The production of ethylene by red raspberry (Rubus idaeus L.cv. Glen Clova) fruit increased climacterically during development.The concentration of ethylene within green fruit was low butincreased substantially as fruit abscission and ripening commenced.The receptacle contained higher concentrations than the drupeletsat all stages measured. In the mature ripening fruit the ethyleneconcentrations were found to be physiologically significant,and would accelerate the abscission of large green non-abscisingfruit if supplied as a fumigant. The addition of ethylene toripe fruit did not accelerate abscission, probably because saturatinglevels occurred naturally within these fruit. Reduction of ethylenesynthesis rates using the inhibitor of ethylene production aminoethoxyvinylglycine(AVG) reduced the rate of abscission zone weakening which occursin detached large green fruit. The rate of ethylene productionwas found to be dependent on the supply of the precursor l-aminocyclopropane-l-carboxylicacid (ACC). This only accumulated to any extent in those ripefruit with high rates of ethylene production. Citrus fruits have two abscission zones (AZ), named A (in the pedicel) and C (in the calyx). Early fruitlet abscission takes place exclusively through AZ A, while at June drop it is progressively inactivated and AZ C begins to operate. In previous work, it has been demonstrated that carbohydrate and ethylene regulate fruit drop through abscission zone C. In this paper, we have analysed the effect of these two factors in developing fruitlets of Satsuma mandarins (Citrus unshiu [Mak.] Marc.) cv. Okitsu to elucidate their involvement on abscission through AZ A. The data indicated that ACC content and ethylene production of fruitlets paralleled abscission rates. Sucrose supplementation increased fruit set, although did not counteract the abscising effect induced by ACC. Branch girdling of terminal fruitlets carrying several leaves significatively reduced ethylene production and abscission rates, and increased sugar content. Pedicel girdling showed the opposite. Taken together, the results revealed that the carbohydrate content may be a biochemical signal involved in the mechanisms controlling abscission through AZ A. The evidence also showed parallelisms between ethylene and its activation. As the induction of higher ethylene levels after the period of AZ A activity, however, was not able to promote fruit drop, it is also concluded that solely ethylene is not sufficient to activate abscission.  相似文献

Physiological characterization of ethylene-suppressed Cantaloupe Charentais melons ( Cucumis melo var. cantalupensis Naud cv. Védrantais) revealed that some ripening-associated events, like degreening of the rind and cell separation in the peduncular abscission zone, are totally dependent on ethylene. By contrast, some other ripening events, like softening and membrane deterioration, depend only partially on ethylene and display some ethylene-independent components. Application of increasing levels of exogenous ethylene on these antisense 1-aminocyclopropane-1-carboxylic acid oxidase fruits enabled the determination of the gradual sensitivity of various ripening pathways to the hormone. The threshold level of ethylene capable of physiological activity varied from 1 ppm for degreening of the rind to 2.5 ppm for softening, membrane deterioration and cell separation in the peduncular abscission zone. Up to a saturating dose of 5 ppm, the extent of rind degreening was proportionally related to the level of applied ethylene. The saturating levels of ethylene for flesh softening (2.5 ppm) and for membrane deterioration and cell separation in the peduncular abscission zone (5 ppm) were much lower than the internal ethylene found at the climacteric peak of wild-type fruit (over 100 ppm). The cessation of ethylene treatment resulted in a complete arrest of the rind degreening and peduncular cell separation indicating that both ripening pathways are completely dependent on ethylene. On the contrary, softening and membrane deterioration, though significantly slowed upon removal of ethylene treatment, continued to proceed in the absence of the hormone, thereby unmasking the ethylene-independent component of softening and membrane deterioration. The presence of ethylene-independent components in the regulation of individual pathways indicates that the ripening of climacteric fruit involves a substantial portion of non-climacteric regulation.  相似文献

Inconsistency of cropping is an important problem for UK sweet cherry production. Premature fruit abscission in Prunus can reduce yields severely, however, the environmental cues and hormonal signals that trigger abscission have not been identified. Auxin (IAA) is known to delay abscission by reducing the sensitivity of cells in the abscission zone to ethylene, a promoter of abscission. Therefore, the capacity for polar auxin transport (PAT) through sweet cherry pedicels was examined in relation to fruit abscission. Cherry ‘spurs’ (short shoots) with similar leaf areas and different fruit numbers were phloem-girdled to restrict assimilate movement. Abscission from spurs with many fruit (eight or more) occurred within 14 days of girdling, whereas abscission from spurs with few (two) fruit was minimal. The pedicels’ capacity for PAT in spurs with different fruit numbers was determined 1, 3 and 9 days after girdling (DAG). Fruit were analysed for endogenous IAA concentration 3, 5, 7 and 9 DAG. PAT inhibitors 2,3,5-triiodobenzoic acid or 1-N-naphthylphtalamic acid were applied to pedicels of fruit not expected to abscise, i.e. on spurs with few fruit. The effect of these inhibitors on fruit abscission was determined 14 DAG. The proportion of the transported [3H]-IAA was lower from the outset in pedicels from spurs with many fruit. By 9 DAG, symptoms of fruit abscission were apparent and 40% less [3H] -IAA was transported through pedicels on spurs with many fruit. Fruit endogenous IAA concentrations were similar in the two groups of spurs. Application of PAT inhibitors shortly after girdling increased fruit abscission by 30%. The results suggest that although a decline in PAT is not the only cause of fruit abscission, the maintenance of PAT contributes to fruit retention.  相似文献

Tomato fruit ripening and ethylene production were inhibited following treatment with methyl bromide (MB). Methyl bromide significantly delayed ripening initiation in mature-green (MG) fruit and retarded the rate of ripening of turning (T) fruit as measured by color development and flesh softening. Treatment with MB caused an initial transient burst of ethylene production, but the subsequent ripening-associated increase in ethylene was delayed. Ethylene treatment partially overcame MB inhibition in MG fruit but had no affect on T fruit. The inhibition of ethylene production by MB appears to be due to lack of formation of 1-aminocycloprone-1-carboxylic acid (ACC) in MG fruit, whereas in T fruit lack of conversion of ACC to ethylene is indicated. A key feature of MB inhibition of ripening in tomato appears to be reduced sensitivity to ethylene.  相似文献

Ethylene promotes fruit ripening, including softening. The fruit of melting-flesh peach (Prunus persica (L). Batsch) cultivar 'Akatsuki' produces increasing levels of ethylene, and the flesh firmness softens rapidly during the ripening stage. On the other hand, the fruit of stony hard peach cultivars 'Yumyeong', 'Odoroki', and 'Manami' does not soften and produces little ethylene during fruit ripening and storage. To clarify the mechanism of suppression of ethylene production in stony hard peaches, the expression patterns of four ethylene biosynthesis enzymes were examined: ACC synthases (Pp-ACS1, Pp-ACS2, and Pp-ACS3) and ACC oxidase (Pp-ACO1). In the melting-flesh cultivar 'Akatsuki', Pp-ACS1 mRNA was dramatically induced after harvesting, and a large amount of ethylene was produced. On the other hand, in stony hard peaches, Pp-ACS1 mRNA was not induced during the ripening stage, and ethylene production was inhibited. Since Pp-ACS1 mRNA was induced normally in senescing flowers, wounded leaves, and wounded immature fruit of 'Yumyeong', Pp-ACS1 was suppressed only at the ripening stage, and was not a defect in Pp-ACS1. These results indicate that the suppression of fruit softening in stony hard peach cultivars was caused by a low level of ethylene production, which depends on the suppressed expression of Pp-ACS1.  相似文献

Seedlings of tomato fruit ripening mutants were screened for their ability to respond to ethylene. Ethylene induced the triple response in etiolated hypocotyls of all tomato ripening mutants tested except for one, Never ripe (Nr). Our results indicated that the lack of ripening in this mutant is caused by ethylene insensitivity. Segregation analysis indicated that Nr-associated ethylene insensitivity is a single codominant trait and is pleiotropic, blocking senescence and abscission of flowers and the epinastic response of petioles. In normal tomato flowers, petal abscission and senescence occur 4 to 5 days after the flower opens and precede fruit expansion. If fertilization does not occur, pedicel abscission occurs 5 to 8 days after petal senescence. If unfertilized, Nr flowers remained attached to the plant indefinitely, and petals remained viable and turgid more than four times longer than their normal counterparts. Fruit development in Nr plants was not preceded by petal senescence; petals and anthers remained attached until they were physically displaced by the expanding ovary. Analysis of engineered 1-aminocyclopropane-1-carboxylate (ACC) synthase-overexpressing plants indicated that they are phenotypic opposites of Nr plants. Constitutive expression of ACC synthase in tomato plants resulted in high rates of ethylene production by many tissues of the plant and induced petiole epinasty and premature senescence and abscission of flowers, usually before anthesis. There were no obvious effects on senescence in leaves of ACC synthase overexpressers, suggesting that although ethylene may be important, it is not sufficient to cause tomato leaf senescence; other signals are clearly involved.  相似文献

The physiological effects of reduced ethylene synthesis in a transgenic tomato (Lycopersicon esculentum) line expressing 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase enzyme have been examined. Fruit from the transgenic line 5673 ripen significantly slower than control fruit when removed from the vine early in ripening. In contrast, fruit that remain attached to the plants ripen much more rapidly, exhibiting little delay relative to the control. Ethylene determinations on attached fruit revealed that there was significantly more internal ethylene in attached than detached fruit. The higher ethylene content can fully account for the observed faster on-the-vine ripening. All of the data are consistent with a catalytic role for ethylene in promoting many, although not all, aspects of fruit ripening. Biochemical analyses of transgenic fruit indicated no significant differences from controls in the levels of ACC oxidase or polygalacturonase. Because transgenic fruit are significantly firmer than controls, this last result indicates that other enzymes may have a significant role in fruit softening.  相似文献

'Gloster 69' apples are unusual because they do not accumulate ethylene during storage at 2 kPa O2 at 1.5–3.5°C with continuous ethylene removal. Their ethylene physiology and the extent of various ripening processes in storage were investigated. Ethylene production and l-aminocyclopropane-l-carboxylic acid (ACC) remained low for up to 200 days, and both increased on transfer of fruit to 15°C. The increase in ACC could be stimulated by ethylene treatment of apples after storage. In spite of this evidence that fruit remained preclimacteric, some softening and production of soluble pectin and volatile esters occurred at 3.5°C. These processes were suppressed at 1.5°C, but chlorophyll, starch, malate and sucrose losses and increases in glucose and fructose occurred at both temperatures.  相似文献

Aminoethoxyvinylglycine (AVG) inhibits 1-aminocyclopropane-1-carboxylic acid (ACC) synthase, and thus blocks ethylene synthesis. Preharvest foliar application of AVG to apple (Malus domestica Borkh.) fruit retards several key events of maturation including climacteric ethylene production, starch conversion to sugars, fruit softening, and abscission zone development. Although the impact of AVG on apple fruit maturation is well known, the biochemical basis of these effects is not well understood. The effects of AVG application on Redchief Delicious apple fruit maturation were studied. AVG applied four weeks prior to harvest significantly reduced internal ethylene levels, amylose degradation, and accumulation of sucrose, glucose, and sorbitol. Because AVG application coincidentally inhibited starch degradation and the increase in internal ethylene, we investigated the enzymatic basis of starch mobilization in apple fruit. Amylase activity was somewhat reduced in AVG-treated fruit. Amylase activity was less in AVG-treated fruit during the early stages of starch mobilization. Starch phosphorylase activity increased dramatically during the later stages of starch mobilization, but was not affected by AVG treatment. Soluble starch synthase activity was also unaffected by AVG treatment and remained constant throughout the eight-week harvest period. Moreover, AVG did not affect the levels of amylopectin, fructose, malate, ascorbate, citrate, or anthocyanin. These results suggest that apple fruit ripening has both ethylene-dependent and -independent processes occurring simultaneously.  相似文献

'La France' pear ( Pyrus communis L.) fruit stored at 1°C for 1 month (short-term storage) before transfer to 20°C softened and developed a melting texture during ripening, whereas fruit stored for 5 months (long-term storage) before transfer to 20°C softened but did not develop a melting texture. To clarify the mechanisms involved in fruit softening and textural changes, the cDNAs encoding cell-wall hydrolases were isolated by RT-PCR, and their expression and localization were investigated in 'La France' pears. Genes encoding three polygalacturonases (PG; EC 3.2.1.15), four pectin methylesterases (PME; EC 3.1.1.11), one α -arabinofuranosidase (ARF; EC 3.2.1.55), three β -galactosidases (GAL; EC 3.2.1.23), and two endo-1,4- β - d -glucanases (Cel; EC 3.2.1.4) were isolated. Among these 13 isolated genes, PcPG1 was the only gene for which the mRNA expression levels increased in both the short- and long-term stored fruits. This suggested that PcPG1 is involved in fruit softening rather than in the development of the melting texture. In contrast, the expression levels of PcPG3 , PcPME1 , PcPME2 , PcPME3 , PcGAL1 , PcGAL2 , and PcCel2 increased during ripening only in the short-term stored fruit. These genes might thus be involved in the development of the melting texture.  相似文献
