The tryptic peptides comprising the aminoethylated β-hemoglobin chain of Macaca nemestrina were isolated by paper electrophoresis and chromatography. The results of stains for specific amino acids, comparison of peptide maps with those produced by aminoethlated β chains from human hemoglobin A, and amino acid analyses of all peptides in the macaque β chain support the conclusion that the primary structure of the β chain of M. nemestrina is identical to that of the corresponding chains of M. fuscata fuscata and M. fascicularis. The apparent evolutionary conservatism of macaque β chains is discussed in relation to the wide degree of structural variation previously observed among the α-hemoglobin chains of several species of Macaca.  相似文献

One hemaglobin was found in the adult rhesus monkey (Macaca mulatta) by the technique of isoelectric focusing. In addition to the adult hemoglobin, one fetal hemaglobin was observed in neonates of the same species. The alpha and non-alpha globins of rhesus monkey hemoglobins showed similar electrophoretic mobilities as those of human hemoglobin A by cellulose acetate membrane electrophoresis. It appeared that the rhesus monkey hemoglobin system is a good model system for the study of human hemoglobin development.  相似文献

The amino acid sequences of 28 soluble tryptic peptides from chorismate mutase/prephenate dehydratase from Escherichia coli K12 have been determined. Together with the four unique cysteine-containing peptides sequenced by Gething and Davidson ((1976) Eur. J. Biochem. 71, 327-336) this accounts for approximately 75% of the total sequence expected for this protein. A high frequency of identify between some of the peptides suggests the possibility of gene duplication during the evolution of the structural gene for the enzyme.  相似文献

The amino acid sequence of the alpha-chain of trout hemoglobin (Hb) IV is given, thus completing the primary structure of the hemoglobin component of trout's blood characterized by the Root effect. The trout Hb IV alpha-chain consists of 142 amino acid residues; comparison with the corresponding sequences from human and carp hemoglobins shows differences of 50.0 and 35.9%, respectively. A difference of 39.6% is found with the alpha-chain of trout Hb I, the other major hemoglobin component of trout blood, devoid of heterotropic effects.  相似文献

S-Carboxymethylated L-asparaginase was digested with trypsin and the resulting peptides were isolated by using gel filtration, ion exchange column chromatography and paper chromatography. Among the peptides thus isolated, 27 peptides were considered not to overlap and the sum of the amino acids from these 27 peptides is in good agreement with amino acid composition of the enzyme. The amino acid sequences of the peptides were determined by fragmentation with various enzymes and subtractive Edman degradation.  相似文献

Summary The amino acid compositions of the tryptic peptides of the following carnivore hemoglobin chains have been determined: gray fox (Urocyon cineroargenteus); raccoon (Procyon lotor); polar bear (Thalarctos maritimus); coati mundi (Nasua nasua) chain; coati mundi (Nasua narica) two chains; cat (Felis catus) chain; and lion (Panthera leo) chain. These provide a basis for future sequencing of these hemoglobins and construction of an evolutionary tree. Adrenal glands from Rhesus monkeys (Macaca mulatta) of 160 days gestation, newborn, 2 months-old infants or 6 months-old infants were excised and prepared, by a collagenase digestion, as a cell suspension. The cells were incubated with 10 pg/ml, 100 pg/ml or 1 ng/ml of a peptide of the ACTH/pro-opiomelanocortin 'family', 57K, 31K, 20K, alpha MSH, ovine-CLIP or gamma LPH either in the presence or absence of 166 pg/ml ACTH1-39. The production by cortisol and androstenedione was measured by radioimmunoassay. Using the steroid production by aliquots of the cell suspension with either no stimulating agent or ACTH1-39 alone as controls, the net influence of these different peptides on basal or ACTH1-39-stimulated production was observed. alpha MSH, ovine-CLIP and gamma LPH had no influence on either basal or stimulated cortisol or androstenedione production. Corticotrophic peptides of 57K, and 20K and pro-opiomelanocortin each had a steroidogenic activity alone, in all age groups. In the fetal and newborn monkeys' adrenal cells, peptides of 57K and 20K at 1 ng/ml had an inhibitory influence on ACTH1-39 stimulated cortisol and androstenedione production. The influence of the 20K peptide is partially inhibitory as the steroidogenic potential of this peptide is not additive with that of ACTH1-39. These results show that, as observed in other species, that the ACTH/pro-opiomelanocortin range of peptides are inhibitory to the action of ACTH1-39 in the developing adrenal.  相似文献

The complete amino acid sequence of the gamma chain from the major one of two fetal hemoglobins from the rhesus monkey, Macaca mulatta, was determined by automated, stepwise degradation of selected fragments produced by cleavage at methionyl and tryptophanyl residues and at the single aspartylprolyl bond. The minor fetal hemoglobin is single aspartylprolyl bond. The minor fetal hemoglobin is similar to human Hb F1 in relative electrophoretic and chromatographic properties and in the level at which it is found (about 12% of the total Hb F). On these grounds, we assume that this minor component contains, like Hb FI, gamma chains that differ from those of the major component by virtue of acetylation of their amino-terminal glycyl residues. Although the gamma chains of most antropoid primates examined to date are structurally heterogeneous and, hence, appear to be encoded by nonallelic genes, no sign of structural heterogeneity was detected at any position in the major gamma chain from M. mulatta. Thus, if nonallelic gamma-chain genes exist in this species, the chains encoded by them may be identical in sequence. The gamma chain from M. mulatta is but the sixth primate gamma chain whose primary structure has been fully characteerized. The slight extent of structural divergence among these chains (the four chains from various species of Old World monkeys differ from one another by no more than two substitutions, while the human and cercopithecoid gamma chains differ at no more than five sites) attests to the conservative nature of gamma-chain evolution among the higher primates.  相似文献

The technique of isoelectric focusing on immobilized pH gradients (IPG) has been applied to the analysis of tryptic digests of alpha- and beta-chains of human hemoglobin. Using peptides purified by RP-HPLC as a reference, it was possible to create a peptide map in the single IEF dimension. Unfortunately, it was not possible to find experimental conditions (medium for migration and staining) which would allow the detection of peptides of less than 10-12 residues. Almost all the bands visible on the gel could be assigned to known peptides. In order to obtain these results the IPG runs were performed in 8 M urea containing 0.5% carrier ampholytes and the gel stained with colloidal Coomassie brilliant blue G-250, in the presence of a high-salt concentration and at acidic pH.  相似文献
