We examined the community-environment relationships of lotic macroinvertebrates in near-pristine headwater streams, and the correlation between patterns in taxonomic and functional structure at two regional extents. The across-ecoregion scale comprised five ecoregions spanning all of Finland, while the within-ecoregion scale comprised of north boreal and middle boreal ecoregions. We expected that taxonomic structure should exhibit stronger relationships than functional structure to spatial gradients, while the reverse should be true for local environmental factors. We found some support for this notion, because spatial variables were marginally more important for taxonomic than functional structure. Furthermore, within the two ecoregions, local environmental variables were slightly more important for functional than taxonomic structure. Geographical location (i.e. spatial variables) was more influential at the across-ecoregions extent than within the two ecoregions. Largely the same local environmental variables accounted for variation in both taxonomic and functional structure, including water pH, nutrients, colour, and stream size. Similar responses to local environmental features likely contributed to the significant correlation between patterns in taxonomic and functional structure. It has been suggested that functional traits may be rather insensitive to natural variation, yet our functional categorization showed distinct variation along local environmental, catchment, and spatial gradients across near-pristine streams. Thus, natural ecological filters should be accounted for prior to the examination of the effects of anthropogenic filters on stream macroinvertebrate community structure.  相似文献

We find that both Brevundimonas diminuta and Serratia marcescens can grow through sterilizing grade filter membranes of different membrane polymer compositions. Although this passage does not occur on a consistent basis, generation of “grow-through positive” results indicate that grow-through can occur stochastically at basal levels. This observation argues that the following risk mitigation strategies during pharmaceutical aseptic processing are warranted: minimization of processing times, and monitoring, minimizing and characterizing pre–filter bioburden.  相似文献

Eleven 16S rRNA gene clone libraries including 34 archaeal and 72 bacterial phylotypes were constructed from total 708 clones of hydrothermal vent prokaryotes trapped by 0.2- and 0.1-μm-pore-size filters. Crenarchaeota and Proteobacteria phylotypes dominated the archaeal and bacterial populations, respectively. Novel unaffiliated phylotypes occurred only in the 0.1-μm-trapped populations.  相似文献

The effect of temperature on the flow of diluted blood [Hct = 0.21], through 5 μm Nuclepore filters, is described by the Arrhenius equation with an energy of activation of 27.7 kJ/mol. Plasma, diluted with PBS, is almost three times less sensitive to temperature, with an energy of activation of 9.8 kJ/mol, while red cells are of intermediate sensitivity, with an energy of activation of 14.7 kJ/mol. The most sensitive elements to changes in temperature are leukocytes, with energies of activation of 31 kJ/mol and 35 kJ/mol for fast-flowing leukocytes (granulocytes and lymphocytes) and slow-flowing leukocytes (monocytes) respectively. Hence, the major determinants of the decline in filterability of blood through micropore filters are the leukocytes. This effect is compounded when blood is kept for 10 minutes or more at 10° C due to activation of granulocytes, which leads to permanent pore blocking when the affected blood is filtered at room temperature. The combination of increased passage time of leukocytes through peripheral areas at abnormally low temperatures and subsequent activation might influence the flow of blood in non-affected tissues. The effect of temperature on the filterability of red blood cells through 3 μm filters is not described by the Arrhenius equation and the deviations are seen as a gradual change of slope rather than a sharp break between two straight lines. The data are consistent with a gradual shift in rate limiting step away from the entry event into pores, which dominates at low temperature but becomes progressively less important at elevated temperatures. The changing parameter is probably the volume of the red cell, which is less important when flow is measured through 5 μm pores.  相似文献

Two identical biotrickling filters named BTFa and BTFb were run in parallel to examine their performances in removing hydrogen sulfide. BTFa was filled with ceramic granules, and BTFb was filled with volcanic rocks. The results showed that BTFb was more robust than BTFa under acidic conditions. At empty bed residence times (EBRTs) of 20 and 15?s, the removal efficiency of BTFa was close to 100%. At EBRTs of 10 and 5?s, the removal efficiency of BTFa slightly decreased. The removal efficiencies of BTFa decreased by different degrees at the end of each stage, dropping to 94%, 81%, 60%, and 71%, respectively. However, the H(2)S removal efficiency in BTFb consistently reached 99% throughout the experiment. Pyrosequencing analyses indicated that members of Thiomonas dominated in both BTFs, but the relative abundance of Acidithiobacillus was higher in BTFb than in BTFa.  相似文献

Besides its principal maximum, the spatial frequency characteristic curve of the complex visual cortical receptive field of curarized cats also has additional maxima and also negative regions, as predicted by the theory of piecewise Fourier analysis. Comparison of responses of the complex receptive field to sinusoidal gratings completely and incompletely contained in the field and comparison of responses to sinusoidal and square-wave gratings indicate that the receptive field, as a spatial frequency filter, has linear properties. The response of the complex receptive field rises with an increase in the number of periods of the sinusoidal grating. Several periods of optimal frequency match the complex field. Receptive fields tuned to a broad band of spatial frequencies were found in neuron columns. The response of chrysanthemum plants to gibberellic acid (GA3) and daminozide, grown under 6% CuSO4 and water (control) spectral filters, was evaluated to determine the involvement of gibberellins in regulation of plant height under CuSO4 filters. The CuSO4 filter increased the red (R)/farred (FR), and blue (B)/R ratio (R=600–700 nm; FR =700–800 nm; B=400–500 nm) of transmitted light. PPF under 6% CuSO4 filter was reduced by about 34% compared to PPF under water filter which averaged about 750 μM·m?2·s?1. Control plants were shaded with Saran Wrap to ensure equal PPF as in the CuSO4 chamber. GA3 application increased plant height under both the control and CuSO4 filter, but the height increase under the CuSO4 filter was about 20% greater than that under the control filter. Daminozide treatment reduced plant height under the control and CuSO4 filter, but the height reduction in control plants was slightly greater than under the CuSO4 filter. The height reduction caused by daminozide was prevented by GA3 application in plants grown under the control or CuSO4 filter. The results suggest that GA3 may be partially involved in height reduction under CuSO4 filters.  相似文献

For olfaction to occur, signal molecules must move through the environment from the source to the receptor cells. As molecules approach receptor structures they pass through a boundary layer surrounding those receptor structures. Within boundary layers the interaction between the forces causing chemical dispersion changes. To investigate how the boundary layer changes the dynamics of the chemical signals, we measured chemical dynamics within the boundary layer around the moth antennae using microelectrodes. The results showed that the boundary layer amplified three aspects of the chemical signal: peak height, peak onset, and decay time. Spectral analysis of turbulent signals showed that the temporal aspects of the chemical signal were altered. The boundary layers around the male and female antennae have different effects on the spectrum of chemical temporal fluctuations. Specifically, at a flow speed of 0.12 m s−1, the analysis showed distinct amplification patterns for each sex. Thus, the fluid flow around the antennae functions as a filter, altering the structure of the chemical signal that is arriving at the receptors. The results illustrated in this study show that male and female moths have different physical filters that can alter the information that can be extracted from odor plumes. The widely used partial synthesis of phospholipids via deacylation of naturally occurring phospholipids, followed by reacylation with fatty acid anhydrides, is accompanied by phosphoryl migration. The resulting mixture of α- and β-phospholipids was separated by short-column chromatography. Milder acylation procedures in which no phosphoryl migration occurs, were developed. 1,2-Dilinoleoyl-sn-glycero-3-phosphocholine was prepared in 50% yield by acylation of sn-glycero-3-phosphocholine (GPC) with N-linoleoylimidazole. Detailed NMR and infrared spectra of α- and β-phosphatidylcholines (PCs) and -ethanolamines (PEs) are reported and the differences between isomers discussed.  相似文献

Immobile plants and immobile modular animals outlive unitary animals. This paper discusses competing but not necessarily mutually exclusive theories to explain this extreme longevity, especially from the perspective of phenotypic plasticity. Stem cell immortality, vascular autonomy, and epicormic branching are some important features of the phenotypic plasticity of plants that contribute to their longevity. Monocarpy versus polycarpy can also influence the kind of senescent processes experienced by plants. How density-dependent phenomena affecting the establishment of juveniles in these immobile organisms can influence the evolution of senescence, and consequently longevity, is reviewed and discussed. Whether climate change scenarios will favour long-lived or short-lived organisms, with their attendant levels of plasticity, is also presented.  相似文献

Abstract

The current reconsideration of the place in nature of human beings unfortunately continues to be an acrimonious one. All too often the debate is more akin to a warlike encounter where each side attempts to gain control or the upper hand than a search for points of agreement. Given this context, it is important to entertain views that emanate from different cultural traditions as a way to infuse the debate with new life. Students of Native American culture have consistently pointed out that the essential concepts of life balance and reciprocity represented there may serve as useful points of consideration as we struggle with the appropriate relationships with animals and nature. Copper and iron play important roles in a variety of biological processes, especially when being chelated with proteins. The proteins involved in the metal binding, transporting and metabolism have aroused much interest. To facilitate the study on this topic, we constructed two databases (DCCP and DICP) containing the known copper- and iron-chelating proteins~ which are freely available from the website http://sdbi.sdut.edu.cn/en. Users can conveniently search and browse all of the entries in the databases. Based on the two databases, bioinformatic analyses were performed, which provided some novel insights into metalloproteins.  相似文献

1. 3R-[2-(14)C]Mevalonate was incorporated into geranyl and neryl beta-d-glucosides in petals of Rosa dilecta in up to 10.6% yield, and the terpenoid part was specifically and equivalently labelled in the moieties derived from isopentenyl pyrophosphate and 3,3-dimethylallyl pyrophosphate. A similar labelling pattern, with incorporations of 0.06-0.1% was found for geraniol or nerol formed in leaves of Pelargonium graveolens The former results provide the best available evidence for the mevalonoid route to regular monoterpenes in higher plants. 2. Incorporation studies with 3RS-[2-(14)C,(4R)-4-(3)H(1)]-mevalonate and its (4S)-isomer showed that the pro-4R hydrogen atom of the precursor was retained and the pro-4S hydrogen atom was eliminated in both alcohols and both glucosides. These results suggest that the correlation of retention of the pro-4S hydrogen atom of mevalonate with formation of a cis-substituted double bond, such as has been found in certain higher terpenoids, does not apply to the biosynthesis of monoterpenes. It is proposed that either nerol is derived from isomerization of geraniol or the two alcohols are directly formed by different prenyltransferases. Possible mechanisms for these processes are discussed. 3. The experiments with [(14)C,(3)H]mevalonate also show that in these higher plants, as has been previously found in animal tissue and yeast, the pro-4S hydrogen atom of mevalonate was lost in the conversion of isopentenyl pyrophosphate into 3,3-dimethylallyl pyrophosphate.  相似文献

Zinc is an essential trace element crucial for the function of more than 300 enzymes and it is important for cellular processes like cell division and apoptosis. Hence, the concentration of zinc in the human body is tightly regulated and disturbances of zinc homeostasis have been associated with several diseases including diabetes mellitus, a disease characterized by high blood glucose concentrations as a consequence of decreased secretion or action of insulin. Zinc supplementation of animals and humans has been shown to ameliorate glycemic control in type 1 and 2 diabetes, the two major forms of diabetes mellitus, but the underlying molecular mechanisms have only slowly been elucidated. Zinc seems to exert insulin-like effects by supporting the signal transduction of insulin and by reducing the production of cytokines, which lead to beta-cell death during the inflammatory process in the pancreas in the course of the disease. Furthermore, zinc might play a role in the development of diabetes, since genetic polymorphisms in the gene of zinc transporter 8 and in metallothionein (MT)-encoding genes could be demonstrated to be associated with type 2 diabetes mellitus. The fact that antibodies against this zinc transporter have been detected in type 1 diabetic patients offers new diagnostic possibilities. This article reviews the influence of zinc on the diabetic state including the molecular mechanisms, the role of the zinc transporter 8 and MT for diabetes development and the resulting diagnostic and therapeutic options.  相似文献

Small interfering RNA (siRNA) and microRNA (miRNA) are small RNAs of 18-25 nucleotides (nt) in length that play important roles in regulating gene expression. They are incorporated into an RNA-induced silencing complex (RISC) and serve as guides for silencing their corresponding target mRNAs based on complementary base-pairing. The promise of gene silencing has led many researchers to consider siRNA as an anti-viral tool. However, in long-term settings, many viruses appear to escape from this therapeutical strategy. An example of this may be seen in the case of human immunodeficiency virus type-1 (HIV-1) which is able to evade RNA silencing by either mutating the siRNA-targeted sequence or by encoding for a partial suppressor of RNAi (RNA interference). On the other hand, because miRNA targeting does not require absolute complementarity of base-pairing, mutational escape by viruses from miRNA-specified silencing may be more difficult to achieve. In this review, we discuss stratagems used by various viruses to avoid the cells' antiviral si/mi-RNA defenses and notions of how viruses might control and regulate host cell genes by encoding viral miRNAs (vmiRNAs).  相似文献
