Levels of testosterone (T) and LH in the peripheral serum of male and female rabbits were measured and compared following coitus. Blood was collected by heart puncture from restrained, unanesthetized animals of both sexes. In male rabbits, basal serum T levels were highly variable, ranging from 131 to 12,149 pg/ml, and if low preceding coitus they tended to rise; whereas, if high, they usually dropped as they did in nonmated males subjected to repeated heart punctures. In contrast, basal serum LH levels in males were quite constant (mean +/- SE, 1993 +/- 152 pg/ml) and were not significantly altered after coitus unless blood T levels had been drastically lowered by two priming doses of estradiol benzoate. In intact does, on the other hand, copulation which resulted in ovulation induced an approximately 20-fold increase in serum LH concentration which was sustained for about 4 hr. Postcoital elevations in serum LH also occurred in estrogen-primed intact and estrogen-primed ovariectomized does. Under the conditions of our experiments, the parallel elevations in serum LH and T observed postcoitally in the female rabbit could not be demonstrated in the male.  相似文献

The hypothalamic-pituitary-gonadal (HPG) axis and the somatotropic axis are influenced by nutritional factors. Calorie restriction (CR) extends lifespan but suppresses both the HPG and the somatotropic axes. Since most CR studies use a fairly severe (40%-60%) reduction of calorie intake, we hypothesized that a milder CR (20%) might not be deleterious to reproduction in male mice. To test this hypothesis, we evaluated the effects of 20% CR on testicular testosterone content and on testicular expression of genes that are relevant to testicular function and reproductive competence, including insulin-like growth factor-I, cytochrome P450 aromatase (Cyp19a1), androgen receptor, luteinizing hormone receptor, follicle-stimulating hormone receptor, cytochrome P450c17 and 3-beta-hydroxysteroid dehydrogenase/isomerase. To relate CR effects to the activity of the somatotropic axis, we have used growth hormone-resistant GHR knockout mice as well as transgenic mice overexpressing GH. Mild CR did not affect testosterone levels in testis homogenates and had little effect on expression of the examined genes in the reproductive organs. Altered activity of the GH/insulin-like growth factor-1 axis had a major impact on the parameters analyzed. The results also suggest that expression of several key genes involved in the control of testicular function is preserved under conditions of mild CR and encourage speculation that mild regimens of CR can produce longevity benefits without impairing reproduction.  相似文献

The effect of successive HCG or Gonadoreline injections on some reproductive parameters in 21 young male rabbits, aged from 4 to 4.5 months, has been studied. Animals were randomly allocated in three groups of seven to receive weekly (on each Monday) i.m. injections of 20 mg Gonadoreline, 50 IU HCG, or 0.5 ml Saline solution, respectively. The ejaculate from each male was collected and analysed twice a week (Tuesday and Friday). Higher sperm volumes were observed on the first week after Gonadoreline administration compared to the other groups (p < 0.05), and lower values during the second week in the Control group vs. Gonadoreline group (p < 0.05). Sperm volume increases after the 3rd week in Control animals (p < 0.05). When considering the global period studied, mean sperm volumes achieved after Gonadoreline treatment were higher than in HCG or Control group. Despite the random distribution of the animals to each treatment, and although throughout the experiment more ejaculates were discarded in the Gonadoreline group, the final number of doses obtained per ejaculate was higher in this group followed by HCG treated animals. Globally, the mean sperm concentration of the Control group during the entire studied period was significatively lower compared to the other groups (p < 0.001). No sperm motility differences were detected among the groups (p < 0.1174). HCG injections caused a significative increase in plasma testosterone concentration during the first and second week (p < 0.001) and similar plasma levels were observed in all groups afterwards. No direct plasma testosterone increasing levels owed to Gonadoreline injections were detected, probably due to the frequency of blood sampling (once a week).  相似文献

Plasma LH and testosterone levels did not differ significantly between high and low fertility rams before or after sexual stimulation (ejaculation or teasing). Repeated stimulation caused significant elevation of mean plasma LH and an almost significant rise in testosterone concentration. Plasma testosterone peaks followed those of LH after 30-60 min. A single sexual stimulation did not always cause an LH peak or increase the mean LH level.  相似文献

Three social groups of laboratory-housed talapoin monkeys (Miopithecus talapoin) consisting of four adult males and four or five adult females, were observed over a 4-year period. All females were ovariectomized and given estradiol implants at intervals to render them sexually attractive; except for two castrated males with testosterone implants, all males were intact. Sexual and aggressive interactions were recorded, and testosterone levels were measured in plasma taken from males twice weekly. In each group males formed a linear dominance order, defined in terms of the direction of aggression between animals. The hormonal responses of intact males were monitored with respect to the presence of attractive females, access to these females, and transfer from the social group to isolation. In all groups the behavioral and endocrine responses of males to these treatments were rank related. In some instances, rising in rank was associated with elevated testosterone and falling in rank with decreased testosterone; these hormonal changes were associated with changes in sexual and aggressive interactions. The effects of sexual and aggressive behavior on plasma testosterone titers are discussed.  相似文献

We examined the affect of handling on reproductive signal expression in male threespine stickleback, Gasterosteus aculeatus. Spectrophotometric techniques have become popular methods for measuring fish color, but the measurements require handling of the subjects in order to make measurements. Fish can undergo rapid physiological color change in response to stress. As such, handling fish may induce color changes and measured colour may not be representative of signal appearance under non-stressful conditions. We measured the reflectance characteristics of the opercular, ventro-lateral and dorso-lateral areas of 41 reproductive males from Hotel Lake, BC, Canada using spectroradiometry immediately upon capture and again at 2, 3, 4, 5, 10, 15, and 20 min post-capture. Repeated measures ANOVA revealed that post-capture handling did not affect opercular or ventro-lateral reflectance characteristics over the 20 min period. However, we did observe a change in dorso-lateral reflectance characteristics of parental males over the experiment. The results of our study show that handling does not influence reflective properties of two key components of stickleback reproductive signals during a time frame that is reasonable for taking field measurements of signal color.  相似文献

It has been observed for several years that males of many mammalian species exhibit higher prevalence and intensity of parasitic infections than females. It has been theorized that this sexual dichotomy may be due to the immunosuppressive properties of testosterone. The administration of exogenous testosterone to laboratory animals often results in higher yields of several parasitic nematodes, lending credence to this hypothesis. We sought to determine if worm burdens in individual mice were related to serum testosterone concentrations. We found that in a cage of five male mice, one or two individuals exhibited high circulating testosterone levels, while the rest had low to undetectable levels. Serum testosterone did not correlate with worm recoveries. The implications of these findings are twofold. First, our data suggest that high endogenous testosterone does not necessarily decrease the ability of mice to resist nematode infection and second, that results obtained from administration of exogenous testosterone should be interpreted with caution.  相似文献

The role of ANG II in the arterial baroreflex control of renal sympathetic nerve activity (RSNA) in eight term-pregnant (P) and eight nonpregnant (NP) conscious rabbits was assessed using sequential intracerebroventricular and intravenous infusions of losartan, an AT1 receptor antagonist. The blood pressure (BP)-RSNA relationship was generated by sequential inflations of aortic and vena caval perivascular occluders. Pregnant rabbits exhibited a lower maximal RSNA reflex gain (-44%) that was primarily due to a reduction in the maximal sympathetic response to hypotension (P, 248 +/- 20% vs. NP, 357 +/- 41% of rest RSNA, P < 0.05). Intracerebroventricular losartan decreased resting BP in P (by 9 +/- 3 mmHg, P < 0.05) but not NP rabbits, and had no effect on the RSNA baroreflex in either group. Subsequent intravenous losartan decreased resting BP in NP and further decreased BP in P rabbits, but had no significant effect on the maximal RSNA reflex gain. ANG II may have an enhanced role in the tonic support of BP in pregnancy, but does not mediate the gestational depression in the arterial baroreflex control of RSNA in rabbits.  相似文献

We exposed rats to circularly polarized 50 Hz magnetic fields to determine if plasma testosterone concentration was affected. Previous experiments indicate that magnetic fields suppress the nighttime rise in melatonin, suggesting that other neuroendocrine changes might occur as well. Male Wistar-King rats were exposed almost continuously for 6 weeks to magnetic flux densities of 1,5, or 50 μT. Blood samples were obtained by decapitation at 12:00 h and 24:00 h. Plasma testosterone concentration showed a significant day-night difference, with a higher level at 12:00 h when studied in July and December, but the day-night difference disappeared when concentrations were studied in April. In three experiments, magnetic field exposure had no statistically significant effect on plasma testosterone levels compared with the sham-exposed groups. These findings indicate that 6 weeks of nearly continuous exposure to circularly polarized, 50 Hz magnetic fields did not change plasma testosterone concentration in rats. © 1994 Wiley-Liss, Inc.  相似文献

Total and free plasma testosterone (T) were determined in male homosexuals. While total T showed no significant difference to a heterosexual control group, apparently free T in plasma was significantly lower (p less than 0.001) in 35 male homosexuals (mean +/- SD = 10.7 +/- 3.3 ng/100 ml) than in 38 male heterosexuals of similar age (13.3 +/- 4.5 ng/100 ml).  相似文献

Opioid peptides have been implicated in the mediation of intermale conflict. The effect of naloxone upon tube restraint-induced target biting was investigated since a high correlation between bite scores and intermale aggression has been claimed. No significant modification of target biting behaviour by naloxone was produced, in mice either naive or habituated to the apparatus (high and low baselines of responding respectively) suggesting no involvement of opioid peptides in this paradigm. The results are discussed in terms of controllability of the presumably stressful situation and the relation of restraint-induced biting to conspecific aggression.  相似文献

Patients with either Cushing's or Nelson's syndrome, both of which are associated with markedly increased levels of beta-endorphin immunoreactivity in plasma, were given tests of pain sensitivity, mood, and cognition following administration of 10 mg naloxone and saline on separate days. No differences were found between naloxone and saline administrations on any of these psychological measures. These results suggest that immunoreactive beta- endorphin in plasma does not exert opiate effects.  相似文献

Blood samples were withdrawn every 20 min from 3 conscious intact and 2 castrated mature males during non-consecutive periods of 12 h during the light and dark phases of the lighting schedule (intact dogs) and of 11 h during the light period (castrated dogs). In the intact dogs testosterone concentrations ranged from 0.4 to 6.0 ng/ml over the 24-h period. LH concentrations varied from 0.2 to 12.0 ng/ml. In all animals, LH peaks were clearly followed, after about 50 min, by corresponding testosterone peaks, but no diurnal rhythm could be established. LH concentrations in the castrated dogs were high (9.8 +/- 2.7 (s.e.m.) ng/ml), and still showed an episodic pattern in spite of the undetectable plasma testosterone levels.  相似文献

Although evidence is accumulating that prenatal testosterone (T) compromises reproductive function in the female, the effects of excess T in utero on the postnatal development of male reproductive function has not been studied. The aim of this study was to assess the influence of prenatal T excess on age-related changes in pituitary and gonadal responsiveness to GnRH in the male sheep. We used the GnRH agonist, leuprolide (10 microg/kg), as a pharmacologic challenge at 5, 10, 20 and 30 weeks of age. These time points correspond to early and late juvenile periods and the prepubertal and postpubertal periods of sexual development, respectively. LH and T were measured in blood samples collected before and after GnRH agonist administration. The area under the response curve (AUC) of LH increased progressively in both controls and prenatal T-treated males from 5 to 20 weeks of age (P<0.01). The LH responses in prenatal T-treated males were lower at 20 and 30 weeks of age compared to controls (P<0.05). AUC-T increased progressively in control males from 5 through 30 weeks of age and prenatal T-treated males from 5 to 20 weeks of age. The T response in prenatal T-treated males was higher at 20 weeks compared to controls of same age but similar to controls and prenatal T-treated males at 30 weeks of age (P <0.05). Our findings suggest that prenatal T treatment advances the developmental trajectory of gonadal responsiveness to GnRH in male offspring.  相似文献

Astrocytes are considered the third component of the synapse, responding to neurotransmitter release from synaptic terminals and releasing gliotransmitters--including glutamate--in a Ca(2+)-dependent manner to affect neuronal synaptic activity. Many studies reporting astrocyte-driven neuronal activity have evoked astrocyte Ca(2+) increases by application of endogenous ligands that directly activate neuronal receptors, making astrocyte contribution to neuronal effect(s) difficult to determine. We have made transgenic mice that express a Gq-coupled receptor only in astrocytes to evoke astrocyte Ca(2+) increases using an agonist that does not bind endogenous receptors in brain. By recording from CA1 pyramidal cells in acute hippocampal slices from these mice, we demonstrate that widespread Ca(2+) elevations in 80%-90% of stratum radiatum astrocytes do not increase neuronal Ca(2+), produce neuronal slow inward currents, or affect excitatory synaptic activity. Our findings call into question the developing consensus that Ca(2+)-dependent glutamate release by astrocytes directly affects neuronal synaptic activity in situ.  相似文献

In social species with low rates of direct male competition levels of corticosteroids should not correlate with social status. Male spotted hyenas acquire social status by observing strict queuing conventions over many years, and thus levels of male-male aggression are low, and male social status and tenure are closely correlated. In this study, we investigated whether the low rate of direct male competition in spotted hyenas was reflected in fecal corticosteroid levels of adult males in the Serengeti National Park. Also, interactions with dominant females may influence corticosteroid levels of males, and it has been suggested recently that males with a long tenure (high rank) are more stressed by females than males with a short tenure (low rank). We tested whether there is a difference in the likelihood of being aggressively challenged by dominant females between long-tenured and short-tenured males. Short-tenured males were more likely to elicit an aggressive response by females than long-tenured males, but previous work suggests that they also interacted less frequently with females, thus avoiding putting themselves in a potentially stressful situation. Thus, as expected, the comparison of males in three different clans revealed no correlation between social status or tenure and fecal corticosteroid levels. However, males of the largest clan had the highest levels of fecal corticosteroids, possibly reflecting higher rates of social interactions in larger clans.  相似文献

Testosterone plays an important role in territorial behavior of many male vertebrates and the Challenge Hypothesis has been suggested to explain differences in testosterone concentrations between males. For socially monogamous birds, the challenge hypothesis predicts that testosterone should increase during male–male interactions. To test this, simulated territorial intrusion (STI) experiments have been conducted, but only about a third of all bird species investigated so far show the expected increase in testosterone. Previous studies have shown that male black redstarts (Phoenicurus ochruros) do not increase testosterone during STIs or short-term male–male challenges. The aim of this study was to evaluate whether black redstarts modulate testosterone in an experimentally induced longer-term unstable social situation. We created social instability by removing males from their territories and compared the behavior and testosterone concentrations of replacement males and neighbors with those of control areas. Testosterone levels did not differ among replacement males, neighbors and control males. Injections with GnRH resulted in elevation of testosterone in all groups, suggesting that all males were capable of increasing testosterone. We found no difference in the behavioral response to STIs between control and replacement males. Furthermore, there was no difference in testosterone levels between replacement males that had expanded their territory and new-coming males. In combination with prior work these data suggest that testosterone is not modulated by male–male interactions in black redstarts and that testosterone plays only a minor role in territorial behavior. We suggest that territorial behavior in species that are territorial throughout most of their annual life-cycle may be decoupled from testosterone.  相似文献
