Three‐dimensional reconstructions of a hematopoietic organ (HPO) from Bombyx mori larva were undertaken using light and electron microscopy. Each compact islet varied in sizes, but in the central area of the HPO their size became smaller. Compact islets and loose islets were made up of prohemocytes, plasmatocytes, and reticular cells, but there were differences in the proportions of these cells. Within the cytoplasm of reticular cells and within their cell projection, vacuoles were observed. Cell proliferation occurs primarily in the compact islets, and differentiation associated with the reticular cells occurs primarily in the loose islets. It can be inferred that reticular cells have a significant influence on proliferation and differentiation associated with hematopoiesis. According to the results of the 3‐D reconstruction, one reticular cell is in contact with eight or nine hemocytes. Each reticular cell is presumably of approximately ten hemocytes. A brief review is presented of the current state of ultrastructure, cytochemistry, and physiology of the hemocytes and meso- and metathoracic peri-imaginal-wing organs in silkworms. According to the accepted morphological classification, five circulating types of hemocytes are recognized in Bombyx mori as well as in Antheraea pernyi. They are prophemocytes or stem cells, plasmatocytes or pre-differentiated cells and three specialized cells, granulocytes, spherule cells and oenocytoids. During post-embryonic development the last four types are the most common in the circulating hemolymph. Plasmatocytes are considered to be pluripotent cells from which granulocytes, spherule cells and oenocytoids are derived. Contrary to the situation in most insects the plasmatocytes are not phagocytic in Antheraea. The granulocytes are efficient phagocytes. Both plasmatocytes and granulocytes are involved in pinocytosis. Another possible function of the granulocytes is hemolymph coagulation. The function of the spherule cells which contain a paracrystalline material (muco- or glycoproteins) is by no means clear. The phenoloxidase activity found within the cytosol of oenocytoids appears effective against the natural monophenol and diphenol substrates. The involvement of oenocytoids in the complex metabolism of phenols and particularly in the production of plasma phenolases has been reported. The mitotic division of five circulating hemocyte types is well known and was long regarded as the only mechanism of postembryonic hemocyte production. We present for silkworms, experimental evidence of the hemocytopoietic function of the meso- and metathoracic organs surrounding the imaginal wing discs. Ablation experiments demonstrate that the mitotic activity of free hemocytes is unable to maintain the normal hemocytogram in the absence of the two paris of organs. These organs are typically divided into cell islets ensheathed by a connective tissue membrane. Two types of islets may be classified by the disposition of the cells : the compact islets or aggregations of stem cells and the reticulate islets which are mainly composed of hemocytes at different steps of differentiation. The relative number of prohemocytes in the total hemocyte population ranges from 84 to 97 p. cent in organs of Antheraea pernyi. This well-defined cell type appears to be the major hemocyte type in hemocytopoietic organs. In Antheraea, the mitotic index (the relative number of mitotic hemocytes in the total cell population) varies from 0.5 to about 3 p. cent. Finally, our data direct attention to cyclic functional changes such as mitotic divisions and hemocyte differentiation which run parallel to the molting cycle.  相似文献

The fine structure of the hemopoietic tissue and its detailed reticular organization in the mealworm beetle, T. molitor were examined using light and scanning electron microscopes. The major hemopoietic tissues in the abdomen were located on the upper surface of the dorsal diaphragm which continuous over the ventral wall of the heart. Histologic characteristics of this hemopoietic tissues are dense clusters of cells. They are irregular in outline and are not surrounded by any connective tissue sheath. The hemopoietic tissue of this insect is consisted of three cellular components which are the reticular cells, hemocytic stem cells and several kinds of mature hemocytes. The reticular cells had numerous cytoplasmic processes and forming a complex network. The stem cells give rise to differentiating hemocytes of the different cell lineages. Mature hemocytes within this hemopoietic tissue are originated from the stem cells and differentiated into several types of hemocytes including prohemocytes, plasmatocytes, and granulocytes.  相似文献

We followed the fate of microcultured Bombyx mori prohemocytes in vitro. Prohemocytes isolated from larval hemolymph (day 1 of 4th instar) were maintained for 4-11 days in serum-free MGM-450 medium and some of them underwent mitotic division. Over 60% of the non-dividing prohemocytes differentiated to plasmatocytes or granulocytes. Some of the granulocytes subsequently transformed to spherulocytes. Of the dividing prohemocytes, 59.2% of the daughter cells differentiated into other types of hemocytes such as plasmatocytes, granulocytes and spherulocytes, and the remainder divided into new prohemocytes. Four of these renewed prohemocytes generated daughter cells composed of plasmatocytes and granulocytes. These results suggest that prohemocytes possess the properties of stem cells, and that plasmatocytes and spherulocytes may be terminally differentiated cells, whereas granulocytes, at least in part, may be a transient form of spherulocyte. Oenocytoids were not produced, suggesting that the lineage of oenocytoids differs from that of other types of hemocytes and that it is determined before release from hemopoietic organs.  相似文献

Hemopoiesis in orthopteran insects occurs in a hemopoietic organ that is located bilaterally along the aorta. This organ is also known as a reticulo-hemopoietic organ because of the rich presence of reticular cells. This study was performed to further elucidate hemopoiesis in the reticulo-hemopoietic organ of an orthopteran, Euprepocnemis shirakii. We focused on the question why reticular cells are so abundant (35% of cells in hemopoietic organ). Interestingly, 21% of these reticular cells surrounded hemocytes with their reticular cytoplasm. The surrounded hemocytes were distinguished by their different size and darkly stained nucleus. These cells were characterized by immunostaining using antibodies against several types of hemocytes: 45% of the surrounded hemocytes were CD34+, and these positive cells were double stained (over 85%) when immunostained by another hemopoietic pluripotent cell marker, Sca-1. Transmission electron microscopic analysis showed that reticular cells surrounded hemocytes containing large nuclei and poorly developed cytoplasmic organelles. This strongly suggests that the reticular cells surround hemopoietic stem cells. Additionally, surrounded hemopoietic progenitor cells are undergoing apoptosis as indicated by the TUNEL assay. The enclosed apoptotic cells are engulfed and then phagocytosed by reticular cells. Our results suggest that reticular cells are related to the differentiation and apoptosis of hemopoietic stem cells.  相似文献

Cell based therapy for the treatment of type 1 diabetes is limited by the overall shortage of donor organs for transplantation. This is the rationale for the research on the generation of insulin-producing beta cells from an inexhaustible source of cells such as the stem cells. Stem cells are progenitor cells which possess the capacity of self-renewing and differentiation in fully mature cells depending on the culture conditions. The fundamental question is how to make terminally matured pancreatic beta cells. During the last years different approaches for the neogenesis of beta cells have been described using embryonic stem cells, adult stem cells residing in the pancreas, or other nonpancreatic cell types. Although fully functional islets have not yet been derived from any stem cells, the use of stem cells is still the most promising approach on the way to establish a treatment protocol for the cure of type 1 diabetes in the future.  相似文献

A neurotumor of a peripheral nerve origin, RT4, was induced by subcutaneous injection of a newborn BDIX strain rat with ethylnitrosourea. The tumor cells were adapted to cell culture and, after about 2 months, were found to consist of cells displaying four distinct morphologies (AC, B, D, and E). Clonal lines of the four cell types were established. Their morphological stability suggested that there was a stem cell type (AC) which differentiated into other types of cells (B, D, and E). Presumptive stem cell lines were established from single cells. In 10 of the 12 clonal stem cell lines, colonies of differentiated cells appeared in approximately 40 days. One stem cell culture was maintained for 55 days, and all three differentiated cell types resulted in the same culture. The differentiated cells are morphologically indistinguishable from the cells isolated from the original tumor. We are thus able to demonstrate a stem cell in the RT4 neurotumor apparently capable of multipotential differentiation in vitro. We call this phenomenon cell type conversion. The stem cells (AC) and one type (D) of the differentiated cells produce a nervous system specific protein, S100 protein, while its production is arrested when the stem cells differentiate into two other cell types (B and E). No appreciable levels of neurotransmitter synthesizing enzymes (choline acetyltransferase, tyrosine hydroxylase, and glutamic acid decarboxylase) are detected in any cells. The present study shows that hemocytic granular cells synthesize and secrete type IV collagen (ColIV) in the silkworm Bombyx mori (B. mori) and suggests that these cells play roles in the formation of basement membrane, the encapsulation of foreign bodies, and the metamorphic remodeling of the gut. The full- and partial-length cDNA of B. mori prolyl 4-hydroxylase alpha subunit (BmP4Halpha) and B. mori ColIV (BmColIV) were cloned, respectively. In situ hybridization and immunocytochemistry on larval tissues and cells identified hemocytic granular cells as the cells that express mRNAs and proteins of both BmP4Halpha and BmColIV. Immunohistochemistry and immunocytochemistry demonstrated that BmColIV was present in the basement membrane and in the secretory granules of granular cells, respectively. Granular cells in culture secreted BmColIV without accompanying the degranulation and discharged it from the granules when the cells were degranulated. Nylon threads were inserted into the hemocoel of larvae. Granular cells concentrated around the nylon threads and encapsulated them as a self-defense reaction. BmColIV was found to be a component of the capsules. Furthermore, the present study showed that actively BmColIV-expressing granular cells accumulated around the midgut epithelium and formed BmColIV-rich thick basal lamina-like structures there in larval to pupal metamorphosis.  相似文献

Crustaceans possess blood cells (hemocytes) that mediate organismal defense and are analogous to vertebrate leukocytes. In order to more fully characterize these types of cells, hemocytes of the branchiopod crustacean, Artemia franciscana, were analyzed. The data indicate that Artemia have one type of hemocyte, ranging in morphology from compact and spherical to flat and spreading when examined in vitro. Electron microscopy revealed many cytoplasmic granules in the hemocytes and only a limited number of other membrane-bound organelles. Centrioles and microtubules were also visible in thin sections of chemically fixed samples. The cytoplasm of spherical hemocytes was completely labeled by general antitubulin antibodies, but in flattened hemocytes packing of cytoskeletal elements was less tight and individual microtubules were observed. Probing of Western blots disclosed acetylated, tyrosinated, and detyrosinated tubulin isoforms in hemocyte homogenates, the first characterization of posttranslationally modified tubulins in this cell type. Acetylated tubulin was restricted to a subset of microtubules, whereas tyrosinated microtubules were displayed more abundantly. Staining obtained with antibody to detyrosinated tubulin was unusual because it was limited to the perinuclear region of hemocytes. Incubation of blood cells with a monoclonal antibody to gamma-tubulin yielded fluorescent dots sometimes in pairs, a pattern characteristic of centrosomes. The findings support the conclusion that Artemia hemocytes undergo rapid morphogenesis in vitro accompanied by extensive rearrangement of their microtubules, the latter probably indicative of cytoskeletal changes that occur during cell movement and phagocytosis. Additionally, the hemocytes contain posttranslationally modified alpha-tubulins and centrosome-associated gamma-tubulin, both with the potential to influence microtubule organization and function.  相似文献

A cytological observation, using conventional fixing and staining, is made on the hemopoietic tissue in the crab, Carcinus maenas. The hemopoietic organ is formed by nodules grouping different cell types; nodules are surrounded by a limiting layer including collagenous filaments and material looking like basal lamina. Some fibrocytes and semi-granulous hemocytes are lining this limited layer. These hemocytes, more or less flattened, are transforming in fibrocytes. Fibroblast-like cells, with well developed intercellular junctions, are the first cell type: their dedifferentiation gives rise to isolated mitoting cells. We have named these mitoting cells "hemocytoblast". They are stem cells for hyaline hemocytes. Fibroblast-like cells can be compared with "reticular cells" in Insects. Uncertainty exists as to the formation and evolution of nodules.  相似文献

To understand the inhibitory effects of high temperature on insect growth at the cellular level, we investigated the influence of high temperature on the proliferation and division of larval hemocytes in the silkworm, Bombyx mori. Although the total number of hemocytes in the larval body increased enormously over time at 26 degrees C, no increase was observed at 38 degrees C. The number of mitotic hemocytes in circulation increased between days 1 and 2 of the fourth larval stage at 26 degrees C, whereas fewer hemocytes were observed at 38 degrees C. Laser scanning cytometry revealed that the DNA content of hemocytes collected from the fourth-stadium larvae was predominantly 2C, 4C, and 8C, and the proportion of each type of hemocyte changed dynamically with development during the fourth instar. Specifically, the proportion of hemocytes with a higher DNA content increased gradually during the feeding phase then decreased during the molting phase at 26 degrees C; in contrast, no decrease was observed at 38 degrees C. The heat-induced accumulation of 8C hemocytes was mainly detected in granulocytes and plasmatocytes. These data suggest that high temperatures induce a G(2) arrest in larval hemocytes.  相似文献

Expression of prophenoloxidase mRNA during silkworm hemocyte development   总被引:3,自引:0,他引:3  
Two clones encoding different prophenoloxidase isoforms were amplified by polymerase chain reaction of RNA from the hemocytes of an experimental strain of Bombyx mori. The nucleotide sequences of the clones and the deduced amino acid sequences were confirmed to be nearly identical to those of the orthologous clones previously obtained from a commercial race of B. mori. Northern blot hybridization using these clones as probes demonstrated that the prophenoloxidase mRNA in the hemocytes is expressed in a stage-specific manner during the final larval instar and pupal stage, showing a peak one day before pupation in males and on the day of pupation in females. A sexual difference was also observed when the content of prophenoloxidase protein in the hemolymph (including hemocytes) was measured by an enzyme-linked immunosorbent assay.  相似文献

Spain ranks number one in organ donors (35 per million per yr). Although the prevalence of diabetes is low (100,000 type 1 diabetic patients and 2 million type 2 diabetic patients), the expected number of patients receiving islet transplants should be estimated at 200 per year. Islet replacement represents a promising cure for diabetes and has been successfully applied in a limited number of type 1 diabetic patients, resulting in insulin independence for periods longer than 3 yr. However, it has been difficult to obtain sufficient numbers of islets from cadaveric donors. Interesting alternatives include acquiring renewable sources of cells using either embryonic or adult stem cells to overcome the islet scarcity problem. Stem cells are capable of extensive proliferation rates and are capable of differentiating into other cell types of the body. In particular, totipotent stem cells are capable of differentiating into all cell types in the body, whereas pluripotent stem cells are limited to the development of a certain number of differentiated cell types. Insulin-producing cells have been obtained from both embryonic and adult stem cells using several approaches. In animal models of diabetes, the therapeutic application of bioengineered insulin-secreting cells derived from stem cells has delivered promising results. This review will summarize the different approaches that have been used to obtain insulin-producing cells from embryonic and adult stem cells and highlights the key points that will allow in vitro differentiation and subsequent transplantation in the future.  相似文献

Type 1 diabetes is a debilitating condition, affecting millions worldwide, that is characterized by the autoimmune destruction of insulin-producing pancreatic islets of Langerhans. Although exogenous insulin administration has traditionally been the mode of treatment for this disease, recent advancements in the transplantation of donor-derived insulin-producing cells have provided new hope for a cure. However, in order for islet transplantation to become a widely used technique, an alternative source of cells must be identified to supplement the limited supply currently available from cadaveric donor organs. Stem cells represent a promising solution to this problem, and current research is being aimed at the creation of islet-endocrine tissue from these undifferentiated cells. This review presents a summary of the research to date involving stem cells and cell replacement therapy for type 1 diabetes. The potential for the differentiation of embryonic stem (ES) cells to islet phenotype is discussed, as well as the possibility of identifying and exploiting a pancreatic progenitor/stem cell from the adult pancreas. The possibility of creating new islets from adult stem cells derived from other tissues, or directly form other terminally differentiated cell types is also addressed. Finally, a model for the isolation and maturation of islets from the neonatal porcine pancreas is discussed as evidence for the existence of an islet precursor cell in the pancreas.  相似文献

目的:通过观察家蚕Bombyx mori吞噬细胞的微细结构,来确定拟绛色细胞是否也具有吞噬功能。方法:用荧光小球微量注射家蚕pnd pS品系的幼虫,经荧光染色剂丫啶橙和碘化丙啶染色循环血细胞后,在荧光显微镜下观察并扫描拍摄。结果:观察发现除颗粒细胞和浆血细胞外,一些原血球细胞(血干细胞)和拟绛色细胞(多酚氧化酶)也能吞噬荧光小球。在拟绛色细胞里还发现许多和颗粒细胞一样的能被丫啶橙染色的颗粒。尽管在小球细胞中没有发现被吞噬的荧光小球,但该类血球有比较多的能被丫啶橙染色的大颗粒,这表明它们可能是已经被吞噬的凋亡小体。结论:除颗粒细胞和浆血细胞外,一些原血球细胞和拟绛色细胞也能吞噬荧光小球。说明拟绛色细胞也具有吞噬功能。  相似文献

Electron microscopical studies indicate that midgut alterations observed in larval Amphimallon majalis during Bacillus popilliae invasion were due to infection of epithelium, wound repair, and hemocytic encapsulation. Hemocytes bearing angular cytoplasmic granules formed the capsule which adhered to the basal lamina subjacent to the mesenteronic lesion. Exocytosis of hemocyte granules into bacilli-containing endocytic invaginations of plasma membranes and vacuoles was observed. These hemocytes as well as mesenteronic cells exhibited bactericidal capabilities.  相似文献
