The two membrane-bound respiratory nitrate reductases of Escherichia coli are encoded by distinct operons at two different loci, chlC and chlZ, on the chromosome. The chlZ locus includes a narK homologue, narU, encoding a nitrite extrusion protein, and narZYWV encoding nitrate reductase Z. No apparent homologue to the narXL operon has been found. Homology between narU and narK on the one hand and narZYWV and narGHJI on the other hand is limited to the coding regions.  相似文献

The number of rRNA cistrons is measured by filter saturation hybridization in different stocks of D. hydei, where the wild-type X chromosome has one nucleolus organizer (NO) and the wild-type Y has two separated NO's. (see PDF) females having no X chromosomal NO show an rDNA content exceeding that of a Y chromosome. An even greater increase in the rRNA cistron number is measured in two translocation stocks where the (see PDF) is combined with one half of a Y and, therefore, each stock contains only one of the two Y chromosomal NO's. But when the same Y fragments are brought together with a wild-type X chromosome they lose about one-half of their rRNA cistrons within one generation. Males with two complementary Y fragments but having no X chromosomal NO show a considerably higher rDNA content than the (see PDF) females, although both are equal in respect of their NO number. Consideration is given to related phenomena in Drosophila melanogaster.  相似文献

The type VI secretion system (T6SS) is a versatile secretion machine dedicated to various functions in Gram-negative bacteria, including virulence toward eukaryotic cells and antibacterial activity. Activity of T6SS might be followed in vitro by the release of two proteins, Hcp and VgrG, in the culture supernatant. Citrobacter rodentium, a rodent pathogen, harbors two T6SS gene clusters, cts1 and cts2. Reporter fusion and Hcp release assays suggested that the CTS1 T6SS was not produced or not active. The cts1 locus is composed of two divergent operons. We therefore developed a new vector allowing us to swap the two divergent endogenous promoters by Ptac and PBAD using the λ red recombination technology. Artificial induction of both promoters demonstrated that the CTS1 T6SS is functional as shown by the Hcp release assay and confers on C. rodentium a growth advantage in antibacterial competition experiments with Escherichia coli.  相似文献

The musculature of male genitalia was studied hitherto only in two species of Tephritidae, one species of Platystomatidae, one species of Pallopteridae, and three species of Ulidiidae of the superfamily Tephritoidea. The split of the hypandrium from one structure into three (the hypandrium and two lateral sclerites) is traced. The hypandrial origin of the lateral sclerites of the hypandrial complex is shown based on the localization of muscle attachment sites. The subepandrial origin of the inner lobes of the surstyli is also confirmed.  相似文献

Precious corals are soft corals belonging to the family Coralliidae (Anthozoa: Octocorallia: Alcyonacea) and class Anthozoa, whose skeletal axes are used for jewelry.The family Coralliidae includes ca. 40 species and was originally thought to comprise of the single genus Corallium. In 2003, Corallium was split into two genera, Corallium and Paracorallium, and seven species were moved to this newly identified genus on the bases of morphological features. Previously, we determined the complete mitochondrial genome sequence of two precious corals Paracorallium japonicum and Corallium konojoi, in order to clarify their systematic positions. The two genomes showed high nucleotide sequence identity, but their gene order arrangements were not identical. Here, we determined three complete mitochondrial genome sequences from the one specimen of Mediterranean Corallium rubrum and two specimens of Corallium elatius coming from Kagoshima (South Japan). The circular mitochondrial genomes of C. rubrum and C. elatius are 18,915 bp and 18,969–18,970 bp in length, respectively, and encode 14 typical octocorallian protein-coding genes (nad16, nad4L, cox13, cob, atp6, atp8, and mtMutS, which is an octocoral-specific mismatch repair gene homologue), two ribosomal RNA genes (rns and rnl), and one transfer RNA (trnM). The overall nucleotide differences between C. konojoi and each C. elatius haplotype (T2007 and I2011) are only 10 and 11 nucleotides, respectively; this degree of similarity indicates that C. elatius and C. konojoi are very closely related species. Notably, the C. rubrum mitochondrial genome shows more nucleotide sequence identity to P. japonicum (99.5%) than to its congeneric species C. konojoi (95.3%) and C. elatius (95.3%). Moreover, the gene order arrangement of C. rubrum was the same as that of P. japonicum, while that of C. elatius was the same as C. konojoi. Phylogenetic analysis based on three mitochondrial genes from 24 scleraxonian species shows that the family Coralliidae is separated into two distinct groups, recovering Corallium as a paraphyletic genus. Our results indicate that the currently accepted generic classification of Coralliidae should be reconsidered.  相似文献

FOF1 ATP synthases are rotary nanomotors that couple proton translocation across biological membranes to the synthesis/hydrolysis of ATP. During catalysis, the peripheral stalk, composed of two b subunits and subunit δ in Escherichia coli, counteracts the torque generated by the rotation of the central stalk. Here we characterize individual interactions of the b subunits within the stator by use of monoclonal antibodies and nearest neighbor analyses via intersubunit disulfide bond formation. Antibody binding studies revealed that the C-terminal region of one of the two b subunits is principally involved in the binding of subunit δ, whereas the other one is accessible to antibody binding without impact on the function of FOF1. Individually substituted cysteine pairs suitable for disulfide cross-linking between the b subunits and the other stator subunits (b-α, b-β, b-δ, and b-a) were screened and combined with each other to discriminate between the two b subunits (i.e. bI and bII). The results show the b dimer to be located at a non-catalytic α/β cleft, with bI close to subunit α, whereas bII is proximal to subunit β. Furthermore, bI can be linked to subunit δ as well as to subunit a. Among the subcomplexes formed were a-bI-α, bII-β, α-bI-bII-β, and a-bI-δ. Taken together, the data obtained define the different positions of the two b subunits at a non-catalytic interface and imply that each b subunit has a different role in generating stability within the stator. We suggest that bI is functionally related to the single b subunit present in mitochondrial ATP synthase.  相似文献

The taxa of the Interiores species group are treated as two species, one of which is divided into two subspecies.Vernonia missurica, a widespread taxon, has large flower heads (32-58 flowers per head), obtuse involucral bract tips, and a characteristic flavonoid Chromatographic profile.Vernonia baldwinii ssp.baldwinii, occurring throughout the Ozarks and Ouachitas, andV. baldwinii ssp.interior of the Central Plains possess smaller flower heads (17-34 flowers per head), acute or acuminate involucral bracts, and a different Chromatographic profile.Vernonia baldwinii ssp.baldwinii is distinguished from ssp.interior by the recurved involucral bract tips and by the possession of more flowers per head. Morphological data reveal that an intergradation zone exists between the population systems of the two subspecies along the edges of the Ozarks and Ouachitas. Although much gene exchange occurs between these two taxa, they maintain a level of genetic integrity that permits their ready recognition throughout wide ranges. All three taxa have a gametic chromosome number ofn = 17 and hybridize extensively with sympatric species.  相似文献

Examination of type material ofVerbesina standleyi andCalea pachyphylla shows that the two taxa are conspecific. The older name,C. pachyphylla, has long been associated with the quite remote,C. orizabaensis. The latter is indistinguishable fromC. rupestris andC. guatemalensis; accordingly, these are placed in synonymy.Calea orizabaensis consists of two infraspecific taxa, var.orizabaensis and var.websteri; the latter is described as new.  相似文献

Ilex, the largest genus of dioecious woody plants, is a good study system to assess the role of hybridization in speciation and evolution. Ilex dabieshanensis, a tree endemic to Dabieshan Mountains region, was initially described as a new species. Based on morphological intermediacy and sympatric distribution with its putative parental species, I. cornuta and I. latifolia, we proposed it as a natural hybrid between them. In this study, we sequenced one chloroplast intergenic spacer (trnH-psbA) and two nuclear genes (gapC and nepGS) in I. dabieshanensis and its putative parental species to test the hybrid origin hypothesis. Our results showed that there were one to two differentially fixed sequence differences between I. cornuta and I. latifolia at the two nuclear genes. Twelve of the 14 individuals of I. dabieshanensis exhibited additivity in chromatograms on these differentially fixed sites at both nuclear genes, and the remaining two exhibited additivity in chromatograms on the fixed site at only the nepGS gene. Except one haplotype of I. cornuta at the nepGS gene, all of the haplotypes of I. cornuta at the two nuclear genes were well separated from those of I. latifolia, and most haplotypes of I. dabieshanensis were shared with those of I. cornuta and I. latifolia. Phylogenetic analysis of these haplotypes was largely consistent with haplotype network analysis. I. cornuta and I. latifolia differed by two nucleotide substitutions in the chloroplast intergenic spacer, and 12 individuals of I. dabieshanensis had the same sequences as I. latifolia, while the remaining two were identical with I. cornuta. The molecular data provide convincing evidence for the hybrid origin of I. dabieshanensis and asymmetrical direction of hybridization. One haplotype of I. cornuta at the nepGS gene was nested with those of I. latifolia, indicating introgression to I. cornuta.  相似文献

The sesquiterpene lactones isolated from species in the genus Artemisia have been reviewed in an attempt to better understand the phylogeny and systematics of the four sections (subgenera), Abrotanum, Absinthium, Dracunculus and Seriphidium, proposed by Besser in 1829. The absence of hair on the receptacle is the only morphological characteristic separating species of Abrotanum from the species of Absinthium. There are no chemical characteristics segregating the species in these two subgenera since both produce eudesmanolides and guaianolides that are identical or biosynthetically similar. This suggests that the two subgenera could be combined into one (Artemisia) as proposed by Poljakov. The subgenus Seriphidium is composed of two geographical groups, one in the Old World and the other in the New World. The Old World species almost exclusively produce sesquiterpene lactones in the eudesmanolide class whereas the New World species (section Tridentatae) produce eudesmanolides and guaianolides, many of the latter being identical or structurally related to the sesquiterpene lactones in New World Abrotanum species. The chemical data in conjunction with geographic distributions suggest that the subgenus Seriphidium is polyphyletic and that the section Tridentatae originated from Abrotanum. Consequently, the Tridentate should be recognized as a subgenus separate and distinct from the Old World Seriphidium. There was insufficient information from the subgenus Dracunculus for interpretation.  相似文献

There is no general agreement on relationships within Xylophagoidea (Diptera, Brachycera). The musculature of the male genitalia of Xylophagus cinctus (De Geer) (Xylophagidae, the most primitive family of Brachycera) is described and compared with that of some other Xylophagoidea: Exeretonevridae (Exeretonevra angustifrons Hardy), Coenomyiidae (Anacanthaspis biafasciata Röder), and Rhagionidae (Rhagio montanus Becker, Chrysopilus dives Loew, and Ch. helvolus Meigen) discussed earlier (Ovtshinnikova, 1989, 1998; Palmer et al., 2000). In spite of the differences in the structure of the genital sclerites, Xylophagidae possess all the muscles found in Coenomyiidae and Rhagionidae. The musculature of the male genitalia of Xylophagus cinctus includes two muscle pairs of the aedeagus sheath (M1 and M2); three muscle pairs of the ejaculatory complex (M30, M31, and M32); one muscle pair of the gonocoxites (M33); two muscle pairs of the gonostyli (M27 and M28); one muscle pair of the proctiger (M21), one muscle pair of the cerci (M29); two pairs of the tergosternal muscles (M5 1 and M5 2); and two pairs of the pregenital muscles (M18 and M19). Muscles of the family Exeretonevridae are mostly the same, except for the muscles of the cerci M29, proctiger M29, and pregenital muscles M18 and M19, that are subdivided into two parts. This fact and also a different degree of the development of muscles M32 and M5 2 clearly distinguish Exeretonevridae from closely related families. The attachment places of the muscles of the aedeagus sheath M2 and of the gonostyli M28, as well as the split character of the tergosternal muscle pair M5 1 makes it possible to distinguish two sister groups, Xylophagidae plus Exeretonevridae, versus Coenomyiidae plus Rhagionidae. It should be noted that the muscles of the male genitalia of Xylophagidae, Exeretonevridae, Coenomyiidae, and Rhagionidae possess similar plesiomorphic characters, and these families should be united into the superfamily Xylophagoidea. This superfamily is the most primitive superfamily of Brachycera Orthorrhapha and possesses the most stable set and arrangement of male genital muscles within the entire suborder. An improved dendrogram of the phylogenetic relationships between the known groups of Xylophagoidea is proposed on the basis of the structure of male genital muscles.  相似文献

Detailed study of the ultrastructures on the cortical lists of retiolitids occurring after the lundgreni event has been undertaken. The extremely well preserved material from the Go?dap borehole of Poland (Baltica) enables classification of clathrial lists into two types—with single and double seams. Significantly different seam development is observed between two groups of retiolitids occurring before and after the lundgreni event. The study of seams and increments on the lists enables reconstruction of the membrane arrangement of the rhabdosomes. Reconstruction of the proximal ends, the arrangement of the ancora umbrella, and the layout of the proximal ventral and lateral orifices of five genera, Neogothograptus, Holoretiolites, Plectograptus, Spinograptus, and Quattuorgraptus n. gen., is presented. The new genus Quattuorgraptus is separated from Spinograptus because of the different development of the ancora umbrella and the width of the lateral walls of the rhabdosome.  相似文献

We have investigated the occurrence of dosage compensation in D. willistoni and D. pseudoobscura, two species whose X chromosome is metacentric with one arm homologous to the X and the other homologous to the left arm of chromosome 3 of D. melanogaster. Crude extracts were assayed for isocitrate dehydrogenase (XR), glucose-6-phosphate dehydrogenase (XL?), 6-phosphogluconate dehydrogenase (XL?), and α-glycerophosphate dehydrogenase (chromosome 2) in D. willistoni, and for esterase-5 (XR), glucose-6-phosphate dehydrogenase (XL?), 6-phosphogluconate dehydrogenase (XL?) and amylase (chromosome 3) in D. pseudoobscura. Our results indicate that a mechanism for dosage compensation is operative in both arms of the X chromosome of these two species.  相似文献

Three members of the genus Borrelia (B.burgdorferi, B.garinii, B.afzelii) cause tick-borne borreliosis. Depending on the Borrelia species involved, the borreliosis differs in its clinical symptoms. Comparative genomics opens up a way to elucidate the underlying differences in Borrelia species. We analysed a low redundancy whole-genome shotgun (WGS) assembly of a B.garinii strain isolated from a patient with neuroborreliosis in comparison to the B.burgdorferi genome. This analysis reveals that most of the chromosome is conserved (92.7% identity on DNA as well as on amino acid level) in the two species, and no chromosomal rearrangement or larger insertions/deletions could be observed. Furthermore, two collinear plasmids (lp54 and cp26) seem to belong to the basic genome inventory of Borrelia species. These three collinear parts of the Borrelia genome encode 861 genes, which are orthologous in the two species examined. The majority of the genetic information of the other plasmids of B.burgdorferii is also present in B.garinii although orthology is not easy to define due to a high redundancy of the plasmid fraction. Yet, we did not find counterparts of the B.burgdorferi plasmids lp36 and lp38 or their respective gene repertoire in the B.garinii genome. Thus, phenotypic differences between the two species could be attributable to the presence or absence of these two plasmids as well as to the potentially positively selected genes.  相似文献

The taxonomy of Chamaedrilus glandulosus (Michaelsen, 1888) s. l., most commonly known previously as Cognettia glandulosa, is revised. A recent molecular systematic study has shown that this taxon harbours two cryptic, but genetically well separated lineages, each warranting species status. In this study these two lineages are scrutinized morphologically, on the basis of Michaelsen’s type material as well as newly collected specimens from Central and Northern Europe. Chamaedrilus glandulosus s. s. is redescribed and Chamaedrilus varisetosus sp. n. is recognized as new to science. The two species are morphologically very similar, differing mainly in size, but seem to prefer different habitats, with Chamaedrilus glandulosus being a larger aquatic species, and Chamaedrilus varisetosus being smaller and mainly found in moist to wet soil.  相似文献

On Kilimanjaro, Tanzania two rather different forms of Chlorophytum comosum were observed. One form occurred on higher altitudes, the other on lower altitudes. Morphological, molecular and ecological studies conclude that the two forms both are entitled to species recognition. The high altitude form must still be referred to C. comosum s.l., while the low altitude form is a new species and it is described as C. rhizopendulum Bjorå & Hemp sp. nov. The two species inhabit mutually completely exclusive habitats on Kilimanjaro. Whereas C. comosum has a wide ecological potential, C. rhizopendulum has narrow ecological demands as a highly adapted specialist. The two species are easily distinguished morphologically. Molecular studies show that C. comosum is of polyphyletic origin, and the species complex needs revision.  相似文献
