From 1972 to 1977 a large laboratory effort was devoted to determining data on efficacy, safety, environmental impact (on nontarget organisms), and some preliminary field work using several isolates of Bacillus sphaericus. The B. sphaericus strains were found to be specific in their mosquito larvicidal activity, not causing mammalian toxicity nor apparent perturbation of the environment. During this period several fermentation and industrialization problems were investigated so that by 1978, using new strains and cultures, it was possible to have prepared kilogram amounts of an active dry stable powder, of strain 1593, for field evaluation. These field evolution. These field evaluations are presently still in progress. Control has been seen particularly against Culex, Anopheles, and Psorophora species, with some what less control aganst Aedes species. Unlike the agriculturally oriented Bacillus thuringiensis candidates, B. sphaericus bacterial cell, which is digested in the larval midgut (within a peritrophic membrane), releasing a toxin as early as 15 min after ingestion. Subsequent death of the larva ensues. Recent evidence suggests that applied B. sphaericus powder will survive in aquatic situations (ditches, ponds, and tree holes) for at least nine month. Comparisons of the B. sphaeicus strains with recently isolated strains of B. thuringiensis (var. israelensis), the latter being particularly active against Aedes species, indicates that they may be useful complements of each other in overall mosquito control strategies. The recent isolation of several new strains of B. thuringiensis, from WHO-CCBC accessions from Roumania, indicate that although the B. thuringiensis isolate is a rare event when compared to the occurrence of B. sphaericus isolates (they usually occur together in accessions from which B. thuringiensis is isolated), several new useful strains of B. thuringiensis should be anticipated. The longevity of the B. thuringiensis strains in the wild has not yet been investigated.  相似文献

Aedes aegypti Act4 is a paralog of the Drosophila melanogaster indirect flight muscle actin gene Act88F. Act88F has been shown to be haploinsufficient for flight in both males and females (amorphic mutants are dominant). Whereas Act88F is expressed in indirect flight muscles of both males and females, expression of Act4 is substantially female-specific. We therefore used CRISPR/Cas9 and homology directed repair to examine the phenotype of Act4 mutants in two Culicine mosquitoes, Aedes aegypti and Culex quinquefasciatus. A screen for dominant female-flightless mutants in Cx. quinquefasciatus identified one such mutant associated with a six base pair deletion in the CxAct4 coding region. A similar screen in Ae. aegypti identified no dominant mutants. Disruption of the AeAct4 gene by homology-dependent insertion of a fluorescent protein marker cassette gave a recessive female-flightless phenotype in Ae. aegypti. Reproducing the six-base deletion from Cx. quinquefasciatus in Ae. aegypti using oligo-directed mutagenesis generated dominant female-flightless mutants and identified additional dominant female-flightless mutants with other in-frame insertions or deletions. Our data indicate that loss of function mutations in the AeAct4 gene are recessive but that short in-frame deletions produce dominant-negative versions of the AeAct4 protein that interfere with flight muscle function. This makes Act4 an interesting candidate for genetic control methods, particularly population-suppression gene drives targeting female viability/fertility.  相似文献

Efficacy of two tropical plant extracts for the control of mosquitoes   总被引:10,自引:0,他引:10  
The larvicidal activity of seed and leaf extracts of Calophyllum inophyllum and leaf extract of Rhinacanthus nasutus on the juveniles of Culex quinquefasciatus , Anopheles stephensi and Aedes aegypti was determined. Ethyl acetate (EA) soluble fractions of C. inophyllum and petroleum ether (PE) fraction of R. nasutus extracts showed very high larvicidal activity. Irrespective of the species of the mosquitoes tested, at concentrations ranging from 3.91 to 9.39 ppm, 9.04 to 35.49 ppm and 13.21 to 28.92 ppm of the active fractions of seed and leaf extracts of C. inophyllum and the leaf extract of R. nasutus , respectively, killed 50% of the treated larvae. The fractions also interfered with adult emergence. Exposure of the mosquitoes at any stage of their larval development to less than 0.86, 5.49 and 6.81 ppm of the active fractions of seed and leaf extracts of C. inophyllum and the leaf extract of R. nasutus , respectively, inhibited 50% of the treated larvae from emerging as adults. Experiments under laboratory and semi-field conditions showed that the activity of the extracts persisted for up to 10 days.  相似文献

In anti-malaria operations the use of DDT for indoor residual spraying has declined substantially over the past 30years, but this insecticide is still considered valuable for malaria control, mainly because of its low cost relative to alternative insecticides. Despite the development of resistance to DDT in some populations of malaria vector Anopheles mosquitoes (Diptera: Culicidae), DDT remains generally effective when used for house-spraying against most species of Anopheles, due to excitorepellency as well as insecticidal effects. A 1990 cost comparison by the World Health Organization (WHO) found DDT to be considerably less expensive than other insecticides, which cost 2 to 23 times more on the basis of cost per house per 6 months of control. To determine whether such a cost advantage still prevails for DDT, this paper compares recent price quotes from manufacturers and WHO suppliers for DDT and appropriate formulations of nine other insecticides (two carbamates, two organophosphates and five pyrethroids) commonly used for residual house-spraying in malaria control programmes. Based on these 'global' price quotes, detailed calculations show that DDT is still the least expensive insecticide on a cost per house basis, although the price appears to be rising as DDT production declines. At the same time, the prices of pyrethroids are declining, making some only slightly more expensive than DDT at low application dosages. Other costs, including operations (labour), transportation and human safety may also increase the price advantages of DDT and some pyrethroids vs. organophosphates and carbamates, although possible environmental impacts from DDT remain a concern. However, a global cost comparison may not realistically reflect local costs or effective application dosages at the country level. Recent data on insecticide prices paid by the health ministries of individual countries showed that prices of particular insecticides can vary substantially in the open market. Therefore, the most cost-effective insecticide in any given country or region must be determined on a case-by-case basis. Regional coordination of procurement of public health insecticides could improve access to affordable products.  相似文献

Use of an electrostatic sprayer for control of anopheline mosquitoes   总被引:3,自引:0,他引:3  
The Electrodyn sprayer was compared with a compression sprayer (Hudson X-pert) for residual application of cypermethrin, a pyrethroid insecticide, to control the malaria vectors Anopheles arabiensis Patton and An. funestus Giles in experimental huts at Magugu in Tanzania. The time taken for hut spraying, 2-2.5 min per hut, was similar for both types of sprayer. Two or three huts were treated internally with cypermethrin at 40 or 80 mg a.i./m2 using Electrodyn formulation for comparison with 80 mg a.i./m2 using wettable powder formulation. Each of the twelve huts (including five untreated controls) was fitted with window exit traps and either louvre or verandah traps for mosquito sampling. The Electrodyn sprayer was fitted with a pair of elbowed deflectrodes to direct the positively charged spray droplets onto walls and ceiling. All treatments gave 94-100% mortality-rates of indoor-resting anophelines throughout the evaluation period of 11 weeks post-spray. Reductions of An.arabiensis and An.funestus females by 10-42% and 62-91%, respectively, in rooms and by 72% and 51% in exit traps indicated that cypermethrin deterred mosquitoes from entering the huts. Overall mortality-rates of mosquitoes were 66% of both species in huts treated with 40 mg/m2 Electrodyn, 43% An.funestus and 71% An.arabiensis due to 80 mg/m2 Electrodyn formulation and 49% An.funestus and 64% An.arabiensis due to 80 mg/m2 WP formulation (no significant differences). It is concluded that the Electrodyn sprayer with deflectrodes is a convenient and effective means of residual house-spraying with pyrethroid insecticide for malaria vector control.  相似文献
