labeling technique revealed no intracellular disposition of SC-2. Combined treatment of lignans with SC-2 enhanced the intracellular transport of schisandrin B and deoxyschisandrin but decreased that of gomisin C, resulting in alteration of cell-killing bioactivity. The Second Law of Thermodynamics allows this type of unidirectional transport. Conclusively, SC-2 alters the transport and cell killing capability by a “Catcher-Pitcher Unidirectional Transport Mechanism”.  相似文献

分别于2006年7月、10月和2007年4月,对全光照、林缘和林下3种光环境下夏蜡梅及群落中35种主要植物叶的灰分含量和去灰分热值进行测定,分析它们在光合作用中对太阳能的固定和积累能力。不同光环境下夏蜡梅叶灰分含量的大小顺序都是全光照>林缘>林下,10月3种光环境下夏蜡梅叶灰分含量均高于4月和7月,均差异显著。4月、7月和10月,林下和全光照环境下的叶去灰分热值呈先下降后上升的趋势,而林缘中叶去灰分热值却逐渐下降。群落各层植物的灰分含量大小顺序为草本层>灌木层>间层>乔木层,夏蜡梅灰分含量略低于灌木层灰分含量的平均值;各层次植物平均去灰分热值大小顺序为乔木层>灌木层>草本层>间层,夏蜡梅平均去灰分热值低于间层。多数植物不同季节叶的灰分含量、去灰分热值均以10月最高。  相似文献

米氏冰草和羊草都是根茎型禾草,是典型的无性系植物。采取单位面积挖掘取样法,对呼伦贝尔沙地植被次生演替过程中的米氏冰草和羊草种群分株年龄结构进行调查。结果表明,在单生和混合群落中,米氏冰草分株分别由3和4个年龄级组成,羊草均由2个年龄级组成;米氏冰草分株数量和生物量年龄结构变化基本一致,即随着生长年限的延长,呈减少低龄级和增加高龄级比例的趋势,使单生群落中始终为增长型,但接近于稳定型,混生群落中由增长型变为稳定型。羊草分株数量和生物量年龄结构在连续2a均为明显的增长型。羊草各龄级平均单株生产力均高于米氏冰草,平均是米氏冰草的5.2倍。米氏冰草和羊草种群分株年龄结构的变化,蕴含着米氏冰草种群的优势地位将被羊草种群取代的趋势。  相似文献

Schisandra chinensis (Turcz.) Baill. is one of the important traditional medicinal plants in East Asia. It is a dioecious plant with aromatic flowers. The female and male flowers of S. chinensis possess slightly different fragrance characteristics. The overall scent of S. chinensis flowers is quite similar to that of Syringa dilatata (Korean lilac) flowers. Hence, this study aimed to understand the aromatic profile of the hexane extract from female and male flowers of S. chinensis and to compare their profile with the hexane extract of Korean lilac flowers. The chemical composition of hexane extract was determined by gas chromatography and mass spectrometry (GC-MS) analysis. In total, 67 different components were detected in the hexane extract of female (48) and male flowers (51) of S. chinensis; 32 of which were common to both female and male flowers. In regards to gender difference, 16 components were found only in female flowers, and 19 components were found only in male flowers. The results revealed that the most abundant components in the hexane extract of both female and male flowers were lilac alcohol C (9.53 and 7.00%), lilac alcohol A (6.55 and 5.71%), n-hexadecanoic acid (6.21 and 6.96%), linoleic acid (5.14 and 7.61%), β-elemene (5.12 and 1.99), and lilac aldehyde D (4.13 and 4.97%). The data suggest that the major compounds in the hexane extract of S. chinensis flowers were generally similar, but they varied quantitatively according to gender. The presence of 10 components in both S. chinensis and Korean lilac flowers may be responsible for their similar fragrance characteristics. It could be concluded that the different fragrance characteristics of these flowers may be due to the presence of several gender-specific aromatic compounds in minor percentages.  相似文献

BackgroundParidis Rhizoma (PR) is a famous traditional herbal medicine. Apart from two officially recorded species, viz. Paris polyphylla Smith var. yunnanensis (Franch.) Hand. - Mazz. (PPY) and P. polyphylla Smith var. chinensis (Franch.) Hara (PPC), there are still many other species used as folk medicine. It is necessary to understand the metabolic differences among Paris species.PurposeTo establish a strategy that can discover species-specific steroidal saponin markers to distinguish closely-related Paris herbs for quality and safety control.MethodsA new strategy of molecular-networking-guided discovery of species-specific markers was proposed. Firstly, the ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) was applied to obtain the MS and MS/MS data of all samples. Then, molecular networking (MN) was created using MS/MS data to prescreen the steroidal saponins for subsequent analysis. Next, the principal component analysis (PCA) and orthogonal partial least square discriminant analysis (OPLS-DA) models were established to discover potential markers. Finally, the verification, identification and distribution of chemical markers were performed.ResultsA total of 126 steroidal saponins were screened out from five species using MN. Five species were classified successfully by OPLS-DA model, and 18 species-specific markers were discovered combining the variable importance in the projection (VIP) value, P value (one-way ANOVA) and their relative abundance. These markers could predict the species of Paris herbs correctly.ConclusionThese results revealed that this new strategy could be an efficient way for chemical discrimination of medicinal herbs with close genetic relationship.  相似文献

Separate benzocyclooctadiene lignans were isolated from the berries of Schisandra chinensis in milligram quantities on analytical reverse phase (RP) HPLC by an automated repeat-injection method and shown to have anti-proliferative activity against human colorectal cancer cells. Structures of the compounds were determined by a combination of NMR and mass spectrometry. Stereospecific NMR assignments for gomisin-N and deoxyschisandrin, gave more complete and accurate data than previously reported, based on 600 MHz 2D HSQC, DQF-COSY and HMBC data. Comparison of coupling constants and HMBC crosspeak intensities with calculated and X-ray crystal structures confirmed their stereochemistry and conformation. Analysis of structure–activity relationships revealed the importance of key structural determinants. The S-biphenyl configuration of gomisin N, the most active lignan, correlated with increased anti-proliferative activity, while the presence of a hydroxyl group at the C7 position reduced or abolished this activity. Increased activity was also observed when a methylenedioxy group was present between C12 and C13. The percent yield of the most active compounds relative to the starting plant materials was 0.0156% for deoxyschisandrin and 0.0173% for gomisin N. The results of these studies indicate that automated repeat-injection method of analytical HPLC may provide a superior alternative to the standard semi-preparative HPLC techniques for separation of complex mixtures.  相似文献

在夏蜡梅(Sinocalycanthus chinensis)主要分布地设置11个代表性样地,研究了种群结构与空间分布格局,并对种群密度与主要环境因子进行相关性分析。结果表明,大多数夏蜡梅种群幼年阶段的个体较丰富,种群自然更新良好;样地群落夏蜡梅以集群分布为主;在透光率低、林分郁闭的群落中夏蜡梅聚集强度高,但种群更新不良、幼苗缺乏。因此,夏蜡梅适合在相对开敞的遮阴环境下生长,过于郁闭林地应采取适度间伐上层乔木、疏除过密灌木的抚育保护措施。  相似文献

Glechoma longituba is a widely used traditional Chinese medicine (TCM) in treating various diseases; however, the in vivo integrated metabolism of its multiple bioactive components remains unknown. In this paper, ultra-performance liquid chromatography (UPLC) coupled to quadrupole time-of-flight (QTOF) and the MetaboLynx™ software combined with mass defect filtering (MDF) together provide unique high throughput capabilities for drug metabolism study, with excellent MS mass accuracy and enhanced MSE data acquisition. This rapid automated analysis method was successfully applied for screening and identification of the constituents absorbed and metabolized studies of G. longituba extract after oral administration to rats. The results showed that 21 parent components of G. longituba extract were absorbed into the blood circulation of the rats and a total of 80 metabolites of 9 parent compounds were tentatively detected in vivo by their MS spectra obtained at low or high collision energy scan with the comparison of the authentic standards and literature data. The developed method was simple and reliable, revealing that it could be used to rapid screen and identify the structures of active components responsible for pharmacological effects of G. longituba and to better clarify its action mechanism. This work suggests that the integrative metabolism approach makes a useful template for drug metabolism research of TCM.  相似文献

Many researchers have studied the potential medicinal properties of galls from Rhus chinensis because of the importance of these galls in East Asian traditional medicine. Gall formation induced by a parasitic aphid species (Schlechtendalia chinensis) occurs via a well-documented developmental progression, and traditional medicinal efficacy is thought to be maximal during a specific portion of this cycle. To investigate seasonal changes of metabolites in the galls of R. chinensis, we collected samples from the galls and leaves of R. chinensis at sites in Mt. Jiri and Mt. Cheonma in Korea between May and December, 2011. Samples were extracted and analyzed gas chromatography mass spectrometry (GC-MS) and liquid chromatography quadrupole time-offlight mass spectrometry (LC-QTOF-MS) to monitor metabolic changes. Multivariate analyses such as principle components analysis (PCA) and orthogonal partial least square discriminant analysis (OPLS-DA) were used to find patterns in metabolite profile changes and the responsible substances for seasonal fluctuations. LC-QTOF-MS analyses showed differences of metabolites in same organisms depending on seasons, locations, and biological interactions. Additional GC-MS analyses identified approximately 28 metabolites including sugars, amino acids, and organic acids. Shikimic acid and gallic acid appear to be the major compounds contributing to the seasonal variability in metabolic profiles of R. chinensis leaves and galls. In addition, we found that shikimic acid and gallic acid content in R. chinensis galls were the highest during wintertime.  相似文献

Taxus chinensis var. mairei (Lemée et Lévl) Cheng et L.K. Fu is an evergreen tall tree ubiquitous to the southeastern region in China. The first chemical study on this species was published in 1987. Since then about 163 compounds including taxoids and non-taxane compounds were isolated from seeds, root, bark and leaves of this species. This review summarized the chemical investigation on T. chinensis var. mairei (Lemée et Lévl) Cheng et L.K. Fu in the past 20 years. T. chinensis var. mairei (Lemée et Lévl) Cheng et L.K. Fu was assigned as a variation of T. chinensis (Pilger) Rehd. in Flora of China. However, present chemotaxonomic data would suggest that these two plants might belong to two different chemotypes; further genetic investigation and comprehensive chemical studies on them are warranted to address this issue.  相似文献
