Transpiration inhibition by stored xylem sap from well-watered maize plants   总被引:3,自引:0,他引:3  
There is increasing evidence that a chemical signal exists in xylem sap of plants subjected to water deficits which influences physiological responses in plant shoots. An important method of studying this signal is the transpiration response of excised leaves exposed to xylem sap collected from plants. However, Munns et al [Plant, Cell & Environment 16, 867–877] cautioned that transpiration inhibition is observed when xylem sap collected from wheat and barley is stored before determining physiological activity. The objective of the study reported here was to determine if transpiration inhibition develops in maize sap collected from well-watered plants when the sap is stored under various conditions. It was found that storage of maize sap collected from well-watered plants for only 1 d at -20°C resulted in the development of substantial transpiration inhibition in bioassay leaves. Storage of sap at 4°C resulted in the development of the effect after 2 weeks, while storage at ?86°C showed only small transpiration inhibition after 3 weeks. The major source of the transpiration inhibition was the development of a substance in the stored sap that resulted in physical blockage of the transpiration stream in bioassay leaves. However, a small signal component may also have developed in the stored sap. Because of the possibility of ionic activity under freezing conditions at ?20°C, calcium was studied for its potential involvement in the transpiration inhibition. However, the calcium concentrations found to inhibit transpiration were nearly an order of magnitude larger than the calcium concentrations observed in xylem sap.  相似文献

Rice flour was stored at 15 °C/9 months, at 35 °C/14 days, or dry-heated at 120 °C/20 min. The breadmaking properties baked with this rice flour/fresh gluten flour deteriorated. In addition, the rice flour was mixed with oil in water vigorously, and oil-binding ability was measured. Every rice flour subjected to storage or dry-heated at 120 °C showed higher hydrophobicity, owing to changes in proteins. Then, proteins in the stored rice flour were excluded with NaOH solution, and bread baked with the deproteinized rice flour showed the same breadmaking properties as unstored rice flour/fresh gluten flour. The viscoelasticity of wheat glutenin fraction decreased after the addition of dry-heated rice flour in a mixograph profile. DDD staining increased Lab in color meter, which suggested an increase in SH groups in rice protein. The increase in SH groups caused a reduction in wheat gluten protein resulting in a deterioration of rice bread quality.  相似文献

Rooster spermatozoa were stored at 25, 5, or ?196 °C in either TC199, a pyruvate-lactate mouse ova culture medium, or as undiluted semen. There was a linear decrease in percent of motile sperm during storage at 25 or 5 °C in all cases, and a curvilinear decrease with increasing storage times at ?196 °C. Percent of motile sperm present after increasing storage time suggested pyruvate-lactate is a better extender than TC199 at the three storage temperatures studied. Pullets inseminated with 1 × 108 motile sperm using fresh sperm diluted in TC199 or pyruvate-lactate, or stored 24 hr at 5 or ?196 °C produced 68.7, 74.1, 20.6, and 10.8% fertile eggs, respectively. The differences in fertility between controls or between samples stored at 5 and ?196 °C were not significant. However, fertility from sperm stored at 5 and ?196 °C was significantly lower (p < .05) than both control groups. Thus, it can be concluded that TC199 or pyruvate-lactate may be used to dilute fresh rooster semen collections prior to insemination. In contrast, fertility of rooster sperm is not satisfactorily maintained after 5 or ?196 °C storage for 24 hr in a pyruvate-lactate extender.  相似文献

Aims: Producing granular cultures of obligate aphid pathogen Pandora nouryi for improved sporulation and storage. Methods and Results: Small millet–gel granules were made of the mixtures of 80–95% millet powder with 5–20% polymer gel (polyacrylamide, polyacrylate or acrylate‐acrylamide copolymer) and inoculated with mycelia at 30 mg biomass g?1 dry granules plus 87·5% water, followed by static incubation at 20°C for 4–12 days. The fungus grew well on 12 preparations but best on that including 10% copolymer. An 8‐day culture of this preparation discharged maximally 58·5 × 104 conidia mg?1 granule at 100% RH and was capable of ejecting conidia at the nonsaturated regimes of 86–97% RH. During storage at 6°C, granular cultures with >85% water content had twofold longevity (120 days) and half‐decline period (34–36 days) of those stored at room temperature. The steadily high water content preserved the cultures better than that decreasing at 6°C. However, conidia from 70‐day‐stored granules were less infective to Myzus persicae nymphs than those from fresh ones based on their LC50s. Conclusions: The millet–gel granules had higher sporulation capacity than reported Pandora cultures and a capability of spore discharge at nonsaturated humidity. In roach Rutilus rutilus growth ceases below a temperature threshold of 12° C. This cessation of growth is accompanied by a reduction in feeding. Do roach decrease feeding in the cold because of reduced energy demand, caused by the decelerating effect of low temperature on metabolism and growth, or is feeding directly limited by low temperatures, leading to reduced growth rates? It was found that at low temperatures the intake and digestion of food may be limited by reduced activities of digestive enzymes. Trypsin, amylase and γ‐glutamyl transferase showed a negative compensation with respect to temperature, resulting in very low activities at acclimation temperatures of ≤12° C. Trypsin activity, falling from 400·5 ± 131·2 U g?1 fresh mass of the gut at 27° C to 12·5 U g?1 fresh mass at 4° C, displayed the strongest linear correlation with growth rates, suggesting that trypsin activities may set a limit to growth in the low temperature range. If protein digestion is limiting at low temperatures, this should be reflected in reduced concentrations of amino acid in the white muscle. The size of the total amino acid pool was not affected by temperature acclimation and ranged between 19·2 ± 6·2 and 25·2 ± 3·6 µmol g?1 fresh mass of the white muscle. A decrease, however, was found of several amino acids, mainly of threonine and glutamine, in the low temperature range. Low concentrations of the essential amino acid threonine (0·14 ± 0·03 µmol g?1 fresh mass at 12° C and 0·12 ± 0·05 µmol g?1 fresh mass at 4° C) were probably due to nutritional or digestional limitations and may therefore have resulted from reduced trypsin activity in the cold. The non‐essential amino acid glutamine, however, can be endogenously synthesized and its low level observed at 4° C (0·16 ± 0·09 µmol g?1 fresh mass) was not necessarily a result of low trypsin activities. It is more likely that low temperatures impair glutamine synthesis. The possibility that glutamine concentrations may be down regulated under conditions when anabolic processes are not advantageous is discussed.  相似文献

This study compared different temperatures and dormancy‐reversion procedures for preservation of Phakopsora pachyrhizi uredospores. The storage temperatures tested were room temperature, 5°C, ?20°C and ?80°C. Dehydrated and non‐dehydrated uredospores were used, and evaluations for germination (%) and infectivity (no. of lesions/cm2) were made with fresh harvested spores and after 15, 29, 76, 154 and 231 days of storage. The dormancy‐reversion procedures evaluated were thermal shock (40°C/5 min) followed or not by hydration (moist chamber/24 h). Uredospores stored at room temperature were viable only up to a month of storage, regardless of their hydration condition. Survival of uredospores increased with storage at lower temperatures. Dehydration of uredospores prior to storage increased their viability, mainly for uredospores stored at 5°C, ?20°C and ?80°C. At 5°C and ?20°C, dehydrated uredospores showed increases in viability of at least 47 and 127 days, respectively, compared to non‐dehydrated spores. Uredospore germination and infectivity after storage for 231 days (7.7 months), could only be observed at ?80°C, for both hydration conditions. At this storage temperature, dehydrated and non‐dehydrated uredospores exhibited 56 and 28% of germination at the end of the experiment, respectively. Storage at ?80°C also maintained uredospore infectivity, based upon levels of infection frequency, for both hydration conditions. Among the dormancy‐reversion treatments applied to spores stored at ?80°C, those involving hydration allowed recoveries of 85 to 92% of the initial germination.  相似文献

The activity of the soluble enzyme CPK and the membrane bound enzyme Na+-K+ ATPase as a function of storage temperature, time of storage and cryoprotectant type and concentration in canine myocardial tissue was invesigated. Activity of CPK is well preserved at ?196 °C and ?79 °C and falls off during one month storage at ?40 °, ?20 °, and 0 °C. Na+-K+ ATPase demonstrates a greater liability. After an initial cryoprotectant “activation,” activity drops. In all cases, however, addition of the cryoprotectant preserved activity better than in samples stored only in buffer.  相似文献

Potato mop-top virus (PMTV) was transmitted by inoculation of sap to twenty-six species in the Solanaceae or Chenopodiaceae and to Tetragonia expansa; species in eleven other plant families were not infected. The virus was cultured in inoculated leaves of Nicotiana tabacum cv. Xanthi-nc or in N. debneyi. Diagnostic local lesions were produced in Chenopodium amaranticolor. In winter, ten solanaceous species were slowly invaded systemically but the first leaves infected were those immediately above inoculated leaves. When transmitted to Arran Pilot potato by the vector Spongospora subterranea, PMTV induced all the main types of shoot and tuber symptoms found in naturally infected plants. Isolates of PMTV from different sources differed considerably in virulence. PMTV-containing tobacco sap lost infectivity when heated for 10 min at 80 °C, diluted to 10-4, or stored at 20 °C for 14 weeks. Infectivity was partially stabilized by 0·02% sodium azide. When sap was centrifuged for 10 min at 8000 g, infectivity was mainly in the sediment. Infective sap contained straight rod-shaped particles about 20 nm wide, with lengths up to 900 nm and crossbands at intervals of 2·5 nm. Many of the particles were aggregated side-to-side, and the ends of most seemed damaged. The slight infectivity of phenol-treated leaf extracts was abolished by pancreatic ribonuclease. The present cryptogram of PMTV is R/*:*/*:E/E:S/Fu.  相似文献

The effects of human albumin preparations on oxidative energy metabolism and lipid svnthesis were investigated in rat liver slices incubated with sodium [1-14C]acetate as precursor. Labeled CO2 production and incorporation of precursor into the major lipid classes was increased 2 to 3-fold by fresh preparations of albumin (fraction V), and by defatted fraction V, whereas highly purified cystalline albumin was less active. Albumin preparations from various commercial suppliers varied widely in activity. Activity of fraction V was preserved during storage at ?20°C, and gradually lost at +3°C in the course of 1 year. In contrast, defatted fractions rapidly lost activity in storage at both temperatures. After 1 year in storage at +3°C, albumin preparations became inhibitory to CO2 production and lipid synthesis. The results suggest that commercial albumin used in metabolic studies, and in clinical situations may have unpredictable or undesirable effects related to state of purity and storage conditions of the protein.  相似文献

Aims: To determine the efficacy of X‐ray processes in inactivating L. monocytogenes levels in smoked catfish during storage at 5°C and to determine the effects of X‐ray doses on controlling the growth of spoilage bacteria on smoked catfish during storage at 5°C for up to 5 weeks. Methods and Results: Smoked catfish fillets inoculated with L. monocytogenes were treated with 0·0–2·0 kGy X‐ray and stored at 5°C for 5 weeks. The negative controls (uninoculated/untreated) and uninoculated samples treated with the lowest (0·1 kGy) and highest (2·0 kGy) doses were stored at 5°C and tested for psychrotrophs count during the 5 weeks of storage. The initial L. monocytogenes population on smoked catfish was significantly (P < 0·05) reduced to undetectable level by a treatment of 1·0 kGy or higher. The initial psychrotrophs count on smoked catfish was significantly reduced from 4·7 CFU g?1 to below the detectable level by a treatment with 2·0 kGy. Conclusions: Smoked catfish treated with 2·0 kGy X‐ray had no detectable L. monocytogenes throughout 35 days of storage at 5°C. A treatment with 2·0 kGy X‐ray also kept the levels of psychrotrophs in the smoked catfish within the acceptable level until 35 days. Significance and Impact of the Study: The results of this investigation indicate that X‐ray at 2·0 kGy can eliminate L. monocytogenes and extend the shelf life of smoked catfish stored at refrigeration temperature.  相似文献

This article reports the effect of cold storage at 4 °C from December to April on chestnut cuttings. Rooting was found to increase after four months preceded by treatment with 4000 mg l-1 IBA. ThePhaseolus test of the biological activity of extracts of the cuttings showed the inhibition exerted by the acid fraction of the fresh (December) cuttings to have been replaced by a rootpromoting effect in cuttings stored for 4 months at low temperature. Pepino mosaic virus, a new potexvirus from pepino (Solanum muricatum)   总被引:1,自引:0,他引:1  
Pepino mosaic virus (PepMV), a previously undescribed virus, was found in fields of pepino (Solanum muricatum) in the Canete valley in coastal Peru. PepMV was transmitted by inoculation of sap to 32 species from three families out of 47 species from nine families tested. It caused a yellow mosaic in young leaves of pepino and either a mild mosaic or symptomless infection in 12 wild potato species, five potato cultivars and potato clone USDA 41956 but S. stoloniferum and potato cultivars Merpata and Revolucion reacted with severe systemic necrotic symptoms. The virus was transmitted by plant contact but not by Myzus persicae. It was best propagated and assayed in Nicotiana glutinosa. Sap from infected N. glutinosa was infective after dilution to 10-1 but not 10-6, after 10 min at 65°C but not 70°C and after 3 months at 20°C. PepMV had filamentous particles with a normal length of 508 nm; the ends of some seemed damaged. Ultra-thin sections of infected leaves of N. glutinosa revealed many inclusions containing arrays of virus-like particles some of which were banded or whorled; small aggregates of virus-like particles were also common. The virus was purified by extracting sap from infected leaves in a solution containing 0·065 M disodium tetraborate, 0·435 M boric acid, 0·2% ascorbic acid and 0·2% sodium sulphite at pH 7·8, adding silver nitrate solution to the extract, and precipitating the virus with polyethylene glycol followed by two cycles of differential centrifugation. Particles of PepMV normally yielded two proteins with molecular weights of 26 600 and 23 200, but virus obtained from infective sap aged overnight yielded only the smaller protein suggesting that it was a product of degradation of the larger one. The virus is serologically related to two potexviruses, narcissus mosaic and cactus X and its properties are typical of the potexvirus group.  相似文献

Abstract: The effects of storage at 4°C on the antagonist and agonist binding properties of the muscarinic acetyl-choline receptor from fresh surgical and frozen autopsy samples from human cerebral cortex were studied. The number of 1-[3H]3-quinuclidinyl benzilate binding sites and their affinities were stable up to 51 h, both when stored as pieces of intact nonfrozen tissue and as a homogenate. The agonist binding properties as measured by the ability of the muscarinic agonist carbachol to compete with l-[3H]3-quinuclidinyl benzilate were also stable up to 51 h when the tissue was stored in the form of pieces. The affinity for carbachol decreased when the tissue was stored as a homogenate. The frozen autopsy samples showed no significant differences in binding properties in comparison with fresh neurosurgical tissue.  相似文献

The effects of harvesting frequency on productivity, nutrient storage and uptake, and detritus accumulation by water hyacinth (Eichhornia crassipes /Mart/ Solms) cultured outdoors in nutrient-enriched waters were evaluated for a period of 13 months. Significant differences in hyacinth standing crop and productivity were measured with harvesting regimes of 1, 3 (harvest at maximum density) and 21 harvests over a 13-month period. The average plant standing crop decreased from 65 to 20 kg (fresh wt) m−2 for systems with 1 and 21 harvests, respectively. Total harvested plant biomass was 67 kg (fresh wt) m−2, 110 kg (fresh wt) m−2 and 162 kg (fresh wt) m−2 for 1, 3 and 21 harvests, respectively. The mean net productivity increased from 7·7 to 16·5 and 24·5 g (dry wt) m−2 day−1 for 1, 3 and 21 harvests, respectively. Nutrient storage in water hyacinth biomass (live, dead and detrital) at the end of the study decreased from 93 to 46 and 30 g N m−2, and from 20 to 12 and 5 g P m−2, for 1, 3 and 21 harvests, respectively. For the system with one harvest, 46% of the stored N and 25% of the stored P were recovered in dedrital tissue at the bottom of the tank. For the systtem with 21 harvests, only 11% of the stored N and 15% of the stored P were recovered in detrital tissue at the bottom of the tank. Ammonium-N and soluble reactive P concentrations in the water column were significantly higher for the treatment with one harvest compared to the treatments with 3 and 21 harvests.  相似文献

The conditions of promotion of autolysis of three strains of Lactobacilli were investigated. The autolysis of L. acidophilus, L. helveticus and L. casei at exponential phase was remarkably enhanced by freezing storage at ?20°C overnight.

The turbidity decrease of L. acidophilus’ cell suspension corresponds to the increase of cell free nitrogen compounds, glucosamine and DNA component. All these compounds were more rapidly released from the cells stored at ?20°C than those stored at 3°C. The cells which were harvested at the exponential phase had higher autolytic activity than those at stationary phase. The storage of the cells at ?20°C for 2 days or more could effectively promote the autolysis.

The activity was increased by Ca2+ or Mg2+. Optimum pH of the autolytic enzyme of L. acidophilus was 6~7.  相似文献

Seeds of lettuce (Lactuca sativa L.) and sunflower (Helianthusannuus L.) were stored hermetically at 35 °C with 11 differentmoisture contents between 1·3 and 6·9%, and between1·3 and 7·1% of fresh mass, respectively. Germinationand vigour (mean germination time, root length, seedling dryweight) were determined after storage for 0, 8, or 16 weeks(sunflower) or 0, 8, 16, or 48 weeks (lettuce) in these environmentsfollowed by various humidification treatments (to avoid imbibitioninjury). The range of seed storage moisture contents over whichdeterioration was minimized depended upon the criterion of deteriorationused, and varied somewhat between species. Comparison of theseranges for seeds stored for the longest durations showed thatfor some criteria seed performance was poorer (P < 0·05)at both the lowest and highest moisture contents investigatedthan at certain of the intermediate storage moisture contents(e.g, most rapid germination occurred in sunflower followingstorage at 2·2-4·7% moisture content), whereasfor other criteria all the drier storage moisture contents weresuperior to the more moist (e,g. greatest seedling growth occurredin sunflower following storage at 1·3-5·1% moisturecontent). But none of these results suggested that lettuce andsunflower seeds stored hermetically at 2·5-3·0%or 2·2-2·5% moisture content, respectively, wereless vigorous than at any other moisture content tested. Inboth species, these storage moisture contents are in equilibriumwith about 8-10% relative humidity (r.h.) at 20 °C, whichis similar to and indeed marginally less than the 10-13% r.h.recommended following earlier studies on the longevity of seedsin hermetic storage at much warmer temperatures. Thus, theseresults show no evidence that the optimum seed moisture contentfor storage increases with decrease in temperature, at leastover the range 35-65 °C, as has been suggested elsewhere.We conclude that the international recommendation for the long-termseed storage for genetic conservation at 5 ± 1% moisturecontent should not be revised upwardly, and that in situationswhere refrigeration cannot be provided storage at even lowermoisture contents is worthy of further investigation for thoseseeds in which desiccation at 20 °C to equilibrium at 10%r.h. results in moisture contents well below 5%.Copyright 1995,1999 Academic Press Helianthus annuus L., sunflower, Lactuca sativa L., lettuce, desiccation, seed storage, seed vigour  相似文献

Blastospores of three strains of Metarhizium anisopliae were stored in 18 liquids at 4°C, 20°C and 35°C for 18 weeks, 12 weeks or 9 days respectively. Viability was quantified by determination of their germination. In bioassays the virulence of stored blastospores was studied using adults and third instars of Locusta migratoria migratorioides (R. & F.) and compared to those of freshly produced blastospores and conidia. Generally, there was great variability in the viability of blastospores, depending on the fungal strain and the liquids used. Blastospores survived best at 4°C in 10% hydroxyethyl starch; for example, germination of M. anisopliae strain 97 still amounted to more than 80% after storage for 18 weeks. Other suitable liquids were deionized water, 25% Ringer's solution and 1% sodium alginate. The viability of blastospores stored at 20°C was considerably shorter than at 4°C. During storage for 12 weeks at 20°C the best protective liquids for M. anisopliae strain 97 were 25% Ringer's solution (43% germination), deionized water (23%) and 10% hydroxyethyl starch (23%). At 35°C, 45% of M. anisopliae strain 97 blastospores still germinated after storage for 7 days in 25% glycerol. The bioassays revealed that the virulence of blastospores after storage was comparable to that of fresh ones and even better than that of fresh conidia. In general, the LT50 was about 4–6 days at an alternating day/night temperature of 28/20°C.  相似文献

The purpose of this work was to evaluate the effects of ethylene action blockade and cold storage on the ripening of ‘Golden’ papaya fruit. Papayas harvested at maturity stage 1 (up to 15% yellow skin) were evaluated. Half of the fruits, whether treated or not treated with 100 nL L−1 of 1-methylcyclopropene (1-MCP), were stored at 23°C, while the other half were stored at 11°C for 20 days prior to being stored at 23°C. Non-refrigerated fruits receiving 1-MCP application presented a reduction in respiratory activity, ethylene production, skin color development and pectinmethylesterase activity. Even with a gradual increase in ethylene production at 23°C, fruits treated with 1-MCP maintained a high firmness, but presented a loss of green skin color. Cold storage caused a decrease in ethylene production when fruits were transferred to 23°C. The results suggest that pulp softening is more dependent on ethylene than skin color development, and that some processes responsible for loss of firmness do not depend on ethylene.  相似文献

【目的】评估土壤长期保存(4个月)对土壤微生物群落代谢活性的影响。【方法】采用Biolog? EcoPlateTM生态板研究4 °C风干保存和?20 °C低温冻存的农田土壤和森林土壤中微生物群落的碳源利用模式。【结果】与新鲜土壤样品相比,长期保存的土壤样品的微生物群落对碳源的利用能力大大降低,其多样性、均匀度和Simpson指数均降低;风干保存和低温冻存两者对土壤微生物的碳源利用的影响没有显著差异;除风干保存的土壤样品中利用多聚物类的微生物类群的代谢活性外,两种保存方法显著降低微生物群落的代谢活性,降低幅度为54.5%–99.8%。【结论】长期保存土壤可能会导致对微生物群落信息的低估,土壤微生物代谢活性研究的最佳样品为新鲜 土壤。  相似文献

One-month-old fruits of Acer ginnala with winged pericarp attached gave 44% germination and this was not increased by cold treatment at 4°C for 0, 10, 20, or 30 days, gibberellic acid treatment at 0, 1, 10, 100 or 1000 mg litre-1, or ethephon treatment at 0, 2, 20, 200 or 2000 mg litre-1. After 6 months of storage at 20–25 °C, germination of untreated fruits fell to 5% but could be restored to that of 1-month-old fruits by incubation at 4 °C for 30 days. After 9 months storage, no germination occurred in untreated fruits. Cold treatment (30 days at 4 °C partially restored germination (26%). Treatment with either gibberellic acid (1000 mg litre-1) and 30 days at 4 °C (40%) or ethephon (100 mg litre-] and 30 days at 4 °C improved germination (69%). The combination of all three treatments, i.e. 100 mg litre-1 gibberellic acid, 100 mg litre-1 ethephon and 30 days at 4 °C, optimised germination (86%). Thus, dormancy of A. ginnala developed during storage but could be reversed by a combination of treatment with low temperature and growth regulators. The highest germination (86%) was achieved after low temperature and growth regulator treatment of stored fruit.  相似文献
