Uniformly labelled [14C]glucose was introduced into the xylem of segments of willow stem. Forty-eight hours later sieve-tube sucrose was collected via servered aphid stylets, and the distribution of radioactivity in the hexose moieties of this was compared with the distribution in those of sucrose extracted from the segment. Very little correlation was found between the two sets of values, indicating possible inversion during loading. This lack of correlation could not be attributed to contributions to the sieve-tube sucrose from pools of labelled hexoses in the segment. Further experiments, however, showed quite high degrees of correlation between sieve-tube sucrose and sucrose extracted from the wood, indicating that the latter tissue was a major source of sieve-tube sucrose. This conclusion was substantiated in experiments in which sieve-tube exudate, obtained from stem segments, was compared with exudate obtained from the isolated bark of the segment. In other experiments, stylets were established on stem segments, then on isolated pieces of bark obtained from these segments. Sucrose and potassium exudation rates fell by as much as 50% on removing the bark from the segment. It was not possible to formulate a precise figure for the contribution of the wood to stylet exudation owing to injury effects and the complexity of the experimental system. No firm evidence could be found in support of the view that sucrose is inverted during loading of sieve elements from the storage cells of the stem.  相似文献

ABSTRACT. The paper describes a radio frequency microcautery operating at 27 MHz. Unusually high power is used to achieve a high rate of temperature rise in the tissue in order to leave fluid transporting vessels unblocked, e.g. aphid stylets. The power is adjustable down to levels appropriate to normal radio cautery applications and a timer is provided for single pulse operation. A special-purpose thermionic valve is used to generate the power which is fed to a remote probe.  相似文献

On the basis of the distribution of radioactivity between sucrose,organic phosphates, and hexose in sieve-tube exudate obtainedvia severed stylets, it has been postulated (Peel and Ford,1968), that two pathways exist for the movement of sugars fromthe bathing solution into the sieve elements of bark strips.These pathways were designated the direct and the indirect.Theexperiments described in the present paper show that the distributionof radioactivity in stylet exudate when labelled sugars aremoving by the indirect pathway may be influenced by the simultaneoustransport of other solutes. Phosphates moving by either thedirect or indirect pathway can influence the activity distributionin stylet exudate, as can sugars moving by the direct pathway.  相似文献

Ricinus communis L. seedlings exuded pure phloem sap from the cut hypocotyl for several hours. Throughout the entire exudation period proteins were present in the phloem exudate at a constant concentration ranging from 0.11 to 0.41 mg·ml–1 depending on the culture conditions and the age of the seedlings. Manipulation of the nutrient supply at the cotyledons after removal of the endosperm did not change the protein concentration in the exudate. Comparison of sieve-tube exudate proteins (STEPs) with soluble proteins extracted from the hypocotyl and the cotyledons showed a unique abundance of small proteins in the exudate, with molecular weights ranging from 10 to 25 kDa. Bands at 18, 19 and 20 kDa were especially dominant. The proteins found transiently in the xylem exudate, which might represent proteins secreted at the wound surface, were different in pattern. Two-dimensional separation of STEPs revealed that more than 100 distinct polypeptides occurred in the sieve-tube exudate, most of them slightly acidic with isoelectric points ranging from 4 to 6 and a few basic ones around 8. [35S]Methionine fed to the cotyledons led to labelling of STEPs, demonstrating their rapid synthesis. The mature, functional sieve-tube system in higher plants is dependent upon protein import from the companion cells to maintain a functional long-distance transport system. Soluble proteins present within the sieve-tube lumen were investigated by analysis of sieve-tube exudates which revealed the presence of distinct sets of polypeptides in seven monocotyledonous and dicotyledonous plant species. Antibodies directed against sieve-tube exudate proteins from Ricinus communis L. demonstrated the presence of shared antigens in the phloem sap collected from Triticum aestivum L., Oryza sativa L., Yucca filamentosa L., Cucurbita maxima Duch., Robinia pseudoacacia L. and Tilia platyphyllos L. Specific antibodies were employed to identify major polypeptides. Molecular chaperones related to Rubisco-subunit-binding protein and cyclophilin, as well as ubiquitin and the redox proteins, thioredoxin h and glutaredoxin, were detected in the sieve-tube exudate of all species examined. Actin and profilin, a modulator of actin polymerization, were also present in all analyzed phloem exudates. However, some proteins were highly species-specific, e.g. cystatin, a protease-inhibitor was present in R. communis but was not detected in exudates from other species, and orthologs of the well-known squash phloem lectin, phloem protein 2, were only identified in the sieve-tube exudate of R. communis and R. pseudoacacia. These findings are discussed in terms of the likely roles played by phloem proteins in the maintenance and function of the enucleate sieve-tube system of higher plants. ABSTRACT. A technique is described for cutting the mouth-parts of feeding aphids with a radio frequency probe, and for collecting plant sap from the severed ends of the stylets without contamination. The technique is a significant improvement on previous methods used, and will allow reliable quantitative analysis to be made of plant sap on which the aphids feed.  相似文献

Sieve-tube exudate protein (STEP) from Ricinus communis L. seedlings consists of a characteristic set of more than 100 different polypeptides, against which a complex antiserum was raised. This antiserum cross-reacted with dominant protein species (molecular weights 10–30 kDa) present in the sieve-tube exudate and, to a lesser extent, with proteins in tissue extracts of Ricinus and a wide range of other plant species. For further elucidation of the nature of individual STEPs in the sieve tubes the anti-STEP serum was used to screen a cDNA expression library constructed from Ricinus cotyledon mRNA. Two clones that differed in the 3′ untranslated region encoded a protein of 11 kDa which showed striking homology to bacterial and eucaryotic glutaredoxin sequences. Glutaredoxin activity was confirmed for the recombinant protein after overexpression in Escherichia coli and characterised in detail in sieve-tube exudate. Michaelis Menten constants (K m) for reduced glutathione and cysteine were 2 mM and 50 μM, respectively. Besides l-cysteine, dehydroascorbate and protein disulphides were also reduced by the activity present in the sieve-tube exudate. Glutathione, which is the obligate donor of reduced thiols for glutaredoxin, was present in sieve-tube sap in millimolar concentrations (up to 3 mM) with a ratio of total to oxidised glutathione of 3:1. It is suggested that glutaredoxin and glutathione in sieve tubes prevent oxidative damage and may be involved in redox regulation of sieve-tube proteins. In the rhizosphere, plants are exposed to a multitude of different biotic and abiotic factors, to which they respond by exuding a wide range of secondary root metabolites. So far, it has been unknown to which degree root exudate composition is species‐specific and is affected by land use, the local impact and local neighborhood under field conditions. In this study, root exudates of 10 common grassland species were analyzed, each five of forbs and grasses, in the German Biodiversity Exploratories using a combined phytometer and untargeted liquid chromatography‐mass spectrometry (LC‐MS) approach. Redundancy analysis and hierarchical clustering revealed a large set of semi‐polar metabolites common to all species in addition to species‐specific metabolites. Chemical richness and exudate composition revealed that forbs, such as Plantago lanceolata and Galium species, exuded more species‐specific metabolites than grasses. Grasses instead were primarily affected by environmental conditions. In both forbs and grasses, plant functional traits had only a minor impact on plant root exudation patterns. Overall, our results demonstrate the feasibility of obtaining and untargeted profiling of semi‐polar metabolites under field condition and allow a deeper view in the exudation of plants in a natural grassland community.  相似文献

Experiments are described which attempt to clarify the quantitative relationship between sieve-tube loading of sucrose, and ATP-turnover rates in the phloem of willow (Salix viminalis L.). Two experimental approaches have been made towards the solution of this problem. In the first of these the respiratory breakdown of 14C-sugars was measured in segments of willow stem when no sieve-tube transport was taking place, and also under conditions where transport was occurring in response to the feeding of individuals of the aphid Tuberolachnus salignus (Gmelin). An increase in respiratory activity, measured by the output of 14CO2, was found to occur as a consequence of transport. Since the rate of sieve-tube sugar loading could be measured by the production of honeydew from the aphids, and by making assumptions concerning the production of ATP in respiration, it was concluded that the stoichiometry of sucrose loading was 1.9 mol ATP · (mol sucrose)-1. A somewhat higher value of 2.5 mol ATP · (mol sucrose)-1 was found using the second approach. In this, attempts were made to measure ATP turnover rates using [32P]orthophosphate supplied to strips of willow bark which bore exuding aphid stylets.  相似文献

Abscisic acid (ABA) was identified by combined gas liquid chromatography-mass spectrometry in sieve-tube exudate collected from the cut stylar ends of white lupin fruit. Water stress caused an increase in ABA levels in leaf, seed and pod tissues and phloem exudate. When compared with levels in extracts of these tissues, the concentration of ABA in sieve-tube sap was very high. The willow aphid Tuberolachnus salignus feeds by inserting itsstylets into a single sieve element. If the insect is severedfrom its proboscis whilst feeding, sap from the sieve tube exudesfrom the cut end of the stylet canal and may continue for manyhours. By attaching a capillary tube the rate of exudation canbe measured and the sap analysed. Normally the rate of exudationwas about 1 mm.3 per h. This means a movement of water and sucroseof 100 cm./h. or 100 sieve elements per min. The sap was foundto contain between 5 and 15 per cent, sucrose, up to 0.4 percent, raffinose, no reducing sugars, and about 0.5 per cent,amino-acids. Girdles some distance above and below exuding stylets failedto stop exudation (Tables I and II). This indicates a rapidsealing of the cut ends of the sieve tube and a switch-overin source of supply from the leaves to storage cells in thestem. The use of isolated stem segments and even irrigated stripsof bark followed. Using these, the following points have beenestablished, (a) There is no polarity of movement (Table IV);(b) a minimal length of about 16 cm. of stem or 800–1,000sieve elements are needed to give the full exudation rate (Fig.5); (c) when the D.P.D. on the inside of the bark is raised(Figs. 6, 7, and 8), the rate of exudation declines, but theconcentration of the exudate rises and exudation continues,indicating a maintenance of turgor, even in face of a D.P.D.of 20 atm. or more. Secretion of sugar into the sieve tube continuesalbeit slowly even against a sieve tube concentration of 50per cent. These findings are discussed in relation to current theoriesof translocation and are considered to favour the mass flowhypothesis.  相似文献

We performed electron-microscopic examination of structural diurnal changes in the lumen of sieve tubes and the vacuolar system of corresponding companion cells and changes induced by the experimental blockage of assimilate export from the leaf by its cold-girdling. For these investigations, Cucurbita pepo L. and Helianthus annuus L. plants were used, that is, plant species from groups of symplastic and apoplastic plants, which differ in the type of companion cells and a mode of phloem terminal loading. The examinations showed the complete identity of changes in the electron texture of the sieve-tube lumens and companion-cell vacuoles in both plant species in the course of a day, when the level of assimilates changed, or after export blockage. Similar changes in the structure of the vacuolar labyrinths were stated in the companion cells under normal conditions and after cold-girdling, as related to the rate of sieve-tube loading with the vacuolar exudate. Vacuolar expansion and starch accumulation developing in response to changes in the assimilate level in the evening and after cold blockage of the assimilate export occurred in different types of cells, as dependent on their position in the symplast domains. However, the rate of the process similarly depended on the balance between assimilate synthesis and export. Synchronous changes in the texture of the sieve-tube lumen and companion-cell vacuoles were observed within each complex, but asynchronous changes occurred in different complexes. We suggested this phenomenon for recognizing the particular complexes, when they are grouped in a bundle. We observed no signs of cytoplasm or protein synthetic machinery in the sieve tubes. We concluded that the sieve-tube lumen and vacuoles of companion cells are common in nature. Similar electron texture of the images of the companion-cell vacuolar labyrinth and tube lumens, their connection through the lateral sieve fields, morphological modifications of the companion-cell vacuolar system as dependent on the activity of sieve tube loading—all of these facts imply the continuity of these transport compartments and fluxes in them and the similarity in the composition of the exudates from companion-cell vacuoles and phloem tubes.  相似文献

Antibodies were raised against lectin purified from the sieve-tube exudate of Cucurbita maxima. Immunocytochemistry, using peroxidase-labelled antibodies and Protein A-colloidal gold, was employed to determine the location of the lectin within the tissues and cells of C. maxima and other cucurbit species. The anti-lectin antibodies bound to P-protein aggregates in sieve elements and companion cells, predominantly in the extrafascicular phloem of C. maxima. This may reflect the low rate of translocation in these cells. Under the electron microscope, the lectin was shown to be a component of P-protein filaments and was also found in association with the sieve-tube reticulum which lines the plasmalemma. The anti-lectin antibodies reacted with sieve-tube proteins from other species of the genus Cucurbita but showed only limited reaction with other genera. This study investigated advantages and drawbacks of two sieve-tube sap sampling methods for comparison of phloem proteins in powdery mildew-infested vs. non-infested Hordeum vulgare plants. In one approach, sieve tube sap was collected by stylectomy. Aphid stylets were cut and immediately covered with silicon oil to prevent any contamination or modification of exudates. In this way, a maximum of 1muL pure phloem sap could be obtained per hour. Interestingly, after pathogen infection exudation from microcauterized stylets was reduced to less than 40% of control plants, suggesting that powdery mildew induced sieve tube-occlusion mechanisms. In contrast to the laborious stylectomy, facilitated exudation using EDTA to prevent calcium-mediated callose formation is quick and easy with a large volume yield. After two-dimensional (2D) electrophoresis, a digital overlay of the protein sets extracted from EDTA solutions and stylet exudates showed that some major spots were the same with both sampling techniques. However, EDTA exudates also contained large amounts of contaminative proteins of unknown origin. A combinatory approach may be most favourable for studies in which the protein composition of phloem sap is compared between control and pathogen-infected plants. Facilitated exudation may be applied for subtractive identification of differentially expressed proteins by 2D/mass spectrometry, which requires large amounts of protein. A reference gel loaded with pure phloem sap from stylectomy may be useful for confirmation of phloem origin of candidate spots by digital overlay. The method provides a novel opportunity to study differential expression of phloem proteins in monocotyledonous plant species.  相似文献

To access phloem sap, aphids have developed a furtive strategy, their stylets progressing towards sieve tubes mainly through the apoplasmic compartment. Aphid feeding requires that they overcome a number of plant responses, ranging from sieve tube occlusion and activation of phytohormone-signalling pathways to expression of anti-insect molecules. In addition to bypassing plant defences, aphids have been shown to affect plant primary metabolism, which could be a strategy to improve phloem sap composition in nutrients required for their growth. During compatible interactions, leading to successful feeding and reproduction, aphids cause alterations in their host plant, including morphological changes, modified resource allocation and various local as well as systemic symptoms. Repeated salivary secretions injected from the first probe in the epidermal tissue up to ingestion of sieve-tube sap may play a crucial role in the compatibility between the aphid and the plant.  相似文献

The cotyledons of castor bean (Ricinus communis L.) act as absorption organs for amino acids, which are supplied to the medium. The analysis of the sieve-tube sap, which exudes from the cut hypocotyl, demonstrated the ability of the cotyledons to load particular amino acids into the phloem and to reject the loading of others. The sieve-tube sap of cotyledons, which were embedded in the endosperm, contained 150 mM amino acids, with 50 mM glutamine as the major amino acid, and 10–15 mM each of valine, isoleucine, lysine and arginine. Removal of the endosperm led to a drastic decline in the amino-acid content of sieve-tube sap down to 16 mM. Addition of single amino acid species to the medium increased the amino acid concentration in the sieve-tube sap in specific manner: glutamine caused the largest increase (up to 140 mM in exudate), glutamate and alanine smaller increases (up to 60 mM), and arginine the smallest. In addition, the amino acid composition of the sieve-tube sap changed, for instance, glutamine or alanine readily appeared in the sieve-tube sap upon incubation in glutamine or alanine, respectively, whereas glutamate was hardly discernible even in the case of incubation with glutamate; arginine was loaded into the sieve tubes only reluctantly. In general, glutamine and alanine accumulated four- to tenfold in the sieve tubes. The uptake of amino acids and of sucrose into the sieve tubes was interdependent: the loading of sucrose strongly reduced the amino acid concentration in the sieve-tube exudate and loading of amino acids decreased the sucrose concentration. Comparison of the concentrations of various amino acids on their way from the endosperm via the cotyledon-endosperm interface, through the cotyledons and into the sieve tubes showed that glutamine, valine, isoleucine and lysine are accumulated on this pathway, whereas glutamate and arginine are more concentrated in the cotyledons than in the sieve tubes. Obviously the phloem-loading system has a transport specificity different from that of the amino acid uptake system of the cotyledon in general and it strongly discriminates between amino acids within the cotyledons.  相似文献

Many insect folivores sever veins or cut trenches before feeding on leaves that emit fluid from latex, resin, or phloem canals. The behaviors reportedly function to reduce the insect's exposure to exudate. This hypothesis was tested with three species that sever leaf midribs and two species that trench. In each case, the insect's cuts reduced exudation at the distal feeding site by at least 94% relative to an adjacent control leaf. However, most of the insects contacted exudate during the vein-cutting/trenching operation, which could potentially negate any benefits received during feeding. To estimate how much exudate red milkweed beetles (Tetraopes tetrophthalmus) ingest, I weighed beetles before and after vein cutting, and simulated beetle feeding on leaves with and without vein cuts. By severing veins, the beetles decreased their total ingestion of exudate by at least 92%.  相似文献

Comparisons were made between the free amino acid composition in leaf exudates and that in pure phloem sap, using twin samples taken from a single leaf of two oat (Avena sativa L.) and three barley (Hordeum vulgare L.) varieties. Leaf exudate was collected in a 5 mm EDTA-solution (pH 7.0) from cut leaf blades and phloem sap was obtained through excised aphid (Rhopalosiphum padi L.) stylets. Fluorescent derivatives of amino acids were obtained using 9-fluorenylmethyl chloroformate and were separated by means of high performance liquid chromatography. The total concentration of free amino acids varied considerably in the exudate samples. There was no correlation between the total amino acid content in the exudate samples and that of the corresponding phloem sap samples, but the amino acid composition of the corresponding samples was highly correlated (median R2-value 0.848). There was only limited between-plant variation in phloem sap amino acid composition. Nevertheless, in comparisons involving all samples, many of the amino acids showed significant correlations between their relative amounts in exudate and phloem sap. The results presented here indicate that the exudate technique holds great promise as an interesting alternative to the laborious and time-consuming stylet-cutting technique of obtaining samples for comparative studies of phloem sap.  相似文献

Unmodified samples of barley (Hordeum vulgare) sieve tube sap have been obtained by severing the stylets (stylectomy) of feeding aphids and collecting the exuding liquid. Primers were designed to direct the amplification of a series of specific cDNAs encoding barley proteins selected because of their significance in sieve tube function. mRNA encoding the H(+)/sucrose co-transporter SUT1, a putative aquaporin and the H(+)/ATPase PPA1 were detected in sieve tube sap. These mRNA species appear to be present at very low concentrations. mRNA encoding the potassium transporter HAK1 could not be detected. The results strongly suggest that some mRNA species are imported into sieve elements, which are enucleate, from neighbouring companion cells.  相似文献

This literature review concerns insect responses to fire, compared to other feasible and appropriate conservation managements of open habitats. Many insect groups decline markedly immediately after fire, with the magnitude of reduction related to the degree of exposure to the flames and mobility of the insect. Niche diversity is lower in recently burned habitat, and the rate of insect increase following fire also relates to the species' ability to gain access to the regrowing vegetation. Postburn flora can be quite attractive to some recolonizing insects, possibly to some degree a result of fire-caused insect mortality which provides plants with short-term release from insect herbivory. Insect declines may follow immediately after mowing, but usually of lesser degree and shorter duration than after a fire of comparable timing and size. Season and scale of cutting may affect how much and which species showed positive or negative responses. Cut areas offer the vegetational structure and composition preferred by some insects, but cutting – or cutting at certain scales, seasons, or frequencies – may also be unfavorable for some species. Heavy grazing results in niche and assemblage simplification. Nonetheless, some invertebrates prefer the short turfs and bare ground resulting from heavier grazing. Other species vary in whether they peak in abundance and diversity in intermediate, light, or no grazing. In comparisons of mowing/haying and grazing regimes of similar compatibility with maintenance of the same habitat types, responses of particular species and species groups varied as to whether they had a preference for one or the other. Characteristics associated with insect responses to fire related to the degree of exposure to lethal temperature and stress experienced in the post-fire environment, suitability of post-treatment vegetation as habitat, and ability to rebuild numbers in the site (from survivors and/or colonizers). These factors appear equally useful for explicating insect responses to other managements such as haying, mowing, and grazing. By contrast, the assumption that the most habitat-restricted species will be most adapted to ecological forces believed to be prevalent in that ecosystem appears less efficacious for predicting insect management preferences.  相似文献
