Although disease-resistance polymorphisms are common in natural plant populations, the mechanisms responsible for this variation are not well understood. Theoretical models predict that balancing selection can maintain polymorphism within a population if the fitness effects of a resistance allele vary from a net cost to a net benefit, depending upon the extent of pathogen damage. However, there have been a few attempts to determine how commonly this mechanism operates in natural plant-pathogen interactions. Ipomoea purpurea populations are often polymorphic for resistance and susceptibility alleles at a locus that influences resistance to the fungal pathogen, Coleosporium ipomoeae. We measured the fitness effects of resistance over three consecutive years at natural and manipulated levels of damage to characterize the type of selection acting on this locus. Costs of resistance varied in magnitude from undetectable to 15.5%, whereas benefits of resistance sometimes equaled, but never exceeded, these costs. In the absence of net benefits of resistance at natural or elevated levels of disease, we conclude that selection within individual populations of I. purpurea probably does not account completely for maintenance of this polymorphism. Rather, the persistence of this polymorphism is probably best explained by a combination of variable selection and meta-population processes.  相似文献

Although fitness costs associated with plant defensive traits are widely expected, they are not universally detected, calling into question their generality. Here, we examine the potential for life‐history trade‐offs associated with herbicide resistance by examining seed germination, root growth, and above‐ground growth across 43 naturally occurring populations of Ipomoea purpurea that vary in their resistance to RoundUp ® , the most commonly used herbicide worldwide. We find evidence for life‐history trade‐offs associated with all three traits; highly resistant populations had lower germination, shorter roots, and smaller above‐ground size. A visual exploration of the data indicated that the type of trade‐off may differ among populations. Our results demonstrate that costs of adaptation may be present at stages other than simply the production of progeny in this agricultural weed. Additionally, the cumulative effect of costs at multiple life cycle stages can result in severe consequences to fitness when adapting to novel environments.  相似文献

We examined the genetic basis of resistance to the rust pathogen Coleosporium ipomoea in three host species: Ipomoea purpurea, I. hederacea, and I. coccinea (Convolvulaceae). In crosses between resistant and susceptible individuals, second-generation selfed offspring segregated in ratios that did not differ statistically from the 3:1 ratio indicative of single-gene resistance with the resistant allele dominant. One out of three crosses between resistant individuals from two different populations revealed that resistance loci differed in the two populations, as evidenced by the production of susceptible individuals among the S(2) generation. These results suggest that gene-for-gene interactions contribute substantially to the dynamics of coevolution in this natural pathosystem. They also suggest that evolution of resistance to the same pathogen strain may involve different loci in different Ipomoea populations.  相似文献

Because disease resistance is a hallmark signature of pathogen-mediated selection pressure on hosts, studies of resistance structure (the spatial distribution of disease resistance genes among conspecific host populations) can provide valuable insights into the influence of pathogens on host evolution and spatial variation in the magnitude of their effects. To date few studies of wild plant-pathogen interactions have characterized resistance structure by sampling across the host's biogeographic range, and only a handful have paired such investigations with studies of disease levels under natural conditions. I used a greenhouse cross-inoculation experiment to characterize genetic resistance of 16 populations of California dwarf flax (Hesperolinon californicum) to attack by multiple samples of the rust fungus Melampsora lini. I documented a latitudinal cline in resistance structure, manifest across the host's biogeographic range, which mirrored almost identically a cline in infection prevalence documented through field surveys of disease in study populations. These results provide empirical evidence for clinal patterns of antagonistic selection pressure, demonstrate that such patterns can be manifest across broad biogeographic scales, and suggest that rates of disease prevalence in wild plant populations may be tightly linked to the distribution of host resistance genes. Tests for local adaptation of the fungus revealed evidence of the phenomenon (significantly greater infection in sympatric plant-fungal pairings) as well as the potential for substantial bias to be introduced into statistical analyses by spatial patterns of host resistance structure.  相似文献

为鉴定引起广西种植区白及锈病的病原菌种类且筛选抗锈病的白及资源,该研究对白及锈病病原菌进行分离,并采用形态学和分子生物学的方法对病原菌进行鉴定,同时通过人工接种病原菌法对23份白及进行锈病抗性评价以及筛选抗锈病的白及资源。结果表明:(1)从白及感病叶片中分离的锈病病原菌X2夏孢子呈金黄色,卵圆形或椭圆形,大小为(21.43~30.95)μm ×(13.10~19.05)μm; 冬孢子呈橘红色、红褐色,倒卵形或棍棒状,大小为(17.25~30)μm ×(5.5~6.65)μm。(2)把菌株X2全长689 bp的ITS序列(OQ826009)与GenBank 已登陆的序列进行相似性分析发现,菌株X2与Coleosporium sp.(KY783686.1)匹配度最高,序列一致性为95.86%,但系统发育树表明,X2与Coleosporium bletiae(MN108161.1,OP363680.1)聚为一类群; 结合形态学和分子生物学的方法,鉴定菌株X2为Coleosporium bletiae。(3)人工接种菌株X2 14 d后,23份白及的病情指数范围在0~70.7之间,并根据病情指数将 23 份白及划分为 6 个抗性等级,即表现为免疫的白及1份,病情指数为0; 表现为高抗的白及4份,病情指数为1.7~4.7; 表现为抗病的白及6份,病情指数为5.6~9.4; 表现为中抗的白及5份,病情指数为12.7~18.3; 表现为感病的白及5份,病情指数为32.0~49.1; 表现为高感的白及2份,病情指数为62.2~70.7。综上认为,表现为免疫和高抗的5份白及(分别来自云南红河、广西恭城、广西百色、贵州遵义、湖北宜昌)病情指数低、抗锈病能力强,可推广应用或作为培育抗锈病优良种质的亲本材料。该研究结果为后续开展白及锈病的有效防治与致病机理研究提供了支撑。  相似文献

Theory predicts that hosts and pathogens will evolve higher resistance and aggressiveness in systems where populations are spatially connected than in situations in which populations are isolated and dispersal is more local. In a large cross‐inoculation experiment we surveyed patterns of host resistance and pathogen infectivity in anther‐smut diseased Viscaria alpina populations from three contrasting areas where populations range from continuous, through patchy but spatially connected to highly isolated demes. In agreement with theory, isolated populations of V. alpina were more susceptible on average than either patchily distributed or continuous populations. While increased dispersal in connected systems increases disease spread, it may also increase host gene flow and the potential for greater host resistance to evolve. In the Viscaria–Microbotryum system, pathogen infectivity mirrored patterns of host resistance with strains from the isolated populations being the least infective and strains from the more resistant continuous populations being the most infective on average, suggesting that high resistance selects for high infectivity. To our knowledge this study is the first to characterize the impacts of varying spatial connectivity on patterns of host resistance and pathogen infectivity in a natural system.  相似文献

1.  Plants are simultaneously attacked by multiple herbivores and pathogens. While some plant defences act synergistically, others trade-off against each other. Such trade-offs among resistances to herbivores and pathogens are usually explained by the costs of resistance, i.e. resource limitations compromising a plant's overall defence.
2.  Here, we demonstrate that trade-offs can also result from direct negative interactions among defensive traits. We studied cyanogenesis (release of HCN) of lima bean (Fabaceae: Phaseolus lunatus ) and effects of this efficient anti-herbivore defence on resistance to a fungal pathogen (Melanconiaceae: Colletotrichum gloeosporioides ).
3.  Leaf tissue destruction by fungal growth was significantly higher on high cyanogenic (HC) lima bean accessions than on low cyanogenic (LC) plants. The susceptibility of HC accessions to the fungal pathogen was strongly correlated to reduced activity of resistance-associated polyphenol oxidases (PPOs) in leaves of these plants. LC accessions, in contrast, showed high PPO activity, which was correlated with distinct resistance to C. gloeosporioides .
4.  Experimentally applied, gaseous HCN reduced PPO activity and significantly increased the size of lesions caused by C. gloeosporioides in LC leaves.
5.  Field observations of a wild lima bean population in Mexico revealed a higher infection rate of HC compared to LC plant individuals. The types of lesions observed on the different cyanogenic plants in nature were similar to those observed on HC and LC plants in the laboratory.
6.   Synthesis. We suggest that cyanogenesis of lima bean directly trades off with plant defence against fungal pathogens and that the causal mechanism is the inhibition of PPOs by HCN. Our findings provide a functional explanation for the observed phenomenon of the low resistance of HC lima beans in nature.  相似文献

Fitness costs of resistance are among the most widely discussed explanations for the evolution of induced resistance, but studies on induced resistance to pathogens are scarce and contradictory. In the present study the influence of nitrogen supply, length of the growing period and competition on the seed production of Arabidopsis in response to treatment with the chemical resistance elicitor BION® was investigated. BION® treatment elicited resistance to the bacterial pathogen Pseudomonas syringae, and biochemical changes after BION® treatment were similar to those observed after bacterial infection. Induced plants grew more slowly during the first week after resistance induction, for which they then compensated by exhibiting faster growth than controls. Whether or not induced plants produced less seeds than controls depended on growing conditions. Costs, no costs and even higher seed production by induced plants were observed in experiments differently combining abiotic conditions. A higher seed production by induced plants arose particularly when the vegetation period was short, most probably a consequence of senescence-related processes that had been activated by resistance elicitation. Induced plants, however, produced less seeds when competing with controls and experiencing a full growing period. Studies controlling only some of the critical environmental factors can easily lead to apparently contradictory results, which in fact represent different outcomes of a complex interplay of factors.  相似文献

Coinfection, whereby the same host is infected by more than one pathogen strain, may favor faster host exploitation rates as strains compete for the same limited resources. Hence, coinfection is expected to have major consequences for pathogen evolution, virulence, and epidemiology. Theory predicts genetic variation in host resistance and pathogen infectivity to play a key role in how coinfections are formed. The limited number of studies available has demonstrated coinfection to be a common phenomenon, but little is known about how coinfection varies in space, and what its determinants are. Our aim is to understand how variation in host resistance and pathogen infectivity and aggressiveness contribute to how coinfections are formed in the interaction between fungal pathogen Podosphaera plantaginis and Plantago lanceolata. Our phenotyping study reveals that more aggressive strains are more likely to form coinfections than less aggressive strains in the natural populations. In the natural populations most of the variation in coinfection is found at the individual plant level, and results from a common garden study confirm the prevalence of coinfection to vary significantly among host genotypes. These results show that genetic variation in both the host and pathogen populations are key determinants of coinfection in the wild.  相似文献

Staphylococcus aureus is a ubiquitous gram-positive bacterium that can cause superficial to serious systemic infections in animals and humans. Here we report the development of a plant infection model to study the pathogenesis of this bacterium. Three global regulatory mutants, RN6911 (agr-), ALC 488 (sarA-) ALC 842 (sarA-/agr-) and an alpha-toxin mutant defective in biofilm formation (DU1090) which are attenuated in animal pathogenesis, were also attenuated in their ability to infect plants, suggesting that these regulators that mediate synthesis of virulence factors essential for animal pathogenesis are also required for plant pathogenesis. Further, using Arabidopsis plants altered in defense responses such as the transgenic lines NahG [defective in salicylic acid (SA) accumulation], and 35S-LOX2- (defective in jasmonic acid production and hyper-accumulator of SA), and mutants ics1 (depleted in SA accumulation), and npr1-1 (non-expressor of pathogenesis-related protein) we show that resistance of Arabidopsis to typical plant pathogens and the animal pathogen S. aureus is conserved and is mediated by SA. The data presented here suggest that Arabidopsis thaliana resistance to S. aureus is mediated either by a direct effect of SA on the pathogen, specifically one that affects the attachment/aggregate formation on the root surface and reduces the pathogen's virulence, or by SA-dependent, NPR1-independent host responses.  相似文献

Parasites are ubiquitous features of living systems and many parasites severely reduce the fecundity or longevity of their hosts. This parasite‐imposed selection on host populations should strongly favor the evolution of host resistance, but hosts typically face a trade‐off between investment in reproductive fitness and investment in defense against parasites. The magnitude of such a trade‐off is likely to be context‐dependent, and accordingly costs that are key in shaping evolution in nature may not be easily observable in an artificial environment. We set out to assess the costs of phage resistance for a plant pathogenic bacterium in its natural plant host versus in a nutrient‐rich, artificial medium. We demonstrate that mutants of Pseudomonas syringae that have evolved resistance via a single mutational step pay a substantial cost for this resistance when grown on their tomato plant hosts, but do not realize any measurable growth rate costs in nutrient‐rich media. This work demonstrates that resistance to phage can significantly alter bacterial growth within plant hosts, and therefore that phage‐mediated selection in nature is likely to be an important component of bacterial pathogenicity.  相似文献

Resistance genes (R genes) in plants mediate a highly specific response to microbial pathogens, often culminating in localized cell death. Such resistance is generally pathogen race specific and believed to be the result of evolutionary selection pressure. Where a host and pathogen do not share an evolutionary history, specific resistance is expected to be absent or rare. Puccinia psidii, the causal agent of myrtle rust, was recently introduced to Australia, a continent rich in myrtaceous taxa. Responses within species to this new pathogen range from full susceptibility to resistance. Using the myrtle rust case study, we examine models to account for the presence of resistance to new encounter pathogens, such as the retention of ancient R genes through prolonged ‘trench warfare’, pairing of resistance gene products and the guarding of host integrity.  相似文献

The process of cellularization of the endosperm was studied in Ipomoea purpurea Roth and Cytinus hypocistis L. In both the examined species normal cytochinesis, involving the formation of the phragmoplast, characterizes the change from the nuclear to cellular condition. Nevertheless, some ultrastructural aspects of the cell wall initiation seem to indicate that the modality of freely-growing walls cannot be excluded. The hypothesis of a unique method of wall initiation for the endosperm of the nuclear type, formulated by some Authors, is discussed.  相似文献

Fitness costs are frequently invoked to explain the presence of genetic variation underlying plant defense across many types of damaging agents. Despite the expectation that costs of resistance are prevalent, however, they have been difficult to detect in nature. To examine the potential that resistance confers a fitness cost, we examined the survival and fitness of genetic lines of the common morning glory, Ipomoea purpurea, that diverged in the level of resistance to the herbicide glyphosate. We planted a large field experiment and assessed survival following herbicide application as well as fitness of the divergent selection lines in the absence of the herbicide. We found that genetic lines selected for increased resistance exhibited lower death compared to control and susceptible lines in the presence of the herbicide, but no evidence that resistant lines produced fewer seeds in the absence of herbicide. However, susceptible lines produced more viable seeds than resistant or control lines, providing some evidence of a fitness cost in terms of seed germination, and thus potential empirical support for the expectation of trait trade‐offs as a consequence of adaptation to novel environments.  相似文献

 The inheritance of resistance to southern rust (caused by Puccinia polysora Underw.) was investigated in two F2:3 populations derived from crossing two temperate-adapted, 100% tropical maize (Zea mays L.) inbred lines (1416-1 and 1497-2) to a susceptible Corn Belt Dent hybrid, B73Ht×Mo17Ht. The inbred lines possess high levels of resistance to southern rust and may be unique sources of resistance genes. Heritability for resistance was estimated as 30% and 50% in the two populations from regression of F2:3 family mean scores on F2 parent scores, and as 65% and 75% from variances among F2:3 families on a single-plot basis. RFLP loci on three chromosomal regions previously known to possess genes for resistance to either southern rust or common rust (P. sorghi Schw.) were used to localize genes affecting resistance to southern rust in selected genotypes of both populations, and to estimate their genetic effects. A single locus on 10S, bnl3.04, was associated with 82–83% of the variation among field resistance scores of selected F2:3 families in the two populations. Loci on chromosomes 3 (umc26) and 4 (umc31) were significantly associated with resistance in the 1497-2 population, each accounting for 13–15% of the phenotypic variation for F2:3 field scores. Multiple-marker locus models, including loci from chromosomes 3, 4, and 10 and their epistatic interactions, accounted for 96–99% of the variation in F2:3 field scores. Similar results were obtained for resistance measured by counting pustules on juvenile plants in the greenhouse. An attempt was made to determine if the major gene for resistance from 1416-1 was allelic to Rpp9, which is also located on 10S. Testcross families from the cross (1416-1×B37Rpp9)×B14AHt were evaluated for resistance to southern rust in Mexico. Neither source of resistance was completely effective in this environment, preventing determination of allelism of the two genes; however, both sources of resistance had better partial resistance to southern rust than did B14AHt.
