Transfer of immunity to horse erythrocytes (HRBC) by immune lymphoid cells was performed to analyze the kinetics of adoptive immunity in the clonal ginbuna crucian carp, Carassius auratus langsdorfti . Immune lymphoid cells were intravascularly transferred to the unprimed recipients and then recipients were evaluated by measuring the antibody titre of the plasma. Antibody productivity was most successfully conferred by splenic cells, followed by pronephric and mesonephric cells, taken from immune donors 7 days post-immunization, while transferability by thymic cells was lacking or very low, even if possible. Peak response of plaque-forming cells (PFCs) was observed at 5–7 days after the first injection, and the maximum number of PFCs at peak response was almost the same in all organs examined, such as the pronephros, mesonephros, spleen and the thymus. Direct correlation between transferability and number of PFCs was not observed on individuals, although the peak of transferability corresponded to that of the PFC response. Preliminary experiments of cell transfer by separated pronephric cells showed that the lymphocyte-rich fraction was more effective than the bottom fraction containing fewer lymphocytes in transferring immune reactivity. These results suggest that cells involved in transferring immune reactivity are B lymphocytes, composed of different developmental stages and distributed differently in the different lymphoid organs.  相似文献

A better understanding of the immune responses in fish elicited by oral immunisation is of importance for the development of new and effective oral vaccines for cultured fish. In the present study, we characterized specific cell-mediated cytotoxic responses in isogeneic ginbuna crucian carp (Carassius auratus langsdorfii) following oral immunisation with cellular antigens. Trinitrophenyl- (TNP) or dinitrophenyl- (DNP) modified syngeneic and allogeneic cells were used for studying the fine specificity and genetic restriction of orally-induced cytotoxic cells. Hapten-specific cytotoxic responses were detected in peripheral blood leukocytes (PBLs) of fish orally immunised with haptenated syngeneic cells. PBLs from orally immunised fish had cytolytic activity for haptenated syngeneic cells, but they showed little reactivity against both haptenated and unmodified allogeneic targets. Similarly, oral immunisation of fish with hapten-modified allogeneic cells did not induce hapten-specific cytotoxic cells which can lyse haptenated syngeneic targets. Although ginbuna crucian carp possess spontaneous cytotoxic cells that are capable of killing mammalian tumour cells, cold target inhibition studies suggested that such spontaneous cytotoxic cells were not involved in the killing of haptenated syngeneic targets. Oral immunisation of fish with haptenated syngeneic cells also induced hapten-specific cytotoxic memory responses. Oral administration of haptenated fixed cells also effectively induced hapten-specific cytotoxic cells in the treated fish. These findings suggest that oral immunisation with antigens can elicit antigen-specific cytotoxic cells that are capable of recognizing antigens in an MHC-restricted manner. In addition, our results provide indirect evidence that fish possess a mechanism for taking up exogenous non-replicating antigens from the alimentary tract and generating antigen-specific cytotoxic cells.  相似文献

Cytotoxic T cells (CTLs) constitute an important component of the specific effector mechanism in killing against microbial-infected or transformed cells. In addition to these activities, recent studies in mammals have suggested that CTLs can exhibit direct antimicrobial activity. Therefore, the present investigation was conducted to find out the microbicidal activity of CD8α+ T cells of ginbuna crucian carp, Carassius auratus langsdorfii. The CD8α+ T cells from immunised ginbuna exhibited the antibacterial activity against both facultative intracellular bacteria and extracellular bacteria. The maximum reduction of viable count of pathogens was recorded with effector (sensitized) cells and target (bacteria) ratio of 10:1 co-incubated for a period of 1–2 h at 26 °C when effector cells were derived from ginbuna 7 days after one booster dose at 15th day of primary sensitization/immunisation. Sensitized CD8α+ T cells are found to kill 92.1 and 98.9% of Lactococcus garvieae and Edwardsiella tarda, respectively. No significant difference in the bacterial killing activity could be recorded against facultative intracellular bacteria and extracellular bacteria. The specificity study indicated the non-specific killing of bacteria. CD8α+ T cells from E. tarda immunised ginbuna exhibited 40% of non-specific killing activity against L. garvieae and those from L. garvieae immunised ginbuna showed 42.7% of non-specific killing activity against E. tarda. Furthermore, CD4+ T cells also killed 88% and 95.7% of L. garvieae and E. tarda, respectively. In addition to T cell subsets, surface IgM+ cells also killed both types of pathogens. Therefore, the present study demonstrated the direct antibacterial activity of CD8α+, CD4+ T-cells and surface IgM+ cells in fish.  相似文献

This study investigated the gynogenetic cytobiological behavior of the third gynogenetic generation (G3), which was generated from the diploid eggs produced by the second gynogenetic generation (G2)of red crucian carp × common carp, and determined the chromosomal numbers of G3, G2×scatter scale carp and G2×allotetraploid hybrids of red crucian carp × common carp. The results showed that the diploid eggs of G2 with 100 chromosomes, activated by UV-irradiated sperm from scatter scale carp and without the treatment for doubling the chromosomes, could develop into G3 with 100 chromosomes.Similar to the first and second gynogenetic generations (G1 and G2), G3 was also diploid (2n=100) and presented the hybrid traits. The triploids (3n=150) and tetraploids (4n=200) were produced by crossing G2 with scatter scale carp, and crossing G2 with allotetraploids, respectively. The extrusion of the second polar body in the eggs of G2 ruled out the possibility that the retention of the second polar body led to the formation of the diploid eggs. In addition, we discussed the mechanism of the formation of the diploid eggs generated by G2. The establishment of the diploid gynogenesis clonal line (G1, G2 and G3) provided the evidence that the diploid eggs were able to develop into a new diploid hybrid clonal line by gynogenesis. By producing the diploid eggs as a unique reproductive way, the diploid gynogenetic progeny of allotetrapioid hybrids of red crucian carp × common carp had important significances in both biological evolution and production application.  相似文献

In this study, we conducted a cross of white crucian carp(♀)×red crucian carp(♂)(WR), and characterized the morphology, reproduction and genetics of the progeny. Different from parents, WR with the gray color showed the hybrid morphological traits of both parents. WR possessed normal gonads producing mature eggs or sperm, and exhibited high fertilization rate(90.2%) and high hatchery rate(80.5%), which contributed to produce and enlarge the population. WR with the same DNA content as parents was a diploid fish with 100 chromosomes(2n=100). Amplified ITS of 45 S r DNA, in WR the sequences consisting of 884 bp bases of the entire ITS-1 region, 5.8 S region, and entire ITS-2 region. The sequences showed high similarity between WR and its parents and leaned towards male inheritance. In WR, NTS of 5S r DNA consisted of three length types with total 654 bp bases. From sequence analysis of NTS, WR shared 94.2% and 95.1% similarities with their female and male parent, respectively. Sequence analysis of ITS and NTS revealed that there existed recombination and variation in the hybrid progeny, which was the genetic base for adaptation and speciation. In conclusion, we obtained WR from hybridization and it exhibited hybrid traits in morphology and variation in genetic composition showing essential difference with its parents. The obtainment of WR has important significance in fish genetic breeding.  相似文献

Clonal ginbuna crucian carp is, a naturally gynogenetic fish, and is a useful model animal for studying T-cell-mediated immunity. To gain molecular information on MHC class I molecules from this species, we have identified four types of MHC class I (caauUA-S3n, caauUF-S3n, caauZE-S3n, and caauZB-S3n) and five beta 2-microglobulin (β(2)m) (caauβ2m-1a, caauβ2m-1b, caauβ2m-2, caauβ2m-3a and caauβ2m-3b) by an expressed sequence tag (EST) analysis and using homology cloning with degenerated primers. Like UA class I genes in other cyprinid fish, the caauUA-S3n shows features of classical MHC class I, such as conservation of all key amino acids interacting with antigenic peptides, and ubiquitous tissue expression. A phylogenetic analysis shows that the β(2)m-1 and β(2)m-2 isoforms are clustered with those of other cyprinid fishes, while β(2)m-3 isoforms make a cluster that is separated from a common ancestor of salmonid and cyprinid fishes. This finding suggests that the β(2)m isoforms of ginbuna cruician carp comprise two lineages and may possess different functions. The MHC class I and β(2)m sequences from one clonal strain will facilitate our understanding of the interaction of MHC class I with β(2)m in teleosts.  相似文献

As part of an experimental study of the direct and indirect effects of piscivory on prey fish, the diets of crucian carp Carassius carassius were compared across sections of a divided pond; two sections were stocked with crucian carp alone and two with crucian carp plus perch Perca fluviatilis . Analysis of crucian gut contents indicated that the composition of invertebrate prey did not differ in the presence v . absence of perch. However, crucians, particularly small individuals (<10cm) that were most vulnerable to predation, displayed a lower intake of invertebrate prey in sections with perch. Although diet composition differed between crucians caught in inshore v. offshore habitats (with habitat use being related to crucian size and the presence or absence of perch), no clear pattern existed between habitat and total food intake. Overall, the major effects of predators on the diet of crucian carp appeared to be caused by increased ecological density (resulting from confinement of small crucians inshore) and reduced activity levels, rather than simple shifts to safer habitats.  相似文献

Fishes show a remarkable diversity of shapes which have been associated with their swimming abilities and anti-predator adaptations. The crucian carp (Carassius carassius) provides an extreme example of phenotypic plasticity in body shape which makes it a unique model organism for evaluating the relationship between body form and function in fishes. In crucian carp, a deep body is induced by the presence of pike (Esox lucius), and this results in lower vulnerability to gape-limited predators, such as pike itself. Here, we demonstrate that deep-bodied crucian carp attain higher speed, acceleration and turning rate during anti-predator responses than shallow-bodied crucian carp. Therefore, a predator-induced morphology in crucian carp enhances their escape locomotor performance. The deep-bodied carp also show higher percentage of muscle mass. Therefore, their superior performance in escape swimming may be due to a combination of higher muscle power and higher thrust.  相似文献

Summary The pancreatic islets ofCarassius carassius have been studied by electron microscopy. 1. Besides A-, B- and D-cells, two new cell types, the fourth and the fifth, have been identified. The fourth cell type is numerous; it occurs interposed among the other types of islet cells or in small clusters. The secretory granules (90–280 mg in diameter) are round or oval and usually with much lower electron density than α- and δ-granules. The secretory granules of the fifth type of cell (approximately 140–240 mμ in diameter) contain finely granular material and an electron dense core that is round or often tetra- or hexagonal. 2. The islet cells with clear cytoplasmic matrix generally contain large numbers of fine, agranular and cored vesicles 400–680 ? in diameter. They appear, in bead-like chains, or randomly scattered throughout the cytoplasm, or often clustered in aggregates close to the secretory granules and show evidence of incorporation into the secretory granules. The two types of vesicles may be formed by constriction or pinching-off of the tubular smooth endoplasmic reticulum.  相似文献

An improved triploid crucian carp (ITCC) was produced by crossing improved tetraploids (G1×AT, ) with improved red crucian carp (IRCC, ), which were obtained by distant crossing and gynogenesis. The biological characteristics of ITCC, including the number and karyotype of chromosomes, gonadad and pituitary structures, phenotype, and growth rate are reported. ITCC possessed 150 chromosomes with the karyotype 33m+51sm+33st+33t. In the breeding season, both ovary-like and testis-like gonads of ITCC were unable to produce normal mature gametes. The ultrastructure of the pituitary of ITCC showed that most of the endocrine granules in gonadotrophic hormone (GTH) cells had not been re-leased, providing endocrinological evidence for the sterility of ITCC. Compared with triploid crucian carp (TCC) produced by mating Japanese crucian carp with allotetraploid hybrids, ITCC not only re-tained the excellent traits of fast growth rate and sterility, but also acquired improved morphological characteristics, including higher body, shorter tail and smaller head.  相似文献

目的 :研究迷走神经感觉传入信息对Mauthner细胞 (M细胞 )兴奋性的影响。方法 :刺激鲫鱼迷走神经 ,并运用微电极穿刺技术记录鲫鱼M细胞胞内电位变化。结果 :在M细胞胞内记录到分级的、复合的兴奋性突触后电位(EPSP) ,分为第一成分和第二成分。随着刺激强度的增大 ,EPSP的幅度增大 ,反应持续时间延长。当刺激强度足够大时 ,在第一成分或第二成分的基础上可爆发动作电位。结论 :①刺激迷走神经可引起M细胞顺向激活 ,这与以往的观点不同 ;②从迷走神经到M细胞的感觉传入通路可能由含有兴奋性和抑制性成分的不同种类的神经链构成 ,M细胞的兴奋性取决于兴奋和抑制之间的相互关系  相似文献

PCR-RFLP analysis of the ND3/ND4L/ND4 and 12S/16S rRNA regions and nucleotide sequence variation of the cytochrome b gene were used to study the mtDNA divergence in species of the family Cyprinidae, to examine the phylogenetic relationships of the species, and to identify their taxonomic status. The results indicated that an ancestral form diverged into silver crucian carp and crucian carp after its separation from the common carp lineage. The divergence of continental Carassius auratus gibelio and Japanese C. auratus cuvieri occurred more recently. Two well distinguishable mtDNA phylogroups, suggesting divergent evolution, were observed in continental C. auratus gibelio populations. The divergence was possibly related to the formation of two silver crucian carp groups with different types of reproduction, triploid gynogenetic and diploid gonochoric. At the same time, the results supported the high probability of current genetic exchange between the forms. In view of these findings and high morphological similarity of the two forms, they were not considered to be separate species.  相似文献

Crucian carp (Carassius carassius) develop a deeper body in response to chemical cues from piscivores. This change in body morphology has been suggested to be a predator-induced defence. Here we investigate the possible benefits of the induced body morphology in laboratory experiments. Pike foraging behaviour when feeding on crucian carp of different body depths was recorded using video. Further, in a preference experiment pike were allowed to choose between shallow-bodied and deep-bodied crucian carp of similar lengths. Crucian carp body morphology did not affect predatory behaviours (activity, searching, following, observing, capture success) in northern pike, but an increase in crucian carp body depth led to an increase in handling time in pike. In the preference experiment, pike preferred shallow-bodied crucian carp over deep-bodied. Thus, a change in body morphology, induced by the presence of piscivores, benefits crucian carp by increasing piscivore handling times and an avoidance of the deep-bodied phenotype.  相似文献

对26尾三倍体湘云鲫的线粒体tRNA-Thr基因、tRNA-Pro基因和部分控制区的核苷酸序列进行了测定,获得26条长度为837—839 bp的同源基因序列,共发现65个多态性核苷酸变异位点,多态位点比例为0.077,定义了8种单元型。在湘云鲫8种单元型中确认了DNA复制终止相关的序列TAS、中央保守区序列(CSB-F、CSB-E和CSB-D)和保守序列CSB1,8种单元型含有3—5个TAS序列。在65个变异位点中,大部分序列变异为转换,8种单元型之间的序列差异在0.1%—6.3%之间。该研究为三倍体湘云鲫的繁殖和遗传改良提供了一些有价值的信息。  相似文献

Cell proliferation and gill morphology in anoxic crucian carp   总被引:1,自引:0,他引:1  
Is DNA replication/cell proliferation in vertebrates possible during anoxia? The oxygen dependence of ribonucleotide reductase (RNR) could lead to a stop in DNA synthesis, thereby making anoxic DNA replication impossible. We have studied this question in an anoxia-tolerant vertebrate, the crucian carp (Carassius carassius), by examining 5'-bromo-2'-deoxyuridine incorporation and proliferating cell nuclear antigen levels in the gills, intestinal crypts, and liver. We exposed crucian carp to 1 and 7 days of anoxia followed by 7 days of reoxygenation. There was a reduced incidence of S-phase cells (from 12.2 to 5.0%) in gills during anoxia, which coincided with a concomitant increase of G(0) cells. Anoxia also decreased the number of S-phase cells in intestine (from 8.1 to 1.8%). No change in the fraction of S-phase cells ( approximately 1%) in liver was found. Thus new S-phase cells after 7 days of anoxia were present in all tissues, revealing a considerable rate of DNA synthesis. Subsequently, the oxygen-dependent subunit of crucian carp RNR (RNRR2) was cloned. We found no differences in amino acids involved in radical generation and availability of the iron center compared with mouse, which could have explained reduced oxygen dependence. Furthermore, the amount of RNRR2 mRNA in gills did not decrease throughout anoxia exposure. These results indicate that crucian carp is able to sustain some cell proliferation in anoxia, possibly because RNRR2 retains its tyrosyl radical in anoxia, and that the replication machinery is still maintained. Although hypoxia triggers a 7.5-fold increase of respiratory surface area in crucian carp, this response was not triggered in anoxia.  相似文献
