Angiogenin (Ang), one of the most potent angiogenic factor, is related with the growth and metastasis of numerous tumors. This paper presents a very simple and label-free square-wave voltammetry (SWV) aptasensor to detect angiogenin, in which an anti-angiogenin-aptamer was used as a molecular recognition element, and the couple ferro/ferricyanide as a redox probe. At the bare gold electrode, the redox couple (K4[Fe(CN)6]/K3[Fe(CN)6]) can be very easily accessed to the electrode surface to give a very strong SWV signal. At the anti-angiogenin/Au electrode surface, when angiogenin was added to the electrochemical cell, the binding of the analyte results in less availability for a redox reaction, which led to smaller SWV current. To quantify the amount of angiogenin, current suppressions of SWV peak were monitored using the redox couple of an [Fe(CN)6]4−/3− probe. The plot of signal suppression against the logarithm of angiogenin concentration is linear with over the range from 0.01 nM to 30 nM with a detection limit of 1 pM. The aptasensor also showed very good selectivity for angiogenin without being affected by the presence of other proteins in serum. It is the first time to use a very simple method to detect the cancer marker. Such an aptasensor opens a rapid, selective and sensitive route for angiogenin detection and provides a promising strategy for other protein detections.  相似文献

Individual and simultaneous determination of 50 nM uric acid (UA) and ascorbic acid (AA) using enlarged, citrate-stabilized gold nanoparticles (AuNPs) self-assembled to 2,5-dimercapto-1,3,4-thiadiazole (DMT) monolayer modified Au (Au/DMT) electrode by an amperometric method is described for the first time. Self-assembly of AuNPs on the electrode surface was confirmed by atomic force microscopy (AFM), attenuated total reflectance FT-IR and diffuse reflectance spectral measurements. The electron transfer reaction (ETR) of [Fe(CN)6]3−/4− was blocked at Au/DMT electrode, whereas it was restored with a peak separation of 200 mV after the attachment of AuNPs on the Au/DMT (Au/DMT/AuNPs) electrode, which was confirmed from the ETR of the [Fe(CN)6]3−/4− redox couple. When the self-assembled AuNPs were enlarged by hydroxylamine seeding, the ETR of [Fe(CN)6]3−/4− was improved significantly with a peak separation of 100 mV. Tapping mode AFM showed that the average size of the enlarged-AuNPs (E-AuNPs) was 50-70 nm. The E-AuNPs modified electrode catalyzes the oxidation of AA and UA, separates their voltammetric signals by 200 mV, and has excellent sensitivity towards AA and UA with a detection limit of 50 nM. The practical application of the modified electrode was demonstrated by measuring the concentration of UA in blood serum and urine.  相似文献

Electronic absorption and 8.0 T magnetic circular dichroism (MCD) spectra are reported for M(CN)8 4−, M=Mo(IV) and W(IV), in aqueous solution and M(CN)8 3−, M=Mo(V) and W(V), in acetonitrile solutions. In addition some absorption and MCD spectra are reported for the M(CN)8 3− ions embedded in thin poly methyl methacrylate (PMMA) plastic films at temperatures from 295 to 10 K. The temperature dependence of the MCD spectra confirms the presence of C terms. The solution and PMMA spectra for the both Mo and W complexes in either the IV or V oxidation states are remarkably similar to each other for the same oxidation state and are interpreted within a D2d structural framework for the isotropic environment. The weak bands below 3.0 μm−1 (1 μm−1=104 cm−1) for the M(IV) complexes are assigned as metal-localized ligand field (LF) transitions. LF transitions are also suggested for weaker unresolved absorption between 3.0 and 3.6 μm−1 for the M(V) ions. The intense bands above 3.6 μm−1 for M(IV) and 4.6 μm−1 for M(V) complexes are interpreted as metal to ligand charge transfer (MLCT) from the metal b1(x2y2) HOMO to CN-based π * orbitals. The prominent intense bands observed below 4.5 μm−1for the M(V) complexes are assigned as ligand to metal charge transfer (LMCT) from occupied non-bonding or weakly π bonding CN orbitals to the half-filled b1(x2y2) HOMO.  相似文献

Photoluminescence spectra of synthesized crystals of the mixed-metal heterobimetallic Ba{[Ag(CN)2]2−2x[Pt(CN)4]x} (x = 0 → 1) were recorded at 300, 150 and 77 K. Emission of the mixed-metal system containing both [Pt(CN)4]2− and [Ag(CN)2] ions shows an emission band due to Pt-Ag interaction. This new peak (424 nm) is not seen for pure Ba[Pt(CN)4] or Ba[Ag(CN)2]2. Theoretical DFT and extended Hückel molecular orbital population calculations shows that in heterometallic dimers, the HOMO correspond to dz2 of the metals with a weakly antibonding interaction between the moieties while the LUMO π*(CN) orbital shows a stronger bonding character between monomers. This indicate that the 424 nm emission peak corresponds to a MLCT (d → π*) transition with some metal to metal charge transfer character (Pt to Ag).  相似文献

Exposure of winter rye leaves grown at 20°C and an irradiance of either 50 or 250 μmol m−2 s−1 to high light stress (1600 μmol m−2 s−1, 4 h) at 5°C resulted in photoinhibition of PSI measured in vivo as a 34% and 31% decrease in ΔA820/A820 (P700+). The same effect was registered in plants grown at 5°C and 50 μmol m−2 s−1. This was accompanied by a parallel degradation of the PsaA/PsaB heterodimer, increase of the intersystem e pool size as well as inhibition of PSII photochemistry measured as Fv/Fm. Surprisingly, plants acclimated to high light (800 μmol m−2 s−1) or to 5°C and moderate light (250 μmol m−2 s−1) were fully resistant to photoinhibition of PSI and did not exhibit any measurable changes at the level of PSI heterodimer abundance and intersystem e pool size, although PSII photochemistry was reduced to 66% and 64% respectively. Thus, we show for the first time that PSI, unlike PSII, becomes completely resistant to photoinhibition when plants are acclimated to either 20°C/800 μmol m−2 s−1 or 5°C/250 μmol m−2 s−1 as a response to growth at elevated excitation pressure. The role of temperature/light dependent acclimation in the induction of selective tolerance to PSI photoinactivation is discussed.  相似文献

For the purpose of developing a direct label-free electrochemical detection system, we have systematically investigated the electrochemical signatures of each step in the preparation procedure, from a bare gold electrode to the hybridization of label-free complementary DNA, for the streptavidin-modified electrode. For the purpose of this investigation, we obtained the following pertinent data; cyclic voltammogram measurements, electrochemical impedance spectra and square wave voltammogram measurements, in Fe(CN)6 3−/Fe(CN)6 4− solution (which was utilized as the electron transfer redox mediator). The oligonucleotide molecules on the streptavidin-modified electrodes exhibited intrinsic redox activity in the ferrocyanide-mediated electrochemical measurements. Furthermore, the investigation of electrochemical electron transfer, according to the sequence of oligonucleotide molecules, was also undertaken. This work demonstrates that direct label-free oligonucleotide electrical recognition, based on biofunctional streptavidin-modified gold electrodes, could lead to the development of a new biosensor protocol for the expansion of rapid, cost-effective detection systems.  相似文献

A thermodynamic study of the inclusion process between 2-chlorobenzophenone (2ClBP) and cyclomaltoheptaose (β-cyclodextrin, β-CD) was performed using UV–vis spectroscopy, reversed-phase liquid chromatography (RP-HPLC), and molecular modeling (PM6). Spectrophotometric measurements in aqueous solutions were performed at different temperatures. The stoichiometry of the complex is 1:1 and its apparent formation constant (Kc) is 3846 M−1 at 30 °C. Temperature dependence of Kc values revealed that both enthalpy (ΔH° = −10.58 kJ/mol) and entropy changes (ΔS° = 33.76 J/K mol) are favorable for the inclusion process in an aqueous medium. Encapsulation was also investigated using RP-HPLC (C18 column) with different mobile-phase compositions, to which β-CD was added. The apparent formation constants in MeOH–H2O (KF) were dependent of the proportion of the mobile phase employed (50:50, 55:45, 60:40 and 65:35, v/v). The KF values were 419 M−1 (50% MeOH) and 166 M−1 (65% MeOH) at 30 °C. The thermodynamic parameters of the complex in an aqueous MeOH medium indicated that this process is largely driven by enthalpy change (ΔH° = −27.25 kJ/mol and ΔS° = −45.12 J/K mol). The results of the study carried out with the PM6 semiempirical method showed that the energetically most favorable structure for the formation of the complex is the ‘head up’ orientation.  相似文献

A detailed investigation on the oxidation of aqueous sulfite and aqueous potassium hexacyanoferrate(II) by the title complex ion has been carried out using the stopped-flow technique over the ranges, 0.01≤[S(IV)]T≤0.05 mol dm−3, 4.47≤pH≤5.12, and 24.9≤θ≤37.6 °C and at ionic strength 1.0 mol dm−3 (NaNO3) for aqueous sulfite and 0.01≤[Fe(CN)6 4−]≤0.11 mol dm−3, 4.54≤pH≤5.63, and 25.0≤θ≤35.3 °C and at ionic strength 1.0 or 3.0 mol dm−3 (NaNO3) for the hexacyanoferrate(II) ion. Both redox processes are dependent on pH and reductant concentration in a complex manner, that is, for the reaction with aqueous sulfite, kobs={(k1K1K2K3+k2K1K4[H+])[S(IV)]T]/([H+]2+K1[H+]+K1K2) and for the hexacyanoferrate(II) ion, kobs={(k1K3K4K5+k2K3K6[H+])[Fe(CN)6 4−]T)/([H+]2+K3[H+]+K3K4). At 25.0 °C, the value of k1′ (the composite of k1K3) is 0.77±0.07 mol−1 dm3 s−1, while the value of k2′ (the composite of k2K4) is (3.78±0.17)×10−2 mol−1 dm3 s−1 for aqueous sulfite. For the hexacyanoferrate(II) ion, k1′ (the composite of k1K5) is 1.13±0.01 mol−1 dm3 s−1, while the value of k2′ (the composite of k2K6) is 2.36±0.05 mol−1 dm3 s−1 at 25.0 °C. In both cases there was reduction of the cobalt(III) centre to cobalt(II), but there was no reduction of the molybdenum(VI) centre. k22, the self-exchange rate constant, for aqueous sulfite (as SO3 2−) was calculated to be 5.37×10−12 mol−1 dm3 s−1, while for Fe(CN)6 4−, it was calculated to be 1.10×109 mol−1 dm3 s−1 from the Marcus equations.  相似文献

The kinetics of electron transfer from cytochrome c2 to the primary donor (P) of the reaction center from the photosynthetic purple bacterium Rhodobacter sphaeroides have been investigated by time-resolved absorption spectroscopy. Rereduction of P+ induced by a laser pulse has been measured at temperatures from 300 K to 220 K in a series of specifically mutated reaction centers characterized by altered midpoint redox potentials of P+/P varying from 410 mV to 765 mV (as compared to 505 mV for wild type). Rate constants for first-order electron donation within preformed reaction center–cytochrome c2 complexes and for the bimolecular oxidation of free cytochrome c2 have been obtained by multiexponential deconvolution of the kinetics. At all temperatures the rate of the fastest intracomplex electron transfer increases by more than two orders of magnitude as the driving force −ΔG° is varied over a range of 350 meV. The temperature and ΔG° dependences of the rate constant fit the Marcus equation well. Global analysis yields a reorganization energy λ = 0.96 ± 0.07 eV and a set of electronic matrix elements, specific for each mutant, ranging from 1.2 10−4 eV to 2.5 10−4 eV. Analysis in terms of the Jortner equation indicates that the best fit is obtained in the classical limit and restricts the range of coupled vibrational modes to frequencies lower than ∼200 cm−1. An additional slower kinetic component of P+ reduction, attributed to electron transfer from cyt c2 docked in a nonoptimal configuration of the complex, displays a Marcus type dependence of the rate constant upon ΔG°, characterized by a similar value of λ (0.8 ± 0.1 eV) and by an average electronic matrix element smaller by more than one order of magnitude. In all of the mutants, as the temperature is decreased below 260 K, both intracomplex reactions are abruptly inhibited, their rate being negligible at 220 K. The free energy dependence of the second-order rate constant for oxidation of cyt c2 in solution suggests that the collisional reaction is partially diffusion controlled, reaching the diffusion limit at exothermicities between 150 and 250 meV over the temperature range investigated.  相似文献

Crystal structure of [ReO2(4-MeOpy)4][PF6] (4-MeOpy = 4-methoxypyridine) complex has been examined by the single crystal X-ray analytical method. This complex shows a trans-dioxo geometry (average Re-O bond length = 1.766(2) Å) and its equatorial plane is occupied by four 4-MeOpy molecules (average Re-N bond length = 2.156(4) Å). Electrochemical reaction of [ReO2(4-MeOpy)4]+ in CH3CN solution containing tetra-n-butylammonium perchlorate as a supporting electrolyte has been studied using cyclic voltammetry at 24 °C. Cyclic voltammograms show one redox couple around 0.65 V (Epa) and 0.58 V (Epc) [versus ferrocene/ferrocenium ion redox couple, (Fc/Fc+)]. Potential differences between two peaks (ΔEp) at scan rates in the range from 0.01 to 0.10 V s−1 are 65 mV, which is almost consistent with the theoretical ΔEp value (59 mV) for the reversible one electron transfer reaction at 24 °C. The ratio of anodic peak currents to cathodic ones is 1.04 ± 0.03 and the (Epa + Epc)/2 value is constant, 0.613 ± 0.001 V versus Fc/Fc+, regardless of the scan rate. Spectroelectrochemical experiments have also been carried out by applying potentials from 0.40 to 0.77 V versus Fc/Fc+ with an optically transparent thin layer electrode. It was found that the UV-visible absorption spectra show clear isosbestic points at 228, 276, and 384 nm, and that the electron stoichiometry is evaluated as 1.03 from the Nernstian plot. These results indicate that the [ReO2(4-MeOpy)4]+ complex is oxidized reversibly to the [ReO2(4-MeOpy)4]2+ complex. Furthermore, it was clarified that the [ReO2(4-MeOpy)4]2+ in CH3CN has the characteristic absorption bands at 236, 278, 330, 478, and 543 nm and their molar absorption coefficients are 4.3 × 104, 4.5 × 103, 1.0 × 104, and 6.1 × 103 M−1 cm−1 (M = mol dm−3), respectively.  相似文献

When Tris–SO4was used as an extraction buffer for phosphoenolpyruvate carboxylase (PEPC) from leaves of the C4plant Cynodon dactylon(L.) Pers., a higher extractable activity was obtained as compared to Tris–HCl, especially at low phosphoenolpyruvate concentrations and an assay pH of 7.2. The Tris–SO4-extracted PEPC activity was stable under dilution and remained unchanged for at least 24 h at 22°C. This enzyme was less sensitive to both activation by glucose-6-phosphate and inhibition by L-malate. The effects of Tris–SO4could be attributed to its preferential exclusion from the enzymic protein domain and, therefore, to a shifting of this oligomeric enzyme to a more aggregable form that is more stable and active.  相似文献

High-temperature-induced inhibition of the acceptor side of Photosystem II (PS II) was studied in tobacco thylakoids using oxygen evolution, chlorophyll a (Chl a) fluorescence and redox potential measurements. When thylakoids were heated at 2 °C/min from 25 to 50 °C, the oxygen evolving complex became inhibited between 32 and 45 °C, whereas the acceptor side of PS II tolerated higher temperatures. Variable Chl a fluorescence decreased more slowly than oxygen evolution, suggesting that transitions between some S-states occurred even after heat-induced inhibition of the oxygen evolving activity. 77 K emission spectroscopy reveals that heating does not cause detachment of the light-harvesting complex II from PS II, and thus the heat-induced increase in the initial F0 fluorescence is due to loss of exciton trapping in the heated PS II centers. Redox titrations showed a heat-induced increase in the midpoint potential of the QA/QA -) couple from the control value of –80 mV to +40 mV at 50 °C, indicating a loss of the reducing power of QA -). When its driving force thus decreased, electron transfer from QA -) to QB in the PS II centers that still could reduce QA became gradually inhibited, as shown by measurements of the decay of Chl a fluorescence yield after a single turnover flash. Interestingly, the heat-induced loss of variable fluorescence and inhibition of electron transfer from QA -) to QB could be partially prevented by the presence of 5 mM bicarbonate during heating, suggesting that high temperatures cause release of the bicarbonate bound to PS II. We speculate that both the upshift in the redox potential of the QA/QA -) couple and the release of bicarbonate may be caused by a heat-induced structural change in the transmembrane D1 or D2 proteins. This structural change may, in turn, be caused by the inhibition of the oxygen evolving complex during heating.  相似文献

Under optimal freeze-drying conditions, solutions exhibit a cake-like porous structure. However, if the solution temperature is higher than the glass transition temperature of the maximally freeze-concentrated phase (Tg′) during drying phase, the glassy matrix undergoes viscous flow, resulting in cake collapse. The purpose of the present study was to investigate the effect of cake collapse on the integrity of freeze-dried bull spermatozoa. In a preliminary experiment, factors affecting the Tg′ of conventional EGTA buffer (consisting of Tris–HCl, EGTA and NaCl) were investigated in order to establish the main experimental protocol because EGTA buffer Tg′ was too low (−45.0 °C) to suppress collapse. Modification of the EGTA buffer composition by complete removal of NaCl and addition of trehalose (mEGTA buffer) resulted in an increase of Tg′ up to −27.7 °C. In the main experiment, blastocyst yields after ooplasmic injection of freeze-dried sperm preserved in collapsed cakes (drying temperature: 0 or −15 °C) were significantly lower than those of sperm preserved in non-collapsed cake (drying temperature: −30 °C). In conclusion, freeze-dried cake collapse may be undesirable for maintaining sperm functions to support embryonic development, and can be inhibited by controlling both Tg′ of freeze-drying buffer and temperature during the drying phase.  相似文献

The tetranuclear and pentanuclear mixed-valence coordination compounds Na[(NC)5FeII-μ(CN)-PtIV(NH3)4-μ(NC)-FeII(CN)4-μ(CN)-RuIII(NH3)5], or FePtFeRu, and [RuIII(NH3)5-μ(NC)-FeII(CN)4-μ(CN)-PtIV(NH3)4-μ(NC)-FeII(CN)4-μ(CN)-RuIII(NH3)5](OSO2CF3)2, or RuFePtFeRu, were synthesized and characterized by IR and UV-Vis spectroscopy, electron microprobe analysis (EPMA), inductively coupled plasma (ICP), and cyclic voltammetry (CV). Both molecules exhibit FeII → PtIV intervalent charge transfer (IVCT) absorptions in the 400-450 nm range and FeII → RuIII transition(s) between 750 and 950 nm. The energies, intensities, and half-widths of these transitions correspond well with those of model compounds. The cyclic voltammogram of FePtFeRu between 0.00 and 0.90 V versus SCE exhibits two quasi-reversible Fe waves at 0.56 and 0.74 V versus SCE, while that for RuFePtFeRu has only one Fe redox event at 0.72 V versus SCE. When the potential of the working electrode is scanned negative of −0.38 V versus SCE, however, both complexes undergo an ECE (electrochemical-chemical-electrochemical) mechanism whereby the electrochemical reduction of Ru(III) is followed by a double electron transfer to reduce Pt(IV) to Pt(II). Upon reduction to Pt(II), the cyanide bridges break and the complexes dissociate into smaller fragments. Irradiation of the FeII → PtIV IVCT transition in both compounds leads to a photolysis solution that contains dissociated Fe(II)-Ru(III) as one of its products. Irradiation of the FeII → RuIII IVCT transition yields a similar UV-Vis spectrum, suggesting that the same intermediate is common to both photolysis mechanisms. The implications of this research within the larger context of multiple electron transfer are also discussed.  相似文献

A conserved tryptophan residue located between the A1B and FX redox centres on the PsaB side of the Photosystem I reaction centre has been mutated to a glycine in Chlamydomonas reinhardtii, thereby matching the conserved residue found in the equivalent position on the PsaA side. This mutant (PsaB:W669G) was studied using EPR spectroscopy with a view to understanding the molecular basis of the reported kinetic differences in forward electron transfer from the A1A and the A1B phyllo(semi)quinones. The kinetics of A1 reoxidation due to forward electron transfer or charge recombination were measured by electron spin echo spectroscopy at 265 K and 100 K, respectively. At 265 K, the reoxidation kinetics are considerably lengthened in the mutant in comparison to the wild-type. Under conditions in which FX is initially oxidised the kinetics of charge recombination at 100 K are found to be biphasic in the mutant while they are substantially monophasic in the wild-type. Pre-reduction of FX leads to biphasic kinetics in the wild-type, but does not alter the already biphasic kinetic properties of the PsaB:W669G mutant. Reduction of the [4Fe-4S] clusters FA and FB by illumination at 15 K is suppressed in the mutant. The results provide further support for the bi-directional model of electron transfer in Photosystem I of C. reinhardtii, and indicate that the replacement of the tryptophan residue with glycine mainly affects the redox properties of the PsaB bound phylloquinone A1B.  相似文献

The electronic spectrum of CuII(dps)2 in CH3CN with dps=3,5-diisopropylsalicylate shows a ligand field absorption at λmax=711 nm (ε=140 M−1 cm−1), and a phenolate to Cu(II) ligand-to-metal charge transfer (LMCT) band at λmax=428 nm (ε=950). LMCT excitation of CuII(dps)2 leads to the reduction of Cu(II) to Cu(I). Copper(II) disappears with φ=2.8×10−3 at λirr=436 nm.  相似文献

The porphyrin, meso-5-(pentafluorophenyl)-10, 15, 20-tris(4-pyridyl)porphyrin has been used to synthesize two new metalloporphyrin complexes. Insertion of copper(II) into the porphyrin center gives the copper(II) porphyrin. Coordination of three [Ru(bipy)2Cl]+ moieties (where bipy = 2,2′-bipyridine) to the pyridyl nitrogens of the copper(II) porphyrin gives the target complex. Electronic transitions associated with the copper(II) porphyrin and the triruthenium copper(II) porphyrin include an intense Soret band and a less intense Q-band in the visible region of the spectrum. An intense π-π∗ transition in the UV region associated with the bipyridyl groups and a metal to ligand charge transfer (MLCT) band appearing as a shoulder to the Soret band are observed for the ruthenated copper(II) porphyrin. Electrochemical properties associated with the multimetallic complex include a redox couple in the cathodic region with E1/2 = −0.86 V versus Ag/AgCl attributed to the porphyrin and a redox couple in the anodic region E1/2 = 0.88 V versus Ag/AgCl due to the RuIII/II couple. DNA titrations indicate the triruthenium copper(II) porphyrin interacts with DNA potentially through a groove binding mechanism. Irradiation of aqueous solutions of the target complex and supercoiled DNA at a 10:1 base pair to complex ratio with visible light above 400 nm indicates that the complex causes nicking of the DNA helix.  相似文献

Measuring and monitoring of protein oxidation modifications is important for biopharmaceutical process development and stability assessment during long-term storage. Currently available methods for biomolecules oxidation analysis use time-consuming peptide mapping analysis. Therefore, it is desirable to develop high-throughput methods for advanced process control of protein oxidation. Here, we present a novel approach by which oxidative protein modifications are monitored by an indirect potentiometric method. The method is based on adding an electron mediator, which enhances electron transfer (ET) between all redox species and the electrode surface. Specifically, the procedure involves measuring the sharp change in the open circuit potential (OCP) for the mediator system (redox couple) as a result of its interaction with the oxidized protein species in the solution. Application of Pt and Ag/AgCl microelectrodes allowed for a high-sensitivity protein oxidation analysis. We found that the Ru(NH3)62+/3+ redox couple is suitable for measuring the total oxidation of a wide range of therapeutic proteins between 1.1 and 13.6%. Accuracy determined by comparing with the known percentage oxidation of the reference standard showed that percentage oxidation determined for each sample was within ±20% of the expected percentage oxidation determined by mass spectrometry.  相似文献

The synthesis of an unsymmetrical phenanthroline-based ligand, 2-methyl-9-(3,5-dimethylpyrazolylmethyl)-1,10-phenanthroline (L), and its cupric [Cu(II)] (1) and cuprous [Cu(I)] (2) complexes, are reported. The X-ray structures of each of these Cu complexes show distinct changes in coordination environments consistent with the geometrical preferences of the two oxidation states. In the solid-state, the Cu(II) complex (1) adopts a geometry best described as trigonal bipyramidal, while the Cu(I) complex (2) consists of a single dicationic dimer in which the ligand bridges between two copper ions, separated by 4.26 Å. The two Cu(I) coordination sites differ in 2 with one copper center complexed in a trigonal planar geometry and the other copper in a distorted tetrahedral environment; the latter coordination results from an additional CH3CN ligand. Complex 1 exhibits a reversible redox process at −0.34 V versus Fc/Fc+ in CH3CN, attributable to the Cu2+/Cu+ couple, while the dimeric Cu(I) complex (2) does not display this redox couple on the CV timescale. Oxygen evolution is inhibited when leaves of chilling-sensitive plants like cucumber are treated at 0 °C in the dark. The activity is restored by moderate illumination at room temperature. We examined the changes in the redox state of the Mn-cluster in cucumber leaves in the processes of dark-chilling inhibition and subsequent light-induced reactivation by means of thermoluminescence (TL). A TL B-band arising from S2QB charge recombination in PSII was observed upon single-flash illumination of untreated leaves, whereas four flashes were required to yield the B-band after dark-chilling treatment for 24 h. This three-step delay indicates that over-reduced states of the Mn-cluster such as the S−2 state were formed during the treatment. Fitting analysis of the flash-number dependence of the TL intensities showed that the Mn-cluster was more reduced with a longer period of the treatment and that S−3 was the lowest S-state detectable in the dark-chilled leaves. Measurements of the Mn content by atomic absorption spectroscopy showed that Mn atoms were gradually released from PSII during the dark-chilling treatment but re-bound to PSII by illumination at 30 °C. Thus, dark-chilling inhibition of oxygen evolution can be ascribed to the disintegration of the Mn-cluster due to its over-reduction. The observation of the S−3 state in the present in vivo system strongly suggests that S−3, which has been observed only by addition of exogenous reductants into in vitro preparations, is indeed a redox intermediate of the Mn-cluster in the processes of its disintegration and photoactivation.  相似文献
