Cell wall polysaccharides from black currants and bilberries were characterised in three approaches. First, compositions of skin, pulp, and seeds show the distribution of polysaccharides over these tissues. A sequential extraction of cell wall material with different aqueous extractants informs about the extractability of the different polysaccharides, viz. pectins, hemicellulose, and cellulose. Finally, by isolation of cell wall polysaccharides from juice and press cakes obtained by the conventional juice manufacturing. The polysaccharide distribution was followed during juice processing. The main difference between bilberries and black currants is the dominant sugar residue in seeds: mannose for black currants and xylose for bilberries. Most of the hemicellulolytic sugars and cellulose can be found back in the press cake. The sugar composition of the press cake is similar to the composition of the residue after sequential extraction. Black currants contain more pectic sugars than bilberries. Consequently, a commercial enzyme used during processing releases more pectic material into the juice.  相似文献

It has been suggested in the literature that the glycolytic enzymes are organized into a multi-enzymic complex. We have evaluated this hypothesis for the phosphotriose-glycerate phosphate group of glycolytic enzymes of muscle using sucrose density gradient centrifugation, gel filtration, and ultrafiltration. Attempts were made to avoid dilution and changes in pH. The ratio of activities of the phosphotriose-glycerate phosphate group of enzymes was similar to that found in several other tissues that has led to their designation as a constant proportion group of enzymes. However, no evidence was obtained that they exist as a multi-enzymic complex in chicken breast muscle. As the pH of the press juice is raised to 7.0 and the temperature to 25°C, association occurs between some components in the muscle press juice as evidenced by a blocking of the pores of an ultrafiltration membrane. This association, however, does not involve the enzymes of the phosphotriose-glycerate phosphate group.  相似文献

Objective: The present study compared surface electromyographic (sEMG) activity obtained from the submental muscle group for a tongue press and a head lift exercise as potential therapeutic exercises for dysphagic elderly. Materials and methods: Fifty‐three healthy volunteers with a mean age of 35.3 participated in this study. Subjects were required to perform an isometric task, pressing their tongue against the hard palate, and an isotonic task requiring sustained lingual force against the hard palate. Pressure sensors were used to measure the amount of lingual pressure against the hard palate. Submental sEMG data from these tasks were compared with those obtained from the isometric and isotonic aspects of a head lift exercise. Results: No sEMG differences were identified between the isometric tongue press task and head lift exercise. Isotonic tongue press exercises resulted in significantly higher maximum and mean sEMG values compared with the isotonic head lift exercise (p < 0.05). The submental sEMG activity from the tongue press exercise was equal (isometric) to, or greater (isotonic) than comparable muscle activation obtained during the head lift exercise. Conclusions: The tongue press exercise may be less strenuous than the head lift exercise while achieving the same therapeutic effect.  相似文献

Modifications to a Pollahne leaf sap press are described which allow it to be used for bioassay purposes without risk of cross contamination. Constructional details are given for manual and automatic versions which utilise hot and cold water to sterilise and cool the working parts of the press. Two compressed air guns which are used together to inoculate indicator plants, are also described. The first incorporates carborundum powder into a jet of air and causes controlled wounding of plant epidermal cells to permit virus entry. The other applies sap inoculum as a finely divided spray to the wounded plants. The variables affecting the use of these guns are discussed. Improvements in sensitivity of 64 fold, compared to manual methods were obtained and potential to develop this to 500fold demonstrated.  相似文献

This study was undertaken to develop techniques for measuring absolute rates of sterol synthesis in extrahepatic tissues in vitro and to estimate the magnitude of the errors inherent in the use of various 14C-labeled substrates for such measurements. Initial studies showed that significant errors were introduced when rates of synthesis were estimated using [3H]water since about 20 nmol of water were bound to each mg of tissue cholesterol isolated as the digitonide. This source of error could be eliminated by subtracting apparent incorporation rates obtained at 0 degrees C from those obtained at 37 degrees C or by regenerating and drying the free sterol. In a second set of experiments, the H/C incorporation ratio in cholesterol was determined in the liver by measuring the absolute rates of hydrogen and acetyl CoA flux into sterols. The ratio of 0.69 +/- 0.03 was found to be independent of the rate of hepatic cholesterol synthesis, the rate of hepatic acetyl CoA generation, or the source of the acetyl CoA. In a third set of studies, rates of incorporation of [3H]water or 14C-labeled acetate, octanoate, and glucose into digitonin-precipitable sterols were simultaneously measured in nine different extrahepatic tissues. Assuming that the H/C ratio measured in the liver also applied to these tissues, the [3H]water incorporation rates were multipled by the reciprocal of the H/C ratio to give the absolute rates of sterol synthesis in each tissue. When these were compared to the incorporation rates determined with the 14C-labeled substrates the magnitude of the errors in the rates of sterol synthesis obtained with these substrates in each tissue could be assessed. Only [14C]octanoate gave synthesis rates approaching 100% of those obtained with [3H]water and this occurred only in the intestine and kidney; in the other extrahepatic tissues this substrate gave rates of 6--66+ of the absolute rates. Rates of [14C]acetate incorporation in sterols varied from 4 to 62% of the [3H]water incorporation rates while those obtained with [14C]glucose were only 2--88% of the true rates. These studies document the large and highly variable errors inherent in estimating rates of sterol synthesis in extrahepatic tissues using 14C-labeled substrates under in vitro conditions.  相似文献

Cartilaginous and connective tissues were exposed to a moderate-intensity IR laser treatment with IR radiometric temperature control. The isotherms of water sorption by dry preparations of intact and laser-treated tissues were obtained and calculated in terms of well known models. It was shown that the sorption capacity of a monolayer of tissues decreases with increasing temperature during laser treatment. Changes in the supramolecular structure of the extracellular matrix of tissues (loss of regularity) upon IR laser exposure are discussed.  相似文献

Anaerobic co-digestion of press mud with water or sewage at ratios of 1:7.5, 1:10 and 1:12.5 were performed in continuously fed UASB reactors for hydrogen production. At a constant hydraulic retention time of 30 h, the specific hydrogen production rate was 187 mL/g volatile solids (VS) reduced during maximum biohydrogen production of 7960 mL/day at a 1:10 ratio of press mud to sewage. Chemical oxygen demand (COD) and VS reductions of 61% and 59% were noted on peak biohydrogen yield. A pH range of 5-6 was suitable at ambient temperature for entire process; a lower pH was inhibitory. Co-digestion of acidic press mud with sewage controlled pH for fermentation. Hence press mud can be exploited for biohydrogen production.  相似文献

Twelve experienced male weight lifters performed a rebound bench press and a purely concentric bench press lift. Data were obtained pertaining to 1) the benefits to concentric motion derived from a prior stretch and 2) the movement frequency adopted during performance of the stretch-shorten cycle (SSC) portion of the rebound bench press lift. The subjects also performed a series of quasi-static muscular actions in a position specific to the bench press movement. A brief perturbation was applied to the bar while these isometric efforts were maintained, and the resulting damped oscillations provided data pertaining to each subject's series elastic component (SEC) stiffness and natural frequency of oscillation. A significant correlation (r = -0.718, P less than 0.01) between maximal SEC stiffness and augmentation to concentric motion derived from prior stretch was observed. Subjects were also observed to perform the SSC portion of the rebound bench press movement to coincide with the natural frequency of oscillation of their SEC. These results are interpreted as demonstrating that the optimal stiffness in a rebound bench press lift was a resonant-compliant SEC.  相似文献

Water proton longitudinal NMR relaxation times were measured in various tissues of healthy and tumor-bearing mice. Measurements were performed as a function of the Larmor frequency nu in the range 6-90 MHz, and at two temperatures (theta + and theta -) bracketing the 'freezing transition', at which the major part of the water signal disappears. At both temperatures, 1/T1 behaves according to: 1/T1 = A/square root nu + B A and B are obtained at theta + and theta -, and yield the proportion of bound water, which is convincingly identified with non-freezable water. The proportions found lie around 6% for tumors and 12% for other tissues. Discrimination between tissues via T1 is demonstrated to be essentially due to the bound water proportion. Bound water on the one hand and free water on the other hand behave similarly in all tissues including tumors. The activation energy for free water is found to be identical to that of pure water, although relaxation times are markedly different. It is noticed that determining the bound water proportion by signal intensity measurements at theta + and theta - is less reliable than by the T1 method.  相似文献

Biomass in the form of agricultural residues is becoming popular among new renewable energy sources, especially given its wide potential and abundant usage. Pyrolysis is the most important process among the thermal conversion processes of biomass. In this study, the various characteristics of bio-oils acquired under different pyrolysis conditions from safflower seed press cake (SPC) were identified. The elemental analyses and calorific values of the bio-oils were determined, and then the chemical compositions of the bio-oils were investigated using chromatographic and spectroscopic techniques such as column chromatography, 1H NMR, FTIR and GC. The fuel properties of the bio-oil such as kinematic viscosity, flash point, density, water content and ASTM distillation were also determined. Chemical compositions of bio-oils showed that some quantities of hydrocarbons were present, while oxygenated and polar fractions dominated. The bio-oils obtained from safflower seed press cake were presented as an environmentally friendly feedstock candidate for biofuels and chemicals.  相似文献

The diurnal variation in leaf water potential of wheat underfield conditions is determined by means of the Scholander pressurechamber and the J14 press. Experimental work was carried outon several days during both the vegetative and reproductivegrowth stages. An exponential relationship (v = 4.09 e0.05x),with coefficient of determination r2 = 0.81, is found to existbetween the J14 press and Scholander pressure chamber measurements.Both the Scholander pressure chamber and J14 press appear capableof following hourly variations in leaf water potential provided8 and 16 leaves per sample are used for the two instrumentsrespectively.  相似文献

Young barley seedlings were stressed using nutrient solutions containing NaCl or polyethylene glycol and measurements were made of leaf growth, water potential, osmotic potential and turgor values of both growing (basal) and nongrowing (blade) tissues. Rapid growth responses similar to those noted for corn (Plant Physiology 48: 631-636) were obtained using either NaCl or polyethylene glycol treatments by which exposure of seedlings to solutions with water potential values of −3 to −11 bars effected an immediate cessation of leaf elongation with growth resumption after several minutes or hours. Latent periods were increased and growth resumption rates were decreased as water potential values of nutrient solutions were lowered. In unstressed transpiring seedlings, water potential and osmotic potential values of leaf basal tissues were usually −6 to −8 bars, and −12 to −14 bars, respectively. These tissues began to adjust osmotically when exposed to any of the osmotic solutions, and hourly reductions of 1 to 2 bars in both water potential and osmotic potential values usually occurred for the first 2 to 4 hours, but reduction rates thereafter were lower. When seedlings were exposed to solutions with water potential values lower than those of the leaf basal tissues, growth resumed about the time water potential values of those tissues fell to that of the nutrient solution. After 1 to 3 days of seedling exposure to solutions with different water potential values, cumulative leaf elongation was reduced as the water potential values of the root medium were lowered. Reductions in water potential and osmotic potential values of tissues in leaf basal regions paralleled growth reductions, but turgor value was largely unaffected by stress. In contrast, water potential, osmotic potential, and turgor values of leaf blades were usually changed slightly regardless of the degree and duration of stress, and blade water potential values were always higher than water potential values of the basally located cells. It is hypothesized that blades have high water potential values and are generally unresponsive to stress because water in most of the mesophyll cells in this area does not exchange readily with water present in the transpiration stream.  相似文献

The rates of emergence of wheat and lupin were measured in 13 field experiments on water repellent sands. Conventional sowing was compared with furrow sowing either with or without the use of a press wheel and several rates of banded wetting agent. Measurements included, severity of water repellence, plant emergence, rainfall, soil temperature at sowing and, at one site, the area of wet soil after sowing. All ameliorative techniques improved emergence, with responses being greatest when seeds were sown into dry soil. Compared with conventional sowing, furrow sowing increased wheat and lupin emergence by an overall average of 16 and 41%, respectively. The benefits were greater at the drier sites. Increases in emergence due to the use of a press wheel were sometimes small, although they always occurred (1–19%). It was visually observed that press wheel use gave more uniform seeding depth, reduced clods and ensured more accurate placement of banded wetting agent. Banded wetting agent consistently improved wheat and lupin emergence, particularly where early rains were light and press wheels were used. The wetting agent increased the cross-sectional area of wet topsoil (0–10 cm) which was positively related with increased wheat emergence (R2 = 0.91). At 0.5 L ha−1 of banded wetting agent, the soil along the furrow was four times wetter than without wetting agent. Wetting agent at 0.5 and 1 L ha−1 (with press wheels) increased wheat emergence by 6 and 11% and lupin emergence by 13 and 11%, respectively. The high rates of banded wetting agent gave highest plant densities. Grain yield was only measured at three sites. Furrow sowing did not increase grain yield, however, press wheels use with furrow sowing increased grain yield by 30%. Banded wetting agent increased grain yield and they were positively correlated. The highest rate increased grain yields by a further 9% above press wheels and furrow sowing. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Two model systems were used to study the freezing of water in small diameter pores. Water in pores having a diameter of less than 100 nanometers froze at lower temperatures than bulk water. Data obtained with a range of pore sizes were consistent with predicted values based on equations developed by Mazur (1965 Ann NY Acad Sci 125: 658-676), and Homshaw (1980 J Soil Sci 31: 399-414). The addition of solutes lowered the freezing point of water in small pores. We propose that the freezing behavior of water in small pores may account for some of the freezing patterns observed in plant tissues. In tissues where cells are tightly packed, share common walls, and lack intercellular spaces, the presence of water in cell wall microcapillaries would alter the freezing temperature of tissue water, impede the spread of ice, and facilitate supercooling.  相似文献

The velocity, attenuation and frequency content of the mechanical pulse wave propagation in gels of various water contents, in normal tissues from various sites and in oedematous tissues from different patients were investigated. The properties of the propagated pulse wave depend on the water content of the gel and the viscoelastic properties of the tissues. From the dependence of the pulse wave propagation velocity on elasticity, viscosity and density, information may be obtained concerning the effects of oedema on the mechanical properties of tissue.  相似文献

The ESR method was used to study free-radical disturbances in tissues of mouse liver and spleen exposed in vitro to gamma-radiation and fission neutrons. It was shown that both types of radiation induced damages to basic chemical components of the cell, namely, DNA, lipids, proteins, and water. The radiochemical yields of each radical registered were obtained and RBE of neutrons were evaluated with a reference to the formation in tissues of radicals of each type. A stable cellulose paper system was studied to relate water distribution data, as obtained previously from plant tissues, to the analysis of freezing energy. Water distribution data for the cellulose system were obtained by several techniques and were coordinated with calorimetric data. The effect of the cellulose system on the latent heat of freezing was evaluated to estimate activation energies as functions of the amount of associated liquid water. Similar activation energies of water phase transitions in critical plant tissue systems may be heritable characteristics that affect freezing stress. Adhesion energy, that develops between ice and hydrophilic polymer systems as they compete for liquid water in a complex interface, was suggested as one possible source of freezing stress. This does not occur in frost desiccation.  相似文献

The details of a convenient laboratory press are presented anda procedure by which plant sap is readily obtained is described.The pressing procedure has been shown to release 53–74per cent. of the total plant water as expressed sap. Preliminary observations on the uptake and translocation ofantibiotics by higher plants obtained by examining expressedsap are described. It is shown that the assay of expressed sapprovides a measure of the griseofulvin and chloramphenicol concentrationin leaves of plants grown in solutions of these antibioticsthat does not differ significantly from the value obtained bythe assay of water extracts. Extraction of leaves from plants grown in griseofulvin solutionswith organic solvents demonstrates a much greater antibioticcontent than is indicated by the assay of expressed sap. Toexplain this difference a ‘free’ antibiotic fraction,obtainable in expressed sap or water extracts, and a ‘bound’fraction recovered from the leaf only by organic solvent extractionare postulated.  相似文献

Raf. (Fabaceae). Sodium chloride (NaCl) was added to the soil and salinity was maintained at 0.3, 1.9, 3.9, 6.0 and 7.9 dS m?1. A negative relationship between seedling emergence and salt concentration was obtained. Salinity caused reduction in water content and water potential of tissues (leaves, stems, tap roots and lateral roots) that resulted in internal water deficit to plants. Consequently, shoot and root elongation, leaf expansion and dry matter accumulation in leaves, stems, tap roots and lateral root tissues of seedlings significantly decreased in response to increasing concentration of salt. Proline content in tissues was very low. There were no effective mechanisms to control net uptake of Na on root plasma membrane and subsequently its transport to shoot tissues. Potassium content significantly decreased in tissues in response to salinisation of soil. This tree species is a moderate salt-tolerant glycophytic plant. Nitrogen and calcium content in tissues significantly decreased as soil salinity increased. Phosphors content in tissues exhibited a declining trend with increase in soil salinity. Changes in tissues and whole-plant accumulation pattern of other elements tested, as well as possible mechanisms for avoidance of Na toxicity in this tree species in response to salinisation, are discussed.  相似文献
