The objective of this study was to evaluate pollen dispersal inBrassica napus (oilseed rape). The selectable marker, used to follow pollen movement, was a dominant transgene (bar) conferring resistance to the herbicide glufosinate-ammonium. Transgenic and non-transgenic plants of the cultivar Westar were planted in a 1.1 ha field trial, with the transgenic plants in a 9 m diameter circle at the centre, surrounded by non-transgenic plants to a distance of at least 47 m in all directions. A 1 m circle of non-transgenic plants was sown in the centre of the transgenic area to allow estimation of the level of pollen dispersal when plants were in close contact. Honeybee hives were placed at the trial site to optimize the opportunity for cross-pollination. During the flowering period, regular observations were made of the number of plants flowering and the number and type of insects present in 60 1 m2 areas. These areas were located uniformly around the plot at distances of 1, 3, 6, 12, 24, 36 and 47 m from the edge of the 9 m circle of transgenic plants. Seed samples were harvested from each of the 7 distances so that approximately 20% of the circumference of the plot was sampled at each distance. The centre non-transgenic circle was also sampled. Plants were grown from the seed samples and sprayed with glufosinate to estimate the frequency of pollen dispersal at each distance. In order to screen enough samples to detect low frequency cross-pollination events, seed samples were tested in the greenhouse and on a larger scale in the field. Results were confirmed by testing progeny for glufosinate resistance and by Southern blot analysis. The estimated percentage of pollen dispersal in the non-transgenic centre circle was 4.8%. The frequency was estimated to be 1.5% at a distance of 1 m and 0.4% at 3 m. The frequency decreased sharply to 0.02% at 12 m and was only 0.00033% at 47 m. No obvious directional effects were detected that could be ascribed to wind or insect activity.  相似文献

Genetically engineered pollen with a visible marker gene could be useful to monitor the movement of transgenic pollen provided there are no negative physiological or fitness effects of expressing such a gene. In this study, we measured the fitness of Nicotiana tabacum cv. Xanthi pollen expressing the marker gene green fluorescent protein (GFP). Average pollen tube germination frequencies and pollen tube growth rates in vitro were measured in three different types of plants: (1) plants producing GFP in pollen cells only (LAT59-GFP), (2) plants synthesizing GFP under the control of a constitutive promoter (CaMV 35S) in which no GFP was produced in pollen, and (3) non-transgenic plants. Pollen synthesizing the GFP protein did not differ significantly in average pollen germination frequencies from pollen without GFP (P=0.65). Average pollen tube growth rates over a 5-h period did not differ significantly between transgenic and non-transgenic types (R2=0.89, 0.98, and 0.95, respectively, for GFP-tagged, 35S-GFP, and wild type). Overall, GFP expression in pollen grains of tobacco was not found to have an effect on pollen fitness under the controlled experimental conditions of this study.  相似文献

Gene flow from transgenic oilseed rape (BRASSICA NAPUS) might not be avoidable, thus, it is important to detect and quantify hybridization events with its relatives in real time. Data are presented showing the correlation between genetically linked green fluorescent protein (GFP) with BACILLUS THURINGIENSIS (Bt) CRY1AC gene expression in hybrids formed between transgenic B. NAPUS "Westar" and a wild Chinese accession of wild mustard (B. JUNCEA) and hybridization between transgenic B. NAPUS and a conspecific Chinese landrace oilseed rape. Hybrids were obtained either by spontaneous hybridization in the field or by hand-crossing in a greenhouse. In all cases, transgenic hybrids were selected by GFP fluorescence among seedlings originating from seeds harvested from B. JUNCEA and the Chinese oilseed rape plants. Transgenicity was confirmed by PCR detection of transgenes. GFP fluorescence was easily and rapidly detected in the hybrids under greenhouse and field conditions. Results showed that both GFP fluorescence and Bt protein synthesis decreased as either plant or leaf aged, and GFP fluorescence intensity was closely correlated with Bt protein concentration during the entire vegetative lifetime in hybrids. These findings allow the use of GFP fluorescence as an accurate tool to detect gene-flow in time in the field and to conveniently estimate BT CRY1AC expression in hybrids on-the-plant.  相似文献

 In order to help establish a basis for the assessment of gene flow associated with the large-scale release of transgenic oilseed rape, we previously designed a method which makes it possible to retrieve the average pollen dispersal of a single plant from that of a large source plot. The ‘individual’ pollen distribution thus obtained is less dependent on the experimental design than pollen distributions usually published and could therefore be used to model the possible escape of a transgene from commercial transgenic crops. In this study we report on a field experiment set up to study the pollen dispersal from an herbicide-resistant transgenic variety of oilseed rape and to test the applicability of the method on the experimental data. Two techniques were used to determine the individual pollen dispersal, and their outcomes are compared. The results suggest that approximately half of the pollen produced by an individual plant fell within 3 m and that the probability of fertilisation afterwards decreased slowly along a negative exponential of the distance. Comparison with the global pollen distribution from the source plot indicates that pollen-dispersal distributions based on dispersal from whole plots instead of individual plants would have underestimated the proportion of pollen that was dispersed over average or long distances. In this study, the pollen‐active LAT59 promoter from tomato was used to express a green fluorescent protein (GFP) encoding gene in Nicotiana tabacum (tobacco) pollen. This promoter is preferentially expressed in anthers and pollen. Pollen in transgenic plants segregated in a 1 : 1 Mendelian ratio, and the plants were polymerase chain reaction (PCR)‐positive. GFP‐tagged pollen was developed as a tool for tracking the movement of transgenic plant pollen in the environment. Specifically, it should be a useful tool for characterizing the spatial distribution patterns of transgenic pollen, to determine pollination mechanisms, to monitor the effects on nontarget organisms, and to monitor gene flow in field conditions.  相似文献

One of the main insect pests in oilseed rape is the pollen beetle (Brassicogethes aeneus (Fabricius), syn. Meligethes aeneus). To maximize efficiency of control of this pest, insecticides are required that ideally, not just prevent yield losses by bud feeding of overwintered pollen beetles, but simultaneously minimize the reproduction of the pest, thereby reducing the size of the following generation infesting next year’s oilseed rape. The neonicotinoid active substance thiacloprid is known to reduce bud infestation with eggs and larvae. However, the mechanisms by which this occurs as well as the effects of other active substances are not known. In this study, the effects of the neonicotinoid insecticide Biscaya (a.i. thiacloprid) and the pyrethroids Mavrik (tau-fluvalinate) and Karate Zeon (lambda-cyhalothrin) applied at the bud stage of winter oilseed rape with recommended field rates on infestation of buds with eggs and larvae of pollen beetles were tested in field trials in Germany in 2013–2015. In additional greenhouse experiments, it was investigated whether effects on bud infestation were caused by lethal effects on pollen beetle or by insecticidal residues on plants causing sublethal effects. In the field trials, application of Biscaya and Mavrik significantly reduced the percentage of buds containing eggs and larvae in contrast to Karate Zeon. In 2014 and 2015, 14 days after application, bud infestation on the main raceme was reduced by 86 and 82%, respectively, in Biscaya-treated plots and by 51 and 71%, respectively, in Mavrik-treated plots compared to the untreated plots. In the greenhouse experiments, the lowest percentage of bud infestation with eggs and larvae was recorded on Biscaya-treated plants whereas on Mavrik-treated plants, there was no significant difference compared with the control. The results of the field trials show that Biscaya and Mavrik reduced oilseed rape bud infestation with eggs and larvae of pollen beetles primarily by lethal effects on overwintered pollen beetles or by repellency. However, Biscaya had additional effects on egg laying, which was supported by the greenhouse experiments.  相似文献

Abstract. The behavioural response of adults of the pollen beetle, Meligethes aeneus F. (Coleoptera: Nitidulidae), to the odour of one of their host-plants, oilseed rape ( Brassica napus L.), was observed in the laboratory using a fourarmed airflow olfactometer, and under semi-natural conditions using a markrelease–recapture technique. Oilseed rape leaf and flower odour was attractive to pollen beetles in the olfactometer and they dispersed upwind from the centre of a circular array (radius 20 m) of yellow water traps, baited with extracts of oilseed rape leaves or flowers. The dispersal of pollen beetles in the absence of oilseed rape odour was predominantly downwind and crosswind. The percentage recapture of pollen beetles was significantly greater in traps baited with rape flower extracts than leaf extracts. Pollen beetles can use odour-mediated upwind anemotaxis to locate oilseed rape plants from distances of at least 20 m over open ground.  相似文献

It has been suggested that there is a link between the relatively recent expansion in the farming of oilseed rape (Brassica napus L.) and an increase in the incidence of respiratory illness in eastern Scotland. However, there is limited evidence of the extent to which one possible cause of this increased illness, the pollen of oilseed rape, is disseminated from fields of this crop. The present study used sedimentation traps and a portable volumetric air sampler to study the transport of pollen from rape fields in Fife, and the results confirmed the widely held view that most of the pollen travels only a short distance from its source. In this case relatively low quantities ofBrassica pollen were detected more than 200 m from fields of rape. This may have implications for the formulation of advice for those who appear to suffer from an allergic reaction to rape crops.  相似文献

Transgenic oilseed rape ( Brassica napus ) plants may remain as 'volunteer' weeds in following crops, complicating cultivation and contaminating crop yield. Volunteers can become feral as well as act as a genetic bridge for the transfer of transgenes to weedy relatives. Transgenic mitigation using genes that are positive or neutral to the crop, but deleterious to weeds, should prevent volunteer establishment, as previously intimated using a tobacco ( Nicotiana tabacum ) model. A transgenically mitigated (TM), dwarf, herbicide-resistant construct using a gibberellic acid-insensitive (Δ gai ) gene in the B. napus crop was effective in offsetting the risks of transgene establishment in volunteer populations of B. napus . This may be useful in the absence of herbicide, e.g. when wheat is rotated with oilseed rape. The TM dwarf B. napus plants grown alone had a much higher yield than the non-transgenics, but were exceedingly unfit in competition with non-transgenic tall cohorts. The reproductive fitness of TM B. napus was 0% at 2.5-cm and 4% at 5-cm spacing between glasshouse-grown plants relative to non-transgenic B. napus . Under screen-house conditions, the reproductive fitness of TM B. napus relative to non-transgenic B. napus was less than 12%, and the harvest index of the TM plants was less than 40% of that of the non-transgenic competitors. The data clearly indicate that the Δ gai gene greatly enhances the yield in a weed-free transgenic crop, but the dwarf plants can be eliminated when competing with non-transgenic cohorts (and presumably other species) when the selective herbicide is not used.  相似文献

Abstract. The measurements of Yes'kov & Sapozhnikov (1976) suggest that electrostatic potentials on foraging honeybees can reach hundreds of volts. Pollen grains of oilseed rape, Brassica napus L., subjected experimentally to potentials of this order, jumped a distance that increased approximately as the square of the voltage, between two pin electrodes on which, in some experiments, were impaled an anther or stigma of oilseed rape or a freshly-killed honeybee. Most floral surfaces were insulated, but there was a low-impedance path to earth via the stigma, and the electrostatic field due to an approaching charged bee must therefore concentrate there. Thus, if electrostatic potentials of this magnitude occur in nature they may increase the chance that pollen from bees will reach the stigma rather than other floral surfaces, as well as enabling pollen to jump from anther to bee and from bee to stigma across an air gap of the order of 0.5 mm.  相似文献

Competitive ability of transgenic oilseed rape transformed with a pea lectin gene was estimated by comparisons of its performance when grown in a mixture with its non-transgenic counterpart and when grown singly, with and without pollen beetles present. The experiments were carried out in cages, once with bumblebees as pollinators and once without. In the absence of herbivory but with the presence of bumblebees, singly grown plant lines without lectin generally performed better than lines with lectin. Pollen beetles affected plant growth and reproduction, but there were no consistent differences between the lectin and non-lectin plant lines indicating that the transgenic trait did not protect plants from pest attack. Herbivory reduced the number of seeds when bumblebees were present. In the absence of bumblebees, however, plants produced more seeds with pollen beetles than without, indicating that some pollination was carried out by the beetles. Efficient pollination affected the competitive abilities of the lines; lectin lines were more competitive with bumblebees present and the reverse was true when bumblebees were absent. In the presence of bumblebees, lectin lines gained from being grown mixed with its non-transgenic counterpart. Because the transgenic plants expressed pea lectin in developing pollen it is possible that pollen quality in those plants was reduced, which may explain why the lectin lines had an advantage over non-lectin lines when exchange of pollen between the two plant types was facilitated by bumblebees.  相似文献

Assaying for transgenic pollen, a major vector of transgene flow, provides valuable information and essential data for the study of gene flow and assessing the effectiveness of transgene containment. Most studies have employed microscopic screening methods or progeny analyses to estimate the frequency of transgenic pollen. However, these methods are time-consuming and laborious when large numbers of pollen grains must be analyzed to look for rare transgenic pollen grains. Thus, there is an urgent need for the development of a simple, rapid, and high throughput analysis method for transgenic pollen analysis. In this study, our objective was to determine the accuracy of using flow cytometry technology for transgenic pollen quantification in practical application where transgenic pollen is not frequent. A suspension of non-transgenic tobacco pollen was spiked with a known amount of verified transgenic tobacco pollen synthesizing low or high amounts of green fluorescent protein (GFP). The flow cytometric method detected approximately 75% and 100% of pollen grains synthesizing low and high amounts of GFP, respectively. The method is rapid, as it is able to count 5000 pollen grains per minute-long run. Our data indicate that this flow cytometric method is useful to study gene flow and assessment of transgene containment.  相似文献

The cultivation of genetically modified (GM) herbicide resistant oilseed rape (Brassica napus) has increased over the past few years. The transfer of herbicide resistance genes via pollen (gene flow) from GM crops to non-GM crops is of relevance for the realisation of co-existence of different agricultural cultivation forms as well as for weed management. Therefore the likelihood of pollen-mediated gene flow has been investigated in numerous studies. Despite the difficulty to compare different experiments with varying levels of outcrossing, we performed a literature search for world-wide studies on cross-fertilisation in fully fertile oilseed rape. The occurrence and frequency of pollen-mediated intraspecific gene flow (outcrossing rate) can vary according to cultivar, experimental design, local topography and environmental conditions. The outcrossing rate from one field to another depends also on the size and arrangement of donor and recipient populations and on the ratio between donor and recipient plot size. The outcrossing levels specified in the presented studies are derived mostly from experiments where the recipient field is either surrounding the donor field (continuous design) or is located as a patch at different distances from the donor field (discontinuous design). Reports of gene flow in Brassica napus generally show that the amount of cross-fertilisation decreases as the distance from the pollen source increases. The evidence given in various studies reveals that the bulk of GM cross-fertilisation occurs within the first 10 m of the recipient field. The removal of the first 10 m of a non-transgenic field facing a GM crop might therefore be more efficient for reducing the total level of cross-fertilisation in a recipient sink population than to recommend separation distances. Future experiments should investigate cross-fertilisation with multiple adjacent donor fields at the landscape level under different spatial distributions of rapeseed cultivars and different cropping systems. The level of cross-fertilisation occurring over the whole field is mainly important for co-existence and has not been investigated in agricultural scale experiments until now. Potential problems with herbicide resistant oilseed rape volunteers arising from intraspecific gene flow can be largely solved by the choice of suitable cultivars and herbicides as well as by soil management.  相似文献

Understanding patterns of pollen movement at the landscape scale is important for establishing management rules following the release of genetically modified (GM) crops. We use here a mating model adapted to cultivated species to estimate dispersal kernels from the genotypes of the progenies of male-sterile plants positioned at different sampling sites within a 10 x 10-km oilseed rape production area. Half of the pollen clouds sampled by the male-sterile plants originated from uncharacterized pollen sources that could consist of both large volunteer and feral populations, and fields within and outside the study area. The geometric dispersal kernel was the most appropriate to predict pollen movement in the study area. It predicted a much larger proportion of long-distance pollination than previously fitted dispersal kernels. This best-fitting mating model underestimated the level of differentiation among pollen clouds but could predict its spatial structure. The estimation method was validated on simulated genotypic data, and proved to provide good estimates of both the shape of the dispersal kernel and the rate and composition of pollen issued from uncharacterized pollen sources. The best dispersal kernel fitted here, the geometric kernel, should now be integrated into models that aim at predicting gene flow at the landscape level, in particular between GM and non-GM crops.  相似文献

Information on the extent of transgene dispersal by pollen to adjacent potato plots and to related weed species is an important requisite for risk assessment; a procedure followed before novel transgenic plants are evaluated under field conditions. The purpose of the investigation was to determine the frequency of cross-pollination between potato (Solanum tuberosum) plants at different distances, using a kanamycin resistnace transgene (nptII) as a selectable marker. All potato plants were from the variety Désirée. Non-transgenic potato plants, used as potential recipients of transgene-containing pollen, were planted in 12 sub-plots, at distances of 0–20 m from the nearest transgenic potato plants. Seeds harvested from the non-transgenic plants were screened for resistance to kanamycin, and molecular methods were used to confirm that resistant progeny contained thenptII gene. Where transgenic and non-transgenic potato plants were in alternate rows (leaves touching), 24% of seedlings from the non-transgenic parent plants were kanamycin-resistant. Comparable seedlings from plants at up to 3 m distance had a resistance frequency of 2%, at 10 m the frequency was 0.017% and at 20 m no resistant progeny were observed. Plants of the weed speciesS. dulcamara andS. nigrum were also planted close to the transgenic potatoes to test for evidence of hybridization, and no kanamycin-resistant seedlings were observed among progeny fromS. dulcamara andS. nigrum. This investigation provided evidence that the extent of gene dispersal from transgenic potatoes to non-transgenic potatoes falls markedly with increasing distance, and is negligible at 10 m. There was, also, no evidence of transgene movement from potato toS. dulcamara andS. nigrum under field conditions. These data will be valuable in defining genetic isolation procedures for the early field evaluation and the use of novel transgenic potato genotypes.  相似文献

Pollinating insects are not only important in wild plant pollination, but also in the production of a large number of crops. Oilseed rape production is increasing globally due to demands for biofuels which may have impacts on pollinating insects which visit the crop and on the pollination services delivered to co-flowering wild plants. In this study, we tested (1) the degree of pollinator sharing between oilseed rape and native wild plants in field margins and hedgerows and (2) the effects of oilseed rape on the quality of pollination service delivered to these wild plants. We found large overlap between flower visitors of wild plants and oilseed rape, but the composition of species overlap differed with respect to each wild plant species. Nearly all individual visitors caught on both the crop and foraging on wild species carried crop pollen, but more than half the insects also carried pollen from wild plants. However, very little oilseed rape pollen was deposited on wild plant stigmas. This shows that (1) oilseed rape overlaps in pollinator niche with most co-flowering wild plants, and (2) crop pollen deposition on wild plant stigmas is low which may indicate that it is unlikely to cause reductions in seed set of wild plants, although this was not measured here. Furthermore, wild plants in field margins and hedgerows are important sources of alternative forage for pollinating insects even when a crop is mass flowering, and we suggest maintenance and augmentation of field margins and hedgerows to provide alternative forage for pollinator conservation to continue provision of pollination services to both crops and wild plants.  相似文献

During an early stage of migration into a field of oilseed rape in the spring of 1988, more pollen beetles were observed on the double-low variety, Ariana, than on the adjacent single-low varieties, Bienvenu and Mikado. More beetles were found on open flowers on terminal racemes than on yellow and green flower buds on lateral racemes. Five alkenyl, three indolyl and two aromatic glucosinolates were identified in the floral tissues of oilseed rape. The major compounds were progoitrin (2-OH, 3-butenyl) and glucobrassicanapin (4-pentenyl). There were no differences in the types and concentrations of glucosinolates present in the terminal and lateral racemes of any variety, and only small differences between single- and double-low varieties. Concentrations of glucosinolates in floral tissues of crops grown with a little nitrogen were twice those of well-fertilised crops. There was no association between the number and distribution of pollen beetles on plants and the amounts or types of glucosinolates present in floral tissues.  相似文献

Abstract.  1. Pollen is considered to be an important dietary component for many species of flower-feeding herbivores. Its influence on oviposition site selection by the pollen beetle Meligethes aeneus , and on the development of its larvae was investigated.
2. The effects of pollen presence and absence on adult, egg, and larval incidence in the field, and on larval development in the laboratory were compared through the use of Synergy, a composite hybrid oilseed rape Brassica napus variety comprising male-fertile (with pollen) and male-sterile (without pollen) plants.
3. In the field, adult females were more abundant on male-fertile plants during flowering, and a greater proportion of male-fertile than male-sterile buds were accepted for oviposition. These data indicate a possible role of pollen in oviposition site selection by female pollen beetles.
4. The numbers of first instar larvae on the two plant lines did not differ; however, more second instars were found on male-fertile than on male-sterile flowers. This suggests a greater larval survival on male-fertile plants, possibly due to the more readily available food resources and better nutrition afforded by the presence of pollen.
5. Laboratory experiments confirmed that a diet which included pollen improved survival to adulthood and resulted in heavier pupae and adults; however, pollen was not obligatory for larval survival and development.
6. The pollen beetle, previously thought to be an obligate pollen feeder, is therefore more generalist in its requirements for development. These findings may relate to the nutritional and behavioural ecology of other flower-feeding herbivores.  相似文献

Abstract.  1. Although pollen is a vital nutritional resource for honey bees, Apis mellifera , the influence of pollen quality on their foraging behaviour is little understood.
2. In choice-test experiments, bees showed no innate pollen-foraging preferences, but preferred oilseed rape Brassica napus pollen over field bean Vicia faba pollen after previous foraging experience of oilseed rape.
3. The free amino acid content of oilseed rape and field bean pollen was compared using high-performance liquid chromatography. Oilseed rape pollen contained a greater proportion of the most essential amino acids required by honey bees (valine, leucine, and isoleucine) than field bean, suggesting that oilseed rape pollen is of greater nutritional quality for honey bees than is field bean pollen.
4. Honey bee foraging preferences appeared to reflect pollen quality. The hypothesis that pollen amino acid composition affects the foraging behaviour of honey bees is discussed.  相似文献
