 The eyes of different larval stages of Carinaria lamarcki were examined ultrastructurally. In all larval stages the eyes consist of a cornea, a lens and an everse retina. The photoreceptors in young larvae are exclusively of the ciliary type. In old larvae, however, two types of photoreceptors are present and the retina is composed of two segments: a posterior segment with altered ciliary photoreceptors (=type I sensory cells) and an anterior segment with what are presumably rhabdomeric photoreceptors (=type II sensory cells). The anterior retina is interpreted here as an accelerted character. Furthermore, the arrangement of the pigment granules changes during the long larval development being cup shaped in young larvae versus ribbon shaped in old larvae. The findings allow for the conclusions that: (a) the ciliary photoreceptors are correlated with the long larval period of Heteropoda and that (b) the eyes are altered continuously during the larval cycle. Visceral larva migrans: migratory pattern of Toxocara canis in pigs.   总被引:4,自引:0,他引:4  
The migratory pattern of Toxocara canis was investigated following infection of pigs with 60000 infective eggs. Groups of six pigs were slaughtered at 7, 14 and 28 days after infection (p.i.), and the number of larvae in selected organs and muscles was determined by digestion. A group of uninfected pigs was used as negative controls for blood parameters and weight gain. Toxocara canis migrated well in the pig, although the relative numbers of larvae recovered decreased significantly during the experiment. On day 7 p.i., high numbers of larvae were recovered from the lymph nodes around the small intestine and to some extent also from the lymph nodes around the large intestine, and from the lungs and the liver. On day 14, the majority of larvae were recovered from the lungs and the lymph nodes around the small intestine, and by day 28 p.i. most larvae were found in the lungs. Larvae were recovered from the brain on days 14 and 21, with a maximum on day 14 p.i. No larvae were found in the eyes. Severe pathological changes were observed in the liver and lungs, especially on day 14 p.i.; also, development of granulomas was observed in the kidneys. Finally, a strong specific antibody response towards T. canis L2/L3 ES products was observed from day 14 p.i. until termination of the experiment, and the maximum eosinophil response was observed 14 days p.i. The pig is a useful non-primate model for human visceral larva migrans, since T. canis migrate well and induce a strong immunological response in the pig. However, the importance of the pig as a paratenic host is probably minor, because of the relatively early death of most of the larvae.  相似文献

The distribution of Toxocara canis larvae in the eye was determined for mice given a single challenge dose (C-mice) as compared to mice similarily challenged after 2 or 3 previous infections (SC-mice). Controls were mice given only the 2 or 3 previous infections and uninfected mice. Eyes were observed in situ during a 34 day post-challenge period to compare inflammatory responses in the anterior eye; histologic examination of serial sections of the eyes of these mice was done at the end of this period. In situ observations showed that lesions in the anterior eye converted from hemorrhagic to white cell more rapidly in the SC-mice; white cell lesions were predominant in SC-mice as early as day 5, whereas similar predominance in C-mice was not noted until days 16–21. Histology revealed that approximately 90% of these eyes were infected. Worm burdens per eye correlated more closely with total dose per mouse than with the effects of immunization. Histologic study showed that 90% of larvae observed were in the retina, but that most lesions were in the uveal tissues which harbored only 0·8% of the total number of larvae.  相似文献

The cerebrally innervated eyes of metamorphically competent larvae, newly metamorphosed larvae, and adults ofAporrhais pespelecani are ultrastructurally investigated and compared. The eyes are composed of a lens, a cornea, and an everse retina. In adults, a humour is located behind the lens. The retina consists of two different types of cells: sensory cells and supportive cells. The present study confirms earlier results and demonstrates that the distal part of the sensory cells is altered during ontogenesis. In metamorphically competent larvae, the sensory cells are exclusively ciliary. In newly metamorphosed larvae and in adults, however, the sensory cells are of the mixed type, bearing both cilia and microvilli. Furthermore, the findings confirm that both the supportive and corneal cells, as well as the distal supportive cell processes which are restricted to the eyes of adults are involved in lens formation.  相似文献

 The eyes of different larval stages and juveniles of Atlanta peroni are generally composed of a cornea, a lens and a retina. In juveniles a distinct pigmented shield is visible and an enormous humour is located behind the lens. Larvae have only two sensory cells and the photoreceptors are of the ciliary type. In juveniles a striking feature is the shape of the retina. It is ribbon-shaped and new sensory cells are present which are arranged in three rows. The photoreceptors are of the ciliary type as well. Contrary to the arrangement in larvae, the ciliary plasmalemma in juveniles forms numerous lamellar stacks. In accordance with the sensory cells the stacks are organized in three parallel rows. The lamellae of adjacent stacks within a row overlap each other. The latter unique feature has not yet been found in any other representative of the Heteropoda. These findings demonstrate that (a) the eyes are altered during the development from larvae into juveniles, (b) the larval sensory cells are reduced and replaced by new sensory cells in juveniles and (c) the eyes of juvenile and adult A. peroni are well adapted for their life as visual predators. Although Toxocara canis, an important pathogen of ocular disease, tends to migrate to the eye, the precise migratory route has yet to be determined experimentally. Mongolian gerbils, Meriones unguiculatus, known as a useful animal model for human toxocariasis, were used to investigate the migration route toward the eyes. Infective larvae of T. canis were directly inoculated into the intracranial region. Haemorrhagic lesions or larvae were observed in 56.3% of cases. Histopathologically, a larva was observed in the optic nerve of gerbils 6 days after inoculation, and two larvae were found in the optic chiasma in the gerbils having a haemorrhage in the retina 9 days after inoculation. These results indicate that T. canis migrates from the brain to the eye through the optic nerve. Considering these data and previous studies showing that the ocular changes appear as early as 3 days of infection in the oral-administrated gerbils, there are two phases in the migration to the retina: a haematogenous early phase and an optic nerve route late phase.  相似文献

Alternative migration routes of Ascaris suum in the pig   总被引:1,自引:0,他引:1  
Experiments were conducted to investigate possible alternative routes of extraintestinal migration of Ascaris suum larvae in the pig. Pigs were infected with A. suum via injection of newly hatched larvae into cecal veins (i.v.), into cecal lymph nodes (LN), or intraperitoneally (i.p.), and control animals were inoculated orally with infective eggs (p.o.). Two pigs per inoculation route were necropsied on days 1, 4, and 13 postinoculation. The numbers of liver lesions and the percentage of larvae recovered was considerably greater in pigs inoculated i.v. or p.o. on each necropsy day. However, irrespective of inoculation route, at least a proportion of larvae passed through the livers and were able to complete migration to the small intestine by day 13. The results indicate that larval penetration of the intestinal wall is not necessary for liver-lung migration and that passage through the liver may be favorable for migrating A. suum larvae, although a delayed arrival in the small intestine cannot be ruled out for larvae following alternative routes.  相似文献

Hepatic lesions in experimental Campylobacter jejuni infection of mice   总被引:2,自引:0,他引:2  
Mice orally infected with Campylobacter jejuni developed focal infiltrative necrotic lesions in the liver, as determined by both histology and liver function tests. The initial histopathological feature was a focal infiltrative lesion in the parenchyma and portal triads. Foci of infiltrative lesions became necrotic between days 30 and 60 post-inoculation (p.i.). During this period, portal infiltrates increased in severity. From month 4 p.i., focal areas of infiltrative necrosis in the liver parenchyma became extensive. Study of liver function demonstrated mild elevations of transaminases, alkaline phosphatase and lactic dehydrogenase, and also the presence of hypoalbuminaemia. Although histopathological changes of the liver became gradually more marked after day 30 p.i., liver functions of infected mice were most affected at 2 months p.i. The capacity of C. jejuni to induce hepatic lesions seemed to be related to that of organisms to persist in the gall bladder; there was no correlation between biliary carriage in infected mice and positive faecal culture.  相似文献

Eleven male two-month-old Holstein calves were used to determine the pathological changes induced by a Cooperia punctata infection. After weaning, ten calves received a single oral dose of 45,000 C. punctata infective larvae. One calf remained as a non-infected control. Groups of two calves were killed on days 7, 14, 21, 28 and 35 post-infection (p.i.) for determination of worm burdens and histopathological evaluation. The small intestine was sub-divided into three sections of approximately equal length, and representative samples of mucosa were fixed in 10% formalin, cut, and stained with haematoxylin-eosin. Samples of intestinal contents and mucosal digests were taken and fixed in 10% formalin for an estimation of total worm burdens. An increase in the number of adult parasites and a decrease in the number of larvae were observed with time (P<0.001). A higher concentration of worms was found in the first segment of the small intestine during the five weeks of observation. Histology showed larvae in the intestinal mucosa on day 7 p.i., with a discrete increase in the cellular response. Adult worms and a marked cellular infiltrate with eosinophils and neutrophils were present on day 21 p.i., and these persisted until day 35 p.i. Microcysts resulting from worm destruction were observed from day 21 p.i.  相似文献

African catfish Clarias gariepinus hatched with morphologically immature features; however, sensory organs developed rapidly with fish growth. Although the eyes of newly hatched larvae were immature without pigment, in 2 day‐old larvae, the retina of the eyes had already developed except for the rod cells. No free neuromasts were observed in newly hatched larvae. In 1 day‐old larvae, however, free neuromasts were observed on the head and trunk. Free neuromasts increased with larval growth. Newly hatched larvae had simple round‐shaped otic vesicles; however, all sensory epithelia of the inner ear were observed until the larvae were 3 days old. Two day‐old larvae swam horizontally, had sharp teeth, commenced ingesting rotifers and also artificial feed (small‐size pellets) under both light and dark conditions; by then the larvae already had many taste buds. Three day‐old larvae showed negative phototaxis and cannibalism by eating their conspecifics. Most of the free neuromasts observed in this study had the peculiar feature of many microvilli around the sensory cells on the apical surface. Detected free neuromasts as ordinary type lateral‐line organs were not observed in previous reports in teleosts. In 10 day‐old larvae, there were two lines of free neuromasts on the flank and lower edge of the trunk; presumptive canal neuromasts were oval shaped and had begun to sink under the skin. The direction of maximum sensitivity of the neuromasts was parallel with the longitudinal axis of their elliptical apical surface.  相似文献

The chronology of the life cycle of Trichostrongylus retortaeformis (Zeder, 1800) (Nematoda, Trichostrongyloidea) is studied in its natural host Oryctolagus cuniculus. The free living period lasted 5 days at 24 degrees C. Worm-free rabbits were each infected per os with T. retortaeformis larvae. Rabbits were killed at 12 h post-infection (p.i.) and every day from one day to 13 days p.i. By 12 h p.i., all the larvae were exsheathed and in the small intestine. The third moult occurred between 3 and 5 days p.i. and the last moult between 4 and 7 days p.i. The prepatent period lasted 12 to 13 days. The patent period lasted five and a half months. The four known life cycles of species of Trichostrongylus in ruminants were compared with that of T. retortaeformis. No significant difference was found except in the duration of the prepatent period. These similarities in the life cycles confirm the previously formulated hypotheses on the relationship between the parasites of the two host groups (Durette-Desset, 1985).  相似文献

Enterohepatitis was produced in Mongolian gerbils by intragastric inoculation with Tyzzer's organism from natural infection of a gerbil. Death occurred in 50 to 60% animals 5 to 7 days postinoculation (p.i.). On day 3 p.i., when a few necrotic foci appeared in the liver, a large amount of bacterial antigen was present within ileocecal enterocytes and reticuloendothelial cells of the Peyer's patches. Neutrophil and monocyte infiltration was found in the lamina propria. On day 5 or 6 p.i. there was severe necrotizing and hemorrhagic ileotyphlocolitis. Bacterial antigen was abundant within not only enterocytes, but also smooth muscle cells of the ileum and jejunum as well as reticular cells of the mesenteric lymph nodes. On day 7 p.i. the intestinal lesions subsided in the presence of fewer bacteria, while necrotizing hepatitis became well developed. The results indicated that Mongolian gerbils were highly susceptible to the oral route of infection with the Tyzzer's organism.  相似文献

Ontogenetic change in the visual acuity of Seriola lalandi larvae was measured using both behavioural and anatomical techniques. Visual acuity improved over early development (day 4 to day 7 post-hatch), although for all three larval ages examined estimates of anatomical acuity were consistently lower (higher acuity) than estimates of behavioural acuity. At hatching the eyes of larval kingfish were characterized by an undifferentiated retina surrounding a spherical lens, by day 4 post-hatch the eyes appeared to be functional, the retina was fully pigmented and the optic nerve had made contact with the optic tectum. Analysis of prey search behaviour indicated that larvae employ a saltatory type search behaviour in which brief stationary periods are interspersed with repositioning movements. The mean reactive angle increased between day 4 and day 7 post-hatch indicating that the horizontal visual field was expanding with development, thereby increasing the search area of larvae. Pre-strike distances of early larvae were substantially less than one body length, being constantly around a 1/3 of a body length for all larval ages examined.  相似文献

Toxoplasma gondii is a protozoan parasite that is globally widespread and infects man and animals. With the aim of studying the influence of toxoplasmosis on male reproductive parameters, we investigated sperm motility, concentration and morphology of male rats experimentally infected by T. gondii. The GT F1 strain of T. gondii tissue cysts were fed at a dose of 5 × 103 tissue cysts per rat by oral gavage in an experimental group of 42 healthy adult male Wistar rats, while 42 male rats were used as controls. On days 10, 20, 30, 40, 50 and 60 post-inoculation (p.i.) 7 rats from each group were anesthetized. The body weight of each animal was recorded, then epididymis and testes were immediately removed, weighed and semen evaluation was undertaken. Weight of the right epididymis was significantly decreased on day 30 p.i., sperm motility was significantly decreased on days 10, 20, 30, 40, 50 and 60 p.i. and sperm concentration was significantly decreased on days 10, 30, 40 and 60 p.i. A marked increase of sperm abnormalities was noticed on days 30 and 40 p.i. No pathological lesions were detected either in the pituitary gland or the testes. In this study it was found that toxoplasmosis can affect main reproductive parameters in male rats, which are the most predictive of their fertilizing capacity.  相似文献

Since the 1970s, cytomegalovirus (CMV) infection has been associated with atherosclerotic disease. However, the exact contribution of the virus remains uncertain. In this article we describe both a direct and indirect immune-mediated effect of the virus on the disease process. Eight-week-old apolipoprotein E (apoE) knockout mice were infected with mouse CMV (MCMV) or mock injected, and they were sacrificed at 2 and 20 weeks post-injection (p.i.) to study atherosclerosis, vascular wall IFNgamma and TNFalpha expression and MCMV spread. To study plasma IFNgamma and TNFalpha levels, blood was collected at 1, 2, 4 and 6 days p.i. in addition to days of sacrifice. Plasma cytokine levels were increased after MCMV infection at early time points and decreased to mock levels at 2 and 20 weeks p.i. At 2 weeks p.i., more aortic arch samples showed local cytokine expression after MCMV infection. The number of early atherosclerotic lesions and the percentage of mice containing early lesions were increased at 2 weeks p.i., while at 20 weeks p.i., the MCMV-induced effect on atherogenesis was seen on the late lesions. In conclusion, MCMV infection induces a systemic immune response reflecting an indirect effect of MCMV infection on atherosclerosis in addition to a local aortic immune response reflecting a direct effect of the virus on the atherosclerotic process.  相似文献

The immunological, haematological and enzymatic responses to the inoculation in pigs of 100,000 embryonated eggs of Toxocara canis were studied. Fifteen females were inoculated and three remained as controls. Haematological values were analysed from day 7 p.i. until day 126 p.i. In the inoculated group, white blood cells were raised on day 14 p.i. and eosinophil values on days 7, 14, 21, 35 and 49 p.i. showing significant differences compared with controls (P < 0.05). Absolute eosinophil counts (per ml) presented two rises, the first on days 7, 14 and 21 p.i. and the second on days 35 and 49 p.i. Blood biochemistry was maintained within normal values. Serological examination by ELISA to determine antibody levels against Toxocara canis L2/L3 excretory-secretory (ES) antigens showed values higher than the positive cut-off (1:32) from day 7 p.i. and until the end of the study on day 126 p.i., presenting two peaks: one on day 28 p.i. and the second covering days 49 to 56 p.i. Western blots of sera of inoculated animals presented, from day 7 p.i., two polypeptide bands of 55 and 70 kDa MW and, from day 56 p.i., an additional band of 120 kDa MW, all of which persisted until the end of the study. Immunological responses were sustained over time. No direct correlation was observed between the rise in eosinophils and antibody titres. To validate the conclusions, more studies are required on the polypeptide bands.  相似文献

Chick embryos were injected on the fifth day of incubation with 75 ng cis-diamminedichloroplatinum II (cisplatin) and killed at daily intervals. Bilateral microphthalmia appeared in 88% of the surviving embryos; the decrease in eye size was noticeable 2 or 3 days after injection. Coinciding with this, macroscopic, histological, and ultrastructural changes started to appear in the ciliary body: ciliary processes failed to form and the cells in the inner layer of the ciliary epithelium underwent degenerative changes. Changes in the retina appeared somewhat later. Despite the decreased growth rate of the whole eye the neural layer of the retina continued to grow rapidly; as a result, it formed numerous folds and acquired a glandular appearance. In the most severe cases the rapidly growing retina would invade the ciliary region and replace completely the degenerated inner layer of the ciliary epithelium. It has been shown by previous authors that intraocular pressure is a determinant of eye expansion and also that the secretion of water and ions by the ciliary epithelium is important for the maintenance of that intraocular pressure. On this basis, our results are interpreted as indicating that the primary lesion induced by cisplatin was in the ciliary epithelium and that microphthalmia was the consequence of decreased pressure. It is also concluded that the retinal changes were due to the fact that the retina continued to grow despite the lack of expansion of the eye as a whole.  相似文献

Nasal one-third sized fragments were created from fully differentiated larval Xenopus eyes (stage 47). At various times post-surgery, animals were injected with tritiated thymidine. All animals were fixed 1 day post-injection. Animals injected 1 day post-surgery showed limited healing and thymidine labeling in the retina. In animals injected 1 week post-surgery, heavy thymidine label was localized in the ventrotemporal retina in a "thickened" neuroepithelium internal to the extending pigmented retinal epithelium. In contrast, the dorsal retina showed no apparent extra labeling, but rather resembled normal ciliary margin. In animals injected 1 month post-surgery, the eye fragment regained normal size and retinal layering, and the label was restricted to the ciliary margin. Regenerative growth associated with healing appeared to have been completed by this time. This extra cell division that occurs during the 1st month post-surgery may underlie novel axonal targeting properties shown by nasal one-third sized fragments. For example, these findings are consistent with the idea that pattern duplication of the visuotectal projection in nasal one-third sized eye fragments occurs via intercalary cell division during healing and regeneration.  相似文献
