The micropropagation of neem (Azadirachta indica) was accomplished by culture of buds from crown branches of a mature tree, basal-sprouts of another mature tree and a single juvenile plant. Cultures derived from these three different sources showed significant variation in in vitro response. In case of the crown and basal-sprout explants, addition of 12.5 μM PVP-40 in the establishment medium controlled leaching of phenol growth inhibitors. Phenolic leaching was not observed in juvenile explants. DKW medium (with 0.22 μM BA) was significantly better than MS for shoot proliferation. Shoot cultures of crown branch origin did not elongate and eventually died after the third subculture. In the presence of 4.9 μM IBA in half-strength DKW, 90% of the shoots and 100% of basal-sprout and seedling explant origin, formed roots. Plantlets from both explant types showed 90% survival after acclimatization.  相似文献

What is probably the world’s largest neem (Azadirachta indica) plantation is on 10 sq km in the Plains of Arafat, Saudi Arabia, where 50,000 trees have been planted recently. The project, designed to provide shade to the ca. 2 million Muslim pilgrims who camp there annually for “Haj” rites, has economic potential. Pest-control materials, soap, toothpaste, and pharmaceutical products are among items that could be produced for home consumption and export. Introduced into the country more than 40 yr ago, the tree has acclimated itself remarkably well to the local hot and arid conditions. La plus grande plantation de neem (Azadirachta indica) entourne dix kilometres carrés dans les Plaines d’Arafat, Arabie Séoudite, où 50.000 arbres ont étés plantés récemment. Le projet, créé pour fournir ombrage aux 2 millions de pèlerins musulmans qui y campent annuellement pour les rites de “haj,” a potentiel économique. Les articles que l’on pourra produire pour consommation domestique et exportation incluent les matériels pour régler des pestes, le savon, le dentifrice, et les produits pharmaceutiques. Aquaneem, an emulsified product prepared from the neem (A. indica) kernel was tested against four pathogenic bacteria of fish (i.e. Aeromonas hydrophila, Pseudomonas fluorescens, Escherichia coli and Myxobacteria spp.) to test its efficacy as an antibacterial agent. Growth inhibitory property of the product at 10, 15 and 20 ppm has been noticed and recorded. The percentage reduction of bacterial cell population was noted to be maximum on 9th day at 20 ppm concentration (i.e. 70.14%, 74.15% and 61.75% for A. hydrophila, P. fluorescens and E. coli respectively) with the only exception of myxobacteria which showed maximum reduction percentage (63.90%) on 15th day. Among all the bacteria tested A. hydrophila, P. fluorescens and Myxobacteria spp. exhibited maximum sensitivity to Aquaneem in terms of percentage reduction of bacterial cell population in comparison to E. coli.  相似文献

A methanol extract of neem oil indicated antifeedant activity at 200 μg/disc by a no-choice bioassay against Reticulitermes speratus Kolbe. The extract was purified by recycling HPLC to isolate 11 compounds of variable termite antifeedant potency. Deacetylgedunin was the most active compound (95% protective concentration or PC95 = 113.7 μg/disc). This was followed by salannin, gedunin, 17-hydroxyazadiradione, nimbandiol, azadiradione, deacetylsalannin, and deacetylnimbin, with PC95 estimates of 203.3, 218.4, 235.6, 245.4, 827.5, 1373.1, and 1581.2 μg/disc, respectively. Epoxyazadiradione, 17-epiazadiradione, and nimbin were not active (PC95 estimates were beyond the bioassay limits). It could be presumed, by comparing their structures and activities, that the furan ring, αβ-unsaturated ketone and hydroxyl group each played an important role in determining the antifeedant activity.  相似文献

Seventeen limonoids (tetranortriterpenoids), 1 – 17 , including three new compounds, i.e., 17‐defurano‐17‐(2,5‐dihydro‐2‐oxofuran‐3‐yl)‐28‐deoxonimbolide ( 14 ), 17‐defurano‐17‐(2ξ‐2,5‐dihydro‐2‐hydroxy‐5‐oxofuran‐3‐yl)‐28‐deoxonimbolide ( 15 ), and 17‐defurano‐17‐(5ξ‐2,5‐dihydro‐5‐hydroxy‐2‐oxofuran‐3‐yl)‐2′,3′‐dehydrosalannol ( 17 ), were isolated from an EtOH extract of the leaf of neem (Azadirachta indica). The structures of the new compounds were elucidated on the basis of extensive spectroscopic analyses and comparison with literature. Upon evaluation of the cytotoxic activities of these compounds against leukemia (HL60), lung (A549), stomach (AZ521), and breast (SK‐BR‐3) cancer cell lines, seven compounds, i.e., 1 – 3, 12, 13, 15 , and 16 , exhibited potent cytotoxicities with IC50 values in the range of 0.1–9.9 μM against one or more cell lines. Among these compounds, cytotoxicity of nimonol ( 1 ; IC50 2.8 μM ) against HL60 cells was demonstrated to be mainly due to the induction of apoptosis by flow cytometry. Western blot analysis suggested that compound 1 induced apoptosis via both the mitochondrial and death receptor‐mediated pathways in HL60 cells. In addition, when compounds 1 – 17 were evaluated for their inhibitory activities against melanogenesis in B16 melanoma cells, induced with α‐melanocyte‐stimulating hormone (α‐MSH), seven compounds, 1, 2, 4 – 6, 15 , and 16 , exhibited inhibitory activities with 31–94% reduction of melanin content at 10 μM concentration with no or low toxicity to the cells (82–112% of cell viability at 10 μM ). All 17 compounds were further evaluated for their inhibitory effects against the Epstein? Barr virus early antigen (EBV‐EA) activation induced by 12‐O‐tetradecanoylphorbol‐13‐acetate (TPA) in Raji cells.  相似文献

We have shown earlier that Neem (Azadirachta indica) bark aqueous extract has potent antisecretory and antiulcer effects in animal models and has no significant adverse effect (Bandyopadhyay et al., Life Sciences, 71, 2845-2865, 2002). The objective of the present study was to investigate whether Neem bark extract had similar antisecretory and antiulcer effects in human subjects. For this purpose, a group of patients suffering from acid-related problems and gastroduodenal ulcers were orally treated with the aqueous extract of Neem bark. The lyophilised powder of the extract when administered for 10 days at the dose of 30 mg twice daily caused a significant (p < 0.002) decrease (77%) in gastric acid secretion. The volume of gastric secretion and its pepsin activity were also inhibited by 63% and 50%, respectively. Some important blood parameters for organ toxicity such as sugar, urea, creatinine, serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, albumin, globulin, hemoglobin levels and erythrocyte sedimentation rate remained close to the control values. The bark extract when taken at the dose of 30-60 mg twice daily for 10 weeks almost completely healed the duodenal ulcers monitored by barium meal X-ray or by endoscopy. One case of esophageal ulcer (gastroesophageal reflux disease) and one case of gastric ulcer also healed completely when treated at the dose of 30 mg twice daily for 6 weeks. The levels of various blood parameters for organ toxicity after Neem treatment at the doses mentioned above remained more or less close to the normal values suggesting no significant adverse effects. Neem bark extract thus has therapeutic potential for controlling gastric hypersecretion and gastroesophageal and gastroduodenal ulcers.  相似文献

Azadirachta indica (neem) exudate gum was treated with pronase for 48 h followed by chromatography on TEAE-cellulose and the major polysaccharide was isolated. The polysaccharide covalently associated with remnant protein, was homogeneous as indicated by rechromatography on TEAE-cellulose, paper electrophoresis, gel chromatography under dissociating conditions on Bio-Gel P-l00 and P-300. The monosaccharide units, galactose, arabinose, glucuronic acid, fucose and glucosamine were present in a molar ratio of 86 ∶ 70 ∶ 30 ∶ 10 ∶ 1. Thirteen amino acids constituted the protein portion. The linkage between the polysaccharide and the protein was a glucosaminyl asparginyl bond. Limited hydrolysis showed that fucose and arabinose were at the non-reducing ends of the polysaccharide and galactose and glucuronic acid were in the central core.  相似文献

The antibacterial activity of Karanj (Pongamia pinnata) and Neem (Azadirachta indica) seed oil in vitro against fourteen strains of pathogenic bacteria was assessed. Using the tube dilution technique, it was observed that 57.14 and 21.42% of the pathogens were inhibited at 500 microl/ml; 14.28 and 71.42% at 125 microl/ml; and 28.57 and 7.14% at 250 microl/ml of Karanj and Neem oils, respectively. The activity with both the oils was bactericidal and independent of temperature and energy. Most of the pathogens were killed more rapidly at 4 degrees C than 37 degrees C. The activity was mainly due to the inhibition of cell-membrane synthesis in the bacteria.  相似文献

Human pancreatic α-amylase (HPA) inhibitors offer an effective strategy to lower postprandial hyperglycemia via control of starch breakdown. Limonoids from Azadirachta indica known for their therapeutic potential were screened for pancreatic α-amylase inhibition, a known anti-diabetic target. Studies were carried out to reveal their mode of action so as to justify their hypoglycemic potential. Of the nine limonoids isolated/semi-synthesized from A.indica and screened for α-amylase inhibition, azadiradione and exhibited potential inhibition with an IC50 value of 74.17 and 68.38 μM, respectively against HPA under in vitro conditions. Further screening on AR42J α-amylase secretory cell line for cytotoxicity and bioactivity revealed that azadiradione and gedunin exhibited cytotoxicity with IC50 of 11.1 and 13.4μM. Maximal secreted α-amylase inhibition of 41.8% and 53.4% was seen at 3.5 and 3.3μM, respectively. Michaelis-Menten kinetics suggested a mixed mode of inhibition with maltopentaose (K i 42.2, 18.6 μM) and starch (K i 75.8, 37.4 μM) as substrate with a stiochiometry of 1:1 for both azadiradione and gedunin, respectively. The molecular docking simulation indicated plausible π-alkyl and alkyl-alkyl interactions between the aromatic amino acids and inhibitors. Fluorescence and CD confirmed the involvement of tryptophan and tyrosine in ligand binding to HPA. Thermodynamic parameters suggested that binding is enthalpically and entropically driven with ΔG° of -21.25 kJ mol-1 and -21.16 kJ mol-1 for azadiradione and gedunin, respectively. Thus, the limonoids azadiradione and gedunin could bind and inactivate HPA (anti-diabetic target) and may prove to be lead drug candidates to reduce/control post-prandial hyperglycemia.  相似文献

Witches' broom and pod rot are the two most devastating diseases of cocoa in South America and Africa, respectively. Their control by means of phytosanitation and chemical fungicides is labor-intensive, costly and, in many cases, environmentally undesirable. Therefore efforts are made in order to identify alternative, environmentally safe and cost-efficient methods for the control of these pathogens. Promising candidates are components of the neem tree (Azadirachta indica), that have been used for centuries in Asia as insecticides, fungicides, anticonceptionals in popular medicine. Here we report about tests on the effect of various concentrations of extracts from neem leaves on growth of mycelia of Crinipellis and Phytophthora and on germination of spores of Crinipellis. We show a 35% growth reduction of mycelia of Phytophthora on neem leaf extract media, whereas growth of mycelia of Crinipellis was not affected, even at the highest concentration of neem leaf extracts used (35%). However, the most dramatic effect of neem leaf extracts is observed on Crinipellis spore germination, here the extracts (20-35%) reduced germination almost completely. Based on these results, we believe that the neem tree might be a source for the production, on small and medium scale, of an effective and cheap formulation for the control of Crinipellis and Phytophthora.  相似文献

Effects of feeding of powdered dry leaves of A. indica (AI) were investigated on humoral and cell mediated immune responses, in a flock of broilers which had survived an outbreak of infectious bursal disease (IBD). AI (2 g/kg) treatment significantly enhanced the antibody titres against new castle disease virus (NCDV) antigen and also potentiated the inflammatory reactions to 1. Chloro-2,4-di- nitro benzene (DNCB) inskin contact test. The results indicate that AI could be beneficial in immunosuppressed condition like IBD, in poultry.  相似文献

The active principle in a methanolic extract of the laboratory-grown cyanobacterium, Fischerella sp. isolated from Neem (Azadirachta indica) tree bark was active against Mycobacterium tuberculosis, Enterobacter aerogenes, Staphylococcus aureus, Pseudomonas aeruginosa, Salmonella typhi, Escherichia coli as well as three multi-drug resistant E. coli strains in in vitro assays. Based on MS, UV, IR 1H NMR analyses the active principle is proposed to be hapalindole T having the empirical formula C21H23N2ClSO and a molecular weight of 386 with the melting point range 179–182 °C. The estimated production of Hapalindole T from the cyanobacterium is 1.25 mg g−1 lyophilized biomass. It is suggested that cyanobacteria colonizing specialized niches such as tree bark could be an antibacterial drug resource.  相似文献

A laboratory experiment was conducted to examine the potentiality of a natural resource neem (Azadirachta indica) seed kernel powder (NSKP) to reduce the urease and nitrification activities in different soils (viz., normal, acid, and sodic) at contrasting moisture (1:1 soil to water and field capacity) and temperature regimes (10 degrees C and 37 degrees C). Results have revealed that application of NSKP with urea did not exhibit any urease inhibitory property in normal and sodic soils, but in acid soil it had maintained higher concentration of urea than the urea alone treated samples for two weeks after application. At 37 degrees C and under field capacity moisture level, urea hydrolysis was more rapid than at 10 degrees C and under waterlogged (1:1) conditions. The NSKP has showed variable effects (4-28%) to inhibit nitrification during 7-21 days after application, depending upon the soil types, temperature and moisture regimes. The nitrification activity was significantly low in acid soil followed by normal and sodic soils. The present study suggests that NSKP has the potential to retard the urease activity in acid soil, and nitrification in all the soils, and thus it may be used along with urea for the better use of applied -N.  相似文献

The density of neem trees in parts of Malindi District in eastern Kenya was assessed to determine the sustainability of their use in the local woodcarving industry. Size and age structure of neem populations under different ownership were analyzed to establish the Annual Allowable Cut. Neem is an excellent carving wood and the available quantities along the Kenyan coast are sufficient to sustainably supply the industry of the whole country. However, because the resource comes from small farm holdings and the supply system to carving groups is not well established, further efforts are required to introduce a management regime that could be certified under the standards of the Forest Stewardship Council.  相似文献

The antisecretory and antiulcer effects of aqueous extract of Neem (Azadirachta indica) bark have been studied along with its mechanism of action, standardisation and safety evaluation. The extract can dose dependently inhibit pylorus-ligation and drug (mercaptomethylimidazole)-induced acid secretion with ED(50) value of 2.7 and 2 mg Kg(-1) b.w. respectively. It is highly potent in dose-dependently blocking gastric ulcer induced by restraint-cold stress and indomethacin with ED(50) value of 1.5 and 1.25 mg Kg(-1) b.w. respectively. When compared, bark extract is equipotent to ranitidine but more potent than omeprazole in inhibiting pylorus-ligation induced acid secretion. In a stress ulcer model, it is more effective than ranitidine but almost equipotent to omeprazole. Bark extract inhibits H(+)-K(+)-ATPase activity in vitro in a concentration dependent manner similar to omeprazole. It offers gastroprotection against stress ulcer by significantly preventing adhered mucus and endogenous glutathione depletion. It prevents oxidative damage of the gastric mucosa by significantly blocking lipid peroxidation and by scavenging the endogenous hydroxyl radical ((z.rad;)OH)-the major causative factor for ulcer. The (z.rad;)OH-mediated oxidative damage of human gastric mucosal DNA is also protected by the extract in vitro. Bark extract is more effective than melatonin, vitamin E, desferrioxamine and alpha-phenyl N-tert butylnitrone, the known antioxidants having antiulcer effect. Standardisation of the bioactive extract by high pressure liquid chromatography indicates that peak 1 of the chromatogram coincides with the major bioactive compound, a phenolic glycoside, isolated from the extract. The pharmacological effects of the bark extract are attributed to a phenolic glycoside which is apparently homogeneous by HPLC and which represents 10% of the raw bark extract. A single dose of 1g of raw extract per kg b.w. (mice) given in one day and application of 0.6g raw extract per kg b.w. per day by oral route over 15 days to a cumulative dose of 9g per kg was well tolerated and was below the LD(50). It is also well tolerated by rats with no significant adverse effect. It is concluded that Neem bark extract has therapeutic potential for the control of gastric hyperacidity and ulcer.  相似文献

Research into the systems of agriculture based on vegetative fallows should begin from enquiries into the shrubs and trees most efficient in restoring soil productivity and providing firewood, timber, fodder and other products essential to meeting basic needs at the rural household and community level. Local processing of such products can give rise to the establishment of thriving village industries and thus generate employment and additional income. Neem tree,Azadirachta indica, of the semiarid regions is one outstanding example of a species which is not only an efficient soil ameliorant but can also provide a series of products highly competitive in price and quality with petroleum-based synthetics. Enough scientific evidence has been gathered from several hundred references on the subject to warrant the launching of a well coordinated multidisciplinary research and development programme leading to agricultural, industrial and commercial exploitation of this tree. Proposals for the implementation of several projects within the programme are briefly outlined in this paper.  相似文献

Neem (Azadirachta indica) extract (NE) and NE combined with copper sulfate and boric acid (NECB) were examined for their protective effect against fungal deterioration of mango (Mangifera indica) and rain tree (Albizia saman) wood. Growth of the white-rot fungus Schizophyllum commune was completely inhibited on solid medium containing 1.8% (w/w) NE or 5% (w/w) NECB. The average weight losses of NE and NECB treated wood blocks inoculated with S. commune were respectively 4.7% and 3.3% for M. indica and 4.1% and 3.0% for A. saman. These numbers were significantly lower compared to those obtained in the untreated condition. Similar observations were also noticed in the case of field tests. An average increase of life span of M. indica and A. saman due to NE treatment compared to untreated samples was about 6–7 times higher. Therefore, both NE and NECB treatments are promising preservation options for enhancing the durability of both M. indica and A. saman woods.  相似文献

The neem tree (Azadirachta indica) contains promising pest-control substances found effective against many economically important pests. These materials are easy to process by village-level industries and easy to use by limited-resource farmers, thereby offering potential for crop protection and off-farm income generation. The tree’s numerous pharmacological and other complementary uses make it doubly attractive for incorporation in large-scale rural development efforts.  相似文献

The objectives of this study were to estimate the outcrossing rate and to explain genetic consequences of the development of seed in the endocarp in a natural population of neem in Bangladesh. Cotyledons of germinated open-pollinated seeds of individual trees were analyzed by starch-gel electrophoresis to examine allozymes. Three loci with clear Mendelian segregation were used to estimate outcrossing rate. A multilocus mixed mating model was used to evaluate the mating system. The population exhibited high outcrossing rates both for multilocus (tm=0.90±0.024) and mean single-locus (ts=0.92±0.020) estimates. The difference between these two parameters (tm–ts=0±SE 0.038) was insignificant, indicating that there was no ’biparental inbreeding’ in the population. The degree of variance of the estimates of multilocus outcrossing rates decreased when two or more loci were included. In order to elucidate the significance of polycarpy a total of 471 seeds were counted out of 440 endocarps. This mechanism appears to be a possible way of avoiding inbreeding.
