The effects of different spectral region of excitation and detection of chlorophyll (Chl) a fluorescence at room temperature on the estimation of excitation energy utilization within photosystem (PS) 2 were studied in wild-type barley (Hordeum vulgare L. cv. Bonus) and its Chl b-less mutant chlorina f2 grown under low and high irradiances [100 and 1 000 μmol(photon) m−2 s−1]. Three measuring spectral regimes were applied using a PAM 101 fluorometer: (1) excitation in the red region (maximum at the wavelength of 649 nm) and detection in the far-red region beyond 710 nm, (2) excitation in the blue region (maximum at the wavelength of 461 nm) and detection beyond 710 nm, and (3) excitation in the blue region and detection in the red region (660– 710 nm). Non-photochemical quenching of maximal (NPQ) and minimal fluorescence (SV0), determined by detecting Chl a fluorescence beyond 710 nm, were significantly higher for blue excitation as compared to red excitation. We suggest that this results from higher non-radiative dissipation of absorbed excitation energy within light-harvesting complexes of PS2 (LHC2) due to preferential excitation of LHC2 by blue radiation and from the lower contribution of PS1 emission to the detected fluorescence in the case of blue excitation. Detection of Chl a fluorescence originating preferentially from PS2 (i.e. in the range of 660–710 nm) led to pronounced increase of NPQ, SV0, and the PS2 photochemical efficiencies (FV/FM and FV′/FM′), indicating considerable underestimation of these parameters using the standard set-up of PAM 101. Hence PS1 contribution to the minimal fluorescence level in the irradiance-adapted state may reach up to about 80 %.  相似文献

Two rice chlorophyll (Chl) b-less mutants (VG28-1, VG30-5) and the respective wild type (WT) plant (cv. Zhonghua No. 11) were analyzed for the changes in Chl fluorescence parameters, xanthophyll cycle pool, and its de-epoxidation state under exposure to strong irradiance, SI (1 700 μmol m−2 s−1). We also examined alterations in the chloroplast ultrastructure of the mutants induced by methyl viologen (MV) photooxidation. During HI (0–3.5 h), the photoinactivation of photosystem 2 (PS2) appeared earlier and more severely in Chl b-less mutants than in the WT. The decreases in maximal photochemical efficiency of PS2 in the dark (Fv/Fm), quantum efficiency of PS2 electron transport (ΦPS2), photochemical quenching (qP), as well as rate of photochemistry (Prate), and the increases in de-epoxidation state (DES) and rate of thermal dissipation of excitation energy (Drate) were significantly greater in Chl b-mutants compared with the WT plant. A relatively larger xanthophyll pool and 78–83 % conversion of violaxanthin into antheraxanthin and zeaxanthin in the mutants after 3.5 h of HI was accompanied with a high ratio of inactive/total PS2 (0.55–0.73) and high 1–qP (0.57–0.68) which showed that the activities of the xanthophyll cycle were probably insufficient to protect the photosynthetic apparatus against photoinhibition. No apparent difference of chloroplast ultrastructure was found between Chl b-less mutants and WT plants grown under low, LI (180 μmol m−2 s−1) and high, HI (700 μmol m−2 s−1) irradiance. However, swollen chloroplasts and slight dilation of thylakoids occurred in both mutants and the WT grown under LI followed by MV treatment. These typical symptoms of photooxidative damage were aggravated as plants were exposed to HI. Distorted and loose scattered thylakoids were observed in particular in the Chl b-less mutants. A greater extent of photoinhibition and photooxidation in these mutants indicated that the susceptibility to HI and oxidative stresses was enhanced in the photosynthetic apparatus without Chl b most likely as a consequence of a smaller antenna size.  相似文献

In our search for new natural photosynthetic inhibitors that could lead to the development of “green herbicides” less toxic to environment, the diterpene labdane-8α,15-diol (1) and its acetyl derivative (2) were isolated for the first time from Croton ciliatoglanduliferus Ort. They inhibited photophosphorylation, electron transport (basal, phosphorylating and uncoupled) and the partial reactions of both photosystems in spinach thylakoids. Compound 1 inhibits the photosystem II (PS II) partial reaction from water to Na+ Silicomolibdate (SiMo) and has no effect on partial reaction from diphenylcarbazide (DPC) to 2,6-dichlorophenol indophenol (DCPIP), therefore 1 inhibits at the water splitting enzyme and also inhibits PS I partial reaction from reduced phenylmetasulfate (PMS) to methylviologen (MV). Thus, it also inhibits in the span of P700 to Iron sulfur center X (FX). Compound 2 inhibits both, the PS II partial reactions from water to SiMo and from DPC to DCPIP; besides this, it inhibits the photosystem I (PS I) partial reaction from reduced PMS to MV. With these results, we concluded that the targets of the natural product 2 are located at the water splitting enzyme, and at P680 in PS II and at the span of P700 to FX in PS I. The results of compounds 1 and 2 on PS II were corroborated by chlorophyll a fluorescence.  相似文献

Photosynthetic parameters, growth, and pigment contents were determined during expansion of the fourth leaf of in vitro photoautotrophically cultured Nicotiana tabacum L. plants at three irradiances [photosynthetically active radiation (400–700 nm): low, LI 60 μmol m−2 s−1; middle, MI 180 μmol m−2 s−1; and high, HI 270 μmol m−2 s−1]. During leaf expansion, several symptoms usually accompanying leaf senescence appeared very early in HI and then in MI plants. Symptoms of senescence in developing leaves were: decreasing chlorophyll (Chl) a+b content and Chl a/b ratio, decreasing both maximum (FV/FM) and actual (ΦPS2) photochemical efficiency of photosystem 2, and increasing non-photochemical quenching. Nevertheless, net photosynthetic oxygen evolution rate (P N) did not decrease consistently with decrease in Chl content, but exhibited a typical ontogenetic course with gradual increase. P N reached its maximum before full leaf expansion and then tended to decline. Thus excess irradiance during in vitro cultivation did not cause early start of leaf senescence, but impaired photosynthetic performance and Chl content in leaves and changed their typical ontogenetic course.  相似文献

Agronomic traits, photosynthetic pigments, gas exchange, and chlorophyll (Chl) fluorescence parameters of red stem buckwheat (Fagopyrum dibotrys Hara) mutants induced by γ-radiation were compared with green control at seedling stage. Plant height, number of first-class branches, and rhizome biomass were inhibited significantly (p<0.01). Chl a, Chl b, and Chl a+b contents decreased with elevated dose of γ-rays, while increasing carotenoid content indicated that buckwheat was capable of adjusting to the radiation damage. Decrease in net photosynthetic rate was the result of both stomatal and non-stomatal limitations. Fluorescence parameters, such as F0, Fm, Fv/Fm, Fv/F0, ΦPS2, electron transport rate, and photochemical quenching declined significantly (p<0.01) as compared with control due to photoinhibition, while non-photochemical quenching increased to enhance thermal dissipation. Lower parameters implied that leaf tissue was damaged significantly by high dose of γ-radiation and therefore leaf senescence was accelerated.  相似文献

Quantitative characteristics of photosynthetic electron transport were evaluated in vivo on the basis of the multi-exponential analysis of OJIP fluorescence transients induced by saturating actinic light. The OJIP fluorescence curve F(t), measured in Chlamydomonas reinhardtii cells, was transformed into the (1 − F O/F(t)) × (F V /F M)−1 transient, which is shown to relate to PS 2 closure. We assumed that kinetics of PS 2 closure during OJIP rise reflects time-separated processes related to the establishment of redox equilibrium at the PS 2 acceptor side (OJ), PQ pool (JI), and beyond Cyt b/f (IP). Three-exponential fitting was applied to (1 − F O/F(t)) × (F V /F M)−1 transient to obtain lifetimes and amplitudes of the OJ, JI, and IP components of PS 2 closure, which were used to calculate overall rates of reduction and re-oxidation of the PS 2 acceptor side, PQ pool, and intermediates beyond Cyt b/f complex. The results, obtained in the presence of inhibitors, oxidative reagents, and under different stress conditions prove the suggested model and characterize the introduced parameters as useful indicators of photosynthetic function.  相似文献

In Escherichia coli, the F1FO ATP synthase b subunits house a conserved arginine in the tether domain at position 36 where the subunit emerges from the membrane. Previous experiments showed that substitution of isoleucine or glutamate result in a loss of enzyme activity. Double mutants have been constructed in an attempt to achieve an intragenic suppressor of the b arg36→ile and the b arg36→glu mutations. The b arg36→ile mutation could not be suppressed. In contrast, the phenotypic defect resulting from the b arg36→glu mutation was largely suppressed in the b arg36→glu,glu39→arg double mutant. E. coli expressing the b arg36→glu,glu39→arg subunit grew well on succinate-based medium. F1FO ATP synthase complexes were more efficiently assembled and ATP driven proton pumping activity was improved. The evidence suggests that efficient coupling in F1FO ATP synthase is dependent upon a basic amino acid located at the base of the peripheral stalk.  相似文献

The effect of salt concentration (NaCl) on growth, fluorescence, photosynthetic activities and pigment content of the cyanobacterium Arthrospira platensis has been investigated over 15 days. It has been observed that high NaCl concentration induces an increase of the growth, photosynthetic efficiency (α), phycobilin/chlorophyll ratio and a slight decrease of dark respiration and compensation points. Moreover, high NaCl concentration enhances photosystem II (PSII) activity compared to photosystem I (PSI). Results show that the phycobilin-PSII energy transfer compared to the chlorophyll-PSII (F695,600/F695,440) increases. However, data obtained about the maximal efficiency of PSII photochemistry are controversial. Indeed, the Fv/Fm ratio decreases in salt adapted cultures, while at the same time the trapping flux per PSII reaction center (TR0/RC) and the probability of electron transport beyond QA (0) remain unchanged at the level of the donor and the acceptor sites of PSII. This effect can be attributed to the interference of phycobilin fluorescence with Chl a when performing polyphasic transient measurements.  相似文献

The peripheral stalk of F1F0 ATP synthase is composed of a parallel homodimer of b subunits that extends across the cytoplasmic membrane in F0 to the top of the F1 sector. The stalk serves as the stator necessary for holding F1 against movement of the rotor. A series of insertions and deletions have been engineered into the hydrophilic domain that interacts with F1. Only the hydrophobic segment from {val-121} to {ala-132} and the extreme carboxyl terminus proved to be highly sensitive to mutation. Deletions in either site apparently abolished enzyme function as a result of defects is assembly of the F1F0 complex. Other mutations manipulating the length of the sequence between these two areas had only limited effects on enzyme function. Expression of a b subunit with insertions with as few as two amino acids into the hydrophobic segment also resulted in loss of F1F0 ATP synthase. However, a fully defective b subunit with seven additional amino acids could be stabilized in a heterodimeric peripheral stalk within a functional F1F0 complex by a normal b subunit.  相似文献

Chromatic photoacclimation and photosynthesis were examined in two strains of Acaryochloris marina (MBIC11017 and CCMEE5410) and in Synechococcus PCC7942. Acaryochloris contains Chl d, which has an absorption peak at ca 710 nm in vivo. Cultures were grown in one of the three wavelengths (525 nm, 625 nm and 720 nm) of light from narrow-band photodiodes to determine the effects on pigment composition, growth rate and photosynthesis: no growth occurred in 525 nm light. Synechococcus did not grow in 720 nm light because Chl a does not absorb effectively at this long wavelength. Acaryochloris did grow in 720 nm light, although strain MBIC11017 showed a decrease in phycobilins over time. Both Synechococcus and Acaryochloris MBIC11017 showed a dramatic increase in phycobilin content when grown in 625 nm light. Acaryochloris CCMEE5410, which lacks phycobilins, would not grow satisfactorily under 625 nm light. The cells adjusted their pigment composition in response to the light spectral conditions under which they were grown. Photoacclimation and the Q y peak of Chl d could be understood in terms of the ecological niche of Acaryochloris, i.e. habitats enriched in near infrared radiation. Simultaneous measurements of chlorophyll (Chl) fluorescence and CO2 assimilation (A) in Vicia faba leaves were taken during the first weeks of growth to evaluate the protective effect of 24-epibrassinolide (EBR) against damage caused by the application of the herbicide terbutryn (Terb) at pre-emergence. V. faba seeds were incubated for 24 h in EBR solutions (2 × 10−6 or 2 × 10−5 mM) and immediately sown. Terb was applied at recommended doses (1.47 or 1.96 kg ha−1) at pre-emergence. The highest dose of Terb strongly decreased CO2 assimilation, the maximum quantum yield of PSII photochemistry in the dark-adapted state (F V/F M), the nonphotochemical quenching (NPQ), and the effective quantum yield (ΔF/FM) during the first 3–4 weeks after plant emergence. Moreover, Terb increased the basal quantum yield of nonphotochemical processes (F 0/F M), the degree of reaction center closure (1 − q p), and the fraction of light absorbed in PSII antennae that was dissipated via thermal energy dissipation in the antennae (1 − FV/FM). The herbicide also significantly reduced plant growth at the end of the experiment as well as plant length, dry weight, and number of leaves. The application of EBR to V. faba seeds before sowing strongly diminished the effect of Terb on fluorescence parameters and CO2 assimilation, which recovered 13 days after plant emergence and showed values similar to those of control plants. The protective effect of EBR on CO2 assimilation was detected at a photosynthetic photon flux density (PFD) of 650 μmol m−2 s−1 and the effect on ΔF/FM and photosynthetic electron transport (J) was detected under actinic lightings up to 1750 μmol m−2 s−1. The highest dose of EBR also counteracted the decrease in plant growth caused by Terb, and plants registered the same growth values as controls.  相似文献

Using two different inbred lines of Momordica charantia (bitter gourd), Y-106-5 and Z-1-4, the cell membrane stability, leaf water potential, pigment contents and the chlorophyll a fluorescence were investigated with different low night temperature (LNT) treatments over a 7 day time period and the sequent a 7 day recovery. Under LNT treatments, electrolyte leakage increased in both inbred lines and it increased more significantly in Y-106-5 plants than that in Z-1-4. The content of Chl b and total Chl decreased, while the Chl a/b ratio increased in stressed plants of the two lines. Almost all LNT treatments induced little change in Chl a content in Z-1-4 whereas obvious decreases in 5 and 8°C treated Y-106-5 plants were observed. Chilling changed the water status of plants and induced decreases of leaf water potential (LWP) in 5 and 8°C treated plants. LNT treatments also resulted in changes in the chlorophyll fluorescence parameters in bitter gourd leaves. The potential PSII activity (F v/F o) was reduced obviously by LNT stress and showed more sensitive to LNT than the maximum quantum efficiency of PSII primary photochemistry (F v/F m). The efficiency of open PSII centers exhibited a slight decrease whereas the photochemical quenching efficient (q P) was affected more seriously by LNT stress in both two inbred lines. The allocation of energy was rearranged by LNT stress. The light fraction used for PSII photochemistry (P) was reduced, while that used for heat dissipation (D) and the third fraction of absorbed light defines excess energy (E) increased due to the chilling stress. The impacts of LNT stress on bitter gourd generally increased with the number of LNT chilling and the severe night chilling. Plants were little affected by 12°C night chilling and the most acute damage was found in 5°C night chilling treatments. A 7 day recovery mitigated the adverse effects of LNT for both lines and almost all LNT treated plants restored to control levels except 5°C night chilling treated Y-106-5 plants. The two lines have a variance in tolerance to LNT stress and display obvious differences of phenotypes under extreme conditions.  相似文献

A pure, active cytochrome b 6 f was isolated from the chloroplasts of the marine green alga, Bryopsis corticulans. To investigate and characterize this cytochrome b 6 f complex, sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE), absorption spectra measurement and HPLC were employed. It was shown that this purified complex contained four large subunits with apparent molecular masses of 34.8, 24, 18.7 and 16.7 kD. The ratio of Cyt b 6 to Cytf was 2.01 : 1. The cytochromeb 6 f was shown to catalyze the transfer of 73 electrons from decylplastoquinol to plastocyanin–ferricyanide per Cyt f per second. α-Carotene, one kind of carotenoid that has not been found to present in cytochrome b 6 f complex, was discovered in this preparation by reversed phase HPLC. It was different from β-carotene usually found in cytochrome b 6 f complex. The configuration of the major α-carotene component was assigned to be 9-cis by resonance Raman spectroscopy. Different from the previous reports, the configuration of this α-carotene in dissociated state was determined to be all-trans. Besides this carotene, chlorophyll a was also found in this complex. It was shown that the molecular ratios of chlorophylla, cis and all-trans-α-carotene to Cyt f in this complex were 1.2, 0.7 and 0.2, respectively.  相似文献

The effects of shade on the growth, leaf photosynthetic characteristics, and chlorophyll (Chl) fluorescence parameters of Lycoris radiata var. radiata were determined under differing irradiances (15, 65, and 100% of full irradiance) within pots. The HI plants exhibited a typical decline in net photosynthetic rate (P N) during midday, which was not observed in MI- and LI plants. This indicated a possible photoinhibition in HI plants as the ratio of variable to maximum fluorescence (Fv/Fm) value was higher and the minimal fluorescence (F0) was lower in the, and LI plants. Diurnal patterns of stomatal conductance (g s) and transpiration rate (E) were remarkably similar to those of P N at each shade treatments, and the intercellular CO2 concentration (C i) had the opposite change trend. Under both shading conditions, the light saturation point, light compensation point and photon-saturated photosynthetic rate (P max) became lower than those under full sunlight, and it was the opposite for the apparent quantum yield (AQY). The higher the level of shade, the lower the integrated daytime carbon gain, stomatal and epidermis cell densities, specific leaf mass (SLM), bulb mass ratio (BMR), leaf thickness, and Chl a/b ratio. In contrast, contents of Chls per dry mass (DM), leaf area ratio (LAR), leaf mass ratio (LMR), leaf length, leaf area and total leaf area per plant increased under the same shade levels to promote photon absorption and to compensate for the lower radiant energy. Therefore, when the integrated daytime carbon gain, leaf area and total leaf area per plant, which are the main factors determining the productivity of L. radiata var. radiata plant, were taken into account together, this species may be cultivated at about 60∼70% of ambient irradiance to promote its growth.  相似文献

Diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem 2 (PS2) as well as Chl content were analyzed in Reaumuria soongorica (Pall.) Maxim., a perennial semi-shrub during dehydration and rehydration. The net photosynthetic rate (P N), maximum photochemical efficiency of PS2 (variable to maximum fluorescence ratio, Fv/Fm), quantum efficiency of non-cyclic electron transport of PS2, and Chl content decreased, but non-photochemical quenching of fluorescence and carotenoid content increased in stems with the increasing of drought stress. 6 d after re-hydration, new leaves budded from stems. In the re-watered plants, the chloroplast function was restored and Chl a fluorescence returned to a similar level as in the control plants. This improved hydraulic adjustment in plant triggered a positive effect on ion flow in the tissues and increased shoot electrical admittance. Thus R. soongorica plants are able to sustain drought stress through leaf abscission and keep part of Chl content in stems.  相似文献

We measured the responses of pigments and chlorophyll a fluorescence parameters of the Antarctic leafy liverwort Cephaloziella varians to snowmelt during austral spring 2005 at Rothera Point on the western Antarctic Peninsula. Although no changes to the concentrations of UV-B photoprotective pigments were detected during snowmelt, chlorophyll and carotenoid concentrations and maximum photosystem (PS)II yield (F v /F m) were respectively 88, 60 and 144% higher in the tissues of the liverwort that had recently emerged from snow than in those under a 10 cm depth of snow. A laboratory experiment similarly showed that effective PSII yield increased rapidly within the first 45 min after plants sampled from under snow were removed to an illuminated growth cabinet. The pigmentation and PSII yields of plants during snowmelt were also compared with those of plants in January, during the middle of the growing season at Rothera Point. During snowmelt, plants had lower F v /F m values, chlorophyll a/b ratios and concentrations of UV-B photoprotective pigments and carotenoids than during mid-season, suggesting that although there is some recovery of PSII activity and increases in concentrations of photosynthetic pigments during snowmelt, the metabolism of C. varians is restricted during this period.  相似文献

Photosynthesis, photorespiration, and chlorophyll (Chl) fluorescence in green and red Berberis thunbergii leaves were studied with two different measuring radiations, red (RR) and “white” (WR). The photosynthetic and photorespiration rates responded differently to the different radiation qualities, which indicate that the carboxylase and oxygenase activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) were affected. Differences in photosynthetic rate between the two color leaves were less under RR than under WR. However, this reduced difference in photosynthetic rate was not correlated with the stomatal response to the measuring radiation qualities. Compared with the WR, the RR reduced the differences in dark-adapted minimum and maximum fluorescence, steady-state fluorescence, light-adapted maximum fluorescence, and actual photochemical efficiency (ΦPS2) of photosystem 2 (PS2), but enlarged the difference in non-photochemical quenching between the two color leaves. Differences in both maximum quantum yield of PS2 and ratio of ΦPS2 to quantum yield of CO2 fixation between the two color leaves were similar under the two measuring radiations. To exclude disturbance of radiation attenuation caused by anthocyanins, it is better to use RR to compare the photosynthesis and Chl fluorescence in green versus red leaves.  相似文献

Syntrichia caninervis Mitt. is the dominant species in the moss crusts of the Gurbantunggut Desert, Northwestern China. We experimented with this species under controlled environmental conditions. Modulated chlorophyll (Chl) fluorescence was used to test the speed of recovery as evidenced by the time course of photosynthetic activity following remoistening. Transmission electron microscopy was used to explore the cytological characteristics of the leaf cells. Minimum and maximum fluorescence (F0 and FM) and photosynthetic yield (FV/FM) of photosystem II (PSII) recovered quickly when shoots were remoistened in the dark. This was especially the case of FV/FM; within the first minute of remoistening this reached 90% or more of the value attained after 30 min. These physiological changes were closely paralleled by cytological changes that indicated no damage to membranes or organelles. Correlation analysis showed that Chl fluorescence decreased both above and below a narrow moisture optimum. Our results underline the capability of S. caninervis to photosynthesize after remoistening. Utilizing precipitation events such as dew, fog, rain, and melting snow allows S. caninervis to survive and grow in a harsh desert environment.  相似文献

In lichens, ribitol is known as a carbon storage compound, an osmotic agens involved effectively in cell compartments protection during dehydration of lichen thalli and as a cryoprotective compound. In our study, we investigated the effect of ribitol on photochemical processes of photosynthesis in foliose lichens [Lasallia pustulata (L.) Mérat., Umbilicaria hirsuta (Sw. ex Westr.) Hoffm.] at low temperature. The effects of three concentrations of ribitol, added externally to thalli segments on several chlorophyll (Chl) fluorescence parameters, were evaluated. The 72 h exposition to 8, 16, and 26 mM ribitol led to a concentration-dependent increase in FV/FM, decrease in non-photochemical quenching (NPQ) but no change in quantum yield of photosystem II photochemistry (ΦPSII) values at −5 °C). At higher temperature (0, +5 °C), no effect of ribitol addition on the photosynthetic parameters was apparent.  相似文献

Recent studies demonstrated that titanium dioxide nanoparticles (TiO2 NPs) could significantly promote photosynthesis and plant growth, but its mechanism is still unclear. In this article, we studied the mechanism of light absorption and transfer of chloroplasts of Arabidopsis thaliana caused by TiO2 NPs treated. The results showed that TiO2 NPs could induce significant increases of light-harvesting complex II (LHCII) b gene expression and LHCII II content on the thylakoid membrane in A. thaliana, and the increases in LHCII were higher than the non-nano TiO2 (bulk-TiO2) treatment. Meanwhile, spectroscopy assays indicated that TiO2 NPs obviously increased the absorption peak intensity of the chloroplast in red and blue region, the fluorescence quantum yield near 680 nm, the excitation peak intensity near 440 and 480 nm and/or near 650 and 680 nm of the chloroplast. TiO2 NPs treatment could reduce F 480/F 440 ratio and increase F 650/F 680 ratio and accelerate the rate of whole chain electron transport and oxygen evolution of the chloroplast. However, the photosynthesis improvement of the non-nanoTiO2 treatment was far less effective than TiO2 NPs treatment. Taken together, TiO2 NPs could promote the light absorption of chloroplast, regulate the distribution of light energy from PS I to PS II by increasing LHCII and accelerate the transformation from light energy to electronic energy, water photolysis, and oxygen evolution.  相似文献
