Genetic engineering of plant resistance characteristics against fungi may unintentionally influence traits that are important for plant–herbivore interactions. We studied the palatability of transgenic birch (Betula pendula), aspen (Populus tremula) and hybrid aspen (P. tremula x tremuloides) genetically modified with the aim to improve fungal disease resistance, to selective mammalian herbivores in cafeteria tests. Roe deer (Capreolus capreolus) were fed with transgenic birch carrying a sugar beet chitinase IV gene. In the experiment with roe deer, none of the six transgenic birch lines differed significantly from the wild-type control in the proportion of consumed plant biomass. Correlation analyses suggested that sugar content did not guide the feeding preferences of roe deer but revealed a positive correlation between starch content and proportion of mass consumed. However, the variation in starch content could not be related to the level of transgene expression. Mountain hares (Lepus timidus) were fed with plant material from chitinase transgenic birch and aspen and hybrid aspen that carried a pinosylvin synthase gene from Scots pine. One transgenic birch line was significantly less palatable to hares than the wild-type control. The results of this study suggest that plant genotype may be related to the palatability of plant material when transgenic and wild-type woody plant material is used as winter food for hares. The results of this case study did not reveal changes in the palatability of the studied transgenic lines that could be readily related to the functioning of the used transgenes.  相似文献

From 1998 to 2000, serum samples of 80 shot European brown hares (Lepus europaeus) from Argentina were examined for antibodies against European brown hare syndrome virus (EBHSV) and 80 spleen samples were tested for EBHSV-antigen by enzyme linked immunosorbent assay (ELISA). Nine hares were positive for EBHSV-antigen. Antibodies against EBHSV were detected in only one individual. Based on negative staining electron microscopy of spleen homogenates, we observed calicivirus in one of five EBHSV-antigen positive hares. However, EBHS has not been reported to cause abnormal mortality in these hares. This is the first report of antibodies to EBHSV, EBHSV-antigen, and electron microscopy findings in free-ranging European brown hares from South America.  相似文献

From 1997 to 2000, complete necropsy and histopathologic investigations were performed on 157 free-ranging European brown hares (Lepus europaeus) found dead throughout Switzerland. Organ samples of all these individuals (157 livers and 107 spleens available) were tested for European brown hare syndrome virus (EBHSV)-antigen by enzyme-linked immunosorbent assay (ELISA) test kit. Furthermore, 60 additional blood samples were tested for antibodies against EBHSV by ELISA. In addition, liver samples of 87 free-ranging mountain hares (Lepus timidus) hunted in 1996 were tested for EBHSV-antigen. In two European brown hares from southern Switzerland lesions suggestive of changes induced by EBHSV were present, and high titers of EBHSV-antigen were detected in both liver and spleen samples of these animals. Based on negative staining electron microscopy investigations of liver and spleen homogenates, we observed calicivirus in one antigen-positive hare. Low EBHSV-antigen titers were found in three additional European brown hares from central and western Switzerland, but EBHS-lesions were absent. Antibodies against EBHSV were not detected in any of the sera of European brown hares, and EBHSV-antigen was not found in the samples of mountain hares. This is the first report of EBHS in European brown hares from Switzerland.  相似文献

European brown hare populations have declined during the last decades. Agricultural intensification has been identified as a relevant driver of this process and agri-environment schemes have been implemented to foster biodiversity in agricultural landscapes. Because species-specific outcomes of measures strongly depend on tailored design of the policy framework and the local management, while changing climate may pose additional challenges, policy and management need science-based information of which landscape composition should be promoted to achieve set biodiversity goals.Here, we used direct observations of European brown hares over 20 years for evaluating the effects of landscape composition and weather conditions on European brown hare density. For the first time, our analysis compared the estimates of machine learning (gradient boosting machine) and linear mixed models in terms of importance of a wide range of explanatory variables for European brown hare densities and effect trends.Scattered woody vegetation, as represented by the two variables transitional woodland-shrub and small woody features, was on top rankings among the predictors and greater proportions of these elements were accompanied by sharp increases of European brown hare density. Also warmer winter temperature had a positive effect.We conclude that promoting scattered woody vegetation in agricultural landscapes is a powerful tool for improving European brown hare habitat quality. Particularly with the increasing dynamic in agriculture due to climate change, incentives and regulations that create a long-lasting heterogeneity in the landscape composition through near-natural elements can support the population of this popular mammal.  相似文献

  • 1 The European roe deer Capreolus capreolus is a typical faunal element of the Holocene. It was already present in Europe at least 600 000 years ago and it has been known from both glacial and interglacial phases since then. With nearly 3000 fossil and subfossil records, it is one of the most frequent mammals in the Late Quaternary.
  • 2 During the Middle and Late Weichselian Pleniglacial, the distribution of the roe deer was not restricted to the Mediterranean peninsulas but repeatedly reached regions of central Europe. In contrast to that, roe deer records from the Last Glacial Maximum (LGM, 21.0–14.5 ka 14C BP) are largely confined to the Mediterranean peninsulas – with the exception of south‐western France and the surroundings of the Carpathians where several records attest to its occurrence during the LGM.
  • 3 During the Greenland Interstadial 1 (12.5–10.8 ka 14C BP), the species' distribution extended further north and the roe deer appeared north of the Alps and reached regions of central Germany. This seems to be correlated with the abrupt change to more favourable environmental conditions during this period. It is very likely that the roe deer disappeared north of the Alps during the Younger Dryas cooling (10.8–10.0 ka14C BP). The northern regions of the central European lowlands were recolonized by roe deer during the late Preboreal 9.7–9.5 ka 14C BP for the first time since the Weichselian Glacial.
  • 4 The combined pattern of genetic data and fossil records of European roe deer suggests several regions in the Iberian peninsula, southern France, Italy and the Balkans as well as in the Carpathians and/or eastern Europe as glacial refugia. It further suggests that C. capreolus might have recolonized most parts of central‐northern Europe out of one or more eastern European (not Balkan) and/or Carpathian refugia. This recolonization wave might have blocked immigration from the traditional Mediterranean areas.

The European roe deer (Capreolus capreolus) is an interesting model for molecular ecology studies because of its abundance and adaptability across a range of environments (including human‐modified habitats), and because of its increasing impacts on agricultural crops and on regenerating forests. We identify polymorphic microsatellites in two managed populations of roe deer in France by using cross‐species amplification of primers from other Cervids and from Bovids. Of the 62 primer pairs tested, 45 amplified microsatellites in roe deer, and 34 were polymorphic. Eleven primer pairs were selected for multiplex gel‐loading for routine genotyping of the studied populations.  相似文献

In this study, we demonstrate that the mountain hare and roe deer compete with each other. This was determined using "natural experiments" of populations found in sympatry and allopatry on the islands along the west coast of Norway. We demonstrate that both species occupy the same habitats, share the same food resources and that food availability is limited. Two browsing species as different in size as roe deer and mountain hare might be expected to partition the available vegetation (e.g. woody scrub) in terms of height above ground level. However, from the evidence collected, the feeding-height-separation hypothesis must be rejected as an explanation for ecological separation between roe deer and mountain hares because there was extensive height overlap in resource utilisation by both species and neither species changed its feeding height in response to the presence of the other. Total browse utilisation did not increase when both species were together; rather, species-specific browse utilisation declined when the other species was present. However, the foraging behaviour of each herbivore varied significantly between the allopatric and sympatric sites. When both herbivores were present, the clip diameter of shoots browsed by mountain hares declined to match those selected by roe deer, while roe deer switched from a browse-dominated diet to a diet dominated by winter-green gramineae. The change to smaller-diameter shoots likely resulted in the hare increasing its intake of digestion-inhibiting or toxic secondary metabolites, while the alternative choice of digging through the snow like roe deer to reach the winter-green gramineae is a practice considered energetically too costly for hares. On this basis, we conclude that the enforced switch to a nutritionally inferior diet by mountain hares at the sympatric sites may result in changes to growth rate and body size which consequently impact on mortality and may explain the competitive superiority of the roe deer.  相似文献

Introgressive hybridization is a widespread evolutionary phenomenon which may lead to increased allelic variation at selective neutral loci and to transfer of fitness‐related traits to introgressed lineages. We inferred the population genetic structure of the European roe deer (Capreolus capreolus) in Poland from mitochondrial (CR and cyt b) and sex‐linked markers (ZFX, SRY, DBY4 and DBY8). Analyses of CR mtDNA sequences from 452 individuals indicated widespread introgression of Siberian roe deer (C. pygargus) mtDNA in the European roe deer genome, 2000 km from the current distribution range of C. pygargus. Introgressed individuals constituted 16.6% of the deer studied. Nearly 75% of them possessed haplotypes belonging to the group which arose 23 kyr ago and have not been detected within the natural range of Siberian roe deer, indicating that majority of present introgression has ancient origin. Unlike the mtDNA results, sex‐specific markers did not show signs of introgression. Species distribution modelling analyses suggested that C. pygargus could have extended its range as far west as Central Europe after last glacial maximum. The main hybridization event was probably associated with range expansion of the most abundant European roe deer lineage from western refugia and took place in Central Europe after the Younger Dryas (10.8–10.0 ka BP). Initially, introgressed mtDNA variants could have spread out on the wave of expansion through the mechanism of gene surfing, reaching high frequencies in European roe deer populations and leading to observed asymmetrical gene flow. Human‐mediated introductions of C. pygargus had minimal effect on the extent of mtDNA introgression.  相似文献

Macroscopic and histological study of a case of unilateral hydrocephalus in a young wild European brown hare (Lepus europaeus) is reported here. Morphological changes were mostly massive dilatation of the left lateral ventricle, atrophy of the cortex and a striking reduction of the corresponding white matter. Lymphocytic cell infiltrates and hemosiderin-laden macrophages in the choroid plexus and meninges also were observed. Although the exact cause of the lesions was not found all findings suggested a hydrocephalus of possible traumatic, infectious or congenital hypoplasia origin.  相似文献

The digestive capacity of free-living moose Alces alces (L.) and roe deer Capreolus capreolus (L.) for four plant species was studied with in vitro technique. Moose were found to digest all species better than roe deer. Individual variations were also smaller for moose. In moose the level of digestibility was the same both summer and winter, while roe deer shot in winter digested winter samples of bilberry Vaccinium myrtillus L. and to some extent Scots pine Pinus silvestris L. better than did summer roe deer. This was taken as evidence for a seasonal adaptation in the roe deer rumen. Digestibility of the plant material studied showed a strong negative correlation to the fiber content. The results are discussed in relation to winter problems for roe deer and possible food competition between moose and roe deer.  相似文献

In the early 1800s, the European roe deer (Capreolus capreolus) was probably extirpated from Switzerland, due to overhunting and deforestation. After a federal law was enacted in 1875 to protect lactating females and young, and limiting the hunting season, the roe deer successfully recovered and recolonized Switzerland. In this study, we use mitochondrial DNA and nuclear DNA markers to investigate the recolonization and assess contemporary genetic structure in relation to broad topographic features, in order to understand underlying ecological processes, inform future roe deer management strategies, and explore the opportunity for development of forensic traceability tools. The results concerning the recolonization origin support natural, multidirectional immigration from neighboring countries. We further demonstrate that there is evidence of weak genetic differentiation within Switzerland among topographic regions. Finally, we conclude that the genetic data support the recognition of a single roe deer management unit within Switzerland, within which there is a potential for broad‐scale geographic origin assignment using nuclear markers to support law enforcement.  相似文献

The progressive decline in the hare population across Europe has been associated with the occurrence of European brown hare syndrome (EBHS), a highly contagious disease considered endemic in all European countries. This study aimed to evaluate the in-field temporal dynamics of European brown hare syndrome virus (EBHSV) infection in wild European brown hares (Lepus europaeus) and to test the influence of population density on EBHS seroprevalence. A total of 512 blood samples were collected from free ranging hares captured for restocking in seven different areas of the province of Brescia (Northern Italy) during seven consecutive years (2006–2013) and tested using a competitive ELISA. A generalized linear mixed model estimated the yearly effects of population density on EBHS prevalence. Of the 512 tested, 344 (67.2 %) tested positive for EBHSV antibodies, with the annual seroprevalence ranging from 94.3 to 35.8 %. The prevalence was 3.303 times higher in areas with a density of over 15 hares/km2 and declined over the years. The results indicate the ongoing transmission of the virus in the tested brown hare population. Since the eradication of EBHS in a wild population is not feasible, a strategy aimed at promoting the endemic stability of the virus through density-dependent mechanisms could be applied; however, this seems more difficult in practice than in theory and would most likely require a very high density of brown hares.  相似文献

The extant taxa of central and northern Europe are commonly believed to derive from Pleistocene ancestors, who moved to the north from three separate glacial refugia: the Iberian and Italian peninsulae, as well as the southern Balkans. The issue of postglacial dispersal patterns was addressed through the investigation of population structure and phylogeography of the European roe deer, Capreolus capreolus . The genetic diversity in 376 individuals representing 14 allegedly native populations across their European range was assessed, using ten autosomal microsatellite loci and restriction fragment length polymorphisms of the mitochondrial D-loop and NADH dehydrogenase 1 gene segments. Our results suggest the existence of three major genetic lineages of roe deer in Europe. One comprises populations in the south-western limit of the species' distribution (i.e. Iberia), where an internal substructure splits a northern from a southern sublineage. A second lineage includes populations of southern and eastern Europe, as well as a separate sublineage sampled in central-southern Italy, where the existence of the subspecies Capreolus c. italicus was supported. In central-northern Europe, a third lineage is present, which appeared genetically rather homogeneous, although admixed, and equally divergent from both the eastern and western lineages. Current patterns of intraspecific genetic variation suggest that postglacial recolonization routes of this cervid to northern Europe could be due to range expansion from one or more refugia in central-eastern Europe, rather than proceeding from the Mediterranean areas.  © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 88 , 85–100.  相似文献

A substantial portion of today’s biodiversity is attributed to the climatic oscillations of the Pleistocene Ice Ages. Gradual but dramatic climate changes were accompanied by expansion, contraction, and isolation of populations, promoting the accumulation of genome differences and adaptations in refugial populations and resulting in allopatric differentiation in a variety of taxa. In the present study, partial mitochondrial DNA sequences of the widely distributed European brown hare (Lepus europaeus) were analyzed to test whether the species’ present genetic structure is the result of postglacial re-colonization of Europe from Asia Minor (clade A) and the Balkans (clade B) only, as suggested previously, or if additional refugia are likely. Analyses indicated the presence of an additional refugium (Italy, clade I). The genealogic network of Italian hares displayed the tree-like structure expected from refugial populations, whereas central European brown hare haplotypes revealed a clear star-phylogeny indicative of past-bottleneck population growth. This population size expansion, which was confirmed by mismatch analysis, was estimated to have occurred ∼50–55 thousand years ago (kya). The divergence of clade A* from the remaining matrilines is estimated at 239 kya, whereas the divergence of the ancestors of clades B* and I from A* occurred about 128 kya. Dispersal is frequently more prevalent in one sex compared to the other. Greenwood proposed that patterns of sex-biased dispersal among birds and mammals are linked to their mating strategies. For species where males defend resources rather than females, he predicted female-biased dispersal, because males should remain at their birth site where they are familiar with the distribution of the resources that they must defend. Greenwood's hypothesis has been extensively supported among birds, where most species exhibit a resource-defence mating strategy. However, almost no equivalent information is available for mammals as males generally defend mates in this group. An exception is the European roe deer, a resource-defence mating ungulate. We thus tested Greenwood's hypothesis on this atypical mammalian model, looking for female-biased dispersal using sex-specific inter-individual genetic distances. We conclusively show that gene flow is not higher among females compared to males in the studied roe deer population, and hence that dispersal is not female-biased, suggesting that male mating strategy is not the primary selective force driving the evolution of dispersal in roe deer. We discuss the role of female mate choice and intra-sexual competition as possible alternative selective pressures involved.  相似文献
