The rheological properties of aqueous solutions of guar gum (GG) and hydroxyethyl guar gum (HEG) have been investigated. The flow properties of these polysaccharide solutions were studied at the shear rate in the range 1.5–1310s−1 using a Rheotest-2 viscometer. The flow of these polysaccharide solutions was described by equation of state based on Cross model. The basic rheological parameters, like zero shear rate viscosity (ηo), elasticity modulus (Go) and relaxation time (glo) were calculated using simple and established relations. Master viscosity curves indicated that the molecular weight distribution of native guar gum has been changed by hydroxyethylation under specified reaction conditions. The effect of concentration and temperature on ηo and λo has been studied, and the relations among these were established by simple equations.  相似文献

A water-soluble seed gum was isolated from seed endosperm of Cassia javahikai. The acid-catalyzed fragmentation, methylation, selective enzymatic degradation and periodate oxidation suggested a heteropolymeric structure for the polysaccharide. The polysaccharide was shown to have a linear chain of β(1 → 4) linked d-mannopyranosyls units with side chains of α(1 → 6) d-galactopyranosyl units. Grafting of polyacrylamide onto the gum was performed using K2S2O8/ascorbic acid redox system in presence of Ag+ as catalyst at 35 ± 2 °C. The viscosity of the gum solution increased on grafting and the grafted gum was observed to resist biodegradation for more than 256 h. Thermogravimetric analysis revealed that grafted gum was more thermally stable than native gum.  相似文献

Type 2 diabetes mellitus (T2DM) is associated with increased plasma triglyceride (TG) concentrations, but African Americans (AA) have lower plasma TG than Caucasians (CC). We evaluated the hypothesis that obese AA women have lower plasma TG than obese CC women do because of differences in lipid kinetics. Eleven AA and 11 CC obese women with T2DM, matched on body mass index (BMI) (AA = 37 ± 1, CC = 37 ± 1 kg/m2), age, duration of diabetes, percentage body fat, and insulin sensitivity (SI, determined by an intravenous glucose tolerance test), were studied. Plasma TG concentration (AA = 1.14 ± 0.11, CC = 1.88 ± 0.18 mmol/l), FFA rate of appearance (Ra) into plasma (AA = 419 ± 27, CC = 503 ± 31 µmol·min−1), and total VLDL-TG secretion rate (AA = 18 ± 2, CC = 29 ± 4 µmol·min−1) were lower in AA than CC women (all P < 0.05). In contrast, plasma total apolipoprotein (apo)B-100 concentration (AA = 1,542 ± 179, CC = 1,620 ± 118 nmol/l) and VLDL-apoB-100 secretion rate (AA = 1.3 ± 0.1, CC = 1.3 ± 0.1 nmol·min−1) were similar in both groups, so the molar ratio of VLDL-TG secretion rate to VLDL-apoB-100 secretion rate was lower in AA women than in CC women. VLDL-TG concentration was lower in AA women due to lower total VLDL-TG secretion rate. However, the VLDL-apoB-100 secretion rate was the same in both groups, demonstrating that AA women secrete smaller VLDL particles containing less TG than do CC women.  相似文献

Guar gum (G) is a simple characterized branched polysaccharide, which is frequently used in food industries. We prepared the gum C-glycosylated derivative (GG), and its sulphated derivative (SGG), aiming to characterize their cancer chemopreventive, and anti-inflammatory properties. Estimation of cancer chemopreventive activity, specifically anti-initiation, including the modulation of carcinogen metabolism and the antioxidant capacity, revealed that GG was a potent anti-initiator, where it inhibited not only the carcinogen activator enzyme, cytochrome P450 1A (CYP1A), but also induced the carcinogen detoxification enzymes glutathione-S-transferases (GSTs), while SGG inhibited both CYP1A and GSTs. SGG was an effective radical scavenger than GG against hydroxyl, peroxyl, and superoxide anion radicals. GG and SGG were found to modulate the macrophage functions into an anti-inflammatory pattern. Thus, both enhanced the macrophage proliferation and phagocytosis of fluorescein isothiocyanate (FITC)-zymosan; however, they also inhibited strongly the nitric oxide generation and tumor necrosis factor-alpha secretion in lipopolysaccharide (LPS)-stimulated RAW macrophage 264.7. Unexpectedly, both GG and SGG dramatically inhibited the binding affinity of FITC-LPS to RAW 264.7, as indicated by flow cytometry analysis. GG and SGG exhibited a significant anti-proliferative activity against human hepatocellular carcinoma cells (Hep G2), and only SGG was specifically cytotoxic for human breast carcinoma cells (MCF-7), but neither was significantly cytotoxic for human lymphoblastic leukemia cells (1301). SGG led to a major disturbance in cell cycle phases of Hep G2 cells as indicated by concomitant arrest in S- and G2/M-phases, a disturbance that was associated with an induced cell death as a result of necrosis, but not apoptosis in both GG- and SGG-treated cells. Taken together, the modified gums could be used as an alternative of G in health food industries to provide cancer prevention in risk populations.  相似文献

The amino acid compositions of the proteinaceous components of the gum exudates from Prosopis alba, P. chilensis, P. glandulosa, P. laevigata, P. torreyana and P. velutina, and for a sample of commercial gum mesquite, are presented. In agreement with data published previously for the polysaccharide components of their gums, only minor differences in composition are shown by these species. The amino acid compositions are characterized by very high proportions of hydroxyproline and by high proportions of proline and serine; these three amino acids account for 62.5% of those present in the gum from Prosopis velutina. The amino acid compositions of these Prosopis gums are remarkably similar to that established recently for the gum from Acacia senegal (gum arabic).  相似文献

Australian gum specimens from Acacia saliciformis, A. xanthina, A. rostellifera, A. murrayana (two specimens differing in the mode of initiation of gum exudation), A. georginae, A. cyclops, A. implexa, and an un-named species (Maslin ‘P31’) have been analysed. The first four of these are placed within Bentham's Series 1, subseries 6F, A. georginae within subseries 7E, and the remainder within subseries 7F. These data extend considerably the ranges of the analytical parameters reported previously for phyllodine species. The molecular weights of the gums from A. cyclops and A. implexa are much higher than those reported earlier for South African specimens; this may affect some taxonomic deductions based on their examination. The gum composition of A. saligna can no longer be regarded as atypical of a phyllodinous species; a suggestion that A. saligna should be transferred to the section Juliflorae may require reconsideration. The major difference between the specimens of gum from A. murrayana lies in their nitrogenous content. Data are reported for the amino acid compositions of the gums from A. saliciformis and A. xanthina.  相似文献

The purpose of this study was to identify endogenous factors that induce gummosis and to show their role in gummosis in tulip (Tulipa gesneriana L. cv. Apeldoorn) stems. Using procedures to detect endogenous factors that induce gum in the stem of tulips, jasmonic acid (JA) and methyl jasmonate (JA-Me) were successfully identified using gas–liquid chromatography–mass spectrometry. Total amounts of JA and JA-Me designated as jasmonates in tulip stems were also estimated at about 70–80 ng/g fresh weight, using deuterium-labeled jasmonates as internal standards. The application of JA and JA-Me as lanolin pastes substantially induced gums in tulip stems with ethylene production. The application of ethephon, an ethylene-generating compound, however, induced no gummosis although it slightly affected jasmonate content in tulip stems. These results strongly suggest that JA and JA-Me are endogenous factors that induce gummosis in tulip stems.  相似文献

Many plant gums, such as gum arabic, contain hydroxyproline-rich glycoproteins (HRGPs), which are also abundant components of the plant cell extracellular matrix. Here we expressed in transgenic BY2 Nicotiana tabacum (tobacco) cells, a synthetic gene encoding a novel HRGP-based gum, designated gum arabic-8 or (GA)(8). (GA)(8) encoded eight repeats of the consensus polypeptide sequence of gum arabic glycoprotein (GAGP): Gly-Pro-His-Ser-Pro-Pro-Pro-Pro-Leu-Ser-Pro-Ser-Pro-Thr-Pro-Thr-Pro-Pro-Leu, in which most of the Pro residues were posttranslationally modified to hydroxyproline (Hyp). (GA)(8) was expressed as a green fluorescent protein (GFP) fusion protein targeted to the culture medium, (GA)(8)GFP. The culture of the transgenic cells in a 5-L bioreactor showed that the production of (GA)(8)GFP was cell growth-associated. The extracellular yield of (GA)(8)GFP was 116.8 mg/L after 14 days of culture and accounted for 87% of the total fusion protein expressed. (GA)(8)GFP was purified from the culture medium by a combination of hydrophobic interaction, gel permeation, and reversed phase chromatography. Biochemical characterization indicated that the amino acid composition of the (GA)(8) module, after removal of GFP by proteolysis, was virtually identical to that of predicted by the GAGP consensus sequence and that carbohydrate, which occurred as arabinogalactan polysaccharides and small oligoarabinosides O-linked through the Hyp residues, accounted for 84% of the molecules' dry weight. Functional assays showed that (GA)(8) exhibited low viscosity in aqueous solution similar to native GAGP. However, neither GFP alone nor the (GA)(8) module could emulsify orange oil. However, the fusion protein (GA)(8)GFP possessed 1.28-fold better emulsification properties than native GAGP. This work demonstrates the feasibility and potential of a synthetic gene approach to the de novo design of novel glycoprotein-based gums and emulsifiers.  相似文献

A gum that exudes from the wounded trunk of the New Zealand native tree Meryta sinclairii has been isolated. The gum was completely precipitated by the β-glucosyl Yariv reagent and was thus determined to be an arabinogalactan-protein (AGP). It contained >95% w/w carbohydrate and only 2% w/w protein with a high level of hydroxyproline. SEC-MALLS showed that the gum had a weight-average molecular weight of 4.45×106 Da compared with 6.02×105 Da for gum arabic. Constituent sugar and linkage analyses were consistent with polymers comprised of a highly branched backbone of 1,3-linked galactopyranosyl (Galp) residues, with side-chains made up of arabinofuranose- (Araf) containing oligosaccharides, terminated variously by rhamnopyranosyl (Rhap), arabinopyranosyl (Arap), Galp and glucuronopyranosyl (GlcpA) residues. Analysis by one-dimensional and two-dimensional 1H and 13C NMR experiments confirmed the linkage analyses. The structure of the gum is discussed in comparison with the structure of gum arabic and other AGPs.  相似文献

Analytical data are presented for the polysaccharide of six gum specimens from Acacia tortuosa, a tropical American Gummiferae species. These data show that the gums were basically similar but differed in the sugar composition reported for most Acacia gum exudates studied so far. The presence of traces of xylose (<1%) and the absence of rhamnose in the gum specimens studied was very unusual.  相似文献

Acacia gum exudates are proteinaceous polysaccharides; their protein content ranges from ca 0.2 to 45%.The data presented show that the amino acid compositions of the gums from 12 phyllodinous species (10 from Bentham's sub-series Uninerves racemosae, two from sub-series Juliflorae) also vary considerably, particularly in respect of their hydroxyproline content (55 residues per 1000 residues in A. aestivalis gum, 287 residues per 1000 in A. saliciformis gum). The proportions of some other amino acids, e.g. alanine, aspartic acid, proline and serine also vary considerably, but the proportions of others, e.g. cystine, methionine, histidine, threonine, tyrosine and valine, are remarkably constant. The amino acid composition of gums with a very low protein content (e.g. A. victoriae and A. mycrobotrya) is similar to that for a highly proteinaceous gum (A. tumida). There are, however, considerable differences between the amino acid compositions of the gums from A. saligna and A. pycnantha (South African and Western Australian specimens). This strengthens previous chemotaxonomic evidence, based on the polysaccharide parameters of their gums, that these two species are not as close taxonomically as was originally believed from morphological considerations.  相似文献

Australian gum specimens from Acacia aestivalis, A. chrysella, A. jennerae and A. microbotrya (five specimens differing slightly in some morphological characters) have been studied. These species, placed within Bentham's Series 1, subseries 6F (Uninerves racemosae) are closely related, forming part of the recognized A. microbotrya group. The five specimens from A. microbotrya show minor variations, similar in extent to those established previously for gums from other species. The gums from A. chrysella and A. jennerae are similar to those from A. microbotrya in chemical composition. The gum from A. aestivalis differs from those from A. microbotrya, A. chrysella and A. jennerae in two main respects: it is more acidic and has a much higher methoxyl content. Thus significant differences in gum composition can be shown by some species that differ only slightly in morphological characters. Data for the amino acid compositions of the proteinaceous components of the gums from A. aestivalis, A. jennerae and A. microbotrya, differ considerably from those for the gums from other species belonging to the Uninerves racemosae, e.g. A. saliciformis and A. xanthina, which are much more viscous and have higher proteinaceous contents containing much higher proportions of the amino acids commonly involved in linkages with sugars. Of the closely related species studied, A. aestivalis is closer to A. microbotrya than A.jennerae in terms of the amino acid compositions of their gums, a reversal in the relative affinities shown by their polysaccharide parameters. Thus amino acid compositions are of interest chemotaxonomically and also in terms of the tertiary structures of Acacia gum exudates.  相似文献

The seeds of Cassia occidentalis, an annual weed occurring throughout India, is a rich source of galactomannan gum. The gum derived from seed endosperm can be potentially utilized in a number of industries to replace the conventional gums. With a view to utilize the gum for broader applications, carbamoylethylation of C. occidentalis seed gum was carried out with acrylamide in presence of sodium hydroxide under different reaction conditions. Variables studied were concentration of sodium hydroxide, acrylamide, gum–solvent ratio, reaction time and temperature. The nitrogen content, carboxyl content and total ether content were determined. The optimum condition for preparing carbamoylethyl C. occidentalis seed gum (%N=2.57) comprised concentration of acrylamide (0.070 mol), sodium hydroxide (0.125 mol), C. occidentalis seed gum (0.03 mol) at 30 °C for 3 h. Rheological properties of carbamoylethyl C. occidentalis seed gum solution showed non-Newtonian pseudo-plastic behavior, relatively high viscosity, cold water solubility and solution clarity vis-à-vis unmodified C. occidentalis seed gum.  相似文献

Analytical data are presented for the water-soluble gum exudates from Brachystegia glaucescens, B. spiciformis, and Julbernardia globiflora. They are acidic polysaccharides containing glucuronic acid, 4-O-methylglucuronic acid and galacturonic acid, together with galactose, minor amounts of arabinose, and relatively high proportions of rhamnose. The exudate from B. glaucescens is of particular interest in having high molecular weight, high intrinsic viscosity, and a high methoxyl content. The nitrogen content of all three gums is low, but amino acid analysis shows that proteinaceous components are involved, as in the gum exudates from other genera.  相似文献

Twelve genes coding for assembly, acetylation, pyruvylation, polymerization, and secretion of the polysaccharide xanthan gum are clustered together on the chromosome of the bacterium Xanthomonas campestris. These genes (gumBCDEFGHIJKLM) are sufficient for synthesis of xanthan gum when placed in bacteria from a different genus, Sphingomonas. The polysaccharide from the recombinant microorganism is largely indistinguishable, structurally and functionally, from native xanthan gum. These results demonstrate that a complex pathway for biosynthesis of a specific polysaccharide can be acquired by a single inter-generic transfer of genes between bacteria. This suggests the biological and commercial feasibility of synthesizing xanthan gum or other polysaccharides in non-native hosts. Novel guar gum-g-poly(sodium acrylate)/rectorite (GG-g-PNaA/REC) superabsorbent nanocomposites were prepared in aqueous solution using guar gum (GG), partially neutralized acrylic acid (NaA), acidified rectorite (H+-REC) and organified rectorite (CTA+-REC) by cetyltrimethylammonium bromide (CTAB) as raw materials, ammonium persulfate (APS) as initiator and N,N′-methylenebisacrylamide (MBA) as crosslinker. FTIR spectra confirmed that NaA had been grafted onto GG chains and the OH groups of REC participated in polymerization reaction. Exfoliated nanocomposite was obtained for H+-REC and intercalated structure was formed for CTA+-REC as shown by XRD results. SEM observations show REC has been uniformly dispersed in polymeric matrix. Effects of HCl concentration, organification degree of CTA+-REC and content of REC on swelling capabilities were investigated and the swelling kinetics of nanocomposites was evaluated. Results indicate that modifying REC by acidification and organification can improve swelling properties of the resultant nanocomposites, and GG-g-PNaA/CTA+-REC exhibited higher swelling capability and swelling rate contrast to GG-g-PNaA/H+-REC.  相似文献

Benzo[a]pyrene is a polycyclic aromatic hydrocarbon (PAH) associated with potent carcinogenic activity. Mutagenesis induced by benzo[a]pyrene DNA adducts is believed to involve error-prone translesion synthesis opposite the lesion. However, the DNA polymerase involved in this process has not been clearly defined in eukaryotes. Here, we provide biochemical evidence suggesting a role for DNA polymerase η (Polη) in mutagenesis induced by benzo[a]pyrene DNA adducts in cells. Purified human Polη predominantly inserted an A opposite a template (+)- and (−)-trans-anti-BPDE-N2-dG, two important DNA adducts of benzo[a]pyrene. Both lesions also dramatically elevated G and T mis-insertion error rates of human Polη. Error-prone nucleotide insertion by human Polη was more efficient opposite the (+)-trans-anti-BPDE-N2-dG adduct than opposite the (−)-trans-anti-BPDE-N2-dG. However, translesion synthesis by human Polη largely stopped opposite the lesion and at one nucleotide downstream of the lesion (+1 extension). The limited extension synthesis of human Polη from opposite the lesion was strongly affected by the stereochemistry of the trans-anti-BPDE-N2-dG adducts, the nucleotide opposite the lesion, and the sequence context 5′ to the lesion. By combining the nucleotide insertion activity of human Polη and the extension synthesis activity of human Polκ, effective error-prone lesion bypass was achieved in vitro in response to the (+)- and (−)-trans-anti-BPDE-N2-dG DNA adducts.  相似文献

This study examined the effects of different gums viz. gum arabic (GA), guar gum (GG), k-carrageenan gum (KG), and xanthan gum (XG) on rheological and 3D printing characteristics of vitamin D (Vit D) enriched orange concentrate (OC) wheat starch (WS) blends. The textural and microstructural properties of printed objects from above mixture were evaluated and compared. The addition of gums induced an increase in apparent viscosity, storage modulus (G′), and loss modulus (G″) of the OC-WS mixtures, while GA decreased the apparent viscosity and G′. Nuclear magnetic resonance (NMR) analysis of 3D printed samples revealed that the movement of transverse time (T2) toward closer to 0 ms indicated an increase in immobilized and bound water populations suggesting the gel formation. The slight shift toward shorter wavelength in FT-IR results for the broadband centered around 3400 cm?1 after addition of gums possibly caused an increase of G′ and load bearing capacity of the blends. 3D printing characteristics revealed that the objects printed using KG containing blend possessed maximum fidelity to the target geometry and good loading bearing capacity, preventing collapsing over time due to the proper G′ value. At tanδ of 0.238, OC-WS-KG mixture achieved the best printing condition. Higher tanδ of GA (0.038) containing samples led to an unwanted collapse of the printed constructs. The objects printed using KG also exhibited the smoothest visible surface as well as microstructure and best mastication properties. Considering the studied features, vitamin D enriched OC with WS-KG was found to be the best match for orange fruit concentrate-based 3D food printing. This work demonstrates the novel ways to develop fortified 3D printed foods.  相似文献

The gum polysaccharides from Terminalia sericea and T. superba have been analysed. They have a complex sugar composition, containing galacturonic, glucuronic, and 4-O-methylglucuronic acids as well as galactose, arabinose, rhamnose, mannose and xylose. The exudates from T. sericea and T. superba are remarkably similar in composition, particularly with respect to their proportions of neutral sugars and total uronic acid content, although T. sericea gum contains considerably more 4-O-methylglucuronic acid than T. superba. Both gums are very viscous and dissolve readily to give solutions of good colour.!!  相似文献

 The species richness of putative ectomycorrhizal (EM) fungi fruiting in blue gum (Eucalyptus globulus Labill.) plantations in Western Australia was investigated in relation to plantation age. Eleven plantations, 1–8 years old, were selected for study and two native Eucalyptus forest sites in the same region were chosen for comparison. Sporocarps of 44 species of putative EM fungi were collected from the 13 sites. Of these, 30 species were found in blue gum plantations. The number of fungal species was highly positively correlated with plantation age and inversely correlated with soil pH. Young plantations (1–5 years) had 2–9 fungal species and were overwhelmingly dominated by species of Laccaria and Scleroderma. In older plantations (6–8 years), the relative abundance of sporocarps of each species within the fungal community decreased, accompanied by an increase in the number of fungal species (12–17 per site). A brief survey of the two native eucalypt forests in this region revealed a much higher number of fungal species than that observed in plantations. In plantations, species of Descolea, Laccaria, Pisolithus and Scleroderma typically fruited in young plantations. Species of epigeous fungi of the genera Boletus, Cortinarius, Hydnum, Inocybe, Lactarius, Paxillus, Russula and hypogeous fungi, including species of Descomyces, Hysterangium and Mesophellia, were found only in older plantations, or in native forests. Some of the fungi that fruit in young plantations are now being evaluated for use in commercial spore inoculation programs to increase the species diversity of EM fungi in exotic eucalypt plantations.
