Spatial and temporal distribution of zeatin and zeatin riboside was studied in the ovaries of amphimictic (Triticum aestivum L.) and apomictic (Taraxacum officinale Web.) species during the period from the quiescence release of the egg cell to its first division. This period was divided into three stages: (1) no signs of division of the central cell, (2) division of the central cell and no signs of division of the egg, and (3) division of the egg. Cytokinin content was determined in ovary and its four sections (basal, medial, apical, and ovule) using an original modification of ELISA. Apomicts and amphimicts were shown to be similar in the pattern of changes in the cytokinin content in basal and medial zones but differed substantially in that of apical section and ovule.  相似文献

We quantified various endogenous cytokinins during wheat (Triticum aestivumL.) and dandelion (Taraxacum officinaleWeb.) ovary development. Wheat ovaries were studied at the following developmental stages: the mature embryo sac with eight nuclei (stage 1), the interphasic zygote 12 h and 24 h after fertilization (stage 2), and the onset of zygote division (stage 3). The dandelion ovaries were studied at the stage of the mature embryo sac (stage 1), in the interphase of the parthenogeneticaly developing ovule (stage 2), and during its first division (stage 3). The material was analyzed by the method of competitive solid-phase immunoenzyme assay (ELISA) using peroxidase-labeled anti-rabbit antibodies. The onset of embryogenesis in wheat and dandelion ovules was accompanied by the substantial rearrangement of their hormonal complexes, which preceded the morphogenetic processes leading to seed formation. This implies that the hormonal system of the whole maternal plant is involved in the induction of embryogenesis. The final stages of embryogenesis depend on the hormonal systems in the flower, ovary, and ovule.  相似文献

In G2 peas senescence only takes place in long days. In order to determine the role of cytokinins in this process the endogenous cytokinins from vegetative shoots of G2 peas were characterized using gas chromatography-mass spectroscopy following purification by HPLC. Cytokinins were extracted and purified with and without the addition of 15N labelled internal standards of several cytokinins to estimate cytokin content by isotope dilution in the mass spectra. Samples without internal standards were bioassayed after HPLC. Bioassays showed the presence of zeatin, zeatin riboside and zeatin-0-glucoside. The presence of zeatin was confirmed by its mass spectrum of its permethylated derivative. Tentative identification of zeatin riboside, zeatin-0-glucoside, dihydrozeatin, and dihydrozeatin-0-glucoside was obtained by the coincidence of the major ion for the permethylated natural and 15N labelled internal standards on GC-MS, and the similar coincidence of ions for permethylated zeatin riboside-0-glucoside by direct probe MS. There was no indication of the presence of significant quantities of zeatin-7-glucoside or zeatin-9-glucoside. The amounts in the tissue ranged from 200–1000 ng/kg fresh weight for each cytokinin and about 2–4 g/kg fresh weight for total cytokinins. Roots are recognised as the major sites of cytokinin synthesis and shoots receive a continuous supply of cytokinins from the roots. Although reports are available on the xylem mobility of putative free bases and their ribosides, relatively few studies on the phloem mobility of cytokinins have been reported. The origin of phloem-mobile cytokinins is uncertain but there is evidence which implicates a recirculation from the root source. This study is the first report in which zeatin and zeatin riboside from the root pressure exudate and phloem sap of Ricinus have been identified by full-scan GC-MS and quantified by GC-MS selective-ion-monitoring. In this study, the concentration of cytokinins in root pressure exudate was similar, but lower, and in the phloem sap higher than that reported previously. The concentration of cytokinins quantified in the phloem sap confirms their transport in the sieve tubes. The relatively high concentration of zeatin riboside detected in the root pressure exudate and of zeatin detected in the phloem sap indicate a possible vascular recirculation of these hormones.  相似文献

In vitro propagation of cauliflower has generally been achieved through axillary shoot proliferation of curd explants on Murashige and Skoog (MS) medium supplemented with an auxin and a cytokinin. Recently, it has been shown (Vandemoortele 1999) that a soaking in sucrose (-2 MPa for 24 h) of cauliflower curd explants, before culture without any growth regulator, also induced axillary branching. The later procedure avoids the phenomenon of hyperhydricity in the shoots formed. Axillary shooting obtained by the two methods appears to be mediated by modifications of internal cytokinin levels. The osmotic pretreatment did not influence auxin levels, but induced a zeatin and a zeatin riboside levels increase. Curd explants cultured with the usual procedure (on MS medium supplemented with 5 μmol/L BA and 0.5 μmol/L NAA) showed a zeatin and zeatin riboside levels increase of the same magnitude and a higher one for isopentenyl adenine and isopentenyl adenosine. The modification of the cytokinin status in the curd explants subjected to a short osmotic pretreatment thus should be less favourable for hyperhydricity.  相似文献

Summary We successfully micropropagatedHesperaloe parviflora from mature plants. Shoot cultures were directly initiated from mature plants using pedicel bud explants on a modified Murashige and Skoog medium containing Nitsch and Nitsch vitamins and 1 μM zeatin riboside. Axillary shoot multiplication from established cultures was most responsive to changing concentrations of N6-benzyladenine (BA) with the greatest production of 6 μM BA. Growing shoots on a medium supplemented with 6 μM BA for 6 wk and then transferring cultures to a 1 μM BA medium for 6 more wk increased the number of transferable shoots, but not significantly. However, our data predicts that the maximum number of transferable shoots produced from a single microshoot would occur on media with 5.4 μM zeatin riboside. Shoots rooted easilyin vitro orex vitro and rooted shoots were easily acclimatized. The methods described in this paper are being used to commercially micropropagateH. parviflora.  相似文献

Data are presented on the cytokinin status of seeds and seed components, at different stages of development in Phaseolus coccineus L., as determined with the soybean callus growth bioassay: A change in cytokinin types according to developmental stage occurred: from biologically very active less polar types (zeatin=Z) at early stages to more polar types (zeatin glucoside=Z9G and zeatin riboside=Zr), with relatively low biological activity, at intermediate and late stages of seed development: When cytokinins were analyzed separately in embryos (embryo proper) and suspensors at two embryonic stages: heart-shaped (A) and middle cotyledonary embryos (stage B) respectively, it was found that: i) at stage A, the suspensor showed cytokinin activity at the level of Z, 2iPA (2-isopentenyladenosine) and Zr, whereas more polar cytokinins (Z9G, Zr) were present in the embryo; ii) at stage B, when the embryo seems to become autonomous for cytokinin supply, there was a relative abundance of active cytokinins (Z, 2iPA) in the embryo to which Z9G activity in the suspensor corresponded. Zeatin and zeatin riboside were identified by full-scan gas chromatography-mass spectrometry (GC-MS) in xylem sap of clonal apple rootstocks (M.27, M.9 and MM.106). These rootstocks exhibit a wide range of control over tree size when grafted to a common scion. The concentrations of zeatin and zeatin riboside were measured by GC-MS selected ion monitoring (SIM) in shoot xylem sap and root pressure exudate obtained from these rootstocks and from trees of Fiesta scion grafted onto the rootstocks. Zeatin was the predominant cytokinin in xylem sap from the dwarfing rootstocks, M.27 and M.9, while zeatin riboside was the predominant cytokinin in xylem sap from the more invigorating rootstock MM.106. Cytokinin concentrations (ng ml–1) in root pressure exudate and shoot xylem sap, (i.e. from above the graft union in composite trees), increased with increasing vigour of the rootstock, irrespective of whether the plants were non-grafted rootstocks, or were composite plants of Fiesta scion grafted onto the rootstocks. Cytokinin content (ng shoot–1) of shoot sap differed with rootstock; the more invigorating (MM.106) had greater amounts of cytokinins than the more dwarfing (M.9 and M.27) rootstocks. These results are discussed in relation to possible influences of roots on the growth of shoots via cytokinin supplies in the xylem sap.  相似文献

Phaseolus coccineus embryos at the heartshaped and the middle cotyledonary stages were cultured in vitro on media added with different concentrations of zeatin (Z) or zeatin riboside (Zr). Growth of early embryos was clearly favored by concentrations of Z from 10-8 M to 10-5 M, lower concentrations having no effect. Zr also promoted in vitro growth of early embryos, but in concentrations from 10-12 M to 10-10 M, higher concentrations being inhibitory. 甘蔗幼叶片,叶鞘和幼茎在含有2.4-D的培养条件下脱分化的情况有明显差异。它们除了在形态学上有区别外,这三种材料原有的细胞分裂素和ABA的水平明显不同。在幼叶片脱分化及愈伤组织形成过程中,内源细胞分裂素和ABA的水平也发生了明显变化。据此我们认为在甘蔗组织培养中2.4-D可能通过调节内源激素的水平及其相互作用,引起培养物中某些生理生化过程发生改变,从而进行脱分化和愈伤组织形成。  相似文献

Tomato transformation and regeneration were analysed and optimized. Cotyledon explants from Lycopersicon esculentum cv. UC82B, were infected by Agrobacterium tumefaciens strain LBA4404 harbouring the neomycin phosphotransferase (NPTII) reporter gene. The effects of phenolic compounds, vitamins and growth regulators on plant transformation and regeneration were studied. Increasing the vitamin thiamine concentration from 0.1 mg l–1 in standard medium to 0.4 mg l–1 decreased the chlorophyll lost that accompanied the expansion of necrotic areas in cotyledon explants. Optimal shoot regeneration rate was obtained with a balanced concentration of 0.5 mg l–1 auxin indolelacetic acid (IAA) and 0.5 mg l–1 cytokinin zeatin riboside. Finally, when the phenolic acetosyringone was present in the co-culture medium at 200 µM, confirmed transgenic lines reached 50% of antibiotic resistant shoots. Under the above conditions, the transformation efficiency reached 12.5%.  相似文献

Summary The zeatin and zeatin riboside content of tomato (Lycopersicon esculentum Mill.) root exudates were determined at different stages of development. Zeatin riboside was found to be the major translocational form of cytokinin in the xylem during early vegetative growth. During flower bud formation this cytokinin decreased markedly in concentration so that, at anthesis, there was no appreciable difference in the zeatin and zeatin riboside concentration in the root exudate.  相似文献

The hormonal status of the Taraxacum officinale Web. ovary was quantitatively assayed for the first time during early stages of embryogenesis. Apparent concentrations of endogenous cytokinins were measured using two systems of enzyme-linked immunosorbent assay (ELISA). The ELISA systems differed from one another by the specificity for the main endogenous forms of zeatin. The specificity of two heterological ELISA systems based on zeatin- and kinetin-specific antisera was studied. A new immunochemical approach to the problem of differential quantitative determination of natural zeatin forms is suggested. This approach does not require preliminary separation of experimental samples into individual fractions. True concentrations of zeatin and zeatin riboside in the T. officinale ovary were calculated based on the average values of apparent concentrations of endogenous cytokinins. When the embryo sac maturation had been completed, there was a threefold increase in the zeatin riboside concentration within the following 12 h. By the time of the first division of an unfertilized ovicell (i.e., within the next 12 h), there had been a twofold decrease in the zeatin riboside concentration. Therefore, at early stages of division of the unfertilized ovicell the zeatin riboside concentration virtually returned to the initial level. In contrast to zeatin riboside, there was a steady trend toward an increase in the zeatin concentration in the T. officinale ovary. Within the first 12 h and the next 12 h after completion of the embryo sac maturation, the zeatin concentration was increased 1.5-fold and 2-fold, respectively. The results of this work provide a pioneering insight into the dynamics of various natural forms of zeatin during the reproductive process. The immunochemical approach to quantitative monitoring of various natural forms of zeatin and their dynamics during embryogenesis suggested in this work can be extended to similar biological, medical, and agricultural problems of differential determination of low-molecular-weight agents of similar structure but different biological activity.  相似文献

Effects of N6-benzyladenine, kinetin, zeatin, N6-benzyladenosine, kinetin riboside, zeatin riboside, jasmonic acid, indole-3-acetic acid, indole-3-butyric acid, indole-3-propionic acid, abscisic acid and gibberellic acid on proliferation of hyphae of arbuscular mycorrhizal (AM) fungus Glomus fistulosum were studied under axenic conditions in vitro. The growth of intraradical hyphae of G. fistulosum was fully suppressed by 30 μM indole-3-acetic acid, but a perceptible decrease in the proliferation of the hyphae was observed already at 3 μM. Because such concentration is near the concentrations common in root tissues in vivo, the effect may be biologically significant. Similar effect was also observed for Glomus mosseae. Inhibitory effects of abscisic acid and cytokinins occurred only at very high, non-physiological concentrations. Ribosylated cytokinins showed stronger inhibition effects than their non-ribosylated counterparts. No stimulation of proliferation of hyphae by any plant hormone tested was observed. Grain filling patterns and their relationships withzeatin (Z), zeatin riboside (ZR), indole-3-acetic acid(IAA) and gibberellin (GA) contents in the grains androots during grain development were examined in sixrice (Oryza sativa L.) genotypes grown in thefield and in water culture. Three grain fillingpatterns based on the filling rate of superior andinferior spikelets were observed, i.e., fastsynchronous: all spikelets started filling early andfast at the early filling stage; slowsynchronous: all spikelets filled slowly at the earlyfilling stage and reached the maximum filling ratelate; and asynchronous: superior spikeletsstarted filling and reached the maximum filling ratemuch earlier than the inferior ones. The order ofgrain filling percentage in the three types of grainfilling patterns was: fast synchronous >asynchronous > slow synchronous. Changes in Z + ZRcontents in the superior and inferior spikelets wereassociated with the grain filling patterns. Grainfilling percentage was significantly correlated withZ + ZR contents in the grains and roots at the earlyand middle grain filling stages. IAA and GA(GA1 + GA3 + GA4)contents in the grains and roots were notsignificantly correlated with grain fillingpercentage. The results suggest that cytokinins in thegrains and roots during the early phase of graindevelopment play an important role in regulating grainfilling pattern and consequently influence grainfilling percentage.  相似文献

Comparative study on fertilization process in Pinus sylvestris, Pinus mugo and in their putative hybrid swarm individuals was done involving pre-zygotic and post-zygotic stages. The amount of surviving ovules from open pollination reflecting the mode of interaction between pollen grains and nucellar tissue of an ovule averaged at 8.1 of sound ovules per conelet in Pinus sylvestris, 7.3 ovules in the hybrid swarm population and at 4.9 ovules in Pinus mugo. A strong correlation was observed between the number of surviving ovules and the proportion of germinating seeds in the compared species and hybrids. Normal course of embryogenesis in Pinus sylvestris and Pinus mugo contrasted with increased frequency of disturbances observed in the hybrid swarm individuals. The differential survival rates of the ovules and deviations from typical pattern of embryogenesis are discussed from the standpoint of cross-ability relationship between Pinus sylvestris and Pinus mugo.  相似文献

Involvement of cytokinins (CKs) in axillary bud growth of miniature rose was studied. Variation in root formation and axillary bud growth was induced by two indole 3-butyric acid (IBA) pretreatments in two cutting sizes. At six physiological developmental stages around the onset of axillary bud growth, concentrations of CKs were determined in both root and axillary bud tissue by liquid chromatography combined with electrospray tandem mass spectrometry (LC-ESP-MS/MS). Chronological early onset of axillary bud growth occurred in long cuttings pretreated at low IBA concentration, whereas physiological early root formation was associated with long cuttings and high IBA concentration. The CKs zeatin (Z), isopentenyl adenine (iP), zeatin riboside (ZR), dihydrozeatin riboside (DHZR), isopentenyl adenosine (iPA), zeatin O-glucoside (ZOG), zeatin riboside O-glucoside (ZROG), zeatin riboside 5-monophosphate (ZRMP), and isopentenyl adenosine 5-monophosphate (iPAMP) were detected. Concentrations of CKs in axillary bud tissue far exceeded those in root tissue. Indole 3-butyric acid pretreatment influenced the concentration of CKs in axillary bud tissue more than did cutting size, whereas pretreatments only slightly affected CKs in root tissue. The dominant CKs found were iPAMP and ZR. An early and large increase in iPAMP indicated rapid CK biosynthesis in rootless cuttings, suggesting that green parts, including the axillary bud, can synthesize CKs. At the onset of axillary bud growth an increase in concentration of Z, ZR, ZRMP, ZOG, and ZROG was largely coincident with a decrease in iPAMP, iPA, iP, and DHZR. After the onset of axillary bud growth, CK content largely decreased. These results strongly indicate a positive role for CKs in axillary bud growth, and presumably ZRMP, ZR, and Z are active in miniature rose.  相似文献

The stability of [3H]zeatin riboside supplied to freshly excised tobacco pith explants was found to be inversely related to -naphthaleneacetic acid concentration in the incubation medium. At higher concentrations of -naphthaleneacetic acid greater breakdown of [3H]zeatin riboside was indicated by higher levels of degradative metabolites (adenine, adenosine and adenosine nucleotides) formed. This auxin effect on cytokinin metabolism appears to be mediated, at least in part, through cytokinin oxidase. The results of in-vitro assays carried out with partially purified enzyme from corn kernels substantiale this conclusion.
