Responses of 93 neurons to isolated presentation of a single click and a series of 10 clicks with following frequency of 1000 Hz and responses of 66 neurons after the click had become a positive conditioned stimulus, and a series of 10 clicks had become a differential, negative stimulus, were investigated in chronic experiments on cats. Formation and realization of differential inhibition of an instrumental food reflex was shown not to lead to strengthening of inhibition in the auditory cortex, and the process of differential inhibition itself within the primary auditory cortex is not essentially an inhibitory process. Identical changes were found in responses of auditory cortical neurons to both positive and negative conditioned stimuli after training, evidence that neurons of the primary auditory cortex play a similar role in realization of the instrumental reflex and in its differential inhibition. It is suggested that the presence of groups of neurons responding by excitation or inhibition only to presentation of a stimulus with definite informative value is of great importance for differentiation.A. Evoked responses (EPs) of the auditory and the sensorimotor cortical areas were studied in experiments on cats during formation of a positive food-procuring conditioned reflex to a rhythmic sequence of clicks and during formation of the differentiation inhibition in response to the clicks of the same parameters, but with different frequency. In the positive conditioned reflex the EPs amplitude in the sensorimotor cortex increases; they are enriched by late components and in their general configuration they become much similar to EPs in the auditory area. The EPs patterns both in the auditory and in the sensorimotor areas considerably change in response to the differentiation stimulus, but differently in each of the sites.  相似文献

In tests on dogs the influence has been studied of bilateral electrolytic lesion of the caudate nuclei heads on realization of acquired conditioned instrumental defensive reactions to spatially separated sound signals (clicks series) under dichotic stimulation. It is shown that caudatectomy does not influence the dogs differentiation of the side of monaural sound stimulation, but leads to their absolute disability for a long time to differentiate right- and left-side positions of the sound image, modelled by changes in interaural difference in time of sound signal arrival at binaural stimulation.  相似文献

The responses of motor cortex neurons in the cat to the presentation of a single auditory click and a series of 10 clicks presented with 1,000/sec frequency were studied under conditions of chronic experiments before and after the development of an instrumental food reflex. After reflex development a single presentation of a positive conditioned stimulus (single click) markedly influenced for 7 sec the appearance of instrumental movements. At the same time, the immediate responses of motor cortex neurons to presentation of the conditioned auditory stimulus had no impact on the appearance in the motor cortex of discharges leading to the realization of instrumental movements. Consequently, motor cortex neurons do not require activation from afferent sensory inputs for the generation of such discharges. The immediate neuronal responses to conditioned stimulation did not inhibit the realization of the instrumental reflex. It is proposed that they are associated with the realization of motor function in the unconditioned defensive response evoked by the presentation of an auditory stimulus. The presence or absence of responses to auditory conditioned stimulation was dependent upon the signal meaning of the stimulus, its physical parameters, and the degree of excitability of the animal.A. What is the spatial and temporal nature of odor representations within primary olfactory networks at the threshold of an animal's ability to discriminate? Although this question is of central importance to olfactory neuroscience, it can only be answered in model systems where neural representations can be measured and discrimination thresholds between odors can be characterized. Here, we establish these thresholds for a panel of odors using a Pavlovian paradigm in the moth Manduca sexta. Moths were differentially conditioned to respond to one odor (CS+) but not another (CS-) using undiluted odorants to minimize salience-dependent learning effects. At 24 and 48 h postconditioning, moths were tested for the presence of a conditioned response (CR) with a blank, then the CS+ and CS- (pseudorandomly) across a 5-log step series of increasing concentration. Results identified discrimination thresholds and established that differential CRs to the CS+ and CS- increased with stimulus concentration. Next, 3 separate groups of moths were differentially conditioned at either one-log step below, at, or one log step above the identified discrimination threshold. At 24 and 48 h postconditioning, moths were tested sequentially with a blank, the concentration used for conditioning, and then undiluted odor. Conditioning at one log step below the discrimination threshold established a CR, indicating both stimulus detection and learning, but was insufficient to establish evidence of discrimination. Moths conditioned at the discrimination threshold were able to discriminate but only when stimulated with undiluted odors, indicating learning, but discrimination measures were hampered. When conditioned above the discrimination threshold, moths had no difficulty in discriminating. These results establish methods for psychophysical characterization of discrimination and indicate that differential conditioning at lowered concentrations biases threshold measures.  相似文献

We examined the local field potential of the hippocampus to monitor brain states during a conditional discrimination task, in order to elucidate the relationship between ongoing brain states and a conditioned motor reflex. Five 10-week-old Wistar/ST male rats underwent a serial feature positive conditional discrimination task in eyeblink conditioning using a preceding light stimulus as a conditional cue for reinforced trials. In this task, a 2-s light stimulus signaled that the following 350-ms tone (conditioned stimulus) was reinforced with a co-terminating 100-ms periorbital electrical shock. The interval between the end of conditional cue and the onset of the conditioned stimulus was 4±1 s. The conditioned stimulus was not reinforced when the light was not presented. Animals successfully utilized the light stimulus as a conditional cue to drive differential responses to the identical conditioned stimulus. We found that presentation of the conditional cue elicited hippocampal theta oscillations, which persisted during the interval of conditional cue and the conditioned stimulus. Moreover, expression of the conditioned response to the tone (conditioned stimulus) was correlated with the appearance of theta oscillations immediately before the conditioned stimulus. These data support hippocampal involvement in the network underlying a conditional discrimination task in eyeblink conditioning. They also suggest that the preceding hippocampal activity can determine information processing of the tone stimulus in the cerebellum and its associated circuits.  相似文献

Studies of olfactory function show that disruption of GABA A receptors within the insect antennal lobe (AL) disrupts discrimination of closely related odors, suggesting that local processing within the AL specifically enhances fine odor discrimination. It remains unclear, however, how extensively AL function has been disrupted in these circumstances. Here we psychophysically characterize the effect of GABA A blockade in the AL of the moth Manduca sexta. We used 2 GABA A antagonists and 3 Pavlovian-based behavioral assays of olfactory function. In all cases, we used matched saline-injected controls in a blind study. Using a stimulus generalization assay, we found that GABA A disruption abolished the differential response to related odors, suggesting that local processing mediates fine odor discrimination. We then assessed the effect of GABA A antagonist on discrimination thresholds. Moths were differentially conditioned to respond to one odor (reinforced conditioned stimulus [CS+]) but not a second (unreinforced conditioning stimulus [CS-]) then tested for a significant differential conditioned response between them across a series of increasing concentrations. Here, GABA A blockade disrupted discrimination of both similar and dissimilar odor pairs as indicated by generally increased discrimination thresholds. Finally, using a detection threshold assay, we established that GABA A blockade also increases detection thresholds. Because detection is a prerequisite of discrimination, this later finding suggests that disrupted discrimination may be due to impairment of the ability to detect. We conclude that the loss of ability to detect and subsequently discriminate is attributable to a loss of ability of the AL to provide a clear neural signal from background.  相似文献

Latent inhibition (LI) is a reduction in the rate of acquisition of a conditioned response that results from repeated preexposure of an animal to a conditioned stimulus (CS). The present experiment was conducted to assess the effect of bilateral lesions of 5-OT terminals of the nucleus accumbens on LI in a conditioned passive avoidance response paradigm. The lesions were produced by administration of 5,7-DHT and resulted in disruption of LI. Sham-operated animals displayed the delay of conditioning (LI) in comparison with the non-preexposed controls. Disruption of the LI was prevented by systemic injection of haloperidol. Involvement of 5-HT substrate of the nucleus accumbens and its interaction with dopaminergic system in the process of the LI development is discussed.  相似文献

The course of reversal learning in go -- no go avoidance reflex differentiation in both cats and dogs was analysed in terms of interrelations between drive and instrumental reflex activity. When signalling properties of the conditioned stimuli were reversed, marked changes in the drive state of experimental animals occurred. The increase of the fear drive influenced both the transfer of the instrumental response to the new positive conditioned stimulus and the extinction of the response to the previously positive stimulus. The quality of the conditioned stimuli and their reflexogenic strength exerted clear effects on the fear drive and on the course of reversal learning. Prefrontal lesions affected the drive state and the conditioned reflex activity of the animals during the reversal learning.  相似文献

Characteristics of the mismatch negativity (MMN) were studied by presenting the subjects with four blocks of stimuli containing standard series of clicks (90%) simulating a stationery sound image located in the head midline, and one of three different deviant series of clicks (10%) simulating either a stationary sound image located near the left ear or a moving sound image which shifted from the head midline to the left ear or in the opposite direction. All the deviant stimuli elicited the MMN with the minimal peak amplitude and the greatest latency evoked by the deviant series of clicks simulating the sound image moving from the head midline to the left ear. These findings suggest that the MMN may be considered as a pre-perceptual physiological measure of the discrimination accuracy for the sound signals with various spatial locations.  相似文献

In this paper we present a method of isolation and morphological and physiological characterization of groundwater bacteria based on numerical taxonomy and cluster analysis, and using a miniaturized test system (microtiter plates). Bacteria were isolated randomly on P-agar, and each strain was characterized in regard to 155 features. The media for biochemical differentiation are listed as well as methods of morphological discrimination. 246 strains of heterotrophic and oligotrophic bacteria, isolated from five water samples from different depths of the saturated groundwater area, were used for optimizing media and test reactions.  相似文献

Previous studies have shown that deep cerebellar nuclei (DCN)-lesioned mice develop conditioned responses (CR) on delay eyeblink conditioning when a salient tone conditioned stimulus (CS) is used, which suggests that the cerebellum potentially plays a role in more complicated cognitive functions. In the present study, we examined the role of DCN in tone frequency discrimination in the delay eyeblink-conditioning paradigm. In the first experiment, DCN-lesioned and sham-operated mice were subjected to standard simple eyeblink conditioning under low-frequency tone CS (LCS: 1 kHz, 80 dB) or high-frequency tone CS (HCS: 10 kHz, 70 dB) conditions. DCN-lesioned mice developed CR in both CS conditions as well as sham-operated mice. In the second experiment, DCN-lesioned and sham-operated mice were subjected to two-tone discrimination tasks, with LCS+ (or HCS+) paired with unconditioned stimulus (US), and HCS− (or LCS−) without US. CR% in sham-operated mice increased in LCS+ (or HCS+) trials, regardless of tone frequency of CS, but not in HCS− (or LCS−) trials. The results indicate that sham-operated mice can discriminate between LCS+ and HCS− (or HCS+ and LCS−). In contrast, DCN-lesioned mice showed high CR% in not only LCS+ (or HCS+) trials but also HCS− (or LCS−) trials. The results indicate that DCN lesions impair the discrimination between tone frequency in eyeblink conditioning. Our results suggest that the cerebellum plays a pivotal role in the discrimination of tone frequency.  相似文献

Neuronal activity in g. proreus was studied during classical secretory conditioning and its differentiation in dogs. Three types of changes in neuronal reaction pattern were identified during differential conditioned stimulus compared to the pattern observed during positive conditioned stimulus. It has been shown that signal significance of the conditioned stimulus may be coded by specific distribution of interspike intervals in response to different conditioned stimuli. In situation of successive inhibition or disinhibition of the differentiation some neurones displayed simultaneous appearance of interspike intervals specific for responses to current and previously presented conditioned stimuli.  相似文献

The rat's ability to vary its whisking "strategies" to meet the functional demands of a discriminative task suggests that whisking may be characterized as a "voluntary" behavior--an operant--and like other operants, should be modifiable by appropriate manipulations of response-reinforcer contingencies. To test this hypothesis we have used high-resolution, optoelectronic "real-time" recording procedures to monitor the movements of individual whiskers and reinforce specific movement parameters (amplitude, frequency). In one operant paradigm (N = 9) whisks with protractions above a specified amplitude were reinforced (Variable Interval 30 s) in the presence of a tone, but extinguished (EXT) in its absence. In a second paradigm (N = 3), rats were reinforced on two different VI schedules (VI-20s/VI-120s) signaled, respectively, by the presence or absence of the tone. Selective reinforcement of whisking movements maintained the behavior over many weeks of testing and brought it under stimulus and schedule control. Subjects in the first paradigm learned to increase responding in the presence of the tone and inhibit responding in its absence. In the second paradigm, subjects whisked at significantly different rates in the two stimulus conditions. Bilateral deafferentation of the whisker pad did not impair conditioned whisking or disrupt discrimination behavior. Our results confirm the hypothesis that rodent whisking has many of the properties of an operant response. The ability to bring whisking movement parameters under operant control should facilitate electrophysiological and lesion/behavioral studies of this widely used "model" sensorimotor system.  相似文献

Neuronal response in the cat association cortex (area 5) to conditioned and non-conditioned acoustic stimulation was investigated. Numbers of neurons responding to a conditioned acoustic stimulus according to the traditional reflex pattern were twice as high. Numbers of inhibitory neuronal responses to the stimulus increased when instrumental reflex occurred. Neurons were found which only reacted to a conditioned acoustic stimulus in the absence of conditioned reflex movement occurring with instrumental food reflex. Although findings do not exclude the possibility of this cortical area contributing to the analysis of sensory signals and evaluation of their biological significance, it might be supposed that its main functional property lies in its involvement in the process of initiating behavioral response to a conditioned response.A. The paper deals with the character of changes in the EP form recorded from the visual cortex of a rat during elaboration of a system of positive and negative (with different degree of precision) conditioned defensive reflexes. It has been shown that in a state of a consolidated habit of pain stimulus avoidance in a shuttle box a negative component with peak latency up to 70 msec develops in EP to the differentiation stimulus of 0.5 c/s. This component is much less pronounced to the 2 c/s conditioned stimulus. When a more precise differentiation stimulus (1 c/s) was presented to the animal in this state the development of the studied component depended on the subsequent behavioural reaction: it developed if the animal reacted in the same way as to the differentiation stimulus. This fact suggests that the generation of this component is connected with the inhibitory processes developing at the time when the animal is choosing its behavioural reaction.  相似文献

Evoked potentials (EPs) to clicks were recorded in the ectosylvian and sigmoid gyri of the cortex, in the medial geniculate body, and in the median nucleus of the thalamus by means of permanently implanted electrodes in experiments on 14 cats. A motor component of the behavioral response (conditioned food reflex) developed in response to the same stimulus (clicks). In the first group of animals the region of destruction was confined to the globus pallidus. In these animals EPs to clicks appeared during the first days after the operation in all structures, although in some tests they were reduced. Meanwhile the behavioral response to clicks was absent and showed no signs of spontaneous recovery. It could be formed (retraining) later in the experiments. In the second group of animals the globus pallidus was not completely destroyed, but neighboring areas of the entopeduncular nucleus and internal capsule were coagulated. In these cats more marked changes in behavioral and electrophysiological indices were observed. EPs to clicks disappeared in some cases for 25–30 days, subsequently returning to normal. The role of the globus pallidus as a "non-specific" polymodal structure in the analysis of sensory information is discussed.A. A. Experiments investigated a Pavlovian conditioning situation where the presence and absence of the stimulus are reversed temporally with respect to the presentation of a reward. Instead of a conditioned stimulus (e.g. odor) signaling the presence of a reward, the stimulus (e.g. odor) is present in the environment except just prior to the presence of the reward. Thus, the absence of the stimulus, or offset of the stimulus (e.g. absence of odor), serves as a conditioned stimulus and is the reward cue. Honey bees (Apis mellifera) were used as a model invertebrate system, and the proboscis‐conditioning paradigm was used as the test procedure. Using both simple Pavlovian conditioning and discrimination‐learning protocols, animals learned to associate the onset of an odor as conditioned stimuli when paired with a sucrose reward. They could also learn to associate the onset of a puff of air with a sucrose reward. However, bees could not associate the offset of an order stimulus with the presentation of a sucrose reward in either a simple conditioning or a discrimination‐learning situation. These results support the model that a very different cognitive architecture is used by invertebrates to deal with certain environmental situations, including signaled avoidance.  相似文献

The effects of external stimulus presentation on neuronal spike response induced in area 7 by conditioned and unconditioned stimuli were investigated in waking cats. Pointing reaction arising in response to the action of external stimuli led to the phasing out of spike response to unconditioned cutaneous stimulation, as well as depression or complete suppression of response in area 7 neurons to conditioned stimulus presentation, despite occurrence of conditioned reflex motion. External stimuli no longer exerted an inhibitory effect on the neuronal spike response induced by protracted use of conditioned and unconditioned stimuli. The fact that the conditioned lever-pressing reflex could occur either following or without neuronal spike response in area 7 would raise the question of whether neurons belonging to this area always participate directly in the triggering and occurrence of conditioned reflex motion.A. In 2-9 months dogs the influence was studied of ablation of the hippocampus dorsal area on formation and preservation of conditioned taste aversion (CTA), elaborated by combination of 30% sucrose solution with i.p. injection of 0.28 M LiCl solution. Lesion of the hippocampus dorsal area does not prevent acquisition after the first pairing of conditioned taste aversion in puppies and adult dogs. Heterogeneous influences are observed after hippocampus lesions on the process of CTA extinction in animals of different ages. Acquaintance with conditioned stimulus before CTA acquisition accelerates the process of its extinction in hippocampectomized indiviuals, but less than in animals with an intact hippocampus.  相似文献
