The ultrastructure of the membrana granulosa (MG) of rat preovulatory follicles was examined using stereological techniques. Organelles studied were nuclei, mitochondria, lipid droplets (LD), lysosomes, and smooth and rough endoplasmic reticulum (SER, RER). The peripheral region of the MG contained the greatest volume of mitochondria, LD and SER, organelles associated with steroidogenesis. The volume of RER, an organelle associated with protein production, was greatest in the cumulus oophorus. These results corroborate previous analyses and demonstrate that the rat MG is composed of discrete subregions.  相似文献

The nematode Angiostrongylus cantonensis is a rat lungworm, a zoonotic pathogen that causes human eosinophilic meningitis and ocular angiostrongyliasis characteristic of rat lungworm (RLW) disease. Definitive diagnosis is made by finding and identifying A. cantonensis larvae in the cerebral spinal fluid or by using a custom immunological or molecular test. This study was conducted to determine if genomic DNA from A. cantonensis is detectable by qPCR in the blood or tissues of experimentally infected rats. F1 offspring from wild rats were subjected to experimental infection with RLW larvae isolated from slugs, then blood or tissue samples were collected over multiple time points. Blood samples were collected from 21 rats throughout the course of two trials (15 rats in Trial I, and 6 rats in Trial II). In addition to a control group, each trial had two treatment groups: the rats in the low dose (LD) group were infected by approximately 10 larvae and the rats in the high dose (HD) group were infected with approximately 50 larvae. In Trial I, parasite DNA was detected in cardiac bleed samples from five of five LD rats and five of five HD rats at six weeks post-infection (PI), and three of five LD rats and five of five HD rats from tail tissue. In Trial II, parasite DNA was detected in peripheral blood samples from one of two HD rats at 53 minutes PI, one of two LD rats at 1.5 hours PI, one of two HD rats at 18 hours PI, one of two LD rats at five weeks PI and two of two at six weeks PI, and two of two HD rats at weeks five and six PI. These data demonstrate that parasite DNA can be detected in peripheral blood at various time points throughout RLW infection in rats.  相似文献

We have developed a simple method for assessing the oxidative metabolic burst of peripheral blood leukocytes with a minute amount of whole peripheral blood by flow cytometry according to the method of Bass et al. with some modification. By this method, we can measure the H2O2 production by both granulocytes and monocytes in the same blood sample. The oxidative product formation by peripheral blood neutrophils can be monitored sequentially in the same mouse infected with E. coli. The mice infected intravenously with 0.1 LD50 of the bacteria showed increased basal activities from an early stage of infection; those infected intraperitoneally with the same dose of the bacteria showed a delayed enhancement. In case of infection with 0.01 LD50, the enhanced basal activities lasted for only a short period of time. The H2O2 production was correlated well with the clearance of the infected bacteria. These results demonstrated that the oxidative-product formation by peripheral blood neutrophils is affected by both the route and the dose of infection.  相似文献

The aim of this study was to compare changes in activity of acetylcholinesterase (AChE) in the brain and motor endplates of rat after administration of soman and tabun. We took brain and diaphragm from laboratory rats administered a median lethal dose (LD(50)) of soman or tabun. Enzyme activity of AChE was studied in selected structures of brain and in motor endplates in the diaphragm. Histochemical detection of AChE by Karnovski and Roots with simultaneous histochemical detection of alkaline phosphatase in case of brain sections was used. The highest activity of AChE in the control group was found in the striatum, amygdaloid nuclei, substantia nigra, superior colliculi, and motor nuclei of cranial nerves V, X a XII. LD(50) of both nerve agents dramatically decreased the activity of AChE in the structures studied--both brain and diaphragm. After intoxication by either agent, activity in above mentioned nuclei was characterized as low or focally moderate. Very low activity was seen in some structures (CA3 field of hippocampus, some nuclei of the tegmentum and cerebellar cortex). We found minimal differences in the histochemical picture of soman or tabun intoxication, apart from the striatum and the superior colliculi which showed stronger inhibition by tabun.  相似文献

A study was made of the dynamics of changes in the number of reticulocytes in the peripheral blood and of reticulocytograms in rats subjected to partial (front or hind body parts) X-irradiation with LD100/10, LD50/30, and LD0/30. The degree of the post-irradiation reticulocytopenia and modification of reticulocytograms was a function of radiation dose as observed 1-3 h following irradiation of the front part and 48 h after irradiation of the hind part of the animal body.  相似文献

Ceruloplasmin administered 60 min before irradiation diminished cAMP and cGMP levels, which were increased by irradiation at LD50 and LD100, and normalized cAMP/cGMP ratio in the rat liver during the first 24 h following irradiation with a dose of 6.24 Gy. The content of phospholipids increased and that of cholesterol decreased under the effect of ceruloplasmin leading to normalization of the molar cholesterol/phospholipid ratio in the rat liver on the 7th day of radiation sickness (LD50, 6.24 Gy).  相似文献

1. Male rats were injected intravenously with amounts ranging from 0.08 to 111.0mumoles of [(7)Be]beryllium sulphate/kg. body wt. The distribution in the rat and the subcellular distribution of beryllium in the liver were determined. 2. Within the entire dose range a higher specific activity of beryllium was present in a mitochondrial fraction containing the lysosomes. Purification of this fraction confirmed that beryllium is taken up by lysosomes. 3. With doses approaching the LD(50), beryllium was also found in increasing amounts to be present in the liver cell nuclei. Beryllium also showed affinity towards isolated cell nuclei in vitro. Evidence is presented that they have one class of binding sites for beryllium. Mitochondria have less affinity for beryllium. 4. No evidence could be obtained of an affinity of beryllium for DNA or RNA by fractionation of nuclei and dialysis experiments. 5. The presence of beryllium in liver cell nuclei may be relevant to the effects of beryllium on nuclear structure and function.  相似文献

目的跑台急性运动疲劳动物模型的建立及评价。方法选取清洁级雄性Wistar大鼠24只(8周龄)作为实验对象。采用多级递增负荷跑台运动方案(跑台坡度为0°,负荷分为三级)建立一次性力竭跑台运动动物模型。尾静脉取血,分别测定大鼠在安静、运动30 min、运动90 min、力竭、恢复30 min、恢复90 min各时间点外周血葡萄糖(GLU)、乳酸(LD)、尿素(BU)、丙二醛(MDA)浓度和肌酸激酶(CK)、超氧化物歧化酶(SOD)活性。结果一次性力竭运动过程中大鼠行为能力和运动能力、血液代谢产物及能量物质呈现阶段性的动态变化。外周血LD、BU、MDA浓度及CK活性均较安静时显著性增高(P<0.01,P<0.05);GLU浓度、SOD活性较安静时显著降低(P<0.01,P<0.05)。各指标的变化特征说明大鼠已达到运动疲劳状态。结论建立了大鼠一次性力竭跑台运动模型,并客观动态评价了大鼠在运动疲劳产生、发展、恢复等不同阶段各指标的变化特点及规律。该模型可用于后续运动疲劳机制的相关研究。  相似文献

Levels of the amino acids GABA and glutamine were determined in the whole brain of the white albino rat Rattus norvegicus after daily injection of 1/2, 1/4, 1/8, 1/16, 1/32 and 1/100 LD50 of cyolane. With 1/2 LD50 an increase in the level of both GABA and glutamine in the brain was recorded. Dose levels of 1/4 and 1/8 LD50 caused an increase in the level of GABA and a decrease in glutamine concentration followed by an increase from the 7th and 11th days for 1/4 and 1/8 LD50, respectively. The induced increase in GABA level started from the 2nd week for 1/16 and 1/32 LD50 and from the 3rd week for 1/100 LD50. Dose levels of 1/16, 1/32 and 1/100 LD50 caused a fluctuating increase in glutamine concentration starting from the 2nd, 3rd and 6th weeks, respectively, which was followed by a fluctuating decrease at the 9th week for 1/32 and 1/100 LD50. These findings support previous findings that the enhanced transformation of glutamic acid to GABA and glutamine is a result of a disturbance in the metabolism of the glutamic acid-GABA and the glutamic acid-glutamine systems in the rat brain.  相似文献

Inhibition of the hippocampus by the medial amygdala is mediated by vasopressin-like peptide. Because vasopressin has action on the periphery as well as the brain, we conducted experiments to evaluate the relationship between possible peripheral actions and the central effects of the endogenous peptide. In the acutely anesthetized rat, peptide-mediated inhibition of the hippocampus is not associated with significant changes in heart rate, blood pressure or body temperature. Peripheral injections of peptide agonist fail to evoke the central inhibition, and peripheral injections of peptide antagonist fail to block the central inhibition. Stimulation of central nuclei that contain vasopressin or a similar peptide also fail to duplicate the effect of stimulating the amygdala. We conclude that the peptidergic transmission is independent of peripheral causes or correlates.  相似文献

The relationship between acute toxicity in rats (LD50 values) and indicators of potential health hazards in humans was investigated, based on a chemical population-based paradigm (i.e. the "chemical diversity approach"). These structure-activity relationship-based analyses indicate that high toxicity in rats (i.e. a low LD50 value) is not a good predictor of health effects in humans. In fact, it was found that high acute toxicity to minnows, as well as toxicity to cultured cells, showed significantly greater associations with the potential for health effects than rat LD50 values.  相似文献

The purpose of this study was to examine bivariate relationships among cancer slope factor (CSF) and acute toxicity in rats and salmonid fish. Chemicals (n=43) were selected based on the availability of both oral CSF and acute toxicity data (rat oral median lethal dose [LD50] or salmonid median lethal concentration [LC50]). Rat oral LD50, salmonid LC50, and oral CSF data were log-transformed, and a Bonferroni-adjusted alpha level was set at 0.05 for subsequent correlation analysis. A significant correlation was observed between CSF and rat oral LD50 (r=?0.61) but not for CSF and salmonid LC50 (r=?0.29). Moreover, rat and fish acute toxicity were not significantly correlated (r=0.38). The significant correlation between CSF and rat oral LD50 compares favorably with published results reported in related studies. Accordingly, these results support prediction of carcinogenic potency, expressed as oral CSF, based in part on acute toxicity in rats.  相似文献

A comparative study of chromosome structures of bone marrow and peripheral blood cells has been carried out in 8 rhesus macaques (2-7 years of age), 4 of which survived after prolonged low-capacity (3.87 microA/kg) gamma irradiation, the total dose being 7.97 Gy (LD50/60). It has been established that prolonged low-capacity gamma irradiation was of a high mutagenic activity. Various tissues of irradiated monkeys showed differences in the frequency (4 months) and types (4-33 months) of aberrations within the period of 4 to 33 months following irradiation. Mutagenic effect characteristic of the early period after the irradiation was retained in the peripheral blood of irradiated monkeys within the period of observation.  相似文献

The effect of subchronic pyridostigmine pretreatment on the toxicity of soman, in the absence of supporting therapy (atropine, oxime, and (or) anticonvulsant), as well as its effect on muscarinic cholinoceptor binding characteristics was assessed in the rat. Pretreatment with pyridostigmine by means of an implanted Alzet osmotic minipump for a 5-day total exposure dose of 12 mg/kg inhibited whole blood acetylcholinesterase activity by 73%. This pyridostigmine pretreatment lowered the soman LD50 from 104 micrograms/kg in control animals to 82 micrograms/kg. In addition, the time to onset of soman-induced convulsions in pyridostigmine pretreated animals was significantly (p less than 0.001) reduced. Pyridostigmine pretreatment produced no significant effect on muscarinic cholinoceptor binding in brain or ileum. Lower doses of pyridostigmine pretreatment inhibited acetylcholinesterase activity (65 and 25%); however, LD50 and time to onset of convulsions following soman (140 micrograms/kg) were not significantly different from controls.  相似文献

Rabbits were treated with cyclophosphamide and 5-fluorouracil, myelosuppressive cytostatic agents, applied with a single dose of 1/3 LD50 or daily doses of 1/30 LD50 given for 14 days. Functional tests for evaluation of granulopoiesis were regularly performed at standard intervals and were following: leukocytosis, bone marrow picture with mitotic index, 3H-thymidine incorporation in vitro followed by autoradiography of labeled promyelocytes and myelocytes, serum lysozyme activity, mobilization of granulocyte reserve pool by staphylococcal alpha-toxin, cytochemistry of granulocytes, phagocytosis ability and Nitro-BT reduction. It has been found that 6-10 days after application of cytostatics, a marked depression of proliferation of young granulocyte forms and lowered reserve pool, are regularly observed. This was followed by spontaneous regeneration of granulopoiesis. No changes were noted in functional tests of mature granulocytes in peripheral blood. It is suggested that for investigation of the impairment of granulopoiesis after application of cytostatic agents, most suitable is evaluation of mobilization of the bone marrow reserve pool, lysozyme activity in blood serum and labelling of promyelocytes and myelocytes with 3H-thymidine in vitro.  相似文献

This work investigated the effects of chronic cadmium (Cd) exposure combined with γ-ray irradiation on the cytotoxicity and genotoxicity of peripheral blood cells and bone marrow cells in rats. Results showed that when the rats were exposed to low dose (LD) Cd of 0.1mg CdCl?/(kgd) for 8 and 12 weeks, the Cd concentration in blood reached to 135-140 μg/L and no toxic effects on peripheral blood lymphocytes, white blood cells (WBC) and granulocyte-monocyte (GM) progenitor cells were observed except polychromatic erythrocytes (PCE) of bone marrow. Moreover, this chronic LD Cd exposure significantly decreased irradiation-induced micronucleus (MN) formation and hypoxanthine-guanine phosphoribosyl transferase (hprt) mutation in lymphocytes and PCE, while the combination of LD Cd exposure and irradiation induced the additive metallothionein (MT) protein expression in bone marrow cells. When the rats were exposed to a high dose (HD) Cd of 0.5mg CdCl/?(kgd) for 8 and 12 weeks, the blood Cd level approached to 458-613 μg/L and an inflammatory response was induced, meanwhile, MN formation and hprt mutation were markedly increased, and the ratio of PCE/NCE (normochromatic erythrocyte) was significantly decreased. Furthermore, when the rats were exposed to HD Cd plus 2 Gy irradiation, additive toxic effects on MN formation, hprt mutation, PCE damage and GM progenitor cell proliferation were observed, while this combination treatment resulted in an obvious reduction of MT protein compared to HD Cd group. In conclusion, chronic exposure to LD Cd induced the adaptive response to irradiation in the genotoxicity of peripheral blood lymphocytes and PCE of bone marrow by the up-regulation of Cd-induced MT protein, but the combination of HD Cd exposure and irradiation generated the additive effects on the cytotoxicity and genotoxicity in peripheral blood lymphocytes and bone marrow cells.  相似文献

Metacid 50, the commercial grade of methyl parathion (O,O-dimethyl-O-4-nitrophenyl phosphorothionate), a commonly used organophosphorus insecticide, was tested for its genotoxicity in Swiss albino mice using the sperm abnormality assay. Sperms of albino mice were examined at two time intervals, 1 week and 5 weeks after a single acute oral treatment with the pesticide at four dose levels, viz., 75.0, 37.5, 18.75 and 9.375 mg/kg body weight corresponding to 1/2 LD50, 1/4 LD50, 1/8 LD50 and 1/16 LD50 values respectively. A dose-related statistically significant increase in the percentage of abnormal sperm observed indicates the genotoxic potency of methyl parathion.  相似文献

Individual and group differences were revealed in the capacity of rat peripheral blood lymphocytes of DNA repair in response to the DNA damaging test-effect of irradiation in vitro. The survival rate of rats irradiated with a dose inducing 50-per cent death was in a definite relationship with the repair capacity of blood lymphocyte DNA in each animal individual prior to irradiation.  相似文献

We describe a limiting dilution (LD) culture system in which cell sorter-purified CD4+ (and CD8+) peripheral blood T cells are cocultured with irradiated, anti-CD3 mab-producing OKT3 hybridoma cells. Under these conditions, one out of 2-3 CD4+ (and CD8+) T cells is induced to clonal proliferation. In striking contrast to previously described LD culture systems, every growing CD4+ cell clone displayed cytotoxic activity when tested in a lectin-facilitated 51Cr release assay against P815 target cells. This contrasts with the development of cytotoxic CD4+ T cells in alloantigen-stimulated LD cultures, where only one out of 15-20 proliferating CD4+ cells killed P815 in the presence of PHA, and one out of 300-500 proliferating CD4+ cells displayed alloantigen-specific cytotoxic activity. Furthermore, we have established antigen-specific proliferating CD4+ T cell clones which do not exert antigen-specific cytotoxicity but can be cytotoxic when crosslinked to target cells via lectin or monoclonal antibody (anti-CD3, anti-TCR). Our results show that a previously unrecognized large fraction (at least 30-50%) of all peripheral blood CD4+ T cells can give rise to cytotoxic effector cells. The mode of CD4+ T cell activation (OKT3 hybridoma versus alloantigen) thus determines whether the intrinsic cytotoxic capacity of CD4+ T cells is functionally activated or not.  相似文献

1. In nuclei isolated from cells of the B50 rat neuroblastoma line the stimulatory effect of methyl mercury on alpha-amanitin-sensitive RNA synthesis is very much reduced compared to the stimulatory effect in HeLa nuclei (see: Frenkel G. D. and Randles K. (1982) Specific stimulation of alpha-amanitin-sensitive RNA synthesis in isolated HeLa nuclei by methyl mercury. J. biol. Chem. 257, 6275-6279). 2. The stimulatory effect of another mercury compound, p-hydroxymercuribenzoate, was also much less pronounced in the B50 nuclei. 3. Similar results were obtained with nuclei isolated from B50 cells which had been induced to differentiate by exposure to dibutaryl cyclic AMP. 4. Nuclei isolated from cells of another rat neuroblastoma line (B35), and nuclei from cells of a human neuroblastoma line both exhibited levels of stimulation similar to that of HeLa nuclei. 5. The B50 and HeLa cells were also compared as to their sensitivity to other effects of methyl mercury.  相似文献
