Summary Genetic traits which are disadvantageous or deleterious in primitive cultures, of hunters and food gatherers, have very low frequencies in almost all cases. There are a few special exceptions, such as the sickle cell trait. Some deleterious traits under genetic control are rare in primitive populations but have fairly high frequencies in populations with long histories or prehistories of civilization, such as Chinese, Japanese or European populations. The Darwinist might suggest that these traits have less selective disadvantage, that is, lower rates of selective elimination, in civilized habitats; i.e. that the various changes in habitat accompanying civilization have caused the rigor of natural selection to relax, in part or perhaps completely.Several cases are discussed of traits under genetic control or genetic influence which demonstrate this phenomenal contrast, between low frequencies among primitive populations and high frequencies among civilized populations. Population data are presented for 4 such cases: colorblindness, myopia, abnormality of the nasal septum and abnormally small external opening of the tear duct.
Zusammenfassung Mit einigen bekannten Ausnahmen, z. B. Sichelzellanämie, kommen genetische Merkmale, die bei den Urvölkern von Jägern und Sammlern ungünstig oder sogar schädlich wirkten, mit niedriger Häufigkeit vor. Genetisch bedingte schädliche Merkmale, die selten unter Urvölkern gefunden werden, kommen jedoch häufiger vor bei Bevölkerungen, die eine lange Geschichte bzw. Vorgeschichte der Zivilisation besitzen, wie z. B. in China, Japan oder Europa. Ein Darwinist würde behaupten, daß bei zivilisierten Wohnorten solche Merkmale selektiv weniger nachteilig wirken, d. h., daß die selektive Ausscheidung langsamer vorgeht. Anders gesagt: der mit der Zivilisation verbundene Wechsel des Wohnortes bildet einen Grund für eine Milderung der Strenge der natürlichen Auslese.Es werden einige genetisch bestimmte oder genetisch beeinflußte Merkmale besprochen, die den Unterschied zwischen den Häufigkeiten (niedrig bei Urvölkern, hoch bei zivilisierten Völkern) aufzeigen,. Für 4 solcher Fälle: Farbensinnstörungen, Kurzsichtigkeit, Nasenscheidenwand-Deformitäten und anomale kleine Außenöffnungen der Tränenwege werden die Befunde aus den Bevölkerungen dargestellt.

Summary The gene for -CGTase from an alkalophilic bacterium, Bacillus sp. #1011, was cloned in an Escherichia coli phage D69 and recloned in an E. coli plasmid pBR322 and a B. subtilis plasmid pUB110. An E. coli recombinant plasmid pTUE202 and a B. subtilis plasmid pTUB703 were selected from ten plasmids, because the transformants by each of the two plasmids produced the highest amount of extracellular -CGTase in each strain. The plasmids were stably maintained and expressed in each bacterial strain. A common DNA region of approximately 2.5 kb was defined in the ten plasmids, and the enzymatic activity was lost when a part of the common region was deleted. The major product of hydrolysis from starch by the -CGTases of E. coli [pTUB202] and B. subtilis [pTUB703] was -CD as in the case of the enzyme of the parental Bacillus sp. #1011.Abbreviations -CGTase -cyclodextrin synthetase - -CD -cyclodextrin - -CD -cyclodextrin - -CD -cyclodextrin - [] designates plasmid-carrier state  相似文献

The probit technique for analysis of subjective assessments of thermal sensation is described. It enables transition temperatures from any selected thermal sensation to the adjacent thermal sensation (e.g. from neutral to warm) to be identified. A transition temperature is defined as that temperature at which the maximum number of people would change their assessment from one thermal sensation to the next. Thus if a seven-point scale of thermal sensation is used, six transition temperatures are possible. Increments between them will not necessarily be identical, as would be assumed in a linear regression analysis. The method has been applied in three studies: laboratory studies by the Kansas State University, field studies in Port Moresby, and field studies in Melbourne. In the first it is shown that men are more thermally tolerant than women, in the second it is shown that in Port Moresby the preferred temperatures of Melanesians are 2°C higher than those of Caucasians and in the third it is suggested that subjects in Melbourne have a slightly lower preferred temperature than predicted by Fanger (1972).  相似文献

Summary The mutagenic action of mitomycin C (MMC) on the chromosomes of two reconstructed karyotypes of barley was studied. MMC-induced chromatid aberrations were found to be distributed non-randomly along the chromosomes. The regions situated next to the secondary constrictions of chromosomes 6 and 7 appeared to be clearly pronounced aberration hot spots. In these segments, intercalary deletions and duplication-deletions were the most frequently induced aberration types. The comparative analysis of the frequency and localization of MMC-induced aberrations in the chromosomes of the two karyotype variants, which differ from each other by the position of the hot spot segments, provided new evidence about the influence of the segment transposition on the hot spot expressivity. The most remarkable finding obtained in the study is that the size of the segment involved in both intercalary deletions and duplication-deletions proved to be strongly affected by the structural peculiarity of the reconstructed chromosome. The possible reasons underlying this finding are discussed.  相似文献

Synopsis 3- and 17-hydroxysteroid dehydrogenase activities were studied histochemically in the male reproductive tract of the domestic fowl. 3-hydroxysteroid dehydrogenase was NAD+-linked and was capable of metabolizing the three substrates used, namely, pregnenolone, 17-hydroxypregnenolone and dehydroepiandrosterone. 17-hydroxysteroid dehydrogenase oxidized testosterone in the presence of NAD+ or NADP+. The pattern of distribution of formazan granules was essentially the same with all the substrates used, and they were located in the Leydig cells, seminiferous tubules and the lining epithelia of the entire excurrent duct system of the testis except the rete testis. The activity of both enzymes appeared to be highest in the ductuli efferentes and decreased distally along the tract. The evidence suggests that steroid synthesis may occur in the epithelial lining of the excurrent ducts as well as in the cells of the testis.  相似文献

In most individuals two HLA-DR genes are expressed from each chromosome. One of these genes encodes one of the classical DR specificities, while the other encodes either of the supertypic DRw52/DRw53 specificities. In addition to these genes usually one or two DR pseudogenes are present. In contrast, the DRw8 chromosomal region only contains a single DR gene. To determine the relationship of this single gene to the multiple DR genes of other DR specificities, comparisons of Southern genomic blots were carried out. In this analysis genomic clones for each individual DR chain locus were included. The DR w8 gene was indistinguishable from the DR III gene of DR3 cells (encoding DRw52), suggesting that it is closely related to the latter gene. The functional implications of this finding are discussed.  相似文献

NADH coenzyme Q reductase (EC 1.6.5.3) has been suggested in the literature to be inactivated by ischaemia. In the present study, NADH coenzyme Q reductase activity was localized in unfixed cryostat sections of ischaemic rat livers and quantified using image analysis. In vitro ischaemia was induced by storage of rat liver fragments for 30, 60, and 120min at 37°C. In vivo ischaemia was provoked by clamping the afferent vessels of median and left lateral liver lobes for 60min followed by 30, 60 and 180min of reperfusion. NADH coenzyme Q reductase activity was demonstrated with the tetrazolium salt method in the presence of polyvinyl alcohol. Final reaction product was found in liver parenchymal cells and its distribution was homogeneous within liver lobules. Only low amounts of final reaction product were formed when the incubation was performed in the absence of the substrate NADH. A non-linear relation was found between the absorbance and incubation time when the reaction was performed in the presence of NADH. Therefore, the initial velocity was taken as the true rate of enzyme activity. A linear relationship was found for the initial velocity and section thickness up to 6µm followed by a levelling off. Electron microscopically, NADH coenzyme Q reductase activity was localized at the outer and inner membranes of mitochondria. In vitro ischaemia up to 120min did not affect NADH coenzyme Q reductase activity. At 30min reperfusion after in vivo ischaemia for 60min enzyme activity was slightly decreased in certain foci which also showed diminished lactate dehydrogenase activity. A further decrease of enzyme activities in foci was observed at 180min reperfusion after ischaemia. It is concluded that NADH coenzyme Q reductase activity is not sensitive to ischaemia. Furthermore, it is likely that the enzyme leaks from liver parenchymal cells into the circulation during reperfusion after ischaemia.  相似文献

The deconvolution method, i.e., reconstruction of noise-free discrete amplitude distributions of excitatory postsynaptic potentials (EPSPs), has been tested in computer experiments. A generalized quantal model without constraints on distribution of transmitter release probabilities (p) was used. Complex binomial amplitude distributions with a number of discrete components n between 2 and 10 and with different p were simulated. The distance between the discrete components (i.e., the quantum value, v) was determined from simulated distributions with the number of amplitudes N equal to 1000 or 100. For samples with N=100, mean v values were calculated from 10 distributions. The solutions obtained by deconvolution were close to (within ±20% of) the simulated v values when the standard deviation of the noise Sn0.5v. For Sn=0.5–2.0v, v grew in proportion to Sn and was overestimated. A similar correlation was found for v calculated from amplitudes of minimal EPSPs recorded from CA1 area neurons of guinea pig hippocampal slices. However, no significant correlation between v and Sn was found for v>135 µV, i.e., for estimates exceeding the total mean value. A substantial increase in the mean quantal content (m) with a relatively slight increase in v during longterm potentiation (LTP) was confirmed for those cases. Methods of elimination of erroneous v estimates were considered. Rejection of cases with v>2.5Sn (when N=1000) or v>3Sn (when N=100) eliminated erroneous estimates with a high probability but demanded very low noise levels. A new criterion for recognition of correct estimates is suggested. It is based on artificial contamination of the measured amplitudes by the Gaussian noise (noise addition) and on the double-step dependence of v on Sn described above. The kinetics of P680+ reduction in oxygen-evolving spinach Photosystem II (PS II) core particles were studied using both repetitive and single-flash 830 nm transient absorption. From measurements on samples in which PS II turnover is blocked, we estimate radical-pair lifetimes of 2 ns and 19 ns. Nanosecond single-flash measurements indicate decay times of 7 ns, 40 ns and 95 ns. Both the longer 40 ns and 95 ns components relate to the normal S-state controlled Yz P680+ electron transfer dynamics. Our analysis indicates the existence of a 7 ns component which provides evidence for an additional process associated with modified interactions involving the water-splitting catalytic site. Corresponding microsecond measurements show decay times of 4 s and 90 s with the possibility of a small component with a decay time of 20–40 s. The precise origin of the 4 s component remains uncertain but appears to be associated with the water-splitting center or its binding site while the 90 s component is assigned to P680+-QA recombination. Weed assemblages from late medieval cornfields have been studied for the first time in northern Switzerland. Eleven samples from at least two different grain stores were investigated. The samples were collected from the carbonised remains of six wooden houses built in the late 13th century A.D. and which burnt down in the middle of the 15th century. The weed floras found in the spelt (Triticum spelta) and oats (Avena sativa) indicate a high botanical diversity in the cornfields at harvest time. Although oats are normally a summer crop and spelt a winter crop, both summer and winter crop weeds (as well as many different present-day grassland taxa) were found in each type of grain. Most of the weeds found were perennial plants, which was interpreted as an indication of both extensive tillage of the arable land and application of the three-field rotation system (Dreifelderwirtschaft). The spectra of the two palaeophytocoenoses (assemblages of ancient plant remains) studied suggest that the phytosociological method may not be reliable for classification of the late medieval remains into summer and winter crop weed communities. These findings should provide a better understanding of the development of anthropogenic plant communities, and in particular, the development of crop weed communities.  相似文献

Growth at 25°C and pH 5.50 favour the production of-glucosidase. De-fatted oilseed flour and Tween 80 enhanced the production of-glucosidase, Lactose, gentibiose, gentibiose-acetate, laminarabiose and xylobiose induced-glucosidase activity. Precipitation of the culture filtrate with (NH4)2SO4 resulted in 26-fold purification with 67% recovery. The optimum pH and temperature for activity were 5.0 to 5.4 and 55°C respectively. The enzyme was stable at 40°C with half-life at 12 h at 50°C. TheK m andV max for the hydrolysis ofp-nitrophenyl--d-glucoside at 40°C H 5.0 are 0.28mm and 0.60 U/mg protein, respectively.  相似文献

The receptive fields of complex neurons within area 18 of the cerebral cortex of the cat were determined by a computer-assisted method using a moving light bar substantially shorter than the long diameter of the receptive field as a visual stimulus. The visual cells repeatedly generated nerve impulses when the stimulus crossed well-defined active points within their receptive fields. Outside of these active points, the cells remained silent. It is suggested that the receptive fields are formed by a discontinuous accumulation of such active points. When the electrical activities of two neighbouring visual neurons are recorded simultaneously, their active points do not coincide. In addition, some active points were located outside the most prominent excitatory part of the receptive field of the studied cells. Individual visual cells typically differ in the number and distribution of active points. Since these cells best respond to a stimulus moving in a certain direction, it is suggested that they may act as direction of movement and/or velocity detectors. Alternate firing of a number of neighboring cells connected to a distributed pattern of peripheral receptors may form a system which is able to code for velocity and direction of the moving stimulus.  相似文献

The synthesis, characterization, thermal behavior and magnetic properties of a number of bis[2-(4-alkyloxyphenyl)malondialdehyde] nickel(II) complexes are reported. X-ray studies of the nickel(II) complexes were performed. It was found that the nickel(II) complexes show liquid-crystalline smectic A phases over a broad temperature range with low melting points. The temperature-dependent magnetic susceptibility measurements of the bis[2-(4-decyloxyphenyl)malondialdehyde] nickel(II) complex were carried out in the range of 4.2–480 K. The temperature dependent magnetic susceptibilities and magnetic moments of this compound (eff=3.27 B at 300 K) indicate that the nickel centers are octahedrally coordinated. Models for the molecular arrangement in the crystalline and liquid-crystalline phases are discussed on the basis of the magnetic data. In spite of the oxygen bridge between the nickel centers, no exchange interactions were found in the crystalline and liquid-crystalline phases.  相似文献

Summary The development of the sessile pseudoheliozoan Clathrulina elegans Cienkowski (Desmothoraca, Rhizopoda, Protozoa) was followed by light and electron microscopy. The cell cycle comprises a heliozoan-like stage sitting in an organic perforated capsule, an amoeboid resp. biflagellate stage ensuring dispersal, and an encysted stage. Special reference is given to the morphogenetic processes during the flagellate-to-heliozoan transformation. The differentiation of the stalk is described in detail and a hypothesis of the morphogenesis of the capsule is presented. Granules in the filopods displaying a jerky movement, are identified as a hitherto underscribed type of haptocyst-like organelles. These kinetocysts are shown to be a useful marker for studying the turnover of the plasma membrane at organelle level. Evidence is presented that in the flagellate stage a surplus of plasma membrane is stored in numerous lacunae thus facilitating the rapid transformation into the heliozoan stage with its comparatively large surface area. The morphogenetic significance of microtubules during the formation of the stalk, the latter being an asymmetric extracellular secretion product, is elucidated. The role of microtubule nucleation sites, the precisely timed assembly and disassembly of microtubules and other factors which probably control development are discussed to some extent.The author thanks Mrs. G. Summary Sporulation of S. cerevisiae MAT/MATa was accompanied by a novel pattern of protein synthesis as shown by the disappearance of some mitotic polypeptides and by the appearance of a new set of meiotic polypeptides. Inhibition of mitochondrial protein synthesis by erythromycin within the 1st h caused the disappearance of several meiotic polypeptides. These meiotic polypeptides were also sensitive to cycloheximide and were localized in the cytosol, demonstrating that they were not mitochondrial translational products. Since erythromycin affected neither protein synthesis nor sporulation in a mitochondrially inherited er y mutant, we conclude that mitochondrial protein synthesis is needed for the expression of some nuclear genes during sporulation.  相似文献

Summary Some simple formulae were obtained which enable us to estimate evolutionary distances in terms of the number of nucleotide substitutions (and, also, the evolutionary rates when the divergence times are known). In comparing a pair of nucleotide sequences, we distinguish two types of differences; if homologous sites are occupied by different nucleotide bases but both are purines or both pyrimidines, the difference is called type I (or transition type), while, if one of the two is a purine and the other is a pyrimidine, the difference is called type II (or transversion type). Letting P and Q be respectively the fractions of nucleotide sites showing type I and type II differences between two sequences compared, then the evolutionary distance per site is K = — (1/2) ln {(1 — 2P — Q) }. The evolutionary rate per year is then given by k = K/(2T), where T is the time since the divergence of the two sequences. If only the third codon positions are compared, the synonymous component of the evolutionary base substitutions per site is estimated by K'S = — (1/2) ln (1 — 2P — Q). Also, formulae for standard errors were obtained. Summary Myotendinous junctions in the myotomal tail muscles of the tadpole of Rana rugosa were examined by electron microscopy. At the site of the myotendinous junction, the sarcolemma is covered on its sarcoplasmic aspect by the connecting filament layer and the attachment layer, and on the extracellular aspect by the intermediary layer and the external lamina, with associated collagen fibrils. The intermediary layer consists of filamentous structures which closely resemble microfibrils (Hanak and Böck, 1971), spine-like or thread-like profiles (Korneliussen, 1973) and intermediary layer (Nakao, 1975a, b) in the myotendinous junctions of other vertebrate skeletal muscles.Particularly interesting is the fact that all the coverings and linings of the sarcolemma, including the external lamina, are completely absent in the terminal segment of the finger-like sarcolemmal invagination characteristic of the myotendinous junction. Furthermore, special types of coupling between a sac of sarcoplasmic reticulum and a part of the sarcolemmal invagination are frequently observed. These couplings always occur along the region of the sarcolemma where the external lamina is absent. The couplings show features similar to those of the triad, such as SR feet , scalloped SR membranes and granular content of the SR sac, suggesting that they are analogous and functionally similar to the triad and other equivalent structures.  相似文献

Summary Heat-shock protein (HSP) gene expression in two wheat lines cv Mustang (heat-tolerant) and cv Sturdy (heat-susceptible) were analyzed to determine if wheat genotypes differing in heat tolerance also differ in in-vitro HSP synthesis (translatable HSP mRNAs) and steady-state levels of HSP mRNA. Several sets of mRNA were isolated from seedling leaf tissues which had been heat-stressed at 37 °C for various time intervals. These mRNAs were hybridized with HSP cDNA or genomic DNA probes (HSP17, 26, 70, 98, and ubiquitin). Protein profiles were compared using in-vitro translation and 2-D gels. The Northern slot-blot data from the heat-stress treatment provide evidence that the heat-tolerant cv Mustang synthesized low molecular weight (LMW) HSP mRNA earlier during exposure to heat shock and at a higher level than did the heat-susceptible cv Sturdy. This was especially true for the chloroplast-localized HSP. The protein profiles shown by 2-D gel analysis revealed that there were not only quantitative differences of individual HSPs between the two wheat lines, but also some unique HSPs which were only found in the Mustang HSP profiles. The high level of RFLP between the two wheat lines was revealed by Southern blot hybridization utilizing a HSP17 probe. These data provide a molecular basis for further genetic analysis of the role of HSP genes in thermal tolerance in wheat.  相似文献
