Glyoxylate decarboxylation during photorespiration   总被引:4,自引:0,他引:4

At 25° C under aerobic conditions with or without gluamate 10% of the [1-14C]glycollate oxidised in spinach leaf peroxisomes was released as 14CO2. Without glutamate only 5% of the glycollate was converted to glycine, but with it over 80% of the glycollate was metabolised to glycine. CO2 release was probably not due to glycine breakdown in these preparations since glycine decarboxylase activity was not detected. Addition of either unlabelled glycine or isonicotinyl hydrazide (INH) did not reduce 14CO2 release from either [1-14C]glycollate or [1-14C]glyoxylate. Furthermore, the amount of available H2O2 (Grodzinski and Butt, 1976) was sufficient to account for all of the CO2 release by breakdown of glyoxylate. Peroxisomal glycollate metabolism was unaffected by light and isolated leaf chloroplasts alone did not metabolise glycollate. However, in a mixture of peroxisomes and illuminated chloroplasts the rate of glycollate decarboxylation increased three fold while glycine synthesis was reduced by 40%. Although it was not possible to measure available H2O2 directly, the data are best explained by glyoxylate decarboxylation. Catalase reduced CO2 release and enhanced glycine synthesis. In addition, when a model system in which an active preparation of purified glucose oxidase generating H2O2 at a known rate was used to replace the chloroplasts, similar rates of 14CO2 release and [14C]glycine synthesis from [1-14C]glycollate were measured. It is argued that in vivo glyoxylate metabolism in leaf peroxisomes is a key branch point of the glycollate pathway and that a portion of the photorespired CO2 arises during glyoxylate decarboxylation under the action of H2O2. The aim of this study was to investigate the physiological basis of increased root exudation from Festuca rubra, in response to defoliation. The hypothesis, that assimilate supply to roots is a key determinant of the response of root exudation to defoliation was tested by imposing CO2-deplete (< 50 mol mol–1) atmospheres to F. rubra. This was done as a non-destructive means of preventing supply of new assimilate to roots of intact and defoliated plants. F. rubra was grown in axenic sand systems, with defoliation and CO2-depletion treatments applied to plants at 14 and 35 days after planting. Root exudation and NO3 uptake were quantified throughout, and post-treatment uptake and allocation of N were determined from the distribution of 15N label, supplied as 15NO3 . Defoliation of F. rubra resulted in significantly (P <0.01) increased root exudation, CO2-depletion did not result in increased exudation from plants of either age. When treatments were applied to F. rubra after 14 days, defoliation and CO2-depletion each reduced NO3 uptake significantly (P <0.05). However, in older plants, uptake of NO3 was less sensitive to defoliation and CO2-depletion. The results indicate that increased root exudation following defoliation is not related directly to reduced assimilate supply to roots. This was evident from the lack of effect of CO2-depletion on root exudation, and the absence of correlation between root-C efflux and the rate of NO3 uptake. The physiological basis of increased exudation following defoliation remains uncertain, but may be dependent on physical damage, either directly or as a consequence of systemic responses to wounding.  相似文献

Previous reports have indicated positive effects of enriched rhizosphere dissolved inorganic carbon on the growth of salinity-stressed tomato (Lycopersicon esculentum L. Mill. cv. F144) plants. In the present work we tested whether a supply of CO2 enriched air to the roots of hydroponically grown tomato plants had an effect on nitrogen uptake in these plants. Uptake was followed over periods of 6 to 12 hours and measured as the depletion of nitrogen from the nutrient solution aerated with either ambient or CO2 enriched air. Enriched rhizosphere CO2 treatments (5000 μmol mol-1) increased NO3 - uptake up to 30% at pH 5.8 in hydroponically grown tomato plants compared to control treatments aerated with ambient CO2 (360 μmol mol-1). Enriched rhizosphere CO2 treatments had no effect on NH3 + uptake. Acetazolamide, an inhibitor of apoplastic carbonic anhydrase, increased NO3 - uptake in ambient rhizosphere CO2 treatments, but had no effect on NO3 - uptake in enriched rhizosphere CO2 treatments. Ethoxyzolamide, an inhibitor of both cytoplasmic and extracellular carbonic anhydrase, decreased NO3 - uptake in ambient rhizosphere CO2 treatments. In contrast, a CO2 enriched rhizosphere increased NO3 - uptake with ethoxyzolamide, although not to the same extent as in plants without ethoxyzolamide. It is suggested that a supply of enriched CO2 to the rhizosphere influenced NO3 - uptake through the formation of increased amounts of HCO3 - in the cytosol. Atmospheric CO2 enrichment is expected to often benefit plant growth, despite causing global warming and nitrogen (N) dilution in plants. Most plants primarily procure N as inorganic nitrate (NO3?) or ammonium (NH4+), using membrane‐localized transport proteins in roots, which are key targets for improving N use. Although interactive effects of elevated CO2, chronic warming and N form on N relations are expected, these have not been studied. In this study, tomato (Solanum lycopersicum) plants were grown at two levels of CO2 (400 or 700 ppm) and two temperature regimes (30 or 37°C), with NO3? or NH4+ as the N source. Elevated CO2 plus chronic warming severely inhibited plant growth, regardless of N form, while individually they had smaller effects on growth. Although %N in roots was similar among all treatments, elevated CO2 plus warming decreased (1) N‐uptake rate by roots, (2) total protein concentration in roots, indicating an inhibition of N assimilation and (3) shoot %N, indicating a potential inhibition of N translocation from roots to shoots. Under elevated CO2 plus warming, reduced NO3?‐uptake rate per g root was correlated with a decrease in the concentration of NO3?‐uptake proteins per g root, reduced NH4+ uptake was correlated with decreased activity of NH4+‐uptake proteins and reduced N assimilation was correlated with decreased concentration of N‐assimilatory proteins. These results indicate that elevated CO2 and chronic warming can act synergistically to decrease plant N uptake and assimilation; hence, future global warming may decrease both plant growth and food quality (%N).  相似文献

The role of photorespiration in the foliar assimilation of nitrate (NO3) and carbon dioxide (CO2) was investigated by measuring net CO2 assimilation, net oxygen (O2) evolution, and chlorophyll fluorescence in tomato leaves (Lycopersicon esculentum). The plants were grown under ambient CO2 with ammonium nitrate (NH4NO3) as the nitrogen source, and then exposed to a CO2 concentration of either 360 or 700 µmol mol?1, an O2 concentration of 21 or 2%, and either NO3 or NH4+ as the sole nitrogen source. The elevated CO2 concentration stimulated net CO2 assimilation under 21% O2 for both nitrogen treatments, but not under 2% O2. Under ambient CO2 and O2 conditions (i.e. 360 µmol mol?1 CO2, 21% O2), plants that received NO3 had 11–13% higher rates of net O2 evolution and electron transport rate (estimated from chlorophyll fluorescence) than plants that received NH4+. Differences in net O2 evolution and electron transport rate due to the nitrogen source were not observed at the elevated CO2 concentration for the 21% O2 treatment or at either CO2 level for the 2% O2 treatment. The assimilatory quotient (AQ) from gas exchange, the ratio of net CO2 assimilation to net O2 evolution, indicated more NO3 assimilation under ambient CO2 and O2 conditions than under the other treatments. When the AQ was derived from gross O2 evolution rates estimated from chlorophyll fluorescence, no differences could be detected between the nitrogen treatments. The results suggest that short‐term exposure to elevated atmospheric CO2 decreases NO3 assimilation in tomato, and that photorespiration may help to support NO3 assimilation.  相似文献

Summary The formations of transients in CO2 exchange in the blue-green alga Anacystic nidulans is dependent on the temperature used during the measurements. The algae were grown in a low light intensity (4000 lux) under normal air conditions and measured in the same low CO2 concentration (0.03 vol. %) but under a higher light intensity (10 000 lux). At a temperature of +20°C the stationary rate of CO2 uptake was reached directly. At a temperature of +35°C, on the other hand, a maximum of CO2 uptake could be observed at the beginning of the light period followed by a steady rate of photosynthesis, which was higher than at +20°C. In the beginning of the dark period a CO2 outburst appeared at 35°C.Only at a low temperature (+20°C) did we find a light induced glycollate excretion; after a maximum at 7 1/2 minutes illumination the release of glycollate ceases and the level decreases to a lower value. A similar time course exists during illumination in red light (621 nm, 1.5·10-8 einsteins) and a temperature of +20°C. In blue light (432 nm, 1,5·10-8 einsteins, +20°C) and in white light at a high temperature (+35°C) we could not find any light induced glycollate excretion. Our results are discussed in reference to the photorespiration. We explain the formation of transients in CO2 uptake of Anacystis at a high temperature (+35°C) and in blue light (+20°C) on the basis of the influence of photorespiration.  相似文献

Root proliferation into nutrient rich zones is an important mechanism in the exploitation of soil nutrients by plants. No studies have examined atmospheric CO2 effects on cotton (Gossypium hirsutum L.) root distribution as affected by localized phosphorus (P). Cotton plants were grown in a Troup sand (loamy, thermic Grossarenic Kandiudults) using 17.2-l containers placed in open top field chambers (OTC) under ambient (360 mol mol–1) or enriched (720 mol mol–1) atmospheric CO2 concentrations for 40 days. Equivalent amounts of P were added (150 mg P per kg of soil) to 100, 50, 25, 12.5, and 6.25% of the total soil volume; control containers with no added P were also included. Under extremely low P (controls), cotton was unresponsive to CO2 enrichment. In treatments with both fertilized and unfertilized soil volumes, root proliferation was greater in the unfertilized soil under elevated CO2 conditions. Stimulation of root growth occurred in the P-fertilized soil fraction; the pattern of stimulation was similar under both CO2 levels. Under ambient CO2, cotton plant response was positive (shoot mass, and total root mass and length) when soil P was confined to relatively small proportions of the total soil volume (6.25 and 12.5%). However, elevated CO2 grown plants tended to respond to P regardless of its distribution.  相似文献

Mutant strains of the facultative autotrophic bacterium Alcaligenes eutrophus blocked in glycollate utilization were isolated and characterized. One of the strains, AE161, which lacked glycollate oxidoreductase activity, excreted up to 1.2mol glycollate/mg cell protein per hour during autotrophic growth. This mutant strain was used to study the efficiency of CO2 fixation in terms of how much of the fixed carbon was excreted as glycollate under different conditions. Glycollate excretion was not detected during heterotrophic growth. Only 1% of the total CO2 fixed was excreted as glycollate in an atmosphere of 4% CO2 plus 20% O2. The rate of glycollate excretion showed a large increase and CO2 fixation decreased as the CO2 concentration was lowered. [1-14C]glycollate was oxidised to14CO2 by peroxisomes isolated from leaves of spinach beet about 3 times as rapidly at 35°C as at 25°C; the rate was further increased with rise in temperature to a maximum at 55°C. These increases are shown to be mainly due to the increased H2O2 available to oxidise glyoxylate non-enzymically as a result of the higher temperature coefficient of glycollate oxidase activity relative to that of catalase. These results are compared with similar increases in the rate of14CO2 release between 25°C and 35°C when [1-14C]glycollate was supplied to leaf discs in light or darkness. The effect of varied Zn supply on the pH of the nutrient solution and uptake of cations and anions was studied in cotton (Gossypium hirsutum L.), sunflower (Helianthus annuus L.) and buckwheat (Fagopyrum esculentum Moench) plants grown under controlled environmental conditions in nutrient solutions with nitrate as source of nitrogen. With the appearance of visual Zn deficiency symtoms, the pH of the nutrient solutions decreased from 6 to about 5 whereas the pH increased to about 7 when the plants were adequately supplied with Zn. In Zn deficient plants the pH decrease was associated with a shift in the cation-anion uptake ratio in favour of cation uptake. Of the major ions, uptake of Ca2+ and K+ was either not affected or only slightly lowered whereas NO3 - uptake was drastically decreased in Zn deficient plants. Although the Zn nutritional status of plants hardly affected the NO3 - concentrations in the plants, the leakage of NO3 - from roots of Zn deficient plants into a diluted CaCl2 solution was nearly 10 times higher than that of plants adequately supplied with Zn. In contrast to Zn deficiency, Mn deficiency in cotton plants neither affected NO3 - uptake nor the pH of the nutrient solution.The results indicate that, probably as a consequence of the role of Zn in plasma membrane integrity and nitrogen metabolism, when Zn is deficient in dicotyledonous species net uptake of NO3 - is particularly depressed which in turn results in an increase in cation-anion uptake ratio and a corresponding decrease in external pH. The ecological relevance of this rhizosphere acidification is discussed.  相似文献

Leaf and root tissue of Lolium perenne L., Agrostis capillaris L. and Festuca ovina L. grown under ambient (350 μl l-1 CO2) and elevated (700 μl l-1) CO2 in a continuously 14C-labelled atmosphere and at two soil N levels, were incubated at 14°C for 222 days. Decomposition of leaf and root tissue grown in the low N treatment was not affected by elevated [CO2], whereas decomposition in the high N treatment was significantly reduced by 7% after 222 days. Despite the increased C/N ratio (g g-1) of tissue cultivated at elevated [CO2] when compared with the corresponding ambient tissue, there was no significant correlation between initial C/N ratio and 14C respired. This finding suggests that the CO2-induced changes in decomposition rates do not occur via CO2-induced changes in C/N ratios of plant materials. We combined the decomposition data with data on 14C uptake and allocation for the same plants, and give evidence that elevated [CO2] has the potential to increase soil C stores in grassland via increasing C uptake and shifting C allocation towards the roots, with an inherent slower decomposition rate than the leaves. An overall increase of 15% in 14C remaining after 222 days was estimated for the combined tissues, i.e., the whole plants; the leaves made a much smaller contribution to the C remaining (+6%) than the roots (+26%). This shows the importance of clarifying the contribution of roots and leaves with respect to the question whether grassland soils act as a sink or source for atmospheric CO2. Positive influences of high concentrations of dissolved inorganic carbon (DIC) in the growth medium of salinity-stressed plants are associated with carbon assimilation through phosphoenolpyruvate carboxylase (PEPc) activity in roots; and also in salinity-stressed tomato plants, enriched CO2 in the rhizosphere increases NO?3uptake. In the present study, wild-type and nitrate reductase-deficient plants of barley (Hordeum vulgare L. cv. Steptoe) were used to determine whether the influence of enriched CO2 on NO?3 uptake and metabolism is dependent on the activity of nitrate reductase (NR) in the plant. Plants grown in NH4+and aerated with ambient air, were transferred to either NO3? or NH4+ solutions and aerated with air containing between 0 and 6 500 μmol mol?1 CO2. Nitrogen uptake and tissue concentrations of NO3? and NH4+ were measured as well as activities of NR and PEPc. The uptake of NO?3 by the wild-type was increased by increasing CO2. This was associated with increased in vitro NR activity, but increased uptake of NO3? was found also in the NR-deficient genotype when exposed to high CO2 concentrations; so that the influence of CO2 on NO3? uptake was independent of the reduction of NO3? and assimilation into amino acids. The increase in uptake of NO3? in wild-type plants with enriched CO2 was the same at pH 7 as at pH 5, indicating that the relative abundance of HCO3? or CO2 in the medium did not influence NO3? uptake. Uptake of NH4+ was decreased by enriched CO2 in a pH (5 or 7) independent fashion. Thus NO3? and NH+4 uptakes are influenced by the CO2 component of DIC independently of anaplerotic carbon provision for amino acid synthesis, and CO2 may directly affect the uptake of NO3? and NH4+ in ways unrelated to the NR activity in the tissue.  相似文献

Summary The rate at which H2O2 becomes available during glycollate oxidation for further oxidation reactions, especially that of glyoxylate to formate and CO2, in peroxisomes from spinach-beet (Beta vulgaris L., var. vulgaris) leaves has been determined by measuring O2 uptake in the presence and absence of added catalase. The rates observed under air and pure O2 were sufficient to account for the 14CO2 released from [l-14C]glycollate under these conditions; the two reactions showed similar characteristics. In the course of the reaction, a fall in catalase activity was observed concomitant with an increase in 14CO2 release. Spinach (Spinacia oleracea L.) and barley (Hordeum vulgare L.) were grown in hydroponic culture with varying levels of orthophosphate (Pi). When leaves were fed with 20 mmol·l–1 Pi at low CO2 concentrations, a temporary increase of CO2 uptake was observed in Pi-deficient leaves but not in those from plants grown at 1 mmol·l–1 Pi. At high concentrations of CO2 (at 21% or 2% O2) the Pi-induced stimulation of CO2 uptake was pronounced in the Pi-deficient leaves. The contents of phosphorylated metabolites in the leaves decreased as a result of Pi deficiency but were restored by Pi feeding. These results demonstrate that there is an appreciable capacity for rapid Pi uptake by leaf mesophyll cells and show that the effects of long-term phosphate deficiency on photosynthesis may be reversed (at least temporarily) within minutes by feeding with Pi.Abbreviation Pi orthophosphate  相似文献

The linked utilization of glycollate and L-serine has been studied in peroxisomal preparations from leaves of spinach beet (Beta vulgaris L.). The generation of glycine from glycollate was found to be balanced by the production of hydroxypyruvate from serine and similarly by 2-oxoglutarate when L-glutamate was substituted for L-serine. In the presence of L-malate and catalytic quantities of NAD+, about 40% of the hydroxypyruvate was converted further to glycerate, whereas with substrate quantities of NADH, this conversion was almost quantitative. CO2 was released from the carboxyl groups of both glycollate and serine. Since the decarboxylation of both substrates was greatly in creased by the catalase inhibitor, 3-amino-1,2,4-triazole, and abolished by bovine liver catalase, it was attributed to the nonenzymic attack of H2O2, generated in glycollate oxidation, upon glyoxylate and hydroxypyruvate respectively. At 25–30° C, about 10% of the glyoxylate and hydroxypyruvate accumulated was decarboxylated, and the release of CO2 from each keto-acid was related to the amounts present. It is suggested that hydroxypyruvate decarboxylation might contribute significantly to photorespiration and provide a metabolic route for the complete oxidation of glycollate, the magnitude of this contribution depending upon the concentrations of glyoxylate and hydroxypyruvate in the peroxisomes.  相似文献

In order to better elucidate fixed-C partitioning, nutrient acquisition and water relations of prairie grasses under elevated [CO2], we grew the C4 grass Bouteloua gracilis (H.B.K.) lag ex Steud. from seed in soil-packed, column-lysimeters in two growth chambers maintained at current ambient [CO2] (350 μL L−1) and twice enriched [CO2] (700 μL L−1). Once established, plants were deficit irrigated; growth chamber conditions were maintained at day/night temperatures of 25/16°C, relative humidities of 35%/90% and a 14-hour photoperiod to simulate summer conditions on the shortgrass steppe in eastern Colorado. After 11 weeks of growth, plants grown under CO2 enrichment had produced 35% and 65% greater total and root biomass, respectively, and had twice the level of vesicular-arbuscular mycorrhizal (VAM) infection (19.8% versus 10.8%) as plants grown under current ambient [CO2]. The CO2-enriched plants also exhibited greater leaf water potentials and higher plant water use efficiencies. Plant N uptake was reduced by CO2 enrichment, while P uptake appeared little influenced by CO2 regime. Under the conditions of the experiment, CO2 enrichment increased root biomass and VAM infection via stimulated growth and adjustments in C partitioning below-ground. Our previous work indicated that salinity caused a shift in the predominant site of nitrate reduction and assimilation from the shoot to the root in tomato plants. In the present work we tested whether an enhanced supply of dissolved inorganic carbon (DIC, CO2+ HCO3) to the root solution could increase anaplerotic provision of carbon compounds for the increased nitrogen assimilation in the root of salinity-stressed Lycopersicon esculentum (L.) Mill. cv. F144. The seedlings were grown in hydroponic culture with 0 or 100mM NaCl and aeration of the root solution with either ambient or CO2-enriched air (5000 μmol mol?1). The salinity-treated plants accumulated more dry weight and higher total N when the roots were supplied with CO2-enriched aeration than when aerated with ambient air. Plants grown with salinity and enriched DIC also had higher rates of NO?3 uptake and translocated more NO?3 and reduced N in the xylem sap than did equivalent plants grown with ambient DIC. Incorporation of DIC was measured by supplying a 1 -h pulse of H14CO?3 to the roots followed by extraction with 80% ethanol. Enriched DIC increased root incorporation of DIC 10-fold in both salinized and non-salinized plants. In salinity-stressed plants, the products of dissolved inorganic 14C were preferentially diverted into amino acid synthesis to a greater extent than in non-salinized plants in which label was accumulated in organic acids. It was concluded that enriched DIC can increase the supply of N and anaplerotic carbon for amino acid synthesis in roots of salinized plants. Thus enriched DIC could relieve the limitation of carbon supply for ammonium assimilation and thus ameliorate the influence of salinity on NO?3 uptake and assimilation as well as on plant growth.  相似文献

Summary Plantlets of Nicotiana tabacum (var. Samsun) were grown under CO2 enriched air supplied by a Warburg buffer. Growth of all plant parts was enhanced. The maximum growth increase was found for roots (120%).Addition of 30 g.l-1 sucrose in the medium resulted in a three time faster growth. However, the effect of CO2 enrichment was still positive in these conditions, although less pronounced than in autotrophic cultures.  相似文献

 Oxygen and CO2 fluxes were measured in hydroponically grown mycorrhizal and non-mycorrhizal Triticum aestivum L. cv. Hano roots. The NO3 uptake of the plants was used to estimate the amount of root respiration attributable to ion uptake. Plants were grown at 4 mM N and 10 μM P, where a total and viable mycorrhizal root colonisation of 48% and 18%, respectively, by Glomus mosseae (Nicol. and Gerd.) Gerd. and Trappe (BEG 107) was observed. The O2 consumption and NO3 uptake rates were similar and the CO2 release was higher in mycorrhizal than in non-mycorrhizal wheat. This resulted in a significantly higher respiratory quotient (RQ, mol CO2 mol–1 O2) in mycorrhizal (1.27±0.13) than in non-mycorrhizal (0.79±0.05) wheat. As the biomass and N and P concentrations in mycorrhizal and non-mycorrhizal wheat were the same, the higher RQ resulted from the mycorrhizal colonisation and not differences in nutrition per se. Chaetoceros muelleri (Lemn.) was cultured with nitrite (NO2) or nitrate (NO3) as the sole nitrogen source and aerated with air or with CO2-enriched air. Cells of C. muelleri excreted into the medium nitrite produced by reduction of nitrate when grown with 100 μM NaNO3 as nitrogen source. Accordingly, NO2 concentration reached 10.4 μM after 95 h at the low CO2 condition (aerated with air); while the maximum NO2 concentration was only around 2.0 μM at the high CO2 condition (aerated with 5% CO2 in air), furthermore, after 30 h it decreased to no more than 1.0 μM. NO2 was almost assimilated in 80 h when C. muelleri was cultured at the high CO2 condition with 100 μM NaNO2 as sole nitrogen source. At the high CO2 condition, after 3 h the activity of nitrite reductase was as much as 50% higher than that at the low CO2 condition. It was indicated that enriched CO2 concentration could inhibit nitrite excretion and enhance nitrite assimilation by cells. Therefore, aeration with enriched CO2 might be an effective way to control nitrite content in aquaculture systems.  相似文献
