This study is the most extensive molecular study of the gastropod families Trochidae and Calliostomatidae published to date, in terms of both numbers of taxa and of gene sequences. As a result of Bayesian phylogenetic analyses of molecular sequence data from one nuclear gene and three mitochondrial genes, we propose dramatic changes to Trochidae family systematics, present the first molecular phylogeny for Calliostomatidae and include the first published sequence data for the enigmatic subfamily Thysanodontinae. Our phylogeny demonstrates that within the family Trochidae there is strong support for three subfamilies new to traditional classifications: Alcyninae subfam. nov., Fossarininae and Chrysostomatinae subfam. nov. As proposed, Alcyninae consists only of the nominotypical genus Alcyna, which is sister to all other trochids. The subfamily Fossarininae, as defined here, includes Fossarina, Broderipia, Synaptocochlea and “Royaeximia and probably also Clydonochilus and Minopa. The subfamily Chrysostomatinae comprises the genera Chrysostoma and Chlorodiloma. Additional molecular support is also obtained for recently redefined Trochinae, Monodontinae, and Cantharidinae and for the traditionally recognised subfamilies Umboniinae and Stomatellinae. The subfamily Lirulariinae is not supported by the molecular data, but rather is incorporated into Umboniinae. We also demonstrate that the current concept of the subfamily Margaritinae (previously a trochid subfamily, but recently and provisionally assigned to Turbinidae) is not monophyletic. We provide preliminary evidence that whereas Margarella rosea (previously a member of Margaritinae) belongs in the trochid subfamily Cantharidinae, its presumptive congener M. antarctica is not a trochid, but instead clusters with the thysanodontine genus Carinastele. Based on the phylogenetic placement of C. kristelleae, we agree with previous proposals based on morphological data that Thysanodontinae are more closely related to Calliostomatidae than Trochidae. Both Calliostoma and Carinastele are carnivorous and if a sister relationship can be confirmed between Carinastele and Margarella antarctica it might mean that carnivory evolved twice in Trochoidea. The direction of dietary changes was not investigated in this study, but mapping diet onto the phylogeny suggests that true herbivory is predominantly a derived character. The new classification system also means that five trochid subfamilies are predominantly associated with hard substrata, one with soft substrata (Umboniinae) and two with algae and seagrass (Alcyninae and Cantharidinae). There has been a shift back to hard substrata in one umboniine clade. Two of three clades within Calliostomatidae were predominantly associated with hard substrata, but one Japanese clade is associated with sand. The finding of three new, unidentified species from very deep water means that Trochidae, like Calliostomatidae, now includes species found at bathyal depths. More deep-water species may be found as increased sampling leads to the discovery of new species.  相似文献

The first fossil nemonychid (Nemonychidae) in Burmese amber, belonging to the subfamily Rhinorhynchinae, is described and figured as Burmonyx zigrasi Davis & Engel, gen. n. and sp. n. While this specimen also comprises the first definitive record of the subfamily in the Asian continent, other compression fossils exist at least from the Yixian Formation of China and the Karatau site of Kazakhstan which may also deserve placement within this group. Although several important areas of the body are obscured by the shape and fragmented condition of the amber piece, a sufficient number of features are visible to consider adequate placement within Rhinorhynchinae, including the fairly strongly punctate elytral striae and appendiculate, nearly bifid pretarsal claws.  相似文献

The maize ??-glucosidase (ZmGLU1) hydrolyzes cytokinin-conjugates for releasing active cytokinins and thus plays important roles in cytokinin regulatory processes. ZmGLU1 belongs to glycosyl hydrolases 1 (GH1) gene family with a large number of members, and the gene function of other homologs remains to be investigated. In this study, 47 Arabidopsis, 34 rice, 31 brachypodium, 28 sorghum and 26 maize GH1 protein sequences were collected and subsequently used to construct a phylogenetic tree by Neighbor-Joining method. ZmGLU1 together with its 7 paralogs and 4 sorghum homologs were assigned into a distinct group (named GLU subfamily) with far evolutionary distance to other GH1 members. None of the Arabidopsis, rice and brachypodium gene falling into this group indicated a recent evolutionary emergence of GLU subfamily in some Poaceae plants after the divergence of Poaceae species. Phylogeny and comparative genome analysis revealed that GLU subfamily members of maize and sorghum evolved from a common ancestor, and expanded independently in each species by several duplications after maize-sorghum split. Ka/Ks analysis showed that purifying selection played important roles in maintenance of similar functions among the maize GLU paralogs. In addition, the similar protein properties and cytokinin-dependent gene expressions further suggested the similar functions of ZmGLUs in cytokinin activation. However, the organ-dependent expression of ZmGLUs exhibited diverse patterns, which might contribute to their diverse roles in cytokinin homeostasis. Taken together, this work put new insights into the evolution and expression of ZmGLU genes, and provided the foundation for future functional investigations.  相似文献

The nongreen plastids, such as etioplasts, chromoplasts, etc., as well as chloroplasts, are all derived from proplastids in the meristem. To date, the Min system members in plants have been identified as regulators of FtsZ-ring placement, which are essential for the symmetrical division of chloroplasts. However, the regulation of FtsZ-ring placement in nongreen plastids is poorly understood. In this study, we investigated the division site placement of nongreen plastids by examining the etioplasts as representative in Arabidopsis Min system mutants. Surprisingly, the shape and number of etioplasts in cotyledons of arc3, arc11 and mcd1 mutants were similar to that observed in wild-type plants, whereas arc12 and parc6 mutants exhibited enlarged etioplasts that were reduced in number. In order to examine nongreen plastids in true leaves, we silenced the ALB3 gene in these Min system mutant backgrounds to produce immature chloroplasts without the thylakoidal network using virus induced gene silencing (VIGS). Interestingly, consistent with our observations in etioplasts, enlarged and fewer nongreen plastids were only detected in leaves of parc6 (VIGS-ALB3) and arc12 (VIGS-ALB3) plants. Further, the FtsZ-ring assembled properly at the midpoint in nongreen plastids of arc3, arc11 and mcd1 (VIGS-ALB3) plants, but organized into multiple rings in parc6 (VIGS-ALB3) and presented fragmented filaments in arc12 (VIGS-ALB3) plants, suggesting that division site placement in nongreen plastids requires fewer components of the plant Min system. Taken together, these results suggest that division site placement in nongreen plastids is different from that in chloroplasts.  相似文献

Recently, two novel mammalian aquaporins (AQPs), AQPs 11 and 12, have been identified and classified as members of a new AQP subfamily, the “subcellular AQPs”. In members of this subfamily one of the two asparagine-proline-alanine (NPA) motifs, which play a crucial role in selective water conduction, are not completely conserved. Mouse AQP11 (mAQP11) was expressed in Sf9 cells and purified using the detergent Fos-choline 10. The protein was reconstituted into liposomes, which were used for water conduction studies with a stopped-flow device. Single water permeability (pf) of AQP11 was measured to be 1.72 ± 0.03 × 10− 13 cm3/s, suggesting that other members of the subfamily with incompletely conserved NPA motifs may also function as water channels.  相似文献

The E. coli mechanosensitive (MS) channel of small conductance (EcMscS) is the prototype of a diverse family of channels present in all domains of life. While EcMscS has been extensively studied, recent developments show that MscS may display some characteristics not widely conserved in this protein subfamily. With numerous members now electrophysiologically characterized, this subfamily of channels displays a breadth of ion selectivity with both anion and cation selective members. The selectivity of these channels may be relatively weak in comparison to voltage-gated channels but their selectivity mechanisms represent great novelty. Recent studies have identified unexpected residues important for selectivity in these homologs revealing different selectivity mechanisms than those employed by voltage gated K+, Na+, Ca2+ and Cl- channels whose selectivity filters are housed within their transmembrane pores. This commentary looks at what is currently known about MscS subfamily selectivity and begins to unravel the potential physiological relevance of these differences.  相似文献

The ruminal bacterium Butyrivibrio fibrisolvens is being engineered by the introduction of heterologous xylanase genes in an attempt to improve the utilization of plant material in ruminants. However, relatively little is known about the diversity and distribution of the native xylanase genes in strains of B. fibrisolvens. In order to identify the most appropriate hosts for such modifications, the xylanase genotypes of 28 strains from the three 16S ribosomal DNA (rDNA) subgroups of Butyrivibrio fibrisolvens have been investigated. Only 4 of the 20 strains from 16S rDNA group 2 contained homologues of the strain Bu49 xynA gene. However, these four xynA-containing strains, and two other group 2 strains, contained members of a second xylanase gene family clearly related to xynA (subfamily I). Homologues of xynB, a second previously described xylanase gene from B. fibrisolvens, were identified only in three of the seven group 1 strains and not in the group 2 and 3 strains. However, six of the group 1 strains contained one or more members of the two subfamilies of homologues of xynA. The distribution of genes and the nucleotide sequence relationships between the members of the two xynA subfamilies are consistent with the progenitor of all strains of B. fibrisolvens having contained a xynA subfamily I gene. Since many xylanolytic strains of B. fibrisolvens did not contain members of either of the xynA subfamilies or of the xynB family, at least one additional xylanase gene family remains to be identified in B. fibrisolvens.  相似文献

Argonaute proteins (AGOs) are mediators of gene silencing via recruitment of small regulatory RNAs to induce translational regression or degradation of targeted molecules. Platyhelminths have been reported to express microRNAs but the diversity of AGOs in the phylum has not been explored. Phylogenetic relationships of members of this protein family were studied using data from six platyhelminth genomes. Phylogenetic analysis showed that all cestode and trematode AGOs, along with some triclad planarian AGOs, were grouped into the Ago subfamily and its novel sister clade, here referred to as Cluster 1. These were very distant from Piwi and Class 3 subfamilies. By contrast, a number of planarian Piwi-like AGOs formed a novel sister clade to the Piwi subfamily. Extensive sequence searching revealed the presence of an additional locus for AGO2 in the cestode Echinococcus granulosus and exon expansion in this species and E. multilocularis. The current study suggests the absence of the Piwi subfamily and Class 3 AGOs in cestodes and trematodes and the Piwi-like AGO expansion in a free-living triclad planarian and the occurrence of exon expansion prior to or during the evolution of the most-recent common ancestor of the Echinococcus species studied.  相似文献

Euryte koreana sp. nov. is described from the shallow littoral on the East Coast of South Korea, and represents the first record of the subfamily Euryteinae Monchenko, 1974 in the Pacific Ocean north of the tropics. It belongs to a group of species that have the caudal rami length/width ratio of around four, but differs from all congeners by a number of morphological features. Detailed drawings and extensive scanning electron micrographs of many characters of ornamentation provided for this species should serve as a benchmark for distinguishing closely related species in this subfamily with conservative macro-morphology. A morphologically distinct population from anchialine caves in Mallorca, identified previously as Euryte longicauda Philippi, 1843, is described as another new species: Euryte jaumei sp. nov. To test the phylogenetic relationships of its members and previous hypotheses about generic placement of two species associated with scleractinian corals, several cladistic analyses are performed on all 16 currently recognized species of Euryteinae and two outgroups, Neocyclops australiensis Karanovic, 2008 and Troglocyclops janstocki Rocha and Iliffe, 1994, using 25 morphological characters. All resulting trees suggest a close relationship between the two commensal species and them as a sister clade to all other Euryteinae. They are transferred into a newly erected genus Coraleuryte gen. nov., as C. bellatula (Humes, 1991) comb. nov. and C. verecunda (Humes, 1992) comb. nov., and a revised diagnoses is provided for the genus Euryte Philippi, 1843. Obtained cladograms also show that Ancheuryte Herbst, 1989 is nested deeply within the Euryte clade, so the genus is synonymized and its only species is transferred, as E. notabilis (Herbst, 1989) comb. nov. A key to species of Euryteinae is also provided.  相似文献

Herbarium leaf fragments of the two known species of Orphanodendron (Leguminosae), O. bernalii and O. grandiflorum, were found to contain the quinolizidine alkaloids camoensine (1), camoensidine (2) and guianodendrine (3), supporting the recent phylogenetic placement of the genus in the genistoid clade of subfamily Papilionoideae rather than its traditional placement in subfamily Caesalpinioideae.  相似文献

Nucleotide sequence analysis of cDNA clones coding for field bean legumin precursor polypeptides revealed two different types, called A and B. Although homologous, both types differ in several sequence characteristics. Comparison with similar data from soybean and recent findings from pea leads to the following conclusions: (i) the two types of legumin genes described represent two subfamilies, A and B, which are probably of widespread occurrence; (ii) legumin genes or subunits can best be placed in either subfamily A or B by sequence homology, in addition B-type subunits contain generally fewer (or none at all in V. faba) Met residues as compared to A-type subunits; (iii) members of one subfamily from different species are more homologous than members of either subfamily within a species, therefore the two subfamilies must have arisen long before speciation of the genera Glycine, Pisum and Vicia; (iv) during speciation members of the B-subfamily diverged significantly more than members of the A-subfamily.  相似文献

 T-cell receptors (Tcrs) of higher organisms play a key role in the specific recognition of self and non-self molecules in the immune system. The large number of Tcr variable (V) genes have been organized into V gene subfamilies according to their sequence similarity at the nucleotide and amino acid level. We cloned and characterized four new members of the Tcra-V22 gene subfamily at the genomic level using a simple and sensitive technique that can rapidly clone members of any multi-member gene family. Sequence analysis reveals that the four Tcra-V22 gene subfamily members have more than 98% sequence similarity in their coding regions, at the nucleotide and amino acid levels. However, the intron between the leader and the coding region varies up to 7% between members of the Tcra-V22 gene subfamily. Comparison of the multi-member Tcra-V22 gene subfamily with other multi-member Tcra-V gene subfamilies (V2, V8, and V11), shows that Tcra-V22 is unique in that it has multiple members with nearly identical amino acid sequence and which are not inherently pseudogenes. Sequence similarity analysis of the Tcra-V22 subfamily with the prototypes of all other Tcra-V subfamilies revealed that the Tcra-V22 subfamily has the closest sequence similarity to that of Tcra-V18 (77% at the nucleotide level and 71% at the amino acid level). The reproductive morphology of Sargentodoxa cuneata (Oliv) Rehd. et Wils. is investigated through field, herbarium, and laboratory observations. Sargentodoxa may be either dioecious or monoecious. The functionally unisexual flowers are morphologically bisexual, at least developmentally. The anther is tetrasporangiate, and its wall, of which the development follows the basic type, is composed of an epidermis, endothecium, two middle layers, and a tapetum. The tapetum is of the glandular type. Microspore cytokinesis is simultaneous, and the microspore tetrads are tetrahedral. Pollen grains are two-celled when shed. The mature ovule is crassinucellate and bitegmic, and the micropyle is formed only by the inner integument. Megasporocytes undergo meiosis resulting in the formation of four megaspores in a linear tetrad. The functional megaspore develops into an eight-nucleate embryo sac after three rounds of mitosis. The mature embryo sac consists of an egg apparatus (an egg and two synergids), a central cell, and three antipodal cells. The pattern of the embryo sac development follows a monosporic Polygonum type. Comparisons with allied groups show that Sargentodoxa shares more synapomorphies with the Lardizabalaceae than other Ranunculales. Characteristics of its reproductive morphology are consistent with the placement of Sargentodoxa as the sister group of the remaining Lardizabalaceae. It does not possess a sufficient number of apomorphic characters to justify its separation into a separate family or subfamily. It is best retained as a member of the Lardizabalaceae.  相似文献

The study of the thermodynamic redox behavior of the hemes from two members of the A family of heme-copper oxygen reductases, Paracoccus denitrificans aa3 (A1 subfamily) and Rhodothermus marinus caa3 (A2 subfamily) enzymes, is presented. At different pH values, midpoint reduction potentials and interaction potentials were obtained in the framework of a pairwise model for two interacting redox centers. In both enzymes, the hemes have different reduction potentials. For the A1-type enzyme, it was shown that heme a has a pH-dependent midpoint reduction potential, whereas that of heme a3 is pH independent. For the A2-type enzyme the opposite was observed. The midpoint reduction potential of heme c from subunit II of the caa3 enzyme was determined by fitting the data with a single-electron Nernst curve, and it was shown to be pH dependent. The results presented here for these A-type enzymes are compared with those previously obtained for representative members of the B and C families.  相似文献

PICKLE plays a critical role in repression of genes that regulate development identity in Arabidopsis thaliana. PICKLE codes for a putative ATP-dependent chromatin remodeler that exhibits sequence similarity to members of subfamily II of animal CHD remodelers, which includes remodelers such as CHD3/Mi-2 that also restrict expression of developmental regulators. Whereas animal CHD3 remodelers are a component of the Mi-2/NuRD complex that promotes histone deacetylation, PICKLE promotes trimethylation of histone H3 lysine 27 suggesting that it acts via a distinct epigenetic pathway. Here, we examine whether PICKLE is also a member of a multisubunit complex and characterize the biochemical properties of recombinant PICKLE protein. Phylogenetic analysis indicates that PICKLE-related proteins in plants share a common ancestor with members of subfamily II of animal CHD remodelers. Biochemical characterization of PICKLE in planta, however, reveals that PICKLE primarily exists as a monomer. Recombinant PICKLE protein is an ATPase that is stimulated by ssDNA and mononucleosomes and binds to both naked DNA and mononucleosomes. Furthermore, recombinant PICKLE exhibits ATP-dependent chromatin remodeling activity. These studies demonstrate that subfamily II CHD proteins in plants, such as PICKLE, retain ATP-dependent chromatin remodeling activity but act through a mechanism that does not involve the ubiquitous Mi-2/NuRD complex.  相似文献

Background

The family Reduviidae (Hemiptera: Heteroptera), or assassin bugs, is among the most diverse families of the true bugs, with more than 6,000 species. The subfamily Triatominae (kissing bugs) is noteworthy not simply because it is the only subfamily of the Reduviidae whose members feed on vertebrate blood but particularly because all 147 known members of the subfamily are potential Chagas disease vectors. Due to the epidemiological relevance of these species and the lack of an efficient treatment and vaccine for Chagas disease, it is more common to find evolutionary studies focusing on the most relevant vectors than it is to find studies aiming to understand the evolution of the group as a whole. We present the first comprehensive phylogenetic study aiming to understand the events that led to the diversification of the Triatominae.

Methodology/Principal Findings

We gathered the most diverse samples of Reduviidae and Triatominae (a total of 229 Reduviidae samples, including 70 Triatominae species) and reconstructed a robust dated phylogeny with several fossil (Reduviidae and Triatominae) calibrations. Based on this information, the possible role of geological events in several of the major cladogenetic events within Triatominae was tested for the first time. We were able to not only correlate the geological changes in the Neotropics with Triatominae evolution but also add to an old discussion: Triatominae monophyly vs. paraphyly.

Conclusions/Significance

We found that most of the diversification events observed within the Rhodniini and Triatomini tribes are closely linked to the climatic and geological changes caused by the Andean uplift in South America and that variations in sea levels in North America also played a role in the diversification of the species of Triatoma in that region.  相似文献

The first complete genome sequence of a phage infecting Weissella cibaria (Weissella kimchii) is presented. The bacteriophage ϕYS61 was isolated from kimchi, a Korean fermented vegetable dish. Bacteriophages are recognized as a serious problem in industrial fermentations; however, ϕYS61 differed from many virulent phages associated with food fermentations since it was difficult to propagate and was very susceptible to resistance development. Sequence analysis revealed that ϕYS61 resembles Podoviridae of the subfamily Picovirinae. Within the subfamily Picovirinae, the ϕ29-like phages have been extensively studied, and their terminal protein-primed DNA replication is well characterized. Our data strongly suggest that ϕYS61 also replicates by a protein-primed mechanism. Weissella phage ϕYS61 is, however, markedly different from members of the Picovirinae with respect to genome size and morphology. Picovirinae are characterized by small (approximately 20-kb) genomes which contrasts with the 33,594-bp genome of ϕYS61. Based on electron microscopy analysis, ϕYS61 was classified as a member of the Podoviridae of morphotype C2, similar to the ϕ29-like phages, but its capsid dimensions are significantly larger than those reported for these phages. The novelty of ϕYS61 was also emphasized by the low number of open reading frames (ORFs) showing significant similarity to database sequences. We propose that the bacteriophage ϕYS61 should represent a new subfamily within the family Podoviridae.  相似文献
