Ear photosynthesis may be an important source of C for grain growth in water-stressed plants of cereals. The main objectives of this work were to determine the stability of the photosynthetic apparatus and the photochemical efficiency of ears in plants subjected to post-anthesis drought. Plants of wheat ( Triticum aestivum L. cv. Granero INTA) were grown in pots under a rain shelter and subjected to water stress (soil water potential around −0.6 to −0.8 MPa) starting 4  days after anthesis. Post-anthesis drought substantially accelerated the loss of chlorophyll, Rubisco and the light-harvesting complex of photosystem II (LHCII) in the flag leaf, but the degradation of these photosynthetic components was much less affected by water deficit in awns and ear bracts. Quantum yield of PSII (ΦPSII) decreased in leaves of water-stressed plants. In contrast, ear bracts had a higher ΦPSII than leaves, and ΦPSII of ear bracts did not decrease at all in response to drought. Removing the grains immediately before fluorescence measurements (less than 30 min) slightly reduced ΦPSII, indicating that CO2 supplied by grain respiration may contribute to the high photochemical efficiency of ears in droughted plants. However, other factors may be involved in maintaining high ΦPSII, since even in the absence of grains ΦPSII remained much higher in ear bracts than in the flag leaf. The relative stability of ear photosynthetic components and their relatively high photochemical efficiency may help to maintain ear photosynthesis during the grain filling period in droughted plants.  相似文献

Plants grown in an environment of elevated CO2 and temperature often show reduced CO2 assimilation capacity, providing evidence of photosynthetic downregulation. The aim of this study was to analyse the downregulation of photosynthesis in elevated CO2 (700 µmol mol−1) in nodulated alfalfa plants grown at different temperatures (ambient and ambient + 4°C) and water availability regimes in temperature gradient tunnels. When the measurements were taken in growth conditions, a combination of elevated CO2 and temperature enhanced the photosynthetic rate; however, when they were carried out at the same CO2 concentration (350 and 700 µmol mol−1), elevated CO2 induced photosynthetic downregulation, regardless of temperature and drought. Intercellular CO2 concentration measurements revealed that photosynthetic acclimation could not be accounted for by stomatal limitations. Downregulation of plants grown in elevated CO2 was a consequence of decreased carboxylation efficiency as a result of reduced rubisco activity and protein content; in plants grown at ambient temperature, downregulation was also induced by decreased quantum efficiency. The decrease in rubisco activity was associated with carbohydrate accumulation and depleted nitrogen availability. The root nodules were not sufficiently effective to balance the source–sink relation in elevated CO2 treatments and to provide the required nitrogen to counteract photosynthetic acclimation.  相似文献

Mesophyll cells isolated from Phaseolus vulgaris and Lycopersicon esculentum show decreasing photosynthetic rates when suspended in media containing increasing concentrations of osmoticum. The photosynthetic activity was sensitive to small changes in osmotic potential over a range of sorbitol concentrations from 0.44 M (−1.08 MPa) to 0.77 M (−1.88 MPa). Photorespiration assayed by 14CO2 release in CO2-free air and by 14CO2 release from the oxidation of [1–14C] glycolate also decreased as the osmotic potential of the incubation medium was reduced. The CO2 compensation points of the cells increased with increasing concentration of osmoticum from approximately 60 μ I−11 at −1.08 MPa to 130 μl 1−1 for cells stressed at −1.88 MPa. Changes in photosynthetic and photorespiratory activities occurred at moderate osmotic potentials in these cells suggesting that in whole leaves during a reduction in water potential, non- stomatal inhibition of CO2 assimilation and glycolate pathway metabolism occurs simultaneously with stomatal closure.  相似文献

Variation in stomatal development and physiology of mature leaves from Alnus glutinosa plants grown under reference (current ambient, 360 μmol mol−1 CO2) and double ambient (720 μmol mol−1 CO2) carbon dioxide (CO2) mole fractions is assessed in terms of relative plant growth, stomatal characters (i.e. stomatal index and density) and leaf photosynthetic characters. This is the first study to consider the effects of elevated CO2 concentration on the distribution of stomata and epidermal cells across the whole leaf and to try to ascertain the cause of intraleaf variation. In general, a doubling of the atmospheric CO2 concentration enhanced plant growth and significantly increased stomatal index. However, there was no significant change in relative stomatal density. Under elevated CO2 concentration there was a significant decrease in stomatal conductance and an increase in assimilation rate. However, no significant differences were found for the maximum rate of carboxylation ( V cmax) and the light saturated rate of electron transport ( J max) between the control and elevated CO2 treatment.  相似文献

Carbon dioxide and abscisic acid (ABA) are two major signals triggering stomatal closure. Their putative interaction in stomatal regulation was investigated in well-watered air-grown or double CO2-grown Arabidopsis thaliana plants, using gas exchange and epidermal strip experiments. With plants grown in normal air, a doubling of the CO2 concentration resulted in a rapid and transient drop in leaf conductance followed by recovery to the pre-treatment level after about two photoperiods. Despite the fact that plants placed in air or in double CO2 for 2 d exhibited similar levels of leaf conductance, their stomatal responses to an osmotic stress (0·16–0·24 MPa) were different. The decrease in leaf conductance in response to the osmotic stress was strongly enhanced at elevated CO2. Similarly, the drop in leaf conductance triggered by 1 μ M ABA applied at the root level was stronger at double CO2. Identical experiments were performed with plants fully grown at double CO2. Levels of leaf conductance and carbon assimilation rate measured at double CO2 were similar for air-grown and elevated CO2-grown plants. An enhanced response to ABA was still observed at high CO2 in pre-conditioned plants. It is concluded that: (i) in the absence of stress, elevated CO2 slightly affects leaf conductance in A. thaliana ; (ii) there is a strong interaction in stomatal responses to CO2 and ABA which is not modified by growth at elevated CO2.  相似文献

In May, greenhouse tomato ( Lycopersicon esculentum Mill.) plants near the end of their winter production cycle were shown to exhibit a diurnal photosynthetic decrease. In order to identify the physiological causes of this decline, we compared in May the photosynthetic characteristics of the fifth youngest leaves from tomato plants of different ages corresponding to a winter production (11-month-old plants) and to a spring production (5-month-old plants). Although the leaves were developed simultaneously under the same environmental conditions, only the ones from the winter production showed a diurnal decline of the in situ CO2 assimilation rate (A CO 2). This was accompanied by a decline of internal CO2 and stomatal conductance and by large accumulations of hexoses. When stomatal closure was relieved under saturated CO2 concentration (5%) using a leaf-disc electrode system, the fifth leaves of both tomato cultures had similar maximum quantum efficiency of O2 evolution (Φmax), light-saturated rate of O2 evolution (Pmax) and quantum efficiency of photosystem II (PSII) photochemistry (ΔF/F'm, q P and q N ). We concluded that the diurnal decline of A CO 2 observed in winter tomato production during May originates from a stomatal limitation that is not dependent on environmental conditions but rather related to the developmental stage of the plants.  相似文献

Besides the paucity of information on the effects of drought stress on photosynthesis and yield in sweet potato [ Ipomoea batatas (L.) Lam.], available reports are also contradictory. The aim of this study was to shed light on the effects of long-term restricted water supply on shoot development, photosynthesis and storage root yield in field-grown sweet potato. Experiments were conducted under a rainout shelter where effects of restricted water supply were assessed in two varieties (Resisto and A15). Large decreases in stomatal conductance occurred in both varieties after 5 weeks of treatment. However, continued measurements revealed a large varietal difference in persistence of this response and effects on CO2 assimilation. Although restricted water supply decreased leaf relative water content similarly in both varieties, the negative effects on stomatal conductance disappeared with time in A15 (indicating high drought acclimation capacity) but not in Resisto, thus leading to inhibition of CO2 assimilation in Resisto. Chlorophyll a fluorescence measurements, and the relationship between stomatal conductance, intercellular CO2 concentration and CO2 assimilation rate, indicated that drought stress inhibited photosynthesis primarily through stomatal closure. Although yield loss was considerably larger in Resisto, it was also reduced by up to 60% in A15, even though photosynthesis, expressed on a leaf area basis, was not inhibited in this variety. In A15 yield loss appears to be closely associated with decreased aboveground biomass accumulation, whereas in Resisto, combined effects on biomass accumulation and photosynthesis per unit leaf area are indicated, suggesting that research aimed at improving drought tolerance in sweet potato should consider both these factors.  相似文献

Spruce seedlings [ Picea abies (L.) Karst.] were exposed to a range of concentrations of Zn, Cd, Hg and methyl-Hg for 5 weeks. The chlorophyll and water content of the needles were then estimated. The rates of photosynthesis, transpiration and dark respiration of the intact plant were determined using a Li-cor portable photosynthesis measuring system. Chlorophyll and water contents of needles decreased in response to all metal treatments, as did CO2 uptake. At 1 μ M Cd, 0.1 μ M Hg and 30 and 60 μ M Zn, the decrease in CO2 uptake could be accounted for by decreased chlorophyll concentrations. Decreased transpiration was only found at 5 μ M Cd and 0.01 μ M methyl-Hg. At 5 μ M Cd most of the decrease in CO2 uptake could be explained by decreased chlorophyll levels and stomatal closure induced by water stress. At 0.01 μ M methyl-Hg, besides a decrease in chlorophyll concentration and partial stomatal closure, photosynthetic reactions may have been directly affected. Respiration rates were not influenced by exposure to heavy metals.  相似文献

The hypothesis that increases in cytosolic free calcium ([Ca2+]i) are a component of the CO2 signal transduction pathway in stomatal guard cells of Commelina communis has been investigated. This hypothesis was tested using fura-2 fluorescence ratio photometry to measure changes in guard cell [Ca2+]i in response to challenge with 700 µl l−1 CO2. Elevated CO2 induced increases in guard cell [Ca2+]i which were similar to those previously reported in response to abscisic acid. [Ca2+]i returned to resting values following removal of the CO2 and further application of CO2 resulted in a second increase in [Ca2+]i. This demonstrated that the CO2-induced increases in [Ca2+]i were stimulus dependent. Removal of extracellular calcium both prevented the CO2-induced increase in [Ca2+]i and inhibited the associated reduction in stomatal aperture. These data suggest that Ca2+ acts as a second messenger in the CO2 signal transduction pathway and that an increase in [Ca2+]i may be a requirement for the stomatal response to CO2.  相似文献

The effects of exogenous application of glycinebetaine (GB) (10 m M ) on growth, leaf water content, water use efficiency, photosynthetic gas exchange, and photosystem II photochemistry were investigated in maize plants subjected to salt stress (50 and 100 m M NaCl). Salt stress resulted in the decrease in growth and leaf relative water content as well as net photosynthesis and the apparent quantum yield of photosynthesis. Stomatal conductance, evaporation rate, and water use efficiency were decreased in salt-stressed plants. Salt stress also caused a decrease in the actual efficiency of PSII ( Φ PSII), the efficiency of excitation energy capture by open PSII reaction centers ( F v'/ F m'), and the coefficients of photochemical quenching ( q P) but caused an increase in non-photochemical quenching (NPQ). Salt stress showed no effects on the maximal efficiency of PSII photochemistry ( F v/ F m). On the other hand, in salt-stressed plants, GB application improved growth, leaf water content, net photosynthesis, and the apparent quantum yield of photosynthesis. GB application also increased stomatal conductance, leaf evaporation rate, and water use efficiency. In addition, GB application increased Φ PSII, F v'/ F m', and q P but decreased NPQ. However, GB application showed no effects on F v/ F m. These results suggest that photosynthesis was improved by GB application in salt-stressed plants and such an improvement was associated with an improvement in stomatal conductance and the actual PSII efficiency.  相似文献

We developed and applied an ecosystem-scale model that calculated leaf CO2 assimilation, stomatal conductance, chloroplast CO2 concentration and the carbon isotope composition of carbohydrate formed during photosynthesis separately for sunlit and shaded leaves within multiple canopy layers. The ecosystem photosynthesis model was validated by comparison to leaf-level gas exchange measurements and estimates of ecosystem-scale photosynthesis from eddy covariance measurements made in a coastal Douglas-fir forest on Vancouver Island. A good agreement was also observed between modelled and measured δ 13C values of ecosystem-respired CO2 ( δ R). The modelled δ R values showed strong responses to variation in photosynthetic photon flux density (PPFD), air temperature, vapour pressure deficit (VPD) and available soil moisture in a manner consistent with leaf-level studies of photosynthetic 13C discrimination. Sensitivity tests were conducted to evaluate the effect of (1) changes in the lag between the time of CO2 fixation and the conversion of organic matter back to CO2; (2) shifts in the proportion of autotrophic and heterotrophic respiration; (3) isotope fractionation during respiration; and (4) environmentally induced changes in mesophyll conductance, on modelled δ R values. Our results indicated that δ R is a good proxy for canopy-level C c/ C a and 13C discrimination during photosynthetic gas exchange, and therefore has several applications in ecosystem physiology.  相似文献

To determine how increased atmospheric CO2 will affect the physiology of coppiced plants, sprouts originating from two hybrid poplar clones ( Populus trichocarpa × P. deltoides - Beaupre and P. deltoides × P. nigra - Robusta) were grown in open-top chambers containing ambient or elevated (ambient + 360 μmol mol−1) CO2 concentration. The effects of elevated CO2 concentration on leaf photosynthesis, stomatal conductance, dark respiration, carbohydrate concentration and nitrogen concentration were measured. Furthermore, dark respiration of leaves was partitioned into growth and maintenance components by regressing specific respiration rate vs specific growth rate. Sprouts of both clones exposed to CO2 enrichment showed no indication of photosynthetic down-regulation. During reciprocal gas exchange measurements, CO2 enrichment significantly increased photosynthesis of all sprouts by approximately 60% ( P < 0.01) on both an early and late season sampling date, decreased stomatal conductance of all sprouts by 10% ( P < 0.04) on the early sampling date and nonsignificantly decreased dark respiration by an average of 11%. Growth under elevated CO2 had no consistent effect on foliar sugar concentration but significantly increased foliar starch by 80%. Respiration rate was highly correlated with both specific growth rate and percent nitrogen. Long-term CO2 enrichment did not significantly affect the maintenance respiration coefficient or the growth respiration coefficient. Carbon dioxide enrichment affected the physiology of the sprouts the same way it affected these plants before they were coppiced.  相似文献

LIMITATIONS OF PHOTOSYNTHESIS IN DIFFERENT REGIONS OF THE ZEA MAYS LEAF   总被引:3,自引:0,他引:3  
The progressive development of the photosynthetic apparatus occurring along the length of the Zea mays leaf offers a convenient system with which to examine the limitations to photosynthetic CO2 assimilation during biogenesis of a C4 leaf. Changes in light-induced O2 evolution and CO2 assimilation, chlorophyll content, activity of PEP-carboxylase, NADP-malic enzyme and the 'R5P system' (consisting of d -ribose-5-phosphate-keto isomerase, ATP- d -ribulose-5 phosphate 1-phosphotransferase and d -ribulose-1,5-bisphosphate carboxylase) and fluorescence emission characteristics were examined along the length of the second leaf of 7-day-old plants grown under a diurnal light regime. The results suggest that the major limitation to CO2 assimilation in the leaf sheath lies within the chlorenchyma and is either energy supply for carboxylation or the capacity of key photosynthetic enzymes. In the leaf blade stomatal resistance to CO2 diffusion constitutes a major fraction of the total leaf resistance to CO2 assimilation implicating the stoma as the major limiting factor to photosynthetic CO2 assimilation.  相似文献

Abstract. Poplar shoots ( Populus euramericana L.) obtained from cuttings were exposed for 6 or 8 weeks to NH3 concentrations of 50 and 100 μgm−3 or filtered air in fumigation chambers. After this exposure the rates of NH3 uptake, transpiration, CO2 assimilation and respiration of leaves were measured using a leaf chamber. During the long-term exposure also modulated chlorophyll fluorescence measurements were carried out to obtain information about the photosynthetic performance of individual leaves. Both fluorescence and leaf chamber measurements showed a higher photosynthetic activity of leaves exposed to 100 μg NH3 m−3. These leaves showed also a larger leaf conductance and a larger uptake rate of NH3 than leaves exposed to 50 μg m−3 NH3 or filtered air. The long-term NH3 exposure did not induce an internal resistance against NH3 transport in the leaf, nor did it affect the leaf cuticle. So, not only at a short time exposure, but also at a long-term exposure NH3 uptake into leaves can be calculated from data on the boundary layer and stomatal resistance for H2O and ambient NH3-concentration. Furthermore, the NH3 exposure had no effect on the relation between CO2-assimilation and stomatal conductance, indicating that NH3 in concentrations up to 100 μg m−3 has no direct effect on stomatal behaviour; for example, by affecting the guard or contiguous cells of the stomata.  相似文献

Abstract. The effect of short-term SO2 fumigation on photosynthesis and transpiration of Vicia faba L. was measured at different irradiances and SO2 concentrations. At high irradiances photosynthetic rates were reduced when leaves were exposed to SO2 and the magnitude of the reduction was linearly related to the rate of SO2 uptake through the stomata. Photosynthetic rates stabilized within 2 h after the start of fumigation.
The effect of SO2 on photosynthesis was measured at different CO2 concentrations to analyse the contribution of stomatal and non-stomatal factors to photosynthetic inhibition. Mesophyll resistance to CO2 diffusion increased as a result of SO2 exposure and caused a rapid reduction in photosynthesis after the start of fumigation. Stomatal resistance was not affected directly by SO2 fumigation, but indirectly as a result of a feedback loop between net photosynthesis and internal CO2 concentration.
Analysis of gas-exchange measurements in biochemical terms indicated that photosynthetic inhibition during SO2 exposure can be explained by a stronger reduction in the affinity of RBP carboxylase/oxygenase for CO2 than for O2.  相似文献

The oxygen requirement for stomatal opening in maize plants ( Zea mays L. hybrid INRA 508) was studied at different CO2 concentrations and light intensities. In the absence of CO2, stomatal opening always required O2, but this requirement decreased with increasing light intensity. In darkness, the lowest O2 partial pressure needed to obtain a weak stomatal movement was about 50 Pa. This value was lowered to ca 10 Pa in light (320 μmol m−2 s−1).
On the other hand. in the absence of O2, CO2enabled stomatal opening to occur in the light, presumably due to the evolved photosynthetic O2. Thus, CO2, which generally reduced stomatal aperture, could induce stomatal movement in anoxia and light. The effect of CO2 on stomatal opening was closely dependent on O2 concentration and light intensity. Stomatal aperture appeared CO2-independent at an O2 partial pressure which was dependent on light intensity and was about 25 Pa at 320 umol m−2 s−1.
The presence of a plasmalemma oxidase, in addition to mitochondrial oxidase, might explain the differences in the O2 requirement at various light intensities. The possible involvement of such a system in relation to the effect of CO2 is discussed.  相似文献

Sunflower seedlings ( Helianthus annuus hybrid Select) were grown in a complete nutrient solution in the absence or presence of Cd2+ (10 and 20 μM). Analyses were performed to establish whether there was a differential effect of Cd2+ on mature and young leaves. After 7 d the growth parameters as well as the leaf area had decreased in both mature and young leaves. Accumulation of Cd2+ in the roots exceeded that in the shoots. Seedlings treated with Cd2+ exhibited reduced contents of chlorophyll and CO2 assimilation rate, with a greater decrease in young leaves. The photochemical efficiency of photosystem II (PSII) was not altered by Cd2+ treatment in either mature or young leaves, although during steady-state photosynthesis in young leaves there was a significant alteration in the following parameters: quantum yield of electron transport by PSII (ΦPSII), photochemical quenching ( q P), non-photochemical quenching ( q NP), and excitation capture efficiency of PSII (Φexc).  相似文献

Influences of different nitrogen applications on photosynthesis and utilization of excitation energy were explored by comparing two field-grown wheat ( Triticum aestivum L.) cultivars with high or low grain protein content [High protein cultivar '8901' (HC) and low protein cultivar '1391' (LC)]. High nitrogen application significantly decreased both CO2 assimilation and photorespiration in both cultivars during the early stages after anthesis. However, the actual photosystem II (PSII) efficiency ( Ф PSII) was not significantly different between high, moderate and low nitrogen applications in the HC. As a result, the ratio of Ф PSII to the quantum yield of carbon metabolism ( Φ CO2) measured under non-photorespiratory conditions in the HC was higher under high nitrogen application than under low or medium nitrogen application. The grain protein content of the HC was also increased by high nitrogen application. In contrast, high nitrogen application decreased the actual PSII efficiency in the flag leaves of the LC in the early stages after anthesis and different nitrogen applications did not significantly alter the Ф PSII/ Φ CO2 ratio or grain protein content in the LC. No significant difference was detected in the activity of superoxide dismutase or ascrobate peroxidase between different nitrogen treatments in either cultivar throughout the entire experimental period. These results indicate that more excitation energy is partitioned to nitrogen metabolism in the flag leaves of the HC under high nitrogen application whereas the partitioning of excitation energy in the LC was not affected by nitrogen application.  相似文献

Abstract. A portable apparatus has been constructed to measure simultaneously the quantum yield of CO2 assimilation, light absorption, chlorophyll fluorescence emission and water vapour exchange of attached intact leaves in the field. The core of the instrument is a light-integrating spherical leaf chamber which includes ports for a light source, photosynthetically active radiation sensor, fluorescence probes and gas inlet and outlet manifolds. Measurement of the quantum flux inside the empty chamber and with a leaf present allows determination of leaf absorptance. An open gas exchange system is employed using an infra-red analyser to measure leaf CO2 exchange. Using a DC white light source the quantum yield of CO2 assimilation based on absorbed light (φabs) may be determined rapidly in either ambient air or artificial gas mixtures. Inclusion of capacitance humidity probes into the gas inlet and outlet ports allows simultaneous determination of water vapour exchange and subsequent estimation of stomatal conductance to CO2 and intercellular CO2 concentration. Measurement of fluorescence emission by the sample leaf exposed to white light is achieved by a modulated fluorescence detection system. In addition to determination of the minimal, maximal and variable fluorescence levels, a further analysis allows the photochemical and non-photochemical components of fluorescence quenching, to be estimated. The theory and design of this apparatus is described in detail. The use of the apparatus in the field is demonstrated through a study of the photosynthetic performance of a maize and bean crop during the growing season and by analysis of the photosynthetic performance of crops subjected to nitrogen-stress and a herbicide treatment.  相似文献

Gas exchange, chlorophyll fluorescence and water potentials, together with ascorbate and glutathione concentrations, were studied during moderate and severe drought stress and in response to re-watering in Allocasuarina luehmannii seedlings. Moderate drought stress (MS) decreased stomatal conductance (gs) and net CO2 assimilation rates (A) to ∼40% and ∼60% of control values, respectively, and caused decreases in internal CO2 concentration (Ci) and maximum light use efficiency of light-acclimated photosystem II (PSII) centres (Fv'/Fm'). Severe drought stress (SS) decreased gs and A to ∼5% and ∼15% of the control values, respectively, and caused increases in Ci and PSII excitation pressure (1 − qP), as well as decreases in water potentials, effective quantum yield of PSII (ΦPSII), maximum efficiency of PSII (Fv/Fm) and Fv'/Fm'. Ascorbate and glutathione concentrations remained unaffected by drought treatments, but ascorbate became more oxidised under severe stress. MS seedlings recovered within 1 day (Ci, Fv'/Fm') to 1 week (A, gs) of re-watering. In comparison, SS seedlings had longer-lasting after-stress effects, with recovery of many variables (gs, water potentials, Fv/Fm, ΦPSII, Fv'/Fm') taking between 1 and 3 weeks from re-watering. We found no indication that interaction with antioxidants played a significant role in recovery. In conclusion, A. luehmannii seedlings appear to function normally under moderate drought, but do not seem to have particular metabolic tolerance mechanisms to endure severe drought, which may have implications for its persistence under climate change at the drier margins of its distribution.  相似文献
