ABSTRACT: BACKGROUND: Lignocellulose is the most abundant biomass on earth. However, biomass recalcitrance has become a major factor affecting biofuel production. Although cellulose crystallinity significantly influences biomass saccharification, little is known about the impact of three major wall polymers on cellulose crystallization. In this study, we selected six typical pairs of Miscanthus samples that presented different cell wall compositions, and then compared their cellulose crystallinity and biomass digestibility after various chemical pretreatments. RESULTS: A Miscanthus sample with a high hemicelluloses level was determined to have a relatively low cellulose crystallinity index (CrI) and enhanced biomass digestibility at similar rates after pretreatments of NaOH and H2SO4 with three concentrations. By contrast, a Miscanthus sample with a high cellulose or lignin level showed increased CrI and low biomass saccharification, particularly after H2SO4 pretreatment. Correlation analysis revealed that the cellulose CrI negatively affected biomass digestion. Increased hemicelluloses level by 25% or decreased cellulose and lignin contents by 31% and 37% were also found to result in increased hexose yields by 1.3-times to 2.2-times released from enzymatic hydrolysis after NaOH or H2SO4 pretreatments. The findings indicated that hemicelluloses were the dominant and positive factor, whereas cellulose and lignin had synergistic and negative effects on biomass digestibility. CONCLUSIONS: Using six pairs of Miscanthus samples with different cell wall compositions, hemicelluloses were revealed to be the dominant factor that positively determined biomass digestibility after pretreatments with NaOH or H2SO4 by negatively affecting cellulose crystallinity. The results suggested potential approaches to the genetic modifications of bioenergy crops.  相似文献

The cost efficiency of the biorefining process can be improved by extracting high-molecular-mass hemicelluloses from lignocellulosic biomass prior to ethanol production. These hemicelluloses can be used in several high-value-added applications and are likely to be important raw materials in the future. In this study, steam pretreatment in an alkaline environment was used to pretreat the lignocellulosic biomass for ethanol production and, at the same time, extract arabinoxylan with a high-molecular-mass. It was shown that 30% of the arabinoxylan in barley straw could be extracted with high-molecular-mass, without dissolving the cellulose. The cellulose in the solid fraction could then be hydrolysed with cellulase enzymes giving a cellulose conversion of about 80–90% after 72 h. For wheat straw, more than 40% of the arabinoxylan could be extracted with high-molecular-mass and the cellulose conversion of the solid residue after 72 h was about 70–85%. The high cellulose conversion of the pretreated wheat and barley straw shows that they can be used for ethanol production without further treatment. It is therefore concluded that it is possible to extract high-molecular-mass arabinoxylan simultaneously with the pretreatment of biomass for ethanol production in a single steam pretreatment step.  相似文献

The study documented the potential of isolated filamentous fungus Aspergillus sp. as whole cell biocatalyst for biodiesel production using Sabourauds dextrose broth medium (SDBM) and corncob waste liquor (CWL) as substrates. SDBM showed improvement in both biomass production (13.6 g dry weight/1000 ml) and lipid productivity (23.3%) with time. Lipid extraction was performed by direct (DTE) and indirect (IDTE) transesterification methods. DTE showed higher transesterification efficiency with broad spectrum of fatty acids profile over IDTE. CWL as substrate showed good lipid productivity (22.1%; 2g dry biomass; 48 h) along with efficient substrate degradation. Lipids derived from both substrates depicted high fraction of saturated fatty acids than unsaturated ones. Physical characteristics of fungal based biodiesel correlated well with prescribed standards. CWL derived biodiesel showed relatively good fuel properties (acid number, 0.40 mg KOH/g of acid; iodine value, 11 g I?/100 g oil; density, 0.8342 g/cm3) than SDBM derived biodiesel.  相似文献

The spent liquor (SL) of neutral sulfite semi‐chemical (NSSC) pulping process contains about 8 wt% lignocelluloses that can be extracted and used in the production of value‐added materials. In this work, a flocculation process followed by centrifugation was considered for isolating lignosulfonate and hemicelluloses from SL. It was observed that, by adding 20 mg/g of polydiallyldimethylammuniom chloride (PDADMAC) with 100,000–200,000 g/mol molecular weight to SL, 45% of lignosulfonate and 39% of hemicelluloses were removed at 30°C. The lignocellulose removal was more efficient for the dual flocculation system of low and high molecular weights PDADMAC than for individual PDADMAC systems. Overall, 49% of lignosulfonate, 47% of hemicelluloses and 97% of turbidity were removed from SL from the dual system when 10 mg/g low molecular weight PDADMAC and 10 mg/g high molecular weight PDADMAC were added to the SL at 30°C, subsequently. The thermogravimetric analysis (TGA) of generated flocs showed that all samples had similar thermal behaviour and 13–16 wt% of flocs remained as ash after burning at 700°C in nitrogen. As the flocs are made of lignocellulosic materials and they are thermally stable, they could be used as fillers in paper board production. © 2016 American Institute of Chemical Engineers Biotechnol. A three-stage control strategy independent of the organic substrate was developed for automated substrate feeding in a two-phase fed-batch culture of Cupriavidus necator DSM 545 for the production of the biopolymer polyhydroxybutyrate (PHB). The optimal feeding strategy was determined using glucose as the substrate. A combined substrate feeding strategy consisting of exponential feeding and a novel method based on alkali-addition monitoring resulted in a maximal cell concentration in the biomass growth phase. In the PHB accumulation phase, a constant substrate feeding strategy based on the estimated amount of biomass produced in the first phase and a specific PHB accumulation rate was implemented to induce PHB under limiting nitrogen at different biomass concentrations. Maximal cell and PHB concentrations of 164 and 125 g/L were obtained when nitrogen feeding was stopped at 56 g/L of residual biomass; the glucose concentration was maintained within its optimal range. The developed feeding strategy was validated using waste glycerol as the sole carbon source for PHB production, and the three-stage control strategy resulted in a PHB concentration of 65.6 g/L and PHB content of 62.7% while keeping the glycerol concentration constant. It can thus be concluded that the developed feeding strategy is sensitive, robust, inexpensive, and applicable to fed-batch culture for PHB production independent of the carbon source.  相似文献

Dissolved lignocelluloses from the pre-hydrolysis liquor (PHL) of kraft-based dissolving pulp production process were recovered by adsorption to lime mud produced in the causticizing plant of the kraft process. The adsorption of lignocelluloses was a fast process, and could be completed within one hour. The addition of polydiallyldimethylammonium chloride (PDADMAC) significantly increased the amounts of adsorbed lignin and hemicelluloses, which more than doubled at the PDADMAC dosage of 0.1% (based on the weight of PHL). The measured heating values of the adsorbed lignocelluloses indicate that adsorption of lignocelluloses to lime mud may result in the energy saving of the lime kiln. The process proposed in this study could also be adapted to decrease inhibitor concentrations (lignin and acetic acid) if the dissolved hemicelluloses in the PHL were used to produce value-added products, e.g., ethanol, xylitol, based on the fermentation process.  相似文献

Solid state anaerobic digestion, as a safe and environment-friendly technology to dispose municipal solid wastes, can produce methane and reduce the volume of wastes. In order to raise the digestion efficiency, this study investigated the pretreatment of yard waste by thermal or chemical method to break down the complex lignocellulosic structure. The composition and structure of pretreated yard waste were analyzed and characterized. The results showed that the pretreatment decreased the content of cellulose and hemicelluloses in yard waste and in turn improved the hydrolysis and methanogenic processes. The thermal pretreatment sample (P1) had the highest methane yield, by increasing 88 % in comparison with digesting the raw material. The maximum biogas production reached 253 mL/g volatile solids (VS). The largest substrate mass reduction was obtained by the alkaline pretreatment (P5). The VS of the alkaline-treated sample decreased about 60 % in comparison with the raw material.  相似文献

An immune-enhancing strain, Lactobacillus plantarum Pi06, isolated from a healthy infant was used for biomass production following optimization of the medium in shake-flask culture. Preliminary studies showed that commercial MRS medium and cultivation under static conditions generated higher biomass production than four other tested media with or without a shaking condition. The selected medium composition, consisting of glucose, yeast extract, soy peptone, ammonium citrate, and corn steep liquor, was further optimized using a systematic method that integrated the Taguchi array design and the Box-Behnken method. The response effects of these factors were first investigated using Taguchi design under an L 16 (45) array. The suggested medium composition, derived from Statistica 7.1 using the Taguchi design, was applied to cultivate cells and a biomass of 7.16 g dry cell weight (DCW)/L was obtained. Response surface methodology based on the Box-Behnken method for the three response variables of glucose, yeast extract, and corn steep liquor was then used to further increase the biomass level to 8.94 g DCW/L. The resulting optimum medium consisted of 35 g/L glucose, 35 g/L yeast extract, and 40 mL/L corn steep liquor. Compared with the initial medium, the biomass yield was improved from 4.31 to 8.94 g DCW/L, an enhancement of approximately 107%.  相似文献

This study proposed a novel waste utilization bioprocess for production of lactic acid and fungal biomass from waste streams by fungal species of Rhizopus arrhizus 36017 and R. oryzae 2062. The lactic acid and fungal biomass were produced in a single-stage simultaneous saccharification and fermentation process using potato, corn, wheat and pineapple waste streams as production media. R. arrhizus 36017 gave a high lactic acid yield up to 0.94-0.97 g/g of starch or sugars associated with 4-5 g/l of fungal biomass produced, while 17-19 g/l fungal biomass with a lactic acid yield of 0.65-0.76 g/g was produced by the R. oryzae 2062 in 36-48 h fermentation. Supplementation of 2 g/l of ammonium sulfate, yeast extract and peptone stimulated an increase in 8-15% lactic acid yield and 10-20% fungal biomass.  相似文献

This study focuses on the isolation and characterization of bacteria from municipal waste and peat to determine those bacteria with good potential for modification and decomposition of lignocellulosic biomass for industrial application. Twenty cellulase-producing bacteria belonging to four major phyla - Firmicutes, Actinobacteria, Proteobacteria and Bacteroidetes - were found when screened on carboxymethyl cellulose-containing agar. Six isolates also exhibited activities towards filter paper as the sole carbon source in salt media, while 12 exhibited activities towards xylan when screened on xylan-containing plates. Moreover, 5 isolates survived in and increased the absorbance of 1% black liquor in salt media by an average of 2.07-fold after 21 days of incubation. Similarly, these 5 isolates increased the absorbance of 0.1% pure lignin at 280 nm in salt media, indicating modification of lignin. Additionally, the Fourier transform infrared spectroscopy analysis of 1% barley straw treated for 21 days with these 5 strains showed a preference for consumption of hemicelluloses over lignin; however, a change in lignin was observed. A Bacillus strain (55S5) and a Pseudomonas strain (AS1) displayed the greatest potential for lignocellulose decomposition due to a variety of cellulase activities, as well as xylanase activity and modification of lignin. Several of these isolates have good potential for industrial use in the degradation of lignocellulosic biomass.  相似文献

Hemicelluloses in industrially produced pre-hydrolysis liquor (PHL) were precipitated with ethanol. These PHL-derived hemicelluloses (PHL-EH) and a commercial, pure birch wood xylan sample (powder form) (BWX) were bleached using chlorine dioxide (D(0) and D(1)) and hydrogen peroxide (Ep) in the D(0)EpD(1) sequence, and the chemical compositions, molecular weights and charge densities of the treated samples were assessed. When applied to high-yield pulp (HYP) at 50 mg/g, 26 and 20 mg/g of the bleached PHL-EH and BWX, respectively, were adsorbed without significantly affecting paper properties. These results suggest that semi-bleached hemicelluloses could be used to increase the basis weight of paper products. Furthermore, an integrated process was proposed that converts the kraft-based dissolving pulp production process into a biorefinery unit with dissolving pulp, bleached hemicelluloses and lignin as main products.  相似文献

In the biological pretreatment process, white-rot fungi are mostly used to degrade lignin and carbohydrates in lignocellulosic biomass. In this study, water-soluble hemicelluloses were recovered from birch wood (Betula alnoides) decayed by white-rot fungi (Ganoderma lucidum C7016) for different durations up to 16 weeks. Accordingly, the dimethyl sulfoxide (DMSO)-soluble hemicelluloses were isolated from the untreated birch wood as a comparison. Results showed that the fungal-degraded polysaccharides were acidic hemicelluloses having a high content of uronic acids ranging from 20.6 to 22.5 %. Gel permeation chromatography analysis demonstrated that the recovered water-soluble hemicelluloses had a lower average molecular weight (M w, 15,990–27,560 g?mol?1) than that of the DMSO-soluble hemicelluloses (M w , 33,960 g?mol?1). Fourier transform infrared spectroscopy, scanning electron microscopy, one- and two-dimensional nuclear magnetic resonance spectroscopy also revealed significantly changes between those of fungal degraded and DMSO-soluble hemicelluloses. It was proposed that the hemicelluloses with low molecular weights were easily removed from wood by fungal degradation. This research revealed the changes of hemicelluloses in fungal degradation in the natural environment, which may enable the exploration of novel methods in bioconversion of lignocellulosic biomass for the production of biofuels and biopolymers, in addition to the development of new and better ways to protect wood from biodegradation by microorganisms.  相似文献

Summary In a newly constructed one-vessel dialysis fermentor, a strain of Staphylococcus carnosus TM300 carrying the lipase secretion plasmid pLipPS1 was used to investigate exoenzyme and biomass production. The bacterial culture grows in an inner compartment of 21 volume, separated from a 101 nutrient broth compartment by a conventional dialysis membrane. In order to avoid substrate depletion and to prolong the growth phase, a highly concentrated nutrient broth was used. The biomass production reached 60 g cell dry weight/l. The increase in extracellular lipase concentration was directly coupled with the increase of cell mass and reached a value of 230 mg/l culture supernatant. Harvesting the cells in the late growth phase, the lipase content was about 30% of the total exoproteins in the supernatant.  相似文献

目的:利用甘薯淀粉废水发酵低成本获取微生物油脂。方法:以甘薯淀粉废水为发酵基质,进行菌株筛选、发酵工艺优化及油脂成分的气相色谱分析。结果:筛选出一株刺孢小克银汉霉F7,生物量为19.375g/L,含油量为45.1%。菌株F7发酵第11天生物量达到18.140g/L,含油量达到51.2%,COD去除率87%。研究发现与对照相比,NaAc和KH2PO4对生长、产油以及出水COD去除有显著促进作用,NaAc2g/L时生物量提高了25.4%,含油量提高了4.4%,COD下降了52.0%,KH2PO4的作用稍次之。结论:资源化利用甘薯淀粉废水发酵生产微生物油脂同时降低废水COD是一条可行的途径,可以为生物柴油提供廉价油源。  相似文献

Euglena gracilis is shown to be able to grow on potato liquor as the main medium component leading to an interesting biotechnological product represented by paramylon – a β‐1,3‐glucan – and, at the same time, revaluating an otherwise annoying waste stream of the potato‐starch industry. Paramylon mass fractions of about 75% are obtained for biomass concentrations of 15 g/L during simple batch cultivation under heterotrophic conditions. Supplementation of the growth medium with glucose and the vitamins B1 and B12 are shown to improve growth rate as well as paramylon content. E. gracilis grows best at about 27.5°C without requiring pH control.  相似文献

Staphylococcus aureus cultivated in liquid media containing untreated cornsteep liquor (CSL) and alkali-treated CSL produced similar biomass yields (6.5–6.9 g/L). However, contents of protein A in the biomass was 0.5% and 1.56% for untreated CSL and treated CSL, respectively. Addition of treated CSL at 20 g/L achieved optimal enhancement of protein A production (0.11 g/L). Probable factors associated in treated CSL for the enhanced protein A production are discussed.  相似文献

The aim of this work is to define an extraction process of hemicelluloses from hardwood chips, prior to their transformation into paper pulp by the kraft process. Eucalyptus wood chips were submitted to autohydrolysis and acid hydrolysis treatments. Autohydrolysis means that no extra acid is added during the heating of the wood suspension, whereas sulphuric acid is added for the acid hydrolysis treatment. The liquor to wood ratio was 4, and temperature and time were varied so as to optimise the hemicelluloses extraction. Quantities of xylans varying between 12 and 38 g/l of hydrolysate (or between 9 and 15% on wood) could be extracted.Kraft cooking process was applied to the pre-hydrolysed wood chips. It was shown that they were easier to delignify than untreated wood chips, and the resulting pulp was also easier to bleach. The resulting cellulosic fibres were characterised for their papermaking properties: the higher the pentosan removal, the lower the strength properties of the pulps produced.  相似文献

This work investigated effects of lignocellulose degradation products on cell biomass and lipid production by Cryptococcus curvatus. Furfural was found to have the strongest inhibitory effect. For the three phenolic compounds tested, vanillin was the most toxic, while PHB and syringaldehyde showed comparable inhibitions in the concentration range of 0–1.0 g/L. Generally little significant differences on the relative cell biomass and lipid contents at the same concentrations of tested compounds were observed between glucose and xylose as a sole carbon source. At 1.0 g/L of furfural, the cell biomass and lipid content decreased by 78.4% and 61.0% for glucose as well as 72.0% and 59.3% for xylose, respectively. C. curvatus ceased to grow at concentrations of PHB over 1.0 g/L or vanillin over 1.5 g/L. The strain could survive in the presence of syringaldehyde up to 2.0 g/L for glucose or 1.5 g/L for xylose. The compounds’ negative impact was reduced by an increase in inoculum size and a 10% (v/v) seed was detected to be optimal for cell biomass and lipid production. The results demonstrated C. curvatus could effectively utilize most of the dominant monosaccharides and cellobiose existing in lignocellulosic biomass hydrolysate in the presence of toxic compounds.  相似文献
