The purpose of these experiments is to determine the effects of foliar applications of benzylaminopurine (BAP) and gibberellic acid (GA) on tuber number production of seed potatoes. In field experiments conducted during 1989/90 cv. Mailén was used and BAP, 50 mg·l–1 was foliarly applied at (1) tuber initiation, 36 days after emergence (DAE); (2) 54 DAE; and, (3) 64 DAE. Under glasshouse conditions, in 1991/92 cv. Spunta was used and BAP 50 mg·l–1+GA 50 mg·l–1 were applied 30 and 37 days after planting/transplanting. In 1992 cv. Huinkul, Kennebec and Spunta were used and BAP 50 mg·l–1+GA 50 mg·l–1 and Biozyme (Techic SA), a commercial product with auxin (IAA, 32.2 mg·l–1), gibberellic acid (GA3, 32.2 mg·l–1) and cytokinins (zeatin, 83.2 mg·l–1) at 5 ml·l–1 were applied. In cv. Mailén, a higher tuber number in the seed fraction (<80 g) was found when BAP was applied at each of the three crop stages, while applications 54 and 62 DAE also increased tuber number in the 80–400 g fraction. As a result of BAP applications, tuber yield was also significantly increased. In the glasshouse experiments, cv. Spunta showed a significant increase in minituber production in 3 out of 4 cases, either if the mother plant came from in vitro generated plantlets or minitubers, or if GA + BAP or Biozyme was applied. It can be concluded that the use of these PGRs under both field and glasshouse conditions in cvs. Mailén and Spunta can result in increased tuber number in the seed fraction.  相似文献

Summary Characteristics of a continuous reactor with Propionibacterium cells entrapped with Ca-alginate are described. For a dilution rate D=0.45 h–1, the gel produced 8 g.l–1 for volatile fatty acids and 0.18 g.l–1 for diacetylacetoin. The fiability of the process and its ability to produce propionic acid bacterium starter at the rate 4.109 cells.l–1 gel.h–1 maked it attractive for industrial applications.  相似文献

Eicosapentaenoic (EPA) and docosahexaenoic (DHA) acid productivities from chemostat cultures of an isolate of Isochrysis galbana have been studied. The productivities reached in the interval of dilution rates between 0.0295 h–1 and 0.0355 h–1 were 1.5mg·1–1·h–1 for lipids, 300 g·1–1·h–1 for EPA and 130g1·1–1·h–1 for DHA. Furthermore, light attenuation by mutual shading, and agitation speed influences on growth and fatty acid composition were analysed. A model relating steady-state dilution rates to internal average light intensity has been proposed, the parameter values of which obtained by non-linear regression were: maximum specific growth rate (max)=0.0426 h–1; the affinity of cells to light (Ik) = 10.92 W·m–2; the exponent (n) = 5.13; regression coefficient (r 2)=0.9999. Summary Eicosapentaenoic acid (EPA) volumetric productivity from an outdoor chemostat culture ofPhaeodactylum tricornutum UTEX 640 in a 50-l tubular photobioreactor varies with dilution rate, reaching a maximum of 47.8 mg l–1 d–1 at D=0.36 d–1. Summary The production of -linolenic acid (GLA) by the fungus Mucor rouxii CBS 416.77 was studied on low budget nitrogen and carbon sources, i.e. rape meal, cocos expeller and two types of yeast extract (nitrogen sources), and starch, starch hydrolysate, beet molasses and cocos expeller (carbon sources). As references, Difco yeast extract and glucose were used. In flask cultivations the three yeast extracts were fully interchangeable, while the Difco yeast extract (the most expensive of those tested) gave a higher productivity of GLA in fermentor cultures (14 mg·l–1·h–1). The yield of lipids and GLA were increased in the order yeast extract < rape meal < cocos expeller. Thus the amount of lipid increased from 0.56 to 2.8 g·l–1, and that of GLA from 0.15 to 0.33 g·l–1. Use of beet molasses or cocos expeller as carbon sources gave poor growth. Starch and starch hydrolysate resulted in better productivity of GLA than glucose (4.7 and 4.9 compared to 3.4 mg·l–1·h–1). To investigate the production potential of eicosapentaenoic acid (EPA) by the diatom Nitzschia laevis, the growth characteristics and fatty acid composition of the cells were studied under photoautotrophic, mixotrophic and heterotrophic conditions of growth. The specific growth rate and maximum biomass concentration were respectively 0.466 d–1 and 2.27 g l–1 for mixotrophic culture, 0.344 d–1 and 2.04 g l–1 for heterotrophic culture, and 0.167 d–1 and 0.5 g l–1 for photoautotrophic culture, respectively. As for EPA production, the yield and productivity were respectively 52.32 mg l–1 and 10.46 mg l–1 d for mixotrophic culture, 35.08 mg l–1 and 6.37 mg l–1 d for heterotrophic culture, and 6.78 mg l–1 and 3.39 mg l–1 d for photoautotrophic culture, respectively. Results suggest that mixotrophic culture is the most suitable growth mode for the production of EPA by the diatom Nitzschia laevis. The results are useful for the development of a cost-effective fermentation process for EPA production by Nitzschia laevis.  相似文献

Summary Submerged batch cultivation under controlled environmental conditions of pH 3.8, temperature 30°C, and KLa200 h–1 (above 180 mMO2 l –1 h–1 oxygen supply rate) produced a maximum (12.0 g·l –1) SCP (Candida utilis) yield on the deseeded nopal fruit juice medium containing C/N ratio of 7.0 (initial sugar concentration 25 g·l –1) with a yield coefficient of 0.52 g cells/g sugar. In continuous cultivation, 19.9 g·l –1 cell mass could be obtained at a dilution rate (D) of 0.36 h–1 under identical environmental conditions, showing a productivity of 7.2 g·l –1·h–1. This corresponded to a gain of 9.0 in productivity in continuous culture over batch culture. Starting with steady state values of state variables, cell mass (CX–19.9 g·l –1), limiting nutrient concentration (Cln–2.5 g·l –1) and sugar concentration (CS–1.5 g·l –1) at control variable conditions of pH 3.8, 30°C, and KLa 200 h–1 keeping D=0.36 h–1 as reference, transient response studies by step changes of these control variables also showed that this pH, temperature and KLa conditions are most suitable for SCP cultivation on nopal fruit juice. Kinetic equations obtained from experimental data were analysed and kinetic parameters determined graphically. A unicellular marine picoplankton, Nannochloropsis sp., was grown under CO2-enriched photoautotrophic or/and acetate-added mixotrophic conditions. Photoautotrophic conditions with enriched CO2 of 2800 l CO2 l–1 and aeration gave the highest biomass yield (634 mg dry wt l–1), the highest total lipid content (9% of dry wt), total fatty acids (64 mg g–1 dry wt), polyunsaturated fatty acids (35% total fatty acids) and eicosapentaenoic acid (EPA, 20:53) (16 mg g–1 dry wt or 25% of total fatty acids). Mixotrophic cultures gave a greater protein content but less carbohydrates. Adding sodium acetate (2 mM) decreased the amounts of the total fatty acids and EPA. Elevation of CO2 in photoautotrophic culture thus enhances growth and raises the production of EPA in Nannochloropsis sp.  相似文献

Outdoor chemostat cultures of the marine microalgaIsochrysis galbana at constant dilution rate (0.034 h–1 ) have been carried out under different weather conditions. Steady-state biomass concentrations were 1.61±0.03 kg m–3 in May and 0.95±0.04 kg m–3 in July, resulting in biomass output rates of 0.54 kg m–3 d–1 and 0.32 kg m–3 d–1 in May and July, respectively. Two patterns of daily variation with the solar cycle were observed in the fatty acid content. Saturated and mono-unsaturated fatty acids (16:0 and 16:1n7) show significant variation with the solar cycle, associated with short-term changes in environmental factors. Palmitic and palmitoleic acids are generated during daylight and consumed during the dark period. However, polyunsaturated fatty acids do not show a significant response to the solar cycle and their changes are associated with long-term variation in environmental factors. The maximum EPA productivity was obtained in May, 14.1 g m–3 d–1, which is close to that found in the literature for indoor continuous cultures. Nonetheless, the outdoor EPA content (up to 2.61 % d.wt) was lower than the indoor EPA content from a previous study (5% d.wt).  相似文献

An unsaturated fatty acid auxotroph of the oleaginous yeast Apiotrichum curvatum, named UfaM3, blocked in the conversion of stearic to oleic acid was cultivated in single-stage continuous culture. The influence of consumed carbon to nitrogen ratios (C/N ratios, g g–1) obtained at various dilution rates (D) on fatty acid (FA) accumulation and its profiles were studied. In continuous culture in N-limited medium a maximum FA accumulation of 45.6% (g g–1 of dry biomass) was obtained at an optimal D of 0.049 h–1, recording an efficiency of substrate conversion of 0.48 g g–1 and 0.22 g g–1 for biomass and lipids, respectively. The quality of lipid approached cocoa butter at an optimal C/N ratio of between 20 and 30. The C/N ratio in the incoming medium was 38.5 g g–1 with 30 g l–1 of glucose and both C and N sources were completely consumed at a critical D of 0.07 h–1. Crude rapeseed oil and post-refining fatty acids were used as substrates for oxalic acid production by a mutant of Aspergillus niger. Both the final concentration and the yield of the product were highest at pH 4 to 5. With a medium containing 50 g lipids l–1, production reached a maximum of 68 g oxalic acid l–1 after 7 d. A high yield of the product (up to 1.4 g oxalic acid g–1 lipids consumed) was achieved with oil and fatty acids combined.  相似文献

The conversion of glycerol to 1,3-propanediol by Citrobacter freundii DSM 30040 was optimized in single- and two-stage continuous cultures. The productivity of 1,3-propanediol formation was highest under glycerol limitation and increased with the dilution rate (D) to a maximum of 3.7 g·l–1·h–1. Glycerol dehydratase seemed to be the rate-limiting step in 1,3-propanediol formation. Conditions for the two-stage fermentation process were as follows: first stage, glycerol limitation (250mM), pH 7.2, D=0.1 h, 31° C; second stage, additional glycerol, pH 6.6, D=0.05 h–1, 28° C. When docosahexaenoic acid (DHA)-producing Moritella marina strain MP-1 was cultured in the medium containing 0.5 μ g cerulenin ml−1, an inhibitor for fatty acid biosynthesis, the cells grew normally, but the␣content of DHA in the total fatty acids increased from 5.9–19.4%. The DHA yield of M. marina strain MP-1 cells also increased from 4 to 13.7 mg l−1 by cerulenin treatment. The same effect of cerulenin was observed in eicosapentaenoic acid (EPA)-producing Shewanella marinintestina strain IK-1 grown in the medium containing 7.5 μg cerulenin ml−1, and the cerulenin treatment increased the EPA yield from 1.6 to 8 mg l−1. The use of cerulenin is, therefore, advantageous to increase the content of intracellular polyunsaturated fatty acids (PUFA) in particular PUFA-containing phospholipids in bacterial cells.An erratum to this article can be found at .  相似文献

Summary A procedure leading to the regeneration of whole plants from protoplasts of melon is described. Protoplasts were isolated from cotyledons and leaves of plants grown in vitro. After 14 days of culture, average viability and division rates were respectively 60% and 30% for the two organs, considering total initial protoplasts plated. The manipulation of the exogenous auxin / cytokinin balance in regeneration media enabled to direct morphogenesis towards somatic embryogenesis (1 mg·l–1 2,4-dichlorophenoxyacetic acid and 0.1 mg·l–1 6-benzylaminopurine) or caulogenesis (0.5 mg·l–1 6-benzylaminopurine and 0.5 mg·l–1 kinetin). Contrary to division ability, regeneration capacity was genotype-dependent under our conditions, but the two organs expressed similar division and regeneration capacities. Maltose was superior to sucrose for the development of caulogenic nodules into buds. Summary In an effort to improve the viability of acetone-butanol-ethanol fermentation by extractive fermentation, 63 organic solvents, including alkanes, alcohols, aldehydes, acids, and esters, were experimentally evaluated for biocompatibility with Clostridium acetobutylicum by observing gas evolution from cultures in contact with candidate solvents. Thirty-one of these solvents were further tested to determine their partition coefficient for butanol in fermentation medium. The biocompatible solvent with the highest partition coefficient for butanol (4.8), was poly(propylene glycol) 1200, which was selected for fermentation experiments. This is the highest partition coefficient reported to date for a biocompatible solvent. Extractive fermentations using concentrated feeds were observed to produce up to 58.6 g·l–1 acetone and butanol in 202 h, the equivalent of three control fermentations in a single run. Product yields (based on total solvent products and glucose consumed) of 0.234 g·g–1 to 0.311 g·g–1 and within run solvent productivities of 0.174 g·l–1·h–1 to 0.290 g·l–1·h–1 were consistentwith conventional fermentations reported in the literature. The extended run-time of the fermentation resulted in an overall improvement in productivity by reducing the fraction of between-run down-time for fermentor cleaning and sterilization.  相似文献

Continuous and repeat-batch biofilm fermentations using Actinobacillus succinogenes were performed with immobilized and suspended-cell systems. For the immobilized continuous system, plastic composite supports (PCS) containing 50% (w/w) polypropylene (PP), 35% (w/w) ground soybean hulls, 5% (w/w) dried bovine albumin, 2.5% (w/w) soybean flour, 2.5% (w/w) yeast extract, 2.5% (w/w) dried red blood cells, and 2.5% (w/w) peptone, or PP tubes (8.5 cm in length) were arranged around the agitator shaft in a grid formation. Agitation was controlled at 125 rpm and 150 rpm. Samples were taken at dilution rates of 0.2, 0.4, 0.6, 0.8, 1.0, and 1.2 h–1 and analyzed for succinic acid production and glucose consumption (g l–1). For PCS bioreactors, the highest final succinic acid concentrations (10.1 g –1, 10.4 g l–1) and percentage yields (62.6%, 71.6%) occurred at the dilution rate of 0.2 h–1. PCS disks were evaluated in a repeat-batch biofilm reactor. Suspended-cell batch fermentations were performed in flasks and a repeat-batch bioreactor. The maximum concentration of succinic acid produced was 40 g l–1. Peak succinic acid percentage yields in continuous and repeat-batch fermentations of A. succinogenes were observed in suspended-cell continuous fermentations at a dilution rate of 1.0 h–1 (76.2%) and in PCS repeat-batch fermentations with an initial glucose concentration of 40 g l–1 (86.7%).  相似文献

Two set of isolates were obtained in an isolation/selection programme to select eicosapentaenoic acid (EPA) and/or docosahexaenoic acid (DHA) rich strains ofIsochrysis galbana. EPA content was improved up to an average of 5.3% (d.wt) for the second set of isolates. On the other hand, with the aeration rate, pH and irradiance kept at low levels, the growth rate was slow and EPA synthesis was enhanced, but productivity increased when growth rates were maximum. A model relating steady-state dilution rates in chemostat cultures ofI. galbana to internal average irradiance is proposed. The greatest productivities were obtained between 0.0295 h–1 and 0.0355 h–1 with 300 mg m–3 h–1 for EPA and 130 mg m–3 h–1 for DHA.  相似文献

Styrene was degraded as sole source of carbon and energy by a selected bacterial community in a two-phase aqueous-organic medium (80%:20%, vol/vol). Silicone oil was used to solubilize styrene, which is sparingly soluble in water and to prevent its toxicity toward microorganisms. Preliminary studies with the mixed population in batch cultures indicate that the specific activity and the maximum growth rate at optimal 3H 6.0 were 46 mg·g–1·h–1 and 0.15 h–1, respectively. In pH-regulated chemostat cultures, styrene was degraded at dilution rates ranging from 0.05 to 0.20 h–1. Kinetic parameters and the proportion of each strain in the mixed culture were followed. At 0.20 h–1, only one strain as compared to four initially present, remained in the medium. This strain Pseudomonas aeruginosa, degrades styrene with a specific activity of 293 mg·g–1·h–1. During the cheese-making process, water activity (aw) is one of the essential environmental parameters acting on bacterial growth and metabolic pathways. The influence of aw on Streptococcus diacetylactis growth and lactic acid production was studied. The specific growth rate was linearly related to water availability in the milk medium. The cell behaviour was quite different above and below aw=0.95, which can be considered a limiting value. Below this value, the lactic acid production reached 1.4–6.1 mg·g–1, whereas the specific productivity was 2.0–2.6 mg·10–10 cells·h–1. Changes in the consumption of lactose and amino acids during the different growth phases was completely modified by decreasing the water availability in the medium. Correspondence to: N. Summary Tyromyces sambuceus was cultivated in a stirred tank reactor under semicontinuous conditions for 70 days. Over longer periods of time in submersion, values of 220 and 330 mg·L–1 of 4-decanolide could be maintained; after dispersion of the headspace mycelia, a single peak product concentration of 880 mg·L–1 was attained. The productivity was subject to large variations, peak values were 18 and 11 mg·L–1·h–1. Depending on the culture conditions, specific growth rates of up to 0.05 h–1 were reached.  相似文献
