Oxygen radical absorbance capacity (ORAC) values showed that methanolic extracts of Albizia julibrissin foliage displayed antioxidant activity. High performance liquid chromatography (HPLC) and mass spectrometry (MS) techniques were utilized in the identification of the compounds. The analysis confirmed the presence of three compounds in A. julibrissin foliage methanolic extract: an unknown quercetin derivative with mass of 610 Da, hyperoside (quercetin-3-O-galactoside), and quercitrin (quercetin-3-O-rhamnoside). Fast performance liquid chromatography (FPLC) was employed to fractionate the crude A. julibrissin foliage methanolic extract into its individual flavonoid components. The flavonoids were quantified in terms of mass and their respective contribution to the overall ORAC value. Quercetin glycosides accounted for 2.0% of total foliage.  相似文献

Reverse-phase HPLC coupled with photodiode array detection was used for the simultaneous separation and determination of naturally occurring adrenergic amines (octopamine, synephrine and tyramine) in fruits and dry extracts of Citrus aurantium L. var. amara and in herbal medicines derived therefrom. Synephrine was the main component in fruits (0.10-0.35%) and in dry extracts (3.00-3.08%) and was present in the range 0.25-0.99% in herbal medicines. Flavanones were analysed in the same samples using a reverse-phase HPLC technique which allowed the identification and quantification of neoeriocitrin, narirutin, naringin, hesperidin, neohesperidin, naringenin and hesperetin. C. aurantium fruits and derivatives contained mainly glycosylated flavanones: in particular, naringin and neohesperidin were found to be the major flavonoids and their concentrations ranged from 1.80 to 26.30 and from 3.90 to 14.71 mg/g, respectively. The oxygen radical absorbance capacity (ORAC) assay has been used to quantify the antioxidative properties of phytonutrients in fruit and vegetable extracts. Using aqueous methanol extracts of tea and spinach as a model systems, separation of the components in the extracts by HPLC followed by semiautomatic ORAC analysis of the column fractions permitted the determination of peroxyl-radical-scavenging profiles, demonstrating the relative abilities of the individual extract components to scavenge peroxyl radicals. ORAC values for up to 80 HPLC fractions were measured, confirming the major contribution of epigallocatechin gallate in the peroxyl radical scavenging of green tea extracts. Although the flavonoids in spinach extracts provided resistance to peroxyl radicals, components that did not bind to the HPLC column and simple phenolic compounds may also be important contributors to the total ORAC activity of spinach leaf extracts. Application of these procedures to plants believed to provide certain human health benefits by reducing free radicals may allow the identification and characterization of the specific components responsible for the free-radical-scavenging activities.  相似文献

本研究以总黄酮吸附量和解析率为检测指标,结合动态洗脱考察结果,考察11种树脂对赶黄草总黄酮的富集精制能力。优化树脂类型,选定了对总黄酮进行富集纯化较好的树脂HPD450;采用单因素及正交实验,确定最佳工艺条件:上样量10 mL,以6 mL/min的流速8 BV水、6 BV 80%甲醇溶液洗脱,pH值为6~7。以此方法得到90%以上的赶黄草总黄酮,实验结果良好,总黄酮精制工艺成效显著。用HPLC法精确测定赶黄草中槲皮苷、PGHG和ThA三个指标性黄酮成分,最佳色谱条件为:C18色谱柱,检测波长280 nm,进样量5μL,流动相:乙腈-0.05%磷酸溶液(0~25 min:12%~45%乙腈;25~40 min:45%~70%乙腈)梯度洗脱;精确测定赶黄草全草中3种黄酮成分的含量。采用分光光度法检测,指导优化总黄酮精制工艺,并以HPLC法精确测定三种黄酮代表成分。比较两种方法测定结果相一致,互为补充;故在工业生产时,可以分光光度法指导生产,以HPLC法精确定量测定。  相似文献

亚硝酸盐在食材、食品加工和食品储存过程中普遍存在,它是亚硝基化合物合成的前体物质,在一定条件下亚硝酸盐极易转化为亚硝胺,二者均可致癌[1-2],因此,清除亚硝酸盐和阻断亚硝胺合成对于预防和控制癌症的发生具有重要意义。关于清除亚硝酸盐和阻断亚硝胺合成的研究已有较多报道,一些植物含有的天然活性成分,如植物中所含的多酚和黄酮类物质等,具有抗氧化、抗菌、清除亚硝酸盐和阻断亚硝胺合成的作用。顶羽菊也Acroptilon repens ( Linn.) DC.页[3]、黄花菜(Hemerocallis citrina Baroni)[4]和簕菜也Acanthopanax trifoliatus ( Linn.) Merr.页[5]等提取物具有清除亚硝酸盐和阻断亚硝胺合成的作用;黄俊生[6]从南姜也Alpinia galanga ( Linn.) Willd.页表皮中提取的色素也具有较强的清除亚硝酸钠和阻断亚硝胺合成活性。  相似文献

Hypericum perforatum extracts are used mainly as oral antidepressants. Depending on source the extracts contain various amounts of phenylpropanes, flavonol derivates, biflavones, proathocyanidines, xanthones, phloroglucinoles, some amino acids, naphtodianthrones (hypericines) and essential oil constituents. The therapeutic use of Hypericum perforatum extracts however is limited by their phototoxic potential. It was the aim of the present study to investigate the phototoxic potential of 3 Hypericum perforatum extracts from different sources as well as some of its main constituents. In order to systematically study the phototoxic potential we established a modified neutral red assay utilizing an immortalized human keratinocyte cell line (HaCaT cells) as substrate and UVA irradiation. This modified neutral red assay was found to be a simple and reliable method for detecting phototoxic effects of reference agents and plant extracts. The validity of this method was demonstrated with known phototoxic compounds like chloropromazine and psoralenes like 5-MOP. Hypericum perforatum extracts demonstrated cytotoxicity and photocytotoxicity in a dose and UVA-dose dependent manner. Hypericine itself also evoked severe phototoxic effects and was thus identified as the main phototoxic constituent. Among the tested flavonoids quercitrin was found to be cytotoxic, while rutin unexpectedly demonstrated phototoxicity whereas quercitrin was effective to control the phototoxic activity of Hypericum perforatum extracts.  相似文献

Solid wastes from white vinification, including grape peels, seeds and stems, were used as raw material for the recovery of antioxidant polyphenols. Extractions were performed using non-toxic media composed of water/ethanol mixtures and hydrochloric, acetic or tartaric acid. Recovery efficiency was assessed by monitoring the antioxidant potency of extracts and several indices related to their polyphenolic composition, including total polyphenol, total flavonoid, total flavanol and condensed tannin (proanthocyanidin) content. Among the by-products tested, seeds were shown to contain exceptional amounts of total polyphenols (13.76 g per 100g dry weight), followed by stems (7.47 g per 100g dry weight) and peels (0.97 g per 100g dry weight). Extracts with the highest antioxidant activity from all by-products were obtained with 57% ethanol. Acidification of this medium with 0.1% HCl improved polyphenol recovery and antiradical activity for stem extracts, but it was unfavourable for seed extraction.  相似文献

The oligosaccharide composition in seeds of some bean cultivars (two of Vicia faba L. and three of V. faba minor) have been examined. The sugars were isolated by solvent extraction and the concentrated extracts were analysed by PC and HPLC analyses. Characterization was also performed by thin-layer chromatography, enzymatic hydrolysis assays, GC determination of alditol acetate derivatives obtained after complete acid hydrolysis and FAB-MS of individual sucrose - galactosides. Although sucrose, raffinose, stachyose and verbascose were always identified, great differences in the total sugar content were found among the five cultivars. Sucrose and verbascose were the major components. Their contents were highest in the Muchamiel cv. (2.30 and 3.05 g/100g of dry bean, respectively). The total flavonoid content of leaf extracts (70% ethanol) from fig (Ficus carica L.), carob (Ceratonia siliqua L.) and pistachio (Pistacia lentiscus L.) plants were determined by using reverse phase high-performance liquid chromatography (HPLC)-and analyzed by UV/VIS array and electrospray ionization (ESI)-mass spectrometry (MS) detectors. As a base for comparison, flavonoid type and level were also determined in extracts from soybeans and grape seeds. It was found that the major flavonoids in Ficus are quercetin and luteolin, with a total of 631 and 681 mg/kg extract, respectively. In Ceratonia leaves, nine different flavonoids were detected. The major one was myricetin (1486 mg/kg extract), with a similar level in Pistacia (1331 mg/kg extract, myricetin). The present study is the first to report the presence of the isoflavone genistein in the Pistacia leaf, which was discovered to consist of about a third of the genistein level detected in soybean.  相似文献

以多糖、总黄酮、醇溶物和水溶物的得率及体外抗氧化活性为考察指标,研究了酒蒸和蜜蒸两种炮制方法对玉竹品质的影响。结果表明:酒蒸炮制玉竹的多糖、醇溶物得率最高,蜜蒸炮制玉竹总黄酮、水溶物的得率最高,比未炮制的玉竹(生品玉竹)中相应成分的得率分别提高了43.86%、29.53%、49.46%和34.66%。将多糖、水溶性浸出物、醇溶性浸出物及总黄酮四者得率相加的和进行比较,蜜蒸最好,蜜蒸为111.069%,酒蒸为107.309%,生品玉竹为80.926%。酒蒸炮制玉竹的多糖、水溶物对DPPH自由基的清除率均高于蜜蒸玉竹和生品玉竹,其DPPHIC50分别为0.345±0.019和0.441±0.022 mg·mL-1;蜜蒸炮制玉竹的总黄酮、醇溶物对DPPH自由基的清除率均高于酒蒸玉竹和生品玉竹,其DPPHIC50分别为0.047±0.011和0.199±0.036 mg·mL-1;在浓度为1 mg·mL-1时,蜜蒸玉竹总黄酮对DPPH自由基的清除率最大,为90.29%,超过了浓度为0.05 mg·mL-1的芦丁和槲皮素标品对DPPH自由基的清除率。两种炮制方法均提高了多糖、水溶物、总黄酮3种提取物的还原能力,但是降低了醇溶物的还原能力。  相似文献

Dietary phenolic compounds, ubiquitous in vegetables and fruits and their juices possess antioxidant activity that may have beneficial effects on human health. The phenolic composition of six commercial apple juices, and of the peel (RP), flesh (RF) and whole fresh Red Delicious apples (RW), was determined by high performance liquid chromatography (HPLC), and total phenols were determined by the Folin-Ciocalteau method. HPLC analysis identified and quantified several classes of phenolic compounds: cinnamates, anthocyanins, flavan-3-ols and flavonols. Phloridzin and hydroxy methyl furfural were also identified. The profile of phenolic compounds varied among the juices. The range of concentrations as a percentage of total phenolic concentration was: hydroxy methyl furfural, 4-30%; phloridzin, 22-36%; cinnamates, 25-36%; anthocyanins, n.d.; flavan-3-ols, 8-27%; flavonols, 2-10%. The phenolic profile of the Red Delicious apple extracts differed from those of the juices. The range of concentrations of phenolic classes in fresh apple extracts was: hydroxy methyl furfural, n.d.; phloridzin, 11-17%; cinnamates, 3-27%; anthocyanins, n.d.-42%; flavan-3-ols, 31-54%; flavonols, 1-10%. The ability of compounds in apple juices and extracts from fresh apple to protect LDL was assessed using an in vitro copper catalyzed human LDL oxidation system. The extent of LDL oxidation was determined as hexanal production using static headspace gas chromatography. The apple juices and extracts, tested at 5 microM gallic acid equivalents (GAE), all inhibited LDL oxidation. The inhibition by the juices ranged from 9 to 34%, and inhibition by RF, RW and RP was 21, 34 and 38%, respectively. Regression analyses revealed no significant correlation between antioxidant activity and either total phenolic concentration or any specific class of phenolics. Although the specific components in the apple juices and extracts that contributed to antioxidant activity have yet to be identified, this study found that both fresh apple and commercial apple juices inhibited copper-catalyzed LDL oxidation. The in vitro antioxidant activity of apples support the inclusion of this fruit and its juice in a healthy human diet.  相似文献

为探究黑苦荞的市场利用价值,该研究选择种植于湖北江汉平原低海拔地区的川荞1号和九江苦荞作为材料,分析苦荞籽粒中游离酚、结合酚、总酚、游离黄酮、结合黄酮和总黄酮的含量,利用DPPH自由基法、ABTS自由基法和铁离子还原抗氧化法(FRAP)三种抗氧化测试模型综合评价其体外抗氧化活性,并运用高效液相色谱(HPLC)技术对其酚类物质的组成进行鉴定。结果表明:(1)川荞1号籽粒的总酚和总黄酮含量显著高于九江苦荞,分别为27.38 mg GAE·g~(-1)DW、31.46 mg RE·g~(-1)DW和12.71 mg GAE·g~(-1)DW、14.68 mg RE·g~(-1)DW;其中游离酚与游离黄酮含量显著高于结合酚与结合黄酮含量,均占总酚和总黄酮含量的79%以上,且九江苦荞中结合酚和结合黄酮的含量高于川荞1号。(2)苦荞籽粒中酚类物质主要由芦丁、槲皮素、表儿茶素、山奈酚、山奈酚-3-芸香糖苷和槲皮素-3-O-芸香糖苷-3'-O-吡喃葡萄糖苷等黄酮类化合物组成,其中游离酚以芦丁和槲皮素为主,结合酚以表儿茶素和芦丁为主。(3)苦荞籽粒提取物均具有一定的抗氧化活性,黑苦荞川荞1号游离态DPPH、ABTS和FRAP抗氧化能力值分别为30.14、11.03、18.84 mg TE·g~(-1)DW,高于九江苦荞,而结合态三种抗氧化能力值低于九江苦荞,但黑苦荞川荞1号总抗氧化能力显著高于九江苦荞。在低海拔地区江汉平原,种植的黑苦荞川荞1号籽粒具有较高含量的酚类物质,符合后续的食品加工的生产要求,市场开发前景广阔。  相似文献

The marine phanerogam Syringodium filiforme, known as "manatee grass", is a common species that grows in coastal areas associated to Thalassia testudinum. With the aim to describe some of its possible chemical characteristics, this study was performed with a sample of 1.2 kg, collected in March 2009, in Guanabo beach, Havana, Cuba. The sample was dried (less than 12% humidity) and a total extract prepared; other three extracts were prepared with the use of solvents of increasing polarity. The phytochemical screening and analytical determinations of each fraction were undertaken Total polyphenol content was determined using pyrogallol as reference's standard; chlorophyll a and b and anthocyanin content were also quantified. Total extract and fractions antioxidant activity were evaluated by using the free radical scavenging activity assay with 1,1-Diphenyl-2-Picrylhydrazyl reactive (knowing as DPPH's method). The phytochemical screening of the different extracts detected the presence of high concentrations of flavonoids, phenols, terpenes, antocyanins, reducing sugars and alkaloids. The total extract and methanol fraction showed significant free radical scavenging properties, while the petroleum ether fraction showed moderate activity, and the chloroform fraction and the aqueous soluble precipitate (residual salt) obtained didn't show antioxidant properties against free radicals. The results of this work confirmed the potentialities of this species for biological purposes.  相似文献

The content of biologically active phenolic compounds (total polyphenols, tannins, flavonoids, and phenolic acids) were determined using spectrophotometry in four wild Croatian species of Daphne L. in the family Thymelaeaceae (Daphne alpina, D. cneorum, D. laureola, and D. mezereum). The concentration of total flavonoids (TF) was highest in the leaves of these Daphne species (0.12?C0.51% dry herb weight, DW) whereas the content of other phenolic compounds analyzed were highest in the roots, including total polyphenols (TP; 2.71?C19.03% DW), tannins (T; 1.14?C7.39% DW), and total phenolic acids (TPA; 0.12?C0.87% DW). D. alpina contained the highest amount of polyphenols, with the exception of flavonoids, where maximum concentrations were found in D. laureola. We also examined the antioxidant activity of leaf, stem, and root extracts. All extracts analyzed demonstrated high free radical scavenging activity with the highest concentration in the leaf extracts of D. alpina. Leaf extracts of D. cneorum showed the highest antioxidant activity in a ??-carotene bleaching assay.  相似文献

为探讨贯叶连翘对慢性应激大鼠生长和脑单胺类神经递质的影响,用15只大鼠设置对照组、应激组和贯叶连翘组3组实验。应激组和贯叶连翘组均进行7天的应激刺激后,贯叶连翘组灌胃贯叶连翘10d。实验结束后,取3组大鼠的脑组织,用高效液相色谱法测定高香草酸(HVA)、去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)的含量。结果表明,应激组大鼠日增重明显低于对照组;而贯叶连翘组大鼠的日增重明显高于应激组。应激组大鼠海马、纹状体和前额叶中的HVA、NE、DA和5-HT与对照组间均无显著差异。贯叶连翘组大鼠纹状体中的DA含量明显高于应激组;而前额叶中的DA则明显低于应激组。因此,贯叶连翘对慢性应激引起的大鼠生长受抑有缓解作用,对其脑内单胺类神经递质有部分调节作用。  相似文献

The newly established hyphenated instrumentation of LC/DAD/SPE/NMR and LC/UV/(ESI)MS techniques have been applied for separation and structure verification of the major known constituents present in Greek Hypericum perforatum extracts. The chromatographic separation was performed on a C18 column. Acetonitrile-water was used as a mobile phase. For the on-line NMR detection, the analytes eluted from column were trapped one by one onto separate SPE cartridges, and hereafter transported into the NMR flow-cell. LC/DAD/SPE/NMR and LC/UV/MS allowed the characterization of constituents of Greek H. perforatum, mainly naphtodianthrones (hypericin, pseudohypericin, protohypericin, protopseudohypericin), phloroglucinols (hyperforin, adhyperforin), flavonoids (quercetin, quercitrin, isoquercitrin, hyperoside, astilbin, miquelianin, I3,II8-biapigenin) and phenolic acids (chlorogenic acid, 3-O-coumaroylquinic acid). Two phloroglucinols (hyperfirin and adhyperfirin) were detected for the first time, which have been previously reported to be precursors in the biosynthesis of hyperforin and adhyperforin.  相似文献

In vitro pollen germination and pollen tube growth investigations are valuable tools used in identification of the effects of environmental factors and genotypic differences on pollen viability, pollen germination and tube elongation. In this study pollen viability, in vitro pollen germination capacity, abnormality ratios and tube length in germinated pollens of Hypericum perforatum L. and H. rumeliacum Boiss. were investigated. Both of these species has spheroid-shaped and tricolporate pollen grains. The diameters of Hypericum perforatum and H. rumeliacum pollens were found as 24 +/- 3 microm and 19 +/- 2 microm, respectively. Pollen viability of H. perforatum and H. rumeliacum was found as 83% and 72%, respectively. The germination percentages were found as 12.85% for H. perforatum and 64.42% for H. rumeliacurm. Tube lengths in germinated pollens of both taxa were measured approximately as 95.25 +/- 38 microm in H. perforatum and 165.92 +/- 53 microm in H. rumeliacium 4 h after inoculation. In germinated pollen grains of H. perlbratum and H. rumeliacumn abnormality percentages were determined as 13.23% and 43.97%, respectively. In germinated pollens of these two species, highly significant (P < 0.00001) differences in in vitro germination percents and abnormality percents were observed. Abnormalities such as swollen tube tip, branched tube, spiralled tube and excessive tube formation were observed in pollen tubes. The results of this study showed that there were obvious differences in pollen germinability between these two species growing under the same environmental conditions.  相似文献
