Diagnostic PCR with two specific primer pairs (NEOSSU and 8DDC) were used to monitor the establishment and geographical distribution of Brazilian isolates of Neozygites tanajoae Delalibera, Hajek and Humber (Entomophthorales: Neozygitaceae) released in Benin for the biological control of the cassava green mite, Mononychellus tanajoa (Bondar) (Acari: Tetranychidae). A total of 141 cassava fields were visited and samples of M. tanajoa suspected to be infected by N. tanajoae were collected in 60 fields distributed between the coastal Southern Forest Mosaic (SFM) and the Northern Guinea Savanna (NGS) zones of Benin, West Africa. Analysis of DNA samples of dead mites using the species specific NEOSSU primers revealed the presence of N. tanajoae in 46 fields. The second country specific pair of primers 8DDC revealed the presence of Brazilian isolates of N. tanajoae in 36 fields, representing 78.3% of fields positive for N. tanajoae. Brazilian isolates occurred from SFM to NGS zones in Benin, however, they were concentrated in fields located within former release zones (e.g. Department of Ouémé in the South and Borgou in the North). In contrast, the indigenous African isolates of N. tanajoae were evenly distributed in the sub-humid and humid savannah zones of the country. The mean infection rate of M. tanajoa with indigenous isolates of N. tanajoae was relatively low (5.3%) compared to Brazilian isolates (28%), indicating a higher biocontrol potential of the latter. This first post-release monitoring using PCR techniques showed that the Brazilian strains of N. tanajoae is well established in Benin and spread effectively in this area.  相似文献

Virulence of entomopathogens is often measured at the individual level using a single host individual or a group of host individuals. To what extent these virulence assessments reflect the impact of an entomopathogen on their host in the field remains largely untested, however. A methodology was developed to induce epizootics of the cassava green mite fungal pathogen Neozygites tanajoae under controlled conditions to evaluate population-level virulence of two (one Beninese and one Brazilian) isolates of the entomopathogen—which had shown similar individual-level virulence but different field impacts. In unrepeated separate experiments we inoculated mite-infested potted cassava plants with either 50 or 25 live mites (high and low inoculum) previously exposed to spores of N. tanajoae and monitored the development of fungal infections for each isolate under the same conditions. Both isolates caused mite infections and an associated decline in host mite populations relative to the control (without fungus) in all experiments, but prevalence of the fungus varied with isolate and increased with inoculum density. Peak infection levels were 90% for the Beninese isolate and 36% for the Brazilian isolate at high inoculum density, and respectively 17% and 25% at low inoculum density. We also measured dispersal from inoculated plants and found that spore dispersal increased with host infection levels, independent of host densities, whereas mite dispersal varied between isolates. These results demonstrate that epizootiology of N. tanajoae can be studied under controlled conditions and suggest that virulence tests at the population level may help to better predict performance of fungal isolates than individual-level tests.  相似文献

A survey of the pathogenic fungi associated with mites on cassava in Benin, West Africa, revealed both geographical and seasonal variation in the presence of Neozygites cf. floridana (Weiser and Muma) and Hirsutella thompsonii Fisher on Mononychellus tanajoa (Bondar) and Oligonychus gossypii (Zacher). Few dead and infected mites were found during the dry season, regardless of vegetation zone. In three of 30 surveyed sites, N. floridana was found infecting 1% of the dead M. tanajoa and 2% of the dead O. gossypii, while H. thompsonii was observed infecting 20% of the dead M. tanajoa in a single site. The frequency of sites having infected mites during the wet season was 3.5 times greater than that seen during the dry season. N. floridana infected 10% of the dead M. tanajoa and 19% of the dead O. gossypii on young leaves. Mites infected with N. floridana were found either in the coastal Southern Forest Mosaic (SFM) or in the Northern Guinea Savanna vegetation zones. N. floridana was rare in the low mite densities associated with mature leaves. H. thompsonii was found on 19% and 29% of the dead M. tanajoa on young and mature leaves respectively. All M. tanajoa infected with H. thompsonii on young leaves and mature leaves (75%) were found in the SFM. A single M. tanajoa was the only infected mite found in the Southern Guinea Savanna. Relatively few O. gossypii were infected with H. thompsonii. N. floridana and H. thompsonii were found together in three sites, but never on the same host. Phytoseiids were never found infected with either pathogen. In a regression analysis, the number of dead mites was significantly estimated from the total number of mites for both species, regardless of leaf age. The numbers of dead M. tanajoa on mature leaves were also estimated from the proportion infected with H. thompsonii. The numbers of infected mites on young leaves were estimated from their association with the SFM for M. tanajoa infected with H. thompsonii, and from total mites for O. gossypii infected with N. floridana. On mature leaves, infected mite numbers were estimated from the numbers of dead M. tanajoa infected with H. thompsonii. The merit of introducing more virulent or better adapted isolates of N. floridana to control M. tanajoa in Africa is discussed.  相似文献

Neozygites tanajoae is an entomopathogenic fungus which has been used for biocontrol of the cassava green mite (Mononychellus tanajoa, CGM) in Africa. Establishment and dispersal of Brazilian isolates which have been introduced into some African countries in recent years to improve CGM control was followed with specific PCR assays. Two primer pairs, NEOSSU_F/NEOSSU_R and 8DDC_F/8DDC_R, were used to differentiate isolates collected from several locations in Brazil and from three countries in Africa, Benin, Ghana and Tanzania. The first primer pair enabled the species-specific detection of Neozygites tanajoae, while the second differentiated the Brazilian isolates from those of other geographical origin. PCR assays were designed for detection of fungal DNA in the matrix of dead infested mites since N. tanajoae is difficult to isolate and culture on selective artificial media. Our results show that all isolates (Brazilian and African) that sporulated on mummified mites were amplified with the first primer pair confirming their Neozygites tanajoae identity. The second pair amplified DNA from all the Brazilian isolates, but did not amplify any DNA samples from the African isolates. None of the two primers showed amplification neither from any of the non-sporulating mite extracts nor from the dead uninfected mites used as negative controls. We confirmed that the two primer pairs tested are suitable for the detection and differential identification of N. tanajoae isolates from Brazil and Africa and that they are useful to monitor the establishment and spread of the Brazilian isolates of N. tanajoae introduced into Benin or into other African countries for improvement of CGM biocontrol.  相似文献

Perennial ryegrass plants collected from fields and Italian ryegrass plants grown from seed were selected for resistance to infection by ryegrass mosaic virus (RMV) by repeated manual inoculation. Two of 108 perennial ryegrass plants and one of 150 Italian ryegrass plants were symptomless after seven and nine inoculations respectively. These three plants were propagated vegetatively. Plants of the two perennial ryegrass clones showed no symptoms after further manual inoculations with the initial isolate of RMV, or with an inoculum from infected plants collected from several fields, or after inoculation by viruliferous mites. Electron microscopy and back tests indicated that the plants were virus free. Some plants of the selected Italian ryegrass clone became infected after a further inoculation with mites or sap, but fewer than similarly inoculated unselected plants.  相似文献

To determine the impact of an acarine predator guild on the abundance of a shared herbivorous prey and its principal exotic predator, a series of surveys were conducted in ca. 200 cassava fields in swamp and non-swamp areas in southwestern Benin, West Africa. For each field, the surveys provided data on the density of a pest arthropod, the cassava green mite Mononychellus tanajoa (Bondar), of an introduced and successfully established natural enemy, the apex-inhabiting predatory mite Typhlodromalus aripo DeLeon, and on occurrence of other predator species that inhabit the leaves and share the same prey. These other predators included one exotic species, Typhlodromalus manihoti Moraes, that is successfully established mainly in swamp areas, and two indigenous species, Euseius fustis (Pritchard and Baker) and Typhlodromalus saltus (Denmark and Matthysse), that are commonly found on cassava in Africa. Our aim was to assess the association between the density of M. tanajoa and that of T. aripo, the most successful predator in terms of establishment and abundance, and subsequently determine how this association was affected by the presence or absence of the other predator species. No obvious density-dependent relationship was found by inspecting the scattergrams of T. aripo versus M. tanajoa densities, but high T. aripo densities did not occur when M. tanajoa densities were low and--during the dry season in February--the densities of M. tanajoa steeply increased when T. aripo numbers were low. Given the establishment of T. aripo in all fields, the presence of other species of predatory mites (T. manihoti, both in swamp and non-swamp areas; T. saltus in absence--as well as presence--of T. manihoti in swamp areas; E. fustis in absence of T. manihoti in non-swamp areas) reduced the density of M. tanajoa by a factor 2-3. Thus in all these cases, the presence of an exotic or indigenous species of predatory mite in addition to T. aripo was associated with lower M. tanajoa density. The density of T. aripo was usually positively affected by the presence of other predator species except for T. saltus in presence of T. manihoti that negatively affected the density of T. aripo in swamp areas, an effect likely mediated by either intraguild predation or competition for food. Path analysis showed that indigenous phytoseiid species were more important in suppression of M. tanajoa populations in cassava fields than previously thought. We suggest that the lack of negative effects of the predator species complex is likely due to differential niche use by the various species which reduces interference among the predators. Manipulative experiments are, however, needed to provide details on the relative importance in M. tanajoa suppression by each species within this acarine predator guild.  相似文献

High-N(2)-fixing activities of Frankia populations in root nodules on Alnus glutinosa improve growth performance of the host plant. Therefore, the establishment of active, nodule-forming populations of Frankia in soil is desirable. In this study, we inoculated Frankia strains of Alnus host infection groups I, IIIa, and IV into soil already harboring indigenous populations of infection groups (IIIa, IIIb, and IV). Then we amended parts of the inoculated soil with leaf litter of A. glutinosa and kept these parts of soil without host plants for several weeks until they were spiked with [(15)N]NO(3) and planted with seedlings of A. glutinosa. After 4 months of growth, we analyzed plants for growth performance, nodule formation, specific Frankia populations in root nodules, and N(2) fixation rates. The results revealed that introduced Frankia strains incubated in soil for several weeks in the absence of plants remained infective and competitive for nodulation with the indigenous Frankia populations of the soil. Inoculation into and incubation in soil without host plants generally supported subsequent plant growth performance and increased the percentage of nitrogen acquired by the host plants through N(2) fixation from 33% on noninoculated, nonamended soils to 78% on inoculated, amended soils. Introduced Frankia strains representing Alnus host infection groups IIIa and IV competed with indigenous Frankia populations, whereas frankiae of group I were not found in any nodules. When grown in noninoculated, nonamended soil, A. glutinosa plants harbored Frankia populations of only group IIIa in root nodules. This group was reduced to 32% +/- 23% (standard deviation) of the Frankia nodule populations when plants were grown in inoculated, nonamended soil. Under these conditions, the introduced Frankia strain of group IV was established in 51% +/- 20% of the nodules. Leaf litter amendment during the initial incubation in soil without plants promoted nodulation by frankiae of group IV in both inoculated and noninoculated treatments. Grown in inoculated, amended soils, plants had significantly lower numbers of nodules infected by group IIIa (8% +/- 6%) than by group IV (81% +/- 11%). On plants grown in noninoculated, amended soil, the original Frankia root nodule population represented by group IIIa of the noninoculated, nonamended soil was entirely exchanged by a Frankia population belonging to group IV. The quantification of N(2) fixation rates by (15)N dilution revealed that both the indigenous and the inoculated Frankia populations of group IV had a higher specific N(2)-fixing capacity than populations belonging to group IIIa under the conditions applied. These results show that through inoculation or leaf litter amendment, Frankia populations with high specific N(2)-fixing capacities can be established in soils. These populations remain infective on their host plants, successfully compete for nodule formation with other indigenous or inoculated Frankia populations, and thereby increase plant growth performance.  相似文献

The effect of arbuscular mycorrhizal (AM) fungi on the accumulation and transport of lead was studied in a pot experiment on maize plants grown in anthropogenically-polluted substrate. The plants remained uninoculated or were inoculated with different Glomus intraradices isolates, either indigenous to the polluted substrate used or reference from non-polluted soil. A considerably lower tolerance to the conditions of polluted substrate was observed for the reference isolate that showed significantly lower frequency of root colonisation as well as arbuscule and vesicule abundance. Plants inoculated with the reference isolate also had significantly lower shoot P concentrations than plants inoculated with the isolate from polluted substrate. Nevertheless, inoculation with either indigenous or reference G. intraradices isolate resulted in higher shoot and root biomass and inoculated plants showed lower Pb concentrations in their shoots than uninoculated plants, regardless of differences in root colonisation. Root biomass of maize plants was divided according to AM-induced colouration into brightly yellow segments intensively colonised by AM fungus and non-colonised or only slightly colonised whitish ones. Intensively colonised segments of the isolate from polluted substrate contained significantly higher concentrations of phosphorus and lead than non-colonised ones, which suggest significant participation of fungal structures in element accumulation. 用盆栽法研究了不同土壤含盐量条件下接种不同VAM真菌菌株对棉花、玉米、大豆和甜瓜耐盐性的影响。结果表明,随着土壤中NaCl水平提高,真菌对棉花、玉米、大豆和甜瓜的侵染率下降,其生长量亦均呈递减趋势但对VAM真菌的依赖性则呈明显递增趋势。接种VAM真菌促进了棉花、玉米和大豆的生长,如在NaCl施入量为0~3g·kg-1时,接种M1菌株使棉花干重提高4.6~80.9%;在NaCl施入量为0~2.5g·kg-1时,接种M1菌株的玉米干重比对照增加20~109.6%;NaCl施入量为1g·kg-1时,接种M1和M2两菌株使大豆干重分别增加22.1%和10.2%.不同的VAM真菌菌株对同一植物的耐盐性以及同一种VAM真菌菌株对不同植物的耐盐性的影响程度不同。可以认为,VAM真菌与植物共生对植物在盐渍环境中的生存起着重要的作用;接种VAM真菌可以提高植物在盐渍土壤上的生产能力并减轻植物因盐害造成的产量损失。  相似文献

Three biopesticide parameters were evaluated for a fast-killing isolate (3AP2) and a wild-type isolate (Sf3) of Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV). Both isolates were evaluated for virus production using in vivo methods, for speed of kill based on bioassay of applications to glasshouse-grown and field-grown plants, and for residual insecticidal activity of unformulated virus and an encapsulating formulation to provide UV protection. Two inoculation rates comparing relative in vivo production of the isolates demonstrated 3AP2 inoculated larvae were significantly smaller than Sf3 inoculated larvae at death. At the lower inoculation rate, Sf3 inoculated larvae produced approximately twofold more occlusion bodies as the 3AP2 inoculated larvae. A model system of applications to cabbage plants and a bioassay to observe mortality of neonate S. frugiperda (J.E. Smith) after feeding on samples of treated leaves was used to evaluate speed of kill and residual insecticidal activity. The LT(50) for the 3AP2 isolate was at least 30 h less than the LT(50) for the Sf3 isolate when applied to either glasshouse-grown or field-grown plants. The spray-dried lignin encapsulating formulation provided similar benefits to both virus isolates when exposed to simulated sunlight in the laboratory and to natural sunlight in the field. For treatment applications to field grown cabbage in June, the half-life for efficacy of unformulated virus was <7.5 h compared with a half-life of >26.7 h for encapsulated virus. These results demonstrate that improved technologies can be combined to address characteristics which otherwise can limit the commercial potential of microbial-based biological insecticides.  相似文献

The infectivity per os and by inoculation of ascoviruses isolated from Heliothis zea, Spodoptera frugiperda, and Trichopulsia ni and the effects of infection by these viruses on host growth and development were studied in the species from which these viruses were originally isolated, or in the case of the H. zea isolate, in H. virescens. Mortality caused by all three viral isolates averaged less than 15% in third instars fed doses of 10 to 10(5) viral vesicles per larva incorporated into diet, whereas inoculation with as few as 10 viral vesicles consistently yielded mortality rates greater than 90%. In tests where groups of 10 larvae were inoculated sequentially with a minuten pin that had been inserted into the hemocoel of an infected larva and then dried for up to 24 hr, mortality rates were consistently greater than 90%. The latter results suggest that ascoviruses in the field may be vectored by insect parasites. The studies of the effects of infection on larval growth and development demonstrated a marked loss of appetite and a decrease in weight gain within 24-48 hr of infection in all three isolates. Third instars inoculated at a weight of ca. 15 mg gained little weight after infection and had difficulty molting, but survived in an arrested state of development for 2-5 weeks. Controls pupated within 5-7 days after attaining weights, depending on the host species, in the range of 300-500 mg. Arrested development and lack of weight gain appear to be due to decreased feeding, whereas the long survival of infected larvae is likely due to the limited destruction of host tissues.  相似文献

The cassava green mite, Mononychellus tanajoa (Bondar), is an exotic pest in Africa and is the target of a classical biological control programme. Field data from the Neotropics, where it is indigenous, are presented for the first time, charting the variation in abundance of M. tanajoa over several seasons. This was highly variable, with a characteristic trough mid-year and a peak at the turn of the year. This pattern corresponded positively with rainfall levels, appearing to fit a phenology also characteristic of African studies, where rainfall at the start of the wet season promotes a leaf flush and so growth in M. tanajoa populations. Analyses implied some impact of leaf-inhabiting predatory mites (predominantly Neoseiulus idaeus Denmark & Muma) and a considerable impact of the fungal pathogen Neozygites floridana Fisher on M. tanajoa populations. This pathogen was not observed in the host population for several (generally dry) periods implying survival outside the host, perhaps as resting spores. This is a particularly desirable characteristic of a biological control agent. It is therefore proposed that N. floridana might be of particular use in drier cassava-growing areas where rainfall at the outset of the wet season is not sufficiently intense to cause heavy M. tanajoa mortality but may be sufficient to stimulate epizootics of the fungal pathogen, protecting the flush of new cassava growth.  相似文献

柑橘对丛枝菌根(AM)真菌具有较高的依赖性,从柑橘园土壤中分离筛选的高效促生AM真菌菌株具有重要的应用价值。本研究从广东增城柑橘园(酸橘砧‘红江橙’)土壤中分离的4个AM真菌土著菌株对‘红江橙’幼苗的促生效应。结果表明,4个土著菌株分别是Scutellospora属和Glomus属菌株,根系侵染率为12.7%~29.3%;与不接种对照相比,4个土著菌株不同程度地促进‘红江橙’幼苗的株高、生物量和N、P、K养分含量,菌根依赖性达9.4%~37.1%;主成分分析表明,土著菌株ZCSP-D的促生效应达到常用优良菌株Rhizophagus irregularis的水平。  相似文献

In trials in 1973-5 at the Glasshouse Investigational Unit for Scotland, the yield of fruit from tomato cv. Eurocross BB inoculated at the seedling stage with the Mil-16 attenuated strain of tobacco mosaic virus was 5–8-9-4% greater than that from uninoculated plants which became naturally infected with a severe indigenous strain of the virus within 7–8 wk of planting. The increase in fruit yield, particularly of better grades, resulted in higher gross financial returns (up to 25p/plant) from inoculated plants. The yields from the Mil-16 protected plants were up to 14% greater than those from plants artificially inoculated at the seedling stage with the indigenous severe virus. Inoculation with Mil-16 had little adverse effect on early growth or the rate of fruit development on the first five trusses, but in 1973 the final yield of inoculated plants was depressed c. 5% compared with that from plants substantially free from infection for 14 wk after planting. In 1 year's test no benefit from inoculation with Mil-16 was recorded in cv. Cudlow Cross.  相似文献

Eight native isolates of the entomopathogenic fungus Metarhizium anisopliae (Metschnikoff) Sorokin were obtained by monitoring soils cultivated in a conventional manner. These isolates were compared in three areas: (a) conidial germination, (b) radial growth and sporulation and (c) ability of conidia to infect Tenebrio molitor larvae. All bioassays were carried out at constant temperatures of 10, 15, and 20 °C. Conidia of individual isolates demonstrated differences in germination after a 24-h long incubation at all evaluated temperatures. At 20 °C, the germination ranged from 67 to 100 % and at 15 °C from 5.33 to 46.67 %. At 10 °C, no germination was observed after 24 h; nevertheless, it was 8.67–44.67 % after 48 h. In terms of radial growth, the culture diameters and the associated production of spores of all isolates increased with increasing temperature. At 10 °C, sporulation was observed in three isolates while all remaining cultures appeared sterile. Three weeks post-inoculation, conidia of all assessed isolates caused 100 % cumulative mortality of treated larvae of T. molitor at 15 and 20 °C with the exception of isolate 110108 that induced 81.33 % mortality at 15 °C. At 10 °C, larval cumulative mortality ranged from 6.67 to 85.33 % depending on the isolate. Isolates 110108 and 110111 showed significantly slower outset and a much lower rate of infection at all temperatures compared to other tested isolates of M. anisopliae. The bioassays were carried out with the purpose to sort and select indigenous isolates of M. anisopliae useful as biocontrol agents in their original habitat.  相似文献

It is known that vegetation plays an important role in the retention of heavy metals in salt marshes by taking up and accumulating the metals. In this study, we investigated whether arbuscular mycorrhizal fungi (AMF) increase Cd and Cu uptake and accumulation in the root system of the salt marsh species Aster tripolium L., and whether indigenous AMF isolated from polluted salt marshes have higher capacity to resist and alleviate metal stress in A. tripolium than isolates of the same species originated from non-polluted sites. Plants inoculated with Glomus geosporum, either isolated from a polluted salt marsh site (PL isolate) or from a non-polluted site (NP isolate), and non-mycorrhizal (NM) plants were compared in a pot experiment at four different Cd and Cu concentrations. Cd had no effect in root colonization, whereas high concentrations of Cu decreased colonization level in plants inoculated with the NP isolate. AM colonization did not increase plant dry weight or P concentration but influenced root Cd and Cu concentrations. Inoculation with PL and NP isolates enhanced root Cd and Cu concentrations, especially at highest metal addition levels, as compared to NM plants, without increasing shoot Cd and Cu concentrations. There was no evidence of intraspecific variation in the effects between AMF isolated from polluted and non-polluted sites, since there were no differences between plants inoculated with PL or NP isolate in any of the tested plant variables. The results of this study showed that AMF enhance metal accumulation in the root system of A. tripolium, suggesting a contribution of AMF to the sink of metals within vegetation in the salt marshes.  相似文献

In glasshouse tests, sap from plants infected with 15 different isolates of tomato spotted wilt tospovirus (TSWV) from three Australian states was inoculated to nine genotypes of tomato carrying TSWV resistance gene Sw-5 or one of its alleles. A further two resistant tomato genotypes were inoculated with four isolates each. The normal response in resistant genotypes was development of necrotic local lesions in inoculated leaves without systemic invasion, but 22/752 plants also developed systemic reactions in addition to local hypersensitive ones. Using cultures from two of these systemically infected plants and following four cycles of subculture in TSWV resistant tomato plants, two isolates were obtained that gave susceptible type systemic reactions but no necrotic spots in inoculated leaves of resistant tomatoes. When these two isolates, DaWA-1d and ToTAS-1d, were maintained by repeated subculture for 10 successive cycles in Nicotiana glutinosa or a susceptible tomato genotype, they still induced susceptible type systemic reactions when inoculated to resistant tomato plants. They were therefore stable resistance breaking isolates as regards overcoming gene Sw-5. When resistance-breaking isolate DaWA-1ld multiplied together with original isolate DaWA-l in susceptible tomato, it was fully competitive with the original isolate. However, when DaWA-ld and ToTAS-ld were inoculated to TSWV resistant Lycopersicon peruvianum lines PI 128660R and PI 128660S and to TSWV resistant Capsicum chinense lines PI 152225, PI 159236 and AVRDC CO0943, they failed to overcome the resistance, producing only necrotic local lesions without systemic infection. Thus, although the ease of selection, stability and competitive ability of resistance breaking isolates of TSWV is cause for concern, L. peruvianum and C. chinense lines are available which are effective against them. The effectiveness of the resistance to TSWV in nine tomato genotypes was examined in a field experiment. Spread was substantial in the susceptible control genotype infecting 42% of plants. Resistance was ineffective in cv. Bronze Rebel, 26% of plants developing infection. In contrast, it held up well in the other eight resistant genotypes with only 1–3 or no plants of each becoming infected. Accumulated numbers of Thrips tabaci, Frankliniella occidentalis and F. schultzei were closely correlated with TSWV spread.  相似文献

The behavior of two isolates of Pratylenchus penetrans on six potato clones was assessed to test the hypothesis that these nematode isolates from New York were different. Four potato cultivars (Superior, Russet Burbank, Butte, and Hudson) and two breeding lines (NY85 and L118-2) were inoculated with nematode isolates designated Cornell (CR) and Long Island (LI). Population increase and egression of nematodes from roots were used to distinguish resistance and susceptibility of the potato clones. Based on numbers of eggs, juveniles, and adults in their roots 30 days after inoculation, potato clones Butte, Hudson, and L118-2 were designated resistant to the CR isolate and susceptible to the LI isolate. More eggs were found in the roots of all plants inoculated with the LI isolate than with the CR isolate. The clones NY85 and L118-2 were inoculated with the CR and LI isolates in a 2 x 2 factorial experiment to assess differences in nematode egression. Egression was measured, beginning 3 days after inoculation, for 12 days. The rates of egression were similar for the four treatments and fit linear regression models, but differences were detected in numbers of egressed nematodes. More nematodes of the CR isolate than the LI isolate egressed from L118-2. Differences in egression of females was particularly significant and can be used as an alternative or supplement to reproduction tests to assess resistance in potato to P. penetrans and to distinguish variation in virulence.  相似文献

Groundwater, contaminated with trichloroethylene (TCE) and tetrachloroethylene (PCE), was collected from 13 monitoring wells at Area M on the U.S. Department of Energy Savannah River Site near Aiken, S.C. Filtered groundwater samples were enriched with methane, leading to the isolation of 25 methanotrophic isolates. The phospholipid fatty acid profiles of all the isolates were dominated by 18:1 omega 8c (60 to 80%), a signature lipid for group II methanotrophs. Subsequent phenotypic testing showed that most of the strains were members of the genus Methylosinus and one isolate was a member of the genus Methylocystis. Most of the methanotroph isolates exhibited soluble methane monooxygenase (sMMO) activity. This was presumptively indicated by the naphthalene oxidation assay and confirmed by hybridization with a gene probe encoding the mmoB gene and by cell extract assays. TCE was degraded at various rates by most of the sMMO-producing isolates, whereas PCE was not degraded. Savannah River Area M and other groundwaters, pristine and polluted, were found to support sMMO activity when supplemented with nutrients and then inoculated with Methylosinus trichosporium OB3b. The maximal sMMO-specific activity obtained in the various groundwaters ranged from 41 to 67% compared with maximal rates obtained in copper-free nitrate mineral salts media. This study partially supports the hypothesis that stimulation of indigenous methanotrophic communities can be efficacious for removal of chlorinated aliphatic hydrocarbons from subsurface sites and that the removal can be mediated by sMMO.  相似文献

The human populations of the Brazilian Amazon were formed by interethnic crosses between Europeans, Africans, and Amerindians. The relative contribution of men and women of different ethnic groups was not homogeneous, since the social policies of the first three centuries of Brazilian colonization encouraged mating between European men and indigenous women and, later on, African women. In order to test this model based on historical data, we compared the relative contribution of the Y-DNA and mtDNA of Amerindian and non-Amerindian populations to the formation of the urban population of the town of Belém, in the Amazon region, on the basis of a C→T mutation at locus DYS199 present in 90% of the Amerindian Y-DNA and of five markers that define 99% of the mitochondrial sequences of Amerindians. The contribution of indigenous men to the formation of this population was less than 5%, whereas the contribution of indigenous women was estimated at more than 50% of the mitochondrial sequences of the same population. Thus, the present results demonstrate that the contribution of indigenous women to the formation of the Belém population was 10 times higher than the contribution of indigenous men, a genetic consequence of social behavior and attitudes of the past; our results also help clarify the process of integration of indigenous communities into the urban societies in Brazil and possibly in other countries.
