摘要 目的:寻找具有血栓素A2受体(Thromboxane A2 receptor,TP)抑制作用的选择性环氧合酶-2(Cyclooxygenase-2,COX-2)抑制剂,以期降低其心血管疾病风险。方法:本研究从公开数据库中获取了512种TP抑制剂,通过分子对接、分子动力学模拟和ADMET预测,筛选出化合物TP84。结果:分子对接结果显示,与先前获批的选择性COX-2抑制剂罗非昔布相比,TP84对COX-2的亲和力更高,对环氧合酶-1(Cyclooxygenase-1,COX-1)的亲和力更低;分子动力学模拟进一步表明,模拟过程中TP84与COX-1的结合不稳定,而TP84能稳定结合COX-2,与COX-2的结合自由能是COX-1的3倍;此外,根据ADMET预测,TP84的药物化学、吸收、分布、代谢、排泄和毒性处于类药物候选物的可接受范围内。结论:TP84是一种潜在的低心血管疾病风险选择性COX-2抑制剂。  相似文献

摘要 目的:本研究旨在阐明过表达COX-2的结直肠癌中花生四烯酸代谢与PTEN及其下游通路的相互关系。方法:利用过表达COX-2结直肠癌细胞系CT26和Apc Min/+小鼠腺瘤模型,联合花生四烯酸、COX-2抑制剂NS-398和COX-2基因敲除的干预,采用流式细胞仪检测细胞凋亡、细胞周期及活性氧的产生;Transwell和克隆形成试验观察细胞的迁移和增殖能力;Western blot和免疫组化检测PTEN及其下游相关蛋白的表达。结果:花生四烯酸通过COX-2酶代谢产生活性氧并失活PTEN抑癌基因表达,从而激活PI3K-AKT蛋白促进CT26结直肠癌细胞迁移和增殖,抑制细胞凋亡;而COX-2抑制剂NS-398阻止了花生四烯酸在CT26结直肠癌细胞中的恶性生物学行为。同时,在COX-2基因敲除Apc Min/+小鼠腺瘤组织中,减弱了氧化应激水平,增加了PTEN表达,抑制了PI3K-AKT磷酸化,进一步抑制腺瘤生长,提高小鼠生存率。结论:花生四烯酸通过COX-2酶代谢产生活性氧下调PTEN活性,并激活PI3K-AKT促进结直肠癌生长;COX-2抑制剂可间接促进PTEN表达,抑制结直肠癌生长,能够作为CRC的潜在治疗靶点。  相似文献

目的 基于位点特异性打分矩阵(position-specific scoring matrices,PSSM)的预测模型已经取得了良好的效果,基于PSSM的各种优化方法也在不断发展,但准确率相对较低,为了进一步提高预测准确率,本文基于卷积神经网络(convolutional neural networks,CNN)算法做了进一步研究。方法 采用PSSM将启动子序列处理成数值矩阵,通过CNN算法进行分类。大肠杆菌K-12(Escherichia coli K-12,E.coli K-12,下文简称大肠杆菌)的Sigma38、Sigma54和Sigma70 3种启动子序列被作为正集,编码(Coding)区和非编码(Non-coding)区的序列为负集。结果 在预测大肠杆菌启动子的二分类中,准确率达到99%,启动子预测的成功率接近100%;在对Sigma38、Sigma54、Sigma70 3种启动子的三分类中,预测准确率为98%,并且针对每一种序列的预测准确率均可以达到98%以上。最后,本文以Sigma38、Sigma54、Sigma70 3种启动子分别和Coding区或者Non-coding区序列做四分类,预测得到的准确性为0.98,对3种Sigma启动子均衡样本的十交叉检验预测精度均可以达到0.95以上,海明距离为0.016,Kappa系数为0.97。结论 相较于支持向量机(support vector machine,SVM)等其他分类算法,CNN分类算法更具优势,并且基于CNN的分类优势,编码方式亦可以得到简化。  相似文献

目的 研究构建基于共祖(identity-by-descent,IBD)片段算法预测远亲缘关系分析流程并评估预测准确性。方法 采用高密度单核苷酸多态性(single nucleotide polymorphism,SNP)芯片对253份家系样本进行检测,研究基于IBD片段算法的分析流程进行两两个体间亲缘关系预测,评估预测准确性。随机减少SNP位点,评估位点数对算法预测准确性的影响。结果 IBD片段算法预测1~7级亲缘关系平均置信区间准确率为94.72%,预测可信度为99.77%,6级及以上亲缘关系预测时出现假阴性。随着SNP数量减少,预测准确性会出现一定程度的下降。结论 IBD片段算法可用于7级以内亲缘关系的预测,该算法在群体遗传学、法医遗传学等领域有重要应用价值。  相似文献

摘要 目的:探讨温补肾阳法治疗肾阳虚模型大鼠多尿症状的作用机制。方法:90只雄性SD大鼠随机分成干预组、抑制剂组、空白组、模型组,干预组根据中药剂量分为高剂量组、中剂量组、低剂量组。模型组、干预组及抑制剂组接受肾阳虚模型制备,干预组在成模后每日接受7 g/kg、14 g/kg、28 g/kg剂量中药灌胃,连续灌胃14 d。抑制剂组大鼠接受尾静脉注射通路抑制剂H-89。干预结束后比较各组脏器指数、24 h尿量、24 h尿蛋白水平以及尿液钠离子(Na+)、钾离子(K+)、氯离子(Cl-)浓度、肾脏病理变化、血清醛固酮(ALD)、乙醇脱氢酶(ADH)、促肾上腺皮质激素释放因子(CRF)、大鼠促肾上腺皮质激素(ACTH)、大鼠皮质醇(CORT)、蛋白激酶A(PKA)、蛋白激酶A(cAMP)含量、肾脏组织水通道蛋白2(AQP-2)蛋白表达的变化。结果:温补肾阳法可明显减少肾阳虚模型大鼠尿量,改善临床症状,且具有一定的剂量依赖性(P<0.05)。经过中药干预后大鼠24 h尿蛋白、脏器指数、尿液Na+、Cl-均下降,尿液K+、血清ALD、ADH、CRF、ACTH、CORT、PKA、cAMP含量、肾脏组织AQP-2蛋白表达上调(P<0.05),且抑制剂H-89可阻断该作用。结论:温补肾阳法可明显改善肾阳虚模型大鼠多尿症状,其作用机制可能通过cAMP-PKA-AQP2通路介导。  相似文献

摘要 目的:探讨雌激素受体(ER)、环氧化酶-2(COX-2)在卵巢癌患者中的表达及临床意义。方法:选取经病理手术确诊的120例患者的石蜡卵巢组织切片标本,其中卵巢癌54例,卵巢良性肿瘤66例。采用免疫组化检测ER、COX-2的表达情况。结果:ER和COX-2在卵巢癌组织中的阳性表达率明显高于良性卵巢肿瘤组织(P<0.05)。Ⅳ期卵巢癌组织中ER阳性表达率明显高于Ⅰ期(P<0.05),其余病理分期比较无差异(P>0.05);不同病理分期的COX-2阳性表达率比较无差异(P>0.05)。不同组织学分级卵巢癌组织中ER阳性表达率组组比较均无差异(P>0.05)。Ⅱ级卵巢癌组织中COX-2阳性表达率明显高于Ⅰ级(P<0.05),其余病理分级比较无统计学意义(P>0.05)。伴有转移卵巢癌组织中ER阳性表达率明显高于无转移(P<0.05);伴有转移卵巢癌组织中COX-2阳性表达率与无转移比较无统计学意义(P>0.05);伴有转移卵巢癌组织中ER、COX-2表达免疫组织化学评分法(IHS)评分均高于无转移(P<0.05)。结论:ER和COX-2在卵巢癌组织和卵巢良性肿瘤组织中的表达逐渐上调,且在卵巢癌组织的表达高于卵巢良性肿瘤组织;ER的表达与卵巢癌病理分期和转移相关,COX-2与组织学分级相关,二者对卵巢癌病情的预测具有互补性,可作为生物学指标,对卵巢癌恶性程度进行判断评估。  相似文献

目的 长链非编码RNA在遗传、代谢和基因表达调控等方面发挥着重要作用。然而,传统的实验方法解析RNA的三级结构耗时长、费用高且操作要求高。此外,通过计算方法来预测RNA的三级结构在近十年来无突破性进展。因此,需要提出新的预测算法来准确的预测RNA的三级结构。所以,本文发展可以用于提高RNA三级结构预测准确性的碱基关联图预测方法。方法 为了利用RNA理化特征信息,本文应用多层全卷积神经网络和循环神经网络的深度学习算法来预测RNA碱基间的接触概率,并通过注意力机制处理RNA序列中碱基间相互依赖的特征。结果 通过多层神经网络与注意力机制结合,本文方法能够有效得到RNA特征值中局部和全局的信息,提高了模型的鲁棒性和泛化能力。检验计算表明,所提出模型对序列长度L的4种标准(L/10、L/5、L/2、L)碱基关联图的预测准确率分别达到0.84、0.82、0.82和0.75。结论 基于注意力机制的深度学习预测算法能够提高RNA碱基关联图预测的准确率,从而帮助RNA三级结构的预测。  相似文献

摘要 目的:探讨乳腺癌改良根治术患者术后复发转移的危险因素及血清糖类抗原125(CA125)、环加氧酶-2(COX-2)、可溶性肿瘤坏死因子受体P55(sTNFR-P55)的预测价值。方法:对2014年1月至2016年12月新疆医科大学第一附属医院收治的109例行乳腺癌改良根治术的乳腺癌患者进行前瞻性研究,所有患者术后均随访5年,其中2例失访,107例完成随访。根据5年内患者复发转移情况将其分为复发转移组(n=31)和未复发转移组(n=76)。收集患者入院时的临床病理资料,采用电化学发光法检测术前血清CA125,采用酶联免疫吸附法检测术前血清COX-2、sTNFR-P55。采用logistic回归模型分析患者术后复发转移的影响因素,绘制受试者工作特征(ROC)曲线评估血清CA125、COX-2、sTNFR-P55对术后复发转移的预测价值。结果:复发转移组肿瘤直径>5 cm、浸润性非特殊癌、脉管癌栓、雌激素受体(ER)/孕激素受体(PR)阴性、无内分泌治疗构成比、TNM分期IIIA期、腋窝淋巴结转移数量4~9个构成比高于未复发转移组(P<0.05)。复发转移组血清CA125、COX-2、sTNFR-P55水平高于未复发转移组(P<0.05)。多因素logistic回归分析结果显示,肿瘤直径>5 cm、浸润性非特殊癌、TNM分期IIIA期、脉管癌栓、腋窝淋巴结转移数量4~9个、CA125升高、COX-2升高、sTNFR-P55升高是乳腺癌改良根治术患者术后5年内复发转移的独立危险因素(OR=1.318、1.213、1.223、1.137、1.257、1.241、1.313、1.351,P<0.05)。血清CA125、COX-2、sTNFR-P55均可有效预测乳腺癌术后复发转移,曲线下面积(AUC)分别为0.803、0.749、0.761,三指标联合预测术后复发转移的AUC为0.915,灵敏度和特异度分别为0.94、0.83。结论:肿瘤直径、浸润性非特殊癌、TNM分期、脉管癌栓、腋窝淋巴结转移数量以及术前血清CA125、COX-2、sTNFR-P55异常升高是乳腺癌改良根治术患者术后5年内复发转移的危险因素,术前血清CA125、COX-2、sTNFR-P55联合检测可预测乳腺癌改良根治术后的复发转移风险。  相似文献

目的 从中药筛选具有潜在抑制严重急性呼吸综合征冠状病毒2 (SARS-CoV-2) 活性的成分,进一步从原子水平揭 示其抑制SARS-CoV-2 表面刺突蛋白(S 蛋白) 受体结合域(RBD) 与血管紧张素转化酶2 (ACE2) 结合的内在机制。 方法 检索新型冠状病毒(简称“新冠肺炎”) 治疗中药处方,构建“新冠肺炎中药候选活性成分数据库”。用具有ACE2 抑制活性的小分子化合物构建HipHop药效团模型,并对“新冠肺炎中药候选活性成分数据库”中活性成分筛选。采用分子 对接和分子动力学模拟方法研究候选活性成分与ACE2 的结合方式及其对SARS-CoV-2 S 蛋白与ACE2 识别的影响。 结果 本文通过中药处方挖掘和分子动力学模拟,从143 个新冠肺炎治疗中药处方中筛选出10 种可与SARS-CoV-2 S 蛋白/ 人源ACE2 识别位点结合的中药成分。其中,枇杷叶主要活性成分23-trans-p-coumaryhormentic acid 与ACE2 具有最高的亲和 力,且23-trans-p-coumaryhormentic acid 的结合可有效阻断SARS-CoV-2 S蛋白与宿主细胞ACE2 的结合。结论 本文通过虚 拟筛选发现了SARS-CoV-2 潜在抑制剂分子23-trans-p-coumaryhormentic acid,同时从原子水平预测了其抑制SARS-CoV-2 S 蛋白与ACE2 结合的内在机制,这将为SARS-CoV-2 特异性抗病毒药物的研发提供理论依据。  相似文献

摘要 目的:探讨使用Wnt3a蛋白受体竞争性抑制剂Sfrp2下调Wnt通路的关键蛋白β-catenin的表达对胶质瘤细胞U251线粒体功能以及细胞侵袭能力的影响。方法:使用外源性Sfrp2蛋白处理U251细胞,使用Western Blot技术,Mitotracker线粒体形态学染色,划痕实验观测Sfrp2蛋白对U251细胞Wnt通路的表达,线粒体功能以及细胞迁移能力的影响,并使用GSK-3β抑制剂LiCl上调Wnt通路进一步明确Sfrp2蛋白的作用机制。结果:Sfrp2蛋白可引起Wnt通路的抑制(P<0.05),线粒体分裂(P<0.01)以及细胞迁移能力的下降(P<0.01),而使用LiCl处理后,Wnt通路重现上调(P<0.05),线粒体分裂受到抑制(P<0.05),细胞的迁移能力也再次恢复(P<0.05)。结论:Sfrp2可通过下调Wnt通路引起线粒体分裂进而抑制U251细胞的迁移能力。  相似文献

ObjectiveStudying the diagnostic value of CT imaging in non-small cell lung cancer (NSCLC), and establishing a prognosis model combined with clinical characteristics is the objective, so as to provide a reference for the survival prediction of NSCLC patients.MethodCT scan data of NSCLC 200 patients were taken as the research object. Through image segmentation, the radiology features of CT images were extracted. The reliability and performance of the prognosis model based on the optimal feature number of specific algorithm and the prognosis model based on the global optimal feature number were compared.Results30-RELF-NB (30 optimal features, RELF feature selection algorithm and NB classifier) has the highest accuracy and AUC (area under the subject characteristic curve) in the prognosis model based on the optimal features of specific algorithm. Among the prognosis models based on global optimal features, 25-NB (25 global optimal features, naive Bayes classification algorithm classifier) has the highest accuracy and AUC. Compared with the prediction model based on feature training of specific feature selection algorithm, the overall performance and stability of the prediction model based on global optimal feature are higher.ConclusionThe prognosis model based on the global optimal feature established in this paper has good reliability and performance, and can be applied to the CT radiology of NSCLC.  相似文献

A series of 2-(4-substitutedmethylphenyl)propionic acid derivatives (6a–6m) were synthesized, characterized and evaluated for cyclooxygenase (COX) enzyme inhibitory and antimicrobial activity. Test compounds that exhibited good COX inhibition and antibacterial activity were further screened for their cytotoxicity and genotoxicity. Compounds 6h and 6l showed better COX-1 and COX-2 inhibition when compared to ibuprofen. Inhibition potency of these compounds against COX-2 was very close to that of nimesulide. The compounds 6d, 6h, 6l and 6m displayed promising antibacterial property when compared to chloramphenicol. However, the compound 6l was emerged as the best dual COX inhibitory-antibacterial agent in this study. The ADME prediction of the compounds revealed that they may have a good pharmacokinetic profile. Docking results of the compounds 6h and 6l with COX-1 (PDB ID: 1EQG) also exhibited a strong binding profile.

AimOur aim is to understand the molecular mechanisms of the selective nonsteroidal anti-inflammatory drugs (NSAID), cyclooxygenase-2 (COX-2) inhibitors', higher “priority” to reduce synthesis of the vascular protector, prostacyclin (PGI2), compared to that of nonselective NSAIDs.Main methodsCOX-1 or COX-2 was co-expressed with PGI2 synthase (PGIS) in COS-7 cells. The Km and initial velocity (½t Vmax) of the coupling reaction between COX-1 and COX-2 to PGIS were established. The experiment was further confirmed by a kinetics study using hybrid enzymes linking COX-1 or COX-2 to PGIS. Finally, COX-1 or COX-2 and PGIS were respectively fused to red (RFP) and cyanic (CFP) fluorescence proteins, and co-expressed in cells. The distances between COXs and PGIS were compared by FRET.Key findingsThe Km for converting arachidonic acid (AA) to PGI2 by COX-2 coupled to PGIS is ~ 2.0 μM; however, it was 3-fold more (~ 6.0 μM) for COX-1 coupled to PGIS. The Km and ½t Vmax for COX-2 linked to PGIS were ~ 2.0 μM and 20 s, respectively, which were 2–5 folds faster than that of COX-1 linked to PGIS. The FRET study found that the distance between COX-2-RFP and PGIS–CFP is shorter than that between COX-1-RFP and PGIS–CFP.SignificanceThe study provided strong evidence suggesting that the low Km, faster ½t Vmax, and closer distance are the basis for COX-2 dominance over COX-1 (coupled to PGIS) in PGI2 synthesis, and further demonstrated the mechanisms of selective COX-2 inhibitors with higher potential to reduce synthesis of the vascular protector, PGI2.  相似文献

In this research, we exploited derivatives of thieno[2,3-b]pyridine as dual inhibitors of the key enzymes in eicosanoid biosynthesis, cyclooxygenase (COX, subtypes 1 and 2) and 5-lipoxygensase (5-LOX). Testing these compounds in a rat paw oedema model revealed potency higher than ibuprofen. The most active compounds 7a, 7b, 8b, and 8c were screened against COX-1/2 and 5-LOX enzymes. Compound 7a was the most powerful inhibitor of 5-LOX with IC50?=?0.15?µM, while its p-chloro analogue 7b was more active against COX-2 (IC50?=?7.5?µM). The less desirable target COX-1 was inhibited more potently by 8c with IC50?=?7.7?µM. Surflex docking programme predicted that the more stable anti- conformer of compound (7a) formed a favourable complex with the active site of 5-LOX but not COX-1. This is in contrast to the binding mode of 8c, which resembles the syn-conformer of series 7 and binds favourably to COX-1.  相似文献

AimAvailability of uniformly collected presence, absence, and abundance data remains a key challenge in species distribution modeling (SDM). For invasive species, abundance and impacts are highly variable across landscapes, and quality occurrence and abundance data are critical for predicting locations at high risk for invasion and impacts, respectively. We leverage a large aquatic vegetation dataset comprising point‐level survey data that includes information on the invasive plant Myriophyllum spicatum (Eurasian watermilfoil) to: (a) develop SDMs to predict invasion and impact from environmental variables based on presence–absence, presence‐only, and abundance data, and (b) compare evaluation metrics based on functional and discrimination accuracy for presence–absence and presence‐only SDMs.LocationMinnesota, USA.MethodsEurasian watermilfoil presence–absence and abundance information were gathered from 468 surveyed lakes, and 801 unsurveyed lakes were leveraged as pseudoabsences for presence‐only models. A Random Forest algorithm was used to model the distribution and abundance of Eurasian watermilfoil as a function of lake‐specific predictors, both with and without a spatial autocovariate. Occurrence‐based SDMs were evaluated using conventional discrimination accuracy metrics and functional accuracy metrics assessing correlation between predicted suitability and observed abundance.ResultsWater temperature degree days and maximum lake depth were two leading predictors influencing both invasion risk and abundance, but they were relatively less important for predicting abundance than other water quality measures. Road density was a strong predictor of Eurasian watermilfoil invasion risk but not abundance. Model evaluations highlighted significant differences: Presence–absence models had high functional accuracy despite low discrimination accuracy, whereas presence‐only models showed the opposite pattern.Main conclusionComplementing presence–absence data with abundance information offers a richer understanding of invasive Eurasian watermilfoil''s ecological niche and enables evaluation of the model''s functional accuracy. Conventional discrimination accuracy measures were misleading when models were developed using pseudoabsences. We thus caution against the overuse of presence‐only models and suggest directing more effort toward systematic monitoring programs that yield high‐quality data.  相似文献

目的:探讨Notch信号通路对肝癌细胞迁移能力及钙粘附蛋白E(E-cadherin)、环氧化酶-2(COX-2)表达的影响。方法:体外培养肝癌细胞系(SMMC-7721、MHCC97H)、正常非肿瘤肝细胞系(HL-7702),Transwell小室用于测定细胞的迁移侵袭能力,Western blot蛋白印迹法用于测定Notch1、E-cadherin、COX-2蛋白的表达水平,并采用DAPT阻断Notch信号通路,比较肝癌细胞系与正常非肿瘤肝细胞系的迁移侵袭能力及肝癌细胞中E-cadherin、COX-2蛋白的表达水平的改变。结果:SMMC-7721细胞、MHCC97H细胞的迁移能力强于HL-7702细胞,差异有统计学意义(P0.05);相比于HL-7702细胞,MHCC97H细胞、SMMC-7721细胞中的Notch1、COX-2表达水平均显著升高,E-cadherin的表达水平明显降低(P0.05);DAPT处理后,SMMC-7721细胞、MHCC97H细胞发生迁移的能力均弱于对照组,差异有统计意义(P0.05);DAPT处理后,SMMC-7721细胞、MHCC97H细胞内COX-2、Notch1的表达量明显降低,而E-cadherin的表达水平升高(P0.05)。结论:Notch信号通路参与肝癌细胞迁移过程,其机制可能与E-cadherin、COX-2的表达相关。  相似文献

PurposeTo assess the influence of reconstruction algorithms and parameters on the PET image quality of brain phantoms in order to optimize reconstruction for clinical PET brain studies in a new generation PET/CT.MethodsThe 3D Hoffman phantom that simulates 18F-fluorodeoxyglucose (FDG) distribution was imaged in a Siemens Biograph mCT TrueV PET/CT with Time of Flight (TOF) and Point Spread Function (PSF) modelling. Contrast-to-Noise Ratio (CNR), contrast and noise were studied for different reconstruction models: OSEM, OSEM + TOF, OSEM + PSF and OSEM + PSF + TOF.The 2D multi-compartment Hoffman phantom was filled to simulate 4 different tracers' spatial distribution: FDG, 11C-flumazenil (FMZ), 11C-Methionine (MET) and 6-18F-fluoro-l-dopa (FDOPA). The best algorithm for each tracer was selected by visual inspection. The maximization of CNR determined the optimal parameters for each reconstruction.ResultsIn the 3D Hoffman phantom, both noise and contrast increased with increasing number of iterations and decreased with increasing FWHM. OSEM + PSF + TOF reconstruction was generally superior to other reconstruction models. Visual analysis of the 2D Hoffman brain phantom suggested that OSEM + PSF + TOF is the optimum algorithm for tracers with focal uptake, such as MET or FDOPA, and OSEM + TOF for tracers with diffuse cortical uptake (i.e. FDG and FMZ). Optimization of CNR demonstrated that OSEM + TOF reconstruction must be performed with 2 iterations and a filter FWHM of 3 mm, and OSEM + PSF + TOF reconstruction with 4 iterations and 1 mm FWHM filter.ConclusionsOptimization of reconstruction algorithm and parameters has been performed to take particular advantage of the last generation PET scanner, recommending specific settings for different brain PET radiotracers.  相似文献

A series of 26 selective COX-2 inhibitors which reported previously by our laboratory was selected to generate three-dimensional quantitative structure activity relationship (3D-QSAR) model. Active conformation of each molecule was predicted by docking studies and used for molecular alignment. Activity of 20 molecules as a train set was predicted using three methods including comparative molecular field analysis (CoMFA), CoMFA region focusing (CoMFA-RG) and comparative molecular similarity index analysis (CoMSIA). The best models of CoMFA-RG and CoMSIA revealed correlation coefficients r2 of 0.955 and 0.947, the leave one out cross-validation coefficients q2 of 0.573 and 0.574, respectively. In addition, CoMFA-RG and CoMSIA models were validated by a test set of six molecules with predicted coefficients r2pred of 0.644 and 0.799, respectively. Contour maps of generated models provided fruitful information about structural aspect of molecules that affected their COX-2 inhibitory activity. Based on three models results, steric and electrostatic properties are the most important factors in controlling the activity of the molecules. Results of CoMFA-RG and CoMSIA models were utilized to design new molecules. Comparison of experimental and predicted pIC50 values of designed molecules indicated that CoMFA-RG had the more predictive ability.

目的:探讨胆管癌组织白介素-6(IL-6)、环氧合酶-2(COX-2)和血管内皮生长因子(VEGF)的表达及临床意义。方法:将手术切除并经病理诊断确诊的胆管癌石蜡包埋标本80例纳为胆管癌组,另取癌旁正常胆管组织作为对照组,采用免疫组织化学SP法检测两组组织中IL-6、COX-2、VEGF的表达情况并做比较,分析胆管癌组织中VEGF、COX-2、IL-6阳性表达与临床病理特征关系,采用Spearman等级相关分析胆管癌组织中VEGF、COX-2、IL-6表达的相关性。结果:胆管癌组的VEGF、COX-2、IL-6阳性表达率均显著高于对照组,组间比较差异有统计学意义(P0.05)。胆管癌组织中VEGF、COX-2、IL-6阳性表达率与有无淋巴结转移、TNM分期、分化程度有关(P0.05),而与性别、年龄、肿瘤直径无关(P0.05),其中有淋巴结转移、TNM分期Ⅲ~Ⅳ期、低分化程度的胆管癌患者的VEGF、COX-2、IL-6阳性表达率高于无淋巴结转移、TNM分期Ⅰ~Ⅱ期、中高分化程度的胆管癌患者(P0.05)。Spearman等级相关分析显示,胆管癌组织中VEGF与COX-2、IL-6呈正相关(P0.05),COX-2与IL-6也呈正相关(P0.05)。结论:胆管癌组织IL-6、COX-2、VEGF均呈现高表达,并与胆管癌的生长、转移密切相关,检测IL-6、COX-2和VEGF有助于判断胆管癌疾病进展。  相似文献
