The aim of this study was to synthesize and evaluate plasmid DNA interaction of new platinum(II) complexes with some 2-substituted benzimidazole derivatives as carrier ligands which may have potent anticancer activity and low toxicity. Twelve benzimidazole derivatives carrying indole, 2-/or 3-/or 4-methoxyphenyl, 4-methylphenyl, 3,4-dimethoxyphenyl, 3,4,5-trimethoxyphenyl, 4-methoxybenzyl, 3,4,5-trimethoxybenzyl, 3,4,5-trimethoxystyryl, 3,4,5-trimethoxybenzylthio or dimethylamino ethyl groups in their position 2 and twelve platinum(II) complexes with these carrier ligands were synthesized. The chemical structure of the platinum complexes have been characterized by their elemental analysis and FIR, 1H NMR and mass spectra and their 1H NMR and FIR spectra were interpreted by comparison with those of the ligands. The interaction of all the ligands and their complexes with plasmid DNA and their restriction endonuclease reactions by BamHI and HindIII enzymes were studied by agarose gel electrophoresis. It was determined that complex 1 [dichloro-di(2-(1H-indole-3-yl)benzimidazole)platinum(II)·2H2O] has stronger interaction than carboplatin and complex 10 [dichloro-di(2-(3,4,5-trimethoxystyryl)benzimidazole)platinum(II)·2H2O] has stronger interaction than both carboplatin and cisplatin with plasmid DNA.  相似文献

Three novel cisplatin analogues were synthesized, designed according to an approach which violates the “classical” structure-activity relationship, by replacing the diamine ligands with a planar N donor heterocycle giving a sterically hindered complex. Moreover, the sterical hindrance of antitumor drug candidates potentially makes them less susceptible to deactivation by sulphur-containing proteins and helping to overcome resistance mechanisms. The resulting mononuclear complexes of sterically hindered polidentate heterocyclic N ligands [PtCl(bbp)]Cl (1) [bbp = 2,6-bis(2-benzimidazolyl)pyridine], [PtCl2(dptdn)](H2O) (2) [dptdn = sodium 5,6-diphenyl-3-(2′-pyridyl)-1,2,4-triazine-4″,4″′-disulfonate] and [(dptdn)(dpt)Pt]Cl2(H2O) (3) [dpt = 5,6-diphenyl-3-(2′-pyridyl)-1,2,4-triazine] have been prepared and structurally characterised. Both neutral and ionic complexes are present, with monofunctional (1) and bifunctional Pt(II) moieties (2) and coordinatively saturated Pt(II) ions in the mixed ligand complex (3), whose size and shape enable them to behave as novel scaffolds for DNA binding. All complexes were tested “in vitro” for their biological activity on human HT29 colorectal carcinoma and HepG2 hepatoma cells. The complexes (1) and (3), endowed with a positive charge, showed a potent cytotoxic activity and reduced cell viability with an efficacy higher than that of cisplatin; whilst the neutral bifunctional compound (2) was inactive. IC50 values have been calculated for the active compounds. The cytotoxic effects were confirmed by the accumulation of treated cells in subG0/G1 phase of cell cycle, by the loss of mitochondrial potential (Δψm) and by the chromatin condensation or fragmentation observed by means of fluorescence microscopy after Hoechst 33258 nuclear staining. A study on intracellular platinum uptake in HT29 cell line has been also performed and data obtained strongly suggest that the cytotoxicity of new tested complexes reported in this work is based on a different pharmacodynamic pattern with respect to cisplatin.  相似文献

An array of poly- and mononuclear complexes of Pt(II) with polypyridyl ligands is reported. The framework complexes [(PtCl(2))(2)(bpp)(2)(micro-PtCl(2))](H(2)O)(2) [bpp=2,3-bis(2-pyridyl)pyrazine], [PtCl(2)(micro-tptz)PtClNCPh]Cl [tptz=2,4,6-tris(2-pyridyl)-1,3,5-triazine], and mononuclear PtCl(2)(NH(2)dpt) [NH(2)dpt=4-amino-3,5-bis(2-pyridyl)-1,2,4-triazole] have been prepared and structurally characterized. Both neutral and ionic complexes are present, with bifunctional and monofunctional Pt(II) moieties, whose size and shape enable them to behave as novel scaffolds for DNA binding. Pt(II) complexes were tested for their biological activity. Cell viability assay and flow cytometric analysis demonstrated that these complexes, particularly [PtCl(2)(micro-tptz)PtClNCPh]Cl, were effective death inducers in human colon rectal carcinoma HT29 cells and their cytotoxic activity was higher than that exerted by cisplatin. Morphological analysis of treated HT29 cells, performed by fluorescence microscopy after Hoechst 33258 staining, showed the appearance of the typical features of apoptosis. Moreover, our results suggested that mitochondria are involved in apoptosis induced by Pt(II) complexes in HT29 cells as demonstrated by dissipation of mitochondrial transmembrane potential.  相似文献

A novel Fe(III) Schiff base complex of the [FeL2(NO3)2]NO3 type where L = 2-((pyridin-4-yl)methyleneamino)-3-aminomaleonitrile was synthesized using the reflux and sonochemical methods and their antibacterial and antifungal activity were evaluated. The nanoparticles of iron oxide (Fe2O3) were obtained from the iron nano-structure complex as a precursor after calcination at 600 ˚C for 3 h. All the synthesized compounds were characterized by various spectroscopic techniques. The results of SEM showed that the morphology of iron nano-structure complex was rod-like while the morphology of the Fe2O3 nano powder was spherical. The results of the biological studies indicated that the iron nano-structure complex showed a stronger antibacterial and antifungal efficiency than its bulk complex. Finally, the empirical geometrical parameters of complexes revealed a good agreement with calculated ones at DFT-B3LYP level.  相似文献

Novel p-tolylimido rhenium(V) complexes trans-[Re(p-NC6H4CH3)X2(quin-2-COO)(PPh3)] and cis-[Re(p-NC6H4CH3)X2(quin-2-COO)(PPh3)]·MeCN have been obtained in the reactions of [Re(p-NC6H4CH3)X3(PPh3)2] (X = Cl, Br) with quinoline-2-carboxylic acid. The compounds were identified by elemental analysis IR, UV-Vis spectroscopy and X-ray crystallography. The electronic structures of trans- and cis-halide isomers of [Re(p-NC6H4CH3)Cl2(quin-2-COO)(PPh3)] have been calculated with the density functional theory (DFT) method. Additional information about binding in the compounds [Re(p-NC6H4CH3)Cl2(quin-2-COO)(PPh3)] with cis- and trans-halide arrangement has been obtained by NBO analysis. The electronic spectra of trans and cis isomers of [Re(p-NC6H4CH3)Cl2(quin-2-COO)(PPh3)] were investigated at the TDDFT level employing B3LYP functional in combination with LANL2DZ.  相似文献

Two novel dinuclear nickel(II) complexes [Ni2(ntb)2(μ-tp)(H2O)1.61(CH3OH)0.39](NO3)2·5.13CH3OH·2.25H2O (1) and [Ni2(ntb)2(μ-fum)(H2O)(CH3OH)](NO3)2·6CH3OH·H2O (2) (tp = terephthalate dianion, fum = fumarate dianion, ntb = tris(2-benzimidazolylmethyl)amine) containing tetradentate poly-benzimidazole ligand were synthesized and structurally characterized by IR spectra, UV-Vis, elemental analysis and X-ray crystallography. The Ni(II) ions in 1 and 2 have distorted octahedral geometry with four nitrogen atoms of ntb, one oxygen atom of water and one oxygen atom supplied by the carboxylate group of the bridged dicarboxylato ligand. Complexes 1 and 2 consist of terephthalato- and fumarato-bridged dinickel(II) centers in bis(monodentate) bonding fashion. The Ni?Ni distances are 11.333 Å for 1 and 8.966 Å for 2. The magnetic susceptibility measurements at variable temperature show that two complexes exhibit weak antiferromagnetic interactions between nickel(II) ions with J values of −0.25 cm−1 and −0.36 cm−1, respectively.  相似文献

The synthesis and characterization of several complexes of the composition [{M(terpy)}n(L)](ClO4)m (M = Pt, Pd; L = 1-methylimidazole, 1-methyltetrazole, 1-methyltetrazolate; terpy = 2,2′:6′,2″-terpyridine; n = 1, 2; m = 1, 2, 3) is reported and their applicability in terms of a metal-mediated base pair investigated. Reaction of [M(terpy)(H2O)]2+ with 1-methylimidazole leads to [M(terpy)(1-methylimidazole)](ClO4)2 (1: M = Pt; 2: M = Pd). The analogous reaction of [Pt(terpy)(H2O)]2+ with 1-methyltetrazole leads to the organometallic compound [Pt(terpy)(1-methyltetrazolate)]ClO4 (3) in which the aromatic tetrazole proton has been substituted by the platinum moiety. For both platinum(II) and palladium(II), doubly metalated complexes [{M(terpy)}2(1-methyltetrazolate)](ClO4)3 (4: M = Pt; 5: M = Pd) can also be obtained depending on the reaction conditions. In the latter two compounds, the [M(terpy)]2+ moieties are coordinated via C5 and N4. X-ray crystal structures of 1, 2, and 3 are reported. In addition, DFT calculations have been carried out to determine the energy difference between fully planar [Pd(mterpy)(L)]2+ complexes Ip-IVp (mterpy = 4′-methyl-2,2′:6′,2″-terpyridine; L = 1-methylimidazole-N3 (I), 1-methyl-1,2,4-triazole-N4 (II), 1-methyltetrazole-N3 (III), or 3-methylpyridine-N1 (IV)) and the respective geometry-optimized structures Io-IVo. Whereas this energy difference is larger than 70 kJ mol−1 for compounds I, II, and IV, it amounts to only 0.8 kJ mol−1 for the tetrazole-containing complex III, which is stabilized by two intramolecular C-H?N hydrogen bonds. Of all complexes under investigation, only the terpyridine-metal ion-tetrazole system with N3-coordinated tetrazole appears to be suited for an application in terms of a metal-mediated base pair in a metal-modified oligonucleotide.  相似文献

Three platinum-chloroquine complexes, trans-Pt(CQDP)2(I)2 [1], trans-Pt(CQDP)2(Cl)2 [2] and trans-Pt(CQ)2(Cl)2 [3], were prepared and their most probable structure was established through a combination of spectroscopic analysis and density functional theory (DFT) calculations. Their interaction with DNA was studied and their activity against 6 tumor cell lines was evaluated. Compounds 1 and 2 interact with DNA primarily through electrostatic contacts and hydrogen bonding, with a minor contribution of a covalent interaction, while compound 3 binds to DNA predominantly in a covalent fashion, with weaker secondary electrostatic interactions and possibly hydrogen bonding, this complex also exerted greater cytotoxic activity against the tumor cell lines.  相似文献

BackgroundSchiff base metal complexes are considered promising chemotherapeutic agents due to their potential application in cancer therapy.MethodsThe current work sought to synthesize a brand-new Schiff base ligand obtained from 2-hydroxybenzohydrazide and (E)− 1-(2-(p-tolyl)hydrazono)propan-2-one with metal ions which included Pd(II) and Zn(II) ions. Elemental analyses, FT-IR, mass spectra, 1H NMR, UV-Vis spectrometer, and computational analysis characterized the compound's structure. In vitro, the breast cancer cell line (MCF-7) was tested for its sensitivity to Schiff base (HL) and its Pd(II) and Zn(II) complexes. The half-maximal inhibitory concentration IC50 of the compounds was determined and used to perform the comet assay, which was carried out to reveal the photo-induced DNA damaging ability of the compounds of individual cells. Moreover, the compounds' effects on antioxidant defense systems of enzymes in cells: superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activities and oxidant Malondialdehyde (MDA) were examined in MCF-7 cells.ResultsThe Pd(II) complex displayed approximately the same IC50 as Cisplatin, while Zn(II) complex had better activity than Cisplatin with very low IC50, 1.40 μg/ml. Significant alterations in SOD, CAT, GPx, and MDA production were discovered, inducing oxidative stress, enlarging ROS production, and reducing the antioxidant amount. This change was approximately similar in most compounds. Consequently, it promoted apoptosis, particularly the Zn(II) complex, which demonstrated an improved impact because of its ability to influence the antioxidant defense systems of enzymes, mostly SOD and GPx, besides increasing MDA levels.ConclusionIt can be concluded that Zn(II) complex is the most effective anticancer drug since it induced a very similar genotoxic effect as Cisplatin and has a very low IC50 value.  相似文献

Rhodium(II) complexes [Rh2(μ-OAc)2(OAc)(bpy)(H2O)2]PF6 (1), [Rh2(μ-OAc)2(OAc)(phen)(H2O)2](PF6)·H2O (2), [Rh2(μ-OOCCH3)3(OOCCH3)(phen)] (3) and [Rh2(μ-O2CCH3)3(O2CCH3)(Ph2phen)] (4) (Ph2phen = 4,7-diphenyl-1,10-phenanthroline) have been synthesized and characterized by means of NMR, IR and UV-Vis spectroscopic methods. X-ray structure of complex 4·1.5(CH3COCH3) has been determined and its geometry and electronic structure has been elucidated using OPBE and B3LYP DFT methods. New Pd(II) and Pt(II) complexes [ML2] (HL=a substituted 2,5-dihydro-5-oxo-1H-pyrazolone-1-carbothioamide) have been synthesized by reacting K2MCl4 (M=Pd, Pt) or Pd(OAc)2 with beta-ketoester thiosemicarbazones. The structures of seven of these complexes were determined by X-ray diffraction. Although all exhibit a distorted square-planar coordination with trans- or (in one case) cis-[MN2S2] kernels, their supramolecular arrangements vary widely from isolated molecules to 3D-networks. The in vitro antitumoral assays performed with two HL ligands and their metal complexes showed significant cytostatic activity for the latter, with the most active [ML2] derivative (a palladium complex) being about sixteen times more active than cis-DDP against the cisplatinum-resistant cell line A2780cisR.  相似文献

Mono(trifluoroacetato)platinum(IV) complexes of the formula [PtIV(dach)L3(TFAc)] (dach = trans-(±)-1,2-diaminocyclohexane, TFAc = trifluoroacetate, L = acetate or propionate) could be prepared from the reaction of [PtIV(dach)L3(OH)] with trifluoroacetic anhydride in the presence of a base and the crystal structure of compound 4 was determined by X-ray crystallography. In vitro antitumor activity of complex 4 (ED50 = 3.1 μM) was found to be much higher than carboplatin (ED50 = 10.2 μM).  相似文献

Two new complexes of platinum(II) and silver(I) with acesulfame were synthesized. Acesulfame is in the anionic form acesulfamate (ace). The structures of both complexes were determined by X-ray crystallography. For K2[PtCl2(ace)2] the platinum atom is coordinated to two Cl and two N-acesulfamate atoms forming a trans-square planar geometry. Each K+ ion interacts with two oxygen atoms of the S(O)2 group of each acesulfamate. For the polymeric complex [Ag(ace)]n the water molecule bridges between two crystallographic equivalent Ag1 atoms which are related each other by a twofold symmetry axis. Two Ag1 atoms, related to each other by a symmetry centre, make bond contact with two equivalent oxygen atoms. These bonds give rise to infinite chains along the unit cell diagonal in the ac plane. The in vitro cytotoxic analyses for the platinum complex using HeLa (human cervix cancer) cells show its low activity when compared to the vehicle-treated cells. The Ag(I) complex submitted to in vitro antimycobacterial tests, using the Microplate Alamar Blue (MABA) method, showed a good activity against Mycobacterium tuberculosis, responsible for tuberculosis, with a minimal inhibitory concentration (MIC) value of 11.6 μM. The Ag(I) complex also presented a promising activity against Gram negative (Escherichia coli and Pseudomonas aeruginosa) and Gram positive (Enterococcus faecalis) microorganisms. The complex K2[PtCl2(ace)2] was also evaluated for antiviral properties against dengue virus type 2 (New Guinea C strain) in Vero cells and showed a good inhibition of dengue virus type 2 (New Guinea C strain) replication at 200 μM, when compared to vehicle-treated cells.  相似文献

Reactions of 2-(3,5-dimethylpyrazol-1-ylmethyl)pyridine (L1), 2-(3,5-diphenylpyrazol-1-ylmethyl)pyridine (L2), 2-(3,5-di-tert-butylpyrazol-1-ylmethyl)pyridine (L3) and 2-(3-p-tolylpyrazol-1-ylmethyl)pyridine (L4) with K2[PtCl4] in a mixture of ethanol and water formed the dichloro platinum complexes [PtCl2(L1)] (1), [PtCl2(L2)] (2), [PtCl2(L3)] (3) and [PtCl2(L4)] (4). Complex 1, [PtCl2(L1)], could also be prepared in a mixture of acetone and water. Performing the reactions of L2 and L3 in a mixture of acetone and water, however, led to C-H activation of acetone under mild conditions to form the neutral acetonyl complexes [Pt(CH2COCH3)Cl(L2)] (2a) and [Pt(CH2COCH3)Cl(L3)] (3a). The same ligands reacted with HAuCl4 · 4H2O in a mixture of ethanol and water to form the gold salts [AuCl2(L1)][AuCl4] (5) [AuCl2(L2)][Cl] (6) [AuCl2(L3)][Cl] (7) and [AuCl2(L4)][AuCl4] (8); however, with the pyrazolyl unit in the para position of the pyridinyl ring in 4-(3,5-dimethylpyrazol-1-ylmethyl)pyridine (L5), 4-(3,5-diphenylpyrazol-1-ylmethyl)pyridine (L6) neutral gold complexes [AuCl3(L5)] (9) and [AuCl2(L6)] (10) were formed; signifying the role the position of the pyrazolyl group plays in product formation in the gold reactions. X-ray crystallographic structural determination of L6, 2, 33a, 8 and 10 were very important in confirming the structures of these compounds; particularly for 3a and 8 where the presence of the acetonyl group confirmed C-H activation and for 8 where the counter ion is . Cytotoxicity studies of L2, L4 and complexes 1-10 against HeLa cells showed the Au complexes were much less active than the Pt complexes.  相似文献

Diorganotin(IV) complexes of N-acetyl-l-cysteine (H2NAC; (R)-2-acetamido-3-sulfanylpropanoic acid) have been synthesized and their solid and solution-phase structural configurations investigated by FTIR, Mössbauer, 1H, 13C and 119Sn NMR spectroscopy. FTIR results suggested that in R2Sn(IV)NAC (R = Me, Bu, Ph) complexes NAC2− behaves as dianionic tridentate ligand coordinating the tin(IV) atom, through ester-type carboxylate, acetate carbonyl oxygen atom and the deprotonated thiolate group. From 119Sn Mössbauer spectroscopy it could be inferred that the tin atom is pentacoordinated, with equatorial R2Sn(IV) trigonal bipyramidal configuration. In DMSO-d6 solution, NMR spectroscopic data showed the coordination of one solvent molecule to tin atom, while the coordination mode of the ligand through the ester-type carboxylate and the deprotonated thiolate group was retained in solution. DFT (Density Functional Theory) study confirmed the proposed structures in solution phase as well as the determination of the most probable stable ring conformation. Biological investigations showed that Bu2SnCl2 and NAC2 induce loss of viability in HCC cells and only moderate effects in non-tumor Chang liver cells. NAC2 showed lower cytotoxic activity than Bu2SnCl2, suggesting that the binding with NAC2− modulates the marked cytotoxic activity exerted by Bu2SnCl2. Therefore, these novel butyl derivatives could represent a new class of anticancer drugs.  相似文献

Ruthenium complexes [Ru(mpy)2(DMSO)2] (1) and [Ru(mbtz)2(DMSO)2] (2) containing 2-mercaptopyridine (mpy) and 2-mercaptobenzothiazole (mbtz) have been synthesized. Reactivity of 1 have been examined with 2,2′-bipyridine (bipy), 1,10-phenanthroline (phen), EPh3 (E = P, As) and 1,2-bis(diphenylphosphino)-methane (dppm). It reacted with bipy or phen in DMF to afford [Ru(mpy)2(bipy)] (3) and [Ru(mpy)2(phen)] (4) while, its reaction with EPh3 or dppm in common organic solvents failed to afford products containing EPh3 or dppm. Complexes under investigation have been characterized by elemental analyses, spectral, electrochemical studies and structures of 1-4 have been determined crystallographically. Density functional theory calculations have been performed on 1-4 and the model complex [Ru(mpy)(PMe3)2] (5) using exchange correlation functionals BP86. Optimized bond length and angles are in good agreement with the structural data. The Ru-N and Ru-S bond distances in [Ru(mpy)2]-moiety of 1 are relatively shorter than 5, indicating higher stability of 1 in comparison to 5. The WBI values of Ru-N1, Ru-N2, Ru-S1 and Ru-S2 bonds indicate Ru-mpy bonding trend as 3 > 4 > 1 > 5. There is an overall charge flow in the direction L → [Ru(mpy)2] (L = DMSO, bipy, phen and PMe3). Due to greater ionic character and Pauli repulsive interactions for Ru-PMe3 bond in comparison to Ru-DMSO, the DMSO ligands in 1 may not be substituted by phosphine ligands experimentally.  相似文献

Five new complexes of Pt(II), Pd(II), Co(III) and Ni(II) with 2-pyridine(quinoline)carboxaldehyde selenosemicarbazones were synthesized and characterized. Crystal structures of Pt(II) complex with the pyridine derivative and Co(III) complex with the quinoline derivative were determined. In all complexes the ligands were coordinated through N2Se donor atom set forming either square-planar (Pt, Pd) or octahedral (Co, Ni) geometry. All complexes showed biological activity.  相似文献

A new series of glucose-conjugated Pt(IV) complexes that target tumor-specific glucose transporters (GLUTs) was designed, synthesized, and evaluated for their anticancer activities. All six compounds, namely, A1-A6, exhibited increased cytotoxicity that were almost six fold higher than that of oxaliplatin to MCF-7 cells. These Pt(IV) complexes can be reduced to release Pt(II) complexes and cause the death of tumor cells. Simultaneously, the glycosylated Pt(IV) complexes (30.21–91.33?μM) showed lower cytotoxicity that normal LO2 cells compared with cisplatin (5.25?μM) and oxaliplatin (8.34?μM). The intervention of phlorizin as a GLUTs inhibitor increased the IC50 value of the glycosylated Pt(IV) complexes, thereby indicating the potential GLUT transportability. The introduction of glucose moiety to Pt(IV) complexes can effectively enhance the Pt cellular uptake and DNA platination. Results suggested glucose-conjugated Pt(IV) complexes had potential for further study as new anticancer agents.  相似文献

The benzimidazole derivative Schiff bases and their copper(II) (Cu(II)) mixed-polypyridyl complexes (1-4) have been synthesized and characterized by the spectral and analytical techniques. DNA binding/cleavage studies indicate a stronger binding capability for the complex 4 which is confirmed by the absorbance, viscometric and gel-electrophoresis studies. The photocleavage of plasmid pBR322 DNA reveals that hydroxyl radical (OH(?)) and singlet oxygen ((1)O(2)) are likely to be the reactive species. Analysis of the growth activity shows that the antimicrobial effect of these Schiff bases on Gram-negative bacteria is higher than that on Gram-positive. Furthermore, the complexes having nitro group show an increased antimicrobial effect.  相似文献

A series of octahedral Ru(II) polypyridyl complexes, [Ru(phen)(2)L](2+) (L=R-PIP and PIP=2-phenylimidazo[4,5-f][1,10]phenanthroline) were synthesized and characterized by elementary analysis, (1)H NMR and ES-MS, as well as UV-visible spectra and emission spectra. The antitumor activities of these complexes and their corresponding ligands were investigated against mouse leukemia L1210 cells, human oral epidermoid carcinoma KB cells, human promyelocytic leukemia cells (HL-60) and Bel-7402 liver cancer cells by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. It was found that the complexes [Ru(phen)(2)L](2+) (L=R-PIP) exert rather potent activities against all of these cell lines, especially for the KB cells (IC(50)=4.7+/-1.3 microM). The binding affinities of these Ru(II) complexes to CT-DNA (calf thymus DNA), as well as the DNA-unwinding properties on supercoiled pBR322 DNA were also investigated. The results showed that these Ru(II) polypyridyl complexes not only had an excellent DNA-binding property but also possessed a highly effective DNA-photocleavage ability.
