Marine invasions are a worldwide problem that involves changes in communities and the acclimation of organisms to them. The invasive Chlorophyte Caulerpa racemosa var. cylindracea is widespread in the Mediterranean and colonises large areas from 0 to 70 m in depth. The omnivorous fish Spondyliosoma cantharus presents a high frequency of occurrence of C. racemosa in the stomach contents at invaded areas (76.3%) while no presence of C. racemosa was detected in control areas. The isotopic composition of muscle differed significantly between invaded and non-invaded sites for δ13C (− 16.67‰ ± 0.09 and − 17.67‰ ± 0.08, respectively), δ15N (10.22‰ ± 0.22 and 9.32‰ ± 0.18, respectively) and the C:N ratio (2.01 ± 0.0002 and 1.96 ± 0.009, respectively). Despite the high frequency of occurrence of C. racemosa in the stomach contents of S. cantharus and its important contribution to the δ13C source (20.7% ± 16.2), the contribution of C. racemosa to the δ15N in S. cantharus food sources was very low (6.6% ± 5.8). Other invertebrate prey such as decapods and polychaetes were more important contributors to the δ15N source at both invaded and non-invaded sites. Activation of enzymatic pathways (catalase, superoxide dismutase, glutathione-s-tranferase, 7-ethoxy resorufin O-de-ethylase) but not a significant increase in lipid peroxidation MDA (0.49 ± 0.01 nmol/mg prot at non-invaded and 0.53 ± 0.01 nmol/mg prot at invaded sites) was observed in S. cantharus individuals living in C. racemosa-invaded sites compared with control specimens. The low δ15N contribution values of C. racemosa by S. cantharus together with the toxicity demonstrated by the activation of the antioxidant defences and the important contribution of invertebrate prey to the δ15N could mean that the ingestion of C. racemosa by S. cantharus might be unintentional during the predation of invertebrate preys living underneath the entanglement of the C. racemosa fronds and stolons mats.  相似文献

The kinetic properties of a microsomal gill (Na+,K+)-ATPase from the freshwater shrimp, Macrobrachium olfersii, acclimated to 21‰ salinity for 10 days were investigated using the substrate p-nitrophenylphosphate. The enzyme hydrolyzed this substrate obeying cooperative kinetics at a rate of 123.6 ± 4.9 U mg− 1 and K0.5 = 1.31 ± 0.05 mmol L− 1. Stimulation of K+-phosphatase activity by magnesium (Vmax = 125.3 ± 7.5 U mg− 1; K0.5 = 2.09 ± 0.06 mmol L− 1), potassium (Vmax = 134.2 ± 6.7 U mg− 1; K0.5 = 1.33 ± 0.06 mmol L− 1) and ammonium ions (Vmax = 130.1 ± 5.9 U mg− 1; K0.5 = 11.4 ± 0.5 mmol L− 1) was also cooperative. While orthovanadate abolished p-nitrophenylphosphatase activity, ouabain inhibition reached 80% (KI = 304.9 ± 18.3 μmol L− 1). The kinetic parameters estimated differ significantly from those for freshwater-acclimated shrimps, suggesting expression of different isoenzymes during salinity adaptation. Despite the ≈2-fold reduction in K+-phosphatase specific activity, Western blotting analysis revealed similar α-subunit expression in gill tissue from shrimps acclimated to 21‰ salinity or fresh water, although expression of phosphate-hydrolyzing enzymes other than (Na+,K+)-ATPase was stimulated by high salinity acclimation.  相似文献

The trophic ecology of marine vertebrates has been increasingly studied via stable isotope analysis of body tissues. However, the theoretical basis for using stable isotopes to elucidate consumer–prey relationships remains poorly validated for most taxa despite numerous studies using this technique in natural systems. In this study, we measured stable carbon and stable nitrogen diet-tissue discrimination (Δdt) in whole blood, red blood cells, blood plasma solutes, and skin of leatherback sea turtles (Dermochelys coriacea; N = 7) maintained in captivity for up to 424 days and fed an isotopically consistent control diet with a mean C:N ratio of 2.94:1.00 and an energetic content of 20.16 ± 0.39 kJ g− 1 Dry Mass. We used a random-effect repeated measure model to evaluate isotopic consistency among tissue samples collected on days 276, 348, and 424. Both δ13C and δ15N remained consistent among sampling events in all tissues (all 95% posterior intervals for the slopes of a linear model included zero), indicating that all tissues had fully integrated diet-derived stable isotope compositions. Mean tissue-specific δ13C ranged from − 18.30 ± 0.16‰ (plasma solutes) to − 15.54 ± 0.14‰ (skin), whereas mean δ15N was from 10.06 ± 0.22‰ (whole blood) to 11.46 ± 0.10‰ (plasma solutes). The computed Δdt factors for carbon ranged from − 0.58‰ (plasma solutes) to + 2.25‰ (skin), whereas Δdt for nitrogen was from + 1.49 (red blood cells) to + 2.85 (plasma solutes). As the only discrimination factors available for leatherback turtles, our data will be useful for future interpretations of field-derived stable isotope data for this species. The inherent variability in Δdt values among individuals was low, which supports the value of these data for dietary reconstructions. However, it is important to note that tissue-specific discrimination factors for leatherbacks contrast with the widely accepted values for endothermic species (0–1‰ for C, 3–5‰ for N), and are also different from values established for hard-shelled turtles. This underscores the need for species- and tissue-specific discrimination factors before interpreting trophic studies of wild animals, including marine turtles.  相似文献

The extracellular β-agarase LSL-1 produced by an agar-liquefying, soil bacterium Acinetobacter sp., AG LSL-1 was purified to homogeneity by combination of ion-exchange and size exclusion chromatography with final yield of 44%. The enzyme has a specific activity of 397 U mg−1 protein and with a molecular mass of 100 kDa. The agarase was active in the pH range of 5.0–9.0, optimally at pH 6.0 and temperature between 25 °C and 55 °C and optimal at 40 °C. The enzyme retained 63% of native activity at 50 °C suggesting it is a thermostable. The activity of the agarase was completely inhibited by metal ions, Hg2+, Ag+ and Cu2+, whereas 25–40% of native activity was retained in the presence of Zn2+, Sn2+ and SDS. Neoagarobiose was the final product of hydrolysis of both agarose and neoagarohexaose by the purified agarase LSL-1. Based on the molecular mass and final products of agarose hydrolysis, the β-agarase LSL-1 may be further grouped under group III β-agarases and may be a member of GH-50 family. This is the first report on the purification and biochemical characterization of β-agarase from an agar-liquefying Acinetobacter species.  相似文献

The psychrotolerant bacterium Shewanella sp. G5 was used to study differential protein expression on glucose and cellobiose as carbon sources in cold-adapted conditions. This strain was able to growth at 4 °C, but reached the maximal specific growth rate at 37 °C, exhibiting similar growing rates values with glucose (μ: 0.4 h−1) and cellobiose (μ: 0.48 h−1). However, it grew at 15 °C approximately in 30 h, with specific growing rates of 0.25 and 0.19 h−1 for cellobiose and glucose, respectively. Thus, this temperature was used to provide conditions related to the environment where the organism was originally isolated, the intestinal content of Munida subrrugosa in the Beagle Channel, Fire Land, Argentina. Cellobiose was reported as a carbon source more frequently available in marine environments close to shore, and its degradation requires the enzyme β-glucosidase. Therefore, this enzymatic activity was used as a marker of cellobiose catabolism. Zymogram analysis showed the presence of cold-adapted β-glucosidase activity bands in the cell wall as well as in the cytoplasm cell fractions. Two-dimensional gel electrophoresis of the whole protein pattern of Shewanella sp. G5 revealed 59 and 55 different spots induced by cellobiose and glucose, respectively. Identification of the quantitatively more relevant proteins suggested that different master regulation schemes are involved in response to glucose and cellobiose carbon sources. Both, physiological and proteomic analyses could show that Shewanella sp. G5 re-organizes its metabolism in response to low temperature (15 °C) with significant differences in the presence of these two carbon sources.  相似文献

Seasonal changes of field populations and growth rates of two dinoflagellates, Ceratium furca and Ceratium fusus, were examined in the temperate coastal water of Sagami Bay, Japan. Weekly field sampling was conducted from August 2002 to August 2003, and laboratory experiments were also carried out to investigate effects of temperature, irradiance and photoperiod on the growth rates of these two Ceratium species. In the field, the abundances of both species increased significantly from April to August 2003, were gradually decreased from November 2002 and were not observed in January 2003. C. fusus was able to increase at lower temperatures in February 2003 compared to C. furca. In the laboratory, the two species did not grow at <10 °C or >32 °C. The highest specific growth rate of C. furca was 0.72 d−1 at 24 °C and 600 μmol m−2 s−1. Optimum growth rates (>0.4 d−1) of C. furca were observed at temperatures from 18 to 28 °C and at irradiances from 216 to 796 μmol m−2 s−1. The highest growth rate of C. fusus was 0.56 d−1 at 26 °C and 216 μmol m−2 s−1. Optimum growth rates of C. fusus were observed at the same irradiance rage of C. furca, whereas optimum temperature range was narrower (26–28 °C). The growth curves of both species indicated saturation of the growth rates when light intensity was above 216 μmol m−2 s−1, and did not show photoinhibition at irradiances up to 796 μmol m−2 s−1. The specific growth rates of both Ceratium species were clearly decreased at L:D = 10:14 relative to those at L:D = 14:10 and L:D = 12:12. The present study indicates the two Ceratium species can adapt to a wide range of temperature and irradiance.  相似文献

An intracellular β-xylosidase from the thermophilic fungus Sporotricum thermophile strain ATCC 34628 was purified to homogeneity by Q-Sepharose and Mono-Q column chromatographies. The protein properties correspond to molecular mass and pI values of 45 kDa and 4.2, respectively. The enzyme is optimally active at pH 7.0 and 50 °C. The purified β-xylosidase is fully stable at pH 6.0–8.0 and temperatures up to 50 °C and retained over 58% of its activity after 1 h at 60 °C. The enzyme hydrolyzes β-1,4-linked xylo-oligosaccharides with chain lengths from 2 to 6, releasing xylose from the non-reducing end, but is inactive against xylan substrates. The apparent Km and Vmax values from p-nitrophenyl β-d-xylopyranoside are 1.1 mM and 114 μmol p-nitrophenol min−1 mg−1, respectively. Alcohols inactivate the enzyme, ethanol at 10% (v/v) yields a 30% decrease of its activity. The enzyme is irreversibly inhibited by 2,3-epoxypropyl β-d-xylobioside while alkyl epoxides derived from d-xylose were not inhibitors of the enzyme. The enzyme catalyses the condensation reaction using high donor concentration, up to 60% (w/v) xylose.  相似文献

A rain exclusion experiment simulating drought conditions expected in Mediterranean areas for the following decades (15% decrease in soil moisture) was conducted in a Mediterranean holm oak forest to study the response of leaf δ13C, δ15N, and N concentrations to the predicted climatic changes for the coming decades. Plant material was sampled in 2000, 2003, 2004, and 2005 in eight plots: four of them were control plots and the other four plots received the rain exclusion treatment. Although there was a negative relationship between δ13C and soil moisture, for each species and year, the rain exclusion treatment did not have any significant effect on δ13C, and therefore on the intrinsic water use efficiency (iWUE) of the three dominant species: Phillyrea latifolia, Arbutus unedo, and Quercus ilex. On the other hand, rain exclusion clearly increased the δ15N values in the three species studied, probably indicating higher N losses at the soil level leading to a 15N enrichment of the available N. It suggested that rain exclusion exerted a greater effect on the nitrogen biogeochemical cycle than on the carbon assimilation process. δ15N values were inversely correlated with summer soil moisture in Q. ilex and A. unedo, but no relationship was observed in P. latifolia. This latter species showed the lowest iWUE values, but it was the only species with no decrease in annual basal increment in response to the rain exclusion treatment, and it also had the highest resistance to the hot and dry conditions projected for the Mediterranean basin in the coming decades. The different strategies to resist rain exclusion conditions of these species could induce changes in their competitive ability and future distribution. The losses of N from the ecosystem may further limit plant growth and ecosystem functioning.  相似文献

A novel β-glucosidase from Fusarium proliferatum ECU2042 (FPG) was successfully purified to homogeneity with a 506-fold increase in specific activity. The molecular mass of the native purified enzyme (FPG) was estimated to be approximately 78.7 kDa, with two homogeneous subunits of 39.1 kDa, and the pI of this enzyme was 4.4, as measured by two-dimensional electrophoresis. The optimal activities of FPG occurred at pH 5.0 and 50 °C, respectively. The enzyme was stable at pH 4.0–6.5 and temperatures below 60 °C, and the deactivation energy (Ed) for FPG was 88.6 kJ mo1−1. Moreover, it was interesting to find that although the purified enzyme exhibited a very low activity towards p-nitrophenyl β-d-glucoside (pNPG), and almost no activity towards cellobiose, a relatively high activity was observed on ginsenoside Rg3. The enzyme hydrolyzed the 3-C, β-(1 → 2)-glucoside of ginsenoside Rg3 to produce ginsenoside Rh2, but did not sequentially hydrolyze the β-d-glucosidic bond of Rh2. The Km and Vmax values of FPG for ginsenoside Rg3 were 2.37 mM and 0.568 μmol (h mg protein)−1, respectively. In addition, this enzyme also exhibited significant activities towards various alkyl glucosides, aryl glucosides and several natural glycosides.  相似文献

Escherihica coliumC122::Tn5 cells were γ-radiated (137Cs, 750 Gy, under N2), and lac-constitutive mutants were produced at 36% of the wild-type level (the umC strain was not deficient in spontaneous mutagenesis, and the mutational spectrum determined by sequencing 263 spontaneous lacId mutations was very similar to that for the wild-type strain). The specific nature of the umC strain's partial radiation was determined by sequencing 325 radiation-induced lacId mutations. The yields of radiation-induced mutation classes in the umC strain (as a percentage of the wild-type yield) were: 80% for A · T → G · C transitions, 70% for multi-base additions, 60% for single-base deletions, 53% for A · T → C · G transversions, 36% for G · C → A · T transitions, 25% for multi-base deletions, 21% for A · T → T · A transversions, 11% for G · C → C · G transversions, 9% for G · C → T · A transversions and 0% for multiple mutations. Based on these deficiencies and other factors, it is concluded that the umC strain is near-normal for A · T → G · C transitions, single-base deletions and possibly A · T → C · G transversions; is generally deficient for mutagenesis at G · C sites fro transversions, and is grossly deficient in multiple mutations. Damage at G · C sites seems more difficult for translesion DNA synthesis to bypass than damage at A · T sites, and especially when trying to produced a transversion. The yield of G · C → A · T transitions in the umC strain *36% of the wild-type level) argues that a basic sites are involved in no more than 64% of γ-radiation-induced base substitutions in the wild-type strain. Altogether, these data suggest that the UmuC and UmuD′ proteins facilitate, rather than being absolutely required for, translesion DNA synthesis; with the degree of facilitation being dependent both on the nature of the noncoding DNA damage, i.e., at G · C vs A · T sites, and on the nature of the misincorporated base, i.e., whether it induces transversions or transitions.  相似文献

Much of the historical Everglades has been either lost or degraded as a result of human activities. Among the aquatic habitats that comprise the Everglades landscape mosaic, open-water sloughs support critical ecological functions and appear especially sensitive to both hydrologic and water-quality perturbations. We used a combination of remote sensing and on-the-ground sampling to document spatial changes in the extent and vegetative composition of sloughs along a phosphorus (P) gradient in the northern Everglades. Increasing levels of water and soil P were associated with a decline in slough coverage, loss of the abundant native periphyton community, and a shift in dominant macrophyte species. The characteristic slough macrophyte species Eleocharis cellulosa and Nymphaea odorata exhibited different sensitivities to P enrichment, but both species declined with enrichment as slough habitats were invaded by Typha domingensis, a species that is known to expand aggressively in response to enrichment. A limited amount of open-water habitat occurred in highly enriched areas, but these habitats were maintained largely as a result of airboat disturbance and did not contain characteristic slough vegetation. Many changes in slough coverage and composition occurred in areas where water and soil P concentrations were only marginally higher than background levels. Our findings support the need for Everglades hydrologic restoration efforts to adhere to strict water-quality standards for P to avoid further degradation of this key landscape feature. Protein synthesis is an essential growth process in all animals. Little information is available on post-prandial protein synthesis and even less where different protein sources are compared. Protein synthesis was measured at 4 and 24 h after feeding juvenile barramundi in order to determine the effect of using lupin as a partial protein replacement for fish meal on the post-prandial protein metabolism. Juvenile barramundi (4.3 ±0.6 g) were held in a recirculation system (27 °C, salinity 10‰ and 24 h light) for 15 days. Fish were fed one of two isonitrogenous isoenergetic diets (40% crude protein, 16% lipid and 18.5 GE MJ kg− 1). One diet was formulated with 100% fish meal as the protein source while the other had 45% of the protein replaced with lupin ingredients (lupin kernel meal (Lupinus angustifolius) and lupin protein concentrate). All fish were fed a ration of 6%·d− 1 and feed intake was not significantly different between the two diets. Specific growth rate (SGR) and growth efficiency (in relation to protein (PPV) and energy (PEV)) were 6.5 ± 0.14%·d− 1, 43.8 ± 2.72% and 38.31 ± 1.56%, respectively, and were not significantly different between the two diets. There was no significant difference in protein synthesis between the two diets at 4 and 24 h after feeding, however protein synthesis was significantly higher 4 h after feeding than at 24 h (p = 0.02). Neither growth performance nor protein metabolism was altered by replacing 45% of the protein with lupin protein and indicated this to be a suitable protein source for barramundi feeds.  相似文献

The sediment cores 225514 and 225510 were recovered from 420 and 285 m water depth, respectively. They were investigated for their benthic foraminiferal δ13C during the last 500 years. Both cores were recovered from the southern flank of the Skagerrak. The δ13C values of Uvigerina mediterranea and other shallow infaunal species in both cores indicate that organic matter rain rates to the seafloor varied around a mean value until approximately AD 1950 after which they increased. This increase might result from changes in the North Atlantic Current System and a co-occurring persistently high North Atlantic Oscillation index state in the 1980s to 1990s, rather than from anthropogenic eutrophication. Using δ13C mean values of multiple species, we reconstruct δ13C gradients of dissolved inorganic carbon (DIC) within pore waters for the time periods AD 1500 to 1950 and AD 1950 to 2000. The calculated δ13CDIC ranges, interpreted as indicating total organic matter remineralization due to respiration, are generally bigger in Core 225514 than in Core 225510. Since mean δ13C values of U. mediterranea suggest that organic matter rain rates were similar at both locations, differences in total organic matter remineralization are attributed to differing oxygen availability. However, oxygen concentrations in the overlying bottom water masses are not likely to have differed significantly. Thus, we suggest that organic matter remineralization was controlled by oxygen availability within the sediments, reflecting strong differences in sedimentation rates at the two investigated core sites. Based on the assumptions that tests of benthic foraminiferal species inhabiting the same microhabitat depth should show equal δ13C values unless they are affected by vital effects and that Globobulimina turgida records pore water δ13CDIC, we estimate microhabitat-corrected vital effects for several species with respect to G. turgida: > 0.7‰ for Cassidulina laevigata, > 1.3‰ for Hyalinea balthica, and > 0.7‰ for Melonis barleeanus. Melonis zaandami seems to closely record pore water δ13CDIC.  相似文献

γ-Aminobutyraldehyde dehydrogenase from Escherichia coli K-12 has been purified and characterized from cell mutants able to grow in putrescine as the sole carbon and nitrogen source. The enzyme has an Mr of 195 000±10 000 in its dimeric form with an Mr of 95 000±1000 for each subunit, a pH optimum at 5.4 in sodium citrate buffer, and does not require bivalent cations for its activity. Km values are 31.3±6.8 μM and 53.8±7.4 μM for Δ-1-pyrroline and NAD+, respectively. An inhibitory capacity for NADH is also shown using the purified enzyme.  相似文献

Human α1-antitrypsin (AAT) was produced in the recombinant yeast Saccharomyces cerevisiae ATCC 20699 grown in batch and fed-batch culture. The final biomass concentration and antitrypsin concentration attained were 55 g·L−1 and 1.23 g·L−1, respectively, in the fed-batch. The maximum productivities of biomass and antitrypsin were 1.6 and > 0.04 g L−1h−1, respectively, or substantially greater than the highest productivity values reported in the past. For recovering the antitrypsin, the cell slurry was concentrated 4-fold (231 g·L−1 biomass, 122 min of processing) by cross-flow microfiltration and the cells were disrupted by bead milling (3 passes of 3 min total retention time). The cell homogenate was treated with aluminum chloride or PBS (pH 7) to aid separation of the cell debris by flocculation and sedimentation. The clarified cell homogenate was subjected to ammonium sulfate fractionation to precipitate the recombinant antitrypsin. The AAT precipitated at 45–75% saturation of ammonium sulfate, depending on the age of the homogenate. The crude AAT in the homogenate degraded at room temperature (25°C), with a zero order deactivation rate of 1.815 × 10−3 ± 3.43 × 10−4 g AAT L−1h−1.  相似文献

Arbutin (Ab, 4-hydroxyphenyl β-glucopyranoside) is a glycosylated hydroquinone known to prevent the formation of melanin by inhibiting tyrosinase. An arbutin-α-glucoside was synthesized by the transglycosylation reaction of amylosucrase (AS) of Deinococcus geothermalis (DGAS) using arbutin and sucrose as an acceptor and a donor, respectively. The maximum yield of the arbutin transglycosylation product was determined to be over 98% with a 1:0.5 molar ratio of donor and acceptor molecules (sucrose and arbutin), in 50 mM sodium citrate buffer pH 7 at 35 °C. TLC and HPLC analyses revealed that only one transglycosylation product was observed, supporting the result that the transglycosylation reaction of DGAS was very specific. The arbutin transglycosylation product was isolated by preparative recycling HPLC. The structural analyses using 13C and 1H NMR proved that the transglycosylated product was 4-hydroxyphenyl β-maltoside (Ab-α-glucoside), in which a glucose molecule was linked to arbutin via an α-(1 → 4)-glycosidic linkage.  相似文献

Effects of carbon concentration and carbon to nitrogen (C:N) ratio on six biocontrol fungal strains are reported in this paper. All fungal strains had extensive growth on the media supplemented with 6–12 g l−1 carbon and C:N ratios from 10:1 to 80:1, and differed in nutrient requirements for sporulation. Except for the two strains of Paecilomyces lilacinus, all selected fungi attained the highest spore yields at a C:N ratio of 160:1 when the carbon concentration was 12 g l−1 for Metarhizium anisopliae SQZ-1-21, 6 g l−1 for M. anisopliae RS-4-1 and Trichoderma viride TV-1, and 8 g l−1 for Lecanicillium lecanii CA-1-G. The optimal conditions for P. lilacinus sporulation were 8 g l−1 carbon with a C:N ratio of 10:1 for M-14 and 12 g l−1 carbon with a C:N ratio of 20:1 for IPC-P, respectively. The results indicated that the influence of carbon concentration and C:N ratio on fungal growth and sporulation is strain dependent; therefore, consideration for the complexity of nutrient requirements is essential for improving yields of fungal biocontrol agents.  相似文献

Recent novel mixed blooms of several species of toxic raphidophytes have caused fish kills and raised health concerns in the highly eutrophic Inland Bays of Delaware, USA. The factors that control their growth and dominance are not clear, including how these multi-species HAB events can persist without competitive exclusion occurring. We compared and contrasted the relative environmental niches of sympatric Chattonella subsalsa and Heterosigma akashiwo isolates from the bays using classic Monod-type experiments. C. subsalsa grew over a temperature range from 10 to 30 °C and a salinity range of 5–30 psu, with optimal growth occurring from 20 to 30 °C and 15 to 25 psu. H. akashiwo had similar upper temperature and salinity tolerances but also lower limits, with growth occurring from 4 to 30 °C and 5 to 30 psu and optimal growth between 16 and 30 °C and 10 and 30 psu. These culture results were confirmed by field observations of bloom occurrences in the Inland Bays. Maximum nutrient-saturated growth rates (μmax) for C. subsalsa were 0.6 d−1 and half-saturation concentrations for growth (Ks) were 9 μM for nitrate, 1.5 μM for ammonium, and 0.8 μM for phosphate. μmax of H. akashiwo (0.7 d−1) was slightly higher than C. subsalsa, but Ks values were nearly an order of magnitude lower at 0.3 μM for nitrate, 0.3 μM for ammonium, and 0.2 μM for phosphate. H. akashiwo is able to grow on urea but C. subsalsa cannot, while both can use glutamic acid. Cell yield experiments at environmentally relevant levels suggested an apparent preference by C. subsalsa for ammonium as a nitrogen source, while H. akashiwo produced more biomass on nitrate. Light intensity affected both species similarly, with the same growth responses for each over a range from 100 to 600 μmol photons m−2 s−1. Factors not examined here may allow C. subsalsa to persist during multi-species blooms in the bays, despite being competitively inferior to H. akashiwo under most conditions of nutrient availability, temperature, and salinity.  相似文献

POMC-derived peptides and mRNA have been identified in heart tissue, although POMC processing has not been fully characterized. In the present study, we found that β-lipotropin and ACTH were localized in rat heart, although they were almost entirely converted to β-endorphin- and α-MSH-related peptides. Ion exchange HPLC analysis revealed that β-endorphin(1–31) was further processed to α-N-acetyl-β-endorphin(1–31), which comprised 35.9 ± 0.1% of total immunoreactivity, and smaller amounts of β-endorphin(1–27), β-endorphin(1–26), and their α-N-acetylated derivatives. The predominant α-MSH immunoreactive peptides coeluted with α-MSH and N,O-diacetyl-α-MSH by reverse-phase HPLC, although small amounts of ACTH(1–13)-NH2 were also present. Thus, multiple forms of β-endorphin and α-MSH are localized in rat heart. β-Endorphin(1–31) is a minor constituent, however, indicating that nonopioid β-endorphin peptides predominate.  相似文献

The dynamics of β-xylosidase biosynthesis from Aspergillus niger B 03 was investigated in laboratory bioreactor. Maximum xylosidase activity 5.5 U/ml was achieved after 80 h fermentation at medium pH 4.0. The isolated β-xylosidase was immobilized on polyamide membrane support and the basic characteristics of the immobilized enzyme were determined. Maximum immobilization and activity yield obtained was 30.0 and 6.8%, respectively. A shift in temperature optimum and pH optimum was observed for immobilized β-xylosidase compared to the free enzyme. Immobilized enzyme exhibited maximum activity at 45 °C and pH 4.5 while its free counterpart at 70 °C and pH 3.5, respectively. Thermal stability at 40 and 50 °C and storage stability of immobilized β-xylosidase were investigated at pH 5.0. Kinetic parameters Km, Vmax and Ki were determined for both enzyme forms. Free and immobilized β-xylosidase were tested for xylose production from birchwood xylan. The substrate was preliminarily depolymerized with xylanase to xylooligosaccharides and the amount of xylose obtained after their hydrolysis with free and immobilized β-xylosidase was determined by HPLC analysis. Continuous enzyme hydrolysis of birchwood xylan was performed with xylanase and free or immobilized β-xylosidase. The maximum extent of hydrolysis was 25 and 30% with free and immobilized enzyme, respectively. Immobilized preparation was also examined for reusability in 20 consecutive cycles at 40 °C.  相似文献
