The survival of a braconid parasitoid Habrobracon hebetor was investigated on nucleopolyhedrovirus (NPV)-infected Spodoptera exigua larvae. The second-instar larvae were exposed to 30, 51.4 and 180 PIB/mm2 of Mamestra brassicae NPV (MbMNPV) as under-LD50, LD50 and over-LD50 values, respectively. They were accessible to be parasitized by H. hebetor after 24, 48 and 72 h post-treatment. Infection of the larvae with MbNPV was deleterious to the survival and parasitism of H. hebetor. The survival of H. hebetor in MbNPV-infected S. exigua larvae was dependent on the interval between viral infection and parasitization, as well as on the treatment dose of MbMNPV; very few adults of parasitoid emerged from infected hosts when host larvae were exposed to 180 PIB/mm2 of MbNPV on 72-h interval treatment. The inoculation dose of MbNPV and the timing of parasitoid release had significant effect on the development of H. hebetor on virus-infected hosts. Field applications of virus for biocontrol of S. exigua may lead to substantial mortality of immature parasitoids.  相似文献

Variations in the resistance to nuclear polyhedrosis virus (NPV) were found in three populations of Spodoptera littoralis. The LD50 for the most resistant population was 1.07 × 104 PIB/5th instar larva as compared to 8.4 × 102 and 5.8 × 102 PIB/larva in the other two populations. The effect of NPV persisted in larvae which survived and pupated. Some of the pupae died, and those which survived produced normally shaped adults. While fecundity was sharply reduced in the less resistant populations, the effect on the most tolerant population was less pronounced. A 3-year-old inoculum, stored unprotected from daylight and without cooling, was much less effective even against the most sensitive larval population as compared to a relatively fresh and refrigerated batch. Larvae in their 6th instar proved to be approximately 10-fold more resistant to the NPV than 5th instar ones, while the difference in weight was only about twice. These variations in resistance to NPV are also discussed from the point of view of applying S. littoralis NPV in pest control schemes.  相似文献

The susceptibility of Heliothis armiger larvae of different ages to a commercial nuclear polyhedrosis virus (NPV), Elcar, was determined by bioassay. The median lethal dosage (LD50) increased 150-fold during the first week of larval life at 25°C, i.e., during development to early fourth instar, but daily feeding rate and thus potential virus acquisition also increased. A linear relationship was determined between log LD50 and larval length, indicating that larval length constitutes a useful index for estimating the susceptibility of larval populations. Median lethal times (LT50s) were similar for larvae tested at ages of 0 to 7 days and ranged from 3.6 to 8.0 days at 30°C. The amount of virus produced in a single, infected neonate was equivalent to 1.4 × 106 LD50s for neonates, a 900,000-fold increase on the dose supplied. The data support the practice of directing the NPV against neonates, but, on the basis of larval susceptibility alone, the age of larvae at treatment may not always be critical.  相似文献

Protoplasts of strain 458 (S458) and strain 521 (S521) of Entomophthora egressa had LD50s of 620 and 8.8 cells/insect, respectively, for sixth-instar spruce budworm larvae. Both protoplast strains exhibited biphasic growth profiles with comparable growth rates in the larval hemocoel. The growth rates of the fungal strains increased as the total hemocyte counts declined. Hemocytopenia was greatest and most rapid in larvae containing S521 protoplasts. Protoplasts of S521 exposed to α-mannosidase and β-N-acetylglucosaminidase were as virulent as the control cells. β-Galactosidase reduced protoplast virulence.  相似文献

A relatively fast and simple peroral technique for the bioassay of insect viruses is described in which newly hatched larvae ingest a uniform volume of virus suspension. Three isolates of the Autographa californica nuclear polyhedrosis virus (NPV) and one isolate of the Heliothis zea NPV were used to test the procedure with Trichoplusia ni and H. zea larvae, respectively. Within-assay and between-assay variation was very low with coefficients of variation averaging 0.012 ± 0.006 and 0.20 ± 0.04 for time-mortality and dose-mortality tests, respectively. The synchronous uptake of virus removed the acquisition-time component of the LT50 values while the constant volume improved the accuracy of LD50 values. The procedure was shown to be suitable for a wide variety of lepidopterous species, including Spodoptera frugiperda, S. eridania, Estigmene acrea, Plutella xylostella, Choristoneura fumiferana, Ostrinia nubilalis, Plodia interpunctella, and Pieris rapae.  相似文献

Pseudomonas aeruginosa was identified as a facultative pathogen of red palm weevil. Intra-haemocoelic injection of the pathogen within larvae and pre-pupae was more effective at killing the insects [with a median lethal dose (LD50) of 9×102 to 2×103 bacteria/insect] than inoculation by force feeding (LD50 of 105 to 4×105 bacteria/insect) or by wading the insects in a suspension of the pathogen (LD50 of 105 to 2×105 bacteria/insect). Injection of 3×103 bacteria/insect killed 69% of larvae; small larvae were more susceptible (LD50 of 9×105 bacteria/larva) than either larger larvae (LD50 of 103 bacteria/larva) or pre-pupa. The median time to death of the small larvae following injection of P. aeruginosa was about 6 days but that following force feeding or wading was about 8 days. A secondary invader, Serratia marcescens, had no effect on the pathogenicity of P. aeruginosa but hastened death of larvae by about 3 days.A. Banerjee and T.K. Baculovirus isolates from the same host species often show a considerable degree of variation on phenotypes. The completely sequenced genotypes C1 and G4 of Helicoverpa armigera single-nucleocapsid nucleopolyhedrovirus (HaSNPV) were compared. Bioassay studies suggested that nearly double of HaSNPV G4 virus was required compared with HaSNPV C1 to achieve a similar LD50, and at the LD90 level the insect-killing speed for HaSNPV C1 was quicker than that of HaSNPV G4. The budded virus (BV) production of HaSNPV C1 was nearly two- to threefold higher at 24 and 48 h post-infection (p.i.) than that of HaSNPV G4. However, the kinetics of polyhedral inclusion body (PIB) formation in HzAM1 cells was similar in both the genotypes, which implied that the insect-killing speed was not influenced by PIB formation, but by the kinetics of BV production. The results suggested that the HaSNPV C1 isolate was a better choice than HaSNPV G4 virus for controlling H. armigera.  相似文献

Conventional pesticides have suffered two main drawbacks: (a) broad spectrum of action and (b) selection of target species resistant to the different active ingredients. Thus compounds that are less harmful to the environment and to human health, more specific and that do not induce resistance need to be developed. One alternative are insect growth regulators, such as fluazuron. The present study examined the efficacy of fluazuron (active ingredient of the acaricide Acatak®) and the sensitivity of Rhipicephalus sanguineus nymphs exposed to different doses of this chemical, and determined the lethal doses of fluazuron: 95% – LD95 and 50% – LD50. Different doses of fluazuron were applied in duplicates on the dorsal region of hosts (“pour on”). Distilled water was used in the control group. On the first day after the treatment with fluazuron, hosts were artificially infested with R. sanguineus nymphs. After engorgement, nymphs were removed, placed on Petri dishes, identified, and maintained in BOD incubator for 15 days. Dead R. sanguineus nymphs after the treatment with 13 different doses of fluazuron were quantified and the LD95 was estimated to be 100 mg/kg and LD50, 19.544 mg/kg (12.478–22.636), with a confidence interval of 95%. Nymphs of R. sanguineus were sensitive to fluazuron at various levels, indicating that this insect growth regulator (IGR) may be used to control this parasite in this stage of its biological cycle, reducing the significant damage it causes.  相似文献

Resistance to a nucleopolyhedrosis virus (Baculovirus heliothis) did not develop in laboratory populations of the cotton bollworm, Heliothis zea. A selection pressure of LD50 to 70 was maintained throughout 20 to 25 generations of selection. No significant changes in LD50, slope, or intercept of dose-mortality lines were detected. Laboratory populations under selection were as susceptible to the virus as nonselected or wild populations of H. zea. The resistance ratio (LD50 of selected generation/initial generation) ranged from 0.5 to 1.2.  相似文献

Polyhedral inclusion body protein (PIBP), harvested from the nuclear polyhedrosis virus (NPV) infecting the gypsy moth, Porthetria dispar, illicits the hemagglutination of chicken erythrocytes. Antisera to PIBP, polyhedral inclusion bodies (PIB), and virions (RODS) from the NPV's infecting P. dispar and the European Pine Sawfly, Neodiprion sertifer, inhibits hemagglutination when utilized to neutralize the PIBP from P. dispar. The crossreactivity of antisera to viral components from N. sertifer, a hymenopteran insect, with viral antigens from P. dispar, a lepidopteran insect, demonstrates a serological relationship exists between two viruses which have widely separated host ranges.  相似文献

The fumigant toxicity of 66 plant essential oils to Plutella xylostella (L.) larvae and Cotesia glomerata (L.) adults was examined using a vapor-phase toxicity bioassay and compared with that of dichlorvos. Responses varied according to oil and insect species used. Based on 24 h LD50 values, pennyroyal oil [10.77 mg/filter paper (4.25 cm diameter)] was the most toxic fumigant, followed by rosemary and sage (Dalmatin) oils (15.15 mg/paper). Potent fumigant toxicity was also produced from armoise, buchu leaf, cedarleaf, coriander, eucalyptus, howood, lavender, myrtle, niaouli, peppermint, and rosewood oils (LD50, 21.29–27.31 mg/paper). All essential oils were less effective than dichlorvos (LD50, 0.52 mg/paper). Against adult C. glomerata, dichlorvos (LD50, 0.03 mg/paper) was the most toxic fumigant, whereas the LD50 values of the 14 essential oils ranged from 1.59 to 8.51 mg/paper. Based on selective toxicity ratio (STR, P. xylostella LD50/C. glomerata LD50), the 14 essential oils (STR, 2.5–14.5) are more selective than dichlorvos (STR, 17.3). The essential oils tested merit further study as potential fumigants for the control of P. xylostella in greenhouses because of their selective toxicity to adult C. glomerata and their much greater activity as a fumigant.  相似文献

It has been reported that the susceptibility of insect herbivores to entomoviruses is affected by phytochemicals ingested during the acquisition of viral inoculum on the foliage of host plants. However, the relationship between this susceptibility and phytochemicals is poorly understood. To test this hypothesis of plant‐mediated effects on this susceptibility, we measured the effects of foliage from three plants, soybeans (Glycine max), collards (Brassica oleracea) and water convolvuluses (Ipomoea aquatica), on the susceptibility of larval beet armyworm (Spodoptera exigua) to nucleopolyhedrovirus (NPV), and analysed six foliar chemicals (total phenolics, peroxidase [POD], catalase [CAT], superoxide dismutase [SOD], endochitinase and exochitinase) in the three plants, respectively. The results of exponential modelling indicated that the LD50s (median lethal dose) of NPV to larvae increased with the increase in both phenolics and POD but declined with the increase in four other foliar chemicals, while the opposite trend was found between median lethal time (LT50) of NPV and the six foliar chemicals. This study reveals that phenolics and POD decrease host susceptibility to the entomoviruses and that CAT, SOD, endochitinase and exochitinase increase this susceptibility.  相似文献

The macrophage migration inhibition test (MMI), an in vitro correlate of delayed hypersensitivity, was found to be an effective means of differentiating Trichoplusia ni and Autographa californica multiple embedded nuclear polyhedrosis viruses (NPV). Peritoneal exudate cells from guinea pigs sensitized to virions of T. ni NPV demonstrated significantly different MMI when challenged with T. ni vs A. californica virions. Similarly, when virions of A. californica NPV were employed as the sensitizing antigen, different percentages of MMI were observed in the homologuus versus heterologous challenges.The susceptibility of the fall armyworm, Spodoptera frugiperda, to these two pathogens was very similar as revealed by a comparison of LD50's, slopes, and fiducial limits of dose-mortality regression lines. In contrast, the cabbage looper, T. ni, was much more susceptible to A. californica NPV.The polyhedral sizes, shapes, and virion occlusion patterns of the two species of NPV were virtually indistinguishable.  相似文献

Host-pathogen relationships were studied between the pink bollworm, Pectinophora gossypiella, and a cytoplasmic polyhedrosis virus (CPV). Results showed that the median effective dose (ED50), the dose that infects half the test subjects, was 1.91 × 102 polyhedral inclusion bodies (PIB)/ml of diet. The median lethal dose (LD50) was 1.72 × 105 PIB/ml. Diagnosis for CPV infection was more reliable in adult pink bollworms than in late-instar larvae. Duration of the larval stage increased with viral dose, but duration of the pupal stage was not affected by CPV. Weights of infected male and female pupae were 23.7 and 24.0% less than those of untreated pupae, respectively. Pupal weights were not significantly influenced by increases in the viral dose. Weights of larvae of a given age decreased as dose increased. The effect of CPV on duration of the immature stages and on pupal weight was not significantly influenced by rearing temperatures between 25.0° and 32.5°C. Pupal weight of infected pink bollworms decreased as the duration of the larval stage increased.  相似文献

Changes in the population size and geographical distribution of the mesoparasitic copepod Pennella sp. (Siphonostomatoida: Pennellidae) on the saury, Cololabis saira, were studied for a period of six years (1981–86) in the western North Pacific Ocean and adjacent seas. The parasite was first recorded in the western North Pacific in 1981. Its population size increased rapidly in 1982–83 and declined slightly in 1984. During these years, infected fish were widely distributed in the western and central North Pacific and also found in the Sea of Okhotsk and the Sea of Japan. However, the population declined dramatically in 1985 and its distributional range was reduced. The parasite disappeared in 1986. The observed frequency distributions of parasites on the host population were the Poisson in 1981 and 1985, but those during 1982–1984 were over-dispersed and fitted the negative binomial.Parasites of the saury, Cololabis saira (Brevoort)-IV.  相似文献

In vitro metabolism study was conducted to observe the role of enzymes and its kind responsible for tolerance in metabolism of flupyrazofos to Diamond backmoth (DBM), Plutella xylostella as a susceptible insect and Beet armyworm (BAW), Spodoptera exigua as a tolerance insect. The results indicated that there were some differences between the DBM (LD50, 0.54 ppm) and BAW (LD50, >2,000 ppm). In the microsomal fractions, the 4th instar BAW larvae metabolized almost the same amount (about 38%) of flupyrazofos with or without NADPH, but it increased the amount of flupyrazol in the 3rd instar larvae of DBM. Flupyrazofos-oxon was slightly increased with the addition of NADPH in both species. With the addition of piperonyl butoxide (PB) or Iprobenfos (IBP), the amount of flupyrazol was decreased to one third of flupyrazol produced by NADPH in BAW and one fourth of flupyrazol produced by NADPH in DBM. Some effects of inhibitors, PB and IBP, and co-factor, NADPH, were showed when flupyrazofos was metabolized with or without them. This means that esterase and oxidase are responsible for the tolerance mechanism of flupyrazofos in DBM and BAW.  相似文献

Three cell lines of Heliothis zea and one cell line of Heliothis virescens replicated the singleembedded, nuclear polyhedrosis virus (NPV) of H. Zea, (i.e., Baculovirus heliothis) with concomitant production of polyhedral inclusion bodies (PIB). Between 20 and 60% of the H. zea cells produced PIB, whereas only 3% of H. virescens cells were found to produce PIB. The H. zea cell lines produced 10 to 20 times more PIB than did the H. virescens cell line. The PIB from all cell lines produced typical symptoms of an NPV infection when bioassayed against larvae of H. zea. More than 99% of the total viral activity of the final whole culture was due to the PIB.  相似文献

Third-instar Spodoptera exempta larvae were fed on young maize leaves treated with 20 μl of polyhedral inclusion body (PIB) suspension of concentrations that varied from 1.6 × 102 to 1.6 × 109 PIBs/ml. Daily observations were kept on mortality rates. A probit analysis on the results gave an LD50 value of 48.4 PIBs/larva (lower and upper fiducial limits 39.2 and 59.4 PIBs/larva, respectively), and an LT50 that varied from 146.2 to 221.3 hr, depending on the dosage. LD and LT values obtained show the high pathogenicity of S. exempta nuclear polyhedrosis virus to its host.  相似文献

Antiapoptotic genes of baculoviruses have been shown to prevent virus induced apoptosis in insect cells. Dot blot and Southern hybridizations of EcoRI genomic library and genomic digests of Spodoptera litura nucleopolyhedrosis virus (SlNPV) respectively give strong hybridization signals with antiapoptotic DNA (p35 gene) probe of the prototype Autographa californica nucleopolyhedrosis virus (AcNPV). Both the hybridizations indicate the presence of a homologous gene in the 1.8 kb EcoRI-Y fragment of SlNPV. The sequence of 1.244 kb region of this fragment encompasses an open reading frame coding for a polypeptide of 296 amino acids under sequential early (TATA) and late (TAAG) promoter motifs like that in other baculovirus p35 genes. The putative SlNPV p35 ORF expresses abundantly as a 35 kDa protein in Spodoptera frugiperda (Sf9) cells when allowed to express under the polyhedrin promoter of AcNPV.  相似文献
