Infection of plants with pathogens leads not only to the induction of defence reactions but also to changes in carbohydrate metabolism. In this study, the effects of infection by a virulent and an avirulent strain of P. syringae on spatio-temporal changes in photosynthesis were compared using chlorophyll fluorescence imaging. The maximum PSII quantum yield, effective PSII quantum yield and nonphotochemical quenching were decreased in Arabidopsis leaves infected with either strain. At the same time, the quantum yield of nonregulated energy dissipation was increased. These changes could be detected by chlorophyll fluorescence imaging before symptoms were visible by eye. The effects were restricted to the vicinity of the infection site and did not spread to uninfected areas of the leaf. Qualitatively similar changes in photosynthetic parameters were observed in both interactions. Major differences between the responses to both strains were evident in the onset and time course of changes. A decrease in photosynthesis was detectable already at 3 h only after challenge with the avirulent strain while after 48 h the rate of photosynthesis was lower with the virulent strain. In contrast to photosynthesis, the regulation of marker genes for source/sink relations and the activities of invertase isoenzymes showed qualitative differences between both interactions. Inoculation of the virulent but not the avirulent strain resulted in downregulation of photosynthetic genes and upregulation of vacuolar invertases. The activity of vacuolar invertases transiently increased upon infection with the virulent strain but decreased with the avirulent strain while extracellular invertase activity was downregulated in both interactions.  相似文献

The hypothesis that seasonal changes in sycamore aphid,Drepanosiphum platanoidis (Schr.), recruitment are determined by changes in food quality and aphid population density was tested. There was no clear association between the reproductive activity of the sycamore aphid and the seasonal changes in specific amino acids or groups of amino acids in extracts of sycamore,Acer pseudoplatanus L., leaves. Seasonal changes in reproductive activity tracked the changes in total amino acids of the leaf tissue of the host, but with a short time delay. High numbers of adult aphids appeared to depress reproductive activity. A regression analysis of the results revealed that total amino acids the previous week and current numbers of adult aphids significantly affected sycamore aphid reproductive activity. The results of this analysis support the above hypothesis, that the marked seasonal changes in the total quantity of amino acids in sycamore leaves and intraspecific competition for this resource, through its effect on adult weight, shape the seasonal cycle in the reproductive activity of the sycamore aphid.  相似文献

The effect of kanamycin on the electrophysical parameters of cell suspensions of Escherichia coli K-12 and pMMB33 was investigated. Incubation of the sensitive K-12 strain with kanamycin resulted in significant changes in the orientation spectra (OS) of the cell suspensions; these changes were not revealed in the case of the resistant pMMB33 strain. In the case of the sensitive K-12 strain incubated with different kanamycin concentrations, changes in the OS of the cell suspensions occurred within the 10–1000 kHz frequency range of the orienting electrical field. The most pronounced change in the electrooptical signal was observed at 10 μg/ml of kanamycin. Control experiments were carried out by standard plating on nutrient media. Thus, the OS changes of suspensions in the presence of antibiotics may be used as a test for microbial resistance to such antibiotics.  相似文献

The fungus Harpella melusinae (Harpellales: Trichomycetes) is obligately associated with the midguts of larval Simuliidae (Diptera). The level of infestation of a population of Simulium ornatum by H. melusinae was monitored at a stream in Hampshire, England. Significant temporal changes in the level of infestation were recorded during monthly and weekly collections; a twenty-fold increase being recorded over a nine-day period. Possible mechanisms by which these changes occur are discussed.  相似文献

Seasonal changes in the infestation and dispersion patterns of egg predatory nemerteans on their crab hosts were analyzed. Marked differences in the seasonal patterns of infestation were noted between worm species, yet, common patterns in the aggregation of the worms were found. Worm aggregation increased at the onset of the reproductive seasons of the hosts, and at the nadirs of the reproductive seasons for those hosts with year round breeding. The aggregation patterns of two worm species fluctuated with the physical environment of their estuarine hosts. Salinity changes as a result of seasonal rains may have caused changes in the underlying dispersion patterns of Carcinonemertes epialti on Hemiarapsus oregonensis, and C. mitsukurii on Portunus pelagicus. Lastly, the embryogenic cycle of the host species was significant in shaping the infestation and aggregation patterns of C. epialti on Cancer anthonyi and C. regicides on Paralithodes camtschaticus. Worm immigration and emigration were linked to crab embryogenesis and directly influenced the dispersion patterns of the worms.  相似文献

The inhibitory effect of nystatin and killer toxin on the growth of free and covalently-immobilizedSaccharomyces cerevisiae cells was studied. The resistance of immobilized cells to both agents was accompanied by increased amounts of phospholipids and sterols. Seasonal changes in vanadate sensitive plasma membrane H+-ATPase activity of bilberry (Vaccinium myrtillus L.) and Scots pine (Pinus sylvestris L.) were studied in a period from February to August in northern Finland. The plasma membrane isolation was performed by sucrose gradient centrifugation, and the H+-ATPase activity was assayed by spectrophotometrical determination of released inorganic phosphate. The studied species showed seasonal changes from high winter to low spring activity, indicating probable physiological changes between hardened and dehardened tissue. ATPase activity of bilberry peaked up at the beginning of the growth period, obviously due to active phloem loading of photosynthates.  相似文献

Summary This investigation was aimed at studying the mycoflora present in Huelva (South West Spain), a highly polluted region. The study was carried out using the Cour trap and it is worth pointing out that the chemical treatment performed following the Cour methodology might destroy a certain percentage of spores. We analyzed the presence and biannual changes (10 April 1989 to 8 April 1991) of fungal spores of three genera, seventeen form genera, and four groups lacking taxonomic category, looking at their morphology: ascospores, basidiospores, and unidentified unicellular and multicellular spores. Special attention was given to agriculturally interesting groups. The results obtained were correlated with meteorological parameters (rainfall, minimum temperature and humidity).  相似文献

Pollen morphology and ultrastructure inLactuceae pollen is considered in relation to the accomodation of volume changes, pollination biology and exine-held substances. Echinate pollen grains, such as those ofCatananche, are shown to accomodate volume changes by folding along the colpi and possibly by volume changes in the cavea. The different patterns of echinolophate pollen respond in different ways. Folding along the colpi is important inScorzonera andTragopogon and to a limited extent inCichorium andEpilasia whilst inScolymus the colpi are almost immobilized. Movements of the lacunar floors take over the harmomegathic function to compensate for lack of colpus mobility. Bulging of the intine at the apertures and changes in the size of the cavea may account for part of the volume change accomodated in any pollen type. Echinolophate pollen is interpreted as being a superior means of regulating volume changes with the most economical and mechanically efficient use of wall material which has evolved independently in several tribes ofCompositae.  相似文献

The dynamics of changes in total proteolytic activity and activities of various groups of proteases in the coleoptiles of 3- to 12-day-old wheat seedlings grown in light with and without antioxidant BHT (2,6-di-tert-butyl-4-methylphenol) was studied. It was established that the specialized proteases that easily hydrolyze specific synthetic substrates and the enzymes actively hydrolyzing histone H1 dominate in young coleoptiles of 3- to 4-day-old seedlings. Proteases that degrade equally well the majority of the studied substrates are accumulated in the cells of old coleoptiles of 11- to 12-day-old seedlings. Under the effect of BHT, the plants grown in the light (in comparison with etiolated seedlings) demonstrated a somewhat changed dynamics of proteolytic activity in young coleoptiles and the disappearance of proteases active toward histone H1. An inhibitory analysis revealed a relative domination of cysteine proteases in young coleoptiles at the initial development stage of seedlings, whereas the fraction of serine proteases markedly increased in old coleoptiles. We presume that the revealed quantitative and qualitative changes in the proteolytic apparatus of the coleoptile cells induced by BHT may be largely responsible for the retardant and geroprotective effect of this antioxidant in plants.  相似文献

The metabolic changes in tomato fruits and seeds separately infected with cucumber mosaic virus, Pseudomonas syringae pv. tomato or Botrytis cinerea were investigated cytochemically. The changes of peroxidase (E.C. 1.11.1.7) and β-glucosidase (E.C. 3.2.1.21) were investigated biochemically as well. Tomato fruits were involved in the study because of their high economic value. Tomato seeds were investigated since they have been used most extensively as a model system for studying the physiology and biochemistry of seed development. The diseases caused by the pathogens under study are of special importance for yield reduction in tomato. The three pathogens provoked local changes in the activities of enzymes under study that affect the infected pericarp tissues and neighboring seeds. It was established non-specific and specific responses. The non-specific responses of invaded tissues were expressed as a local enhancement of peroxidase activity in both pericarp tissues and seeds as well as a decrease in activities of: i. enzymes taking part in aerobic and anaerobic respiration, ii. hydrolases esterase and acid phosphatase involved in the basic metabolism as well as an enhancement of their activities in neighboring tissues. Furthermore, it was observed an enhancement of α-galactosidase activity in infected area was observed. The specific responses depending on the type of the pathogen consisted in an enhancement of glucose-6-phosphate dehydrogenase activity by virus infection and an increase of β-glucosidase activity by fungal invasion.  相似文献

77 K absorption and fluorescence spectra are measured during incubation of etiochloroplast isolated from pine cotyledons and suspended in buffers with or without various co-factors (GSH, ATP, CoA) and NADPH. It is shown that, in the absence of co-factors, rapid spectral changes due to denaturation of the protein-pigment complexes occur. The spectral changes differ according to whether denaturation occurs in the light or in the dark. The rate of denaturation of the Ch1-protein complexes is significantly lowered when co-factors are added and a protective effect of NADPH on the PChlide-protein complexes was observed.

The pH dependences of activities of homogenous hydrogenases of Thiocapsa roseopersicina and Desulfomicrobium baculatum in the reaction of hydrogen uptake in solution in the presence of benzyl viologen and the pH dependences of catalytic currents of hydrogen oxidation by electrodes on which these hydrogenases were immobilized were compared. Maximal activities of the hydrogenases from T. roseopersicina and D. baculatum in the reaction hydrogen uptake in solution were observed at pH 9.5 and 8.5, respectively. However, the steady-state current caused by catalytic uptake of hydrogen was maximal for the T. roseopersicina hydrogenase-containing electrode at pH 5.5-6.5 under overvoltage of 30-60 mV, whereas for electrodes with D. baculatum hydrogenase it was maximal at pH 6.0-6.5. Analysis of these data suggests that pH-dependent changes in the hydrogenase activities in solution during hydrogen uptake are due not only to the effect of proton concentration on the enzyme conformation or protonation of certain groups of the enzyme active center, but they are rather indicative of changes in free energy of the reaction accompanying changes in pH.  相似文献

Purified porcine heart lactate dehydrogenase was inactivated and partially unfolded with p-chloromercuribenzoate (pCMB). With the increase of pCMB/enzyme ratio the enzyme was gradually inhibited till almost completely inactivated at the pCMB/enzyme ratio of 20 : 1. Native polyacrylamide gel electrophoresis showed that with the increase of pCMB/enzyme ratio the bands of native enzyme decreased till completely vanished. Meanwhile inactive multiple bands emerged and became thicker, which implied that lactate dehydrogenase became loose. The conformational changes of the enzyme molecule modified with pCMB were followed using fluorescence emission, ultraviolet difference, and circular dichroism (CD) spectra. Increasing pCMB concentration resulted in the decrease of fluorescence emission intensity. The ultraviolet difference spectra of the enzyme modified with pCMB exhibited an increasing absorbance in the vicinity of 240 nm with the increasing concentration of the inhibitor. The changes of the fluorescence and ultraviolet difference spectra reflected the conformational changes of the enzyme. The CD spectrum changes of the enzyme showed that its secondary structure changed as well. These results suggest that pCMB not only inhibits this enzyme but also influences its conformation (partial unfolding).  相似文献

The Jegrznia floodplain has been deprived of flooding since the late 19th century. Human influence has been limited to haymaking and cattle grazing without land reclamation. Therefore, the present vegetation reflects the impact of hydrological changes, without being much disturbed by changes in other factors. A drainage gradient on the former floodplain was examined for nutrient deficiencies and compared with an intact floodplain. For this purpose we analyzed nutrient concentrations in above ground biomass, we measured redox potentials and peat variables for a rich fen vegetation, a well-developed fen meadow as well as an impoverished fen meadow vegetation. According to P:N-ratios in above ground biomass, all vegetation types appear to be limited by phosphorus, whereas the impoverished fen meadow is also limited by potassium. These results are supported by the chemical composition of the peat, redox characteristics and the physical changes in the peat induced by drainage. It is concluded that hydrological changes in combination with an annual mowing regime resulted in a shift from no nutrient limitation to P-and/or K-limitation in the former Jegrznia floodplain. Changes in hydrology at Jegrznia stopped the input of nutrients, increased the leaching of K and turned P into unavailable Al and Fe-complexes. Continued mowing increased P-and K-deficiency.  相似文献

A survey of 51 species fromNicotiana subgg.Tabacum, Rustica andPetunioides has shown that evolution was accompanied by a five-fold variation in nuclear DNA amounts. This variation, however, was not directly correlated with the changes in chromosome number. Drastic rearrangement of karyotypes is characteristic for the evolution ofNicotiana spp. Significant gain or loss in nuclear DNA has often accompanied such changes, but DNA variation has also occurred without significant changes in karyotype arrangements.—The distribution of nuclear DNA is discontinuous inNicotiana, species cluster into DNA groups with consistently regular increments in the mean DNA amounts. The discontinuities are viewed as steady states in terms of genomic balance and biological fitness.—Changes in the amount of nuclear DNA and in the heterochromatin are compared with the morphological, chromosomal and adaptive changes which accompanied speciation in 14 subgeneric sections. The evolutionary significance of DNA variation is discussed.  相似文献

Several studies have shown changes in the patterns of damage from feeding insects associated with changes in palatability and overall consumption as a result of wound-induced chemical changes in plants. This paper describes how the pattern of feeding damage made by the larvae of Spodoptera littoralis Boisd. (Lepidoptera: Noctuidae) on tomato is affected by changes in palatability of the leaves. Two sorts of responses to leaves from plants that had received prior damage were observed. Larvae offered a choice of leaves tended to take fewer meals on leaves from previously-wounded plants than on control leaves, frequently rejecting the former after sampling them. On wounded plants this rejection behaviour was associated with a shift in feeding site towards the base of the plant. However, starved larvae offered only a single excised leaf readily ate leaves from wounded plants but took shorter meals on these leaves than on controls. Although it was not directly tested it is possible that this difference in response reflected changes in food selectivity with a differing level of satiation. The results are considered in relation to the adaptive significance of the plant of changes in within-plant distributions of herbivore damage.  相似文献

Infection of striped mullet (Mugil cephalus) with Aeromonas hydrophila results in an acute septicemic disease. The disease can be experimentally induced by intramuscular injection, skin or gill scarification or by the oral route using pellets purposely seeded with bacteria. The organism was isolated from the blood 1–2 days after infection and from all organs 24 hr or longer after infection. The disease is characterized by early inflammatory and proliferative changes and later necrotic changes. Enteritis and hepatic necrosis are constant findings in aeromonad disease of M. cephalus but surface lesions are not pathognomic for these infections in mullet. Death of infected fish may be attributed to bacterial toxins which cause necrosis of parenchymal organs and soft tissue structure.  相似文献

Dynamics of CO2 evolution at low pressure was studied in barley, maize, pea, wheat and pine seedlings using the gas exchange system with laser photoacoustic spectrometer. The CO2 evolution from plant surfaces to environment increased with decreasing air pressure. Simultaneously the changes in activities of phosphoenolpyruvate carboxylase, glucose-6-phosphate dehydrogenase, glyceraldehyde phosphate dehydrogenase, alcohol-dehydrogenase, isocitrate dehydrogenase, malate dehydrogenase in pea and maize leaves were observed. Growth of the autotrophic nitrifying bacteria Nitrosomonas europaea and Nitrobacter sp. was studied in continuous culture. Steady state growth kinetics of both organisms conformed with that predicted by chemostat theory, modified to account for maintenance energy requirement. Steady state data were used to calculate the maximum specific growth rate, the saturation constant for growth, the true growth yield and the maintenance coefficient. Transient growth was studied by imposing step changes in dilution rate. Step increases resulted in overshoots and oscillations in substrate concentration before establishment of a new steady state while step decreases in dilution rate were followed by monotonic changes in substrate concentration. The size of overshoots in substrate concentration following step increases in dilution rate was dependent on both the magnitude of the increase and of the dilution rate prior to the change.  相似文献
